Ao considerar o contexto dos estudos sobre Turismo de Base Comunitária (TBC) e a evidência da governança como um dos fatores limitantes à consolidação de experiências turísticas de base comunitária, esta tese defende que a governança condiciona a reprodução dos princípios de participação e protagonismo comunitário, centrais às discussões do TBC. Dessa maneira, o objetivo geral desta tese é propor um modelo de análise da governança do TBC (MAG do TBC). Considerando uma perspectiva metodológica qualitativa baseada na teoria adaptativa de Layder (1993; 1998), o caminho metodológico compreendeu quatro fases distintas, nas quais fazem parte: uma pesquisa bibliográfica, duas consultas a especialistas por meio de grupo de discussões e verificação empírica do modelo em uma experiência de turismo consolidada, a Associação Acolhida na Colônia, sediada no município de Santa Rosa de Lima -SC. Ademais, a verificação empírica do MAG do TBC no campo possibilitou a emergência de novos critérios que foram incorporados à versão final do modelo. Os dados oriundos destas fases do estudo foram examinados e discutidos a partir da análise do conteúdo categorial. Desse modo, propõese que a análise da governança em experiências turística de TBC consolidadas seja analisada com base em três dimensões da governança, a saber: participação, transparência e eficácia. A operacionalização da análise destas dimensões se dá por meio da sistematização de elementos constituintes e seus critérios de análise, inerentes a cada dimensão supracitada.
Considering the context of the studies about Communitarian Based Tourism (CBT) and the evidence of governance as one of the limiting factors to the consolidation of the CBT experiences, this thesis defends that the governance influences the reproducing of the principles of participation and communitarian protagonism, central to CBT discussions. This way, the general aim of this research is propose a model of governance analysis of the CBT (MAG of TCB). Considering a qualitative methodology perspective based on adaptive theory of Layder (2993; 1998), the methodological way comprehended four different phases, which are composed by: bibliographical research, two queries to specialists through discussing groups and empiric verification of the model in an experience of consolidated tourism, the Association Acolhida na Colônia, placed in the city Santa Rosa de Lima-SC. Moreover, the empiric verification of the MAG of TBC in the field enabled the emergency of new criteria that were incorporated to final version of the model. The data from these phases of the study were examined and discussed from the analysis of categorical subject. This way, it is proposed that the analysis of governance in consolidated TBC touristic experience is analyzed based in three governance dimensions: participation, transparency and efficiency. The operationalization of these dimensions analysis is given through systematization of constituting elements and its criteria of analysis, inherent to each dimension mentioned above.

Abstract:
A leptospirose é causada por bactérias que pertencem ao gênero Leptospira. Em um estudo realizado por nosso grupo, observou-se que as proteases secretadas por leptospiras patogênicas foram capazes de clivar a molécula C3 do Complemento e seus fragmentos C3b e iC3b, além de Fator B, C4b e C2. Neste trabalho expandimos a análise da atividade proteolítica sobre os componentes do Complexo de Ataque à Membrana (MAC): C6, C7, C8 e C9. Nós observamos que essas proteases clivam todos os componentes do MAC inclusive o complexo solúvel formado e que essas clivagens ocorrem de modo tempo-dependente. Além disso, as clivagens dessas moléculas ocorrem de modo seletivo, pois mesmo utilizando quantidades reduzidas de sobrenadantes ainda foi possível observar produtos de clivagem. A atividade proteolítica foi inibida pela 1,10fenantrolina, indicando a participação de metaloproteases. O reconhecimento de quais moléculas do MAC são clivadas por proteases de leptospiras patogênicas pode contribuir para o desenvolvimento de estratégias terapêuticas na infecção por estes patógenos.
Leptospirosis is a zoonosis caused by spirochetes from the genus Leptospira. In a previous study, our group observed that the proteases secreted by Pathogenic Leptospira were capable of cleaving C3 of Complement, as well as the fragments C3b and iC3b, Factor B (Alternative Pathway), C4 and C2 (Classical and Lectin Pathways). In this work, we analyze the activity of the leptospiral proteases on the components of Membrane Attack Complex (MAC). We observed that the protease cleaves all MAC components including soluble complex formed and that these cleavages occur in a time-dependent manner and in a selective way, since even when reduced quantities of supernatants were used, the cleavage products were still observed. The proteolytic activity was inhibited by 1,10phenanthroline, indicating the participation of metalloproteinases. The recognition that MAC molecules are cleaved by proteases of pathogenic leptospires can contribute to the development of therapeutic strategies for the infection by these pathogens.

