Comptant parmi les figures les plus éminentes de la littérature arthurienne, célèbre au Moyen Âge comme dans les périodes postérieures, Merlin demeure pourtant un personnage polymorphe et contradictoire. Un double angle d’étude permettra de saisir ses ambiguïtés et de constituer une poétique du personnage. D’abord, dans une perspective relationnelle, le bestiaire, c’est-à-dire la faune littéraire, constitue un élément de sa définition. Dans les Suites rétrospectives du Merlin en prose (la Suite dite « Vulgate » la Suite dite « Post-Vulgate » et le Livre d’Artus), romans qui constituent l’aboutissement des proses arthuriennes du XIIIe siècle, cet axe relationnel rencontre ensuite une perspective intertextuelle. Par son extension et sa nature, le bestiaire merlinien se révèle extrêmement variable et difficilement classable, à l’image du personnage. Présidant aux relations entre le personnage et le bestiaire, les paradigmes de l’incarnation et de la voix, ainsi que la dialectique intus/foris, laissent percevoir une complexification croissante de Merlin, qui allie un rôle de vates responsable de la fiction et du bestiaire prophétique et une dimension proprement romanesque à l’issue du Merlin en prose. Enfin, dans les Suites du Merlin en prose, le bestiaire rend aussi bien compte du (re)développement et de la fin conjoints de ces deux aspects du personnage que de la poétique des textes. Par le biais de relations métonymiques, métaphoriques et analogiques avec Merlin, le bestiaire construit ainsi trois conceptions synchroniquement contrastées du personnage, en adéquation avec la tonalité et les objectifs poétiques différents des Suites. One of the most eminent figures of Arthurian literature, renowned in the Middle Ages as in later periods, Merlin remains however a polymorphous and contradictory character. A study focusing on two aspects will allow us to perceive his ambiguities and to form the poetics of the character. Firstly, from a relational perspective, the bestiary, that is to say the literary fauna, constitutes an element of this definition. In Prose Merlin’s retrospective prose sequels (the “Vulgate” Suite, the “Post-Vulgate” Suite and the Livre d’Artus), romances which constitute the apex of thirteenth century Arthurian texts in prose, this relation axis encounters an intertextual perspective. By its extent and its nature the Merlin bestiary reveals itself to be extremely variable and difficult to categorize, as is the nature of the character. The paradigms of incarnation and voice, together with the intus/foris dialectics, that govern the relations between the character and the bestiary, show that Merlin becomes increasingly complex, because he combines a role of vates responsible for the fiction and the prophetic bestiary with a distinctly romantic dimension by the end of the Prose Merlin. Finally, in the Prose Merlin sequels, the bestiary also explains both the (re)development and the end of these two aspects of the character, as well as the poetics of the texts. Through the metonymical, metaphorical and analogical relations with Merlin, the bestiary thus builds up three different synchronically contrasting conceptions of the character, matching the tone and the various poetical purposes in the Suites. Merlin is the protein encoded by the tumor suppressor gene NF2. Deletion or loss-of-function of NF2 leads to neurofibromatosis type 2, a disease characterized by the formation of multiple benign tumors of the nervous system. In addition to the genetic disorder, loss of merlin expression has been found in sporadically occurring schwannomas and meningiomas, as well as in mesothelioma. Merlin has two major isoforms that differ in only one exon at the C-terminal. Previous work hypothesized that isoform 11 is unable to suppress growth. In this thesis, I show that both of the major merlin isoforms are able to suppress growth in multiple cell lines, including mesothelioma. Merlin has been shown to suppress growth through multiple mechanisms, including upstream regulation of the oncogene YAP through stabilization of the Hippo-pathway kinase Lats, allowing Lats to phosphorylate and inhibit YAP. In this thesis I identify an additional mechanism for merlin regulation of YAP, which occurs independently of the Hippo-pathway-based regulation. I show that mesothelioma cells expressing merlin have lower YAP-driven transcriptional activity and that expression of merlin leads to cytoplasmic localization of YAP. Furthermore, I show that this regulation of YAP is not dependent on the major Lats phosphorylation sites, but does require the YAP WW domains. This thesis provides additional insight into how merlin controls cell growth, and into YAP regulation by merlin.

