Abstract:
mAKAP (muscle-selective A-kinase-anchoring protein) co-ordinates a cAMP-sensitive negative-feedback loop comprising PKA (cAMP-dependent protein kinase) and the cAMP-selective PDE4D3 (phosphodiesterase 4D3). In vitro and cellular experiments demonstrate that PKA-phosphorylation of PDE4D3 on Ser-13 increases the affinity of PDE4D3 for mAKAP. Our data suggest that activation of mAKAP-anchored PKA enhances the recruitment of PDE4D3, allowing for quicker signal termination.

Abstract:
The compartmentalization of second messenger-activated protein kinases contributes to the fidelity of hormone-mediated signal transduction events. For example, the cAMP-dependent protein kinase is tethered at specific intracellular locations through association with A-kinase anchoring proteins (AKAPs). We now report the cloning of mAKAP, an anchoring protein found predominantly in heart, skeletal muscle and brain, and whose expression is induced in neonatal ventriculocytes by treatment with hypertrophic stimuli. mAKAP is targeted to the nuclear membrane of differentiated myocytes. Analysis of mAKAP-green fluorescent protein (GFP) fusion constructs revealed that nuclear membrane targeting is conferred by two regions of the protein, between residues 772–915 and 915–1065, which contain spectrin-like repeat sequences. Heterologous expression of the mAKAP targeting sequences displaced the endogenous anchoring protein from the nuclear membrane, demonstrating that mAKAP targeting is saturable. Collectively, these data suggest that a domain containing spectrin-like repeats mediates targeting of the anchoring protein mAKAP and the cAMP-dependent protein kinase holoenzyme to the nuclear membrane in response to differentiation signals.

Abstract:
The physical association of regulatory enzymes and ion channels at relevant intracellular sites contributes to the diversity and specificity of second messenger-mediated signal transduction in cells. mAKAP is a scaffolding protein that targets the cAMP-dependent protein kinase A and phosphodiesterase type 4D3 to the nuclear envelope of differentiated cardiac myocytes. Here we present data that the mAKAP signaling complex also includes nuclear envelope-resident ryanodine receptors and protein phosphatase 2A. The ryanodine receptor is the major cardiac ion channel responsible for calcium-induced calcium release from intracellular calcium ion stores. As demonstrated by a combination of immunohistochemistry and tissue fractionation, mAKAP is targeted specifically to the nuclear envelope, whereas the ryanodine receptor is present at both the sarcoplasmic reticulum and nuclear envelope intracellular membrane compartments. At the nuclear envelope, a subset of cardiac ryanodine receptor is bound to mAKAP and via the association with mAKAP may be regulated by protein kinase A-mediated phosphorylation. By binding protein kinase A and ryanodine receptor, mAKAP may serve as the scaffold for a cAMP- and calcium ion-sensitive signaling complex.

Abstract:
One of the crucial cardiac signaling pathways is cAMP-mediated PKA signal transduction, which is regulated by a family of scaffolding proteins, i.e., A-kinase anchoring proteins (AKAPs). Muscle-specific AKAP (mAKAP) partly regulates cardiac cAMP/PKA signaling by binding to PKA and phosphodiesterase 4D3 (PDE4D3), among other proteins, and plays a central role in modulating cardiac remodeling. Moreover, genetics plays an incomparable role in modifying the risk of cardiovascular diseases (CVDs). Single-nucleotide polymorphisms (SNPs) in various proteins have especially been shown to predispose individuals to CVDs. Hence, we hypothesized that human mAKAP polymorphisms found in humans with CVDs alter the cAMP/PKA pathway, influencing the susceptibility of individuals to CVDs. Our computational analyses revealed two mAKAP SNPs found in cardiac disease-related patients with the highest predicted deleterious effects, Ser 1653 Arg (S1653R) and Glu 2124 Gly (E2124G). Coimmunoprecipitation data in human embryonic kidney-293T cells showed that the S1653R SNP, present in the PDE4D3-binding domain of mAKAP, changed the binding of PDE4D3 to mAKAP and that the E2124G SNP, flanking the 3′-PKA binding domain, changed the binding of PKA before and after stimulation with isoproterenol. These SNPs significantly altered intracellular cAMP levels, global PKA activity, and cytosolic PDE activity compared with the wild type before and after isoproterenol stimulation. PKA-mediated phosphorylation of pathological markers was found to be upregulated after cell stimulation in both mutants. In conclusion, human mAKAP polymorphisms may influence the propensity of developing CVDs by affecting cAMP/PKA signaling, supporting the clinical significance of PKA-mAKAP-PDE4D3 interactions. NEW & NOTEWORTHY We found that single-nucleotide polymorphisms in muscle-specific A-kinase anchoring protein found in human patients with cardiovascular diseases significantly affect the cAMP/PKA signaling pathway. Our results showed, for the first time, that human muscle-specific A-kinase anchoring protein polymorphisms might alter the susceptibility of individuals to develop cardiovascular diseases with known underlying molecular mechanisms.