Abstract:

The most widely used protocol for providing reliable service and congestion control in the Internet is the Transmission Control Protocol (TCP). When the Internet is moving towards more use in mobile applications it is getting more important to know how TCP works for this purpose.

One of the technologies used for mobile Internet is the Enhanced General Packet Radio Service (EGPRS) extension to the popular GSM system. This thesis presents a low-level analysis of TCP performance in an EGPRS system and an overview of existing TCP, GSM and EGPRS technologies.

The bottleneck in an EGPRS system is the wireless link – the connection between the mobile phone and the GSM base station. The data transfer over the wireless link is mainly managed by the complex RLC/MAC protocol.

In this thesis, simulations were made to identify some problems with running TCP and RLC/MAC together. The simulations were made using existing EGPRS testing software together with a new TCP module. The simulation software is also briefly described in the thesis.

Additionaly, some suggestions are given in order to enhance performance, both by changing the EGPRS system and by modifying the TCP algorithms and parameters.

Les systèmes de 3G offrent de nouveaux services support (bearer services) à plus hauts débits pour les modes de transmission "paquet". Ces services vont coexister avec la voix (ou d'autres services temps réels), des scénarios de trafic mixte, voix et données, doivent être envisagés. La norme UMTS permet effectivement aux utilisateurs d'avoir plus d'un service activé simultanément. Les différentes classes de trafic augmentent la complexité de la gestion des ressources radios. Dans ce contexte, deux types de fonctions sont étudiés: l'allocation de TFCI et l'ordonnancement de paquets. Leur impact sur la qualité de service (QoS) ainsi que sur la capacité du système est évalué. On propose des améliorations de ces mécanismes dans le but d'augmenter la capacité du système et par conséquent d'améliorer la QoS des utilisateurs. Les études se restreignent au sens montant, c'est à dire aux transmissions du mobile (User Equipment ou UE) vers le réseau. Un premier mécanisme pour lequel un effort d'amélioration est fait, est l'adaptation du lien radio par variation du débit instantané transmis. On simule le cas d'une transmission multiservice (voix et données). L'UE doit partager un débit global qui lui est alloué entre les différents services activés. Ces derniers sont véhiculés dans des radio bearers (tuyaux supports). À chaque intervalle élémentaire de transmission (Transmission Time Interval, TTI), l'UE sélectionne un sous-débit pour chaque bearer; ceci se fait par la sélection d'un "format de transport" à appliquer pendant la durée TTI. Cette procédure s'effectue dans la couche MAC (Medium Access Control); le résultat de la sélection est une combinaison de formats de transport (Transport Format Combination, TFC) que la couche physique doit utiliser. La procédure, nommée sélection de TFC, permet d'adapter la transmission des différents services aux conditions variables de la propagation radio: elle détermine notablement la performance de transmission. L'algorithme de sélection de TFC est tracé dans ses grandes lignes dans la norme. Un de ses principes est de favoriser le trafic temps réel au détriment des services de données par paquet. Cependant, le trafic temps réel peut être perturbé par le trafic de données sous certaines conditions, en particulier pour les mobiles éloignés de la station de base (Node B). On propose un algorithme de sélection de TFC qui limite ces perturbations et qui offre une plus large zone de couverture aux services temps réels. En plus, il améliore la QoS du service de données et le débit effectif de l'UE sans augmenter sa puissance de transmission. Un autre type d'études concerne l'ordonnancement de paquets entre les différents utilisateurs ou UEs. C'est une procédure qui est contrôlée par la partie fixe du réseau. Nous l'étudions principalement par simulation en considérant divers mécanismes ou variations. Un premier mécanisme est nommé fast Variable Spreading Factor (fast VSF): les UEs distants changent rapidement leur facteur d'étalement (SF) afin de conserver une puissance de transmission constante, ce qui vise à stabiliser l'interférence inter-cellulaire. Un deuxième mécanisme étudié est un accès paquet décentralisé (decentralized mode) utilisant une information en retour sur le niveau global d'interférence dans la cellule. Un troisième mécanisme nommé "fast scheduling" (ordonnancement rapide) raccourcit le cycle d'ordonnancement. Les résultats ont montré que dans le cas de faible ou moyenne charge dans la cellule, le mode décentralisé réduit le délai par paquet jusqu'à 25 %. L'ordonnancement rapide augmente la capacité du système jusqu' à 10%. En plus, il améliore la QoS perçue par les utilisateurs en terme de débit par utilisateur et délai par paquet transmis.

