The objective of this diploma thesis is to design a proposal for managing projects in small and mid-sized companies that will be part of reference Model of Business Informatics MBI. MBI is a reference model for business IT, which is being developed by Department of Information Technologies at the University of Economics in Prague. MBI contains best practices in combination with theoretical background. The proposal includes prepared document templates ready to use in project management. This thesis also compares well-known and most widely used project management standards. The theoretical part is focused on the most widely used project management standards and methodologies. First chapter compares PMBOK, ICB and PRINCE2. This chapter is focused on the most important characteristics, used approaches and differences of these standards. Next chapter is closely focused on PRINCE2 described in terms of defined processes. In the practical part of this diploma thesis is a proposal for managing projects in small and mid-sized companies. The proposal takes the form of tables that matches the MBI model structure. This proposal is available on MBI website and is ready to use in practice. The theoretical part of this thesis is mainly based on the official publications and information from official websites of companies focused on project management. Practical part of this thesis is based on theoretical and practical experience. The usability of the results is evaluated in conclusion.

The purpose of diploma thesis is not only to elaborate the summary of tools already applied in the Internet environment for use of marketing and Public Relations classical music but also the summary of new and emerging tools that could be applied for such use but have not yet been applied in classical music.
Generally, this thesis deals with the chosen theme at user level and describes current applicable means available at the Internet with regard to possible use for institutions functioning in clasical music. It summarizes obtained data on current and emerging technologies and provides a comprehensive and user-friendly survey of given issues.

The subject of this thesis is the analysis of the fields related to culture transformation, that implies the changes in its financing, ownership or legal environment, as a follow up to the change of the cultural paradigm itself and to the comparison of the selected countries, which differ both in their historical development and the present day. The reasons for choosing the topic are described in this thesis, different attitudes to the domain of culture are mentioned, the contemporary process of changing the cultural paradigm is explained, and the cultural politics and systems in the Czech Republic are compared to the situation in France, Great Britain and the USA. Possible alternatives concerning the trend in culture conception in the Czech Republic are outlined, and the methodology leading to obtaining background material for possible liable decision making about the future culture system in the Czech Republic is defined.

One of the key processes of IS/IT service provider is Service Level Management. IT service provider faces a problem how the services should be defined, how a balanced service portfolio should be set up and how the processes covering the service management should be designed to meet strategic goals of the organization. The thesis focuses on IS/IT service portfolio design and management with a particular focus on infrastructure services. General goal of the thesis is to support IS/IT service provider's activities directly linked with service level management, IT financial management and IT service portfolio design. The thesis puts together theoretical sources in the area of IT Governance, generally accepted methodologies and best practices, actual market trends and author's experience. The theoretical part maps current development in the field of IS/IT service management, summarizes relevant theoretical inputs, categorizes IS/IT services, gives an overview of the most commonly used pricing models and presents actual IS/IT market trends, such as SaaS or Utility Computing. The application part, which follows the theoretical part of the thesis, is built upon fundaments of key findings and conclusions of the theoretical part. Application section introduces a method of service level management transformation and a business model suitable for IS/IT service provider. Furthermore a process of IS/IT service portfolio design is introduced. The process is supported by an analytical tool, which enables analysis of provider's financial and infrastructure data. The output of the analysis is a set of scenarios of IS/IT service portfolio including revenue, costs and utilization of resources and technologies. Academic benefit is seen not only in the consolidation of relevant theoretical sources dealing with IT service design and management. The thesis also introduces frameworks for optimization of service level management process and a concept of a IT service provider's business model. A practical contribution of the thesis is seen in a process of IT service portfolio design supported by an analytical tool for provider's real data processing, covering demand, capacity, utilization and financials.

Thesis analyses the current state of waste management of Czech Republic and the issue of production of various kinds of waste. Real data about production and composition of waste flows of Jihlava have been used. In addition, thesis evaluates the impact of local conditions on production of isolated constituents of municipal waste within the town. Influences such as density of population, housing development or form of heating have been monitored. A method on the town level for estimation and evaluation of production of municipal waste is designed in the thesis. Tools such as descriptive statistics, correlative analysis or association rules are used.

This dissertation considers the economic management of organizations funded by Sezimovo Ústí Town Council in the period from 2009 to 2013, namely the nursery school in Lipová Street and the Centre for Culture and Sport. Basic terms required for completing the practical part are defined in the theoretical-methodological part. These terms are associated with non-profit organizations and the economic management and accountancy of budget-funded organisations. The dissertation will provide an analysis of the economic management of the chosen budget-funded organisations, investigating their dependence on council funding and suggesting ways to improve economic efficiency. Professional publications and legal directives concerning this issue have been used as a basis for the theoretical-methodological part. The practical part is supported by secondary data, mainly in the form of accounting reports gained from the chosen budget-funded organisations.

