Abstract:
South African mathematics teachers in the Senior Phase (Grades 8 and 9) were introduced to the pedagogical tool, meaning equivalence reusable learning objects (MERLO), as a formative assessment (FA) strategy to promote and support teachers’ professional growth in using FA practices in the classroom. The cultural-historical activity theory (CHAT) and meta-didactical transposition (MDT) were used to frame the evolution process of teachers’ praxeologies. In this study we used qualitative participatory action research that encompassed 3 phases: pre-MERLO phase, MERLO workshop and post-MERLO phase. The study was conducted in the northeast of Pretoria in the Tshwane district in the Gauteng province, South Africa. Twelve Senior Phase mathematics teachers were purposively sampled in 6 public schools before the workshop training. During the workshop training, only 5 teachers participated due to the COVID-19 outbreak. The data collection techniques included pre- and post-interviews, workshop training sessions, classroom observations, field notes, teachers’ reflective journals, teachers’ lesson plans, learners’ workbooks and learners’ worksheets, and data were analysed using thematic analysis. The findings reveal that the teachers acquired adequate knowledge and skills to effectively structure and integrate the lesson plan of teachers’ didactical praxeologies as FA activities into their mathematics classrooms. The findings also reveal that the learners showed more interest and motivation, were actively involved, developed a deeper understanding of mathematics content, and showed increased autonomy in learning. Future research could involve implementing MERLO in all South African provinces and introducing MERLO to other emerging countries. However, the findings of this study are based on a limited sample of teachers and schools, and the recommendation is that, for future studies, more teachers should be involved in the MERLO professional development.

Straipsnyje apžvelgiami prancūzų fenomenologo M. Merleau-Ponty biografijos ir filosofijos svarbiausi bruožai. Vienintele filosofijos problema M. Merleau-Ponty įvardijo pažinimo klausimą. Jis pabrėžė, jog ir psichologizmas, ir transcendentalizmas neapmąsto žmogaus būties klausimo ir įteisina jo subjektyvumo susvetimėjimą: šis subjektyvumas tiriamas tartum daiktas ir todėl natūralizuojamas; kitu atveju jis tiriamas grynosios galimybės erdvėje lyginant jį su absoliutu. Galiausiai M. Merleau-Ponty galutinai atsisako tiek empirizmo, tiek intelektualizmo pozicijos ir nukreipia visas pastangas į autentiškąją egzistenciją. Jos prasmės atskleidimas reikalauja grįžti į gyvąjį jos patyrimą, kuriame konstituojama ši prasmė. Daroma išvada, kad šios egzistencinės fenomenologijos specifiką sudaro atskiro žmogaus subjektyvumo, kaip būties pasaulyje, aprašymas bei subjektyvumo prilyginimo grynajai sąmonei atsisakymas. Apriorinės žmogaus būties struktūros yra žmogaus kūno būties struktūros. Fundamentalioji struktūra-suvokimas konstituoja ir būtį, ir sąmonę.

L'elaborato vuole esplorare le potenzialità del metodo MERLO nella didattica della matematica. Il metodo MERLO (Meaning Equivalence Reusable Learning Object) nasce negli anni novanta in Canada e solo da qualche anno è stato portato nel contesto italiano, principalmente grazie al Dipartimento di Matematica dell'Università di Torino. Questo metodo per incentivare l'apprendimento concettuale si basa sull'uso di schede con rappresentazioni multisemiotiche con lo scopo di evidenziare un Boundary Of Meaning (BOM) ossia un confine di significato. L'affermazione di cui si vuole indagare la conoscenza e le sue diverse rappresentazioni semiotiche rientrano nel BOM mentre rappresentazioni semiotiche simili ma con un significato diverso ne sono esclusi. Nell'elaborato sono stati analizzati quelli che sono i principali fattori che intervengono nell'apprendimento di un concetto. È stato poi esposto in cosa consiste il metodo MERLO e come è stato trasposto in Italia, per meglio capire le potenzialità e criticità nella sua applicazione è stata riportata una sperimentazione didattica in una classe terza di una scuola secondaria di primo grado. Per avere una visione completa dell'introduzione del MERLO nel contesto della didattica della matematica sono state intervistate alcune docenti che ne fanno uso nella propria didattica e è stato intervistato anche il Professor Kennet che ha collaborato con i creatori del metodo. Al termine dell'elaborato sono state indicate le criticità del metodo MERLO e le sue potenzialità.

