Abstract:

Abstract:
Disasters are defined as events caused either by natural or technological occurrences, that overwhelm the resources that are immediately available to manage or mitigate the impact of the event. Disasters, by their very nature, are newsmakers. With the improvement in telecommunications, the barriers of distance are reduced to the extent that the people of the world all are members of what Marshall MacLuhan called the “global village.” It now is a common practice to watch the effects of a disaster on the other side of the world, from the safety and comfort of the living room, live on television. The capacity to empathize with the victims, and to feel almost a part of the incident, results in tremendous public attention and an urging of governments, not directly affected by the event, to get involved and “do something!”

Abstract:

Abstract. The accelerated techno-scientific development has brought with it profound changes in advanced societies that are particularly felt in teaching in all disciplinary areas. The massive presence of educational resources derived from the development of information technology is gradually integrated into teaching practices, as evidenced by the history of Educational Technology at its stage of recent development. We see how the various teaching materials were conceived, using technological devices, and how teachers integrated them in their practice. Similar to the Gutenberg Galaxy and Marconi Galaxy, consecrated by McLuhan, the Internet Galaxy, acclaimed by Castells reminds us clearly the relationship between technological development and societal, cultural and educational changes. Traditional teaching materials and resources continue to play a key role in teaching, however, the latest technological developments challenge us as teachers and researchers better exploit their use and integration through what they bring as new promoters of better learning. This article aims to identify a set of curricular integration possibilities of ICT in physical education teaching, exploring the potential of video technology, interactive whiteboard (digital blackboard), the Web 2.0 applications that open a very wide range and full of possibilities for teaching physical education.

Abstract:
Melanogenesis is the biological process which the skin pigment melanin is synthesized to protect the skin against ultraviolet irradiation and other external stresses. Abnormal biology of melanocytes is closely associated with depigmented skin disorders such as vitiligo. In this study, we examined the effects of maclurin on melanogenesis and cytoprotection. Maclurin enhanced cellular tyrosinase activity as well as cellular melanin levels. We found that maclurin treatment increased the expression of microphthalmia-associated transcription factor (MITF), tyrosinase-related protein- (TRP-) 1, TRP-2, and tyrosinase. Mechanistically, maclurin promoted melanogenesis through cyclic adenosine monophosphate (cAMP) response element binding (CREB) protein-dependent upregulation of MITF. CREB activation was found to be mediated by p38 mitogen-activated protein kinase (MAPK) or cAMP-protein kinase A (PKA) signaling. In addition, maclurin-induced CREB phosphorylation was mediated through the activation of both the cAMP/PKA and the p38 MAPK signaling pathways. Maclurin-induced suppression of p44/42 MAPK activation also contributed to its melanogenic activity. Furthermore, maclurin showed protective effects against H2O2 treatment and UVB irradiation in human melanocytes. These findings indicate that the melanogenic effects of maclurin depend on increased MITF gene expression, which is mediated by the activation of both p38 MAPK/CREB and cAMP/PKA/CREB signaling. Our results thus suggest that maclurin could be useful as a protective agent against hypopigmented skin disorders.

Abstract:
Benzo[a]pyrene (B[a]P), a polycyclic aromatic hydrocarbon formed during the incomplete combustion of organic matter, has harmful effects. Therefore, much research is ongoing to develop agents that can mitigate the effects of B[a]P. The aim of this study was to examine the effect of maclurin, one component of the branches of Morus alba L., on the B[a]P-induced effects in HaCaT cells, a human keratinocyte cell line. Maclurin treatment inhibited aryl hydrocarbon receptor (AHR) signaling as evidenced by reduced xenobiotic response element (XRE) reporter activity, decreased expression of cytochrome P450 1A1 (CYP1A1), and reduced nuclear translocation of AHR. The B[a]P-induced dissociation of AHR from AHR-interacting protein (AIP) was suppressed by maclurin. Maclurin also inhibited the production of intracellular reactive oxygen species (ROS) induced by B[a]P. In addition, the antioxidant property of maclurin itself was demonstrated by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Furthermore, maclurin activated antioxidant response element (ARE) signaling through enhancement of ARE luciferase reporter activity and the expression of ARE-dependent genes including nuclear factor (erythroid-derived 2)-like 2 (Nrf2) and heme oxygenase-1 (HO-1). Nrf2 activation and its nuclear translocation were promoted by maclurin through p38 MAPK activation. These data indicate that maclurin had antagonistic activity against B[a]P effects through activation of Nrf2-mediated signaling and inhibition of AHR signaling and, suggesting its potential in protecting from harmful B[a]P-containing pollutants.

Badge, Joanne Louise. "The molecular characterisation of Narcissus latent virus and Maclura mosaic virus." Thesis, University of Leicester, 1997. http://hdl.handle.net/2381/35608.

