In this thesis, <i>in situ</i> hybridisation histochemistry was the principal technique applied to the study of a number of specific species of messenger RNA (mRNA) in tissue from normal and diseased post-mortem human brains. Gs and Go are members of the heterotrimeric G protein family whose members are crucial to cellular responsiveness. APP and Goa mRNA levels were found to be reduced in the dentate gyrus of AD sufferers in comparison to controls and were positively correlated in this region irrespective of diagnosis. The APP gene is encoded on chromosome 21 and APP mRNA levels are increased

Multi-messenger astronomy is a very hot topic in the astrophysical community. A messenger is something that carries information. Different astrophysical messenger types are photons, cosmic rays, neutrinos, and gravitational waves. They all carry unique and complementary information to one another. The idea with multi-messenger astronomy is that the more different types of messengers one can obtain from the same event, the more complete the physical picture becomes. In this thesis I study the multi-messenger emission from gamma-ray bursts (GRBs), the most luminous events known in the Universe.

Release of Ca2 + from intracellular stores may be initiated by ci+ itself, or by a Ca:!T-releasing second messenger such as inositol 1,4,5-trisphosphate. Regulation of gene expression is a complex process in mammalian cells with many levels of control. In recent years non-coding RNAs in the form of microRNAs (miRNA) have surfaced as important regulators of protein coding genes, with biologically important roles in development, differentiation and cell growth. In this dissertation the complex interactions between miRNAs and mRNAs in ovarian cancer are investigated using a combination of computational and experimental techniques. In vitro studies and current models predict that increases in levels of miRNA should result in corresponding decreases

The effect of glucose on ribosomal protein mRNA stability was examined. The addition of glucose, to a final concentration as low as 0.01%, resulted in a dramatic, transient increase in ribosomal protein mRNA stability. It has been demonstrated that protein kinase A (PKA) activity plays an important role in the up-regulation of ribosomal protein gene transcription in response to glucose addition. PKA pathway is required for the transient stabilisation of ribosomal protein mRNA following glucose addition, mRNA half-lives were determined in a PKA mutant strain. Abstract:
CHOP, or Gadd153, is a 29 kDa protein that plays a pivotal role in the mediation of cellular stress-induced cell death. The expression of the gene encoding CHOP/Gadd153 is regulated at both the transcriptional and post-transcriptional levels. Compared to transcription, the regulation of Chop gene expression at the post-transcriptional level is much less understood. In this study, the role played by the 3’-untranslated region of CHOP mRNA (3UTRChop) in mediating mRNA stability was examined. A reporter plasmid was constructed so that the mRNA expressed has 3UTRChop as its 3’-untranslated region

Abstract:
Polyribosome aggregation state in growing tissues of barley and wheat leaf or stems of pea and squash was studied in relation to seedling growth and water status of the growing tissue in plants at various levels of osmotic stress. It was found to be highly correlated with water potential and osmotic potential of the growing tissue and with leaf or stem elongation rate. Stress rapidly reduced polyribosome content and water status in growing tissues of barley leaves; changes were slow and slight in the non-growing leaf blade. Membrane-bound and free polyribosomes were equally sensitive to stress
