Citadelle minérale soudano-sahélienne, les Monts Mandara sont sujets à une reprise d’érosion que l’on pourrait corréler à la récurrence d’années sèches entre 1960 et 1991. Parallèlement les sociétés montagnardes ont connu de profonds bouleversements : mutations des systèmes agraires, introduction des cultures commerciales, monétarisation de l’économie, diffusion de l’islam et du modèle culturel peul, accélération des mobilités montagnardes, descente en plaine de certains montagnards. Cette reprise d’érosion doit être envisagée de façon complexe, conformément à la conception holiste des rapports des hommes à leur environnement des paysanneries Mandara. Considérant que les dynamiques des géosystèmes Mandara sont indissociables des mutations territoriales qui animent les formations socio-spatiales Mandara, l’analyse des modèles paysagers montagnards permet de saisir conjointement toutes les forces animant ces espaces. Figures de continuité ou fragments de désordre, ces forces se combinent localement selon les milieux, les sociétés et leurs héritages. La diversité des groupes humains dans des milieux contrastés explique les nuances fortes dans le rapport à l’espace développé par chaque groupe, guidant ses affichages paysagers. C’est dans ce rapport à l’espace, indissociablement idéel et matériel, que résident les éléments de compréhension de la dynamique des milieux et des mutations territoriales localisées. Les mutations des constructions territoriales expliquent plus sûrement la reprise d’érosion que connaissent certains massifs ou piémonts, dans le cadre d’une péjoration climatique qui constitue au final un aléa habituel pour des sociétés enracinées dans leur territoire<br>Soudano-Sahelian mineral citadel, Mount Mandara are subject to a resumption of erosion that we could correlate in the recurrence of dry years between 1960 and 1991. At the same time mountain societies knew profound turnovers: transformations of the agrarian systems, introduction of commercial cultures, the monetarization of the economy, the influence of the Islam and the cultural model Fulani, acceleration of the mobilities, descent in plain of mountain dwellers. This resumption of erosion must be envisaged in a complex way, according to the holistic conception of the relationship of the men to their environment in farming societies Mandara. Considering that the dynamics of ecosystems are inseparable territorial transformations which liven up the socio-spatial formations Mandara, the analysis of the mountain landscaped models allows to seize collectively all the strengths livening up these spaces. Figures of continuity or fragments of disorder, these strengths harmonize locally according to the ecosystems, the societies and their inheritances. The variety of the human groups in contrasted ecosystems explains the strong nuances in the relationship to the space developed by every group, guiding its landscaped displays. It is in this relationship in the space, indossociably imaterial and material, that live the elements of understanding of the dynamics of the ecosystems and the localized territorial transformations. The transformations of the territorial constructions explain more certainly the resumption of erosion which know mountains and piedmont, within the framework of a climatic crisis which constitutes in the end a usual chance for societies rooted in their territory

Abstract:
O Maciço Granítico de Mandira constitui um corpo granítico peralcalino, de aproximadamente 50 \'km POT.2\' de área, nitidamente intrusivo, com forma alongada elipsoidal e eixo maior com direção N4OE, localizado no sul do Estado de São Paulo, na Região Administrativa do Vale do Ribeira. Limita-se a N e NW com as rochas gnáissico-migmatíticas do Complexo Costeiro, as quais são transicionais (a oeste) para os granitos anatéticos ou sintectônicos da Fácies Cantareira. No Extremo NE ocorrem os ectinitos do Complexo Turvo-Cajati. Petrograficamente distinguem-se três unidades do Maciço: Acaraú (\'gama\'A), que ocorre em dois \"stocks\" na porção meridional do plutão e é composta principalmente por mesopertita granito com ferro-hastingsita; Mandira (\'gama\'M), que compõe o corpo principal do Maciço e apresenta composição de mesopertita granito com riebeckita; Mandira 1 (\'gama\'M1), que ocorre em quatro \"stocks\" na peroferia do corpo maior de \'gama\'M e é composta principalmente por granito 3A. Além das três unidades graníticas, ocorrem regiões afetadas em graus variados por processos tardi a pós-magmáticos: greisenização, albitização e feldspatização, por vezes associados a intenso hidrotermalismo. As regiões afetadas pelos processos tardi a pós-magmáticos encontram-se enriquecidas em Zn, Pb e Cu, que geralmente constituem as mineralizações. Esta pesquisa apresenta a caracterização petrográfica, mineralógica e química do maciço a partir de estudos em amostras de rocha (superfície e testemunhos de sondagem). Propõe também uma hipótese para evolução petrogenética do Maciço Granítico Mandira, envolvendo a intrusão e colocação das várias unidades graníticas bem como o estudo dos processos de alteração tardi a pós-magmáticos.