Abstract:
We evaluated the results of endovascular management of patients with mid-basilar artery aneurysm (mBAA). During a seven year period, 14 patients (mean age 39.6 years, male/female ratio 1:1) with mBAA were treated with endovascular techniques at our institute. Pretreatment clinical grades were determined using the Hunt-Hess scale. Outcome was evaluated using the Glasgow Outcome Scale scores (GOS) during a mean follow-up period of 15.6 months (range, three to 70 months). Fourteen patients with 15 mBAAs were treated endovascularly. Four (28.6%) patients died of rebleeding within one day after embolization. In ten mBAAs, immediate postprocedural angiograms showed that complete occlusion was achieved, subtotal occlusion in one, and incomplete occlusion in four. Follow-up angiographic results in ten patients confirmed complete occlusion of 11 aneurysms. Long-term outcome was good (GOS Score 4 or 5) in ten patients (71.4%) and fatal (GOS Score 1) in four (28.6%). Favorable overall long-term outcome can be achieved in 78.6% patients with mBAAs. Endovascular management of mBAAs is an effective treatment in the long-term. In our experience, the natural history of mBAAs is dismal.

Rapid advancements in the infrastructure of Information and Communication Technology (ICT) have led to radically new but ubiquitous technology; cloud computing. Cloud computing has gracefully emerged offering services that possess on-demand scalability, huge computing power, and a utility-like availability, all at a relatively low cost. It has unsurprisingly become a paradigm shift in ICT, gaining adoptions in all forms of application i.e., personal, academic, business, or government. Not only for its cost-effectiveness but also for its inherent ability to meet business goals and provide strategic ICT resources. More recently there have been advances in cloud computing leading to the evolution of newer commercial cloud services, one of which is the Mobile backend as a Service (MBaaS). The MBaaS is important and required for a robust mobile application back-end data storage and management. Its wide adoption and importance stem from its ability to simplify application development and deployment. Also, MBaaS is robust, with the ability to cope with errors by providing nifty tools and other features. These enable rapid scaffolding of mobile applications. This paper reviews Mobile backend as a Service (MBaaS) and provides required background knowledge on some cloud services and their providers to enable stakeholders to make informed decisions and appropriate choices.

Abstract:
The ever-increasing complexity of cyber-physical systems is driving the need for assurance of critical infrastructure and embedded systems. However, traditional methods to secure cyber-physical systems—e.g., using cyber best practices, adapting mechanisms from information technology systems, and penetration testing followed by patching—are becoming ineffective. This paper describes, in detail, Verification Evidence and Resilient Design In anticipation of Cybersecurity Threats (VERDICT), a language and framework to address cyber resiliency. When we use the term resiliency, we mean hardening a system such that it anticipates and withstands attacks. VERDICT analyzes a system in the face of cyber threats and recommends design improvements that can be applied early in the system engineering process. This is done in two steps: (1) Analyzing at the system architectural level, with respect to cyber and safety requirements and (2) by analyzing at the component behavioral level, with respect to a set of cyber-resiliency properties. The framework consists of three parts: (1) Model-Based Architectural Analysis and Synthesis (MBAAS); (2) Assurance Case Fragments Generation (ACFG); and (3) Cyber Resiliency Verifier (CRV). The VERDICT language is an Architecture Analysis and Design Language (AADL) annex for modeling the safety and security aspects of a system’s architecture. MBAAS performs probabilistic analyses, suggests defenses to mitigate attacks, and generates attack-defense trees and fault trees as evidence of resiliency and safety. It can also synthesize optimal defense solutions—with respect to implementation costs. In addition, ACFG assembles MBAAS evidence into goal structuring notation for certification purposes. CRV analyzes behavioral aspects of the system (i.e., the design model)—modeled using the Assume-Guarantee Reasoning Environment (AGREE) annex and checked against cyber resiliency properties using the Kind 2 model checker. When a property is proved or disproved, a minimal set of vital system components responsible for the proof/disproof are identified. CRV also provides rich and localized diagnostics so the user can quickly identify problems and fix the design model. This paper describes the VERDICT language and each part of the framework in detail and includes a case study to demonstrate the effectiveness of VERDICT—in this case, a delivery drone.

