Abstract:
Osmanlı 18. yüzyıldan itibaren eğitim, bilim ve askerî alanda çok sayıda değişim ve dönüşüm yaşamıştır. Modernleşme olarak da adlandırabileceğimiz bu sürecin eğitim alanındaki ilk örnekleri askerî mühendislik okulları olan “Mühendishâne-i Bahr-i Hümâyûn” ve “Mühendishâne-i Berrî-i Hümâyûn”dur. Tanzimat Fermanı’nın ilan edilmesi ile peyderpey İdadî (lise), rüşdiye (ortaokul) ve ibtidai (ilkokul) okulları açılmış ve mühendishânelere hazır öğrenciler yetiştirilmiş ve dolayısıyla 19. yüzyılın ikinci yarısından itibaren bu okullardan çok sayıda başarılı mühendis mezun olmuştur. 1874’te ilk sivil mühendislik okulu olan “Mülkiye Mühendis Mektebi”nin açılması ile beraber mühendislik eğitimi daha da gelişmeye başlamıştır. Bu okullardan mezun olan öğrenciler mühendislik işlerini yürütmekle beraber fen bilimlerindeki modern gelişmeleri yakından takip etmişler ve modern bilimsel gelişmelerin Osmanlı’ya ulaşmasına katkı sağlamışlardır. Mezun olan mühendislerin bir kısmı devlet tarafından yüksek öğrenim için başta Paris olmak üzere önemli Avrupa şehirlerine gönderilmişlerdir. Bu öğrenciler döndüklerinde devletin önemli mevkilerinde istihdam edilmişlerdir. Paris’e gönderilen öğrencilerden biri de Mühendis Mehmet Misbah’tır. 1911 yılında “Mühendis Mekteb-i Âlîsi”nden mezun olan Misbah aynı yıl Paris’e gönderilmiş, 1913’te geri döndüğünde mezun olduğu Mühendishâne’ye hoca olarak atanmış ve 1919 yılına kadar görevini sürdürmüştür. Analitik Geometri ve Düzlem Geometrisi derslerini üstlenen Misbah matematik alanında yirmiye yakın makale yayımlamıştır. Misbah’ın matematik felsefesine ve matematik eğitimine dair görüşleri bu makalenin konusunu oluşturmaktadır. Bu amaçla Misbah’ın Genç Mühendis dergisinde yayımlanan “Felsefe-i Riyâziyyât” isimli makalesi ve Mesâil-i Hendesiye isimli kitabın ilk cildine yazdığı “Mukaddime (Önsöz)”, matematik felsefesi ve matematik eğitimi bağlamında analiz edilmiştir. Misbah mühendishane’de öğrenci olduğu yıllarda Genç Mühendis isimli dergiye 1910-1911 yılları arasında on altı tane yazı göndermiştir. Misbah, Genç Mühendis isimli derginin 1326 Kanûn-i Sânî/Ocak 1911 tarihli 36. sayısının 5-8 sayfaları arasında el yazması şeklinde yayımladığı “Felsefe-i Riyâziyyât” isimli makalesinde matematik felsefesine dair görüşlerini açıklamıştır. Matematiğin tanımlarından yola çıkarak matematiğin ontolojisine ve epistemolojine dair görüşler beyan eden Misbah’ın isim zikretmeden Platon’un ve Aristoteles’in görüşlerinden faydalandığı tespit edilmiştir. Fakat 19. yüzyılda geliştirilen Mantıkçılık, Formalizm ve Sezgicilik gibi güncel matematik felsefesi yaklaşımlarından makalede söz edilmemiştir. Oysa söz konusu dönemde bazı Osmanlı aydınları Öklid-dışı geometrilere ve felsefî sonuçlarına dair bilgilendirici seminerler düzenlemişler ve makaleler yayımlamışlardır. Misbah’ın mühendishâneden arkadaşı olan Muhittin Sırrı Şamlı 1911’de Mesâil-i Hendesiye isimli iki ciltten oluşan bir kitap kaleme almış ve Misbah’tan da kitabın birinci cildine önsöz yazmasını istemiştir. Misbah önsözde matematik eğitimine dair görüşlerini dile getirmiştir. Matematiğin öğretiminde ispatın yer alması gerektiğini ve ispatların bizzat öğrenciler tarafından yapılmasının “muhakeme” becerisini geliştireceğini vurgulamıştır. Misbah’ın ifadelerinden matematik eğitiminde “öğrenci merkezli” bir anlayışa sahip olduğu tespit edilmiştir.

Abstract:
Mehmet Akif Ersoy ve Muhammed İkbal, aynı tarihlerde fakat farklı coğrafyalarda yaşamış iki Müslüman şairdir. Mehmet Akif Ersoy 1936’da, Muhammed İkbal 1938’de vefat etmiştir. Her ikisi de aynı fikirde toplumun geleceği adına aynı ıstırabı paylaşan fikir adamlarıdır. Gönülleri istiklal ve hürriyet aşkıyla tutuşan bu iki şair, aynı zamanda coşkun ruhlara ve iman dolu kalplere de tercüman olmuştur. Bu iki şairin bir diğer ortak yanı ise gençliğe verdikleri önemdir. İkbal, Cavidname adlı eserinin sonunda neşredilen Hitâb be Câvîd adlı hitabede, oğlu Cavid’in şahsında gençliğe rehber olmaya çalışır. Hitâb be Câvîd 136 beyitten oluşan nasihatname türünde yazılmış müstakil bir eserdir. Klasik şiir geleneğinde siyasetname, hikmet, pend-name, vasiyetname ve ibret-name gibi isimlerle anılan nasihat kültürünün devamı niteliğinde bir eserdir. Aynı şekilde Âkif de Safahat’ın altıncı cildinde yer alan Asım adlı şiirinde, Asım’ın şahsında genç nesle yol gösterme gayretindedir. Müstakil bir şiir olan Asım, mesnevi nazım şeklinde kaleme alınmış, manzum hikâyedir. Bu çalışmada İkbal ve Âkif’in, Hitâb be Câvîd ve Asım adlı eserlerinden hareketle, genç nesle tavsiyeleri karşılaştırmalı olarak ele alınmıştır. Yaşadıkları coğrafyalar farklı olsa da aynı medeniyetin bireyi olan Akif ve İkbal genelde aynı görüşleri dile getirmiştir. İkbal, klasik şiir üslûbunu benimsemiş, nasihatlerini de bu yönde sunmuş ve bireysel değerler eğitimi esas alan bir yol çizmiştir. Akif ise şeklen olarak klasik olmakla birlikte, üslup ve yöntem açısından modern bir yol benimsemiş, bireyi toplumun bir parçası olarak görmüş ve değerler eğitimi toplumsal merkezli bir gelişim olarak düşünmüştür.