<br>The Mandira Massive is an alkali granitic body covering an area of approximately 50 km² in the Administrative Region of the \"vale do Ribeira\" in the southern of the São Paulo state. It shows clear intrusive realtions, despite its mapped lensoid shape whose longest axis the regional main structural direction. At N and NW the country rocks belong to the gneiss-migmatitic \"Complexo Costeiro\", transitional (to the west) to syntectonic or anatetic granites of Cantareira facies. At the NE end ectinites of the \"Complexo Turvo-Cajati\" occur. Three units are petrographically distinguished in the massive; 1) Acaraú (\'gama\'A), exposed in two stocks at the meridional part of the pluton, made up essentially of ferrohastingsite bearing mesoperthite granite; 2) Mandira (\'gama\'M) that constitutes the main volume of granite present in the Massive and is composed by riebeckite bearing mesoperthite granite: 3) Mandira 1 (yM1), occurring in four stocks scattered along the Massive border zone and showing a granite 3A composition. In the addition to the above cited units regions were detected wich were variously affected by tardi to post magmatic process; greisenization, albitization and k-feldspatization, sometimes associated to intensive hydrothermal action. Such regions prove to be enriched by Zn, Pb and Cu mineralizations. This research substantiates the present chemical, mineralogical and petrografical knowledge about the Mandira Massive and was carried out from studies on bore core and surface samples. It also presents a hypothesis aiming to explain the Massive petrogenetic evolution comprehending intrusion and emplacement of the several granitic units as well as the tardi to post magmatic alteration wich followed this process.

ABSTRACT OF THE THESIS MANDALA SPRINGS by Juleen Collins Florida International University, 2018 Miami, Florida Professor Debra Dean, Major Professor MANDALA SPRINGS is the small town setting for a story that explores the nature of secrets, lies, revelations, and the damage each can cause. The narrative follows Bodhi MacLachlan, a young woman who struggles with Borderline Personality Disorder, back to the psychiatric hospital where she has resided in-patient multiple times. The long-term association with her psychiatrist becomes complicated when she reveals details of her affair with a secretive man. Meanwhile, she becomes obsessed with uncovering the mysteries of the relationship between a new patient, Scott, and his much younger sister, Ollie, to whom he is guardian. As Bodhi proceeds, she unintentionally but irreparably damages her relationships with each of these characters. Ultimately, she must come to terms with the consequences of her actions. To reflect the complicated, upside down world of psychiatric illness, the story is written in the hybrid form of a playscript-novel.

Abstract:
<p> This depth psychological study explores the possibility that the drawing and subsequent recognition of chaotic looking irregular and misshapen shapes, lines, and marks applied in a lopsided and unbalanced manner within and protruding outside a mandala circle represent the beginning of a psychological development in the patient's psyche. Such a shift in the psyche may set the stage for using mandala psychology to facilitate the psychotherapeutic work of gathering and containing additional destructive and shadowy psychological material. </p><p> A retrospective single case study design is combined with a heuristic approach to discover the effects of producing an asymmetrical mandala, as described above, while following the chaotic mandala images through a change in the plot line of the research/participant's psychological story. Information is collected and outlined for the depth psychotherapy community that describes the process of mandala psychology from this vantage point of unbalanced and chaotic mandalas, illustrating the mandala's usefulness as a container for psychological and emotional chaos. The change in the plot line of the subject's mandala story is compared to and amplified with the structure of the plot line of the prototypical fairytale narrative at the moment when the darkly woven female character of the witch enters the narrative. This mythological component is introduced to enrich the telling of this case study. The archetypal analysis hypothesizes that at the moment the research/subject engaged her chaotic mandalas, the shift she experienced represented the collective and empowering primal energy of the dark energies of the feminine, personified for this study as the witch. Abstract:
Electroporation is a technique that employs electrical pulses to open pores in a cells membrane to allow DNA to enter and be transcribed using the cells machinery. A Bombyx mori odorant receptor (BmOr) promoter will be introduced to the antennae of Manduca sexta pupa using this technique. Vectors containing a BmOr promoter and TagRFP were constructed using pUC19 as the base plasmid by sequential ligations of first the promoter and then TagRFP using common restriction sites. After ligation the vectors were checked by restriction digest to extract and check the size of the insert and by sequencing the insert and comparing to the expected sequence. Vectors BmOR46-TagRFP-pUC19, BmOR41-TagRFP-pUC19, BmOR27-TagRFP-pUC19 were successfully created and are usable in electroporation experiments as demonstrated by successful electroporation of explanted antennal tissue. Future directions include use of GFP instead of TagRFP as the reporter gene and addition of RNAi to the vector to affect development through loss-of-function of a protein for the purpose of identifying some of the roles specific proteins play in olfactory development. Electroporation will be performed in live stage 4 pupa antenna instead of explants to allow for development of the antenna after introduction of the vector.

