Abstract:
The recognition of Gregor Mendel's achievement in his study of hybridization was signalled by the ‘rediscovery’ papers of Hugo de Vries, Carl Correns and Erich Tschermak. The dates on which these papers were published are given in Table 1. The first of these—De Vries ‘Comptes rendus paper—was in French and made no mention of Mendel or his paper. The rest, led by De Vries’ Berichte paper, were in German and mentioned Mendel, giving the location of his paper. It has long been accepted that the first account of Mendel's work in English was given by the Cambridge zoologist, William Bateson, to an audience of Fellows of the Royal Horticultural Society in London on 8 May, 1900. This is based on two sources: the paper ‘Problems of Heredity as a Subject for Horticultural Investigation’, published in the Society's journal later that year and stated as ‘Read 8 May, 1900’, and Beatrice Bateson's account of the event over a quarter of a century later. Of the paper which her husband gave on that occasion she wrote:He had already prepared this paper, but in the train on his way to town to deliver it, he read Mendel's actual paper on peas for the first time. As a lecturer he was always cautious, suggesting rather than affirming his own convictions. So ready was he however for the simple Mendelian law that he at once incorporated it into his lecture.

Abstract:
In early twentieth-century America, the novel technology called “eugenics”—a potent hybrid of biological science, statistical method, and cultural assumptions—won a diverse following of academics, animal breeders, social workers, criminologists, psychiatrists, institutional superintendents, philanthropists, and activists spanning the political spectrum from socialists to white supremacists. Although heirs to the Enlightenment pursuits of science, reason, and a rationally organized state, eugenicists rejected the Enlightenment's egalitarian strain, insisting that hereditary endowment determined social structure. Fusing Darwin's theory of evolution and Mendel's discoveries in plant heredity, eugenicists claimed to find distinct genetic roots for the many problems of personality and society that alarmed their contemporaries: from “feeble-mindedness” and “psychopathy” to “delinquency” and “hypersexuality.” Within the bright lines of a eugenic worldview, the poverty and crime that pervaded an avowedly meritocratic urban-industrial democracy were comprehended as the offspring of hereditary “mental defects,” racial “mongrelization,” and sentimental charitable efforts that, in a vain attempt to reform deviant individuals, had only assured their survival and reproduction.

Abstract:
The implementation of the independent curriculum requires teachers to develop teaching tools in the form of complete and systematic teaching modules. This study aims to produce a valid Mendelian law of inheritance patterns teaching modules. The research method is R&D (Research and development) with the 4D development model. The subjects of this study were two biology lecturers at FMIPA UNP and six biology teachers. The data collection instruments were observation questionnaires, validity questionnaires, and legibility test questionnaires. This study found that the independent curriculum teaching modules in phase F were not yet available because the independent curriculum had just been implemented. Based on the research conducted, it was found that the teaching module on the pattern of inheritance on Mendel's laws based on problem based learning for biology subjects in phase F SMA/MA is valid with an average score of 91.20% and very valid criteria.

Abstract:
DNA analysis is widely applied in solving forensic cases, especially Short Tandem Repat (STR) because of its advantages. In identifying the individual, the National Police compared the individual's DNA with that of his parents. Each anal has a pair of DNA fragments of which half are inherited by the father and the remainder by the mother according to Mendel's Law of Segregation. In this study, we compared DNA typing between the child and the mother with the help of PCR extracted by the Chelex method to find the mother fragment and obtain the father fragment. A child is the biological child of the alleged father if he or she has less than 2 exclusion STR loci. The results of this study revealed that all paternal fragments from the child were identical to the DNA fragments of the alleged father, except for one locus, namely CSF1PO which had a mutation. Mutations in the STR locus lower the paternity index, although it can still be concluded that the child is the biological child of the alleged father.

