The quantum dots are very attractive due their optical properties and the possibility to insert several different functions on their surface. Full textIonic gradient changes are essential in almost every cellular and bacterial metabolic processes. Therefore, any malfunction that perturbs these ion concentrations can induce major illnesses in the affected physiological solution or biological tissue. Amongst all the chemical species influenced by cell metabolism, protons are key to many events; enzyme activity, protein structural changes, and activation of membrane transporters. In this doctoral thesis, pH-sensitive fluorescent nanoparticles were developed and grafted on planar substrates in order to probe the culture medium of different cells in vitro. These innovative analytical tools for biomedical research offer the possibility to image quantitative pH values in microscopy with both spatial (~1 μm) and temporal resolutions (<300 ms). As such, the sensors were optimized in a structural parametric study with regards to the nanoarchitecture design. These particles comprise a metallic core with plasmonic properties to enhance the fluorescence of molecules incorporated into a porous silica shell. The core size, the spacing layer thickness, the spectral overlap between the plasmon and the fluorescence excitation/emission bands, and the sensitive moieties were carefully chosen to take advantage of the plasmonic range of enhancement in concentric architectures. Since the covalent encapsulation of molecules is highly adaptable for many silanized fluorophores, core-multishell architectures were synthesized as a proof of concept which allows simultaneous measurements of pH and halides (Cl-, Br-, I-), and signal normalization with an internal reference onto the same nanoparticle. The ratiometric correction compensates for fluctuations in light intensity and experimental errors with sensor concentration, photobleaching and environmental effects as a function of time. Finally, this analytical strategy was applied in a spectroscopic study of fluorescein-doped nanoparticles as quantitative pH-sensitive markers implemented in biological cell cultures. Particularly, the core-shell nanoarchitectures were functionalized with a silane component and immobilized at the surface of a silica substrate by way of complementary click coupling. These improved microscope coverslips were evaluated as a fluorescent sensing surface for the culture of adhesive excitable cells – e.g. cardiac fibroblasts and neurons, but also for the growth of bacterial biofilms and the study of multiphase interfaces in microfluidic devices.

Full textPerovskite oxides can be used as sensitive element in pH sensors. A high ionic conductivity, mechanical and chemical resistances allow a fast response time and some potential applications in industry, such as food industry. Firstly, the crystallographic structure and the conduction properties of various oxides were studied before testing their pH sensitivity. Secondly, the optimized conditions to obtain a ceramic sensitive membrane starting from powders synthesized by various methods were determined. Moreover, the characteristics of the pH sensor were determined: response time, selectivity, influence of the redox species… Finally, among the studied compounds, the Li0. 30La0. 57TiO3 (LLTO) compound which exhibits the highest sensitivity to the pH was selected as model for studying the mechanism of pH detection. In particular, there mechanisms were suggested: the oxydo-reduction, the ionic exchange and the reaction of the -OH groups at the oxide surface. Experimental results show that the -OH groups at the oxide surface are responsible of the pH sensitivity of the ceramic membranes. The pH detection property by a potentiometric sensor of this type is unique and innovative. This work will allow the development of these pH sensors for practical uses

déduire le rapport h/M, où h désigne la constante de Planck
et M la masse de l'atome considéré, afin d'obtenir la valeur
de la constante de structure fine. Le principe de
l'expérience est le suivant. A partir d'un nuage d'atomes froids,
nous sélectionnons une classe de vitesse subrecul à l'aide d'une
transition Raman. Ensuite, nous accélérons les atomes de manière
cohérente à l'aide d'une onde stationnaire dans le référentiel de
l'atome (cet effet est analogue aux oscillations de Bloch en
physique du solide). Enfin, nous mesurons la vitesse finale des
atomes par une nouvelle transition Raman. La connaissance des
vitesses finale et initiale nous permet de remonter à la quantité
de mouvement transférée et donc à la vitesse de recul de l'atome.
Nous présentons dans ce manuscrit les premiers résultats de ce
travail expérimental.

Full textThe interest in electrochemical microsensors ChemFETs (Chemical Field Effect Transistor) continues unabated, stimulated by its wide range of applications. The simulation, the fabrication and the characterization of different ChemFET's structures (normally-off and normally-on) have been carried out during this work. The technological process and the packaging of normally-on ChemFETs sensors have been successfully developed. An analysis of the influence of the functioning ChemFET's mode on their characteristics (sensitivity, linearity. . . ) has been done using an electrochemical characterization of chemical sensors. Finally, pH-ChemFETs (SiO2/Si3N4 gate) microsensors have been adapted to the ammonium ion NH4+ detection thanks to a polysiloxane (PSX) membrane containing nonactine as ionophore, leading to the realization of pNH4-ChemFETs (SiO2/Si3N4/PSX gate). The ion sensitive film was deposed using photolithography techniques. The performance of the pH-ChemFETs and pNH4-ChemFETs have been experimentally validated for the concentration range pH = [2-11] and pNH4 = [1-5] respectively