Cet essai se propose de montrer comment, dans l'univers imaginaire d'Edgar Quinet, parvient à se constituer autour du personnage de Merlin ce qu'une mythocritique inspirée de Pierre Albouy peut qualifier de mythe littéraire. Du mythe littéraire tel que Pierre Albouy l'a défini, le mythe de Merlin recréé par Edgar Quinet dans son épopée testamentaire (Merlin l'enchanteur, 1860) présent en effet tous les caractères fondamentaux. Il s'enracine dans une tradition ou plutôt dans un faisceau de traditions populaires et savantes, folkloriques (Barzaz-Breiz de la Villemarque), historiographiques et romanesques (Historia Regum Britanniae ; Vita Merlini de Geoffroy de Monmouth ; Brut de Wace ; Merlin de Robert de Boron, suivi de son cortège de suites anonymes. De cette tradition, il retient des thèmes et figures recurrentes (Merlin prophète, poéte, enchanteur tour à tour puissant ou enseveli vivant par la femme qu'il aime) qu'il organise en un récit complexe. Ce dernier, enrichi de significations nouvelles, se transforme chez Quinet, au terme d'une triple palingénèsie, en mythe du moi, mythe de la poétique, mythe de l'histoire universelle. This essay intends to show how, in edgar quinet's imaginary world, what can be called a literary myth - as a mythocriticism inspired by pierre albouy would call it - is gradually built up round the character of merlin. For the myth of merlin, as edgar quinet recreated it in his testamentary epic work (merlin l'enchanteur, 1860), shows all the fundamental characteristics of the literary myth as defined by pierre albouy. It takes root in a tradition or rather a group of traditions either belonging to folklore (barzaz breiz by la villemarque) or again belonging to historiography and romance (historia regum britanniae, vita merlini by geoffroy de monmouth ; brut by wace ; merlin by robert de boron, and all the anonymous works that followed in its wake). Out of this tradition the myth retains recurrent themes and characters (merlin as a prophet, a poet, an enchanter, now powerful, now buried alive by the woman he loves) which are woven into a complex tale, this narrative, enriched with new meanings, changes in quinet's work, at the outcome of a three-fold palingenesy, into a myth of the self, a myth of the poetic and a myth of universal history. Though branching, the romantic myth of merlin is none the less unified in its structure as well as in its meaning. Thus it shows the dynamic coherence that gilbert durand requires of a myth,and it builds itself around the schemas of difference and metamorphosis which reveal the active spell that edgar quinet demands for imagination and mythological creation

Martial Merlin a été Gouverneur général de l'Afrique équatoriale française (de 1909 à 1917), de l'Afrique occidentale française (de 1919 à 1923) et de l'Indochine (de 1923 à 1925). Il est le seul Administrateur colonial à avoir occupé les trois grands Gouvernements généraux de la France d'Outre-mer. L'AEF, l'AOF et l'Indochine étaient des Groupes de colonies : ils furent créés à la fin du XIXe siècle, afin de donner une unité de direction à des colonies jusque là dispersées. L'étude de l'œuvre et de la doctrine coloniales de Merlin nous permet de mieux connaître l'institution du Gouvernement général. Merlin souhaitait que l'Administration fût organisée sur des bases claires. Il a défini les attributions de chaque organe du Gouvernement général et il a souligné le rôle essentiel du Gouverneur général, chargé de donner une unité de direction politique et économique aux territoires regroupés. Merlin souhaitait également que l'Administration s'appuyât sur des bases solides. Il a mis en œuvre une politique de Gouvernement indirect, afin de diriger les populations indigènes par l'intermédiaire de leurs chefs coutumiers. Organe de coordination économique, le Gouverneur général devait assurer la mise en valeur de son Groupe de colonies : Merlin a défini une politique de développement précise, tout en se souciant des moyens de sa politique de développement (il a notamment réglementé le régime de la main d'oeuvre). Martial Merlin was the Governor General of the AEF (French equatorial Africa) from 1909 to 1917, of the AOF (French western Africa) from 1919 to 1923 and of Indochina from 1923 to 1925. He is the only colonial administrator to have served as Governor General of all three organizations of the French colonies. The AEF, AOF and Indochina, each grouping together many colonies, were created at the end of the nineteenth century, in order to impose a coherence to the direction of the colonies, which up until then had been administered separately. A study of the colonial theory expressed in the works of Merlin provides an insight into the institution of the Government General. Merlin affirmed that the administration of the colonies should be organized on a clear basis. He assigned specific attributes to each organ of the Government General, and emphasized that the essential role was to be played by the Governor General, whose duty it was to define a unified policy for the political and economic direction of the various territories. Seeking a firm basis for this administration, Merlin implemented a policy of indirect government, controlling the native population by using their customary chiefs as intermediaries. As an agent of economic coordination, the Governor General was to turn to good account his group of colonies ; Merlin defined a policy of development which included initiatives to insure the means by which to implement that policy (he introduced, for example, a set of labor regulations). As an agent of political direction, the Governor General was to convince the natives of the advantages of the "French peace" ; Merlin defined and implemented a policy of “contact”, in order to gain the support of the natives to the French cause, while at the same time practicing a "defensive" policy, made necessary by the eruption of movements hostile to the French presence