Abstract:
Calpain 3 is known as the skeletal muscle–specific member of the calpains, a family of intracellular nonlysosomal cysteine proteases. It was previously shown that defects in the human calpain 3 gene are responsible for limb girdle muscular dystrophy type 2A (LGMD2A), an inherited disease affecting predominantly the proximal limb muscles. To better understand the function of calpain 3 and the pathophysiological mechanisms of LGMD2A and also to develop an adequate model for therapy research, we generated capn3-deficient mice by gene targeting. capn3-deficient mice are fully fertile and viable. Allele transmission in intercross progeny demonstrated a statistically significant departure from Mendel's law. capn3-deficient mice show a mild progressive muscular dystrophy that affects a specific group of muscles. The age of appearance of myopathic features varies with the genetic background, suggesting the involvement of modifier genes. Affected muscles manifest a similar apoptosis-associated perturbation of the IκBα/nuclear factor κB pathway as seen in LGMD2A patients. In addition, Evans blue staining of muscle fibers reveals that the pathological process due to calpain 3 deficiency is associated with membrane alterations.

Abstract:
Twentieth century biology began with the rediscovery of Gregor Mendel's work on peas and ended with the sequencing of the human genome. In between came Thomas Morgan's studies of fruit flies, the grand synthesis between genetics and evolutionary biology in the 1930s, and Watson and Crick's publication in 1953 of the double helix structure of deoxyribose nucleic acid (“DNA”), the substance in the nucleus of cells, which carries the genetic code of all eukaryotic life. The genetics of the second half of the century focused on learning how DNA coded for proteins, how to splice, clone, and recombine pieces of DNA, and how genetic mutations caused disease. In the late 1980s, a project to identify the actual sequence of all 3.2 billion base pairs of the human genome began. In June 1999, President Clinton and Prime Minister Blair announced that a working draft of the human genome was complete, with the final completed draft to 99.9% accuracy expected in May 2003, fifty years after the publication of Watson and Crick's landmark paper.

Abstract:
Information of legitimacy of oil palm progenies is important to guaranty the quality and to control commercial seeds procedures. A true and legitimate cross will produce progeny which has a combination of their parent's allele. The information could be obtained early in the nursery stage through DNA fingerprinting analysis. Simple Sequence Repeats (SSR) is one of DNA markers used for DNA fingerprinting, since the marker system has advantages to acquire information of allele per individual in population and efficiency diverse allele of progeny and their parents. The aim of the research is to obtain legitimacy of 12 progenies analyzing in the oil palm nursery stage. Thirteen SSR markers were used to analyze 12 crossings number of oil palm. The genotypes data by alleles of SSR inferred and quantified using Gene Marker® Software version 2.4.0 Soft Genetics® LLC and analyzed based on Mendel's Law of Segregation. The result showed based on heredity pattern of progeny and their parent's allele that progenies H were indicated genetically derived from their known parents while progenies from A and G indicated as illegitimate crossing. Probability value for legitimacy of progenies of 9 other crosses has 0.031 and 0.5. Legitimacy analysis of progeny using SSR markers could be used to control the quality of crossing material and earlier selection in the oil palm nursery.

Abstract:
This work aims at the production of an educational object based on a game platform, entitled Genus Project. Our objective is to support and facilitate the understanding of classical mendelian genetics. This project is under a Professional Masters’ program in Scientific, Educational and Technological Formation – PPGFCET – at the Federal University of Technology, Paraná (UTFPR). Game platforms constitute important educational objects and motivational strategies to stimulate students’ learning. Games can also be considered as alternative proposals for the teaching of diverse aspects of scientific knowledge; in addition, they provide high quality interaction between students and teachers, encouraging cooperative relations between them. From the educational aspect, we focused on meaningful learning and active learning methodologies as pedagogical principles for the teaching of classical mendelian genetics at high school level. Hereditary phenomena are often pointed out as a especially difficult subject in the learning of genetics. Therefore, the Genus Project was developed in order to present alternatives to overcome those difficulties by establishing a science fiction scenario for the game – which is inspired by concepts of "gamification" – including traditional exercises and problems in genetics. In this way, the game stimulates students’ interaction, and promotes educational strategies to empower students and make them protagonists of their own learning process.

