ABSTRACT - The Role of the Veterinary Professional in Laboratory Biosafety and Biosecurity: Development of a Biosafety and Biosecurity Manual to the Bromatology and Biological Defence Laboratory of the Portuguese Army - Biosecurity and biosafety should be a priority of all States in the defence and protection of their citizens. Epidemic outbreaks or the threat of bioterrorism demand from Governments and international institutions appropriate strategic measures to fight such risks. In view of the fact that, currently in Portugal, there is a lack of broader laboratory biosafety and biosecurity legislation, which is a key part of this type of strategy, this thesis was elaborated in order to provide the conceptual framework for the design and conception of a biosafety and biosecurity manual and to produce it for use at the Laboratory of Bromatology and Biological Defence of the Portuguese Army. The thesis begins by discussing the conceptual distinction between biosafety and biosecurity. It then proceeds to examine those concepts in the perspective of different organisations and considers its importance in the field of public health. Following the identification of the current challenges in biosafety and biosecurity, an analyses of the responses that have been given to these risks at a global, European and Portuguese levels is done, in particular related to the regulatory framework, norms and conventions between countries. In the Portuguese context, in addition to the approach to the current legislation and the new biosafety standard, were also considered the UPCAST project and the Portuguese Biosafety Laboratory Network. The thesis includes the presentation of the manual, which focuses on infrastructure, laboratory equipment, personal protective equipment, waste handling and disposal, good laboratory practices, among others. Finally, a critical discussion is made of the issues emerged from the discussion of the conceptual framework, seeking to contextualize them in relation to the purposes of the thesis and the guidelines that structured the design and conception of the manual.

Abstract : For many years, memory has fascinated the philosophers, psychologists, and neurobiologists. However, is not clear whether it is possible to obtain improved memory. It is known that acupuncture therapy is well-accepted in the world and its ancient and contemporary interventions exert a strong influence on brain-related pathologies. Previous studies show that acupuncture at acupoint Baihui (GV20) decreased the damage caused by stroke or cerebral ischemia as well as positive effects on neurofunctional rehabilitation and treat conditions such as headaches, dizziness and anxiety in both humans and in animal models. Thus, this study aimed to investigate whether manual acupuncture (MA) in Baihui (VG20) could improve learning and memory in mice. The animals were treated with MA for a minimum of 15 days and subjected to behavioral tests for evaluation of memory, anxiety, depression and pain.Treatment with MA in Baihui (VG20) caused a significant improvement in short and long-term memory in both the test object recognition as inhibitory avoidance when compared with the control and sham groups. In addition, the MA in Baihui (VG20) significantly increased the correct choices in the sixth, seventh and eighth days in eight-arm radial maze and reduced the total errors in the seventh and eighth days compared with the control and sham groups. On the other hand, the MA in Baihui (VG20) showed no anxiolytic activity when analyzed in the test box light/dark and elevated plus maze. However, it was able to promote significant antidepressant activity in the tail suspension test compared with the control and sham groups. In the formalin test, MA showed an antinociceptive effect associated with neurogenic pain, but not in inflammatory pain. Thus, the data from this study demonstrate that stimulation of the acupoint Baihui (VG20) by manual acupuncture induces improvement of learning and memory as well as having important antidepressant activity in mice.