Tujuan dari penelitian ini adalah membuat suatu aplikasi yang dapat membantu masyarakat untuk mencari informasi dan pemesanan jenis kendaraan pariwisata kepada pihak PO bus pariwisata yang ingin digunakan berdasarkan kebutuhan langsung dari smartphone yang mereka miliki. Aplikasi ini dibuat dengan menggunakan perangkat lunak Framework Jquery Mobile, Android Project, SDK Apiomat, Notepad++, browser Mozilla Firefox, dan Adobe PhoneGap. Metode yang digunakan adalah pendekatan Software Development Life Cycle (SDLC), yang terdiri dari fase perencanaan, analisa, perancangan, implementasi dan uji coba. Berdasarkan uji coba, aplikasi ini dapat diakses secara langsung melalui smartphone android dengan teknologi mobile backend as a service (MBaaS) yang terkoneksi internet, dan informasi yang diberikan aplikasi ini adalah mengenai kumpulan nama PO bus, alamat pusat, alamat cabang, fasilitas kendaraan, harga, peta lokasi, kontak telepon dan email yang dimiliki PO bus sesuai informasi yang dibutuhkan masyarakat khususnya yang tinggal di DKI Jakarta.

ABSTRACT Burkholderia pseudomallei is a gram-negative facultative intracellular pathogen that causes melioidosis, an invasive disease of humans and animals. To address the response of this bacterium to iron-limiting conditions, we first performed a global transcriptional analysis of RNA extracted from bacteria grown under iron-limiting and iron-rich conditions by microarrays. We focused our study on those open reading frames (ORFs) induced under iron limitation, which encoded predicted proteins that could be involved in the biosynthesis and uptake of the siderophore malleobactin. We purified this siderophore and determined that it consisted of at least three compounds with different molecular weights. We demonstrated that ORFs BPSL1776 and BPSL1774, designated mbaA and mbaF, respectively, are involved in the biosynthesis of malleobactin, while BPSL1775, named fmtA, is involved in its transport. These genes are in an operon with two other ORFs (mbaJ and mbaI) whose transcription is under the control of MbaS, a protein that belongs to the extracytoplasmic function sigma factors. Interestingly, the transcription of the mbaA, fmtA, and mbaS genes is not controlled by the availability of the siderophore malleobactin.

In order to determine the basic conditions for further research to identify threats to cloud technologies and measures for their counteraction, the article provides an extended analysis of cloud computing as a service. Among the above services, the basic services IaaS, SaaS, PaaS are selected, which are the basis for the existence of more unified services (CaaS, MCaS, DaaS, FaaS, IPaaS, MBaaS, NaaS, SeCaa, DBaaS, MaaS, DBaaS, MaaS, DBaaS, MaaS that increase the scope. Listed are possible service providers, including Ukrainian. It is determined that in order to transfer the work of systems of engineering calculations and computer-aided design (CAD) systems to a cloud platform, a rather new promising service CAEaaS (Cоmputеr Аidеd Еnginееrіng аs а Sеrvіcе) – computer engineering systems as a service. The most popular SAEs are used in the following industries: mechanical engineering and machine tools, defense and aerospace, energy, shipbuilding, semiconductor production, telecommunications, chemical, pharmaceutical and medical industry, construction, production of heating, air conditioning, ventilation. The success of a project decision depends on the awareness of responsible choice in the very first stage. Unfortunately, in support of the CAEaaS cloud service, Ukraine is only making the first steps and can provide the technological needs of enterprises, provided the involvement of foreign suppliers. According to the authors of the article, the future of the Ukrainian industry according to the cloud service SAEaS. Because CAE helps reduce the cost and time of product development, improve product quality and durability. Design decisions can be made based on their impact on performance. Designs can be evaluated and refined using computer modeling rather than physical prototype testing, saving money and time. This article is the basis for further research on the identification of threats to cloud technologies and their response.

Emerging Medical Body Area Networks (MBANs) require new, protected spectrum for clinical applications. This may mean uncoordinated and autonomous operation of multiple MBANs, within the new candidate bands. The question is that how will MBANs coexist as a secondary service with other radio systems? Clinical environment requires balance of robust and efficient wireless techniques to enable coexistence of MBANs and other radio devices where low transmission power MBANs as secondary systems may be vulnerable to interference from incumbent devices transceivers. Physical separation between the MBANs and incumbent radio devices and avoiding the transmission in the same frequency bands among the wireless techniques may be considered. In this paper, we propose interferencemanagement techniques considering such coexistencebetween the MBANs and other radio systems and deal withthe issue of co-existence with primary systems by proposing novel methods for a gateway-to-gateway coordination, to assist the methods described in the first and second part of this paper. Result is improved reliability and Quality of Service for MBANs. These would lead to multiple clinical benefits, including better patient mobility, more monitoring flexibility and extension of monitoring into care areas that are currently unmonitored. Reduced clinical errors and reduced overall monitoring costs are other results.