In questo elaborato vengono riproposti due percorsi di insegnamento e apprendimento svolti all'interno della Scuola di Istruzione Superiore Alessandro Volta di Lodi nell'anno scolastico 2015-2016. Nel primo e stato sviluppato il concetto di angolo, mentre nel secondo è stata affrontata la trigonometria. I due percorsi pongono gli studenti al centro dell'attività didattica, riproducendo in parte il modello Flipped Mastery (Apprendimento Capovolto). Entrambi i percorsi sono stati sviluppati in classi parallele, le attività riguardanti l'angolo sono state proposte in una prima ITIS e in una prima professionale, mentre per lo sviluppo della trigonometria l'attività è stata presentata in una terza ITIS meccanica del diurno e in una terza ITIS elettrotecnica del serale. Alla fine dell'unità sono stati raccolti i dati prodotti dalle due classi e sono stati confrontati i risultati ottenuti.

Le prolusioni e i testi delle lezioni universitarie sono il punto di partenza della ricerca: tali scritti offrono una prospettiva privilegiata – all’interno del mondo giuridico subalpino prima e unitario poi – per valutare se e in che misura questi ‘discorsi giuridici’, per loro natura indirizzati a un pubblico di élite ma non limitato ai soli universitari, abbiano influito non solo sulla vita scientifica, accademica ma pure su quella culturale, politica ed istituzionale. La prima parte della tesi è dedicata ad un’analisi dettagliata del dibattito sull’istruzione legale a partire dalla ‘Riforma Alfieri’ del 1846: si è cercato di offrire un quadro della situazione didattica della Facoltà giuridica torinese nella seconda metà dell’Ottocento, attraverso le parole – ed in specie i testi di lezione – degli stessi protagonisti che si preoccuparono di migliorare la qualità e l’organizzazione dell’insegnamento. Tale cornice aiuta a comprendere meglio il clima e l’ambiente di convinta partecipazione al miglioramento della didattica giuridica in cui si inseriscono le prolusioni e i discorsi inaugurali, che a loro volta rispecchiano le problematiche e gli argomenti topici del periodo, e le lezioni dei docenti della Facoltà subalpina. In proposito emerge l’importante contributo di tre di essi, il Merlo, il Melegari e l’Albini: nella seconda parte della tesi è stato dedicato un capitolo a ciascuno di questi tre giuristi. La scelta è ricaduta su questi tre docenti perché, sebbene molto diversi per origine e percorso di formazione, hanno offerto un diretto contributo personale al miglioramento degli studi legali sia con la collaborazione fattiva ad un perfezionamento legislativo del cursus studiorum legale sia con i loro insegnamenti che erano significativi del nuovo percorso degli studi legali intrapreso dopo la riforma Alfieri del 1846. Il punto di incontro fra i tre studiosi si trova nel comune convinto e profondo interesse per il miglioramento degli studi legali. La lettura dei testi delle loro lezioni ha permesso di poter apprezzare tale sforzo, non solo dal punto di vista dell’impegno per il miglioramento della regolamentazione degli studi nella Facoltà legale, ma anche da quello del contenuto degli insegnamenti.<br>The introducing lectures and the transcriptions of the University of Turin courses in law are the starting point of the research. These documents offer a privileged perspective on the law environment of the subalpine world, and subsequently of united Italy. They allow us to evaluate whether and how these discourses in law, addressed to a privileged public but not restricted to university circles, have had an influence not only on academia but also on the cultural, political and institutional life of the time. The first part of the thesis is dedicated to a detailed analysis of the debate on the structure of legal studies, starting from the 1846 “Alfieri reform”. The goal is to delineate an outline of the situation of didactics of the Turin’s Law School during the second half of the nineteenth century. This is accomplished through the words and the transcriptions of the courses of those same professors that were influential in improving the quality and organization of the teaching. Such analysis helps better understanding the environment of active participation to the amelioration of the didactics of law education in which the inaugural lectures are immersed, reflecting the problems and topics of the time. In the second part of the thesis, the contributions of Professors Merlo, Melegari and Albini are discussed, and a chapter is dedicated to each of them. The analysis of the work of these three scholars is particularly interesting as, even if very different for social origin and academic curriculum, they have offered an important personal input to the enrichment of the law education path, both with an effective collaboration in the legal definition of the cursus studiorum and with their teachings, providing significant examples of the new study path in law (post Alfieri reform). The meeting point between the three scholars is the common interest for the improvement of legal studies. The analysis of the texts of their lectures has allowed this effort to be appreciated, not only under the point of view of the legal definition of the cursus studiorum, but also under that of the contents of their teaching.