Abstract:
The present study adopted a “field-laboratory-field” approach in the assessment of the efficacy of ergonomics interventions specific to two selected tasks evaluated in a South African automotive industry. Initial field testing was conducted in an Eastern Cape (South Africa) automotive plant where high risk areas were identified during walkthrough ergonomics surveys in conjunction with interaction with operators. Temporal factors and working postures of 12 industrial workers were recorded and observed, while physiological and perceptual responses were assessed. Two priority areas were focused upon for analysis, namely the Paintshop and Bodyshop with the former identified as being the more taxing of the two tasks. Responses of 30 students participating in rigourously controlled laboratory simulations were subsequently collected while completing the two tasks, namely the Paintshop Trolley Transfer (PTT) and Car Door Carriage (CDC) for participants. Working postures, kinematic, physiological and perceptual responses were assessed pre- and post-intervention. Following the laboratory experimentation a basic re-evaluation was conducted at the plant to assess whether the proposed changes had a positive effect on working postures, physiological and perceptual responses. The results of the preliminary field investigation revealed a prevalence of awkward working postures and excessive manual work in both areas. Laboratory experimentation revealed a notable reduction in task demands pre- versus post-intervention. The PTT mean lean angle for two-handed pre-intervention pulling observations of 23.7° (±3.51) was reduced to 13.9° (±2.21) post-intervention. Low back disorder (LBD) risk was reduced during the two-handed pull intervention (from 36.8% ±8.03 to 21.7% ±5.31). A significant decrement in heart rate responses from 103 bt.min-1 (±11.62) to 93 bt.min[superscript -1] (±11.77) was recorded during the two-handed symmetrical pushing intervention. The electromyography (EMG) responses for one-handed pushing and pulling pre-intervention showed the highest levels of muscular activity in the right medial deltoid due to an awkward and asymmetrical posture. CDC responses demonstrated that minor changes in the storage height of the door resulted in a significant reduction in sagittal flexion from 28.0° (±4.78) to 20.7° (±5.65). Predictions of average probability of LBD risk were significantly reduced from 50.3% (±5.91) to 39.8% (±5.10) for post-intervention car door lifting. In addition, the greatest reduction in EMG activity as a %MVC was achieved during sub-task ii (reduced from 35.1 to 13.7% and 30.5 to 13.9% for left and right erector spinae respectively) which was associated with the introduction of the transfer trolley for the door transfer phase of the CDC. Re-evaluation in the automotive plant revealed that the most notable change has been the implementation of automated ride on trolleys in the Paintshop. The Bodyshop area has also been modified to allow more effective job rotation and the step into the storage bin has been reduced via a “low-cost” stepping platform. Mean heart rate recordings were reduced from 94 (±9.77) bt.min[superscript -1] to 81 (±3.72) bt.min[superscript -1] in the Paintshop. Overall the results demonstrate the effect of “low-cost” interventions in reducing the physical stresses placed on workers in the automotive industry where much of the work is still done manually.

Abstract:
Promising results from the use of problem-based learning (PBL) as a teaching method in medical programs have encouraged many institutions to incorporate PBL into their curricula. This study investigates how applying hybrid-PBL (H-PBL) in a microbiology laboratory impacts students' higher-order thinking as compared to applying a lecture-based pedagogy. The experimental design compared the learning outcomes of two groups of students: the control group and the H-PBL group, for whom PBL cases comprised 30% of the curriculum. Both groups were taught basic skills for the microbiology lab by the same instructor. Using the traditional teaching style for the control group, the instructor offered each student what they needed for their experiments. The H-PBL group practiced experimental design, data analysis, theory proposal, and created research questions by using six study cases that were closely linked to the area of study. The outcome was measured using a pre- and post- assessment consisting of 24 questions that was designed by following Bloom's taxonomy of learning levels. A one-way ANOVA was used to analyze the data. The results showed that for the first three levels of Bloom's taxonomy— knowledge, comprehension, and application—there were no statistically significant differences between the H-PBL and control group gain scores as determined by a one-way ANOVA. For the knowledge level, f (1, 78) = .232, and p = .632; for the comprehension level, f (1, 78) = .004, and p = .951; and for the application level f (1, 78) =. 028, and p =.863. On the other hand, the gain scores for the three higher levels—analysis, evaluation, and creativity—improved for the H-PBL group. The analysis level showed statistically significant differences, with f (1, 78) = 4.012, and p = .049. Also, there were statistically significant differences in students' performance at the evaluation level, with f (1, 78) = 11.495, and p = .001, and the creativity level, with f (1,78 ) = 23.432, and p = .000. In conclusion, the study results supported the value of incorporating hybrid problem-based learning (H-PBL) into the traditional microbiology laboratory curriculum.