Abstract:
The chymotrypsin and trypsin-like activities from the Manduca sexta midgut were purified, partially characterized and the cDNAs encoding the enzymes were cloned. A putative elastase cDNA was also cloned and the sequence is reported. The purified trypsin and chymotrypsin are maximally active at alkaline pH (10.5-11.0 and 10.5 respectively). Kinetic studies of the chymotrypsin reveal that the K(m) for the synthetic substrate N-succinyl-Ala-Ala-Pro-Phe p-nitroanilide varies only slightly between pH 7.5-11.5 and the Dixon plot shows a kinetically significant pKₐ at 9.2. The specificity of the purified chymotrypsin is the hydrolysis of the peptide bond on the carboxyl side of tyrosine, phenylalanine, tryptophan, histidine, leucine, threonine and glycine. The chymotrypsin is inhibited by TPCK, PMSF, chymostatin and DFP. Three different trypsin cDNAs, two chymotrypsin cDNAs and an elastase cDNA were isolated that encode preproenzymes of 256, 292 and 291 amino acids, respectively. All the encoded activation peptides contain an arginine residue immediately preceding the amino-terminal residue of the mature enzymes which suggests tryptic activation of the zymogens. The encoded trypsins and chymotrypsin contain the highly conserved residues, Asp, His, Ser, involved in catalysis in serine proteases, along with the residues which define the specificity pockets. The encoded elastase contains the residues which define the specificity pocket; however, the "catalytic triad" consists of Asp, His and Thr. All the cDNAs for the midgut enzymes encode a large number of arginines which may stabilize the enzymes, by remaining protonated, in the alkaline midgut of M. sexta. Sequence comparisons show that all sequences are most similar to other invertebrate and vertebrate serine proteases. Northern analysis localizes the trypsin mRNA to the middle and posterior third of the midgut, the chymotrypsin mRNA to the anterior and middle third of the midgut and the elastase mRNA to the middle third of the midgut.

Com o intuito de conhecer, compreender e interpretar as transformações e mudanças que estão em processo na comunidade, decorrentes do reconhecimento oficial da Terra de Quilombo e da criação da Reserva Extrativista, ambas no ano de 2002, a partir das experiências e percepções dos moradores de Mandira. Essa pesquisa visa colaborar com o debate sobre populações quilombolas e a sua relação com o território. Além de contextualizar a formação e a transformação dos quilombos brasileiros, e contribuir para a valorização da identidade cultural quilombola dessa importante comunidade. Para a realização desta pesquisa foram necessários levantamentos bibliográficos para aprofundamento na temática, levantamento de dados secundários no IBGE, ISA, ITESP entre outros. A realização dos trabalhos de campo foi fundamental para a efetivação da pesquisa, o primeiro trabalho de campo realizado em 2014, foi possível conhecer a comunidade e ver que seria possível a realização desta pesquisa. Já o segundo trabalho de campo, realizado em 2015, foram realizadas seis entrevistas e no terceiro, e último trabalho de campo, realizado em 2017, foram realizadas mais 3 entrevistas, por meio de um roteiro semiestruturado, com lideranças e moradores do Mandira. Como resultados, a transformação que salta aos olhos, é referente as construções físicas, desde 2014, o primeiro contato com a comunidade, até 2017, várias construções e mudanças estruturais surgiram e que mudaram a configuração da comunidade. Como a construção do Restaubar Ostra Cataia, dos banheiros de uso público, a reforma e construção da extensão do galpão do centro cultural, muito utilizado em festas e eventos, a piscina, entre outras. Essas novas construções e reformas estão sendo motivadas especialmente pelo turismo de base comunitária que tem se fortalecido em toda a comunidade Mandira. Também foi possível perceber, uma transformação da comunidade no quesito ambiental, no manejo e cuidado do mangue e da extração das ostras, houve uma tomada de consciência no quesito do mangue, pois a extração de ostras estava desenfreada, afetando a capacidade de produção do manguezal. Depois da criação da Cooperostra, seguem um plano de manejo da reserva extrativista, para mitigar o impacto no mangue, que foi causado no passado e hoje já foi revertido. Outra transformação essencial para a comunidade Mandira, foi relacionada a sua identidade cultural, durante muitos anos os moradores sentiam vergonha de serem reconhecidos como mandiranos, pois sofriam racismo e preconceitos, por serem negros, da zona rural e extratores de ostras no manguezal. O quilombo de Mandira deve ser valorizado tanto por sua história, quanto por sua identidade quilombola, como também pelas práticas de conservação da natureza, da Mata Atlântica e do Manguezal.