His Gothic novel The Monk contains all the typical Gothic elements such as a ruined castle, aggressive villain, women in distress, the atmosphere of terror and horror and a lot more. This article analyses and compares to what extent the Gothic elements of the late 18th century survived in the contemporary detective story The Ice House (1993) written by Minette Walters and how these elements have changed.

Caravaggio's seminal ambitious program in the Contarelli Chapel (San Luigi dei Francesi, Rome) representing the Calling and the Martyrdom of St. Matthew (1599-1600) invoked the threat of iconoclasm as it imagined the martyr's body as a violated relic. Counter-Reformation images and hagiographies, such as the engravings in Richard Verstegan's Thééââtre des cruautéés des hééréétiques nostre temps, traduit du latin en franççois (Anvers, 1588) and Giovanni Battista Cavalieri's Ecclesiae Anglicanae Trophaea (Rome, 1584), after paintings by Pomarancio, depicted disintegrated sacred bodies subject both to iconoclastic violence and to confusion with polluted bodies. This conflation of social categories in Caravaggio's paintings led his biographers to blame him for the endemic replacement of classical antiquity by persons off the street. The retrospective account by classical theorists displaced an earlier archaeological project that took the ruined saint's body as its artifact: the scientific archaeology of the Christian relic in the Roman catacombs in Antonio Bosio's Roma Soutteranea (Rome, Latin edition, 1634; Italian edition, 1650). The threat of the disintegration of the composition in Caravaggio's paintings thematized the paradox of the incorruptible yet violated flesh of the martyr.The painting, like the relic, recapitulated the biographical narrative of martyrdom. It also proposed patterns of reception based on contemporary rhetorical models: the descriptive enumeration of Christian humanist ekphrasis based on Prudentius's fourth century Crowns of Martyrdom and articulated in Gregory Martin's late-sixteenthcentury guide to Sacred Rome.

Introduction. In spite of the fact that major researchers of Gregory Palamas’ theology, Robert Sinkewicz and John Demetracopoulos, offered detailed explanations of when and for what ‘The chapters on physics’ of Gregory Palamas were written, it seems that this issue was not finally resolved. Methods. This article, therefore, has two goals. The first is to answer the question why in ‘The chapters on physics’ the polemic against Barlaam and Akindynos, although it occupies a large part of the treatise, is presented as a part of the broader theological and philosophical program of Palamas, and what role does the cosmological part (1–14 chapters) play in the structure of the treatise. In the author’s opinion, such a statement of the question will allow us to understand what Gregory Palamas was guided by when writing the text. The second objective is to offer a social portrait of the addressee of this treatise, delivering some of representatives of the stratum on which the Palamas was oriented. Analysis. In answering the first question, special attention is paid to chapter 81 of the treatise, which, in the author’s opinion, reflects the intent of the entire text, namely, to demonstrate that the logic according to which something indivisible can be thought of as divisible without ceasing to be indivisible, works not only in the case of essence and energy, that is, in theology, but also in cosmology and anthropology. In answering the second question, the focus is on the figure of Matthew Kantakouzenos, who was at the time of writing the treatise in conflict with his father, emperor John VI Kantakouzenos, thereby causing discord in the political party, which at that time was a defender of the ideas of Hesychasm. It was Matthew Kantakouzenos, in the author’s opinion, or the social stratum behind him, who could be the recipients of the treatise. Results. The analysis of the treatise and its historical context shows that it was written largely for political reasons as a program text of Hesychasm, equipped with natural science and philosophical tools, since its recipients were both the broad educated strata of Byzantine society and direct participants in political life in the middle of the 14th century.

The article is devoted to a new interpretation of the well-known postscript in the so-called Ostromir Gospels—the earliest Russian manuscript, written in 1056/1057. In this afterword on fols. 294r and 294v, one of scribes (deacon Gregory by name) twice asked for a blessing for Ostromir the “Posadnik,” who had paid for the creation of this outstanding manuscript. The first blessing requested is the blessing of the Holy Evangelists. Although it is perfectly appropriate for someone who ordered such an expensive Gospel book to ask for blessings from the evangelists, the names of the evangelists were listed in an unusual sequence. The order of names corresponds to the order in which selected fragments of the Gospels (Pericopes) are read in “aprakos” Gospels: John, Matthew, Luke, Mark. The second blessing is quite uncommon. Gregory asks the three biblical patriarchs—Abraham, Isaac, and Jacob—to bless Ostromir. The most reliable explanation of this request is the Christian name of Ostromir, which is mentioned in the scribe’s note—Joseph. In Old Rus’ people considered everyone named Joseph as a bearer of the name of Joseph the All-Comely (Genesis). So Gregory appears to flatter Ostromir implicitly by calling him the all-comely son of Jacob, grandson of Isaac, and great-grandson of Abraham. But to stress the desirable and hoped-for similarity, Deacon Gregory mentions Ostromir-Joseph twice as the first after the Great Duke of Kiev, just as the biblical Joseph was named the first after Pharaoh. This shows that in pre-Mongol Rus’ there was a thorough knowledge of the texts of the Old Testament and that there was a readiness to include these skills in a complicated intellectual game.