Abstract:

Marshall, Katherine C. "An Evaluation of the Water Lifting Limit of a Manually Operated Suction Pump: Model Estimation and Laboratory Assessment." Scholar Commons, 2017. http://scholarcommons.usf.edu/etd/7056.

Full text
Abstract:
With 663 million people still without access to an improved drinking water source, there is no room for complacency in the pursuit of Sustainable Development Goal (SDG) Target 6.1: “universal and equitable access to safe and affordable drinking water for all” by 2030 (WHO, 2017). All of the current efforts related to water supply service delivery will require continued enthusiasm in diligent implementation and thoughtful evaluation. This cannot be over-emphasized in relation to rural inhabitants of low-income countries (LICs), as they represent the largest percentage of those still reliant on unimproved drinking water sources. In that lies the motivation and value of this thesis research- improving water supply service delivery in LICs. Manually operated suction pumps, being relatively robust, low cost, and feasible to manufacture locally, are an important technology in providing access to improved drinking water sources in LICs, especially in the context of Self-supply. It seems widely accepted that the water-lifting limit of suction pumps as reported in practice is approximately seven meters. However, some observations by our research group of manually operated suction pumps lifting water upwards of nine meters brought this “general rule of thumb” limit into question. Therefore, a focused investigation on the capabilities of a manually operated suction pump (a Pitcher Pump) was conducted in an attempt to address these discrepancies, and in so doing, contribute to the understanding of this technology with the intent of providing results with practical relevance to its potential; that is, provide evidence that can inform the use of these pumps for water supply. In this research, a simple model based on commonly used engineering approaches employing empirical equations to describe head loss in a pump system was used to estimate the suction lift limit under presumed system parameters. Fundamentally based on the energy equation applied to incompressible flow in pipes, the empirically derived Darcy-Weisbach equation and Hydraulic Institute Standards acceleration head equation were used to estimate frictional and acceleration head losses. Considering the theoretical maximum suction lift is limited to the height of a column of water that would be supported by atmospheric pressure, reduced only by the vapor pressure of water, subtracting from this the model was used to predict the suction lift limit, also referred to herein as the practical theoretical limit, assuming a low (4 L/min) and high (11 L/min) flow rate for three systems: 1) one using 1.25-inch internal diameter GI pipes, 2) one using 1.25-inch internal diameter PVC pipes, and 3) one using 2-inch internal diameter PVC pipes. In all considered cases, with an elevation equal to sea level, the suction lift limit was estimated to be over nine meters. At a minimum, the suction lift limit was estimated to be approximately 9.4 meters for systems using 1.25-inch internal diameter pipe and 9.8 meters for systems using 2-inch internal diameter pipe, with essentially no discernable effects noticed between pipe material or pipe age. Additionally, laboratory (field) trials using a Simmons Manufacturing Picher Pump and each of the aforementioned pipe specifications were conducted at the University of South Florida (Tampa, FL, USA) to determine the practical pumping limit for these systems. Results from the pumping trials indicated that the practical pumping limit- the greatest height at which a reasonable pumping rate could be consistently sustained with only modest effort, as perceived by the person pumping- for a Pitcher Pump is around nine meters (9 meters when using 1.25-inch internal diameter GI or PVC pipe and 9.4 meters when using 2-inch internal diameter PVC pipe). Therefore, results from this research present two pieces of evidence which suggest that the practical water-lifting limit of manually operated suction pumps is somewhere around nine meters (at sea level), implying that reconsideration of the seven-meter suction lift limit commonly reported in the field might be warranted.