To the extent that this analysis is based on the construction of poetry characterized by tension, both the Brazilian poet and the Portuguese poets take Cesário Verde as a precursor of their poetry, and this is the first common aspect that they present. From a possible relation between the works of the poets, seen starting from the same ideal of poetry, we seek to understand how the works of the mentioned poets are related in an evident form, such as the dialogue established between João Cabral and Sophia de Mello through the poets' own notes, or, in a secret way, as in the relationship established between João Cabral and Alberto Caeiro, in which the dialogue is created from the analysis, elucidating common aspects between them. For such, we use the text O sujeito lírico fora de si, by Michel Collot (2013), for the analysis of the formative moments of the work of João Cabral, of the complete work of Alberto Caeiro and of the three movements of the work of Sophia de Mello, from her relationship with the Brazilian poet. The approximations established between the three are guided by the decentralization of the poetic subject and the consequent refusal of memory, by the rigor of construction marked by the ideals of absence of the subject, the lucidity and concreteness and, specifically, by the hallucination in the work of João Cabral and Sophia de Mello. From the study of this poetry that differs from the subjective lyric poetry, we can also see the construction of the landscape as a particular case of containment of subjectivity, analysis guided by the work Poética e filosofia da paisagem, also by Collot. Without intending to erase the differences that mark the particularity of the poetic project of each of the poets, it was sought to consolidate the provisions to the contrary according to their own statements, understanding from the analysis a poetry of constitutive interaction that associates poetic state and construction process.<br>O presente trabalho tem como objetivo investigar o modo como os poetas João Cabral de Melo Neto, Alberto Caeiro e Sophia de Mello Breyner Andresen reconfiguram, mesmo que de forma diversa, uma tradição lírica fundada na expressão dos sentimentos do poeta, ao realizar uma poesia em que a subjetividade é circunscrita em favor de uma realidade exterior. Na medida em que essa análise se baseia na construção de uma poesia que se caracteriza pela tensão, tanto o poeta brasileiro quanto os poetas portugueses tomam Cesário Verde como precursor de suas poesias, sendo este o primeiro aspecto em comum que apresentam. A partir de uma possível relação entre as obras dos poetas, visto partirem de um mesmo ideal de poesia, procura-se depreender como as obras relacionam-se de forma evidente, a exemplo do diálogo estabelecido entre João Cabral e Sophia de Mello por meio dos apontamentos dos próprios poetas, ou, de forma secreta, como na relação estabelecida entre João Cabral e Alberto Caeiro, em que o diálogo é criado a partir da análise, elucidando-se aspectos comuns entre eles. Para tanto, recorre-se ao texto O sujeito lírico fora de si, de Michel Collot (2013), a partir do qual se empreende a análise dos momentos formadores da obra de João Cabral, da obra completa de Alberto Caeiro e dos três movimentos da obra de Sophia de Mello, a partir de sua relação com o poeta brasileiro. As aproximações estabelecidas entre os três norteiam-se pelo descentramento do sujeito poético e pela consequente recusa da memória, pelo rigor de construção marcado pelos ideais de ausência do sujeito, de lucidez e concretude e, especificamente, pela alucinação na obra de João Cabral e Sophia de Mello. A partir do estudo dessa poesia que difere da poesia lírica subjetiva, constata-se também a construção da paisagem como caso particular de contenção da subjetividade, análise orientada pela obra Poética e filosofia da paisagem, também de Collot. Sem a intenção de apagar as diferenças que marcam de forma particular o projeto poético de cada um dos poetas, buscou-se consolidar as disposições em contrário segundo as afirmações deles próprios, depreendendo-se da análise uma poesia de interação constitutiva que associa estado poético e processo de construção.

