ABSTRACTMatthias Alexander Castrén (1813–1852), a great Finnish researcher and fieldworker, the first professor of the Finnish language, undertook a vast range of studies, geographically from Norwegian Lapland to Siberia and in subjects from linguistics to ethnology. His extensive work in the Russian north made him one of the principal figures of Siberian and Finno-Ugrian studies. Castrén's pioneering contributions in Turkology, Mongolian studies and archaeology are also noteworthy. He spent almost ten years on expeditions outside Finland and during his short life, he died at the age of 39, he managed to collect a vast amount of material, which is set out in 32 volumes of his manuscripts. However, his name is less known than are the names of some of his prominent friends. In English there are only few comments on his travels and work and these are in the context of the history of Finnish science in the 19th century. Published materials of Castrén's work include two volumes in Finnish translated from Swedish, six volumes in Swedish, and twelve volumes in German. Castrén was born in a period which is often regarded as a turning point in Finnish history and as a key period in the formation of the Finnish nation. Presenting an overview of different fields of Castrén's research, this article analyses his scientific contribution along two lines: in the context of the rising national awareness in Finland in the 19th century and of the mainstream developments in the scientific scene of this time. Based on Castrén's travel diaries, correspondence and lectures, the article seeks to contribute to understanding the historical aspects that shaped this great researcher situated as he was between three national traditions of his time: his writing was mostly in Swedish, his research activities were mainly carried out on behalf of the Russian Academy of Sciences, and yet he is most prized as a pioneer of Finnish linguistics, ethnography, archaeology and other disciplines.

Traz a Técnica de Alexander como uma ferramenta para o autoconhecimento e para a liberdade muscular que é um dos elementos do Método de Stanislavski. Concluímos com um relato pessoal de mudança perceptiva na influência do olhar sobre a emissão vocal e atuação.

The Alexander Technique is a method that emerged as a result of Actor Frederick Matthias Alexander’s work on himself to solve the problems he had with his voice during his performance. This technique, which was effective for himself, rapidly became widespread because it was tried by different people over time and positive results were obtained. Today, it is effective not only for music but also for people from different disciplines to use their bodies more comfortably and thus have a natural posture. In this sense, in the study, ten different instructors working in various locations were selected as the study group to reach the findings related to the use of the Alexander Technique and the results of the technique in flute lessons given in the music departments of higher education institutions in Turkey. Framed by the researcher, interview questions with expert opinions were asked, and the data obtained were tabulated and presented with the content analysis method, one of the qualitative research methods.

In the article, the literary works of Baltic German writer Carl Hugenberger have been explored. Anthology of his poetry translations, “Derrigs laika kaweklis” (Useful Pastime, I–II, 1826–1827), has been analysed. The anthology was significant in the emancipation of Latvian literary culture and liberation from moral didacticism as well as the development of the self-sufficient aesthetic value of literature. Thus, the anthology prepared the way for the formation of Latvian national literature in the mid-19th century. Special attention has been turned towards Hugenberger’s translations of poems by Johann Wolfgang Goethe and Friedrich Schiller. Besides, the evaluation and reception of Hugenberger’s works have been explored. The article concludes that despite the innovative role and poetic achievements of Hugenberger’s poetry, it did not gain popularity among wider circles of the Latvian reading public and met criticism regarding the shortcomings in the translation techniques that can be explained by the limits of the underdeveloped Latvian language at the time. The most important episodes in Hugenberger’s biography have been outlined as well as his religious hymns and works of popular enlightenment, including translations of “Schillings-Bücher des Rauhen Hauses”, a book series of German Inner mission, works by Jeremias Gotthelf, August Kotzebue, Gottfried August Bürger et al. Special attention has been paid to previously unidentified originals of Hugenberger’s translations – works by Matthias Claudius, Johann Hinrich Wichern, Heinrich Alexander Seidel, and Adolph Krüger as well as previously underexamined partial translation of Johann Peter Hebel’s “Allemanische Gedichte”. The literary works of Hugenberger have been interpreted within the context of the literary praxis of the late popular enlightenment in the Baltics.