Polymeric nanoparticle suspension is a newly developed oil-displacing agent for enhanced oil recovery (EOR) in low-permeability reservoirs. In this work, SiO2/P(MBAAm-co-AM) polymeric nanoparticles were successfully synthesized by a simple distillation–precipitation polymerization method. Due to the introduction of polymer, the SiO2/P(MBAAm-co-AM) nanoparticles show a favorable swelling performance in aqueous solution, and their particle sizes increase from 631 to 1258 nm as the swelling times increase from 24 to 120 h. The apparent viscosity of SiO2/P(MBAAm-co-AM) suspension increases with an increase of mass concentration and swelling time, whereas it decreases as the salinity and temperature increase. The SiO2/P(MBAAm-co-AM) suspension behaves like a non-Newtonian fluid at lower shear rates, yet like a Newtonian fluid at shear rates greater than 300 s−1. The EOR tests of the SiO2/P(MBAAm-co-AM) suspension in heterogeneous, low-permeability cores show that SiO2/P(MBAAm-co-AM) nanoparticles can effectively improve the sweep efficiency and recover more residual oils. A high permeability ratio can result in a high incremental oil recovery in parallel cores. With an increase of the permeability ratio of parallel cores from 1.40 to 15.49, the ratios of incremental oil recoveries (low permeability/high permeability) change from 7.69/4.61 to 23.61/8.46. This work demonstrates that this SiO2/P(MBAAm-co-AM) suspension is an excellent conformance control agent for EOR in heterogeneous, low-permeability reservoirs. The findings of this study can help to further the understanding of the mechanisms of EOR using SiO2/P(MBAAm-co-AM) suspension in heterogeneous, low-permeability reservoirs.

Abstract Objective The Neurobehavioral Symptom Inventory (NSI) is a 22-item self-report measure of non-specific cognitive and somatic symptoms, which contains an embedded measure of symptom validity (Validity 10 scale [V-10]). The mild brain injury atypical symptom (mBIAS) is a separate 5-item measure of symptom validity designed for use in mild traumatic brain injury evaluation. The current study evaluated the NSI V-10 and mBIAS in relation to the Structured Inventory of Malingered Symptomatology (SIMS) Total score and subtest scores. Method Participants were 338 veterans who completed a parent study at a VA Medical Center. All participants deployed to Iraq and/or Afghanistan, did not have a history of moderate or severe traumatic brain injury, and did not have significant neurological or psychiatric conditions. Participants completed the SIMS and the NSI with mBIAS as part of a larger battery. The sample was 86% male and 57% White, with a mean age of 42 years and education of 15 years. Results AUC values using a higher SIMS Total cutoff score were .887 for the V-10 and .735 for the mBIAS; diagnostic accuracy data at various cutoff scores are presented. For SIMS subscales, the mBIAS had higher correlations to the Psychosis (r = .48) and Low Intelligence (r = .31) scales, whereas the V-10 had higher correlations to the Neurologic Impairment (r = .73), Amnestic Disorders (r = .66), and Affective Disorders (r = .59) scales. Conclusion Current data suggest that the V-10 and mBIAS measure different symptom validity constructs and could therefore be employed in a complimentary manner.