Abstract:
Narcissus latent virus (NLV) and Madura mosaic virus (MacMV) are serologically related. However, they have poor serological relationships with other plant viruses with which they have shared characteristics. Coat protein size, particle shape and structure, mode of vector transmission, cytology and serology proved insufficient to classify them. Molecular techniques were employed in order to create tools for the rapid and accurate classification of plant viruses. A carlavirus-specific PCR primer test failed to amplify NLV or MacMV but confirmed that several other viruses belonged to the carlavirus genus. The nucleotide sequences of part of the nuclear inclusion body (NIb) gene, the complete coat protein gene and the 3' untranslated regions of narcissus latent virus (NLV) and Madura mosaic virus (MacMV) were determined. Deduced amino acid sequences for the Nib and coat protein genes revealed that NLV and MacMV are closely related. Comparison of the NIb sequences with other viruses showed that NLV and MacMV have closer affinities with viruses of the Potyviridae than to those of the carlavirus genus with which they were initially classified. It is proposed that NLV and MacMV may form a new genus within the Potyviridae, the Macluraviruses. The viruses associated with narcissus yellow stripe disease were re-evaluated. In order to identify further members for the new genus a second PCR primer was designed to amplify a region shared by the bymoviruses and macluraviruses. Sequence data obtained for the 3'-terminal region of rice necrosis mosaic virus (RNMV) using a fragment amplified by this primer confirmed that RNMV was a member of the bymovirus genus. MacMV and barley mild mosaic bymovirus replicase sequences were used to transform tobacco plants in an attempt to create transgenic resistance.

The imminence of Panambi s dam construction, in the municipality of Alecrim, Rio Grande do Sul, affecting the municipality of Porto Mauá, which will overflow thousands of hectares of rainforest, Macluratinctoria s habitat and, the great exploitation of the specie in the region led to the initiation of this research work. The specie is commonly known as Tajuva, presents arboreal sized, and belongs to the botanical family Moraceae, native of Rio Grande do Sul. It is a semideciduous specie, with heights between 10 to 30 meters and, up to 100 cm of diameter; short trunk, with a smooth bark, grayish green with many yellowed lenticels, many times, with the presence of branched thorns. The study aimed to the realization of anatomical description, the chemical characterization and the basic and weighted densities of Maclura tinctoria(L) D. Don Ex Steud wood. The five studied exemplars were obtained through environmental license, in accordance with current legislation. Three wood samples, taken from BHD were used for the manufacture of histological slides and for the fibers quantification. To the determination of wood density and the chemical analysis of wood, disks were removed from the trunk and divided into opposing wedges. It were used opposing wedges removed from six pre-established heights (base, DAP, 25%, 50%, 75% and 100% of commercial height). It was used the ASTM standards to determinate the density, and from TAPPI to the chemical analysis of wood. The wood was anatomically described and illustrated with photomicrographs. The woody tissue consist mainly of fibers (60 ± 3,4%) and rays (20 ± 2,3%), totaling approximately 80%. The remaining, in other words, 20% are divided into vessels (12 ± 4%) and axial parenchyma (8 ± 3%). The mean values, to the basic density (pb) and weighted density (ppond), were 0,539 and 0,555 g/cm³, respectively. The macromolecular constituents of heartwood and sapwood were determinate separately. The average content found for extractives (18,92; 6,32%); Klason lignin (17,58; 17,55%); holocellulose (63,45; 78,59%); alpha-cellulose ( 35,44; 44,38%); hemicelluloses (28,01; 34,21%) and ashes (1,08; 1,35%), to the heartwood and sapwood, respectively, totaling 102,42 ± 3,1%. The Tajuva s wood brings together the anatomical characteristics prevalent in Moraceae, framing the specie in their respective botanical family. It is considered an evolved specie anatomically. The density presents a decrease in the values in relation to the axial position of the plant. The heartwood is constituted of phenolic compounds and acids in a higher proportion relative to the sapwood, which presents higher starch contents. Studies about the natural durability and the wood quality are fundamental to indicate the best application of wood.
Popularmente, a espécie é conhecida como Tajuva, apresenta porte arbóreo, e pertence à família botânica Moraceae, nativa do Rio Grande do Sul.

In the course of our ongoing search for new anticancer natural products, two species Garcinia oblongifolia and Garcinia oliveri have been studied. We isolated some new polyisoprenylated benzoylphloroglucinols called oblongifolines and some others, which have already been described like guttiferone F which exhibits inhibitory activity of the disassembly of microtubules in tubulin. Guttiferones B and F have never been isolated before from the genus Garcinia. A third plant belonging to the Maclura genus, M. Tricuspidata, Moraceae revealed a significant cytotoxic activity on KB cells. Some other species which have already been studied were rich in analogues of benzoylphloroglucinols. The specie M. Tricuspidata has an interesting cytotoxic activity on KB cells. We studied this specie and isolated some isoflavones and two of those are new: macluspidatines A et B. We observed a positive cytotoxic activity of 6,8-diprenylorobol on KB cells whereas the activity of wighteone was already described