Abstract Acute myeloid leukemia is a grievous illness. BiTE® (bispecific T cell engager) molecules redirect T cells by engaging CD3 and a tumor-associated antigen (TAA). These molecules have shown clinical efficacy but one mechanism of resistance is loss of a single TAA. We hypothesized that a BiTE® molecule targeting >1 TAA could reduce relapse frequency. FLT3 and CD123, were selected for a dual-targeting BiTE® (dBiTE࣪) molecule.A half-life extended (HLE) CD123-FLT3 dBiTE࣪ molecule was evaluated in vitro, in mouse xenografts, and in non-human primate (NHP) tolerability studies. The molecule had nM affinity for human and NHP FLT3, CD123, and CD3, and pM efficacy in cytotoxicity assays using human T cells or NHP PBMCs. The molecule achieved 100% killing against single-positive (sp) cells (CRISPR-generated isogenic cell lines) at potencies like those of double-positive (dp) cells (7.4 ±. 4.1 pM FLT3sp, 7.1 ±. 3.8 pM CD123sp and 3.4 ±. 1.5 pM dp, n=3). In a mouse xenograft model, the molecule induced significant activity and extended survival > 3 weeks (1.0, 0.1, and 0.01 mg/kg (p ≤ 0.001)). In mice with sp tumors, survival benefit could not be calculated due to high survival, demonstrating that both arms of the molecule are active. In NHP, the molecule had a half-life of 52 hours (0.3 or 3 µg/kg). FLT3 mRNA levels, a marker of FLT3-expressing cells, decreased in blood following dosing. Repeat dosing was not tolerated, and cytokine release was observed. Some cytokines were reduced while others increased. CD123 is reportedly expressed on endothelial cells (ECs), with increased expression in inflammatory conditions. An immunohistochemical survey found that CD123 is expressed on human and NHP monocyte/macrophages and ECs with limited distribution in lymphoid tissues and lamina propria of the gut. We hypothesized that cytokine release following administration may result in increased CD123 expression and in turn, further increased cytokine levels with repeat dosing. CD123 was detected on primary human umbilical vein ECs (HUVECs). In co-cultures of HUVECs and T cells, a CD123 BiTE® molecule induced expression of CD123 on HUVEC cells at concentrations that elicited redirected lysis (12.5 pM), T cell activation, and cytokine secretion. To better understand BiTE®-induced upregulation of CD123 on ECs, HUVECs were cultured with supernatants (SN) from a TDCC assay or recombinant IL-3, IL-6, TNFα, or IFNγ. Assay SN and TNFα induced >2-fold CD123 expression on HUVECs, but not on CD123-negative primary human pulmonary microvascular ECs. These data demonstrate that CD123 expression on ECs was increased upon exposure to a CD123 mono-targeting BiTE® molecule, potentially through BiTE®-induced secretion of TNFα. Additional studies are ongoing.In sum, a CD123-FLT3 HLE dBiTE࣪ molecule was active against both dp- and sp-positive target cells in vitro and in vivo. Careful selection of TAA for dBiTE࣪ molecules is necessary to increase efficacy and maintain safety. Suomen kansalliseepos Kalevala ilmestyi vuosina 1835 (Vanha Kalevala) ja 1849 (Uusi Kalevala). Se on yksi harvoja suomalaisen kirjallisuuden edustajia, joiden voidaan katsoa kuuluvan maailmankirjallisuuteen. Kalevala on Elias Lönnrotin (1802–1884) luomus, ja se on käännetty tähän päivään mennessä yli 60 kielelle. Kalevalan käännökset koostuvat runomuotoisten käännösten lisäksi erilaisista proosakäännöksistä, lyhennetyistä laitoksista sekä kokonaan tai osittain mukautetuista käännöksistä. Tämä artikkeli käsittelee Kalevalan ruotsinkielistä käännöshistoriaa, joka on jäänyt aiemmassa tutkimuksessa vähälle huomiolle. Tutkimustehtävämme on kahtalainen: laadimme kansalliseepoksen ruotsinkielisen käännöshistorian 1800-luvulta 2000-luvulle ja tarkastelemme käännösten sisältämiä peritekstejä ensisijaisesti kääntäjän näkökulmasta. Ajallinen kaari on näin ollen laaja, ja analyysissamme käännöshistoriallinen katsaus edeltää peritekstianalyysia. Kirjallisuus- ja käännöstieteessä peritekstillä tarkoitetaan tekstiä välittömästi ympäröivää tekstimateriaalia (Hosiaisluoma 2003). Selvitämme, mitä peritekstejä Kalevalan ruotsinnoshistoriasta löytyy ja mitä niiden avulla on mahdollista sanoa kääntäjien tekemistä valinnoista. Keskiöön nousevat muun muassa kysymykset kääntäjien strategiasta, intentiosta ja kohderyhmästä. Analysoimme peritekstejä kvalitatiivisen lähiluvun menetelmin ja saamme näin lisätietoa siitä, miten Kalevalaa on käännetty ruotsiksi eri aikakausina. Aineistomme koostuu sekä kirjamuodossa julkaistujen Kalevala-ruotsinnosten ensimmäisistä painoksista (12 kpl) että sanoma- ja aikakauslehdissä ilmestyneistä yksittäisistä runokäännöksistä (23 kpl) 1800-luvulta 2000-luvulle (ks. Laukkanen – Lönnroth 2018, jossa dokumentoimme myös joitakin Kalevalan julkaisemattomia ruotsinnoksia). Tutkimuksemme uutuusarvo on ennen kaikkea vanhemman sanoma- ja aikakauslehdistön hyödyntäminen Kalevalan käännöshistorian ja peritekstien näkökulmasta. Kalevalan kääntäjiä ja käännöksiä tutkineen E. N. Setälän (1909, 166) mukaan oli ”luonnollista, että ruotsi oli se kieli, jolle Kalevalaa ensinnä yritettiin kääntää, ja luonnollista myös, että kääntäjät olivat kaikki suomalaisia”. Käännöshistoriallista tutkimusta Kalevalan ruotsinnoksista ei Setälän toteamuksesta huolimatta kuitenkaan ilmaantunut. Kalevalan yleisen käännöshistorian kannalta keskeisiä julkaisuja ovat Setälän lisäksi Rauni Purasen The Kalevala abroad. Translations and Foreign-language Adaptations of the Kalevala (1985), Pertti Anttosen ja Matti Kuusen Kalevala-lipas (uusi laitos 1999) ja Petja Aarnipuun toimittama Kalevala maailmalla. Kalevalan käännösten kulttuurihistoria (2012). Artikkelimme perustuu siihen tutkimukseen, jonka teimme kootessamme Kalevalan ruotsinkielisten käännösten bibliografiaa vuosilta 1835–2018 (Laukkanen – Lönnroth 2018). Viimeisin Kalevalan käännöshistoriaa käsittelevä tutkimus on Kaisa Häkkisen artikkeli Matthias Alexander Castrénin Kalevalan ruotsinnoksesta (2019). Näkökulmamme on kulttuurihistoriallisesti kontekstualisoiva, mikä korostaa käännösten historian, kirjallisuushistorian ja kulttuurihistorian välistä vuorovaikutussuhdetta (esim. Riikonen 2007, 2010). Emme vertaile käännettyjä tekstejä keskenään, vaan nostamme esiin tekstejä, jotka osaltaan tuovat lisävalaistusta Kalevalan ruotsinkieliseen käännöshistoriaan. Esittelemme käännösten yhteydessä kääntäjiä ja kustantajia sekä analysoimme erityisesti kääntäjien esipuheita ja muita tekstejä, joissa he valottavat käännöstensä funktioita ja kohderyhmiä. Läheskään kaikissa käännöksissä ei kuitenkaan ole varsinaista esipuhetta, mutta niissä on usein esimerkiksi takakansiteksti, joka avaa käännöksen tarkoitusta lähinnä kustantamon näkökulmasta. Suojapaperit sen sijaan melkein poikkeuksetta kadonneet. Peritekstit kertovat kääntäjän näkemyksestä omaan käännöstyöhönsä sekä nivovat käännöksen osaksi omaa aikaansa ja kulttuurista kontekstiaan.