Asker ve eczacı olan Avanzade Mehmed Süleyman 19-20. asrın en üretken yazarlarındandır. Hedef kitlesi genel okuyucu olan yazar edebiyat, sağlık ve ahlak kitapları yanında okültist metinler de kaleme almıştır. Bunlardan biri; Ulûm-ı Hafiyeden Âyine ile Keşf-i İstikbâl (Gizli İlimlerden Ayna ile Geleceğin Keşfi)’dir. Eser yalnızca ayna ile değil çeşitli fal yöntemleri, tefeül ve istihare ile geleceğin öğrenilmesi usullerini de anlatmaktadır. İnsanların nereden gelip nereye gittiklerini merak ettiğini düşünen Avanzade genellikle geleceğin daha çok merak edildiğini, bu sebeple çeşitli vasıtalar aracılığıyla geleceğin öğrenilmesi için büyü ve fal pratiklerinin ortaya çıktığını düşünür. Bu eserinde ise söz konusu usullerden bazılarına yer verdiğini söyler. 20. yüzyılın başında İstanbul’da pek çok popüler fal kitabının basıldığı bilinmektedir. Herhangi bir kalıcılığı olmayan ve kısa sürede unutulan bu metinler hem fal kültürüne hem de dönemin sosyal hayatına dair kayda değer bilgiler içermektedir. Yazar, fal ile ilgili tarihi bilgilerle ve çeşitli hükümdarların fal merakına dair anekdotlarla zenginleştirdiği eserinde, falın hangi amaçlarla bakıldığına dikkat çekmektedir. Âşıklar sevgili tarafından bir karşılık görüp görmeyeceklerini, genç kızlar ise kiminle nişanlanacaklarını öğrenebilmek için fala başvurur ki bu husus hâlâ geçerlidir. Dikkat çeken başka bir husus ise sosyal medya aygıtlarının henüz yaygınlaşmadığı o dönemde uzaktaki insanların sıhhat ve afiyetlerine dair merakların fal yoluyla giderilmeye çalışılmasıdır. Bu durum falın yalnızca bir keyif aracı değil aynı zamanda bir ihtiyaç olduğunu göstermektedir. Bu çalışmada Avanzade’nin söz konusu eseri tanıtılmış ve Latin harfleriyle okuyucuya sunulmuştur.

Considering that energy consumption is constantly increasing along with a gradual decrease in energy resource reserves, energy is the most current and pending issue in today's world. Several academic studies have been conducted on the efficient use of energy resources. For this purpose, the idea of "smart building" has been proposed to use energy more efficiently in buildings. With building automation systems, which are indispensable elements of these smart buildings, the energy consumption of buildings can be greatly reduced. In this study, the lighting efficiency of a building automation system built at a training point in the Marmara University Mehmet Genç campus, Vocational School of Technical Sciences, was examined. The Sauter Vision Center software program was used at the observation center of the system. The results were analyzed in terms of efficiency by comparing the energy values in certain time periods obtained from the program with the energy values calculated in full use. The analysis yielded a lighting efficiency value of 54%, which was found to exceed the values reported in the literature.

Tahtacı Alevileri günümüzde Akdeniz, Ege ve Marmara bölgelerinde dağınık halde yaşamaktadır. Uzun yıllar ağaç işçiliğiyle uğraştıkları için “Tahtacı” olarak adlandırılan bu Alevi zümre, diğer Alevi toplulukları gibi ocak merkezli bir yapılanma gösterir. Tahtacı Alevileri Yanyatır ve Hacı Emirli olmak üzere iki ocağa bağlıdır. Yanyatır Ocağı’nın merkezi İzmir Narlıdere, Hacı Emirli Ocağı’nın merkezi Aydın Kızılcapınar köyüdür. Bu iki ocak içerisinde en fazla talibe sahip olan Yanyatır Ocağı’dır. Çalışma alanımız olan Balıkesir Edremit’e bağlı Doyran köyü ve Kazdağlarındaki Tahtacı yerleşim birimlerinde Yanyatır Ocağı’na bağlı dede ve talip toplulukları yaşamaktadır. Sözlü tarihe göre Tahtacı Alevileri eski adı İda Dağı olan Kazdağlarına Fatih Sultan Mehmet’in davetiyle gelmiştir. Fatih Sultan Mehmet İstanbul’u almak ve Midilli’de çıkan isyanları bastırmak amacıyla kullanacağı gemilerin kerestelerini biçtirmek için Toros Dağlarındaki Tahtacı Alevilerini bölgeye iskân ettirmiştir. 16. yüzyılda bölgeye iskân edilen Tahtacı Alevileri, günümüzde de Kazdağlarında yaşamaktadırlar. 1950’lili yıllara kadar ağaç işçiliği ile hayatlarını sürdüren topluluk üyeleri son elli yılda büyük oranda orman işçiliğini bırakarak farklı mesleklerle hayatlarını sürdürmektedirler.