Abstract:
### ABSTRACT/ - An alternative method using sea sand as a sample disruptor is presented here for the extraction of phenolic compounds from plant material. The sea sand disruption method (SSDM) was compared with MSPD (Matrix solid phase dispersion) and SLE (Solid-liquid extraction) for the extraction of several families of phenolic compounds, namely, xanthones, flavanones, phenolic acids, flavonols and coumarins in plant material from two plants from, Moraceae family, Madura pomifera and Ficus carica, using dichloromethane and methanol: water mixtures as eluents. The extraction yields obtained using SSDM were similar or superior to those obtained with MSPD and SLE, for all compounds extracted. When compared with SLE, SSDM has the advantages of less solvent consumption and less sample preparation time; and when compared with the MSPD method, the use of sea sand avoids the cost of the expensive C8 or C18 solid support materials. Several replicates were performed for each extraction procedure and statistical treatment, ANOVA-single factor, was used to compare the extraction yields obtained by these methods. The extraction procedures were reproducible as the RSD values were less than 5% for almost all compounds and extraction procedures. To evaluate recovery by SSDM, spiking experiments were dope using macluraxanthone for the root bark of Madura pomifera extraction and chlorogenic acid, rutin and psoralen for the Ficus carica leaves extraction. Recoveries above 85% were obtained for all compounds tested. The SSDM method was also applied in the extraction of the Ficus elastica leaves where, as far as we know, it has never been identified any phenolic compound. The antioxidant activity of the three plant extracts, Maclura pomifera root bark, Ficus carica and. Ficus elastica leaves, was, evaluated using DPPH and ABTS methods. The three extracts showed antioxidant activities, proving their capacity to scavenge both the DPPH and ABTS radicals. The M. pomifiera extract proved to be the most powerful antioxidant scavenger while the F. carica extract showed slightly inferior antioxidant potential. The F. elastica extract presented very poor antioxidant activity with a value significantly lower than those obtained for the other evaluated extracts. Since several phenolic compounds have already been identified from these plant extracts, their concentration on the extract was determined, and the compound individual contribution for the total antioxidant activity was determined as well. Macluraxanthone accounted for more than 20 % of the total antioxidant activity of the root bark extract and the chlorogenic acid, rutin, and psoralen accounted for about 30% of the total activity of the Ficus carica leaves extract. Most of the remaining antioxidant activity of the root bark is likely due the activity of the other xanthones already identified in the Maclura pomifera extract, as very little activity is expected for the also known flavanones. The majority of the Ficus carica leaves antioxidant activity is unaccounted for and it must be due to the unknown compounds present in the extract.

The objective of this study was to characterize Maclura tinctoria individuals in anthropic secondary formations, as their growth aspects in diameter and basal area, their morphometric relationships, spatial distribution and crown regularity. Also analyzing the behavior of individuals with more than one trunk, called multitrunks, and establish regression models to estimate basal area increment, crown diameter and height. For this, they were measured in two distinct areas in Porto Mauá, RS, diameter at breast height, total height, crown diameter and punctual density of each individual (using Bitterlich method). The morphometric indices were calculated based on data obtained from the field. The regularity of the crown was obtained by the coefficient of variation of the crown rays. For spatial analysis was calculated K Ripley function. The annual periodic increment in diameter (IPAd) and basal area (IPAg) were obtained by analysis of growth rings. In order to verify if these variables influence increments, Pearson correlation analysis was performed. It was developed mathematical equations, the stepwise method to estimate the increment in basal area, crown diameter and total height. The tendency of multitrunks individuals in relation to the single-stem for all variables was verified using analysis of covariance. The multitrunks trees in general show a similar tendency to the trees with a single stem. The studied species have moderate growth to fast, presenting therefore great potential for management. The spatial distribution indicated groupings to a higher range to 15 meters. Morphometric indices varied widely, demonstrating the plasticity of the species. As the variation in crown rays was extensive, featuring its irregularity. The morphometric variables, did not exert significant influence on the increment in diameter of Maclura tinctoria individuals. This indicates that the growth in diameter is statistically the same regardless of tree form. However, the degree of slenderness and comprehensiveness index correlated with the periodic increment in basal area. It was possible to define equations with good settings for height, crown diameter and increment in basal area of the species. All the elaborate equations had dbh as dependent variable, indicating the strong correlation of this variable with the other characteristics of trees Maclura tinctoria.

Abstract:

Pomiferin is a unique, prenylated isoflavonoid that can be isolated and purified from the fruits of Maclura pomifera (Osage Orange). The molecule typically is isolated with a small amount of a molecule called Osajin which is structurally similar to Pomiferin but lacks an aromatic hydroxyl group. As a consequence, Osajin has been shown to be a less effective antioxidant than Pomiferin. In vitro studies on Normal Human Dermal Fibroblasts demonstrate that Pomiferin is a potent extracellular matrix protein stimulant, showing increases in collagen, elastin and fibrillin expression comparable or superior to equivalent concentrations of retinol. Ex vivo hair follicle assays demonstrate comparable effects on expression of collagen and elastin at Pomiferin concentrations in the range of 0.05–5 ppm. Taken together, the results from the two assays conducted on different models indicate that Pomiferin may be a very interesting ingredient for topical skin and scalp treatments where modulation of the expression of extracellular matrix proteins is important.