<br>The present work had as objective to carry out a study on the quilombola community Mandira, that is located in the Ribeira Valley, in the rural zone of the municipality of Cananéia, south coast of São Paulo. With the purpose of knowing, understanding and interpreting the transformations and changes that are in process in the community, due to the official recognition of Quilombo Land and the creation of the Extractive Reserve, both in the year 2002, based on the experiences and perceptions of the inhabitants of Mandira. This research aims to collaborate with the debate about quilombola populations and their relation with the territory. In addition to contextualizing the formation and transformation of Brazilian quilombos, and contributing to the appreciation of the cultural identity of this important community. For the accomplishment of this research it was necessary bibliographical surveys for deepening in the thematic, survey of secondary data in the IBGE, ISA, ITESP among others. The accomplishment of the fieldwork was fundamental for the accomplishment of the research, the first fieldwork conducted in 2014 was possible to know the community and see that it would be possible to carry out this research. The second fieldwork, conducted in 2015, was conducted six interviews and in the third, and last field work, held in 2017, three more interviews were conducted, through a semi-structured script, with leadership and residents of Mandira. As a result, the transformation that jumps to the eye, refers to the physical constructions, since 2014, the first contact with the community, until 2017, there were various constructions and structural changes that changed the configuration of the community. Like the construction of the Restaurate Ostra Cataia, of the bathrooms of public use, the reform and construction of the extension of the shed of the cultural center, much used in parties and events, the swimming pool, among others. These new constructions and reforms are being especially motivated by community-based tourism that has been strengthened throughout the Mandira community. It was also possible to perceive a transformation of the community in the environmental issue, in the management and care of the mangrove and the extraction of the oysters, there was an awareness of the mangrove as the extraction of oysters was unrestrained, affecting the mangrove production capacity. After the creation of Cooperostra, they follow a plan of management of the extractive reserve, to mitigate the impact in the mangrove, that was caused in the past and today has already been mitigated. Another essential transformation for the Mandira community was related to their cultural identity, for many years the residents were ashamed to be recognized as mandiranos, because they suffered racism and prejudice, because they were black, from the countryside and extractors of oysters in the mangrove. The Mandira quilombo should be valued both for its history, as well as for its quilombola identity, as well as for the conservation practices of nature, Atlantic Forest and Mangue.

Abstract:
Insects such as Manduca sexta must be efficient in obtaining energy stores in order to survive. The main goal of the M. sexta larva is to eat and store enough energy to reach the adult stage, produce eggs (in the case of females), and reproduce. Triacylglycerol (TAG), the most important energy storage molecule, is stored mainly in adipose tissues in lipid droplets, although other tissues are also able to store TAG in similar organelle structures but to a lesser degree. The phosphatidic acid (PA) pathway and the 2-monoacylglycerol (MAG) pathway are both energy-dependant acyl-CoA processes and are the main synthetic pathways by which TAG is synthesized in adipose and other tissues. My research led to the discovery of an energy-independent pathway for the synthesis of TAG that was present in the M. sexta midgut. Based on partial purification, a transacylase/lipase enzyme is present and responsible for DAG and TAG synthesis in the M.sexta midgut. Lipogenesis and lipolysis in adipose tissue involves a series of enzymes. Adiponutrin and desnutrin, two proteins involved in fat homeostasis in humans and mice, have received a lot of attention since their activities are dependent on the fed or unfed state of the animal. In this study, bioinformatics analyses were performed, which allowed the identification of an insect gene that has an ortholog in human and mice that plays an important role in adipose tissue TAG hydrolysis and synthesis. Only one insect gene ortholog was found to be present in the Aedes aegypti (mosquito), Drosophila melanogaster (fruit fly), Anopheles gambiae (mosquito), Bombyx mori (silk worm), and Tribolium castaneum (red flour beetle) genomes corresponding to genes involved in the regulation of TAG metabolism in mice (adiponutrin, desnutrin) and humans (iPLA-epsilon, iPLA-zeta, and iPLA-eta). Expression of the M. sexta calcium-independent phospholipase A2 (iPLA) ortholog has demonstrated that the protein is able to transfer acyl groups between MAGs in an energy-independent manner, similar to that in human iPLAs. This is the first example of a transacylase identified in insects.