O presente trabalho de mestrado foi desenvolvido no âmbito de um estágio na empresa Leroy Merlin e tem como objetivo a criação de uma política mutustock para um conjunto de produtos comercializados nas suas quatro lojas da região de Lisboa (Almada, Alfragide, Amadora e Sintra). A criação de uma política mutustock nasceu da necessidade de ter uma política de gestão de stock eficaz e eficiente. No passado, o stock encomendado era da responsabilidade apenas da loja que efetuava a encomenda e era esta que geria toda a sua organização no respetivo armazém. Para além de não existir transferências entre lojas, cabia a cada loja saber gerir o espaço do armazém, tendo em atenção a mercadoria existente e a mercadoria encomendada. Neste sentido, o principal objetivo deste estágio passou por estudar, analisar e identificar as famílias de produtos que devem ser escolhidas para este processo de mutustock. Isto implicará que estas famílias sejam transportadas da loja para a plataforma logística, e aqui estejam disponíveis para qualquer uma das quatro lojas da região de Lisboa. Por consequência, todas as famílias identificadas passam a ter "responsabilidade" partilhada entre as quatro lojas da região de Lisboa. Com a implementação da política mutustock pretende-se diminuir a quantidade de stock tóxico (stock com prazo médio superior ao valor definido pelo Supply Chain), aumentar eficiência e eficácia do espaço de armazém das lojas da região de Lisboa e respetivas equipas, aumentar o volume de vendas e otimizar os custos de stock.
The present master thesis was developed during an internship at Leroy Merlin and it has as main objective the creation of a mutustock policy for a set of products marketed in the four Leroy Merlin stores in the Lisbon area (Almada, Alfragide, Amadora and Sintra). This mutustock policy was born from the need to have an effective and efficient stock management policy. In the past, each ordered stock were under each store responsibility and the entire stock management were run by each store, meaning no transfers between stores and specific warehouse space management, taking into account the existent and the ordered merchandise. In this sense, the main objective of this stage was to study, to analyze and to identify the families of products chosen and included in this process of mutustock. This will involve transporting these families from the store to the logistics platform and, from here, will be available to any of the four stores in the Lisbon area. Consequently, these families have shared "responsibility" among the four stores. With the implementation of the mutustock policy, the aim is to reduce the quantity of toxic stock (stock with an average term higher than the value defined by the Supply Chain), to increase the efficiency and effectiveness of the store space and to leverage the efficiency of store teams, to increase sales volume and to optimize inventory costs.

O sucesso e o crescimento sustentável das empresas assenta, cada vez mais, na sua capacidade de atracção e manutenção dos actuais clientes, principalmente dos mais lucrativos. Desta forma, torna-se fulcral para qualquer empresa avaliar a satisfação dos seus clientes. O presente estudo pretende analisar a satisfação dos clientes da loja Leroy Merlin de Alfragide e, para isso, tal como é apontado na revisão da literatura, torna-se fulcral a análise dos constructos da qualidade percebida do serviço, assim como da fidelização dos clientes. Para a recolha de informação foi utilizado um inquérito por questionário, sendo que na medição da qualidade do serviço é utilizado o modelo Servperf. A amostra foi selecionada de forma não probabilística e abrangeu 100 clientes da loja Leroy Merlin de Alfragide. Conclui-se que os clientes se encontram satisfeitos com o serviço e, para além de assumirem voltar a fazer compras na loja, recomendam a Leroy Merlin aos seus amigos e familiares.
The success and sustainable growth of companies is increasingly built in capability to attract and maintain current customers, mainly the most lucrative ones. Thereby, it is crucial for any company to evaluate the customer satisfaction. This study aims to analyse the level of satisfaction from Leroy Merlin Alfragide customers by examining constructs of perceived service quality as well as customer loyalty, as pointed out in the literature review. In order to measure the quality of service, the SERVPERF model was used, whereas for the analysis of costumer satisfaction, a questionnaire survey was examined. The analyzed sample was selected on a non probabilistic basis which covered 100 customers from Leroy Merlin Alfragide. It is concluded that customers are satisfied with the service and, in addition to wanting to shop again in the store, they recommend the service to their friends and family.

Study of merlin's character through middle age writings and numerous modern images. We studied his type of seduction which first appeared during the middle age period and also the evolution of this particular metaphor. We also focalized our attention on the evolution of the character himself : from the wild man (the fool on the hill) to an idealistic vision : the wise man. We tried to show the evidence of merlin's own seducing seducing processus : a mixture of ambiguity and pagan reminiscences. Merlin annoys, is attracting ; his whole presence gives a supplementary touch of meaning to the text. Merlin seems to be the fortunate possessor of a knowledge that is missing us : nostalgia of the roots or taste for suspense ?