Abstract Introduction: Hepatocellular Carcinoma (HCC) is considered a prototype of inflammation-derived cancer arising from chronic liver injury. The cellular composition of the HCC tumor immune microenvironment (TiME) has a major impact on cancer biology as the TiME can influence tumor initiation, progress, and response to therapy. Mucosal-associated invariant T (MAIT) cells can represent the most abundant T cell subtype in the human liver and have been found to be impaired in both number and function in liver cancer. These innate-like T cells are assigned crucial roles in regulating immunity and inflammation in the context of infection, albeit their role in HCC remains elusive. Methods: High-dimensional flow cytometry was used to analyze MAIT cell phenotypic changes in murine and human liver cancer. Highly multiplexed immunofluorescence microscopy was used to quantify immune cell infiltration in primary human HCC samples. We developed and validated a comprehensive 37-plex antibody panel for immunofluorescence imaging of human fresh frozen HCC samples. We applied co-detection by indexing (CODEX) technology to simultaneously profile in situ expression of 37 proteins at sub-cellular resolution in 15 HCC patient samples using whole slide scanning. Initial image analysis was performed using HALO quantitative image analysis software. Finally, we established an image analysis pipeline to quantify the MAIT cell interaction network at the HCC invasive front. Results: Profiling of human and murine HCC using flow cytometry and highly multiplexed CODEX imaging revealed substantial dysregulation/aberrant activation of MAITs in liver cancer. In situ phenotyping of 4,500,000 single cells (including 1,500,000 CD45+ immune cells) allowed for the quantification of 20 distinct immune cell phenotype clusters, differential analysis of activation markers and spatial features of each individual cell. CODEX imaging revealed detailed composition of the MAIT cell niche in human liver cancer tissue allowing for further distinct spatial analysis including infiltration and nearest-neighbor analysis. Importantly, flow cytometry data of paired samples correlated well with image-based immune phenotyping. Beyond that, whole slide imaging revealed spatial relationships and interactions within the MAIT cell hub localized in distinct tissue regions. Conclusion: Here, we demonstrate that spatially resolved, single-cell analysis of human liver cancer tissue allows for in-depth characterization of interacting immune cellular programs underlying MAIT cell dysfunction in HCC. Background:The differentiation between rheumatoid arthritis (RA) and psoriatic arthritis (PsA) is sometimes a challenge for rheumatologists in daily clinical practice. Imaging techniques such as MRI could be a helpful tool for this purpose.Objectives:To examine the value of 3 Tesla (T) magnetic resonance imaging (MRI) with a high-resolution 16-channel hand coil for the differentiation between RA and PsA.Methods:A total of 17 patients with active PsA and 27 patients with active RA were evaluated by 3T MRI. Images were analyzed by three readers according to the outcome measures for RA clinical trials (OMERACT) and RA and PsA MRI scores for the presence and intensity of the following MRI features: synovitis, flexor tenosynovitis, bone edema, bone erosion, periarticular inflammation, bone proliferation, and joint space narrowing. A receiver operating characteristics (ROC) curve was established for a calculated prediction model comprising age, gender, and the imaging features ‘periarticular inflammation’ and ‘erosion’ of the metacarpophalangeal (MCP) joint of the 5th finger.Results:PsA could be differentiated from RA by extracapsular inflammatory changes (PsAMRIS sub-score ‘periarticular inflammation’), with a minimal odds ratio (OR) for the outcome ‘not RA’ of 0.06 (p< 0.01) at all MCP joints. The author compares five English translations of Alexander Pushkin's A Journey to Arzrum that we know now. These are works made by Birgitta Ingemanson (1974), Ronald Wilks (1998), David and Ludmila Matthews (2003), Nicholas Pasternak Slater (2013), and Richard Pevear and Larissa Volokhonsky (2016). Four of these translations (the latter ones) have not been included into the latest bibliography of English translations and studies of Pushkin's works (1999). The growing number of translations may indicate an increasing interest to A Journey to Arzrum. This study aims to detect the translators' strategies (the level of domestication or foreignization) and their approach to Pushkin's work; to mark the mistakes and inaccuracies, stylistic features, lexical and syntactic peculiarities of translations and differences between them and between them and the original text. The study also shows how the translators' approach to A Journey to Arzrum changed over time. By pointing out all these we can conclude which type of audience suits each translation most and what the general tendency is. In the article the author gives basic information about these translations and their authors and analyze the examples according to the following sections: formal distinctions and main difference in the approaches; toponyms and ethnonyms; exoticisms and direct speech; stylistic peculiarities of the translations (where the author analyzes bigger, complicated, most stylistically expressive fragments of the text and variations in translating names and titles, along with noting syntactic peculiarities). The majority of inaccuracies the author has found come from David and Ludmila Matthews' translation. At the same time this work is the easiest to perceive for broad (especially non-academic) audience or readers that are not well familiar with Russian and Caucasian culture of that period (foreign-language quotes are translated as well as many exoti-cisms etc.). The other translators tend to save accurately the specificities of Pushkin's text and local peculiarities. The author states that, in general, earlier translators chose domestication as a strategy while the later ones preferred foreignization. The highest degree of foreignization in addition to tendency towards maximal figurativeness of the text is observed in Richard Pevear and Larissa Volokhonsky's version. The latter characteristic sometimes does not clearly match the lucidity of Pushkin's style. Birgitta Ingemanson with rare exceptions shows quite a good balance at this point in her translation. It is also interesting to mention that Nicholas Pasternak Slater tries to correct Pushkin's own inaccuracies. The author concludes that all the translations are done properly. Ingemanson's, Wilks', Pasternak Slater's and Pevear and Volokhonsky's versions are distinguished by a very accurate and thorough approach. However, Matthews' translation is good and unique in terms of accomplishing a specific purpose - being understandable by common readers. This way it serves to the popularization of A Journey to Arzrum among the English-speaking audience.