Insects fed high carbohydrate diets have increased fat storage, and excess fat storage in insects may have fitness costs. M. sexta larvae reared on 5.6% fat diet had lower body mass and growth rate compared to those on 3.4% fat or 0.4% fat diets. Fifth instar caterpillars fed high-fat diet ate less food but had higher approximate digestibility than the low-fat diet fed caterpillars. Analysis of fat body and fecal pellet lipid content showed high-fat diet fed caterpillars stored and excreted more lipids than low-fat diet fed caterpillars. To test the hypothesis that increased dietary fat alters lipid transport, we measured mRNA expression of apolipoproteins I and II, proteins for transporting lipids. Expression of apolipoproteins I and II did not differ with dietary fat. Negative feedback from fat intake could inhibit feeding via endocrine pathways. This research will increase our understanding of the regulation of feeding in caterpillars.

Abstract:
Artificial reefs have been deployed throughout the world for a range of different purposes and thus differ in their characteristics. To better understand how their designs and implementation differ spatially, temporally and according to their stated purpose, an extensive collation of information on artificial reefs is needed. Furthermore, in Australia and particularly Western Australia, artificial reefs are increasingly being deployed to enhance recreational fishing experiences and catch rates of key species. To address these topics, this thesis aimed to conduct a systematic literature review on existing artificial reefs around the world and describe their characteristics. It also aimed to compare and describe the characteristics of the fish fauna on the recently deployed Mandurah artificial reef (south-western Australia) to those on nearby natural habitat. Finally, a comparison of the fish community of the three ‘Fish Box’ artificial reefs in Mandurah, Bunbury and Dunsborough was conducted. The literature review identified 1,074 unique artificial reefs from 71 countries, with 89% located in the northern hemisphere, but with an equal distribution between east and west. A more in-depth investigation of artificial reefs that were intentionally deployed found that these deployments increased markedly after 1965 and were most commonly sunk between depths of 10 and 30 m. Most reefs were designed to enhance faunal communities and/or fisheries. More affluent countries have monitored artificial reefs using more technologically advanced methods including video systems over recent decades. However, many reef deployments have not had an associated monitoring program and those that do are generally too short to detect long-term temporal changes. Fish and the associated faunal assemblages were monitored on an artificial reef and natural (control) site using Baited Remote Underwater Video (BRUVs) between February 2019 and July 2020. A far greater number of species and individuals were observed on the artificial reef than the control site. Additionally, three key recreational fishing species, i.e. Chrysophrys auratus, Seriola hippos and Pseudocaranx dentex, were also found in far greater abundances on the artificial reef. Pelagic fish were also more common on the artificial habitat, likely due to the upward flow diversion produced by the modules and the subsequent increased availability of plankton. While community composition was mainly driven by differences between the artificial reef and control site, the faunas of both sites did change temporally, albeit to a lesser extent. There was a general decrease in the number of species of fish in winter and increase in summer. An aim of this study was to develop a standardized protocol for describing artificial reefs, which is hoped to be adopted globally. If successful, this will allow comparisons between artificial reefs and their outcomes to be made thus increasing our understanding of these reefs and improving their designs. The study also demonstrates that the Mandurah artificial reef supports a distinct faunal community compared with an adjacent natural habitat with no reef, including species that are of recreational importance. A comparison of three ‘Fish Box’ artificial reefs demonstrated that the fauna on the Mandurah reef was the most speciose and harboured the greater number of fish, indicating that site selection has a major influence on the resultant fish community composition and thus should be considered in future reef deployments.