 Tahtacı Alevi topluluklar, kapalı toplum özelliğini en güçlü muhafaza eden Alevi zümrelerdendir. Kazdağlarında yaşayan Tahtacı toplulukları inanç ve ritüel dünyaları bağlamında kapalı toplum olarak varlıklarını sürdürmektedirler. Çalışmamıza konu olan Doyran köyünde de geleneksel Tahtacı kültürüne ait inanç ve pratikler yaşatılmakta ve genç kuşaklara aktarılmaktadır. Makalede sunduğumuz veriler, Doyran köyü örnekleminde Geleneksel Tahtacı Alevi kültürüne ait önemli bir birikimi gözler önüne sermektedir. Doyran köyü özelinde toplanan veriler kendisi de Doyran köylü olan Hasan Taşçinten tarafından uzun yıllar alan derleme çalışmaları ile elde edilmiştir. Çalışmamızda Hasan Taşçinten’in topladığı derleme verilerinin ritüel ve inanç pratikleri ile ilgili olanları tasnif edilmiş, yorum ve analizlerle değerlendirilmiştir.

Abstract:
The aim of the study it to reconstruct the European standard for the protection of patients’ lives in its substantive and procedural aspects. In the case-law of the bodies of the system of the Convention for the Protection of Human Rights and Fundamental Freedoms, the scope of the state authorities’ substantive and procedural obligation to protect the right to life in the health care system was defined for the first time by the European Commission of Human Rights in the decision of 22 May 1995 in Mehmet Işıltan v. Turkey, and then repeated in the case-law of the reformed Court in the decision on the admissibility in Powell v. United Kingdom. The study of the European standard for the protection of patients’ lives traces its history, from Mehmet Işıltan v. Turkey and Powell v. United Kingdom; through developments of the meaning of its substantive limb, as illustrated by Mehmet and Bekir Senturk v. Turkey, Asiye Genc v. Turkey, Aydogdu v. Turkey, and Elena Cojocaru v. Romania; to developments of the meaning of its procedural limb, as exemplified by Calvelli and Ciglio v. Italy, Wojciech Byrzykowski v. Poland, Šilih v. Slovenia, and Gray v. Germany; and finally covers the Court’s attempt to sum up its previous approach to the European standard for the protection of patients’ lives, as expressed in the case of Lopes de Sousa Fernandes v. Portugal.

BackgroundFamilial Mediterranean Fever (FMF) is a rare autosomal recessive illness of periodic fever syndrome that primarily affects the Mediterranean population [1]. No single specific diagnostic test exists to accurately diagnose FMF. Testing of the MEFV gene is the primary investigation of choice to support a diagnosis of FMF [2]. Little is known regarding MEFV testing within Australian cohorts.ObjectivesWe aim to describe the pattern of ordering for FMF genetic testing via the ordering of MEFV gene, and the clinical circumstances surrounding its request in a large Australian referral centre.MethodsWe retrospectively identified all MEFV genetic testing ordered within a large public referral centre over a five-year period. We collected information regarding the clinical scenario indicating genetic testing, presenting symptoms and adherence to Tel HaShomer diagnostic criteria as well as whether the genetic testing impacted the final diagnosis.Results64 MEFV genetic testing were ordered through the five-year period, seven were within the paediatric population. The median age of the cohort was 37, 56.3% were male sex. There is a median of five flares of FMF prior to MEFV testing with median time from last flare being two months. 53.1% of the tests were ordered in an inpatient setting, rest were through outpatient. Rheumatology ordered the most MEFV genetic testing in 67.2% of the cohort, followed by Infectious Diseases in 10.9% and General Medicine in 7.8% of the cohort. 79.7% of the participants satisfied the Tel HaShomer criteria for clinical diagnosis of FMF prior to genetic testing. MEFV genetic testing provided a positive diagnostic result in 21.9% (14 participants), there were seven homozygotes, seven compound heterozygotes and eight heterozygotes. Only one of the 14 participants with a positive MEFV test did not satisfy the clinical diagnostic criteria. A total of 36 mutated alleles were identified and M694V was the most frequent mutation appearing in 47.2% of the mutations identified. Peritonitis was the most common presentation in 67.2% of the cohort. One participant had amyloidosis as their presentation. 43.8% of the cohort was commenced on colchicine prior to genetic testing.ConclusionMEFV testing is most commonly ordered by Rheumatology than other specialities. Bu izdivaçtan Şerife Demir adında bir kızları dünyaya gelmiş, ancak kızları genç yaşta vefat etmiştir. Belgede, kızının ölümünden sonra annesinin sahip olduğu mülklerin bir kısmının miras yoluyla kendisine geçtiği açıkça belirtilmiştir. Çalışma, pek de tanınmamış bir Osmanlı subay, seyyah, yazar, mütercim ve edebiyat öğretmeni olup Cumhuriyet döneminde de eser vermeyi sürdürmüş olan Mehmed Enisî'nin hayatı ve eserleri hakkında yazındaki malumatı düzelterek genişletmektedir. The study corrects and expands the information in the literature about the life and works of Mehmed Enisî, a lesser-known Ottoman officer, traveler, writer, translator, and literature teacher who continued his creative output during the Republican period. Thus, the relations between Enisî, a positivist liberal, translating and printing an anti-socialist literary fiction from English into Ottoman as early as 1924, and his perception of socialism as a threat to his ideology and way of life are interpreted. In addition, in the study, the text that Enisî translated from English into Ottoman Turkish was translated into Turkish with Latin letters and made available to researchers.