Abstract:
--- Over the past years has shown the importance of organizations leads to a quality management system to define the interaction and set controls for each process involved, looking for customer satisfaction and promote the pursuit of continuous improvement, enabling it to adapt quickly in a market which is constantly changing. The aim of this analysis prospective-descriptive is to design a quality manual and procedures which allows standardize documentation for all process in an organization, based on a Certifiable International Standard as ISO 9001:2008. For this reason the norm requires to maintain a quality management system. The norm requires a procedure which gives directives for the control of documents to serve as a means of diffusion on activities that take place in every process within the Organization, also requires to include a procedure that gives directives for the control of records generated in each activity of the Organization, call for a procedure that explain the treatment given to each product that is out of its specifications, requires a procedure that explains the treatment that is given to any non-conformance or potential non- conformance in regards or documentation generated by the system and, call for a procedure to give guidelines to audit the quality management systems in order to check effectiveness and continuous improvement. Similarly, show the importance of having a Quality Manual that allows communicate to the Organization, the quality policy and objectives which was established by the senior management and communicated to each one of the partners, and the processes that are part of the scope of the system and the interaction of each of them, besides the mention of complementary procedures that the Organization has decided to implement. In July of 1999, I wrote to 24 Texas junior and community colleges (and one four-year institution) describing my thesis agenda of a general biology lecture and laboratory syllabus for introductory biology students. I requested the titles and authors of the general biology textbooks and laboratory manuals they were currently using, the publishers of these texts, and the edition of said texts. I then contacted publishers of the various textbooks who, in turn, directed me to the Dallas-area representatives for further inquiries. I assimilated the various authors' general biology topics into a two-semester syllabus of lecture and one semester of laboratory. The document is not a text manuscript, but an all-inclusive listing of a general biology syllabus broken down by subject.

Abstract:

Abstract:
Leukocyte differential count in peripheral blood smear has, ever since the method was developed more than 100 years ago, been one of the most frequently used diagnostics tool in the routine hematology laboratory. The manual differential count of leukocytes using a microscope is still the standard method in most small and medium sized laboratories. Even though the method does not require any expensive instruments it comes at a high cost due to it being labor intensive and time consuming. In recent years the rapid technical advancements has led to the development of automatic or semi-automatic methods in which the leukocytes are differentiated. In this study a method comparison was made between manual leukocyte differential counts, CellaVision DM1200 Vet and Advia 2120 when analyzing 106 fresh, feline blood samples. The general agreement between results was good, especially for the most common leukocytes, such as neutrophils and lymphocytes. Results for eosinophils and monocytes had moderate agreement. The confidence intervals were generally wider when CellaVision DM1200 Vet was compared with Advia 2120, than when CellaVision DM1200 Vet was compared to the manual differential count. Despite the fact that Advia 2120 and CellaVision DM1200 Vet are both faster and often show comparable results to the manual differential count, the light microscopy will remain the gold standard for difficult samples, where there is suspicion of inflammation (band neutrophils), intracellular microorganisms, reactive lymphocytes or if the sample contains a high degree of smudge cells or artifacts.

Abstract:
Traditional chemistry laboratory courses have a manual consisting of “step-by-step” experiments; instructions are given to complete experiments, requiring minimal information/concepts processing to be successful. This experience leaves students unprepared for the real-world, where critical thinking skills are needed to conduct research. This study focused on building analytical techniques, conceptual knowledge, and critical thinking skills used to solve research problems. A new quantitative chemistry laboratory manual was developed to transition students from traditional to inquiry-based experiments, requiring analytical method development. Data showed students having less difficulty using the new manual (0.8281 average difficulty) on method development exam questions and experiments, compared to the traditional manual (0.600 average difficulty). T-test showed significant difference between item difficulty, p = 0.029. Using null hypotheses, the new laboratory manual led to an increase in students’ conceptual knowledge and research skills. They were able to use their knowledge and skills to successfully solve real-world related problems.

Nader, Athir. "Engineering Characteristics of Sensitive Marine Clays - Examples of Clays in Eastern Canada." Thèse, Université d'Ottawa / University of Ottawa, 2014. http://hdl.handle.net/10393/30670.