Abstract:
The ability to spatially restrict specific activities across the cell cortex functionally defines individual cells and tissues. This is achieved, in part, via the assembly of protein complexes that link the plasma membrane to the underlying cortical actin cytoskeleton. The neurofibromatosis type 2 (NF2) tumor suppressor Merlin and closely related ERM proteins (Ezrin, Radixin and Moesin) are a special class of such membrane:cytoskeleton associated proteins that function to organize specialized cortical domains. In addition to their high degree of similarity, mounting evidence suggests that Merlin/ERMs share a functional relationship, which is largely unexplored. Unlike Merlin, the ERMs are not known to inhibit cell proliferation; in fact, Ezrin is thought to promote tumor metastasis. Defining the relationship between Merlin and the ERMs is essential to appreciating their respective roles in cancer development. Here I demonstrate a novel role for Merlin and the ERMs in generating cortical asymmetry in the absence of external cues. Our data reveal that Merlin functions to restrict the cortical distribution of Ezrin, which in turn positions the interphase centrosome in single epithelial cells and 3D organotypic cultures. In the absence of Merlin, ectopic cortical Ezrin yields mispositioned centrosomes, misoriented spindles and aberrant epithelial architecture. Furthermore, in tumor cells with centrosome amplification, the failure to restrict cortical Ezrin abolishes centrosome clustering, yielding multipolar mitoses. Consistent with a functional relationship, I observe a strong genetic interaction between Nf2 and Ezrin in the mouse intestine in vivo. Finally, I begin to address the basis of their functional interaction by testing whether they are coordinately regulated by the Ste-20 like kinase SLK. Altogether, these data uncover fundamental roles for Merlin/ERM proteins in spatiotemporally organizing the cell cortex in vitro and in vivo and suggest that Merlin’s role in promoting cortical heterogeneity may contribute to tumorigenesis by disrupting cell polarity, spindle orientation and potentially genome stability.

Probabilistically Checkable Proofs (PCPs) are an important class of proof systems that have played a key role in computational complexity theory. In this thesis we study the power of PCPs in two new settings: Arthur-Merlin games and communication protocols. In the first part of the thesis, we give a 'PCP characterization' of AM analogous to the PCP Theorem for NP. Similar characterizations have been given for higher levels of the Polynomial Hierarchy, and for PSPACE; however, we suggest that the result for AM might be of particular significance for attempts to derandomnize this class. To test this notion, we pose some 'Randomized Optimization Hypotheses' related to our stochastic CSPs that (in light of our result) would imply collapse results for AM. Unfortunately, the hypotheses appear over-strong, and we present evidence against them. In the process we show that. if some language in NP is hard-on-average against circuits of size 2 [omega](n), en there exist hard-on-average optimization problems of a particularly elegant form. In the second part of the thesis, we study PCPs in the setting of communication protocols. We show a complementary negative result: a distributed PCP protocol using a proof of length f, in which Alice and Bob look at k bits of the proof while exchanging t bits of communication, can be converted into a PCP-free randomized protocol with communication bounded by In both parts of the thesis, our proofs make use of a powerful form of PCPs known as Probabilistically Checkable Proofs of Proximity. and demonstrate their versatility. In our work on Arthur-Merlin games, we also use known results on randomness-efficient soundness- and hardness-amplification. In particular, we make essential use of the Impagliazzo-Wigderson generator; our analysis relies on a recent Chernoff-type theorem for expander walks.

Abstract:
The tumour suppressor Merlin is mutated in the familial cancer syndrome Neurofibromatosis Type 2 (NF2) and in various sporadic tumours. Several lines of evidence show a role for Merlin in mediating contact-dependent inhibition of proliferation. Contact inhibition is essential for normal tissue homeostasis and its deregulation is a hallmark of cancer. Despite extensive efforts the mechanism underlying tumour suppression by Merlin remains elusive. A proteomics approach led to the identification of novel Merlin interacting proteins several of which are involved in gene expression, in particular transcription elongation, RNA processing and histone modifications, and are also deregulated in cancer. Among those are the RNA PolymeraseII-associated factor1 complex (Paf1C), CHD1, TAT-SF1 and spliceosome components. These interactions have been validated and mapped to Merlin’s FERM domain and loss-of-function Merlin mutations found in tumours invariably disrupt these interactions. The interaction of Merlin with the Paf1C is modulated by cell density and is required for Merlin’s tumour suppressor function. We find a remarkable case of tumour suppressor gene hypersensitivity in Merlin-deficient cells that correlates with Merlin’s ability to regulate gene expression. Using genome-wide expression profiling we identify a gene signature associated with growth arrest by Merlin expression that is consistent with Merlin’s role in mediating contact inhibition and suggests a role for Merlin in innate immunity and communication with the microenvironment. By integrating re-expression and knockdown gene signatures a core Merlin signature has been defined that correlates with NF2 mutational status and suggests differential drug sensitivities and potential therapeutic targets for treatment of NF2-related tumours. Using genome-wide occupancy analysis we identify a subset of genes where Merlin expression regulates association of the Paf1C with chromatin of coding regions. Some of these regulated genes are known target genes of YAP, the transcriptional co-activator of the Hippo tumour suppressor pathway. Although YAP expression rescues growth arrest by Merlin, Merlin can regulate expression independently of YAP. A model is proposed where Merlin functions parallel to YAP to regulate gene expression at the elongation level through the Paf1C.