Assim, na anÃlise do projeto literÃrio de Gerardo Mello MourÃo (8.1.1917 - 9.3.2007) esboÃamos, inicialmente, um painel histÃrico e cultural do perÃodo de criaÃÃo de sua obra O Valete de Espadas (1960). Nessa anÃlise, as obras crÃticas que embasaram nossa reflexÃo foram: A AngÃstia da InfluÃncia (1991) e Um Mapa da Desleitura (1995) de Harold Bloom e Palavras da CrÃtica (1992), organizada por Josà Luis Jobim. Na etapa final da pesquisa, observamos os encontros discursivos que Gerardo Mello MourÃo mantÃm com outros escritores, na busca por um espaÃo de enunciaÃÃo autÃnoma.<br>The writerâs imaginary and aesthetical inclinations are inserted in the historical and cultural context which involves them. However, besides experiencing the social tensions of his time, the writer searches for his autonomy as a literary creator, absorbing and reinterpreting the tradition. Thus, in the analysis of literary project of Gerardo Mello MourÃo from (Jan., 8th, 1917 to March, 9th 2007), we outline firstly, a historical and cultural panel of the period his work was created with O Valete de Espadas (1960). Such investigation receives the theoretical critical orientation of Antronio Candidoâs, Literature and Society (2000), Wilson Martinsâ, HistÃria da Intelectualidade Brasileira (1996), Gilberto de Mello Kujawskiâs, A Crise do SÃculo XX (1991), Vitor Manuel de Aguiar e Silvaâs, Teoria da Literatura (1973) Pierre Bourdieuâs discussive pressuposition, The Rules of Art (2005) and Dominique Maingueneau with The context of the Literary work (2001). Moreover, we investigated the philosophical and aesthetical tendency of the period which interrelates with the writerâs critical positioning regarding the world around him as well as with his notion about literary writing. At the light of theoretical categories such as âMisreadingâ (Harold Bloom) and intertextuality (Julia Kristeva), we inquired into the 2nd chapter, the literary and critical work, Gerardo Melo MourÃoâs interviews and speeches with the purpose of learning about a dialogue which is creative and traditional. In such analysis, the critical writings which were base for our reflexions were: A AngÃstia da InfluÃncia (1991) and Um Mapa da Desleitura (1995) by Harold Bloom as well as Palavras da CrÃtica (1992), organized by Josà Luis Jobim. In the final part of the research we observed the discursive intercourse that Gerardo MourÃo keeps with other writers searching for a space of autonomous emancipation.

