The objective of this research was to characterize the percutaneous absorption of the organophosphorous pesticide, malathion, across the skin of bullfrogs (Rana catesbeiana) and marine toads (Bufo marinus) using in vitro models. An established mammalian model for percutaneous absorption, the two-compartment Teflon flow-through diffusion cell assay, was adapted to anuran skin to examine species and anatomical site differences in absorption and partitioning of C<sup>14</sup>-radiolabeled malathion. To examine the interactions of disease and pollution on amphibian populations around the world, I investigated the effect of infection on contaminant susceptibility in pickerel frog, Rana palustris, larvae. I conducted standard 48-hr toxicity tests to examine effect of trematode parasite, Echinostoma trivolvis, infection (0, 10, or 30 cercaria) on the susceptibility of pickerel frog tadpoles to the widely used organophosphate insecticide malathion. C57BL/6 mouse thymocytes were exposed to lindane, malathion, and permethrin, either separately or in mixtures of two pesticides, in concentrations ranging from 37.5 uM to 1mM. Tobacco SABP2 has been shown to display high affinity for salicylic acid (SA) and methylsalicylate (MeSA) and plays an important role in SAR signal development. Using biochemical approach, SABP2 has been shown to demonstrate strong esterase activity in converting MeSA to SA. Recent study shows that tetra fluoroacetophenone, a synthetic analog of SA, competitively inhibits SABP2 esterase activity as well as suppresses SAR signal development in tobacco mosaic virus (TMV)-infected tobacco plants. The three insecticides of interest were lindane (an organochlorine), malathion (an organophosphate) and piperonyl butoxide (PBO; a synergist). Based on minimum cytotoxicity (> LC25), the following concentrations were chosen for the pesticide mixture studies: 70Î¼M lindane (Lind), 50Î¼M malathion (Mal) and 55Î¼M PBO. In the AlamarBlue cytotoxicity assay, individual pesticide and mixtures of malathion/PBO (MP) and malathion/lindane (ML) prompted cytotoxicity with varying intensities (Mal 18.8%, Lind 20.4%, PBO 23.5%, ML 53.6% and MP 64.9%).