Abstract:
The large hadron collider (LHC) uses the most energetic and highest luminosity man made proton beams on Earth. The high luminosity (HL-LHC) [1] upgrade aims to increase the levelled luminosity of the LHC by a factor of five, to 5 · 1034cm−2 s −1 , by increasing the bunch population from 1 to 2.22·1011 protons, and decreasing emittance, and β ∗ . Thus the stored beam energy increases from ≈ 362 MJ to ≈ 675 MJ per beam. All synchrotrons encounter unavoidable proton losses. Protons that populate the beam halo pose a threat to the performance and lifetime of certain hardware, such as superconducting magnets, which in the LHC may be quenched by an impact of ≈ 1 · 106 protons [2]. A multi stage collimation system must operate at an efficiency such that no more than 2 · 10−5% of protons incident on collimators may escape and impact upon these magnets [3]. To predict and protect against proton losses in the HL-LHC, collimation simulations must be performed. MERLIN, a C++ accelerator physics library, has been updated to carry out such simulations for the HL-LHC. Novel materials such as molybdenum graphite have been investigated as collimator materials, and a novel technique - collimation enhancement via a hollow electron lens (HEL) - has been studied. Using detailed simulations the performance and operation of possible collimation upgrades are explored.

O presente trabalho trata de analisar a ressignificação do personagem Merlin, das lendas Arthurianas, em três filmes contemporâneos: Dragonslayer, Star wars e The Matrix – sob o viés da tradução intersemiótica. Através do ciclo do herói de Joseph Campbell, do ciclo de mago de Stephen Ware, bem como das propostas de análises de personagens de Vladimir Propp e Mieke Bal, analisamos o personagem Merlin na obra A Morte de Artur, de Sir Thomas Malory. A partir desta análise, comparamos seu percurso narrativo e suas funções narrativas com as do mago arquetípico de Ware. Em seguida, fazemos o mesmo com os personagens Ulrich, Obi-wan Kenobi e Morpheus, nos três filmes escolhidos, e comparamos estas análises entre os personagens fílmicos e Merlin, para averiguar se estes seriam traduções – embora não-nomeadas – do mago arthuriano.
The present work analyses the ressignification of the character Merlin, from Arthurian legends, in three contemporary movies: Dragonslayer, Star Wars and The Matrix - regarding the point of view of intersemiotic translation. Throughout the Hero Cycle of Joseph Campbell, the Mage Cycle of Stephen Ware, and also the proposals of character analysis from Vladimir Propp and Mieke Bal, the character Merlin is analyzed using the book Le Mort D´Arthur, written by Sir Thomas Malory. From this analysis, the narrative trajectory of Merlin and his narrative functions are compared to the archetypical mage of Stephen Ware. After that, the same process is made for the characters Ulrich, Obi-wan Kenobi and Morpheus, from the three chosen movies, and those analysis are compared with the analysis of Merlin, in order to assure if those characters are or are not intersemiotic translations – although not named by the authors as Merlin´s adaptation – of de Arthurian mage.

Abstract:
Neurofibromatosis type 2 (NF2) is an autosomal dominant inherited condition that predisposes individuals to develop multiple nervous system tumours, primarily schwannoma, meningioma and ependymoma. NF2 is characterised by loss of the tumour suppressor protein Merlin, caused by bi-allelic mutations of the encoding gene NF2 or by loss of heterozygosity. These tumours can occur either sporadically or as part of the NF2 condition. The majority are slow growing and display benign features, but this benignancy renders them largely unresponsive to classic chemotherapeutic agents leaving surgery and radiosurgery as the only remaining treatment options. Depending on their location, NF2-related tumours can cause a number of side effects, including nausea, balance problems, and in some cases hearing and/or vision loss. Phosphorylation is a key regulatory mechanism leading to changes in cell signalling. By identifying phosphoproteins that are significantly activated in tumour cells, novel therapies can be developed aiming to specifically target the phosphorylated status of these proteins thus ‘switching off’ the signalling cascade. The objective of this study is to identify and validate common targets in both Merlin-deficient meningioma and schwannoma to eventually exploit in novel therapeutic approaches. Using phosphoprotein purification followed by mass spectrometry analysis, we identified Signal Transducer and Activator of Transcription 1 (STAT1), phosphorylated at Serine (S) 727 and Tyrosine (Y) 701, PDZ and LIM domain protein 2 (PDLIM2), Heat Shock 70kDa Protein 1A (HSPA1A) and Filamin B (FLNB) as potential common, novel therapeutic targets. We validated these candidates in human primary meningioma and schwannoma tumour cells using a variety of techniques. We also showed that specific 7 knockdown of STAT1 and PDLIM2 was related to a significant decrease in cellular proliferation. Additionally, we performed co-immunoprecipitation using PDLIM2 as the bait protein and identified STAT1, HSPA1A and FLNB as binding partners, suggesting a novel interaction network involving all of the potential targets identified in this study. We also identified activation of several pathways and/ or biological processes in both tumour types that warrant further investigation i.e. endocytosis in schwannoma and the proteasome in meningioma. In conclusion, with our approach we substantially increased the overall body of knowledge regarding the proteome and phosphoproteome of meningioma and schwannoma. We generated a comprehensive set of data that highlighted several potential therapeutic targets and dysregulated pathways which will be further investigated.