Fat body tissue was taken from female Manduca larvae throughout the fifth instar. The samples were prepared for histological study and electron microscopy. Hemolymph samples were collected and analyzed for protein profile and concentration. The data showed that the fat body underwent a series of changes with development. These include an increase in cell size, and accumulation of lipid and glycogen during the feeding phase of the instar, and the formation of protein and urate granules during the wandering stage. Also apparent was the cyclic development of a reticular system on the cell surfaces. Maximum development of the system coincided with the period of highest protein concentration in the hemolymph, while its disappearance is coincident with a drop in hemolymph protein concentration and formation of fat body granules. Thus the fat body plays a synthetic role early in the instar and becomes a storage tissue as pupation approaches.

Abstract:
The major developmental cue and the coordinating factor of metamorphic changes in insects is a group of steroid hormones, the ecdysteroids. Using subtractive hybridization I have isolated five cDNA clones that represent genes whose transcripts are up-regulated prior to pupal ecdysis in the nervous system of the tobacco hornworm, Manduca sexta, after a major ecdysteroid peak. Three of the genes, Mng10, Mng14 and tps9 are expressed primarily in the nervous system, the two other genes, esr16 and esr20 are specific to tracheal epithelial cells. In situ hybridizations showed that the transcripts of the two nervous system specific genes were localized to uniquely identifiable neurosecretory cells in the brain and ventral nervous system. The total number of cells expressing these genes in the CNS is 16-20. The third nervous system specific transcript is localized to small patches along the boundary between the central neuropil and neuronal cell bodies, in abdominal ganglia. The patches are smaller than nuclei, possibly mitochondria, and seem to be localized to large tracheal branches. Sequence analysis of the cDNA clones revealed that esr20, esr16 and Mng10 encoded a protein, with slight similarity to already identified sequences. These similarities, however, were insufficient to propose a function for the Manduca genes. Two of the sequences do not seem to encode a protein and are postulated to function as regulatory or structural RNAs. Statistical analysis of the coding and non-coding sequences showed a clear distinction between their behavior using the Zipf'test. Four of the five genes are regulated by ecdysteroids as predicted by the initial screen, transcript levels are reduced at high 20-HE concentrations. More detailed studies of the regulation of a tracheal-specific gene (esr20) showed that the accumulation of the mRNA requires first the presence then the subsequent absence of the ecdysteroids. The role of the ecdysteroids appears to be permissive and an additional factor is required to initiate the accumulation of the mRNA. The transcript of esr20 appears 16 hours before ecdysis and is removed at ecdysis by a mechanism that requires protein synthesis and possibly influences the stability of the transcript.

Abstract:
To begin to determine the role tyrosine kinases play in the development of the antennal lobe in Manduca sexta, phosphotyrosine levels were examined immunocytochemically during critical periods of development. Tyrosine phosphorylation was found to be highest in the region of the antennal lobe known to contain the growing tips of antennal-lobe neurons and forming synapses. To explore the relationship between synapses and tyrosine kinase activity a marker for synapses was developed for light level microscopy. Manduca synaptotagmin was cloned and sequenced and a polyclonal antibody (SYNT76) was produced against the amino terminal of the predicted peptide sequence. SYNT76 labeled known synaptic neuropils in the brain of Manduca and recognized a single 61 kd protein on immunoblots. Immunocytochemical double labeling of antennal lobes for synaptotagmin and phosphotyrosine showed that although the patterns were very similar there was surprisingly little direct signal overlap. The similarity of immunolabeling suggests that tyrosine kinases may affect synapses in the developing antennal lobe but the lack of dramatic signal overlap shows that this association does not occur at the active zone. A trk-like protein was identified immunocytochemically, using an antibody against mammalian neurotrophin receptors, in the same regions of the developing lobe that contained high levels of phosphotyrosine. Trk-like proteins and phosphotyrosine were also found in the growing processes of cultured antennal lobe neurons. To examine the role of trk-like kinases in process outgrowth of antennal-lobe neurons, a tyrosine kinase inhibitor specific for trk-like kinases (Tyrphostin AG879) was applied to cultured neurons and was found to inhibit process outgrowth from antennal-lobe neurons significantly when compared to cells grown under control conditions. To determine the identity of the antennal-lobe trk-like proteins outside of their kinase domains the process of cloning trk-like messenger RNA was begun with reverse transcriptase polymerase chain reactions (RT-PCR). Northern blot analysis using RT-PCR fragments with significant sequence similarity to mammalian trk receptors identified a single 4.5 kb trk-like transcript in the brain and specifically in the antennal lobe during development. These data suggest that a trk-like kinase may mediate a developmental signal that causes antennal-lobe neurons to grow processes.