The Arabidopsis mutant rcd1 is tolerant to methyl viologen (MV). MV enhances the Mehler reaction, i.e. electron transfer from Photosystem I (PSI) to O 2 , generating reactive oxygen species (ROS) in the chloroplast. To study the MV tolerance of rcd1 , we first addressed chloroplast thiol redox enzymes potentially implicated in ROS scavenging. NADPH-thioredoxin oxidoreductase type C (NTRC) was more reduced in rcd1 . NTRC contributed to the photosynthetic and metabolic phenotypes of rcd1 , but did not determine its MV tolerance. We next tested rcd1 for alterations in the Mehler reaction. In rcd1 , but not in the wild type, the PSI-to-MV electron transfer was abolished by hypoxic atmosphere. A characteristic feature of rcd1 is constitutive expression of mitochondrial dysfunction stimulon (MDS) genes that affect mitochondrial respiration. Similarly to rcd1 , in other MDS-overexpressing plants hypoxia also inhibited the PSI-to-MV electron transfer. One possible explanation is that the MDS gene products may affect the Mehler reaction by altering the availability of O 2 . In green tissues, this putative effect is masked by photosynthetic O 2 evolution. However, O 2 evolution was rapidly suppressed in MV-treated plants. Transcriptomic meta-analysis indicated that MDS gene expression is linked to hypoxic response not only under MV, but also in standard growth conditions. Abstract Epithelial mesenchymal transition (EMT) describes a process by which the epithelial cells acquire mesenchymal properties associated with increased migration, invasion, and resistance to therapy. Targeting the intercellular communications and molecular changes that enable EMT is a rational therapeutic approach in PDAC. Our group has previously reported on KPC-Luc PDAC orthotopic mouse models treated with 1. Sham, 2. paricalcitol plus hydroxychloroquine (PH), 3. gemcitabine (G), and 4. GPH. Single cell RNA sequence (scRNASeq) was performed and analyzed using hdWGCNA (high dimensional Weighted Gene Co-expression Network Analysis) and GSEA (Gene Set Enrichment Analysis) to plot and determine intracellular interactions as well as gene expression changes in various treatment conditions. GPH treatment reduces EMT, which is associated with the downregulation of the essential gene fibronectin (Fn1). Collagen and Fn1 pathways co-expression decreases in GPH-treated KPC-Luc tumors. Cancer-associated fibroblasts (CAFs) appear dominant in Collagen signaling, whereas macrophages mediate Fn1 signaling. GPH treatment reduces the expression interaction strength between ligands and receptors (Collagen-Integrin and Fn1-Cd44 or Fn1-Sdc4) compared to sham, PH, and G. This comprehensive study of cell-cell communication and molecular changes during therapy offers a deep understanding of the molecular underpinnings of PDAC-EMT and metastasis and identifies possible targets for GPH. Abstract Circulating Tumor Cells (CTCs) hold the promise of comprehensive yet noninvasive molecular characterization of cancer cells during the course of tumor evolution and progression. To dissect the epigenetic profile of prostate CTCs, we undertook paired DNA methylation and RNA sequencing of single CTCs isolated from blood specimens of patients with prostate cancer. Across the genome, our study identified 40 core Partial Methylation Domains (PMDs) that arise early in tumorigenesis and are shared by all individual CTCs across multiple patients. Key among these is a single genomic locus harboring multiple interferon-inducible genes along with all the CD1 gene family members, implicated in innate immunity. Based on mouse models, we propose that early biallelic silencing of multiple genes residing within a single PMD may contribute to the escape from immune surveillance in prostate and potentially other cancers. Studies of CTC heterogeneity within individual patients are limited by the very small number of cells that can be isolated from standard 10ml blood volumes. To enable such analyses, we applied a new high throughput microfluidic platform, capable of processing an entire leukapheresis product, interrogating 2-3 L of blood volume equivalent. Applying this technology to patients with metastatic prostate cancer, we successfully enriched thousands of CTCs from individual cases, enabling the characterization of intra-patient heterogeneity at the level of cell morphology and marker expression, chromosome copy number variation and exome-wide genotyping, and transcriptionally-defined tumor cell subpopulations. Such high-volume microfluidic enrichment of CTCs constitutes a new dimension in liquid biopsies. Abstract Cancer-associated alterations in DNA methylation include hypermethylation of focal CpG islands within promoters, as well as hypomethylation of large stretches of gene-poor chromosomal domains, so called Partially Methylated Domains (PMDs). While CpG island hypermethylation is associated with silencing of tumor suppressor genes, the functional consequences of hypomethylation are uncertain. PMDs may be cause or consequence of cancer cell proliferation, and they have been associated with transcriptional de-repression of repetitive sequences, as well as histone modification-triggered silencing of rare protein encoding genes within these domains. In studying Circulating Tumor Cells (CTCs) isolated from blood specimens of patients with prostate cancer, we mapped PMDs at the single cell level, identifying a set of 40 “core PMDs” that are shared across cancer cells, within and across different individuals, as well as in prostate cancer cell lines. By analyzing single nuclei from micro-dissected early-stage prostate tumors, we find that hypomethylation of these core PMDs occurs early during prostate tumorigenesis. Core PMDs are highly enriched for immune-related genes, many of which are co-located within a single chromosomal locus targeted for hypomethylation-associated silencing. Nested between adjacent PMDs, we also identify domains that have retained DNA methylation (Preserved Methylation Islands; PMIs), which have sharply demarcated borders, spanning one or a small number of genes. In contrast to PMDs, PMIs are enriched for cell proliferation-associated genes. Together, these observations suggest that hypomethylation-associated silencing of immune-related genes, with sparing of adjacent proliferation-associated genes, is an early event in prostate cancer development. Abstract Introduction: With the accumulation of large-scale