O objetivo do presente estudo foi avaliar a aptidão aeróbia em testes de caminhada com carga externa aplicada por meio da inclinação da esteira, a partir da relação não linear entre inclinação da esteira e tempo até a exaustão em velocidade fixa. Doze indivíduos do gênero masculino com 23,2 ± 2,7 anos de idade, 74,0 ± 7,9kg de massa corporal e 23,7 ± 2,5kg·(m²)-1 de IMC, realizaram duas etapas de testes de caminhada em esteira ergométrica com velocidade fixa de 5,5km·h-1 em todos os testes e sobrecarga de intensidade aplicada por meio de inclinação da esteira (%). A etapa 1 consistiu de três testes retangulares até a exaustão voluntária, nas intensidades de 18%, 20% e 22% de inclinação, para determinação dos parâmetros do modelo de potência crítica por dois modelos lineares e um hiperbólico. A etapa 2 consistiu na determinação da intensidade correspondente ao máximo estado estável de lactato sanguíneo (MEEL). ANOVA demonstrou que o modelo hiperbólico (15,4 ± 1,1%) resultou em estimativa significativamente menor que os outros dois modelos lineares inclinação-tempo-1 (16,0 ± 1,0%) e hiperbólico linearizado tempo-1-inclinação (15,9 ± 1,0%), porém, houve alta correlação entre os modelos. Os dois modelos lineares superestimaram a intensidade do MEEL (14,1 ± 1,4%), e o modelo hiperbólico, mesmo sem diferença estatística, apresentou fraca correlação, com baixa concordância em relação ao MEEL. Conclui-se que a relação inclinação-tempo até a exaustão, em testes de caminhada, não permitem a estimativa de intensidade de exercício suportável por longo período de tempo.

Abstract:
An earlier study (Meel, 2003) showed that more than 90% of victims of sexual assault in Transkei region, South Africa, were HIV-seronegative at the time of the incident. This was despite the fact that the community had a high prevalence of HIV. In sexual assault cases post-exposure prophylaxis (PEP) is recommended to prevent HIV transmission. Therefore, therapy with zidovudine (AZT) and lamivudine (3TC) is justified. The purpose of this study is to describe the demographic characteristics of the victims, to assess the outcome of HIV transmission and to evaluate the success of PEP after its implementation in Transkei. There were 594 victims of sexual assault during the study period at Sinawe Centre from 2000-2003. Of these, 346 (58.2%) were children under the age of 15 years. Seventeen children (2.9%) were found to be HIV positive at the first test. Among the adults, 58(9.8%) tested HIV positive. Of the 225 who attended after PEP was introduced, only two were found to be HIV seropositive at the time of the incident. A second test was recommended after four weeks and a third after 12 weeks. The majority of the victims did not report for the second test, but all 35 who did come to be tested were seronegative. Seventeen of those were between 11-15 years of age. Only seven victims came for the third test, and they, too, were negative. Nausea and vomiting were the commonest side effects of PEP treatment in four patients and one developed a generalized rash. Only one victim seroconverted.

AbstractObjectiveTo develop a worked example of product reformulation of a very popular ‘junk food’ to meet nutritional guidelines for public health in a ready meal.DesignIndicative survey of popular Margherita pizzas, followed by product reformulation, applying dietary guidelines to generate a single-item pizza meal containing 30 % daily amounts of energy and all nutrients. An iterative process was used; first to optimize nutrient balance by adjusting the proportions of bread base, tomato-based sauce and mozzarella topping, then adding ingredients to provide specific nutrients and consumer tasting.SettingUrban areas of contrasting socio-economic status.SubjectsUntrained unselected adults (n49) and children (n63), assessing pizza at tasting stations.ResultsMost commercial pizzas provide insufficient information to assess all nutrients and traditional Margherita pizza ingredients provide insufficient Fe, Zn, iodine, and vitamins C and B12. Energy content of the portions currently sold as standard range from 837 to 2351 kJ (200 to 562 kcal), and most exceed 30 % Guideline Daily Amounts for saturated fat and Na when a 2510 kJ (600 kcal) notional meal is considered. The ‘nutritionally balanced pizza’ provides the required energy for a single-item meal (2510 kJ/600 kcal), with all nutrients within recommended ranges: Na (473 mg, ∼45 % below recommended level), saturated fat (<11 % energy) and dietary fibre (13·7 g). Most adults (77 %) and children (81 %) rated it ‘as good as’ or ‘better than’ their usual choice.ConclusionsNutritional guidelines to reduce chronic diseases can be applied to reformulate ‘junk food’ ready meals, to improve public health through a health-by-stealth approach without requiring change in eating habits.