Essa investigação recebe a orientação teórico-crítica de: Antônio Cândido, Literatura e Sociedade (2000), Wilson Martins, História da Intelectualidade Brasileira (1996), Gilberto de Mello Kujawski, A Crise do Século XX (1991), Vitor Manuel de Aguiar e Silva, Teoria da Literatura (1973) e os pressupostos discursivos de Pierre Bourdieu, As Regras da Arte (2005) e Dominique Maingueneau, O Contexto da Obra Literária (2001). Além disso, investigamos as correntes filosóficas e estéticas do período, que se inter-relacionam com o posicionamento crítico do escritor acerca do mundo que o rodeia e com sua concepção de escrita literária. À luz de categorias teóricas como “Desleitura” (Harold Bloom) e “Intertextualidade” (Julia Kristeva), examinamos, no 2º capítulo, a obra literária e crítica, as entrevistas e os discursos de Gerardo Mello Mourão, com o intuito de apreendermos um diálogo criativo com a tradição. El grupo es muy reconocido en el país de Panamá por su vasta trayectoria de 67 años en el mercado panameño, en las diferentes industrias donde enfocan su productividad. Principalmente su enfoque es en actividades de producción, procesamiento, distribución y comercialización de alimentos. El análisis se hace para el periodo de junio 2017, donde usamos todos los datos financieros obtenidos de sus estados financieros consolidados publicados. Como empresa pública y cotizando en la bolsa de valores de Panamá, también evaluamos sus estados financieros de los últimos años, enfocándonos principalmente en su estructura de capital (patrimonio, deuda y valor de la empresa), porcentajes de crecimiento de crecimiento previamente vistos y los proyectados. Para hacer una evaluación de valoración adecuada, se pueden usar 2 métodos de valoración de empresa:  Valoración de Empresa por Flujos de Caja Descontados  Valoración de Empresa por Múltiplos En nuestro caso utilizaremos la valoración por método de Múltiplos. Parte esencial para obtener una evaluación correcta es obtener datos de la industria, los cuales podremos usar para comparar a los del Grupo Melo. Para este trabajo, utilizamos la empresa Tyson Food, Inc. como representación de compañía comparativa en la industria. Finalmente, al obtener el valor de la empresa y precio de la acción según método utilizado, comparamos el resultado versus el precio actual al cual se cotiza la acción de la empresa en la Bolsa de Valores de Panamá. La empresa resulta estar ligeramente devaluada según el método. The research presented in this paper is the fruit of an ongoing international collaboration with the goal of enhancing students learning outcomes by implementing and sharing a novel pedagogy for conceptual thinking, and use of an innovative didactical and methodological tool: Meaning Equivalence Reusable Learning Objects (MERLO) that provide student-centered, weekly formative assessments for exploring and discussing conceptual situations in small groups. It was developed, tested, and implemented in Canada at University of Toronto and Ryerson University, as well as in Israel, Italy, Russia, and Australia, in different knowledge domains, including: physics; biology; mathematics; mathematics teacher education; teacher training; developmental psychology; English as a second language; architecture; management; business; project management. Statistical analysis of MERLO data collected since 2002, shows that conceptual thinking enhance learning outcomes and deepens students’ comprehension of the conceptual content of learned material. Conceptual thinking is learnable, and provide metrics to document continuous increase in higher-order thinking skills such as critical conceptual thinking, transfer of knowledge, and problem solving. Pedagogy for conceptual thinking is currently implemented with Brightspace (http://www.brightspace.com/), Integrated Learning Platform (ILP) offered by D2L (http://www.d2l.com/) that supports customizable online pedagogy.