Abstract:

Abstract A discussion of the origin of Mendelism cannot start by following the reasoning that led Mendel and others to the important early discoveries in Mendelism; few historical records exist to provide evidence about such discoveries. Abstract As we discussed in 5, in the early days of Mendelism, what is now referred to as the “independent assortment” of two different traits, or “Mendel’s second law,” was not separated from the law of segregation. In 1900, segregation appeared to hold for dihybrid crosses involving two different traits, as well as in the usual monohybrid cases. Mendel’s crosses of yellow tall peas with green short ones, for example, produced ratios of9 yellow tall:3 yellow short:3 green tall:1 green short in the F2• All possible combinations occurred independently.

Abstract:
Abstract After having been overlooked for 35 years, Gregor Mendel’s laws of genetic transmission were rediscovered in 1900. Mendel had based his work on breeding experiments in peas and flowering plants, but the laws were found to be applicable to other organisms as well. With their rediscovery, Mendel’s laws provided the framework upon which all subsequent work in human genetics has been based. Mendel’s first law is referred to as the independent assortment of alleles. (Alleles are the two different copies of a gene.) Alleles may be dominant or recessive in their influence on the development of characteristics. (Genetic characteristics are commonly referred to as “phenotypes.”) If an allele is dominant, one copy is sufficient to produce the phenotype-that is, to encode it. If it is recessive, two copies are required.

Abstract: Iñaki Abalos develop in “Atlas pintoresco” a research about Modern Movement and its relationship with nature and landscape. The author provides an analysis which unmask the picturesque elements in the work of Le Corbusier. These elements, even being "latent" in its earliest theoretical postulates, gradually and "almost secretly" will take over his way of projecting to conquer all scales of architecture in its final stage. Some of the architectural features of the so-called Paulista School seems to have influences of the later work of Le Corbusier.

Abstract:
The information revolution is radically transforming the very foundation of the ‘fossil city’. A ‘virtual’ macro-urbanism will intersect with an ‘actual’ micro-urbanism, physical and concrete, determining the form of the new urban environment. Within the binomial of macro- and micro- urbanism, urban morphology identifies an interesting socio-building scale that can serve as the basic strategy for sustainable city planning in the twenty-first century. Morphology thus becomes the necessary ‘plug-in’ for registering the different ‘networks’ that characterize the contemporary city – from IT and ‘smart’ devices to energy and environmental systems - translating these networks into building practices, into ‘fabrics’, for the physical city. At this purpose an Urban Design methodology has been developed in order to combine the Urban Morphology tools with those of Sustainability giving particular attention to the topics of the comfort outdoor and the passive environmental control systems. The methodology has then been applied in the Sant Adrià De Besos Waterfront Regeneration Project in Barcelona. Neighbourhood’s size, complexity and localisation, between the sea and a large area of brown fields at the northern gateway of the Catalan capital, has set up an interesting testing bench. A sequence of consecutive steps characterizes the methodology in which morphology, architecture and sustainability intersect one another within a single design process. References Gherri B. Abstract:

Abstract:
BACKGROUND AND OBJECTIVES Over half of childhood tuberculosis (TB) remains undiagnosed yearly. TB culture is often unavailable. WHO recommends Xpert-Ultra as first test for diagnosis of paediatric TB, but microbiological confirmation remains low and often requires invasive procedures. We aimed to determine the utility of Xpert-Ultra in stools and urine samples to diagnose TB in children living with HIV (CLWH) in two high-TB burden settings. METHODS This cross-sectional multicentric study took place at Simão Mendes hospital, Guinea-Bissau, from July 2019 to April 2020, and in Malakal hospitals, South Sudan, from November 2019 to June 2023. Children 6 months to 15 years with suspected TB underwent clinical and laboratory assessment, with one respiratory or extrapulmonary sample (gold standard (GS)), one stool and one urine specimen per patient analysed with Xpert-Ultra. RESULTS A total of 93 HIV-positive children were enrolled from Bissau (n=57) and Malakal (n=36), with 49 (53%) females and median (IQR) age of 3.3 (1.5-10) years. Three-quarters of children had severe acute malnutrition (SAM). A total of 72 (77%) children were on ART at baseline and 26/77 (34%) had CD4 count &lt;200cells/mm3. Confirmation of TB was achieved in 20 (22%); 51 (55%) had unconfirmed TB, and 22 (24%) had unlikely TB. Of 93 children with GS diagnosis, the overall yield of positive TB results was 22% (20/93): 10% (9/90) in pulmonary samples and 20% (1/5) in extrapulmonary samples. A total of 86 and 91 samples were used to evaluate Xpert-Ultra on stools and urine, respectively. Compared to GS, sensitivity and specificity on stools were 87.5% (95%CI:52.9-97.8) and 100% (95%CI: 95.3-100), whereas on urine were 30% (95%CI:10.8-60.3) and 100% (95%CI:95.5-100), respectively. No patients were positive in stools or urine and negative with GS. CONCLUSIONS Xpert-Ultra in stools showed high sensitivity and specificity in HIV-infected children when compared to gold standard. Sensitivity of urine was low, but more research is needed to determine its clinical indication.

Abstract:
INTRODUCTION Tuberculosis (TB) is an important cause of morbidity and mortality in children and over 50% of childhood TB remains undiagnosed every year. As microbiological confirmation is low (&lt;30%), the majority of cases in low and middle-income countries are diagnosed on clinical grounds. Point-of-care ultrasound (POCUS) is a non-invasive bedside tool, and TB-focused POCUS has been validated for diagnosis of TB in adults with HIV. We aimed to describe the performance and findings of TB-focused POCUS for children with presumptive TB at a tertiary care hospital in Guinea- Bissau, a setting with a high burden of HIV, malnutrition and TB. METHODS This observational study took place at Simão Mendes hospital in Bissau, from July 2019 to April 2020. Patients aged between 6 months and 15 years with presumptive TB underwent clinical and laboratory assessment, with at least one sample analysed with GeneXpert Ultra, and unblinded clinician-performed POCUS evaluation. POCUS was used to assess for subpleural nodules (SUN’s), lung consolidation, pleural effusion, pericardial effusion, ascites, liver and splenic focal lesions, and abdominal lymph nodes. Presence of any of these signs prompted a POCUS- positive result. Images and clips were evaluated by an expert reviewer and, if discordant, by a second expert reviewer. ETHICS This study was approved by the MSF Ethics Review Board (ERB) and by the Guinea-Bissau Ministry of Health ERB. RESULTS A total of 139 children were enrolled, with 62 (45%) female and 55 (40%) aged under 5 years. HIV infection and severe acute malnutrition (SAM) were found in 59 (42%) and 83 (60%) of patients, respectively. Confirmation of TB was achieved in 27 (19%); 62 (45%) had unconfirmed TB, and 50 (36%) had unlikely TB. Children with TB were more likely to have a POCUS positive result (83/89; 93%) as compared to children with unlikely TB (17/50; 34%). The most common POCUS signs in TB patients were: lung consolidation (51; 57%), SUN’s (49; 55%), pleural effusion (27; 30%), and focal splenic lesions (25; 28%). In children with confirmed TB (n=27), POCUS sensitivity was 85.2% (95% confidence interval (CI) 67.5-94.1). In those with unlikely TB (n=50), specificity was 66% (95%CI 2.2-77.6). Unlike HIV infection, SAM was associated with higher risk of positive POCUS. Cohen’s kappa coefficient for concordance between field and expert reviewers ranged from 0.6 to 0.9 depending on the POCUS sign, while overall POCUS concordance was 0.8. CONCLUSION We found high prevalence of any POCUS sign in children with TB, as compared to children with unlikely TB. POCUS positivity was independent of HIV status, but not of nutritional status. POCUS concordance between field and expert reviewers was moderate to high. TB-focused POCUS could potentially play a supportive role in the diagnosis of TB in children. Abstract:
