Dissertação de Mestrado em Etologia
Neste trabalho pretendeu-se realizar um estudo preliminar sobre as baleias corcundas (Megaptera novaeangliae) que ocorrem nas águas de S. Tomé, e teve como objectivos registar as ocorrências e abundâncias de baleias corcunda que ocorrem nesta área, descrever e identificar os tipos de grupos que ocorrem nesta área, descrever os comportamentos mais frequentes, identificar os diferentes indivíduos que aqui ocorrem e avaliar se esta área contínua a ser uma zona de reprodução para esta espécie. Durante as 125 saídas realizadas em duas épocas consecutivas (2002 e 2003), foram realizadas 65 amostragens comportamentais em que se registavam os vários comportamentos observados, tipos de grupo, número de animais, posição de GPS dos animais e zona. Fazia-se ainda o registo fotográfico (da zona dorsal e caudal) dos animais avistados para posterior foto-identificação. Observou-se baleias corcundas em 45 saídas, tendo sido contabilizados um total de 46 grupos de baleias. As baleias ocorrem nesta área de Agosto a Novembro. Os tipos de grupos mais frequentemente observados foram os grupos "mãe-cria" e "solitários". Em apenas duas ocasiões foram observados comportamentos relacionados com a corte e o acasalamento com a ocorrência de "grupos competitivos". A área onde foram registados mais avistamentos foi na zona sul do Ilhéu das Rolas. Os comportamentos mais observados à superfície foram a respiração normal, seguida dos batimentos e dos breachings. Nos dois anos foram identificados 18 animais adultos, observou-se que alguns animais permaneciam durante um certo tempo na área de estudo durante a época, no entanto não foram reavistados animais de um ano para outro. Assim este trabalho foi a primeira abordagem sobre as baleias corcunda que ocorrem nesta área. Conseguiu-se obter uma visão geral sobre os tipos de grupos, comportamentos e ocorrências das baleias corcundas nesta área, podendo este estudo vir a servir de ponto de partida para um estudo mais aprofundado e continuado sobre estes animais.

Abstract:
La mise au point de réacteurs nucléaires sous-critiques (Accelerator Driven System - ADS), intrinsèquement sûrs, pour incinérer les déchets nucléaires par transmutation d’actinides de durée de vie longue en radioéléments de plus courte durée de vie, réclament des sources intenses de neutrons. Ces sources sont produites par la technique de spallations « éjections », c’est-à-dire l’interaction d’un faisceau de protons de haute énergie et haute intensité avec une cible contenant des métaux lourds. Compte tenu de la puissance déposée par les protons dans la cible (de l'ordre de quelques MW) seuls des métaux sous forme liquide peuvent à la fois résister aux défauts induits et évacuer la chaleur. Dans ce contexte l'expérience MEGAPIE (MEGawatt PIlot Experiment) fonctionnant avec une fenêtre au niveau de la cible de spallation est pionnière et ouvre la voie vers des cibles de haute puissance. Pour la mise en œuvre de cette cible MEGAPIE, les choix de l’alliage de la cible de spallations, des matériaux de structures de la fenêtre et des parties chaudes sont portés respectivement sur l’eutectique plomb-bismuth, bon rendement neutronique, et l’acier T91 (1%Mo9%Cr en masse), bonne tenue à l’irradiation. Pour les parties froides l’acier 316L a été choisi. Toutefois, malgré sa bonne tenue à l’irradiation, le contact de ces deux alliages (eutectique plomb-bismuth et acier T91) présente quelques inconvénients qui nécessitent d’approfondir la connaissance de leurs propriétés physico-chimiques dans l’environnement MEGAPIE et motive cette présente étude. La contribution de ce travail au développement du réacteur de transmutation a consisté : - d’une part à l'élaboration de revêtements destinés à améliorer les performances de l'acier de structure T91 vis à vis de l'oxydation et de la fissuration par l'eutectique Pb-Bi liquide, - d'autre part à l'étude thermodynamique du système Pb-Bi-(Hg)-Fe-Cr-Al-O pour connaître l’ensemble des équilibres entre phases existant dans les conditions imposées par le fonctionnement du réacteur ADS. Le deuxième volet de ce travail trouve sa justification dans les résultats du premier qui ont mis en évidence le rôle primordial joué par les oxydes mixtes dans l'interaction corrosive entre l'acier et l'eutectique Pb-Bi faiblement oxygéné
The development of sub-critical nuclear reactors (Accelerator Driven System - ADS), intrinsically safe to incinerate nuclear waste by transmutation of long-lived actinides to radionuclides with a shorter existence, requires intense sources of neutrons. These sources are produced by the "ejected" spallation technique, i.e. the interaction of a high energy, high intensity proton beam with a target containing heavy metals. Considering the power of the proton beam transferred to the target (of a few MW), only metals in the liquid form can be cooled in a closed cooling system and resist to nuclear reactor induced by the proton beam. In this context, the MEGAPIE project (MEGawatt PIlot Experiment), which works with a window at the spallation target, is pioneer and opens the way for high-power targets. The choice of the alloy for the MEGAPIE spallation target is a lead-bismuth eutectic, which is efficient for producing neutrons. For the window beam and hot parts, the best choice at present is the T91 steel (1%Mo9%Cr in mass), due to its good irradiation resistance. For cold parts, the 316L steel was chosen. However, the contact of the two alloys (eutectic lead-bismuth and T91 steel) presents some inconveniences that need to be investigated in relation to their physico-chemical properties in the MEGAPIE environment, which motivates this study. The contribution of this work to this subject consisted of : - In a part, the development of coatings designed to improve the chemical and mechanical performances of the T91 steel in contact of the lead-bismuth eutectic liquid alloy, - In another part, the investigation of the thermodynamic properties of the Pb-Bi-(Hg)-Fe-Cr-Al-O system for all phase equilibria within the conditions imposed by the operation of the ADS reactor. The second part of this work is justified by the results, which, for the first time, highlight the role played by the mixed oxide phases in the corrosive interaction between the T91 steel and Pb-Bi eutectic, in a poorly oxygenated environment

Abstract:
Biological evidence collected at crime scenes are often subjected to forensic deoxyribonucleic acid (DNA) testing. During forensic DNA testing the DNA from the evidence and known samples are extracted, purified, amplified using Polymerase Chain Reaction (PCR), and analyzed using capillary electrophoresis (CE). In order to appropriately compare the profile of the suspect to the evidence, it is essential that interpretation parameters and optimized processing schemes are established. This study endeavors to accomplish this by: first, evaluating whether the PCR temperature cycling is detrimental to the amplification process; and second, by establishing and comparing interpretation parameters for two commonly employed short tandem repeat (STR) megaplexes. To evaluate the effects of temperature cycling on downstream signal, a dynamic systems model was developed, validated, and used to test the effects of temperature on DNA damage and the subsequent fluorescence signal. Though DNA is generally thought to be a stable molecule, heat-induced damage does occur. Specifically, this model assesses the damage to the guanine and cytosine bases during temperature cycling. The model conducts the amplification of a single locus during PCR and generates the peak height observed after capillary electrophoresis. The model was designed to assess not only the effects of heat-induced DNA damage but to also incorporate variability in PCR efficiency. The simulated data indicate that heat-induced DNA damage does not significantly reduce the allelic signal. Also, although changes in PCR efficiency introduce variability in the peak heights at all targets, the peak heights observed with and without heat-induced DNA damage are not significantly different. In fact, the variation in PCR efficiency has a larger effect on the number of amplicons produced than does the heat-induced DNA damage. The second part of this study compares two PCR amplification megaplexes, PowerPlex® Fusion and GlobalFiler®, by evaluating their sensitivities, limits of detection, presence of artifacts, heterozygous peak balance, and ability to amplify minor contributors in DNA mixtures. Analysis of single source samples using weighted least squares regression analysis indicates that PowerPlex® Fusion has greater analytical sensitivities and lower limits of detection at comparable dye channels, and both kits display similar heterozygous balance. However, the GlobalFiler® processing scheme produced fewer artifacts for the various single source samples analyzed, particularly at higher target amounts. Also, analysis of two and three person DNA mixtures indicates that both megaplexes perform equally well when detection of the minor contributor is the criterion.

Abstract:
A new type of liquid-metal target is designed for the Spallation Neutron Source in PSI. LBE is selected to be the target material and the primary coolant as well. RELAP5/MOD 3.2 is used to analyze the system thermal hydraulics. The nominal conditions are chosen based on temperature constraints from the design assessments. The steady state results are in the proximity of the design specifications and the heat removal capacity is adequately deployed. The normal thermal hydraulic transients, namely the proton beam and beam interrupt, are studied. A basic PID (Proportional, Integral, Derivative) control is implemented in the RELAP5 for regulating the target temperature. It is found that the control chain works very well for the beam trip in limiting the temperature fluctuations. In a beam interrupt, the proton beam is completely turned off without recovering. The transition from full power to hot-standby is quite smooth, but it becomes oscillatory in the long run due to the timelags in the cooling loops’ responses. An off normal case of target main coolant trip has also been studied. Without the main pump, the target can still be operated in the natural circulation mode, and the control can cope with the normal beam transients and restarting the target from hot standby.

Abstract:
The paper reports first steps in an analysis, using Computational Fluid Dynamics (CFD), of a leak of hot (&gt;250°C) lead bismuth eutectic (LBE) from the MEGAPIE spallation source target into the helium-filled gap between the target hull and a close-fitting, double-walled safety container. Issues addressed are: (1) determining the initial impact pressure of the LBE on the inner shell of the safety container, which could result in the gap between the shells being closed, and its cooling capacity compromised; (2) LBE freezing on the cold shell surface could delay action of the leak detection system, and hence the beam trip control; and (3) heat transfer from the hot LBE to the D2O circulating between the safety container walls could induce boiling and subsequent pressurization of the circuit. Steps in the model development needed for the simulation are described, which include, in particular, the implementation of a liquid/gas interface-tracking model based on the Level Set approach. Results confirm that the safety container will be adequately cooled by the D2O during normal target operation, but first indications are that boiling might occur within the D2O circuit if substantial amounts of LBE leak from the target vessel unless its outer surface is coated with an insulating layer. Pressures generated by the impact of a slug of LBE on the safety window do not threaten its structural integrity, and no delay of the leak detection equipment as a consequence of LBE freezing is anticipated.

Abstract:
An interferometric optical profiler using a Videk Megaplus camera interfacing to an 80386 based computer has been developed for measuring surface roughness and 3-D profiles at high resolution. The camera has 1320×1035 square pixels which are 6.8 µm on a side. Fields of view over 1024×1024 pixels can range from 35 to 4642 µm with spatial sampling intervals of 0.034-4.5 µm. With this camera the system is limited by the optical resolution and not the detector spacing. The computer system consists of an 80386 microprocessor and a floating point coprocessor, 16MB of RAM, and a VGA monitor with software running under Xenix 386. Digital data from the camera are captured using a frame grabber and displayed as live video on a monitor at 7 frames/s. The rms repeatability of measurements made with this instrument is better than 0.7 nm and can be half of this value with averaging.