Objetivo: A literatura demonstra que a imagem corporal é uma dimensão central de auto e heteroavaliação e um foco de preocupação não só para as mulheres, mas também para os homens. Diversos autores têm sublinhado a necessidade de um maior investimento no desenvolvimento e validação de instrumentos de medida para a imagem corporal para o sexo masculino, dado que homens e mulheres se distinguem significativamente em relação às preocupações associadas à aparência física. A Male Body Attitude Scale-Revised(MBAS-R) é a versão revista de um instrumento de medida (MBAS), especificamente desenvolvido para a população masculina, que visa avaliar as atitudes e preocupações em relação à imagem corporal. O presente estudo visa estudar a estrutura fatorial e as propriedades psicométricas da versão portuguesa da MBAS-R.Métodos: O estudo foi conduzido numa amostra constituída por 222 homens da população geral portuguesa, com idades compreendidas entre os 18 e os 65 anos.Resultados: Os resultados de uma série de análises fatoriais confirmatórias demonstraram a adequação da versão portuguesa da MBAS-R (12-itens) com uma estrutura de dois fatores (massa muscular e massa gorda). A análise dos itens da versão portuguesa da MBAS-R revelou que todos os itens contribuem consistentemente para a respetiva subescala e para a medida global. Adicionalmente, tanto a escala total como as subescalas da versão portuguesa da MBAS-R revelaram boa consistência interna. Os resultados mostraram ainda que as preocupações e atitudes em relação à imagem corporal, avaliadas pela MBAS-R, se associam positivamente a vergonha externa, vergonha corporal e inflexibilidade alimentar.Conclusões: Os resultados sugerem que a MBAS-R é uma medida breve e válida para avaliar e caraterizar as preocupações masculinas em relação à imagem corporal de um modo global e, simultaneamente, especificamente em relação a duas dimensões centrais da vivência da imagem corporal (massa muscular e gordura corporal) nesta população.

Abstract. Surfactants in atmospheric aerosols determined as methylene blue active substances (MBAS) and ethyl violet active substances (EVAS). The MBAS and EVAS concentrations can be correlated with surface tension as determined by pendant drop analysis. The effect of surface tension was more clearly indicated in fine mode aerosol extracts. The concentration of MBAS and EVAS was determined before and after ultrafiltration analysis using AMICON centrifuge tubes that define a 5000 Da (5 K Da) nominal molecular weight fraction. Overall, MBAS and to a greater extent EVAS predominates in fraction with molecular weight below 5 K Da. In case of aerosols collected in Malaysia the higher molecular fractions tended to be a more predominant. The MBAS and EVAS are correlated with yellow to brown colours in aerosol extracts. Further experiments showed possible sources of surfactants (e.g. petrol soot, diesel soot) in atmospheric aerosols to yield material having molecular size below 5 K Da except for humic acid. The concentration of surfactants from these sources increased after ozone exposure and for humic acids it also general included smaller molecular weight surfactants.

ABSTRACT The formation of biofilms is thought to play a key role in the environmental survival of the marine bacterium Vibrio cholerae. Although the factors involved in V. cholerae attachment to abiotic surfaces have been extensively studied, relatively little is known about the mechanisms involved in the subsequent maturation of the biofilms. Here we report the identification of a novel gene, which we have named mbaA (for maintenance of biofilm architecture), that plays a role in the formation and maintenance of the highly organized three-dimensional architecture of V. cholerae El Tor biofilms. We demonstrate that although the absence of mbaA does not significantly affect the initial attachment of cells onto the surface, it leads to the formation of biofilms that lack the typical structure, including the pillars of cells separated by fluid-filled channels that are evident in mature wild-type biofilms. Microscopic analysis indicates that the absence of mbaA leads to an increase in the amount of extracellular matrix material in the biofilms. The predicted mbaA product is a member of a family of regulatory proteins, containing GGDEF and EAL domains, suggesting that MbaA regulates the synthesis of some component of the biofilm matrix.

This study examined the job changes of 680 early-career business school graduates. Although a number of anecdotal articles characterize MBAs as overly “careerist” and oriented toward job-hopping, little empirical research has focused on this issue. The research included a direct comparison of job-hopping behavior of MBAs with bachelor S degree graduates, taking into account a number of control variables, including demographic and economic variables. Results indicated that MBAs changed jobs less frequently than bachelor 5 degree graduates, even when a variety of other factors were controlled.

SummaryThere have been many developments in mindfulness-based approaches (MBAs) since the original mindfulness-based stress reduction (MBSR) course was first delivered in the 1980s. There are now a variety of mindfulness approaches, which are used to good effect in a range of psychological and physical health disorders. The addictions field first witnessed the development of relapse prevention techniques more than 30 years ago. MBAs have been adapted for use in relapse prevention and can be offered as a mindfulness-based relapse prevention (MBRP) course. MBSR or mindfulness-based cognitive therapy (MBCT) courses may also be useful for those with addiction problems. In this article, we describe the MBAs that are commonly offered at present. We highlight ways in which these approaches may be useful in the addictions feld. We also make suggestions for clinicians who may be interested in offering MBAs within their own service.