In this study, nutritive values of rapeseed (R), rapeseed meal-expeller A (RM-A), rapeseed meal-expeller B (RM-B) and extracted rapeseed meal (ERM) were compared. The trials were performed using the <i>in sacco</i> method with three steers of the Czech Fleckvieh breed, which were fitted with a permanent ruminal cannula. Nylon bags with samples were incubated in the rumen for 2, 4, 8, 16, 24 and 48 hours. The effective degradability (ED) of crude protein (CP) was calculated at 0.08, 0.06 and 0.04 1/h of rumen particulate outflow rates (<i>k</i>), and the obtained ED values were 65.4, 70.8 and 77.4% for R, 86.7, 88.1 and 89.7% for RM-A, 82.2, 84.4 and 87.0% for RM-B and 56.3, 62.1 and 69.6% for ERM, respectively. The ED values significantly differed between feeds (<i>P</i> < 0.05) for all rumen particulate outflow rates. Disappearances of amino acids (AA) after 16 hours of incubation in the rumen of R, RM-A, RM-B and ERM were determined. In all cases, the concentrations of AA in the feeds determined after incubation in rumen were lower than in the original feeds. A mobile bag technique was used to determine intestinal digestibility. In the experiment, three dry cows fitted with permanent large ruminal cannula and the T-piece cannula in the proximal duodenum were used. The intestinal digestibilities of rumen undegraded CP (DSI) were estimated 30.0% in R, 15.4% in RM-A, 27.6% in RM-B and 65.3% in ERM. The DSI values significantly differ between the feeds (<i>P</i> < 0.05), except for the difference between R and RM-B.

The present study examines the ability of meal plans with ≥4 grain-based servings daily to meet nutrient recommendations using lacto-ovo vegetarian and rice-based cuisines.DesignEighteen plans from each cuisine for three age brackets for both genders were developed. Plans aimed for ≥4 servings of grain-based foods daily, with separate plans for all wholegrain, all refined-grain and half wholegrain–half refined-grain foods. Meal plans followed an isoenergetic approach and were designed to meet specific Australian nutrient reference values and serving sizes.ResultsAll plans met the Recommended Dietary Intake or Adequate Intake for targeted nutrients except for Fe in the rice-based meal plan for females aged ≥19 years (17 mg). In the plans for 14–18 year and ≥19 year age groups, four servings of grain-based foods could be accommodated. In the plans for 9–13 years, increasing the number of grain-based food servings to four reduced micronutrients levels delivered by the total diet. Specific food choices were made to ensure nutrient targets were met across each category for wholegrain and refined-grain plans. The major difference in nutrients between wholegrain and refined-grain foods was found in the vegetarian cuisine, where the meal plans containing whole grains produced on average 30 % higher fibre (38–53 g) levels than those with refined grains (27–40 g).ConclusionsWith careful food selection, meal plans with ≥4 servings of grain-based foods daily can meet nutrient reference values for lacto-ovo vegetarian and rice-based cuisines.

The composition of meals that would meet adults’ nutrient requirements was determined by constructing simulated meals, drawing on the selection of foods available to programmes.SampleIn all, seventy meals, sampled from eighteen programmes serving homeless individuals.ResultsOn average, the meals contained 2·6 servings of grain products, 1·7 servings of meat and alternatives, 4·1 servings of vegetables and fruits and 0·4 servings of milk products. The energy and nutrient contents of most meals were below adults’ average daily requirements. Most meals included both purchased and donated foods; the vitamin C content of meals was positively associated with the percentage of energy from donations. Increasing portion sizes improved the nutrient contribution of meals, but the provision of more milk products and fruits and vegetables was required to meet adults’ nutrient requirements.ConclusionsThe meals assessed were inadequate to meet adults’ nutrient requirements. Improving the nutritional quality of meals requires additional resources.

Abstract:
Meat is an attractive medium for microorganism multiplication due to its composition. The microflora of unpacked meat or of meat packed in wrapping plastic foil is significantly different from the microflora of vacuum-packed or controlled-atmosphere packed meat. Dominant microorganisms of unpacked meat are some species of the genus Pseudomonas sp., namely Pseudomonas fragi, Shewanella putrefaciens, Psychrobacter and Acinetobacter, which largely contribute to meat spoilage. From vacuum-packed or controlled-atmosphere packed meat were isolated mainly Brochotrix thermosphacta, heterofermentative lactobacilli (L. cellobiosus), Carnobacterium sp. and Leuconostoc sp. Characteristic microorganisms of vacuum-packed meat were Leuconostoc carnosum, Leuconostoc gelidum and Carnobacterium divergens. Quantitative proportions of the groups of microorganisms in packed meat are related to meat pH value, plastic foil permeability for oxygen and type of packed product.

Public interest in popular diets is increasing, in particular whole-food plant-based (WFPB) and vegan diets. Whether these diets, as theoretically implemented, meet current food-based and nutrient-based recommendations has not been evaluated in detail. Self-identified WFPB and vegan diet followers in the Adhering to Dietary Approaches for Personal Taste (ADAPT) Feasibility Survey reported their most frequently used sources of information on nutrition and cooking. Thirty representative days of meal plans were created for each diet. Weighted mean food group and nutrient levels were calculated using the Nutrition Data System for Research (NDSR) and data were compared to DRIs and/or USDA Dietary Guidelines/MyPlate meal plan recommendations. The calculated HEI-2015 scores were 88 out of 100 for both WFPB and vegan meal plans. Because of similar nutrient composition, only WFPB results are presented. In comparison to MyPlate, WFPB meal plans provide more total vegetables (180%), green leafy vegetables (238%), legumes (460%), whole fruit (100%), whole grains (132%), and less refined grains (−74%). Fiber level exceeds the adequate intakes (AI) across all age groups. WFPB meal plans failed to meet the Recommended Dietary Allowances (RDA)s for vitamin B12 and D without supplementation, as well as the RDA for calcium for women aged 51–70. Individuals who adhere to WFBP meal plans would have higher overall dietary quality as defined by the HEI-2015 score as compared to typical US intakes with the exceptions of calcium for older women and vitamins B12 and D without supplementation. Future research should compare actual self-reported dietary intakes to theoretical targets.

Abstract Background and aim of the study. Telomerase inhibition is a novel promising anticancer strategy which has been pursued using antisense oligonucleotides and more recently pharmacological inhibitors. BIBR-1532 is a mixed-type non-competitive inhibitor of the catalytic subunit of telomerase (h-TERT). In solid tumor cell lines, BIBR-1532 successfully induced telomere shortening and growth inhibition (Damm K et al, The Embo J, 2001). The activity of BIBR-1532 has not been so far evaluated in hematological cancers. Aim of this study is to evaluate the activity of this compound in cell lines derived from B-cell lymphoid tumors. These neoplasms are an interesting model to investigate BIBR1532 since they show marked differences in their telomere lenght according to GC origin (i.e. GC-derived have long telomeres and extra GC-derived have short telomeres)(Ladetto et al Blood 2004). Methods. We employed two extra-GC derived cell lines (the multiple myeloma KMS-11 and the chronic lymphocytic leukemia JVM-2 provided respectively by M. Massaia MD and C Carlo Stella MD) and two GC-derived cell lines (the Burkitt’s Lymphoma CA46 and the follicular lymphoma cell line DHL16 provided by J.G. Gribben MD). All the four cell lines were h-TERT positive. BIBR-1532 (kindly provided by Klaus Damm MD and Jacques van Meel, PhD, Boehringer Ingelheim Pharma, Germany) was administrated thrice a week to the standard concentration of 10μM. Cells were cultured for at least 80 mitotic divisions both in presence and absence of the agent. Cell growth was assessed three times a week. Aliquots of cultured cells were taken approximately every 20 days for telomere length evaluation. Cells were also assessed using the propidium iodide assay to verify whether growth inhibition was due to apotosis or prolipherative arrest. Results. As expected based on their GC-status, the four cell lines had different baseline telomere length (CA46: 6500bp DHL 16: 7000bp KMS11 3200bp, JVM-2 3500). When cells were cultured without BIBR 1532 no significant telomere shortening occurred over the whole culture period. In the presence of BIBR-1532, cell lines approximately lost 32bp at each mitotic division. After 20 mitoses KMS-11 and JVM-2 underwent marked and progressive growth inhibition with a clear plateau effect after 50 to 55 mitoses. In contrast no growth inhibition was noticed in GC-derived cell lines despite a similar degree telomere erosion. Growth inhibition in cell lines responsive to BIBR-1532 was mostly associated with G1 arrest. Conclusions: These results are the first reported experience on the use of BIBR1532 on B-lymphoid cell lines and indicate that: a) BIBR1532 effectively induces telomere shortening in lymphoid tumors; b) in extra-GC telomere erosion quickly lead to pharmacologically-induced senescence with marked growth inhibition; c) GC-derived lymphoid cell lines are effectively protected from pharmacological senescence induced by BIBR 1532 due to their long telomeres. These results indicate that telomerase inhibitors require extensive evaluation as anti-cancer agents in extra-GC-derived lymphoid tumors.