Full text
Abstract:
Sensitive marine clay in Ottawa is a challenging soil for geotechnical engineers. This type of clay behaves differently than other soils in Canada or other parts of the world. They also have different engineering characteristic values in comparison to other clays. Cone penetration testing in sensitive marine clays is also different from that carried out in other soils. The misestimation of engineering characteristics from cone penetration testing can result. Temperature effects have been suspected as the reason for negative readings and erroneous estimations of engineering characteristics from cone penetration testing. Furthermore, the applicability of correlations between cone penetration test (CPT) results and engineering characteristics is ambiguous. Moreover, it is important that geotechnical engineers who need to work with these clays have background information on their engineering characteristics. This thesis provides comprehensive information on the engineering characteristics and behaviour of sensitive marine clays in Ottawa. This information will give key information to geotechnical engineers who are working with these clays on their behaviour. For the purpose of this research, fifteen sites in the Ottawa area are taken into consideration. These sites included alternative technical data from cone and standard penetration tests, undisturbed samples, field vanes, and shear wave velocity measurements. Laboratory testing carried out for these sites has resulted in acquiring engineering parameters of the marine clay, such as preconsolidation pressure, overconsolidation ratio, compression and recompression indexes, secondary compression index, coefficient of consolidation, hydraulic conductivity, clay fraction, porewater chemistry, specific gravity, plasticity, moisture content, unit weight, void ratio, and porosity. This thesis also discusses other characteristics of sensitive marine clays in Ottawa, such as their activity, sensitivity, structure, interface shear behaviour, and origin and sedimentation. Furthermore, for the purpose of increasing local experience with the use of cone and ball penetrometers in sensitive marine clays in Ottawa, three types of penetrometer tips are used in the Canadian Geotechnical Research Site No. 1 located in south-west Ottawa: 36 mm cone tip, and 40 mm and 113 mm ball tips. The differences in their response in sensitive marine clays will be discussed. The temperature effects on the penetrometer equipment are also studied. The differences in the effect of temperature on these tips are discussed. Correlations between the penetrometer results and engineering characteristics of Ottawa's clays are verified. The applicability of correlations between the testing results and engineering characteristics of sensitive marine clays in Ottawa is also presented in this thesis. Two correlations from the Canadian Foundation Engineering Manual are examined. One of these correlations is between the N60 values from standard penetration testing and undrained shear strength. The other correlation is between the shear wave velocity measurement and site class. Temperature corrections are suggested and discussed for penetrometer equipment according to laboratory calibrations. The significance of the effects due to radical temperature changes in Canada and Ottawa is discussed. Some of the main findings from this research are as follows. • The Canadian Foundation Engineering Manual presents a correlation between standard penetration tests (SPTs) and the undrained shear strength of soils. This relationship may not be applicable to sensitive marine clays in Ottawa. • Another correlation between the site class, shear wave velocity, and undrained shear strength is presented by this same manual which may not be applicable to sensitive marine clays in Ottawa. • The rotation rate for field vane testing as recommended by ASTM D2573 is slow for sensitive marine clays in Ottawa. • Correction factors applied to undrained shear strength from laboratory vane tests may not result in comparable values with the undrained shear strength obtained by using field vane tests. • Loading schemes in consolidation or oedometer testing may affect the quality of the targeted results. • Temperature corrections should be applied to penetrometer recordings to compensate for the drift in the results of these recordings due to temperature changes. • The secondary compression index to compression index ratio presented in the literature may not be the value obtained from this research.

In 1999, the policy of generic medicaments was implemented in Brazil, and the availability of pharmaceutical equivalence essays became strategic for its success. A group of laboratories was identified, evaluated and qualified, as a network, by the National Sanitary Surveillance Agency (Anvisa) to perform such studies. These laboratories were named âcenters for pharmaceutical equivalenceâ (EQFAR), and were integrated to The Brazilian Network of Analytical Laboratories on Health (REBLAS). The criteria for qualification and operation of the centers, as well as to perform standardized pharmaceutical equivalence essays, were established by a set of regulations (laws, resolutions and technical rules). The extent, diversity and complexity of these regulations do not always facilitate its knowledge, comprehensive and correct application for people interested in implementation of those rules on the centers as well as the National Sanitary Surveillance Agency tasks of evaluation, qualification and audition. This material consolidate the content of the regulation in force, systematizing them according the following chapters: referential documentation, human resources, biosafety and laboratorial organization, facilities and installations, quality assurance, equipment, materials, and the principals essays that compose the studies of pharmaceutical equivalence and the preparation of their analytical certification. It aims to be a practical instruction â like a manual â that allows the rapid access to the weighty information and to facilitate the work of people interested in implement a center of pharmaceutical equivalence and for those from the Sanitary Surveillance Agencies