A series of batch die-away tests was conducted to evaluate the decay rates of MBAS and COD under conditions simulating untreated discharge. Under aerobic conditions, decay of MBAS was consistently faster than that of COD, and dilution into river water had no effect on either rate. Under strictly anaerobic conditions, neither MBAS nor COD decreased in concentration. Additional testing examined the fate of 14C-LAS and the results showed that LAS disappeared and was mineralized after a lag period. Half-lives were 8–10 hours for loss of parent and 11–12 hours for mineralization. Addition of 14C-LAS in sewage to systems previously exposed to raw sewage resulted in no lag and shorter half-lives. This work indicates that MBAS and LAS degrade more rapidly than COD, suggesting these materials would be at low levels once a stream had recovered from the addition of untreated sewage.

The present study is an attempt to assess the perceptions of the hiring managers about the managerial skills and attitude of the MBA employees in comparison to the non-MBA employees. A sample of 100 hiring managers, who have been involved in the placement of MBAs, was taken for the purpose of the study. The hiring managers have been found agreeing that employees with MBA degree are better in ‘communication skills’, ‘decision making’, ‘leadership qualities’, ‘handle job related tasks and responsibilities’, ‘learning’ and ‘adapting to changing situations’ than non-MBA employees. The hiring managers have also been found agreeing that MBA employees ‘demand higher pay’, ‘change jobs more frequently’, ‘are more authoritarian’, ‘have unrealistic expectations’, ‘are more loyal to their careers than to their jobs and organizations’ than non-MBA employees. The discriminant analytical approach has been applied to examine the impact of the ‘profile’ and ‘attributes’ examined in the study which clearly differentiate hiring managers who agree with the good managerial skills of MBAs and negative attitude of MBAs from those who disagree with these. In case of ‘profile’ the managers with MBA qualification and regarding the various attributes of MBAs considered in the present study; ‘MBAs are more adaptive to changing situations’ and ‘MBAs change jobs more frequently than non-MBA employees’ have been found evoking significant differences among hiring managers. The findings of the present study can be useful for various stakeholders like, monitoring agencies and education policy makers, faculty and governing bodies of various management institutes for developing required management skills and attitudes among the students.

The careers of MBAs from a top US business school are studied to understand how career dynamics differ by gender. Although male and female MBAs have nearly identical earnings at the outset of their careers, their earnings soon diverge, with the male earnings advantage reaching almost 60 log points a decade after MBA completion. Three proximate factors account for the large and rising gender gap in earnings: differences in training prior to MBA graduation, differences in career interruptions, and differences in weekly hours. While there is a growing body of research on the subject of entrepreneurship education and learning, studies of the executive master of business administration (EMBA) are relatively sparse. This article offers an example of an entrepreneurship course specifically tailored to the more mature and experienced EMBA participants. The curriculum offered in this article is the result of 25 years of teaching, testing and refining of content and learning approaches in a university setting. The argument is that the course, as currently designed, can serve as a template for courses to be conducted in business schools located in similar cultural contexts and economic environments.

Ozone was incorporated into an ultrafiltration system to produce higher quality reclaimed water from domestic laundry wastewater. Characteristics of the wastewater for initial washing waste were 488~2,847 mg/L COD, 62~674 mg/L MBAS, and 38~857 mg/L SS. The wastewater was contacted with ozone in a 10L storage tank and circulated through the membrane module for inner pressurized cross-flow filtration. The concentrate was returned back to the contact tank. The membrane used in this experiment was hollow fiber polysulfone UF membrane with MWCO 5,000 and 10,000. It has an effective filtration area of 0.06m2. The experiment was carried out in two phases with either continuous or intermittent ozone injection. For intermittent ozone injection, the mode of injection interval was changed to 5 min./5 min. and 5 min./10 min. for injection/idling. Ozone was dosed at the concentration of 1.5 mg/L. The permeate quality of UF (MWCO 5,000) was 57 mg/L as COD and 5 mg/L as MBAS at continuous ozone injection with removal of 95% in COD and 96.9% in MBAS. Using UF with MWCO 10,000, it was 93.7% and 95.5% of COD and MBAS, respectively. And using intermittent ozone injection, the removal efficiency was 93% in COD and 93.5% in MBAS without any deterioration in COD and MBAS removal. It could reduce the treatment cost. Using ozone injection, fouling of the membrane was also controlled by increasing organic degradation. The flux of UF (MWCO 5,000 and 10,000) was 0.13 and 0.20 m/d for 3 hour filtration (TMP 40≈45 kPa) without ozone injection. It was increased to 0.18 and 0.24m/d by ozone injection. The reclaimed water quality could be estimated well enough to reuse for rinsing purposes.