pharmacogenomic data such as whole-genome RNA sequencing, copy number and mutation profiles for tens of thousands of samples, further screened with thousands of small molecules and other perturbagens, the question arises how to best leverage partially overlapping datasets generated at different facilities. Notably, Haibe-Kains et al. observed discordance in state-of-the-art pharmacogenomic repositories. As research groups across the world continue to generate drug screens of variable size and quality, the need for approaches that can learn from such partially overlapping experiments and improve the signal to noise ratio emerges with increasing importance. Methods: A previously published Bayesian group factor analysis model was shown to outperform other approaches in predicting drug response and identifying gene signature from pharmacogenomic datasets particularly by leveraging shared information for a given gene across multiple omics assays. Here, we applied the same model in a similar fashion to now learn from shared observations for the same drug across multiple partially overlapping and noisy small molecule screens. We integrate gene expression, mutation and drug response data from the two largest pan-cancer repositories at the Broad and Sanger Institutes, respectively. We train joint models on partially overlapping data from both. We evaluate our performance in three ways: 1) we test if the joint model improves the prioritization of known consensus biomarkers for the drugs shared between the two cohorts; 2) we test if the joint model improves the recapitulation of shared drug mechanism of action as compared to the single dataset models; 3) we evaluate the joint models with respect to pathway enrichment as compared to the single dataset models. Results: We evaluated the performance of our joint model for 5 drugs shared between the 2 resources: selumetinib, tanespimycin, nutlin 3A, mirdametinib and PLX4720. First, we show that training joint models on partially overlapping pharmacogenomic datasets can overall improve gene signature identification by improving the ranks of known consensus biomarkers. Second, we show that the joint model learns a latent representation of the drugs that better recapitulates the underlying known mechanisms of action for the three serine/threonine kinase inhibitors. Finally, we show that the joint model achieved improved pathway enrichment results for the targeted MAPK/ERK signaling pathway. Conclusions: We present an application of a Bayesian group factor analysis model, where we employ a drug-centric prior to transfer information about drugs screened in multiple datasets. Six Helicobacter -like isolates were recovered from 15 gastric mucosa samples of red foxes (Vulpes vulpes) shot by hunters in the surroundings of Ljubljana, Slovenia. Gram-negative, tightly coiled, intensely motile, 7–15 µm long and ≤1 µm wide bacteria grew on the biphasic blood agar plates. By using a genus-specific polymerase chain reaction (PCR), all isolates were confirmed as Helicobacter sp. and subsequently subjected to whole-genome sequencing (WGS). Five isolates showed a genome-wide average nucleotide identity (ANI) value of <95 % to the previously described Helicobacter species and one isolate was classified as Helicobacter felis . In the five unidentified isolates, the 16S rRNA gene sequence similarity to the type strains of all Helicobacter species ranged from 98.6 to 98.9 %. Their taxonomic status was established using a polyphasic taxonomic approach comprising the core genome-based phylogeny, morphological and phenotypic characteristics, including an analysis of matrix-assisted laser desorption/ionisation time-of-flight (MALDI-TOF) mass spectra. Phylogeny revealed the existence of three novel and well-supported clusters, with Helicobacter bizzozeronii and Helicobacter baculiformis being the most closely related species. The isolates also differed from the previously described species in their MALDI-TOF profiles and some biochemical characteristics. In conclusion, the data presented herein indicate that the obtained isolates, excluding H. felis isolate, represent three novel Helicobacter species, for which the names Helicobacter labacensis sp. nov., Helicobacter mehlei sp. nov., and Helicobacter vulpis sp. nov. are proposed, with isolates L9T (=DSM 108823T=CRBIP 111719T), L15T (=DSM 108730T=CCUG 72910T) and L2T (=DSM 108727T=CCUG 72909T) as type strains, respectively.

Abstract Introduction: The discovery of immune checkpoint inhibitors has revolutionized metastatic renal cell carcinoma (RCC) treatment. However, in patients with RCC brain metastases, response rates are low and survival outcomes poor. To understand the tumor microenvironmental differences between primary kidney tumors, extracranial metastases, and brain metastases, we developed a detailed single-cell atlas of RCC brain metastases along with their matched extracranial and primary tumors. Methods: We performed single-nucleus RNA-seq on 27 samples (nearly 200,000 cells) from RCC patients; samples included 14 brain metastases, 8 matched primary kidney tumors, and 5 matched extracranial metastases. We performed multiplex IHC to validate selected transcriptomic findings. We used Nanostring CosMx 960-plex RNA spatial molecular imaging technique on selected samples to validate cellular interactions in a spatial context. Results: We established a multi-tissue single-cell atlas of RCC brain metastases by identifying 9 major and 37 minor malignant, immune, and stromal cell clusters. Brain metastases had higher neuronal and glial cells interacting with immune and tumor cells. Brain metastasis tumor cells were also transcriptomically reprogrammed to adapt to the brain microenvironment through enrichment of MYC targets, MTORC1 signaling, epithelial-mesenchymal transition, fatty-acid metabolism, oxidative phosphorylation, and reactive oxygen species pathways. Moreover, cell-to-cell communication and downstream target gene expression analyses showed that brain metastasis tumor cells expressed ligands and receptors that induce tumor cell proliferation in both autocrine and paracrine fashions. Among T-cell populations, we found fewer proliferating cytotoxic T lymphocytes in the brain than in other sites. Moreover, T cells in brain metastases expressed higher levels of several targetable inhibitory checkpoints than did extracranial metastases. In addition, we found that naïve/memory T cells in brain metastases