Thesis (MA)--Stellenbosch University, 2012.<br>ENGLISH ABSTRACT: Nelson Mandela stands as one of the most powerful symbolic figures of the past century, embodying notions of freedom, peace, racial reconciliation and the struggle against tyranny. As largely uncontested as this image is today, its constitution has by no means been uncomplicated. Before he was incarcerated on Robben Island, Mandela was viewed as a young, militant firebrand within the ANC-led liberation movement, an image which was counterpointed by his patrician lineage, education and professional success as a lawyer. His highly visible embodiment of this complex identity served to elevate him not only to the top of the black Johannesburg social hierarchy, but to the forefront of the liberation struggle. The state-sanctioned view of him was, by contrast, as a terrorist, agitating for the destruction of the state. During his imprisonment on Robben Island, the government sought to entirely expunge his words and likeness from active circulation, which ironically facilitated the process of myth-making around him. After his release from prison, Mandela largely succeeded in claiming agency over his image – the one which still persists in the international public imagination – facilitated in large part by the publication of his autobiography, Long Walk to Freedom, and the numerous acts of reconciliation and diplomacy which he undertook. In writing this thesis, I have sought to trace the process of mythmaking around Mandela, questioning how the disparate, and often contradictory, ideas around him have been narrativised and incorporated into the mythical figure we are familiar with today, both by him and others. I have divided the narrative construction of Mandela into two broad epochs: the ―dominant‖ narrative, which developed from his entry into politics until his release from prison in 1990, and the ―official‖ narrative, which developed from his release from prison. I seek to illustrate the processes by which the dominant narrative was constituted, and how this narrative construct gained increasing ideological currency during his imprisonment on Robben Island. I then seek to illustrate how the numerous, often-conflicting elements of the dominant narrative were ultimately consolidated and largely supplanted by the official narrative, as represented by Long Walk to Freedom, focusing specifically on its theme of progress and maturation. In my conclusion, I argue that many of the ideological elements which fed the mythical construction of Mandela in the dominant narrative, as a youthful, masculinised liberation fighter, persist today. The promise which the Mandela of the official narrative embodied, of South Africa as a ‗miracle‘ nation destined to move beyond the vestiges of Apartheid – including racism, unemployment and poverty – has largely failed to materialise, allowing these elements to gain an ideological currency once more.<br>AFRIKAANSE OPSOMMING: Nelson Mandela word beskou as een van die belangrikste simboliese figure van die afgelope eeu, en hy verteenwoordig begrippe soos vryheid, vrede, rasse-versoening en die stryd teen tirannie. This is a quantitative as well as a qualitative research project, focusing on the quantity and frequency of the meanings and processes under scrutiny. The tool used for the mensuratiion of concentrated attention in four high school groups, totalling 70 teenagers, was Test AC-15. In the concentrated attention test, groups B and C (mandala + test) showed results above the average, while groups D and E (test + mandala) came up with results below the average; thus attesting to the mandala's improvement of concentrated attention. The results also show that circular forms are the most frequent in the mandala's inwards, and that, in the quadrature of the circle, flower and star shapes prevail among the themes found. In the verification and analysis of the groups, it is possible to observe that the drawings, in their majority, are center-aligned, which indicates that the subjects of this research are well-adjusted individuals, with a tendency to be more self-driven and self-centered. The characteristics of drawing styles and pressure, reflecting emotional and mental balance, appear to be normal. Also, the drawings present mostly average aesthetic results, showing signs of intelligence, aptitude for spatial abstraction, and emotional equilibrium. Therefore, it can be said that, in this study, the mandala presented an enriching activity for individuals' psychic life, enhancing concentrated attention in the subjects, and bringing moments of improvement in the inner contact with order and harmony, leading to emotional integration<br>O objetivo deste estudo consiste em verificar se o desenho da mandala melhora a atenção concentrada em adolescentes em sala de aula e se o mesmo apresenta símbolos de conotação religiosa. É uma pesquisa qualitativa