Dynamic risk management is not a much-resolved issue at municipal level, although the consequences of these risks can be very serious for the municipality. The Municipality mainly focuses on situations that are called extraordinary events and the hazards of common activities, mainly associated with the running of the village, are usually neglected. The diploma thesis deals with the analysis of the current state of this issue, which deals with approaches for dynamic risk management, municipalities, methods suitable for risk analysis and the possibility of using software tools in this area. Based on an analysis of the current situation, a general model for dynamic risk management at municipal level is created. The output of the thesis is the above-mentioned general model and its verification in a particular municipality. Additionally, the financial difficulty associated with the implementation of this model is presented.

The thesis deals with an analysis of property in the selected village. The thesis is divided into two parts -- the theoretical-methodological and the practical part. In the theoretical part, there are described particular terms concerning the budget management of municipalities, accounting reform and community property. In another, theoretical-methodological part, there is introduced an approach to the accounting of municipality property in connection with the reform of the state accounting. In the practical part, there is analyzed budget management of town, the property structure at the selected village, chosen procedure of depreciation and there is mentioned the relationship between depreciation and funding of new assets. In the thesis, there are described concrete examples of depreciation. Finally, there is evaluated the whole process and there are mentioned recommendations and objections.

The first part specifies crucial terms and principles related to studied points at issue. In the second part is analysed financial management for the past five years. In the last part are suggested solutions for separated activities in Žďár nad Sázavou according to questionnaire survey. Conclusion consists of synthesis and final summaries of detected information and deficits in financial management in Žďár nad Sázavou.

This work is focused on building a content management system for municipal websites. It compares some proprietary and open source systems. Moreover, it aims to create a new content management system for little-municipal websites. The system is created with a use of PHP script language and MySQL database system. The final application runs on Apache and MySQL servers. This project allows the municipal units to create their own websites easily and quickly. They can define both the structure and the content of the pages. The project is designed to be multi-user. It puts great emphasis on the easiness of use and the blind-friendliness.

This thesis is focusing on market value and cost price of a real estate. The difference between the above-mentioned market value and cost price in relation to a specific real estate is emphasized. To regard and evaluate this difference a database of real estates was needed to be established. Building plots in the region of Opava with a planned construction of a detached house were chosen to establish this database. In all cases, the cost price of the structure and the future market value of the building plot with a completed building of the detached house was known. Based on the database, the main impact of each characteristic of the structure and the building plot on the difference in market value and cost price of particular real estates is evaluated in the main part of this thesis. In it’s last part this thesis comes to the application of ascertained conclusions on the problematic of municipalities management with municipal real estates in the most economical effective way.

The target of the study was an analysis of the current legal environment of the state contributory culture organizations in the Czech Republic and the options for their transformation into non-profit non-governmental organizations. This was demonstrated on the case of the Czech Philharmonic Orchestra, a contributory organization. Apart form the analysis of the documents, mainly analysis of semi-structured expert interviews, inner observations and informal expert interviews were applied in the qualitative research. These methods were the tool for a conclusion whether the current legal form of the Czech Philharmonic Orchestra is appropriate or whether there is a more suitable alternative in the current Czech legal environment. The result of the research is its practical application demonstrated on a model transformation of the Czech Philharmonic Orchestra into a non- profit organization (a specific type of a Czech non-profit non-governmental organization serving the public interest and/or common good), according to the Act on non-profit non- governmental organizations in its amended version.

The diploma thesis evaluates the current status of former common pastures of a study area in southwestern Bohemia. The thesis aims to demonstrate the high biological value of former common pastures and determine predictors of their preservation. For this purpose, a vector layer of former common pastures containing 668 segments was created. This layer was compared with other map layers (aerial images, consolidated layer of ecosystems, Natura 2000 mapping, species database) and various field observations. A database describing a current status of former common pastures was created and 49 preserved pastures were selected, according to predetermined criteria. Comparison of data from the common pastures and the sorrounding landscape confirmed higher biological value of the pastures. They have significantly higher habitat diversity, described by frequency and composition of Natura 2000 habitats. Several ecological and descriptive parameters were tested to find predictors of pastures preservation. The results showed that signifiant predictors of the pastures preservation are ecological parameters (i.e. number of Natura 2000 habitats, number of protected species recorded in database) and also descriptive factors (i.e. size of the pastures, shape, distance from municipality). The main aim of the diploma thesis is to evaluate the fish management of a trout reserve Svitava 2 and to suggest a correction on the basis of results from the performed surveys and information from the Moravian Fisherman's associotion evidence. Field surveys were realized on 3rd and 4th September 2013. During these surveys 12 fish species out of 5 families were demonstrated by electrofishing. Caught fis were released back to the river measuring weight and lenght. The ichthyologic research was evaulated using indi-cators for fish populations. On account of acquired knowledge and data, I recommend following correction: advance quantity biennial brown trout on 200 fish per hectare, reduce rainbow trout 40 fish per hectare and advence quantity biennial qrayling on 200 fish per hectare.