BackgroundGiant cell arteritis (GCA) has the tendency to relapse once treatment is tapered or stopped. Such relapses represent a potential threat to GCA patients as they can lead to severe symptoms and organ damage.ObjectivesTo assess the frequency and type of relapses in patients with GCAMethodsThe Real-Life Treatment and Safety (REATS)-GCA cohort has been established by extracting the data on clinical presentation, inflammatory markers, imaging, comorbidities, treatments and serious adverse events of GCA patients from 6 specialized centres in Germany. We undertook descriptive and survival analyses (Kaplan-Meier), and compared baseline characteristics of participants with vs. without relapse. Ethical approval for the cohort was obtained.ResultsWe included 395 patients with a mean age of 71 years, including 264 (66.8 %) females and 129 (32.7%) males. Diagnosis of GCA was supported by temporal artery ultrasound in 37%, 18F-FDG-PET/CT in 29%, temporal artery biopsy in 14% of patients and by MRI or clinically in the remaining patients. 31% of patients presented with an isolated cranial manifestation and 18% with isolated extracranial manifestations. Most common presenting symptoms were headache (57%), fatigue (55%), weight loss (42%) and polymyalgia (38%) (Table 1). The most common comorbidities at the time of study inclusion were arterial hypertension (68%), followed by osteoporosis (26%). Within a median total follow-up duration of 22.2 (11.7-40.6) months, 97 of the 395 patients relapsed including 15 patients who relapsed more than once. The median (IQR) time to first relapse was 12.5 (7.1-21.8) months. Median relapse-free survival was 7.8 years with a relapse risk of 12% (CI, 9 to 15%) at 1 year and 38% (CI, 30 to 45%) at 5 years (Figure 1). Most common symptoms at relapse were headache (35%), polymyalgia (23%), fatigue (19%) and night sweats (12%) (Table 1). Three patients relapsed with sudden loss of vision. Among the 114 relapses observed, 94 (83%) occurred under prednisolone treatment with a median dose of 7.0 mg/day (IQR 4.0-12.5). 26 (23%) occurred under methotrexate and 14 (12%) under tocilizumab treatment. The leading symptoms at relapse are headache and fatigue, while loss of vision is rare (0.76%). В публикации представлены результаты морфолого-функционального анализа одного из видовкаменных украшений Мурзихинского II могильника – сланцевых подвесок. Исследовав форму, количество отверстий, тип сверления, а также расположение отверстий на подвеске нам удалось выделить 5 морфологических групп и составить типологию данного вида украшений. В ходе функционального анализа с применением трасологического метода нам удалось выявить несколько типов следов на подвесках, характеризующих весь жизненный цикл украшений – от изготовления до погребения. The production of hydrogen via acidic electrochemical water splitting is an important pillar towards more sustainability. The overall water electrolysis is hindered by the slow kinetics of the oxygen evolution reaction (OER). Therefore, the catalyst material for the anode still requires optimization. Hereby, we present two highly active OER catalysts, IrOx and Ir0.4Ru0.6Ox nanoparticles (NPs), prepared by a straightforward, surfactant-free, colloidal synthetic route.1 The metallic NPs are ~1.5 nm in diameter and were deposited on carbon Ketjen Black (CKB) as well as on antimony doped tin oxide (ATO) to investigate the active phase as well as potential influences of the support material. After the colloidal synthesis and supporting, the catalysts were activated, thus undergoing a particle growth of ~1 nm. Their performance was benchmarked in terms of activity and stability using our recently developed electrochemical flow cell called gas diffusion electrode (GDE) setup.2 This novel approach allows closing the experimental gap between the two most common methods, namely the rotating disk electrode (RDE) and the membrane electrode assembly (MEA). Independently from the support, Ir0.4Ru0.6Ox shows a higher activity towards OER than IrOx at three tested temperatures, i.e. 30, 40, 60 °C. The two different support materials were also compared, and it was found that independently from the material, NPs deposited on CKB showed slightly higher activity at low current densities (up to 40 mA mgIr -1). As expected, accelerated potentiostatic stability test reveals a rapid degradation of the IrOx/CKB, while IrOx/ATO and Ir0.4Ru0.6Ox/ATO exhibit similar stability than commercially available Ir black. BackgroundThe efficacy of Upadacitinib (UPA), a selective Janus kinase inhibitor, has been evaluated in the SELECT clinical program 1-6. In addition, recent results from the non-interventional UPwArds study further confirmed UPAs clinical effectiveness regarding standard disease activity scores for rheumatoid arthritis (RA) in a real-world setting 7. However, patient-reported outcomes (PROs) as another cornerstone of clinical decision making yet remain to be addressed in the context of a post-marketing setting. This interim analysis, conducted