ABSTRACT: Accurate prediction of dynamic pressures is essential for wellbore integrity analysis in deep water wells and wells with narrow operational windows. During the start-up of circulation, a higher circulating pressure is needed due to the gelation effect of drilling fluid. A coupled poro-elasto-dynamic model is developed to examine the effects of pump start-up on borehole stability. Firstly, a transient hydraulic model is developed to predict the temporal variation of pressure in flow conduits during the pump ramp-up periods. A constitutive equation is used to describe the time-dependent rheology of gelled fluid. The hydraulic model is then coupled with a wellbore stability model to study the effect of dynamic pressures on wellbore integrity. The method of characteristics is employed to solve the governing equations of the hydraulic model. Failure analysis is performed to evaluate the gelation effect of drilling fluid and pump ramp-up on borehole stability. The results of the hydraulic model are validated through comparison with field data and that of a published study. The results indicate that maximum circulating pressure is initially attained before approaching an equilibrium circulating pressure. A time-dependent failure can occur in the vicinity of the borehole depending on the magnitude of gel strength and pump ramp-up speed. The study concludes that by ignoring the time-dependent characteristics of drilling fluids the magnitude of circulating pressures in early times can be underestimated. Therefore, an integrated framework is suggested to consider the time-dependent effects of drilling fluids on circulating pressure and hence wellbore stability. The developed poro-elasto-dynamic model can be utilized to improve our understanding of the transient effects of drilling fluids on wellbore stability. 1. INTRODUCTION Drilling hydraulics is one of the most important factors to control the safety of drilling operations. It has received special attention in the past through downhole measurement tools to improve downhole pressure monitoring. However, it is not possible to deploy such tools in sensitive and risky areas. In addition, it is required to have a reliable hydraulic model to predict downhole pressure for design purposes. Several hydraulic models have been developed to estimate the frictional pressure losses during drilling fluid circulation under steady-state conditions (Reed and Pilehvari, 1993; Merlo et al. 1995; Mitchell and Miska, 2011). The major rheological models to describe the pressure losses in pipe and annuli include the Bingham-Plastic, Power-law, and Yield Power-law (Herschel-Bulkley) model. Drilling fluids generally exhibit thixotropic behavior due to their mixture composition (Bourgoyne et al. 1990). Nevertheless, the fluid gelation and thixotropic effect are not commonly considered in the majority of the abovementioned studies.

Abstract:
The long-term motivation for this research is to transfer useful traits from a broad based gene pool of wild species into the narrow base of a cultivated crop in Cucumis. Our primary focus was to use polyploid prior to fertilization as a tool to overcome fertility barriers in the cross between C. melo and C. metuliferus. In conducting this research, we explored all combinations of tetraploid and diploid parents, in reciprocal combinations. Pollinations were made in both the field and greenhouse, using emasculated flowers, moneocious females, and open pollination by insect vectors, with morphological selection criteria. After observations of thousands of ovaries, we still have no definitive proof that this hybridization yielded viable embryos. The most promising results came from using tetraploid C. metuliferus, as the maternal parent in the interspecific hybridization, that set fruit were seeds contained small embryos that did not germinate. To obtain fruit set, it was important to rear plants in a cooler sunny greenhouse, as would be found in late winter/early spring. A second interspecific hybrid between wild and cultivated Cucumis, C. hystrix x C. sativus, yielded fertile progeny for the first time, while concomitantly working toward our primary goal. Two distinct treatments were necessary; 1) special plant husbandry was necessary to have the wild species produce fruit in cultivation, and 2) embryo rescue followed by chromosome doubling in vitro was required for fertility restoration. Backcrosses to crop species and resistance to nematodes are compelling areas for further work.