La nécessité d’évaluer l’impact des xénobiotiques du milieu aquatique sur les organismes, justifie la mise en place de nouveaux bioessais et de biomarqueurs afin d’identifier les risques sanitaires et mieux les prévenir. L’utilisation de méthodes alternatives à l’expérimentation animale, telles que les cultures de cellules, est préconisée en toxicologie. C’est dans ce contexte que des cultures primaires de cellules cardiaques de palourde Ruditapes decussatus ont été mises en place afin d’évaluer les effets de trois types de polluants du milieu marin un biocide, une phycotoxine et un herbicide. Les primocultures sont obtenues en routine et restent fonctionnelles in vitro pendant au moins un mois. Parmi les cellules, les cardiomyocytes, dont un certain nombre de caractéristiques électrophysiologiques ont pu être précisées par patch clamp, présentent un intérêt comme bioessai pour des applications en toxicologie. Les réponses cellulaires évaluées in vitro varient selon le polluant testé et de manière dose-dépendante. En particulier, l’étude des voies de signalisation MAP kinases (P38, JNK et ERK) se révèle intéressante
The necessity to evaluate impact of xenobiotics from aquatic environment in marine organisms justifies the implementation of new bioassays and biomarkers to identify health risks for better prevention. The use of alternative methods to animal experiment, such as cell cultures, is recommended in toxicology. In this context, primary cultures of heart cells from clam Ruditapes decussatus was established and used to evaluate the effects of three types of pollutants from marine environment: a biocide, a phycotoxine and an herbicide. Primary cultures were routinely obtained and maintained functional in vitro for at least one month. Among cells, cardiomyocytes identified by electrophysiological properties after study by patch clamp presents an interest as bioassay for applications in toxicology. Cellular responses evaluated in vitro varie with the pollutant and are time and dose dependent. In particular, study of MAPK signaling pathways (P38, JNK and ERK) are of major interest

In wireless mesh networks (WMNs), the dedicated control channel (DCC) approach can support broadcasting and avoid multi-channel hidden terminal problem and deafness problem in the multi-channel environment. However, there are two serious problems in the DCC approach: the control channel bottleneck and dynamic channel selection problem. In this thesis, a protocol, named the time-based Multi-channel MAC (TBM), is proposed to resolve these two challenges. The TBM protocol consists of three components. The first one aims at reducing the control overhead by properly predicting the control initiation time. The second one can dynamically aggregate multiple packets to transmit with a single control process. The final component selects the channel that has the least influence to nearby nodes to improve channel reusability. Simulation results show that TBM can achieve significant improvement in the throughput in comparison with the existing work.