after 250 patients had completed the 6-month follow-up visit, aims to fill this gap.ObjectivesTo evaluate the change of selected PROs over 6 months in patients treated with UPA in a real-world data environment.MethodsUPwArds is a prospective, open-label, multicenter, non-interventional, post-marketing study including adult patients with moderate-to-severe RA (swollen joint count [SJC28] ≥ 3 and inadequate response or intolerance to at least one previous disease-modifying antirheumatic drug). According to the German label, patients were treated with UPA 15 mg once daily, as monotherapy or in combination with methotrexate. For this analysis, the following PROs were included: 0-10 numerical rating scales (NRS) for pain and fatigue, the Health Assessment Questionnaire Disability Index (HAQ-DI), the duration and severity of morning stiffness, the Patient Health Questionnaire 9 (PHQ-9), and the Rheumatoid Arthritis Impact of Disease Questionnaire (RAID). Changes from baseline were evaluated for follow-up periods of 1 month, 3 months, and 6 months. Results are presented for the total sample using descriptive measures reflecting sample size (N), average values (standard deviation) for each assessment and average change scores (standard deviation) for follow-up visits. All data were analyzed as observed, with no imputation of missing data.Results483 patients (369 female, 114 male) were included in the study, with available baseline PRO information for 481 patients. 6-months follow-up data were yet available from 279 patients The baseline average age and disease duration were 58.0 (12.3) years and 9.0 (8.0) years, respectively, whereas the mean initial DAS28-CRP was 4.6 (1.0). At baseline, 60.8% of enrolled patients had previously been treated with biologic or targeted synthetic disease-modifying antirheumatic drugs. Overall, PRO scores improved from baseline throughout month 6 with a considerable amelioration at month 3, which was maintained at month 6. Responses were rapid, with improvement already evident at month 1 (Table 1). Computer- und Videospiele sind heute ein selbstverständlicher Bestandteil der Lebenswelt vieler Kinder und Jugendlicher, aber auch von (jüngeren) Erwachsenen, die mit diesen neuen Medien aufgewachsen sind. Lange Zeit haben elektronische Bildschirmspiele allenfalls sporadische Beachtung gefunden. Weder in der Medienforschung oder Medienpädagogik noch in der breiteren Öffentlichkeit waren sie ein Gegenstand von breiterem Interesse.* In den letzten knapp zehn Jahren sind Video- und Computerspiele allerdings zunehmend in den Fokus der Aufmerksamkeit gerückt. Dabei sind in der massenmedial vermittelten Öffentlichkeit vor allem Amokläufe in Schulen in einen direkten Zusammenhang mit den Vorlieben (der Täter) für bestimmte Computerspiele gebracht worden. Die auch von prominenten Politikern aufgegriffene These lautete, dass gewalthaltige Spiele wie der First Person Shooter «Counterstrike» ein virtuelles Trainingsprogramm für das Töten und daher als wesentliche Ursache solcher Schulmassaker anzusehen seien. Auf der Basis dieser kausalen Wirkungsannahmen bzw. der unterstellten negativen Lern- und Trainingseffekte werden seither immer wieder Forderungen nach einem Verbot solcher «Killerspiele» oder gar nach der Verbannung aller Bildschirmmedien aus den Kinderzimmern abgeleitet. Neben solcher skandalisierter Thematisierung ist aber zunehmend auch eine nüchterne wissenschaftliche Auseinandersetzung zu konstatieren. So haben sich seit Beginn des neuen Jahrtausends die «digital game studies» als interdisziplinäres Forschungsfeld etabliert. Im Jahr 2000 wurde innerhalb der Gesellschaft für Medienwissenschaft z.B. die AG Games gebildet, die sich zu einem wichtigen deutschsprachigen Forum für die wissenschaftliche Beschäftigung mit Computerspielen entwickelt hat, und im Jahr 2002 entstand die internationale Digital Games Research Association (DiGRA), die im September 2009 ihre vierte grosse Konferenz nach 2003, 2005 und 2007 durchgeführt hat (vgl. www.digra.org). Seit 2001 gibt es mit der «Game Studies» eine primär kulturwissenschaftlich ausgerichtete Online-Zeitschrift (vgl. gamestudies.org), und daneben sind zahlreiche Publikationen zu verzeichnen, die zur Strukturierung und Systematisierung des Forschungsfeldes beigetragen haben, etwa die transdisziplinär angelegten Sammel- und Tagungsbände von Wolf & Perron (2003); Fritz & Fehr (2003), Copier & Raessens (2003), Neitzel, Bopp & Nohr (2004), Raessens & Goldstein (2005), Kaminski & Lorber (2006), Vorderer & Bryant (2006), de Castell & Jenson (2007), Kafai et al. (2008), Quandt, Wimmer & Wolling (2008). Ausserdem liegen Monografien vor, die sich um Orientierung sowie empirische oder theoretische Klärungen bemühen (etwa Fromme, Meder & Vollmer 2000, Newman 2004, Juul 2005, Klimmt 2005, Mäyrä 2008, Pearce & Artemesia 2009). Diese wissenschaftlichen Entwicklungen und Arbeiten zeigen, dass die Phase der blossen Skandalisierung oder akademischen Ignorierung der Computerspiele zu Ende geht. Stattdessen kann von einer zunehmenden