Foram instalados dois ensaios de amendoim (Arachis hipogaea. L.), um no município de Ouro Preto D'Oeste e outro em Vilhena, testando o comportamento de cinco cultivares de tipos diferentes (Tatu, Altika, Makap, Penapolis e VC-42) de procedências diferentes. 0 delineamento utilizado foi o de blocos casualizados com 4 repetições (Ouro Preto D'0es-te) e 5 repetições (Vilhena) . Os resultados mostraram que há viabilidade de cultivo de amendoim do tipo Spanish e Virginia em dois municípios testados, e a sua produtividade foi maior no ensaio de Ouro Prelo D'Oeste (2.235 a 2.677 kg/ha de grãos) em relação a de Vilhena (1.060 a 2.220 kg/ha de grãos), provável conseqüência da melhor fertilidade do solo, devido ao cultivo realizado em capoeira de 3 anos recém-desmaiada.

The Schizosaccharomyces pombe stress-activated Sty1p/Spc1p mitogen-activated protein (MAP) kinase regulates gene expression through the Atf1p and Pap1p transcription factors, homologs of human ATF2 and c-Jun, respectively. Mcs4p, a response regulator protein, acts upstream of Sty1p by binding the Wak1p/Wis4p MAP kinase kinase kinase. We show that phosphorylation of Mcs4p on a conserved aspartic acid residue is required for activation of Sty1p only in response to peroxide stress. Mcs4p acts in a conserved phospho-relay system initiated by two PAS/PAC domain-containing histidine kinases, Mak2p and Mak3p. In the absence of Mak2p or Mak3p, Sty1p fails to phosphorylate the Atf1p transcription factor or induce Atf1p-dependent gene expression. As a consequence, cells lacking Mak2p and Mak3p are sensitive to peroxide attack in the absence of Prr1p, a distinct response regulator protein that functions in association with Pap1p. The Mak1p histidine kinase, which also contains PAS/PAC repeats, does not regulate Sty1p or Atf1p but is partially required for Pap1p- and Prr1p-dependent transcription. We conclude that the transcriptional response to free radical attack is initiated by at least two distinct phospho-relay pathways in fission yeast.

Abstract:
In cardiac myocytes, the slow component of the delayed rectifier K+ current ( I Ks) is regulated by cAMP. Elevated cAMP increases I Ks amplitude, slows its deactivation kinetics, and shifts its activation curve. At the molecular level, I Ks channels are composed of KvLQT1/IsK complexes. In a variety of mammalian heterologous expression systems maintained at physiological temperature, we explored cAMP regulation of recombinant KvLQT1/IsK complexes. In these systems, KvLQT1/IsK complexes were totally insensitive to cAMP regulation. cAMP regulation was not restored by coexpression with the dominant negative isoform of KvLQT1 or with the cystic fibrosis transmembrane regulator. In contrast, coexpression of the neuronal A kinase anchoring protein (AKAP)79, a fragment of a cardiac AKAP (mAKAP), or cardiac AKAP15/18 restored cAMP regulation of KvLQT1/IsK complexes inasmuch as cAMP stimulation increased the I Ks amplitude, increased its deactivation time constant, and negatively shifted its activation curve. However, in cells expressing an AKAP, the effects of cAMP stimulation on the I Ks amplitude remained modest compared with those previously reported in cardiac myocytes. The effects of cAMP stimulation were fully prevented by including the Ht31 peptide (a global disruptor of protein kinase A anchoring) in the intracellular medium. We concluded that cAMP regulation of I Ks requires protein kinase A anchoring by AKAPs, which therefore participate with the channel protein complex underlying I Ks.

As the center of Government, in Blok Barunay there are historicalarchaeological remains, such as the tomb of Raden Adipati Aryadikusumah, Raden Arya Panji Subrata, and Pangeran Arya Panjikusumah. Around this location there are suspected archaeological remains, such as Kyai Malanggedang's tomb and other tombs. The existence of these tombs should be examined about its shape, age, and location. Thus the study aims to answer these three things. The method for obtaining an appointment is to conduct a library study, observe directly in the field, and conduct interviews with selected speakers. Based on the study, there is an overview of old tombs with markers of upright rocks at the top or top of the hill. The tombs are derived from the same period, namely Islamic period, at the end of the Nagaratengah kingdom.