Project objectives: 1) Characterization of variation for yield heterosis in melon using Half-Diallele (HDA) design. 2) Development and implementation of image-based yield phenotyping in melon. 3) Characterization of genetic, epigenetic and transcriptional variation across 25 founder lines and selected hybrids. The epigentic part of this objective was modified during the course of the project: instead of characterization of chromatin structure in a single melon line through genome-wide mapping of nucleosomes using MNase-seq approach, we took advantage of rapid advancements in single-molecule sequencing and shifted the focus to Nanoporelong-read sequencing of all 25 founder lines. This analysis provides invaluable information on genome-wide structural variation across our diversity 4) Integrated analyses and development of prediction models Agricultural heterosis relates to hybrids that outperform their inbred parents for yield. First generation (F1) hybrids are produced in many crop species and it is estimated that heterosis increases yield by 15-30% globally. Melon (Cucumismelo) is an economically important species of The Cucurbitaceae family and is among the most important fleshy fruits for fresh consumption Worldwide. The major goal of this project was to explore the patterns and magnitude of yield heterosis in melon and link it to whole genome sequence variation. A core subset of 25 diverse lines was selected from the Newe-Yaar melon diversity panel for whole-genome re-sequencing (WGS) and test-crosses, to produce structured half-diallele design of 300 F1 hybrids (MelHDA25). Yield variation was measured in replicated yield trials at the whole-plant and at the rootstock levels (through a common-scion grafted experiments), across the F1s and parental lines. As part of this project we also developed an algorithmic pipeline for detection and yield estimation of melons from aerial-images, towards future implementation of such high throughput, cost-effective method for remote yield evaluation in open-field melons. We found extensive, highly heritable root-derived yield variation across the diallele population that was characterized by prominent best-parent heterosis (BPH), where hybrids rootstocks outperformed their parents by 38% and 56 % under optimal irrigation and drought- stress, respectively. Through integration of the genotypic data (~4,000,000 SNPs) and yield analyses we show that root-derived hybrids yield is independent of parental genetic distance. However, we mapped novel root-derived yield QTLs through genome-wide association (GWA) analysis and a multi-QTLs model explained more than 45% of the hybrids yield variation, providing a potential route for marker-assisted hybrid rootstock breeding. Four selected hybrid rootstocks are further studied under multiple scion varieties and their validated positive effect on yield performance is now leading to ongoing evaluation of their commercial potential. On the genomic level, this project resulted in 3 layers of data: 1) whole-genome short-read Illumina sequencing (30X) of the 25 founder lines provided us with 25 genome alignments and high-density melon HapMap that is already shown to be an effective resource for QTL annotation and candidate gene analysis in melon. 2) fast advancements in long-read single-molecule sequencing allowed us to shift focus towards this technology and generate ~50X Nanoporesequencing of the 25 founders which in combination with the short-read data now enable de novo assembly of the 25 genomes that will soon lead to construction of the first melon pan-genome. 3) Transcriptomic (3' RNA-Seq) analysis of several selected hybrids and their parents provide preliminary information on differentially expressed genes that can be further used to explain the root-derived yield variation. Taken together, this project expanded our view on yield heterosis in melon with novel specific insights on root-derived yield heterosis. To our knowledge, thus far this is the largest systematic genetic analysis of rootstock effects on yield heterosis in cucurbits or any other crop plant, and our results are now translated into potential breeding applications. The genomic resources that were developed as part of this project are putting melon in the forefront of genomic research and will continue to be useful tool for the cucurbits community in years to come.

Abstract:
This project has generated a systematic survey of DNA polymorphism in Cucumis melo. An RFLP and RAPD survey of the major cultivar groups and botanical varieties of this species has been conducted, with the purpose of assessing the degree of molecular variation and phylogenetic relationships within the melon germplasm and, at the same time, develop sets of markets suitable for mapping the melon genome. Additional activities regarding variation in the melon germplasm in fruit traits and regeneration ability have been initiated as well. The necessary populations required for the development of a molecular map of the C. melo genome have been prepared. An F2 that segregated for 4 viral resistances, powdery mildew resitance and sex type has been derived from a PI 414723 x Topmark cross, and a RILs population has been prepared from it. We have confirmed the resistances in the population and have analyzed the genetic relationships between these resistances. Progress toward the construction of a molecular map of C. melo and the development of markers linked to those traits is described. We have so far screened the first few tens of markers in the F2 population, and many additional ones were screened in DNA bulks prepared from such population.