Medieval popes can scarcely have expected such spectacular results from a bull as Gregory EX achieved in 1237. His bull Cum hora undecima of 1235, a fundamental statement of the Church’s missionary function, gave specific licence to the Dominican William of Montferrat to preach, dispense the sacraments, absolve and excommunicate in the lands of schismatics and heretics of the East. Two years later, Philip, the Dominican provincial of the Holy Land, wrote to the pope announcing the conversion to Rome of the Syrian Orthodox (Jacobite) patriarch of Antioch, Ignatius II, the anticipated conversion of the Nestorian catholicos in Baghdad and possibly also the conversion of the Coptic patriarch. It was a staggering return from a mission only two years old, and represented a triumph for the Dominican Order as well as for the papacy.

Nearly two and a half centuries have passed since the first British Gothic novels began to attract attention with their pages full of monstrous characters, excessive violence, explicit sexual content and all kinds of horrific scenes. For the most part, the reception of this type of literature has been very positive, though not exempt from controversies. This paper seeks to show how, beyond the alluring mystery, inventive plots and attraction of the dark side, British horror fiction appeals to the reader’s inner desires and imagination by means of transgressive political, religious or sexual contents that often defy taboos and social decorum. To illustrate this argument, three well-known authors and texts from three different periods will be discussed: Matthew Gregory Lewis’s The Monk (1796), Joseph Sheridan Le Fanu’s “Carmilla” (1872) and Anthony Burgess’s A Clockwork Orange (1962).

Or nous tenons à démontrer que c’est par le biais d’un mélange du mythe faustien et du moine sadique du roman noir, que Notre-Dame de Paris complique de très loin la signification accordée traditionnellement entre les deux forces opposées du bien et du mal, surtout en ce qui concerne l’archidiacre Claude Frollo. In his work on retranslation, Antoine Berman is probably the theorist who came closest to reflecting on back-translation. This article offers interpretations of two of his premises in ‘La retraduction comme espace de traduction’: that all translations are impaired by forces of non-translation and that this phenomenon is attenuated by retranslation. It is partly to investigate these hypotheses that Berman developed the concept of ‘défaillance’. The article traces the evolution of Berman's notion before demonstrating how the study of ‘défaillances’ across translative layers can be enlightening, by analysing three scenes in Matthew Gregory Lewis’ gothic novel The Monk (1796), Antonin Artaud's French translation (1931), and John Phillips’ back-translation (2003). It argues that the study of back-translations is valuable retrospectively, insofar as it magnifies elements which were underdeveloped in source-texts, and that, in so doing, it has the potential to transform our understanding of the larger trajectory of literary works.

Queer Gothic may be considered one of the most recent subgenres of Gothic literature, being understood as a type of fiction in which body, gender and sexuality assume a central role. I approach the representation of queer sexualities in queer Gothic works, basing my analysis on the theory of abjection, as well as on other authors who work with Gothic fiction and Queer Studies, such as Judith Butler (1990), William Hughes and Andrew L Smith (2009) and Paulina Palmer (2016). My analysis consists of drawing a critical panorama, then commenting on Gothic works such as The Monk (1796), by Matthew Gregory Lewis, and The Fall of the House of Usher (1839), by Edgar Allan Poe, culminating in contemporary works such as the novel The Lazarus Heart (1998), by the trans author Poppy Z. Brite. It is possible to consider that in the aforementioned works Queer desires are articulated with supernatural or psychological phantasmagorias, assuming a liminal aspect which denounces the fragmentation of the Queer subject in face of hegemonic society. On the other hand, the representation of Queer sexualities has subversive potentials, while the hegemonic discourse imposes on them the stigma of abjection.

AbstractThis article takes off from what Lyotard calls ‘the immaterial’, demonstrated in the exhibition Les Immatériaux that he curated at the Centre Pompidou in 1985. It aims at outlining a concept of ‘relational materiality’. According to Lyotard, ‘the immaterial’ is not contrary to material: instead, it is a new industrial material brought about by telecommunication technologies, exemplified by Minitel computers, and serves as basis to describe the postmodern condition. Today this materiality is often referred to as ‘the digital’. In order to enter into a dialogue with Lytoard, and to render his notion of ‘immaterial materials’ contemporary, this article contrasts the concept of relational materiality with some current discourses on digital physics (Edward Fredkin, Gregory Chaitin) and digital textuality (Matthew Kirschenbaum). Against the conventional conception that relations are immaterial (neither being a res nor even having a real esse), and also contrary to a substantialist analysis of materiality, this article suggests that a relational materiality is made visible and explicit under digital conditions. It suggests a reconsideration of the ‘relational turn’ in the early 20th century and the concept of concretisation proposed by Gilbert Simondon. The article concludes by returning to Lyotard’s notion of materialism and his vision of a new metaphysics coming out of this ‘immaterial material’, and offers ‘relational materialism’ as a contemporary response.

Arriving at the Lateran on 8 January 1198, officials conducted Innocent III (born Lotari dei Conti di Segni) ceremonially to his apartments within the palace, there to rest, pray and dine.’ Foremost amongst his concerns was the household, last reformed by Gregory I (590–604). Whilst Innocent clearly adopted Gregory as his model, both for the shaping of his personal life as pope and for his understanding of the papal office, the young pope’s efforts to make his household as exemplary as that of his great predecessor have not received the attention they undoubtedly deserve. Gregory’s finest Life, composed c.875 by John, a Roman deacon, uses material from the early vitae, thus avoiding the ‘scrappy and grudging’ biography of the Liber pontificalis. Instead, John draws extensively on Gregory’s letters and the crumbling but then still extant papyrus volumes of the Registrum to demonstrate how this pope transformed his household into monastery, hospice and refuge. Three centuries later, the author of the Gesta Innocentii or Deeds of Innocent III could do no better than to adapt portions of John’s Life to highlight reforms not evidenced since the sixth century Like Gregory, Innocent wished to restore the ideas of the apostolic age to the Church. And where better to begin the spiritual renewal than within a reformed household? His inaugural sermon as pope on St Matthew’s faithful and wise servant accords perfectly with John the Deacon’s view of Gregory as paterfamilias Domini, head of the Lord’s household. Innocent, therefore, regarded the household not only as a metaphor for the congregation of the faithful but also, like Gregory before him, as a model to be used by missionaries to plant and nurture the faith throughout Christendom. Whilst the ongoing conversion of Livonia would provide Innocent with a rare opportunity to inculcate the Christian household within a pagan society, in the Patrimony of St Peter he diverged from Gregory’s path by purposeful itineration with his familia, thus initiating a public role for the household.

Abstract Lung cancer is one of the top five deadliest cancers, and Black Americans have a higher mortality rate when compared to White Americans. Lung adenocarcinoma (LUAD) is the most common histological lung cancer subtype in all population groups including Blacks. Unfortunately, there are no in vitro model systems to study lung cancer and its treatments for Black subjects. Herein we aim to utilize three-dimensional (3D) printing to develop an in vitro three-dimensional (3D) alveolar model using a lung adenocarcinoma cell line from a Black patient (NCI-H23). The developed model is further utilized to study the therapeutic effects of potential LUAD therapeutic agents, polyisoprenylated cysteinyl amide inhibitors (PCAIs), which target and disrupt the RAS-mediated signaling pathway. KRAS is the most commonly mutated oncogene in LUAD in Blacks, and is mutated in NCI-H23 cells. For comparison, hAEC-FT-C7 cells, which are immortalized human alveolar epithelial cells and do not carry the KRAS mutation, will also be used separately to print the alveolar models and test the effects of PCAIs. The experimental results will be further compared to those using two-dimensional (2D) models to better understand the therapeutic effects of PCAIs on Black LUAD. The resulting knowledge will help design better treatments for ethnically/racially diverse patients in whom the RAS signaling pathway is frequently altered. Citation Format: Nadine L. Abstract Background: Recent advances in the treatment of non-Hodgkin lymphoma (NHL) have transformed the landscape and provided significant benefits to patients. Novel agents that target CD19 and CD20 on B cells, such as bispecific T-cell engagers and chimeric antigen receptor (CAR) T cell therapy, have demonstrated benefit for patients in the relapsed and refractory setting. However, these agents can often be associated with significant toxicity and there is a need for new therapies that can provide clinical benefit while providing an improved safety profile.IGM-2323 is a novel CD20 x CD3 bispecific antibody utilizing an IgM backbone. This allows targeting of up to 10 CD20 binding sites for every CD3 site. In preclinical models, IGM-2323 demonstrated excellent antitumor activity with controlled T-cell activation. IGM-2323 is able to stimulate T cells at a more physiologic level than IgG-based antibodies, which may reduce adverse events typically associated with T-cell engagers and CAR-T, such as CRS and neurotoxicity. Methods: This study is a phase 1/2, multicenter, single-arm clinical trial of IGM-2323 monotherapy for patients with relapsed/refractory NHL. The Phase 1 component consists of a dose escalation and limited dose expansion evaluating multiple titration dose levels. As part of the titration dosing regimen, subjects receive weekly dosing on Day 1, 8, and 15 of each 21- day cycle. Dosing begins at 15 mg and is increased weekly for 3-4 weeks to reach the plateau dose. Patients then stay at the plateau dose until disease progression or unacceptable toxicity. Patients who achieve a response by Week 12 may switch to a less frequent dosing interval of every 3 weeks. The Phase 2 component will randomize patients at two different dose levels (100 mg and 300 mg plateau dose) in two separate indications (R/R DLBCL and R/R FL). Primary endpoints include frequency and severity of adverse events and objective response rate (ORR) based on Lugano criteria. Correlative biomarker studies will evaluate the correlation of clinical benefit with