This work is based on field studies made in the year 2007 – a study of writings held by libraries and various govt. and semi-governmental bodies. Research reveals that many prominent people were buried in the Garut area, among them: Syech Sunan Rohmat (in the tomb of Godog), Raden Wangsa Muhamad (in the tomb of Cinunuk) and Syekh Jafar Sidiq, entombed at Cibiuk, one of the most charismatic religious leaders of his day. This third figure (Jafar Sidiq), and indeed his tomb, greatly helped the spread of Islam and even created Garut as a suitable place of pilgrimage, drawing even pilgrims from beyond Indonesia. The motivation for declaring these three tombs a suitable place for pilgrimage is based on the perception that these tombs are worthy places for meditation and for pondering a better life to come. It may be concluded that a number of religious observations, whether carried out individually or else as part of a group, are of great religious import. These pilgrimages continue to this day and may be reckoned as worthy of association with the greater pilgrimages to the Holy Land. Keywords: religion, pilgrimage, ancient tombs, Garut district. Tulisan ini diawali penelitian lapangan yang dilaksanakan pada tahun 2007, dengan metode survei dilengkapi studi kepustakaan, dan wawancara. Melalui tulisan ini berhasil diungkap bahwa tokoh-tokoh yang dimakam¬kan pada ketiga makam kuna di kawasan Garut, yaitu Syech Sunan Rohmat pada makam Godog, Raden Wangsa Muhamad pada makam Cinunuk, dan Syekh Jafar Sidiq pada makam Cibiuk merupakan tokoh yang kharismatik dan religius yang berpengaruh dalam kehidupan masyarakat setempat. Ketiga tokoh penyebar Islam tersebut makamnya dikeramatkan dan sakral serta ramai dikunjungi para peziarah yang datang baik dari dalam maupun luar kawasan Garut dan bahkan dari luar negeri. Motivasi para peziarah berkunjung pada tiga makam tersebut dilandasi persepi bahwa makam merupakan tempat untuk melakukan tafakur atau tempat yang tepat bagi peziarah yang mengutamakan kehidupan spiritual dengan harapan salah satunya hidup akan lebih baik. Dapat disimpulkan bahwa sejumlah upacara yang dilakukan sendiri-sendiri maupun bersama-sama secara serentak dengan penekanan pada upacara (ritus) berdoa, bersaji, atau upacara berupa pesta tahunan, selamatandan sebagainya hal ini menggambarkan unsur religi. Kegiatan religi tersebut masih terus berlangsung dan melekat pada kegiatan ziarah. Kata Kunci: religi, ziarah, makam kuna, kawasan Garut.

In the Tasikmalaya area many sites are found in the form of tombs and ancient grave complexes from the Islamic era. In archaeological studies, the tomb can be interpreted as a place where the body is buried and also the former location of the event by an important figure. The tomb can be interpreted as a grave and also petilasan. Through the tomb data can be revealed various aspects such as background concepts and figures buried. This study of ancient tombs in Tasikmalaya aims to reveal the background of the concept of site selection and the background of the buried figure. The research method is qualitative research through descriptive type. Disclosure of research problems is obtained through literature study and trace data reinforcement in the form of manuscripts. Ancient tombs in Tasikmalaya are on the plain and on top of a hill. The tomb at the top of the hill shows the continuity of previous cultural concepts, namely that high ground in the form of a hill or mountain is a sacred place. Some figures who are buried have a big role in the welfare of the community in the past, namely the opening of the Cibeureum situ.

<p>The meaning of eating is often reduced to concern objects or properties. Food is quite often understood as merely nutritive materials to fulfill the biological needs in order to survive or to satisfy human psychological desire. From this perspective, eating is tragically seen as killing for living. Eating thus becomes a consumptive and exploitative action. Nevertheless, eating can be a more productive act if food is not only construed as object or property, but as human strategy or means to construct relationships in social life. Inspired by Foucault, this paper shows that the meaning of food ought to be extended from the nutritive intrinsic aspects toward the political or cultural aspects; that is, food as a means to construct subject. In a sense, food governs or normalizes people in their social life. Therefore, food and eating give rise to knowledge, value order, behavioral patterns, lifestyles, or beliefs: they become the creative and transformative energy of the civilization.</p>

This study aims to determine (1) typology of the tomb of Sunan Ampel and (2) tombstone ornaments on the tomb of X in the tomb complex of Sunan Ampel. This study uses qualitative research methods through techniques (1) observation, (2) interviews, (3) and literature studies. Those data obtained are analyzed using archeological theory and the approach to artistic values. The results of this study are (1) typology of the tomb of Sunan Ampel, consisting of three elements: (a) sepulchre; (b) tombstone; and (b) the grave cupola, while (2) the tombstone ornaments on the tomb of X in the Sunan Ampel tomb complex are: (a) curly motifs, (b) vine decorations, and (c) stump decorations. Overall, it can be seen that the tomb complex of Sunan Ampel has the typology of tombs and tombstone ornaments that have characteristics and meanings in it.

Rumah makan adalah istilah umum untuk menyebut sebuah usaha kuliner yang menyajikan hidangan kepada masyarakat dan menyediakan tempat untuk menikmati hidangan tersebut serta menetapkan tarif tertentu untuk makanan dan pelayanannya. Meski umumnya rumah makan menyajikan makanan di tempat, tetapi ada juga beberapa yang menyediakan layanan take-out dinig dan delivery service sebagai salah satu bentuk pelayanan kepada konsumennya. Rumah makan biasanya memiliki spealisasi dalam jenis makanan yang dihidangkannya, baik makanan modern maupun makanan tradisional. Hj.Kholifan ialah salah satu pengusaha yang memiliki peluang membuka bisnis kuliner karna minimnya rumah makan di daerah sekitar lingkungan tempat tinggalnya. Selain memiliki peluang tersebut ia juga melestarikan salah satu kebudayaan di Indonesia dengan mendirikan bisnis kuliner yaitu Rumah Makan Biyunge dengan makanan tradisional khas Jawa. Saat ini Rumah Makan Biyunge sudah memiliki sebuah logo, namun banyak khalayak masih belum mengetahui Rumah Makan Biyunge, maka Rumah Makan Biyunge perlu melakukan redesign logo karena logo yang ada belum mencerminkan rumah makan khas Jawa. Setelah mengetahui bagaimana permasalahan corporate identity dari perusahaan diatas, maka penulis mempunyai sebuah gambaran atau konsep tentang apa yang akan dikerjakan dalam tugas akhir. Penulisan Tugas Akhir ini penulis mengambil judul “Perancangan Logo dan Corporate Identity Rumah Makan Biyunge “Ciri Khas Jawa”.

Selama ini STM dalam mengelola data makam dilakukan dengan pencatatan pada buku sehingga sering terjadi sulitnya dalam pencarian data dan sering tidak tercatatnya data makam di pembukuan. Hal ini yang menjadi permasalahan yang dihadapi oleh pengelola makam yaitu STM. Salah satu STM yang berada pada BPPI adalah STM Nurul Huda dimana dalam mengelola data makam masih dengan pencatatan ke buku sehingga data makam dan layanan informasi makam yang diberikan kepada masyarakat tidak tersampaikan dan layanan menjadi lambat. Berkaitan dengan permasalahan yang dihadapi BPPI STM Nurul Huda, maka tim pelaksana kegiatan pengabdian melalui program PKM menawarkan solusi kepada BPPI STM Nurul Huda untuk membangun sistem informasi pengelolaan data makam sebagai layanan informasi makam kepada masyarakat berbasis Web dan Android. Untuk mendukung agar sistem berjalan sesuai yang diharapkan, maka Tim pelaksana memberikan pelatihan komputer dan teknologi internet berbasis mobile kepada pengurus dan petugas lapangan pemakaman yang bertujuan untuk meningkatkan keterampilan dan pengetahuan pengurus dan tenaga bagian pemakaman dalam pemanfaatan teknologi berbasis web dan android. Dengan dihasilkannya sistem informasi pengelola data makam berbasis web dan android ini berkontribusi dalam memberikan nilai tambah kepada BPPI STM Nurul Huda untuk dapat mengelola data makam secara terkomputerisasi dan memberikan layanan dan informasi data makam dengan lebih cepat dan akurat kepada masyarakat. Disamping itu masyarakat terbantu dalam mencari dan menemukan lokasi makam yang akan dikunjungi dengan memanfaatkan sistem berbasis web dan android.

The human factor or food processor has a big important influence in food preparation because the food processor can displace bacterio of E.coli to food and than the instruments as plate is the one of the important factor that can motivate spreading if deases.</em><em>T</em><em>his research was the quantitative with cross sectional design, that purpose to know relationship between personal hygiene and technique of washing plate toward contamination of bacteria on the plate that use by the food stall at Traditional Market of Angso Duo Jambi City. The technique of take sample with quota sampling as many as 30 food stalls and datum was tested by chi-sqaure test.</em><em> </em><em>The results of the analysis found no relationship between washing techniques and personal hygiene to the number of germs in the cutlery used by food vendors Angso Duo Market Jambi City with a value of 1,000 pp.</em><em></em></p>