Normalisierung und Ausdifferenzierung der akademischen Auseinandersetzung mit diesen neuen Medien und ihren Verwendungsweisen ausgegangen werden, wie sie bei anderen, etablierteren Gegenstandsbereichen (etwa der Film- oder Fernsehforschung) schon länger selbstverständlich ist. Zur Normalisierung und Differenzierung der Debatte soll auch dieses Themenheft der Online-Zeitschrift «MedienPädagogik» auf www.medienpaed.com beitragen, das sich mit den digitalen Spielen und Spielkulturen aus einer primär medienpädagogischen Perspektive befasst und nach den Chancen und Potentialen für informelle wie auch formelle Lern- und Bildungsprozesse fragt. Die Beiträge fokussieren in diesem Spannungsfeld von Spielen und Lernen, von Unterhaltung und Bildung unterschiedliche Aspekte. Die Mehrzahl greift dabei aktuelle Diskussionen über Einsatzmöglichkeiten digitaler Spiele im Bereich des Lernens und der Ausbildung auf, die unter dem Label «Serious Games» oder auch «Game-based Learning» geführt werden (Petko; Bopp; Berger/Marbach; Lampert/Schwinge/Tolks; Malo/Neudorf/Wist; Pfannstiel/Sänger/Schmidt). Daneben widmen sich Beiträge der Frage, wie die Lern- und Bildungsrelevanz der medial-kulturellen Praxen, die sich weitgehend unabhängig von pädagogischer Intervention entfalten, untersucht und verstanden, aber auch pädagogisch unterstützt werden können (Fromme/Jörissen/Unger; Schrammel/Mitgutsch). In einem Beitrag geht es schliesslich darum, Computerspiele selbst zum Gegenstand der pädagogischen Reflexion machen (Biermann). Diese Verteilung spiegelt das Gewicht der Schwerpunkte innerhalb des aktuellen Diskurses über den Zusammenhang von Computerspielen und Lernen/Bildung durchaus angemessen wider. Dominik Petko fokussiert auf formelle Lern- und Bildungskontexte und behandelt die Frage, welche didaktischen Potenziale Computerspiele für den gezielten Einsatz in Schule und Ausbildung aufweisen. Ausgangspunkt ist die Überlegung, dass es für den schulischen Bereich nicht ausreicht, die allgemeinen Lernpotenziale der Spiele auszuweisen und die Muster des spielimmanenten Lernens nachzuzeichnen, wie dies in einigen Publikationen der letzten Jahre geschehe. Um einen Schritt weiterzukommen und zu erreichen, dass digitale Spiele tatsächlich vermehrt in den Unterricht integriert werden, komme es darauf an, den Lehrpersonen zu zeigen, dass der Einsatz solcher Spiele einerseits mit einem vertretbaren Aufwand möglich ist und andererseits zu einem erkenn- und begründbaren Mehrwert führt. Dazu sei im ersten Schritt eine genauere Analyse und Typisierung der Spiele und anschliessend eine Konkretisierung der didaktischen Strategien und Arrangements für die sinnvolle Einbettung in den Unterricht erforderlich. Der Beitrag vermittelt einen ersten systematischen Überblick über entsprechende mediendidaktische Ziele und Ansätze für den Einsatz von Computerspielen in Schule und Unterricht. Matthias Bopp geht aus von der Überlegung, dass Computerspiele die Spielenden generell mit Aufgaben und Herausforderungen konfrontieren, die nur im Rahmen von spielbezogenen Lernprozessen bewältigt werden können. Zudem unterstützen aktuelle Spiele die Spielenden in der Regel systematisch beim Erwerb der erforderlichen Kenntnisse und Fähigkeiten, weisen also ein (zumindest implizites) didaktisches Design auf. Wenn man in Rechnung stellt, dass Video- und Computerspiele für Kinder, Jugendliche und zunehmend auch Erwachsene trotz – oder gerade wegen – der hohen Anforderungen, die sie stellen, höchst faszinierend und motivierend sind, dann bietet es sich an, die Lehr-Lern-Designs digitaler Spiele genauer zu untersuchen, um ihre erfolgreichen Prinzipien auf Lernspiele zu übertragen. Der Beitrag konzentriert sich in diesem Kontext auf die Frage, welche Bedeutung zum einen Rahmengeschichten (Storytelling) und zum anderen parasoziale Interaktionen zwischen Spielenden und virtuellen Spielfiguren für die Spielmotivation haben und welche Rolle ihre gezielte Verwendung beim Design von Lernspielen zur Steigerung oder Aufrechterhaltung der Lernmotivation spielen kann. Daraus werden Empfehlungen für die Gestaltung motivierender Lernspiele abgeleitet und abschliessend an Hand zweier Beispiele veranschaulicht. Johannes Fromme, Benjamin Jörissen und Alexander Unger plädieren dafür, die Bildungspotenziale von Computerspielen – und der neuen, computerbasierten Medien überhaupt – nicht nur in ihrer Verwendung bzw. Verwendbarkeit als didaktische und motivationssteigernde Hilfsmittel für die Vermittlung dieser oder jener Lehrinhalte zu sehen, sondern die Perspektive in verschiedenen Hinsichten zu erweitern. Eine prinzipielle Erweiterung bestehe darin, bei der Frage nach Bildungspotenzialen in der Tradition der humanistischen Bildungstheorie nicht primär den Wissenserwerb, sondern die Steigerung von Reflexivität im Selbst- und Weltverhältnis in den Blick zu nehmen und somit zu fragen, in welcher Weise der Umgang mit Medien hierzu beitragen kann. Und weil Bildung in dieser Tradition grundsätzlich als Selbstbildung verstanden werde, liege es nahe, neben der