blood and tissue biomarkers. In Phase 2, there will be a formal analysis after 30 subjects are evaluable at each dose level and the best dose for each indication will be selected for further evaluation. Beckett's investigations in the history of philosophy are well represented in his notebooks of the late 1920s and early 1930s, which provide a close record of his reading in ancient, medieval, and modern philosophy, as well as in history, literature, and psychology. Numerous scholars – Daniella Caselli, Anthony Uhlmann, Dirk Van Hulle, Matthew Feldman, and David Addyman among others – have carefully delineated the relationship between Beckett's note-taking and his deployment of philosophical sources in his literary texts. Whilst the focus quite rightly tends to fall on Beckett's absorption of Presocratic, Aristotelian, Cartesian, and post-Cartesian philosophy, there are important strands of early medieval philosophy that find expression in his literary work. The philosophy notes housed in the library of Trinity College, Dublin, provide insights into Beckett's reading in medieval philosophy, drawing almost exclusively from Wilhelm Windelband's History of Philosophy. The epoch spanning from Augustine to Abelard saw central concepts in theology and metaphysics develop in sophistication, such as matters of divine identity and non-identity, the metaphysics of light, and the nature of sin. The influence of the Eastern Church Fathers (Gregory of Nyssa, Basil of Caesarea, Maximus the Confessor) on Western metaphysics finds expression in the figuration of light and its relation to knowing and unknowing. This eastern theological inflection is evident in the ‘Dream’ Notebook, where Beckett's notes demonstrate his careful reading of William Inge's Christian Mysticism. These influences are expressed most prominently in various themes and allusions in his early novels Dream of Fair to Middling Women, Murphy, and Watt. The formal experiments and narrative self-consciousness of these early novels also respond to the early medieval transformation of textual form, where the precarious post-classical fruits of learning were preserved in new modes of encyclopaedism, commentary, and annotation. Beckett's overt display of learning in his early novels was arguably a kind of intellectual and textual preservation. But the contest of ideas in his work subsequently became less one of intellectual history and more that of immanent thinking in the process of composition itself.

The indulgence is one of the Church’s mechanisms for encouraging pious practices which has generally had a bad press. This paper is concerned with its use in the veneration of three images: the Veronica, the arma Christi or instruments of the Passion, and the Man of Sorrows with the indulgence referring to the Mass of St Gregory. It addresses in particular the circumstances of the original grant of indulgence (whether real or spurious) and the role played by the indulgences in the transmission of these images in devotional manuscripts in England. The earliest of these indulgenced images, the Veronica, is also the most famous, and the one whose origin is most clearly attested. The Chronica maiora of Matthew Paris tells how the Veronica suddenly reversed itself while being carried in procession in 1216. Innocent III responded by composing a prayer in its honour, with an associated indulgence of ten days for each time the prayer was recited. It is noteworthy that although Innocent’s indulgence is commonly referred to as creating the new category of indulgenced image, the indulgence is attached to the prayer, and there is no suggestion that it was necessary to view the image. As is made clear in the Chronica, the addition of a representation of the image (plate 1) was prompted by the fervour of devotion: people did it for themselves. Thus two related, but not identical, impulses joined together to produce the indulgenced prayer to be said before the image, the model for all other indulgenced images. An early example of the prayer used as part of an exercise in devotional propaganda is found in the Revelations of Mechtild of Hackeborn. These normally followed the liturgical year, and on the day of the exposition of the relic she had a vision, aimed at arousing piety towards the image, in which she saw Christ in Majesty, with those who honoured the holy face with a special prayer approaching him,

Abstract Lung cancer is one of the top five deadliest cancers, and black Americans have a higher mortality rate when compared to white Americans even though they smoke relatively fewer cigarettes. Lung adenocarcinoma (LUAD) is the most common histological lung cancer subtype in all population groups including blacks. There are no in vitro model systems to study lung cancer in black subjects, and there is only one lung adenocarcinoma cell line from a black patient (NCI-H23). Here we aim to address the lack of in vitro models to study black LUAD. Using three-dimensional (3D) biofabrication technologies, we’ve developed a thick vascularized tissue model to recapitulate physiologically relevant heterogeneous human alveoli. Specifically, the blood-air barrier of the alveolar model is simulated with two perfusable channels embedded in a thick vascularized tissue construct: a helical blood channel and a straight air channel. Spatially, the air channel is in the center and is surrounded by the blood channel. Human endothelial cells are seeded onto the inner surface of the blood channel and NCI-H23 cells (which carry a KRAS mutation) or untransformed alveolar epithelial cells derived from black subjects, are seeded on the inner surface of the air channel. The blood and air channels are perfused using cell medium and oxygenated air, respectively. The model is used for the study of potential LUAD therapeutic agents such as polyisoprenylated cysteinyl amide inhibitors (PCAIs), which target and disrupt the RAS-mediated signaling pathway. KRAS is the most commonly mutated oncogene in LUAD in blacks, and our experiments are aimed at ensuring that PCAIs are effective in black LUAD and may offer better treatment outcomes for ethnically/racially diverse patients in whom the RAS signaling pathway is frequently altered and for whom pharmaceutical therapy is necessitated by the late diagnoses. Abstract Lung cancer is a leading cause of cancer-related mortality accounting for 23% of cancer deaths in the US. Non-small cell lung cancer (NSCLCs) comprises 84% of all lung cancer cases, and of these, 40% are lung adenocarcinoma (LUAD). Moreover, black Americans are underrepresented in scientific clinical trials but have a lung cancer incidence rate that is 37% higher than their white counterparts. To address this disparity and lack of effective treatment options, we determined the potency of polyisoprenylated cysteinyl amide inhibitors (PCAIs) on cell viability and its effects on KRAS and its downstream mediators in NCI-H23, a black American cell line that harbors a KRAS mutation. Among the 17 PCAIs that were tested, compound NSL-YHJ-2-27 showed the most potency with an EC50 of 2.7 µM. Western blot analysis was performed to determine the effect of the PCAIs on the levels of Mitogen-Activated Pathway Kinase (MAPK)-related proteins. After 48 h of exposure to 5 μM of NSL-YHJ-2-27, we observed increases of 150, 64.5, and 77% in phosphorylation levels of ERK1/2, MEK1/2, and 90RSK. While the particular 90RSK enzymes whose phosphorylation is/are elevated are not known at this time, 90RSK 3 and 4 are known to be proapoptotic. These findings suggest that the PCAIs may be used to target cancers with KRAS mutations that currently lack effective therapies. In the future, we will use a collection of isogenic cell lines to examine the potency of the PCAIs in LUAD driven by other RAS and related monomeric G-proteins, epidermal growth factors, and their receptors as well as RAS-GAPs. Therapies emanating from these studies have the potential to contribute to mitigating the lung cancer therapeutic inequities. Abstract Lung cancer is a source of racial disparity among Black Americans, especially men. Despite lower tobacco exposure among Black men than White men, Black men are 37% more likely to develop lung cancer. Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer in all ethnic/racial groups, including Blacks. KRASis the most frequently mutated driver gene in Black LUAD (~30%), and NRAS mutations are also seen (1-2%). A given oncogenic mutation could have different effects on a patient based on their racial/ethnic genetic background. Thus, studying the appropriate driver mutations in the correct model and genetic background is essential. Unfortunately, to our knowledge, there is only one LUAD cell line from a Black subject. Therefore, we are deriving immortalized human alveolar epithelial cell (hAEC) lines from Black Americans to develop models to study lung adenocarcinoma in Blacks. The Offringa lab has recently developed a rapid method to generate hAEC lines. We have generated three distinct hAEC lines from de-identified remnant transplant lungs from White Americans. Here, we will use the same method to establish new hAEC lines from de-identified human remnant transplant lungs from Black American subjects. hAECs grow well in 2-dimensional (2D) cultures as epithelial monolayers are capable of forming alveolar-like organoids in 3-dimensional (3D) cultures with fibroblasts and Matrigel. We will introduce the KRAS driver mutation alone or in combination with other oncogenes into the hAECs using CRISPR/Cas9 editing. The cell lines will be used to test KRAS-targeting therapeutics, including polyisoprenylated cysteinyl amide inhibitors (PCAIs). These agents have been shown to disrupt RAS-mediated signaling in ethnically/racially diverse cell lines. Our approach yields new cell lines that provide a valuable system to study LUAD in Black Americans. Introduction: Non-union of intertrochanteric and supracondylar fractures of femur are uncommon which results in pain and functional disability, presenting major surgical challenge. We wish to report on two cases regarding endoprosthesis as management for non-union. Case Presentation: Patient A is 54 years old lady sustained closed comminuted intertochanteric fracture of right femur (31A2.3). She underwent open reduction, intramedullary nail with cerclage wire on day 3 post trauma. At 6 months, there is sign of avascular necrosis of femur head with non-union fracture site. Patient still complaining of pain, on non-weight bearing ambulation. At 1 year, patient underwent right proximal femur replacement. At 3 months post replacement, she is able to full weight bear with walking cane without pain. Patient B is 56 years old gentleman sustained Grade IIIa open comminuted fracture of right supracondylar with intercondylar split (33C2.2). He underwent emergency wound debridement, screw fixation with high tibial pin insertion. At 4 months, he underwent distal locking plate of right femur with synthetic bone grafting. At 8 months post fixation, there is non-union comminuted fracture of supracondylar, still unable to weight bear. At 1.5 year post trauma, patient underwent right distal femur replacement. At 3 weeks post replacement, he is able to full weight bear without aid. Discussion: Most failures of treatment occur in unstable fracture patterns of hip. (1) Endoprosthesis may provide as alternative treatment if internal fixation are recognized to be suboptimal, amount and quality of remaining distal bone stock and level of the non-union. (2,3) In both our cases, patient experienced marked improvement in functional capacity and pain free, which are the hallmarks of a successful salvage procedure. (4) Conclusion: Endoprosthesis can be consider as alternative treatment for non-union intertrochanteric and supracondylar fracture of femur, hence providing stable joint, pain free and early return to daily activities. Polyisoprenylated cysteinyl amide inhibitors (PCAIs) constitute a group of cancer therapy agents designed to specifically disrupt and suppress hyperactive G protein signaling, such as that triggered by RAS mutations. Here we determine the effects of PCAIs on the viability, G-proteins levels, downstream mediators, and apoptosis-related proteins on the KRAS-mutant, Black American-derived lung adenocarcinoma cell line, NCI-H23. Of the 17 PCAIs tested, compounds NSL-YHJ-2-27 and NSL-YHJ-2-46 showed the most potency with EC50 values of 2.7 and 3.3 µM, respectively. To determine the effect of the PCAIs on the levels of Mitogen-Activated Pathway Kinase (MAPK)-related enzymes, western blot analysis was performed. After 48 hours exposure to 5 μM of the compounds, 57 to 150% increases of BRAF, ERK1/2, MEK1/2, and p90RSK phosphorylation along with a 60 to 78% decrease of pCRAF were observed, indicating significant disruptions in RAS signaling. Furthermore, 5 μM of NSL-YHJ-2-27 depleted the singly polyisoprenylated monomeric G-proteins RAC 1/2/3 and CDC42 by 77 and 76%, respectively. The depletion of these key cytoskeletal proteins may account for the observed 52 and 42% reduction in cell migration after 24 h exposure to NSL-YHJ-2-27 and NSL-YHJ-2-46, respectively. Western blot analysis was used to determine the effect PCAIs on caspase activation necessary for the initiation of apoptosis. After treatment with 5 μM of NSL-YHJ-2-27, full-length inactive caspase 3 and 7 levels dropped by 72 and 60%, while cleaved active caspase 3 and 7 levels increased by 274 and 130%, respectively. Treatment with 5 μM of NSL-YHJ-2-46 depleted full-length caspase 7 by 53% and increased cleaved active caspase 3 levels by 83%. These findings clearly show the direct effects of the PCAIs on the RAS signaling pathway, perhaps through the activation of proapoptotic isoforms of p90RSK. Abstract Lung cancer is a source of cancer health disparity among black Americans, especially men. Lung cancer incidence rates and mortality rates are higher in black Americans than in white Americans despite lower tobacco exposure among black men than white men. Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer in all ethnic/racial groups, including blacks. Genetic driver mutations differ among racial/ethnic groups. In addition, the same mutation could have different effects based on the genetic background arising from different racial/ethnic groups. Therefore, it is important to study appropriate driver mutations in the correct genetic background. Protooncogenes from the RAS family, KRAS, NRAS, and HRAS, play a crucial role in cell growth regulation, cell proliferation, and survival. KRAS is the most frequently mutated driver gene in black LUAD (~14%). NRAS mutations are also seen (1-2%). It is therefore important to study LUAD cell lines carrying different RAS mutations in black Americans’ genetic backgrounds. Unfortunately, to our knowledge, there is only one LUAD cell line from a black subject. The NCI-H23 cell line contains a heterozygous KRAS mutation at codon 12. We are generating a collection of isogenic cell lines derived from NCI-H23, tailored to study RAS gene activation in black Americans. We will correct the KRAS mutation to wildtype through CRISPR/Cas9 deletion with co-transfection of the wild-type sequence. However, we expect that the NCI-H23 cells may not tolerate the removal of the KRAS without simultaneous replacement with another driver mutation. Therefore, we will replace the KRAS mutation with either mutated NRAS, HRAS, or EGFR. EGFR functions as a receptor tyrosine kinase in the RAS-RAF-MEK-ERK pathway to control the activation of RAS oncogenes and is also frequently mutated in black LUAD (~8%). We have identified a heterozygous single nucleotide polymorphism, rs11047919, 839 nucleotides downstream from the KRAS mutation which we are using to differentiate between the wild-type and mutated KRAS alleles following CRISPR editing. We will use the new cell lines to test the efficacy of polyisoprenylated cysteinyl amide inhibitors (PCAIs). These agents have been shown to disrupt RAS-mediated signaling in ethnically/racially diverse cell lines. In the future, we will develop immortalized alveolar epithelial cell lines from black subjects to further address the lack of lung cancer models representing black Americans. Abstract Objective: Over 75% of patients with ovarian cancer present with widely metastatic disease. We have previously published that AXL is highly expressed in primary ovarian cancer tumors. However, it is unknown whether AXL expression in metastatic tumors correlates with outcome. Additionally, the role of stromally expressed AXL in metastasis has not been fully evaluated. Thus, the objective of this study is to evaluate