e quantitativa, que privilegia os significados e processos mensurados em termos de quantidade e freqüência. O instrumento utilizado para medir atenção concentrada em quatro turmas do Ensino Médio, que totalizaram 70 adolescentes, foi o Teste AC-15. As turmas B e C (Mandala + Teste) que representa a aplicação do desenho da mandala e depois o teste AC15 apresentaram índice superior à média no teste Atenção Concentrada, enquanto as salas D e E (Teste + Mandala) manifestaram um índice inferior à média; comprovando, assim, que a mandala proporciona maior atenção concentrada. Os resultados mostram, também, que as formas circulares são as mais freqüentes no interior da mandala e que, na quadratura do círculo, entre os temas encontrados, há o predomínio de flor e estrela. Na verificação e análise das turmas, é possível observar que os desenhos, em sua maioria, estão centralizados, indicando que os sujeitos desta pesquisa estão ajustados; mostram-se mais auto-dirigidos e auto-centrados. A caracterização do traço e a pressão, que significa equilíbrio emocional e mental, se apresentam normais. Os desenhos também, em grande parte, são medianos, demonstrando inteligência, capacidade de abstração espacial e equilíbrio emocional. Portanto, podemos dizer que a mandala nesse estudo foi uma atividade enriquecedora para a vida psíquica, aumentando a atenção concentrada dos sujeitos e proporcionando momentos à melhora na integração emocional de contato interior de ordem e harmonia, o que pode levar, à integração

Abstract:
The phenomenon of multi-drug resistance in tumor cells is mediated by the overexpression of a membrane protein called P-glycoprotein. Since its initial discovery in tumor cells, P-glycoprotein-like molecules have been found to transport a wide range of substrates in organisms from microorganisms to humans. To test the hypothesis that a P-glycoprotein-like mechanism is operating in insects, three insect species were used: Manduca sexta, Periplaneta americana and Rhodnius prolixus, the nicotine-resistant M. sexta was the model toxin-resistant insect. Immunostaining with a monoclonal antibody, C219, which recognizes a highly conserved epitope in all P-glycoprotein molecules, demonstrated positive immunolabelling of the Malpighian tubules of M. sexta and R. prolixus and at the blood-brain barrier of the central nervous systems of M. sexta and P. americana. To assess the extent of P-glycoprotein involvement at the blood-brain barrier of M. sexta, I took advantage of the three different stages of the insect's life cycle, the actively feeding larval stage and the non-feeding pupal and adult stages. Using an extracellular recording technique to monitor levels of neural activity in isolated abdominal nerve cords, I demonstrated that, in contrast with larval M. sexta, the CNS of both the pupal and adult stages are both highly nicotine-sensitive. Immunostaining for P-glycoprotein in the metamorphosing CNS illustrated that the distribution of P-glycoprotein in the barrier region changes dramatically when the need for a barrier to dietary neurotoxins is lost. M. sexta Malpighian tubules provided an ideal in vitro assay system to directly test our hypothesis that a P-glycoprotein-like molecule is involved in nicotine transport. Using an isolated Malpighian tubule preparation, the tubules were assayed for their ability, first, to concentrate nicotine in the tubule lumen (confirming the presence of an active transporter in the tubules) and, second, to determine if various drugs interfere with nicotine transport. Tubules bathed in 0.5 mM nicotine were found to concentrate nicotine in the secreted fluid at least 9-fold. Nicotine transport was inhibited with verapamil, a known inhibitor of P-glycoprotein. In addition, nicotine transport was competitively inhibited by another alkaloid, atropine, suggesting that the M. sexta pump may be a non-selective alkaloid pump. To further confirm that the insect pump is a P-glycoprotein-like multi-drug pump, the tubules were tested for their ability to transport a known P-glycoprotein substrate, vinblastine. The tubules were not only able to transport radiolabelled vinblastine, but this transport was also inhibited by verapamil. P-glycoprotein is localized to the digestive tracts of mammals and of invertebrates, where it is thought to act as a defective mechanism to protect organisms from dietary toxins. Immunohistochemical staining for P-glycoprotein demonstrated that it is also expressed in the M. sexta gut. In a parallel study, a monoclonal antibody against a P450 from a DDT-resistant housefly (Waters et al., 1989) was used to localize a cytochrome P450 monooxygenase enzyme (P450 or PSMO) to the M. sexta midgut. The co-localization of P450 and P-glycoprotein to the midgut suggests that larval M. sexta use a combination of P450-mediated metabolism coupled with transport to process dietary toxins.