Fruit quality is determined by numerous genetic traits that affect taste, aroma, texture, pigmentation, nutritional value and duration of shelf-life. The molecular basis of many of these important traits is poorly understood and it’s understanding offers an excellent opportunity for adding value to agricultural products. Improvement of melon fruit quality was the primary goal of the project. The original objectives of the project were: The isolation of a minimum of 1000 fruit specific ESTs. The development of a microarray of melon fruit ESTs. The analysis of gene expression in melon using melon and tomato fruit enriched microarrays. A comprehensive study of fruit gene expression of the major cucurbit crops. In our current project we have focused on the development of genomics tools for the enhancement of melon research with an emphasis on fruit, specifically the first public melon EST collection. We have also developed a database to relay this information to the research community and developed a publicly available microarray. The release of this information was one of the catalysts for the establishment of the International Cucurbit Genomic Initiative (ICuGI, Barcelona, Spain, July 2005) aimed at collecting and generating up to 100,000 melon EST sequences in 2006, leveraging a significant expansion of melon genomic resources. A total of 1000 ESTs were promised under the original proposal (Objective 1). Non-subtracted mature fruit and young fruit flesh of a climacteric variety in addition to a non-climacteric variety resulted in the majority of additional EST sequences for a total of 4800 attempted reads. 3731 high quality sequences from independent ESTs were assembled, representing 2,467 melon unigenes (1,873 singletons, 594 contigs). In comparison, as of June 2004, a total of 170 melon mRNA sequences had been deposited in GENBANK. The current project has thus resulted in nearly five- fold the number of ESTs promised and ca. 15-fold increase in the depth of publicly available melon gene sequences. All of these sequences have been deposited in GENBANK and are also available and searchable via multiple approaches in the public database (http://melon.bti.cornell.edu). Our database was selected as the central location for presentation of public melon EST data of the International Cucurbit Genomic Initiative. With the available unigenes we recently constructed a microarray, which was successfully applied in hybridizations (planned public release by August 2006). Current gene expression analyses focus on fruit development and on comparative studies between climacteric and non-climacteric melons. Earlier, expression profiling was conducted using macroarrays developed at the preliminary stage of the project. This analysis replaced the study of tomato microarray following the recommendations of the reviewers and the panel of the original project. Comparative study between melon and other cucurbit crops have begun, mainly with watermelon, in collaboration with Dr. Amnon Levi (USDA-ARS). In conclusion, all four objectives have been addressed and achieved. In the continuation project that have been approved we plan to apply the genomic tools developed here to achieve detailed functional analyses of genes associated with major metabolic pathway.

In different crop plants, combining ability and heterosis are used as important diagnostic tools for assessing the performance of parental genotypes and their hybrids. This research aimed to evaluate heterotic and combining ability effects in the diallel crosses of melon (Cucumis melo L.) for yield- and quality-related traits. Seven melon (C. melo L.) genotypes were grown and crossed in a complete diallel fashion to produce F1 hybrids. During the 2019 crop season, 49 melon genotypes (7 parents + 42 F1 hybrids) were grown in a randomized complete block design with three replications. Observations were made for seven characters. Analysis of variance revealed significant (P ≤ 0.01) differences among the melon genotypes for harvest age, fruit flesh thickness, fruit total soluble solids, fruit length, and fruit diameter and merely significant differences (P ≤ 0.05) for fruit weight. Combining ability analysis revealed that mean squares due to general combining ability (GCA) were significant for fruit diameter but were nonsignificant for all other traits. However, mean squares due to specific combining ability (SCA) were significant for all traits. The parental genotypes PK-165, PK-464, and PK-669 exhibited the highest and desirable GCA effects for yield and quality traits. Hence, these genotypes could be used to generate high-yielding hybrid/open-pollinated cultivars. GCA:SCA ratios further revealed that the traits of harvest age, fruit flesh thickness, fruit total soluble solids, fruit length, and fruit weight were controlled by dominant gene action, whereas fruit diameter was managed by additive and dominant genes. The majority of the traits were controlled by nonadditive gene action, verifying that the said breeding material could be efficiently used for the production of hybrid cultivars on the basis of heterotic effects.