The Cygnus OB2 Radio Survey (COBRaS) is an e-MERLIN Legacy survey awarded ~ 300 hours worth of observing time. Split between L-(42 hrs) and C-(252 hrs) band, the project aims to push the limits of e-MERLIN’s enhanced capabilities by providing highly sensitive, targeted, deep-field radio maps of the core of the Cygnus OB2 association. The L-band (21cm) observations, completed throughout the Spring of 2014, serve as the focus of the work presented here. This thesis primarily aims to investigate the radio emission from massive (M 8 M⊙ ) stars in the Cyg OB association, in order to obtain a better understanding of their mass-loss rates and binarity. A description of the entire data reduction process of the COBRaS 21cm observations is given, as is a discussion of the mitigation of radio frequency interference (RFI). In the advent of new radio telescopes, the technical skills, research and tools developed throughout this process aims to serve the wider radio astronomy community. Further technical work is presented in the form of novel source extraction and cross-correlation scripts that deliver the COBRaS L-band all source catalogue (CLASC). Reaching resolutions of ~180 mas, precise positions and 21cm flux densities are presented for 41 sources found within a 0.48 x 0.48 deg region, centred on the Cyg OB2 association. In obtaining a 3σ sensitivity limit of ~ 100 μJy, these observations significantly improve upon previous surveys of the region. Accurate determinations of the mass-loss rates of single massive OB stars are used to investigate their stellar wind properties, helping to resolve current discrepancies in the mass-loss rates via clumped and structured hot star winds. Finally, an analysis of the non-thermal radiation from the colliding winds of massive star binaries seeks to clarify our understanding of such systems and their radio emission processes.

Merlin is a tumour suppressor protein that is frequently mutated or downregulated in cancer. Biallelic Merlin inactivation is causative of tumour formation, including schwannoma, meningioma and ependymoma. These tumours can occur sporadically or as part of the genetic condition Neurofibromatosis type 2 (NF2) and cause significant morbidity. The current treatment options are restricted to surgery and radiotherapy, which are invasive and may cause further tumour development. The activity of both the E3 ubiquitin ligase complex Cullin 4 really interesting new gene (RING) E3 ubiquitin ligase- DNA damage binding protein (DDB1) and Cullin 4 associated factor 1 (CRL4-DCAF1) and Kinase suppressor of RAS 1 (KSR1) have been shown to be upregulated in schwannoma to drive tumour growth. KSR1 has also been shown to interact with components of the CRL4-DCAF1 complex. We investigated the expression, interaction and therapeutic potential of targeting these proteins in Merlin deficient schwannoma and meningioma using a primary human cell model and relevant cell lines. We found that DCAF1 and KSR1 protein were overexpressed in schwannoma and meningioma and confirmed that targeting both DCAF1 and KSR1 in meningioma had additive effects on proliferation. We also identified that CRL4-DCAF1 facilitates KSR1 dependent RAF/Mitogen-activated protein kinase (MAPK)/ Extracellular signal regulated kinase (ERK) kinase (MEK)/ERK pathway activity. We showed MLN3651, a neddylation inhibitor that targets ubiquitin ligase activity, reduced proliferation and activated apoptosis in Merlin-deficient tumours. We also showed that Merlin-positive tumours were less sensitive to MLN3651 than Merlin-deficient tumours; therefore, MLN3651 sensitivity may be CRL4-DCAF1-dependent. Finally, combination of MLN3651 and the MEK1/2 inhibitor AZD6244 had additive effects, particularly in meningioma. Combinatorial therapy activated the Hippo pathway, inhibited RAF/MEK/ERK pathway activity and proliferation demonstrating that targeting the activity and downstream pathways of both DCAF1 and KSR1 represents an attractive novel therapeutic strategy in Merlin-deficient tumours.