The aim of this study was to examine the factor structure of the Male Body Attitudes Scale (MBAS) among a sample of 212 adolescent males by conducting a confirmatory factor analysis. The data revealed that while muscularity and low body fat factors were appropriate for the male sample, the height factor was not supported. The composite MBAS-Spanish version (MBAS-S) and its two factors had high internal consistency and were not significantly related to the traditionally used measure of body image (Eating Disorder Inventory–Body Dissatisfaction [BD]), confirming that this subscale only appears to evaluate female body concerns. Correlations with psychological well-being variables (anxiety, emotional regulation, and self-esteem) supported the concurrent validity of the instrument. These findings provide further evidence in support of the hypothesis that BD in males has been inappropriately evaluated by measures designed specifically for females. Despite the fact that our results did not replicate the original factorial structure of the instrument, the MBAS-S presents adequate psychometric properties and can be considered as a valuable assessment tool for BD among adolescent males.

This paper describes the adaptation of evolutionary algorithms (EAs) to the structural optimization of chemical engineering plants, using rigorous process simulation combined with realistic costing procedures to calculate target function values. To represent chemical engineering plants, a network representation with typed vertices and variable structure will be introduced. For this representation, we introduce a technique on how to create problem specific search operators and apply them in stochastic optimization procedures. The applicability of the approach is demonstrated by a reference example. The design of the algorithms will be oriented at the systematic framework of metricbased evolutionary algorithms (MBEAs). MBEAs are a special class of evolutionary algorithms, fulfilling certain guidelines for the design of search operators, whose benefits have been proven in theory and practice. MBEAs rely upon a suitable definition of a metric on the search space. The definition of a metric for the graph representation will be one of the main issues discussed in this paper. Although this article deals with the problem domain of chemical plant optimization, the algorithmic design can be easily transferred to similar network optimization problems. A useful distance measure for variable dimensionality search spaces is suggested.

Metal Big Area Additive Manufacturing (MBAAM) is a novel wire-arc additive manufacturing method that uses a correction-based approach developed at the Oak Ridge National Laboratory (ORNL). This approach is an integrated software method that minimizes the dynamic nature of welding and compensates for build height. The MBAAM process is used to fabricate simple geometry thin walled specimens, using a C-Mn steel weld wire, to investigate the scatter in mechanical properties and correlate them to the underlying microstructure. The uni-axial tensile tests show isotropic tensile and yield properties with respect to building directions, although some scatter in elongation is observed. Large scatter is observed in the Charpy Impact tests. The microstructure characterization reveals mostly homogenous ferrite grains with some pearlite, except for some changes in morphology and grain size at the interface between the build and the base plate. The measured properties and microstructure are compared with the toughness and strength values reported in the literature, and a hypothesis is developed to rationalize the differences. Overall, the MBAAM process creates stable, isotropic, and weld-like mechanical properties in the deposit, while achieving a precise geometry obtained through a real-time feedback sensing, closed loop control system.