the role of tumor and stromal AXL in the key steps of tumor metastasis. Methods: Immunohistochemistry was used to measure AXL expression in a tissue microarray with primary and metastatic high grade ovarian tumors. Attachment assays as well as transwell and spheroid invasion assays were performed on established ovarian cancer cell lines. Mesothelial cell clearance assays utilizing human omentum-cultured mesothelial cells (HPMC) and additional invasion assays utilizing normal omental fibroblasts (NOF) were performed. Results: 214 tumor samples were included from 139 patients: 122 primary and 92 metastatic sites, contributing to 85 matched samples. Most had high-grade serous histology (79%) and stage III-IV disease (79%). High AXL expression in metastatic tumors was associated with decreased overall survival (HRadj 1.8, 95% CI 1.03-3.1), but expression in primary tumors was not (1.1, 0.61-2.0). High vs. low AXL expression in metastases was associated with a decreased platinum-free interval (5.92 vs 15.9 months, P=0.03) and decreased progression-free survival (11.6 vs 18.8 months, P=0.02).Tumor AXL expression correlated with stromal AXL expression (R2=0.50, P<0.001). Human ovarian tumor cells depleted of AXL had decreased attachment (OVCAR3-TPMES, 0.55-fold decrease P<0.001) and invasion in transwell (OVCAR5, 173 vs 262 invasive tumor cells/hpf, P<0.01) and spheroid (ES2, 87% vs 31% area invaded, P<0.0001) invasion assays. Human ovarian cancer cells plated on AXL-deficient HPMCs had less cell clearance than those plated on AXL-expressing HPMCs (ES2, 6.0 vs 3.9 normalized area cleared, P<0.001). Further, human ovarian cancer cells plated on AXL-deficient NOFs had less invasion than tumor cells plated on AXL expressing NOFs (OVCAR5, 173 vs 262 invasive tumor cells/hpf, P<0.01). Conclusions: High AXL expression in metastatic ovarian cancer cells correlates with worse survival and worse response to chemotherapy. Further, AXL expression in both tumor and stromal cells contributes to the steps of ovarian cancer metastasis. The Matthews papers, which are brilliantly chosen, represent only one dimension of his oeuvre; he was in fact a recognized scholar of ancient and medieval philosophy, particularly Plato, Aristotle and Augustine. The present selection draws from his persistent, oddly related inquiry, pursued over the course of almost 40 years, into the theory and practice of conducting philosophical conversations with children, whether inside or outside the classroom.

Abstract Background: Residential segregation has been associated with cancer incidence and mortality. Hispanic/Latinos (HL) experience moderate to high residential segregation. Purpose: This study investigates levels of racial and ethnic residential segregation and racialized economic concentrations at the extremes in the Hispanic Community Health Study/Study of Latinos (HCHS/SOL). Methods: We used baseline data from 16,415 HL adults enrolled in the Hispanic Community Health Study/Study of Latinos between 2008-2011 from the Bronx, NY; Chicago, IL; Miami, FL; and San Diego, CA. Segregation measures were calculated from census tract-level (2006-2010 American Community Survey and 2010 decennial census). We measured residential segregation using the % HL Gini coefficient, to capture variability of HL residents within the census tract, and the isolation index, to capture the probability that HL residents come into contact with other members of the same minority group. We measured racialized economic (race/ethnicity + income) concentration using the Index of Concentration at the Extremes (ICE), to capture spatial social polarization at the extremes. We compared means of HCHS/SOL population characteristics using linear regression and adjusted Wald tests for continuous, binary, and categorical variables, respectively, calculated from weighted complex samples analyses. Results: On average, overall segregation was moderate to high (M±SE): Gini (0.39 ± 0.00); Isolation (0.76 ± 0.01); ICE (race: -0.64 ± 0.01; income: -0.29 ± 0.01; race + income: -0.26 ± 0.01). HL adults who were older (> 65 y: 0.80 ± 0.01; 45-65 y: 0.77 ± 0.008; 18-44 y: 0.75 ± 0.75, p = .000) and foreign/territory-born residing in US <10 years (0.81 ± 0.009; p = 0.000; foreign/territory-born residing in US >=10 years: 0.76 ± 0.01; vs. US born: 0.70 ± 0.01) and preferred Spanish (0.79 ± 0.01; p = .000 vs. English: 0.69 ± 0.007) experienced higher racial/ethnic segregation as measured by the Isolation index. HL of Cuban (0.42 ± 0.006, p = 0.049) vs. all other heritage experienced the as measured by the % HL Gini index. We found higher levels of racialized economic segregation among foreign/territory-born residing in US <10 years (-0.32 ± 0.01) vs. foreign/territory-born residing in US >= 10 years (-0.26 ± .01) and US born (-0.21 ± 0.01; p = 0.000); individuals that preferred Spanish (-0.28 ± 0.01) vs. English (-0.21 ± .01; p = 0.000); and self-reported Cuban heritage (-0.42 ± 0.01; p = .000) vs all other heritage. Conclusion: Using multiple, measures of segregation, we found that HL adults who were older, foreign born, and preferred Spanish experienced moderate and high levels of segregation. It is important for future work to examine the impact of racial/ethnic and economic segregation on social determinants of cancer disparities within segregated environments among diverse HL. Abstract The Poly (ADP-ribose) polymerase (PARP) family has numerous essential functions in cellular processes such as transcription, chromatin remodelling, DNA damage response and repair as