were a favorable prognostic marker for overall survival after craniotomy. Our characterization of myeloid cell populations across the 3 disease sites found fewer dendritic cells and monocytes in the brain compared to other sites. Macrophages in brain metastases more highly expressed an M2 immunosuppressive gene signature than did those in primary RCC tumors. Conclusion: Our findings from the largest single-cell atlas of RCC brain metastases with matched primary and extracranial metastases suggest several unique targetable, immunosuppressive biological mechanisms in the brain microenvironment. These results provide a foundation for a deeper understanding of RCC brain metastasis biology and can serve as a resource for the scientific community to further explore therapeutically targetable tumor and immune-related mechanisms. Abstract Background: Endocrine therapy plus a CDK4/6 inhibitor (ET+CDK4/6i) is the mainstay treatment in first-line ER+/HER2- mBC; however, a subset of patients is unable to tolerate CDK4/6i, and resistance to ET arises. Intrinsic resistance mechanisms include alterations in the PI3K/AKT/mTOR or cell cycle pathways; acquired resistance mechanisms include estrogen receptor gene alpha (ESR1)-mut developing during ET in mBC. In EMERALD, single-agent elacestrant significantly prolonged progression-free survival (PFS) vs standard-of-care (SOC) ET and was associated with a manageable safety profile in patients with ER+/HER2- advanced or mBC previously treated with ET+CDK4/6i, leading to its approval as the first clinically available oral SERD. Elacestrant significantly reduced the risk of progression or death compared with SOC ET by 30% in the overall population (HR = 0.70; 95% CI, 0.55 to 0.88; P = 0.002) and by 45% in patients with ESR1-mutated tumors (HR = 0.55; 95% CI, 0.39 to 0.77; P = 0.0005) [Bidard, 2022]. To provide an option for patients unable to receive CDK4/6i and address resistance, the ELCIN trial aims to evaluate the efficacy and safety of elacestrant in patients with ER+/HER2- mBC who received prior ET but did not receive a prior CDK4/6i in the metastatic setting. Methods: ELCIN (NCT05596409) is a single-arm phase 2 trial. Eligible patients are women or men with ER+/HER2− mBC who received 1-2 prior ET and no prior CDK4/6i or chemotherapy in the metastatic setting. Patients must have measurable disease per RECIST v1.1 or ≥1 lytic or mainly lytic bone lesion (in patients with bone disease only), ECOG PS ≤1, and no active or newly diagnosed central nervous system metastases. Patients will receive elacestrant 345 mg (400 mg elacestrant hydrochloride) once daily. The primary endpoint is investigator-assessed progression-free survival (PFS). Secondary endpoints are objective response rate (ORR), duration of response (DoR), clinical benefit rate (CBR), overall survival (OS), patient-reported outcomes (PROs), health-related quality of life (HRQoL), and safety. Exploratory endpoints include assessing elacestrant efficacy according to ESR1-mutation status at baseline, assessing changes in biomarkers, including allele mutation frequencies in cell-free nucleic acids (cfNAs) in blood and the relationship between efficacy endpoints and allele mutation frequencies (at baseline and post-baseline). Abstract Introduction: Pulmonary thromboembolism (PTE) is a significant clinical issue associated with high morbidity and mortality. Risk factors for thromboembolic events include immobilization, myocardial infarction, cerebrovascular accident, surgery, and trauma. Iron deficiency anemia (IDA) can cause reactive thrombocytosis, increasing thromboembolic risk. This study presents a 30-year-old female admitted for PTE, possibly secondary to IDA. Case Presentation: A 30-year-old female with IDA and heavy menstrual bleeding presented with right-sided chest pain and shortness of breath. Her pain, exacerbated by inspiration, and relieved by leaning forward, had worsened over four days. She also reported dizziness and agonal breathing. On arrival, her vitals included a blood pressure of 95/62 mmHg, heart rate of 110 bpm, respiratory rate of 22 breaths/min, and a temperature of 37°C. Physical examination revealed tachycardia, tachypnea, and diminished breath sounds in the right lower lung field. Laboratory tests showed a hemoglobin level of 4.6 g/dL and a platelet count of 745 × 10^9/L, low ferritin, low serum Iron, and high total iron-binding capacity (TIBC). A CT pulmonary angiogram revealed a wedge-shaped infarct in the right lower lobe, consistent with pulmonary embolism. CT abdomen/pelvis showed a thrombus in the right iliac vein, indicative of deep vein thrombosis. The echocardiogram showed left ventricular ejection fraction >55% with no right chamber strain. A pelvic ultrasound revealed no significant pathology. Treatment included heparin and blood transfusions, increasing hemoglobin to 9.0 g/dL. Hemolytic workup including prothrombin gene abnormalities, Factor V Leiden mutation, and protein C activity was negative. However, Protein S activity was low, and PTT-la positive, implying that the patient may be positive for lupus anticoagulant, later ruled out by serial antibody tests.Subsequently, the patient was discharged home on Eliquis and Ferrous sulfate with outpatient hematology follow-up. Discussion: The association between venous thromboembolism and IDA can be explained by several mechanisms. One study suggests that iron deficiency causes hypercoagulability through reactive thrombocytosis, increased oxidative stress, and altered blood viscosity. Another hypothesis is that the hyperdynamic state in anemic patients leads to vascular endothelial injury due to shear forces. This case highlights the importance of considering IDA as a potential risk factor for thromboembolism, especially in patients without other evident prothrombotic conditions. Conclusion: Iron deficiency may be an underestimated risk factor for thromboembolic events. This case underscores the need for a comprehensive workup for VTE that includes assessing for IDA. Iron replacement therapy could serve as an effective preventive strategy in patients with a history of IDA and those at risk for thromboembolism. Further research is warranted to explore the mechanisms linking IDA with increased thrombotic risk and to establish appropriate preventive measures. In the phase 3 EMERALD trial, Ela significantly prolonged PFS vs standard-of-care (SOC) ET and was associated with a manageable safety profile in pts with ER+/HER2- mBC previously treated with ET+CDK4/6i, leading to its approval as the first clinically available oral SERD. Ela significantly reduced the risk of progression or death vs SOC ET by 30% in the overall population (HR 0.70; 95% CI 0.55-0.88; P=0.002) and by 45% in pts with ESR1-mut tumors (HR 0.55; 95% CI 0.39-0.77; P=0.0005) [Bidard, 2022]. Preclinical studies demonstrated that Ela is equally active in both in vitro and in vivo models of ER+/HER2- breast cancer, regardless of prior exposure to CDK4/6i. Based on preclinical models and clinical efficacy data, Ela may improve clinical outcomes in CDK4/6i-naïve pts and provide a convenient all-oral treatment option if combined with CDK4/6i. Abstract In multiple myeloma (MM), the proteasome inhibitor bortezomib (BTZ) triggers immunogenic cell death (ICD), a critical mechanism by which the drug stimulates an anti-MM immune response. During ICD, the dying cancer cell releases danger signals like calreticulin (CRT), which attract dendritic cells (DCs) to engulf the dying cell and present its antigens to T cells. Cancer cells may employ several strategies to restrain the immunogenic outcome, including interfering with the exposure of CRT on the cell surface. Such resistance mechanisms hinder immune activation and might be responsible for the relapse and poor clinical outcomes observed in MM patients. To uncover resistance mechanisms, we integrated the CRT interactome after ICD induction with transcriptomic data from the IFM/DFCI dataset (n=360) and found that GABARAP interacts with CRT and its lower expression was associated with worse clinical outcomes (p=0.0017). We observed a strong correlation between GABARAP protein levels and the intensity of CRT exposure after BTZ treatment in 10 MM cell lines (R2 : 0.62). Consequently, GABARAP KO in 4 ICD-sensitive cell lines impaired the exposure of CRT, thus diminishing ICD, as assessed by MM cell phagocytosis by DCs and T cell activation both in vitro and in vivo in an immunocompetent model. ICD was restored by add-back experiments using recombinant CRT or GABARAP overexpression. This mechanism was also confirmed in patients, by the single-cell RNA-seq analysis of 80 MM patients in which we found a positive correlation between the expression of GABARAP and the ICD signature. Interestingly, low intratumor expression of GABARAP was associated with a lower T cell infiltration both at the single-cell level and by IHC analysis of bone marrow patient samples (n=10). Importantly, GABARAP gene locus is on chr17p13.1, therefore, while this mechanism is general for all MM cells, it appears especially relevant for those carrying a high-risk deletion of the 17p chromosome. Mechanistically, proteomic analysis, confocal and transmission electron microscopy showed that GABARAP loss significantly altered the morphology of the Golgi apparatus, causing alterations in the vesicular transport systems and autophagy. This effectively stopped CRT transport to the surface, thus hindering ICD. By combining BTZ with autophagy inducers, we could restore vesicular transport of CRT to the cell surface in GABARAP low MM cells and their phagocytosis by DCs. We propose a model by which low levels of GABARAP compromise the vesicular transport of CRT exposure. This impairment results in the inhibition of tumor immunogenicity and the overall outcome of ICD. Consequently, combining an ICD-inducing agent, like bortezomib, with an autophagy inducer, like rapamycin, may improve patient outcomes in MM, where del(17p) is common and leads to unfavorable outcomes. Abstract Histone deacetylase inhibitors (HDACi) can modulate the acetylation status of proteins, influencing the genomic instability exhibited by cancer cells. Poly (ADP ribose) polymerase (PARP) inhibitors (PARPi) have a direct effect on protein poly (ADP-ribosyl)ation, which is important for DNA repair. Decitabine is a nucleoside cytidine analogue, which when phosphorylated gets incorporated into the growing DNA strand, inhibiting methylation and inducing DNA damage by inactivating and trapping DNA methyltransferase on the DNA, thereby activating transcriptionally silenced DNA loci. We explored various combinations of HDACi and PARPi +/- decitabine (hypomethylating agent) in pancreatic cancer cell lines BxPC-3 and PL45 (wild-type BRCA1 and BRCA2) and Capan-1 (mutated BRCA2). Colony formation and drug-mediated inhibition of cell proliferation in pancreatic cancer cell lines and comparison of the effects of each drug combination with the effect of the individual drugs are shown in Table. The combination of HDACi (panobinostat or vorinostat) with PARPi (talazoparib or olaparib) resulted in synergistic cytotoxicity in all cell lines tested. The addition of decitabine further increased the synergistic cytotoxicity noted with HDACi and PARPi, triggering apoptosis (evidenced by increased cleavage of caspase 3 and PARP1). The 3-drug combination treatments (vorinostat, talazoparib, and decitabine; vorinostat, olaparib, and decitabine; panobinostat, talazoparib, and decitabine; panobinostat, olaparib, and decitabine) induced more DNA damage (increased phosphorylation of histone 2AX) than the individual drugs and impaired the DNA repair pathways (decreased levels of ATM, BRCA1, and ATRX proteins). The 3-drug combinations also altered the epigenetic regulation of gene expression (NuRD complex subunits, reduced levels). This is the first study to demonstrate synergistic interactions between the aforementioned agents in pancreatic cancer cell lines and provides preclinical data to design individualized therapeutic approaches with the potential to improve pancreatic cancer treatment outcomes. Abstract Introduction: In HPV- HNSCC, limited durable clinical responses to PD1/PD-L1 blockade may be due to diminished effector cytolytic activity and clonal diversity.1-4 mRNA-4157 is a novel mRNA-based INT that encodes up to 34 neoantigens inducing specific antitumor T-cell activation; durable clinical responses with pembro have been shown in melanoma.5 We assessed INT + pembro in unresectable, metastatic HPV- HNSCC in the Phase 1 mRNA-4157-P101/KEYNOTE-603 study (NCT03313778). Methods: Part C of this study enrolled ≥18-year-old patients (pts) with CPI-naïve, recurrent/metastatic HPV- HNSCC. Eligible pts received 200 mg pembro Q3W IV during a 6-week lead-in, then combined with 1 mg INT ≤9 doses Q3W IM, and until disease progression, unacceptable