The Neurofibromatosis type 2 tumor suppressor gene (NF2) is mutated in inherited and sporadically occurring central nervous system tumors. The NF2 encoded protein, merlin, shares close sequence similarity in its amino-terminal domain to members of the band 4.1 family of membrane-cytoskeletal linkers. Similarities between merlin and this family suggest a role for merlin in regulating cytoskeletal function. Thus, NF2 may be a novel type of tumor suppressor gene that mediates its tumor suppressor function through interactions with the actin cytoskeleton. However, the molecular and cellular functions of this tumor suppressor gene were largely unknown when the work described here began. Mutational analysis of Nf2 in flies has lead to the identification of a dominant-negative allele, which harbors mutations in the amino-terminal domain of the protein. The work presented here demonstrates that expression of a murine analog of this amino-terminal mutant of Nf2 (termed, Nf2BBA) leads to complete transformation of NIH3T3 fibroblasts in culture. Cells that express Nf2BBA display disruptions of the actin cytoskeleton, lack of contact inhibition of growth, and anchorage-independent growth. In addition, Nf2-deficient mouse embryo fibroblasts (MEFs) exhibited similar contact inhibition and cell-matrix adhesion defects to Nf2BBA expressing cells. Nf2BBA cells continue to cycle under normal growth inhibitory conditions, such as serum withdrawal, and exhibit high levels of the cell cycle regulator, cyclin D1. Elevated levels of cyclin D1 are necessary for cellular transformation following Nf2BBA expression. Nevertheless, the exact mechanism by which Nf2BBA results in cellular transformation remains elusive. Recently published studies have revealed that merlin may regulate members of the RhoGTPase
(cont.) family, as absence of Nf2 expression in fibroblasts leads to many phenotypes reminiscent of overactive Rac, such as increased membrane ruffling and increased activity of the c-jun N- terminal kinase (JNK). Our work has extended to the analysis of the role of merlin in the regulation of the Rac pathway. Using rat schwannoma cells and N2-deficient MEFs, we have demonstrated that merlin exerts its inhibitory effects downstream of Rac, through a direct interaction with the p21 activated kinase, Pak. We demonstrate that in the absence of merlin, Pak is active and hyperphosphorylated, and, conversely, when merlin is overexpressed, Pak activity is diminished. The N-terminal half of merlin binds to the functionally conserved Rac/Cdc42 interaction binding (CRIB) domain of Pak. Several models for merlin regulation of Pak activity will be discussed. Finally, the identification of Pak as a kinase that is misregulated in the absence of NF2 may lead to possible avenues for therapeutic intervention.

La Théogonie d'Hésiode, la Bible, l'Historia Regum Britanniae et le Faustbuch sont quelques unes des références fixant les caractères principaux des mythes de Prométhée, du Diable, de Merlin et de Faust. Pour étudier ces récits, nous avons élaboré un système des mythes de la connaissance. Construit sur une confrontation des premières versions et des princiales réécritures des mythes, ce système comporte différents motifs sur le modèle des mythèmes de C. Lévi-Strauss. Privilège divin ou objet d'une quête, la connaissance n'a d'intérêt que si elle est totale et qu'elle permet la création. L'objectif de cette thèse est de dégager l'objet de la connaissance. Dans les mythes étudiés, les champs de la connaissance montrent un éclatement en disciplines diverses (Prologue). Cette multitude se reflète dans le portrait du héros de la connaissance (chap. I). Mû par la libido sciendi, le héros connaît les affres du désir triangulaire (chap. II). L'univers des dieux et Dieu sont ses "modèles obstacles". Rédigée dans la première moitié du XIIIe siècle, la Suite Vulgate est une continuation du Merlin en prose qui constitue la dernière pièce du cycle du Graal. Partagée entre le déroulement de la vie de Merlin, qui lui donne une unité de type biographique, et la peinture de la jeunesse héroïque du roi Arthur, cette suite rétrospective sert de transition vers le Lancelot. Son succès s'affirme aux dépens d'autres continuations du Merlin dont la transmission manuscrite semble par comparaison marginale, et dont le projet demeure parfois inabouti. Ces différentes rédactions exposent la dynamique d'écriture et l'émulation suscitées par le développement de la prose arthurienne et l'effort de mise en cycle. L'étude de la mise en recueil, de la mise en page et de l'illustration des manuscrits comprenant le Merlin et ses suites éclaire le mode de production et de réception de ces oeuvres qui continuent d'être copiées et enluminées tout au long du Moyen Age. Le Merlin et la Suite Vulgate, le plus souvent intégrés à des compilations centrées sur l'histoire du Graal, entretiennent un lien particulier avec le Joseph d'Arimathie, l'Estoire del saint Graal et les Prophéties de Merlin, mais circulent aussi dans des recueils d'ambition didactique ou historique. The Vulgate Sequel to the French prose Merlin was composed in the first half of the XIIIth century and is the last piece of the Grail cycle. It creates a wider story encompassing the life of Merlin and the beginnings of King Arthur’s reign and serves as a prequel to the prose Lancelot. The Vulgate Sequel is the most widespread, but the existence of different sequels demonstrates the poetic dynamism and the rivalry produced by the development of Arthurian prose romances and their cyclification. Our study of the manuscript collections including Merlin and its sequels, of their mise en page and illuminations sheds new lights on the production and reception of works which were copied throughout the Middle Ages. Merlin and its Sequel are mostly transmitted in compilations focusing on the history of the Grail: they have a particular relationship to the Joseph of Arimathia, the Estoire del saint Graal and the Prophecies of Merlin. In some manuscripts, Merlin and its Sequel are copied with texts which do not belong to the Arthurian tradition, such as hagiographies or pastoral works, demonstrating their religious and didactic interest. Other compilations stress their historical and chivalric aspect. The Vulgate Sequel and its illumination accentuate the historical aspect of Merlin while giving an epic flavour to the beginnings of King Arthur's reign. This spirit might contradict the perspective of its integration in the romance Vulgate cycle, but it echoes the missionary dimension of the Estoire del saint Graal and resonates with the military conflicts of Lancelot and of the Mort le roi Artu