well as apoptosis. PARP inhibition blocks base excision repair and results in conversion of SSBs to DNA double-strand break (DSBs). DSBs are the most deleterious form of DNA damage that can be generated by exogenous DNA damaging agents or endogenous replication stress. DSBs can be repaired by homologous recombination repair (HRR) or non-homologous end joining (NHEJ). The physiological importance of HRR is underscored by the observation of genomic instability in HRR-deficient (HRD+) cells and, importantly, the association of cancer predisposition and developmental defects with mutations in HRR genes. PARP1 and PARP2 are required for SSB repair, while PARP1 is also involved in the repair of DNA double-strand breaks (DSBs) and replication fork damage. Recent reports suggest the PARP1 inhibition is sufficient to elicit an anti-proliferative effect and that PARP2 is essential for the survival of hematopoietic stem and progenitor cells in animal models.AZD9574 is a novel brain penetrant PARP1 inhibitor that acts by selectively inhibiting and trapping PARP1 at the sites of SSBs. AZD9574 exhibited >8000-fold selectivity for PARP1 compared to PARP2 and other members of the PARP family (PARP2, PARP3, PARP5a and PARP6) in biochemical assays. While AZD9574 inhibited PARP1 enzymatic activity in all tested cell lines irrespective of the HRR status (IC50 range between 0.3 - 2 nM), colony formation assay in isogenic cell lines pairs confirmed higher potency and selectivity towards HRD+ models (DLD1 BRCA2-/-; SKOV-3 BRCA2-/- and SKOV-3 PALB2-/-). For example, AZD9574 IC50 in the BRCA2-/- DLD1 cells was 1.38 nM compared to IC50 > 40 µM BRCA2wt cells, which corresponds to a ~20,000-fold greater efficacy in the BRCA2-/-cells (ratio of AZD9574 IC50 in BRCA2wt divided by BRCA2-/-) compared to the wild type parental line. In vivo, AZD9574 demonstrated dose-dependent efficacy in a BRCA1 mutant MDA-MB-436 subcutaneous xenograft model. Anti-tumour effects of AZD9574 were manifested by pronounced growth regressions that were durable after treatment withdrawal. An intracranial xenograft model of breast cancer brain metastases was developed to assess the efficacy of AZD9574 in the context of blood-brain barrier penetrance. Treatment of animals with established intracranial lesions using a dose of 3 mg/kg AZD9574 showed sustained tumour growth suppression resulting in a significantly extended survival of tumour-bearing mice. Collectively, we believe that our data support the development of AZD9574 as a potential therapy for patients with HRD+ breast cancer whose disease has spread to the brain. Abstract Background: Childhood cancer survivors are a growing population at elevated risk for premature aging and age-related chronic health conditions (CHCs) compared to the general population. In the general population, diet affects many hallmarks of aging, including inflammation, metabolic dysfunctions, and molecular and epigenetic changes. We examined associations between dietary patterns and risk of premature aging in adult survivors of childhood cancer. Methods: Adult survivors (18-65 years old, mean [SD] age 31 [8.4] years) of childhood cancer enrolled in SJLIFE between 2007 and 2017 completed a 110-item food frequency questionnaire at enrollment (n=2,904). Sociodemographic, cancer, cancer treatments, and health history were abstracted from medical records; CHCs were clinically validated. Factor analysis was performed to identify dietary patterns specific to the study population. Premature aging was assessed using the Deficit Accumulation Index (DAI) based on 45 age-related CHCs and categorized into low (<0.2), medium (0.2-0.34), and high (>0.35) aging risk groups. Odds ratios (OR) and 95% confidence intervals (CI) were estimated using multinomial logistic regressions adjusting for potential confounders, such as sociodemographics, health behaviors, and cancer treatments. Individuals’ factor scores of each pattern were grouped into quintiles, and a median score of each quintile was entered as a continuous term in regression models. Results: 20% of survivors were at medium- and 8% were at high-risk for premature aging. Survivors at high-risk for premature aging were more likely to be female, smokers, have low socioeconomic status, and have received radiation therapy to head and neck, chest, spine, or abdomen compared to those at low premature aging risk. Three dietary patterns were identified: 1) plant-based, 2) fast-food, and 3) Western contemporary. A plant-based diet was characterized by greater intakes of vegetables, fruits, whole grains, and fish. A higher plant-based diet pattern score was associated with a lower risk of premature aging (OR=0.76, 95% CI: 0.62-0.94 for the high-risk group; OR=0.91, 95% CI: 0.79-1.04 for the medium-risk group). A fast-food diet was characterized by greater intakes of sweets, processed meat, refined grains, potato, high-fat dairy, and soda. A fast-food diet was related to a non-significant increased risk of premature aging (OR=1.20, 95% CI: 0.91-1.57 for the high-risk group; OR=1.18, 95% CI: 0.98-1.43 for the medium-risk group). A Western contemporary diet characterized by greater intakes of meat, pasta/rice, pizza, Mexican food, and fruit juice was associated with an increased risk of premature aging (OR=1.36, 95% CI: 1.05-1.78 for the high-risk group; OR=1.04, 95% CI: 0.87-1.26 for the medium-risk group). Conclusion: Our findings suggest that diet plays a role in aging, and a diet rich in plant-based foods may mitigate the risk of premature aging in childhood cancer survivors.