The main aim of this thesis is to evaluate the biological status of species populations of agriculturally important fish species in the district Svitava 5 and the evaluation of the evidence in the district's ten-year series. Ichthyological surveys were carried out in autumn 2015, on 20th and 21st November. At various locations were determined basic physicochemical parameters of the water and found topographical data. Catches were made by using electrical units. All fish were returned gently back to the territory after finding plastic signs. In all localities have been detected 3 fish species from two families. The total abundance ranged from 899 to 3634 ks.ha-1 and the biomass from 58.7 to 250.6 kg per ha -1. Big drop in the number of caught individuals was recorded at grayling, when it reduce the number of catches from 21 ks.ha -1 to 1 ks.ha -1 in the ten-year series.

Presented master thesis deals with the issue of assessment of management in salmonid fishing ground Svitava 4. The main aim was to compare the state of fish community with the state after the last survey from 2003-2004 and to evaluate the economic evidence (level of fishermen's pressure, level of reached catch and the stocking of economically important species) for ten years in a row. The ichthyological survey was carried out in summer season in 2014 at four localities within the fishing ground Svitava 4. The topographic data of particular localities were measured by the modern devices. Also basic physicochemical parameters of water were measured. The catching was realized by electric-fishing gear. After measuring of morphometric characteristics all the fish were returned back to the fishing ground. The survey established the presence of 8 fish species in 5 families. The abundance of fish community varied on particular localities in the range from 338 to 1633 pcs.ha-1, the biomass from 53,6 to 199,2 kg.ha-1. The fish community of concerned stream has gone through significant changes since the last survey. Considerable decrease in abundance was recorded mainly in the grayling population.

Isogenic lines in fish represent a fundamental approach to control the genetic background of experimental animals. All individuals from a given isogenic line share the same genotype. So far, isogenic fish lines have been produced only by repeated uniparental inheritance - androgenesis and gynogenesis. Homozygous progeny is produced in the first generation of uniparental inheritance, and each homozygous individual produces a different isogenic line after second generation of uniparental inheritance. Despite optimized procedures for inducing uniparental inheritance, isogenic lines have been successfully produced in only a few species of fish. Doubled haploids after first uniparental inheritance have affected fitness as well as reproductive performance. Long-term maintenance is considerably problematic even when isogenic line is established already, due to low viability and poor reproductive characteristics. The situation is further complicated by the fact that isogenic lines are usually naturally monosex, thus uniparental inheritance must be re-used for further reproduction, or sex reversal needs to be applied in part of isogenic line. Several types of germ cell manipulation were performed in presented thesis. Protocols for cryopreservation of spermatogonia and oogonia have been developed and optimized to maximize post-thaw viability. The physiological activity of cryopreserved cells was confirmed by transplantation into a surrogate host. Cryopreserved and subsequently transplanted cells retained colonization activity comparable to non-frozen control germ cells. More importantly, male germ cells were able to transdifferentiate from oogonia. The success of transplantation was confirmed by detection of expression of genes associated with gametogenesis in carp by RT-PCR. In the next study, the results of cryopreservation experiments were followed, where sterile goldfish was identified as a suitable host for homozygous carp cells. Germ cells obtained from several homozygous individuals were individually transplanted into sterile goldfish. This procedure has a potential to increase the chance of producing a viable gamete for isogenic line production. Germ cells from homozygotes with affected gametogenesis can be transferred to fully viable recipients, thereby increasing the efficiency of isogenic line production overall. In addition, the use of a goldfish as a surrogate parent will ensure that part of the germline chimeras will be male and female, thus isogenic gametes of both sexes can be obtained and no further intervention for further reproduction of the isogenic line. The suitability of triploid zebrafish, which can potentially be used as recipients for cells from homozygotes to produce isogenic lines, has been confirmed for zebrafish. Spermatogonia and oogonia from diploid donors were transplanted into artificially induced triploid larvae. Donor-derived sperm was were obtained upon maturation of triploid recipients. Transplanted oogonia transdifferentiated into spermatogonia and spermatozoa with female sex chromosomes have been produced, which may be of interesting for further studies of sex determination in zebrafish. A new germline transfer technique has been developed using striped embryos. Donor cells were transplanted from the blastula stage to the swim-up larvae. With this approach, undifferentiated primordial germ cells were able to colonize the genital groove and initiate gametogenesis. After reaching sexual maturity, germline chimeras were obtained with gametes and viable progeny. Although the overall efficacy of this method was lower compared to other transplantation methods, this study may be of relevance for germline rescue in poorly viable embryos or lethal mutants.