Puis, dans notre deuxième partie, nous avons analysé six de ces textes afin de savoir de quelle manière un imaginaire individuel entre en confrontation avec un imaginaire collectif. Enfin, nous avons tenté de démontrer que l'ensemble des œuvres étudiées forme une mythologie à part entière, avec des implications importantes dans la vie sociale. C'est ainsi que nous en avons conclu que les distinctions entre mythe littéraire et mythe dit " primitif " ne sont pas aussi importantes que d'aucuns ont bien voulu l'affirmer
In this work about the myth of Merlin, in order to know what the relation between literature and mythology is, I have tried to demonstrate that modern versions of this legend can contain some mythical elements. Then, in my second part, I have analysed six of these texts, to know how an individual imagination can enter in confrontation with a collective imagination. In myt third part, I have tried to show that the group of all the modern versions builds a real mythology, with some implications on social life. Then, I have concluded that the distinctions between literary myth and“primitive” myth aren't so important as some authors have asserted it

Abstract:
Neurofibromatosis type 2 is an autosomal dominant disease characterized by the formation of schwannomas and other peripheral neuropathies. The nf2 gene encodes the protein Schwannomin, or merlin. Schwannomin (Sch) is a membrane-cytoskeletal linking protein that suppresses cell proliferation at high cell density and modulates cell shape. Sch's tumor suppressive activity is regulated by its localization, conformation, and phosphorylation at serine 518 (S518). Sch's localization is dependent on binding the scaffold protein, paxillin. Phosphorylation of Sch at S518 regulates its conformation and tumor suppressor function. In a negative feedback loop, unphosphorylated Sch restricts cell proliferation downstream of Rac and p21-activated kinase (Pak), whereas Pak-induced phosphorylation inactivates Sch's ability to inhibit Pak and cell proliferation. Little is known about the function of the phosphorylated form of Sch, or the molecular mechanisms leading to its phosphorylation. Here we demonstrate that Sch-S518 phosphorylation is dependent on paxillin-binding and plasma membrane localization in SCs. Phosphorylation of Sch at the plasma membrane is mediated by Cdc42-Pak and results in altered SC morphology and polarity. Moreover, we have identified two extracellular stimuli that trigger Sch-S518 phosphorylation; these are neuregulin (NRG) and laminin, two potent activators of SC proliferation and myelination. NRG promotes Sch-S518 phosphorylation downstream of ErbB2/ErbB3 through PKA, whereas laminin-1 stimulation of β1 integrin promotes Pak- dependent phosphorylation of Sch-S518. Additionally, we find that Sch promotes process formation and elongation in primary and myelinating SCs, independent of Sch S518 phosphorylation. However, Sch phosphorylation was found to influence SC differentiation, as expression of an unphosphorylatable variant, Sch-S518A, facilitated SC myelination, whereas expression of a phospho-mimicking variant, Sch-S518D, reduced the SC's ability to myelinate. Together, these findings have identified receptor-mediated and paxillin-dependent pathways that regulate phosphorylation and inactivation of Sch's tumor suppressor function. Additionally, these results have elucidated novel normal functions for Sch during peripheral nerve development and myelination, and identify novel therapeutic targets for treatment of NF2 and other peripheral neuropathies.