toxicity, or ≤35 total cycles of pembro. Objectives included safety, tolerability and preliminary clinical activity. Peripheral blood was collected longitudinally for both immunogenicity via IFN-γ ELISpot and ctDNA analyses. Next generation sequencing was performed on baseline tumor biopsies. Results: A total of 28 enrolled pts received pembro; 22 received INT + pembro. Most adverse events (AEs) related to INT were grade 1-2; most common were influenza-like illness, injection site pain, and pyrexia. No grade 4/5 AEs were related to INT. Three pts had grade 3 events (pyrexia, increased lipase, and decreased lymphocyte count) attributed to both INT and pembro. Six pts discontinued prior to INT (death/disease progression/deterioration). Response rate for INT + pembro was 27.3% (6/22; 95% CI, 10.7-50.2); disease control rate was 63.6% (14/22; 95% CI, 40.7-82.8) per RECIST V1.1 with best overall response of 2 (9.1%) complete, 4 (18.2%) partial response, and 8 (36.4%) stable disease at data cut-off (May 2023); with median follow-up of 38.4 weeks (range, 7.7-198.3). Median (95% CI) PFS and OS were 15.0 weeks (11.6-38.6) and 107.1 weeks (42.7-NE), respectively. Sustained de novo T-cell induction to targeted neoantigens was observed in 5/5 pts with available samples. A decrease in ctDNA levels post-baseline was observed in 3/4 responders (8 available samples). Differential gene expression from pretreatment tumor samples suggests increased inflamed T-cell-associated gene expression (Benjamini-Hochberg-adjusted descriptive p<0.05, for hallmark inflammatory response) in INT + pembro responders. Conclusions: INT + pembro had a manageable safety profile, showed evidence for activation of immune responses, and induced preliminary clinical responses in pts with HPV- HNSCC. Özel mektuplar aile, eş, dost, akraba, yakın arkadaş ve meslektaşlar arasında yazılan mektuplardır. Mektup türünün edebî anlamda değer kazanması; doğal, içten ve canlı olmasıyla ilişkilidir. Türk Edebiyatında mektup türü uzun bir geçmişe sahiptir. Ancak Batılı anlamda mektup türünün gelişimi Tanzimat Dönemi’nde olmuştur. Sanatçılar mektup yazmakla kalmamış, yazılan mektuplar, başka isimler tarafından bir araya getirilerek kitaplaştırılmıştır. Cumhuriyet döneminde de bu gelenek devam etmiş, tanınmış edebiyatçıların mektupları, kitap bütünlüğünde bir araya getirilmiştir. Bunlardan biri de Mehmet Kaplan’ın arkadaşı Âli Ölmezoğlu’na yazdığı mektuplardır. Zeynep Kerman ve İnci Enginün tarafından bir araya getirilen mektuplar, 1939 – 1944 yılları arasını kapsamaktadır. Tüm bu serüvenine şiirleri eşlik etmiştir. 17 Ağustos 1999'da aramızdan ayrılan Çınarlı, 1950'den 1980'e kadar Hisar Dergisi’nin yöneticiliğini ve baş yazarlığını üstlenmiştir. Sanata ve şiire duyduğu bağlılık, genç yaşlarda ortaya çıkmış, ilk şiirini 1943'te henüz öğrenci iken kaleme almıştır. Bu dönemde hece ölçüsüyle yazdığı şiirleri de bulunmaktadır. Abstract:
Many studies on Mehmet Akif Ersoy, who is also known as "Homeland Poet" and "National Poet" in Turkish literature, show that he is in a structure suitable for realism and idealism, and that he frequently deals with subjects in which social concepts are at the forefront. Mehmet Akif collected all his poems in his work called Safahat, which consists of seven books. Family, which constitutes the core of the society and is the most basic building block that keeps it alive, comes first among the social concepts that he values. In his works, where he analyzes the various situations of the members of the family institution in depth, Akif focuses on the "child" and "young" individuals in this institution and the "education" needs of the new generation created by them. Akif has an intense interest in children and young people, because the way to know and serve a society is to know and understand the child well. The periods in which the individual evolves from childhood to youth and from there to adulthood; It has a critical meaning for the child, family and society. Being aware of this, Akif has been in an effort to create the ideal youth and thus to create the ideal society by emphasizing the family institution, which he almost sanctified and defined as the "heavenly palace" in his works. In this compilation type study, which is based on literature review and literature analysis as a method, based on Mehmet Akif Ersoy's work named Safahat, the concepts of "child", "young", "family" and "education", which are considered important in the progress of society, were evaluated from Akif's perspective and the importance of these concepts was tried to be revealed. When Mehmet Akif Ersoy's point of view on the aforementioned social concepts is examined both from his own works and from the works made about him, it seems possible to say that these views still maintain their validity today and that there are suggestions that can be a solution to many current social and educational problems.

Siyasi kimliği ile milli mücadele tarihinde önemli bir yere sahip olan Mehmet Cevat Dursunoğlu (1892-1970), eğitimci kimliği ile de eğitim tarihimiz içerisinde önemli bir yere sahiptir. Yurt işgal altındayken Erzurum’da faaliyet gösteren Özel Albayrak İlkokulu’nda öğretmenlik yaparak eğitim çalışmalarına başlayan Mehmet Cevat Bey, İstiklal Harbi başarıya ulaştıktan sonra da Maarif Vekaleti bünyesinde çeşitli kademelerde görev yapmıştır. Bu kapsamda, öğretmenlik ve okul müdürlüğü görevlerinden sonra genç Türkiye Cumhuriyeti’nin eğitim sistemine yön veren Maarif Müfettişleri Kongresi, Üçüncü Heyet-i İlmiye Çalışması ve Birinci Milli Eğitim Şurası’na aktif olarak katılmış, İlköğretim ve Ortaöğretim Genel Müdürlüğü görevlerini yürütmüş, bu görevlerinden sonra Bakanlık Müfettişi olarak Berlin’de “Talebe Müfettişi” olarak görevlendirilmiş, söz konusu görevi yürütürken Atatürk’ün emri ile Avrupa’dan birçok ünlü ismin ülkemize gelerek eğitime katkı sağlamasında büyük rol oynamıştır. Yurda döndükten sonra (1935-1942) yılları arasında Milli Eğitim Bakanlığı Teftiş Heyet-i Reisliği görevini yürütmüş ve bu görevinden ayrıldıktan sonra siyasete atılmıştır. Milletvekilliği görevini yürütürken de TBMM Eğitim Komisyonu’nda görev yapmış ve eğitim ile ilgili düşüncelerini Meclis kürsüsünden dile getirmeye çalışmıştır. Bu çalışmada, Mehmet Cevat Dursunoğlu’nun eğitimci kimliği üzerinde durulmuş olup arşiv ve litaratür taraması yapılarak, bu konuda yazılmış eserler ve düzenlenmiş belgeler incelenip irdelenerek çalışma tamamlanmıştır.