formellen, institutionalisierten Bildung dem Bereich der informellen Bildung eine entsprechende Beachtung zu schenken. Der Artikel fragt daher nach dem Beitrag, den Computerspiele – und vor allem community-basierte Praxen – für eine Flexibilisierung von Selbst- und Weltsichten sowie für den Aufbau von Orientierungswissen leisten können, und diskutiert abschliessend, inwiefern solche informellen Bildungspotenziale pädagogisch aufgegriffen und unterstützt werden können. Florian Berger und Alexander Marbach gehen davon aus, dass es angesichts der Popularität und hohen Motivationskraft der Computerspiele zwar nahe liege, ihre pädagogische Verwertbarkeit zu prüfen, dass für den pädagogischen Einsatz der digitalen Spiele aber bisher weder theoretisch fundierte Konzepte noch eine hinreichende Forschung existiere. Insbesondere würden Fragen der technischen Machbarkeit zu wenig beachtet, wobei die Schwierigkeit darin bestehe, dass der jeweilige «State of the Art» für Lernspiele als Massstab schon wegen der begrenzten (finanziellen) Ressourcen ausscheide, andererseits aber ein Mindeststandard erreicht werden müsse, um die notwendige Akzeptanz beim Anwender zu finden. Vor diesem Hintergrund geht der Beitrag zunächst der Frage nach, was die technische, die kulturelle und die pädagogische Qualität eines digitalen Spiels ausmacht, um dann – aus einer primär ingenieurwissenschaftlichen Perspektive – zu diskutieren, wie bei der Gestaltung von Lernspielen eine gute Balance erreicht werden kann. Ralf Biermann betrachtet Computerspiele in seinem Beitrag nicht aus mediendidaktischer, sondern aus medienpädagogischer Perspektive und stellt ein Konzept vor, mit dem die digitalen Spiele selbst zum Gegenstand einer lernorientierten Auseinandersetzung werden. Die leitende Idee ist es, Wege aufzuzeigen und zu erproben, wie Computerspiele in den Bereich der aktiven, projektorientierten Medienarbeit eingebunden werden können, die sich dieser neuen Medien – im Unterschied zu Radio, Presse oder Film – bisher kaum angenommen hat. Das Konzept des Video Game Essays knüpft an der Film- und Videoarbeit an, erweitert es aber um einige neue Elemente, die mit den technischen Besonderheiten der Spiele zu tun haben. Der Ansatz kann als innovative Form der Medienanalyse angesehen werden, bleibt aber bei der Analyse nicht stehen, sondern eröffnet auch weitergehende Handlungs- und Lernpotenziale. Als Einsatzgebiete des Video Game Essays werden die ausserschulische Medienarbeit, die Schul- und die Hochschulausbildung genauer betrachtet. Claudia Lampert, Christiane Schwinge und Daniel Tolks zeichnen in ihrem Beitrag die bisherigen Entwicklungen im Bereich der Serious Games nach, die von anderen Ansätzen des mediengestützten Lernens wie E-Learning, Edutainment und Game-Based Learning abgegrenzt werden, und arbeiten den aktuellen Diskussions- und Forschungsstand auf. Die Potenziale und Grenzen werden am Beispiel zweier Spiele aus dem Gesundheitsbereich (Games for Health) detaillierter diskutiert, für die auch erste empirische Befunde vorliegen. Serious Games gewinnen zwar – nach Ansicht der Autoren/innen vor allem aus Marketinggründen – zunehmend an Bedeutung, allerdings bestehe noch ein erheblicher Forschungs- und Evaluationsbedarf. Sabrina Schrammel und Konstantin Mitgutsch kritisieren, dass im medienpädagogischen Diskurs über Computerspiele der Umstand vernachlässigt werde, dass Spielen eine kulturell geprägte, aktive Auseinandersetzung mit einem Spielgegenstand sei. Ihnen geht es im vorliegenden Beitrag daher darum, die spezifische medial- kulturelle Praktik des Computerspielens zu erfassen bzw. dafür einen geeigneten methodischen und theoretischen Zugang zu entwickeln und vorzustellen. Das Spielen von Computerspielen wird in Anlehnung an den internationalen Diskurs als Transformation und Produktion kultureller Erfahrungen interpretiert, auch um aus den Engführungen der im deutschsprachigen Raum noch dominierenden Mediennutzungs- und Medienwirkungsforschung herauszugelangen. Für die pädagogische Auseinandersetzung wird daraus abgeleitet, dass nicht die didaktische Nützlichkeit, sondern die bildungstheoretische Bedeutung von Computerspielen zu fokussieren sei. Den bisher vorherrschenden teleologischen Lernkonzepten wird hier ein genealogischer Ansatz gegenübergestellt, bei dem die Erfahrungen und Lernprozesse im Zuge der Spielhandlungen selbst thematisiert werden. An einem Beispiel wird abschliessend verdeutlicht, wie das theoretisch-methodische Vorgehen einer hierauf ausgerichteten Analyse ausgestaltet und wie bei einer solchen Analyse die medial-kulturelle Praktik des Computerspielens pädagogisch rekonstruiert werden kann. Steffen Malo, Maik Neudorf und Thorben Wist ordnen ihren Beitrag in den Kontext des Game-based Training (GBT) ein und berichten über das Projekt Alphabit, bei dem es darum geht, computerbasierte Lern- bzw. Trainingsspiele als ergänzendes methodisches Mittel für Alphabetisierungs- bzw. Grundbildungsprogramme einzusetzen. Vorgestellt werden die im Projekt entwickelten konzeptionellen Überlegungen zu den