Zusammenfassung: Einleitung: Das Asperger-Syndrom (AS) ist eine Störung von unsicherer nosologischer Validität, deren Abgrenzung zum sogenannten High-functioning Autismus sehr schwierig ist. Diese Störungen werden bis heute leider erst sehr spät erkannt, oft werden zunächst einige andere Diagnosen gestellt. Ziel der vorliegenden Untersuchung ist es, ein Instrument (MBAS) vorzustellen, welches für die Erfassung dieser Störungen sensibel ist. Methodik: Der Fragebogen wurde insgesamt 91 Probanden (44 mit einer autistischen Diagnose, 47 mit nicht-autistischen Diagnosen) vorgelegt. Ergebnisse: Die Items der MBAS erweisen sich überwiegend als mittelschwer und überwiegend gut trennscharf. Die interne Konsistenz der gesamten Skala ist als sehr hoch zu bewerten (Cronbach’s alpha = .91). Es liegt eine konvergente Validität der Ergebnisse aus der MBAS und dem ADI-R vor (r = 0.61, p = .001) Vergleicht man die gebildeten Gruppen «autistisch» vs. «nicht autistisch» in Bezug auf den Gesamtscore miteinander, so zeigt sich, dass der Fragebogen sehr gut zwischen den beiden Vergleichsgruppen unterscheidet. Bei einer Sensitivität von 95.5% und einer Spezifität von 95.7% kommt es nur zu wenigen Fehlklassifikationen, falsch negative Einschätzungen kamen in unserer Stichprobe lediglich bei sehr jungen Probanden vor. Schlussfolgerungen: Die «MBAS» erweist sich als reliabel und diagnostisch valide. Zum Zwecke der Präselektion oder Generierung von Verdachtsdiagnosen des Autismus auf hohem Funktionsniveau ist dieser Fragebogen gut geeignet.

In recent years, advances in structure-based drug design and the development of an impressive variety of high-throughput screening (HTS) assay formats have yielded an expanding list of protein-protein interaction inhibitors. Despite these advances, protein-protein interaction targets are still widely considered difficult to disrupt with small molecules. The authors present here the results from screening 220,017 compounds from the National Institute of Health’s small-molecule library in a novel p53-hDM2 protein-protein interaction biosensor (PPIB) assay. The p53-hDM2 positional biosensor performed robustly and reproducibly throughout the high-content screening (HCS) campaign, and analysis of the multiparameter data from images of the 3 fluorescent channels enabled the authors to identify and eliminate compounds that were cytotoxic or fluorescent artifacts. The HCS campaign yielded 3 structurally related methylbenzo-naphthyridin-5-amine (MBNA) hits with IC50s between 30 and 50 µM in the p53-hDM2 PPIB. In HCT116 cells with wild-type (WT) p53, the MBNAs enhanced p53 protein levels, increased the expression of p53 target genes, caused a cell cycle arrest in G1, induced apoptosis, and inhibited cell proliferation with an IC50 ~4 µM. The prototype disruptor of p53-hDM2 interactions Nutlin-3 was more potent than the MBNAs in the p53-hDM2 PPIB assay but produced equivalent biological results in HCT116 cells WT for p53. Unlike Nutlin-3, however, MBNAs also increased the percentage of apoptosis in p53 null cells and exhibited similar potencies for growth inhibition in isogenic cell lines null for p53 or p21. Neither the MBNAs nor Nutin-3 caused cell cycle arrest in p53 null HCT116 cells. Despite the relatively modest size of the screening library, the combination of a novel p53-hDM2 PPIB assay together with an automated imaging HCS platform and image analysis methods enabled the discovery of a novel chemotype series that disrupts p53-hDM2 interactions in cells.

ABSTRACT In Vibrio cholerae, the second messenger 3′,5′-cyclic diguanylic acid (c-di-GMP) regulates several cellular processes, such as formation of corrugated colony morphology, biofilm formation, motility, and virulence factor production. Both synthesis and degradation of c-di-GMP in the cell are modulated by proteins containing GGDEF and/or EAL domains, which function as a diguanylate cyclase and a phosphodiesterase, respectively. The expression of two genes, cdgC and mbaA, which encode proteins harboring both GGDEF and EAL domains is higher in the rugose phase variant of V. cholerae than in the smooth variant. In this study, we carried out gene expression analysis to determine the genes regulated by CdgC in the rugose and smooth phase variants of V. cholerae. We determined that CdgC regulates expression of genes required for V. cholerae polysaccharide synthesis and of the transcriptional regulator genes vpsR, vpsT, and hapR. CdgC also regulates expression of genes involved in extracellular protein secretion, flagellar biosynthesis, and virulence factor production. We then compared the genes regulated by CdgC and by MbaA, during both exponential and stationary phases of growth, to elucidate processes regulated by them. Identification of the regulons of CdgC and MbaA revealed that the regulons overlap, but the timing of regulation exerted by CdgC and MbaA is different, suggesting the interplay and complexity of the c-di-GMP signal transduction pathways operating in V. cholerae.
