Full textWe put forward a generic and specific révision of the African Clariid genera Dinotopterus BOULENGER, 1906, endemic to Lake Tanganyika, and Bathyclarias JACKSON, 1959, endemic to Lake Malawi, declared synonymous (Greenwood, 1961). Our current results based on morphometric, meristic, osteologic and morphologic criterions, collected on 462 specimens from the NHM’s (UK), MRAC’s (Belgium), SAIAB’s (South-Africa) and CU’s (USA) collections, confirm our first work of generic level (Anseaume & Teugels, 1999), leading us to reject the synonymy between both genera and to declare the second as valide. PCA and morpho-anatomic observations lead us to recognize one valide species in the genus Dinotopterus, twelve valide and two new species in the genus Bathyclarias. Six hybridation cases are viewed. The underscore of a semi-ossified preanal piece, featured by the Clariidae and the Heteropneustidae, affect the phylogeny of the Siluriformes

Full textGallid herpesvirus2 (GaHV-2) is an oncogenic herpesvirus responsible of T-cell lymphoma in chicken. GaHV-2 infections are divided into a lytic phase, depending on the expression of immediate earky genes like ICP4 and ICP27, and a latent phase characterized by the expression of the long non-coding RNA LAT. In this study, we have shown that the LAT is expressed as several highly spliced transcripts, all placing the microRNA clyster mdv1-miR-M8-M10 in their first intron. One of those microRNAs regulated the expression of ICP4 and ICP27. Studies on the ICP27 promoter allowed us to identify several important response elements (REs), including a GC box and AP1 and CRE REs, and to show that the viral protein VP16 does not transactivate the promoter. We identified a spliced transcript driven by the gK promoter that encodes a truncated ICP27 isoforms. Both isoforms of ICP27 are colocalized with spliceosomal SR proteins in the nucleus, but show a slightly different localization

Full textHerpes simplex virus type 1 (HSV 1) is able to establish latent infection in neuronal tissues after a oral primary infection. Spontaneously or upon stimuli, HSV1 can reactive and induce ocular disease. The mechanisms involved in balance between latent infection and reactivation are not completely understood. In oro-ocular murin model of HSV1 (strain SC16) infection, the amounts of Latent-Associated Transcripts (LATs) and non-sliced ICPO transcripts, which encodes a protein involved in the reactivation process, have a similar pattern of evolution from the acute to the latent stage of infection, with an accumulation in trigeminal ganglia and a decrease in other latency sites. Such striking difference between types of neurones was partially found in the corneal scarification model of HSV1 infection. But, classical patterns of HSV1 latency, i. E. Complete extinction of early and late viral genes expression was not obtained in this model, especially with the KOS strain of HSV1. These results suggest that the biological patterns of HSV1 latency depend on the type of neurons, the viral strain and the murin model

Full textThe Marek disease virus (MDV) is an oncogenic herpesvirus responsible of T-cell lymphoma in chicken. MDV infections are divided into a lytic phase, depending on the expression of immediate early gene like ICP4, and a latent phase characterized by the expression of the long non-coding RNA LAT localized in antisense. In this study, we have shown the differential expression of the cluster of miRNA mdv1-miR-M8-M10 was directly correlated with the alternative splicing of LAT’s intron 1 and more specifically with the first viral mirtron biogenesis by the spliceosome. The location of the mirtron mdv1-miR-M6 inside of the cluster is associated with a two-step biogenesis of the miARN of the cluster. On the other hand, we have identified a dual promoter that responded to Sp1, four poly-A signals and three exons that are responsible of transcriptional regulation of ICP4 transcript. We also have predicted five potential isoproteines for ICP4 and were able to observe by immunodetection that ICP4 was mainly expressed in the cytoplasm of infected cells during the lytic phase or the reactivation one

Full textOne of the strongest selective forces affecting human populations in recent history is the malaria parasite Plasmodium falciparum, which is the cause of a variety of well-established examples of pathogen-induced adaptation in humans. A special form of malaria is pregnancy-associated malaria, which is characterised by the accumulation of infected erythrocytes in the placenta, and causes up to 200,000 maternal and infant deaths every year. The aim of this work is to characterise how this particular form of malaria has shaped human genetic variation. To that end we use methods of both evolutionary genetics and molecular epidemiology, reporting the first large-scale investigation of the genetic basis of placental infection. Our results provide new insights into genes modulating the risk of infection, as well as natural selection acting on cellular pathways involved in the pathogenesis of the disease. Finally, we also provide new data on the genetic structure of affected populations in Sub-Saharan Africa.