Rahmenbedingungen, zur Auswahl der Inhalte, zu unterstützenden instruktionalen Hilfen, zu den Entwicklungsprozessen und zu methodischen Aspekten des spielerischen Lernens in virtuellen Umgebungen. Ausserdem werden erste Ansätze für die Umsetzung präsentiert und offene Forschungsfragen aufgezeigt. Auch Jochen Pfannstiel, Volker Sänger und Claudia Schmitz berichten über ein Projekt, das für die Bildungspraxis konzipiert wurde und auch bereits erprobt wird. Hier geht es um Game- based Learning im Hochschulbereich, genauer: um ein Lernspiel, das ergänzend zu einer Pflichtvorlesung in der Informatik eingesetzt wird, um Studierende dazu zu motivieren, sich während des Studiums intensiver und vertiefend mit der Vorlesungsthematik zu befassen. Ziel ist also ein verbessertes und vor allem nachhaltigeres Verständnis der Vorlesungsinhalte durch spielerische Mittel zu erreichen. Der Beitrag beschreibt das dazu entwickelte Lernspiel und berichtet über die bisherigen Erfahrungen und erste Evaluationsergebnisse. * Eine Ausnahme erscheint allerdings erwähnenswert: In der ersten Hälfte der 1980er Jahre gab es in der Bundesrepublik Deutschland eine Debatte (und einige Forschungsarbeiten) zum Videospiel in Spielhallen mit dem Ergebnis, dass 1985 ein geändertes Jugendschutzgesetz in Kraft trat, das den Zugang zu Glücks- und Videospielautomaten in der Öffentlichkeit neu regulierte und unter 18-Jährigen nicht mehr gestattete. Diese Regelung ist – anders als die 2003 obligatorisch gewordenen Altersfreigaben der USK für Computerspiele auf Datenträgern – unabhängig vom Inhalt der Spiele, und sie ist bis heute in Kraft. Abstract Introduction: Limited number of tumor types have been examined for Orthopedia Homeobox (OTP) expression. In pulmonary carcinoids, loss of expression is a strong indicator of poor prognosis. Here, we investigated OTP expression in 37 different tumor types, and the association between OTP expression and DNA methylation levels in lung neuroendocrine neoplasms. Methods: We analyzed publicly available multi-omics data (whole-exome-, whole-genome-, RNA sequencing, Epic-850K-methylation array) of 58 typical-, 27 atypical carcinoids, 69 large cell neuroendocrine carcinoma and 51 small cell lung cancer patients and TCGA (The Cancer Genome Atlas) data of 33 tumor types. 850K-methylation analysis was cross validated using targeted pyrosequencing on 35 carcinoids. Results: Results showed bimodality of OTP expression in carcinoids (OTPhigh versus OTPlow group, likelihood-ratio test p=1.5x10-2), with the OTPhigh group specific to pulmonary carcinoids while absent from all other cohorts analyzed. Significantly different DNA methylation levels were observed between OTPhigh and OTPlow carcinoids in 12/34 OTP infinium probes (fdr&lt;0.05 & β-value effect size&gt;0.2). Overall, OTPlow carcinoids harbor a high DNA methylation level as compared to OTPhigh carcinoids. OTPlow carcinoids showed a significantly worse overall survival (logrank test p=0.0052). Gene set enrichment analysis for somatically mutated genes associated with hallmarks of cancer showed robust enrichment of three hallmarks in the OTPlow group, i.e., sustaining proliferative signaling, evading growth suppressor, and genome instability and mutation. Conclusion: High OTP expression is a unique feature of pulmonary carcinoids with a favorable prognosis. A fear from Technological Advances mainly automation and artificial intelligence (AI) has been raised since the beginning of the 21st century, that machines will eliminate human beings performing certain tasks. Several models were developed to measure the effect of the Automation & AI on the employment and was applied on several countries. The first model was developed by Carl Benedikt Frey and Michael A. Osborne was called An Occupation-Based Approach, 2013. In which they define several tasks and call it “Engineering-Bottlenecks”. Katharina Dengler, Britta Matthes and Wiebke Paulus, 2014 made The Occupational Tasks model in which they use the assignment of activities explained by Alexandra Spitz-Oener and categorize the non-automatable tasks as: analytical non-routine tasks, interactive non-routine tasks. The last model was A-Task Based Approach by Melanie Arntz, Terry Gregory, 2016 in which they argue that FO Model Ignore that, the occupations consist of many tasks, part of them can be automated and another part cannot be. These models were applied on the level of the countries but not on corporate levels.The purpose of this paper is to combine the above three models and applying them on the Mobile operators in Egypt to determine the effect of Automation and AI on the Employment there, taking into consideration the incremental risk from COVID-19 which influenced the humanity.The conclusion of this research is that 41% of the staff working in the Technology field on the mobile operators, their jobs are subject to automation and COVID-19 will make an incremental Risk of 8%, due to creation of efficiency on performing the un-automatable tasks from home through digital platforms.