Abstract:
Background: Agriculture plays a vital role in the economy of any industrially developing country, including South Africa. In the Eastern Cape of South Africa citrus farming is a significant contributor to the local economy (Johnson et al., 2005). The harvesting phase of citrus farming is performed manually and exposes workers to physical risks, which can lead to the development of musculoskeletal disorders. In particular, the standard harvesting bag comprises of a single shoulder strap and promotes asymmetrical load carriage which results in shoulder and lower back pain complaints. The current study compared the physiological (EMG), perceptual (RPE), usability (PUEU) and productivity effects of two new harvesting bag designs (a hip belt and a backpack bag design) to the standard harvesting bag design. This was performed in a laboratory as well as a field setting. Methods (Laboratory phase): 36 participants (12 males and 24 females) were assigned to one worker group. The “tall ladder worker” group was comprised of only males and the “step ladder worker” and “ground worker” group of females. Each participant was required to simulate a citrus harvesting task while utilizing each of the bag designs on different days. On each day/test session, participants performed three harvesting cycles. Muscle activity was measured throughout the entire testing session and RPE were recorded at the end of each cycle. Results (Laboratory phase): The EMG and RPE results indicate that the backpack design was the most ideal design to reduce asymmetry, while the standard harvesting bag design was the worst. Although not significant, there was greater muscle asymmetry (p=0.109) and a significantly higher perceived exertion when using the standard bag (p=0.0004), in comparison to using the backpack. Methods (Field phase): 17 Xhosa-speaking citrus harvesters (6 females and 11 males) participated in this study. Each harvester worked with one of the three bag designs on a different day. Productivity of each worker was assessed every hour by recording the number of bags filled with fruit and at the end of the shift. A Perceived Usefulness & Ease of Use questionnaire was presented to each participant to obtain feedback on worker acceptance to the new bag designs. Results (Field phase): A general trend in support of the hip belt bag design over the other two bag designs were found, even within the different worker demographic groups (age, sex and worker experience). The workers perceived less exertion (7.98 ± 1.86) and were more productive (9.90 ± 2.11 bags/hour) when using the hip belt design; they also found this bag the most useful (1.02 ± 0.09) and easy to use (1.07 ± 0.25). In contrast, the backpack bag design had significantly poorer responses when compared to the other two bag designs and this was evident in all the dependent variables assessed (RPE, productivity and PUEU). Conclusion: The results from the laboratory phase supported the expectation that the backpack bag design reduces asymmetry and hence, is more suitable than the standard harvesting bag. However, results from the field show that the hip belt bag design was the most preferred and the backpack was the least preferred. Bao & Shahnavaz (1989) highlight the need for ergonomics researcher to convey laboratory findings into the field context. However, as shown by the current study, there are numerous challenges associated with field work, making it difficult for laboratory findings to be successfully conveyed to the field. Limitations and Recommendations: For the laboratory phase of the project, no biomechanical and cardiovascular responses were assessed. However, for a holistic approach, these variables should be considered in future studies. Due to high variability from one harvesting cycle to another, more than three harvesting cycles should also be performed to accurately replicate the harvesting process as done in the field over extended durations of time. For the field phase, data should be collected from more than one citrus farm and thus a larger sample size could be obtained. This would improve the validity of the study. In addition to this, data should be collected for a full working day, especially if environmental conditions are not a hindrance, as well as for a whole season, since workloads vary, depending on the time of the harvesting season.