Quality of populations of brown trout (Salmo trutta m. fario) influence of occurence of freshwater pearl mussel (Margaritifera margaritifera) in river in the Czech Republic. Freshwater pearl mussel is a critically endangered species of animals. It does not have a very specific requirements only on environment but mainly on a specific host for holding glochidia (larval stage of pearl mussel). Thus fish management and migratory permeability of flow for support of existing populations is very important. Basic and essential precondition to support populations of freshwater pearl mussels is the presence of viable populations of brown trout, which represents the only suitable host glochidia. There are five locations with the biggest occurrence of pearl mussel in the Czech Republic - river basins the Blanice, the Teplá Vltava, the Malše, the Rokytnice and the Jankovský potok. The management of these flows is under local organizations of Czech Fishing Union, the National Park and Protected Landscape Area of the Šumava, the Secondary School of Fisheries in Vodňany and Faculty of Fisheries and Protection of Waters. The present management was found in the questionnaire for representatives of farming organizations. According to the answers in the questionnaire, the change of plans of restocking was recommended in the river basins of the Teplá Vltava and the Jankovský potok. There is the cooperation with the Germany was recommended in the river basin of the Rokytnice. The restoration of fish nursery is suitable in the river basin of the Malše. There is the first-rate population of brown trout in the river basin of Blanice, thus the continue in the present fish management for next support of this population in sequence on the populations of freshwater pearl mussel was recommended

The work is divided in several parts. The introduction describes history of protection of citizens. The thesis also deals with the issue of information systems from the general point of view and also in connection with crisis management. The practical part looks into the used information systems by questionnaire survey. The survey included all municipalities and communes and all Regional offices. The output of the research is a list of used applications. There are shown following data for each application ? frequency of an application, a map of an application according to regions, scale evaluation of an application and pros and cons of an application. The evaluation of the research disproved the hypothesis that regions and municipalities and communes have a proper idea about what kind of information system they should use. The following part of the thesis introduces a software application to support crisis management. It is a functional application designed to keep a record of shelters in the town of Písek.

The thesis titled "The Municipality and Protection of the Environment from the Viewpoint of Law" is divided into six chapters. The first chapter is the introduction, which is devoted to the constitutional foundations of protection of the environment. The second chapter deals with various kinds of instruments in environmental protection. The third chapter deals with the position of a municipality in the public administration system. The fourth chapter forms the core of the work. This chapter assesses the role of municipalities in the protection of individual components of the environment and focuses on the analysing of individual component laws. The fifth and sixth chapters are devoted to the relationship of the municipalities to the territorial planning institute and an examination of the regulations issued in a municipality that functions on its own. The conclusion focuses on the summary of the relevant issues in individual areas. In terms of environmental protection, the municipality plays an essential role, particularly thanks to its position and powers that are entrusted to the municipal authorities.

The aim of the diploma thesis is to analyse and evaluate the activities of the selected local action group (LAG) within the region. This paper does not evaluate only the current effectiveness of the LAG's work, but also its impact on regional development. The theoretical part focuses on regional policy, rural development policy, municipal cooperation and explains concepts such as LEADER method or local action group. The practical part describes both the basic characteristics of the LAG's functioning and outlines realized projects, explaining the principle and possibilities of financial support. Based on the questionnaire, the participation of municipalities in the area will be analysed. The answers of representatives of municipalities will outline the effectiveness of selected LAG and outline the impact of interregional cooperation. According to the findings and results, possible improvements will be proposed. The result of this work will be the analysis of the influence of regional cooperation of the selected LAG on the development of the region.

This thesis is focused on the quality management system. The object of the thesis is to analyse the current situation of the quality management system. Based on the results, we suggest measures to increase security and data protection in the selected enterprise. The selected enterprise is GEFOS, a.s., which provides comprehensive services in geodesy, photogrammetry, geoinformation systems and land register. The thesis can be divided into the theoretical part and the practical part. The theoretical part is focused on definition of key concepts in the fields of quality management and information system. The practical part consists of four other separate parts, namely the characteristic of the selected company, an analysis of the current situation of the quality management system, and finally suggestions for improvement of security and data protection. The suggestions are focused on requirements provided by the General Data Protection Regulation (GDPR). Information for the practical part was obtained by document analysis and a semi-structured interview with the company management.
