Abstract:
Background:Mercury is a toxic metal that is present in small amounts in the environment, but its level is rising steadily, due to different human activities, such as industrialization. It can reach humans through the food chain, amalgam fillings, and other sources, causing different neurological disorders, memory loss, vision impairment, and may even lead to death; making its detoxification an urgent task. Methods:Various physical and chemical mercury remediation techniques are available, which generally aim at: (i) reducing its mobility or solubility; (ii) causing its vaporization or condensation; (iii) its separation from contaminated soils. Biological remediation techniques, commonly known as bioremediation, are also another possible alternative, which is considered as cheaper than the conventional means and can be accomplished using either (i) organisms harboring themeroperon genes (merB,merA,merR,merP,merT,merD,merF,merC,merE,merHandmerG), or (ii) plants expressing metal-binding proteins. Recently, differentmerdeterminants have been genetically engineered into several organisms, including bacteria and plants, to aid in detoxification of both ionic and organic forms of mercury. Results:Bacteria that are resistant to mercury compounds have at least a mercuric reductase enzyme (MerA) that reduces Hg+2to volatile Hg0, a membrane-bound protein (MerT) for Hg+2uptake and an additional enzyme, MerB, that degrades organomercurials by protonolysis. Presence of bothmerA andmerB genes confer broad-spectrum mercury resistance. However, merA alone confers narrow spectrum inorganic mercury resistance. Conclusion:To conclude, this review discusses the importance of mercury-resistance genes in mercury bioremediation. Functional analysis ofmeroperon genes and the recent advances in genetic engineering techniques could provide the most environmental friendly, safe, effective and fantastic solution to overcome mercuric toxicity.

Abstract:

Abstract:
ABSTRACT A DNA library of pRJ28, a large linear plasmid encoding mercury resistance, was constructed, and the mercury resistance genes were cloned. The 5,921-bp sequence was analyzed and showed a high degree of similarity to the Streptomyces lividans 1326 mercury resistance operon. Genes merR, merT,merP, and orfIV were found in a similar order and in a single transcription unit. merA andmerB were found to be transcribed in the opposite direction to genes merR, merT, merP, andorfIV, as in S. lividans 1326. A novel putative regulatory gene, orfX, was found 22 bp downstream ofmerA. orfX encodes a 137-amino acid protein with a potential helix-turn-helix motif in the N-terminal domain, characteristic of the MerR family of transcriptional regulators. Transcriptional studies showed that orfX is cotranscribed with merA and merB. It is hypothesized thatorfX plays a role in the regulation of the mercury resistance operon, probably by binding at the MerR operator site.

Pounds, Megan Pounds. "(Un) Forming Nature: Kurt Schwitters's Merz Barn (1947-1948)." Thesis, University of Oregon, 2016. http://hdl.handle.net/1794/20552.

Full text
Abstract:
This thesis centers on Kurt Schwitters’s Merz Barn (1947-1948), exploring the relationship between nature and the Merz principles of formung (forming) and entformung (un-forming) within the context of this late work. The Merz Barn, the last of Schwitters’s Merzbauten, has yet to receive the extensive level of research accorded to its famous Hannover predecessor, resulting in an underdeveloped grasp of the project as a whole within Merzbau scholarship. The present study considers Schwitters’s increasing orientation towards nature as a model for artistic creation to elicit an understanding of the ways in which his paradoxical Merz formula, “Formen heißt entformeln,” evolved during his period of exile. I contend that Schwitters employed the organic processes of natural growth and decay to realize the principles of formung and entformung in his Merz Barn. Furthermore, the sculptural interior underscores the dialectical exchange between forming and un-forming, highlighting the liminal space between the opposing processes.

Abstract:
Plasma protein C inactivates the activated coagulation factors V and VIII. The assay of protein C is important, because a protein C deficiency is associated with a thrombotic tendency. We therefore evaluated 5 commercial assays in 49 normal volunteers, 48 patients suspected of deep vein thrombosis (DVT) of the leg but with negative impedance plethysmography (IPG), and 52 patients with DVT proven by IPG. The assays were rocket electrophoresis (Merz and Dade antibody), ELISA (Boehringer Mannheim), 2 chromogenic activity assays (Behringwerke and Kabi) and a clotting assay (Behringwerke). Coumarin therapy was used by 13 DVT positive, and 3 DVT negative patients. Results are presented in the table.TABLE: In correlation 1 and 2, the assay results all non-coumarin treated individuals (n = 133) were compared with rocket electrophoresis and ELISA resp.In the non-coumarin treated patients, both in the DVT positive and in the DVT negative patient group one protein C deficiency was detected by all assays.Based upon the large assay VC (ROCKET) and normal range (B. CLOT), and poor correlation of the assays with the ELISA (ROCKET, B. CLOT) we conclude that the ELISA, B.CHROM and K.CHROM are to be preferred. However, as B.CHROM does not need a plasma absorption step it is somewhat preferable for activity assays.