Full textThe different compartments of the hydrosphere are major sinks or sources of CO2 and the role of freshwaters in the global carbon cycle has been recently reevaluated as substantial. Freshwaters show variations of the carbonate system variables at hourly time scales and in a large range. A key limitation for the carbonate system studies is the necessity of accurate, in-situ and high frequency measurements that would provide data with suitable spatial and temporal resolutions to enlighten the processes affecting the carbonate system. With the aim of in-situ and high frequency sensor development to assess the freshwaters whole carbonate system with the simultaneous measurement of the pH-Alkalinity couple, spectrophotometric measurement of these parameters is developped. This work focuses mainly on the spectrophotometric alkalinity (Alk) measurement, which is an original contribution. This new method, consists to neutralize all the basic species taken into account in the Alk by a weak acid (formic acid, HFo) mixed with a pH sensitive dye (bromophenol blue, BPB) to measure accurately the pH end point value and thus deduce the Alk of the sample. After a step of laboratory adjustement, the performance of the spectrophotometric methods is assessed in low pH buffer solutions for pH, commercial mineral and spring waters for Alk and natural freshwaters for both, by a comparison to conventional potentiometric measurements and theoretical calculations. PH measurements show a precision of ±0. 005; Alk measurements show a precision of ±7μM. For Alk measurement, the best accuracy is obtained by downplaying the excess acid term and thus its associed uncertainty. Additionnaly, for both pH and Alk, accuracy relies upon the performance of the spectrophotometer, carefully prepared and defined indicator solution and all very controlled and reliable thermodynamic constants with their temperature and ionic strength dependence. After this step that provide strong theoretical basis which precision and accuracy meet the requirements of scientist working on freshwaters systems, the spectrophotometric pH-Alk method was integrated as an in-situ and high frequency sensor. The integration started by a functioning assessment that leads to some modifications on the prototype. Additionnaly, the integration bring to light others optical, mechanical and chemical troubles that were solved before the method validation where previous performances were increased: pH measurements show a precision of ±0. 002 and Alk measurements show a precision of ±3μM. Finally, the prototype sensor was successfully tested on natural freshwaters (a carbonated french lake and the Seine river). These deployment shows the sensor sturdiness but reveal the difficulty of Alk measurement in waters with high turbidity, that necessitate a filtration step before spectrophotometric measurements

Full textIn the last decade, a new rational strategy for fine optimization of industrial interest metabolites production has been achieved by means of metabolic engineering. This field does not only limits to ponctual genomic manipulations in the producing cells, but requires a deeper insight of their phisiology. In this context, a part of carbohydrate (glucose) metabolism in the pristinamycins producing Streptomyces pristinaespiralis has been investigated during trophophase and idiophase. Specific activities for some enzymes belonging to the central metabolism were assayed during batch cultures and showed a general diminution before the antibiotic being excreted. Another basic physiological parameter is the intracellular pH (pHi), which not only affects enzymatic activities but also provides informations about intracellular metabolism. A novel method for its determination has been developped by means of epifluorescence microscopy combined to image analysis. PHi during batch fermentations undergone severe fluctuations correlated to pristinamycins production and succinate and lactate excretions. Finally all parameters measurements were performed during different dilution rates chemostat. Cyclic productions of pristinamycins were observed due to the pristinamycins repression on their own synthesis. This study aUowed quantification of enzymatio contents for different growth rates of non producing cells and revealed pHi changes of a lesser magnitude

Full textPH measurement by integrated microelectronic devices has been developed with the advent of devices like ISFET ( Ion Sensitive Field Effect Transistor). These devices are limited by Nernst law at 60mV/pH of sensitivity. With the development of micro-technology, the production of sensors like SGFET (Suspended Gate Field Effect Transistor) became possible. These structures, realized by surface micromachining and conventional MOS processes, can achieve much higher sensitivity. After a review of different methods of measuring pH and their basic principles, the document describes the manufacturing processes of microelectronics sensors SGFET. Then this work deals with the design and implementation of an automated measurement of pH system. This system is based on sensors SGFET arranged in a matrix. This manuscript presents firstly the various options of sensors bias and in a second part, it develops the system architecture. Finally, first results on matrix are presented. Thus the best measurement protocol to ensure reliability and good sensitivity here is a few seconds duration sampling of the drain current with constant gate voltage and drain voltage. This method has achieved reproducibility in the pH sensitivity and reliability over time that could even lead to measures during a long time on the order of 150 hours. The design of an addressing and automated measurement systeme allowed to take advantage of this matrix aspect by making statistical treatment of measurements leading then to a fully operational system

Full textThis work is part of a sustainable agriculture context that aims to consume fewer and fewer inputs while maintaining increasing biomass production levels, both in terms of quantity and quality. Such levels can only be achieved if cultivated plants can develop under optimal conditions, including access to the mineral elements essential for their growth. Many factors are involved in the availability of nutrients in the soil for crops such as pH or oxidation-reduction potential (Eh). Several authors have shown that plants grow within a specific Eh/pH range and are able to alter these parameters in the rhizosphere to ensure intracellular homeostasis. The final objective of this study is to understand the effects of soil redox potential on mobility and availability of nutrients to the plant. The need to carry out measurements for both Eh and pH, in situ in the soil with plants is a real matter. Measuring soil Eh is essential but complex due to the lack of measurement reliability resulting from high temporal variability and metrological challenges. This work proposes practical advancements for measuring Eh in soils using combined electrodes. The protocol was applied to monitor a sunflowers pot culture (1L) for a 70 days campaign under different soil and climate conditions. The relationships between soil Eh / pH and the availability of nutrients presents in the soil (N, P, K, Ca, Mg, S, Fe, Mn, B, Zn, Cu and Mo) has been analysed. This study has highlighted the impact of nitrogen and iron in soil Eh values. While some elements (N, Fe, Mn, S) are directly impacted by soil Eh and pH variations, others (Mg, Ca, P, Cu, Zn, Mo) have their mobility and availability indirectly affected. Considering all these results, the objective of this experiment was to determine if soil Eh would in any way provide an indicator that could represents the "favourable state" of soil for plants growth. To achieve this objective a new parameter, called Ehcumul, has been proposed, similar to the degree day, which describes, in agronomy, the temperature needs for plants growth. Strong correlations between Ehcumul and sunflower growth revealed the existence of optimal soil Eh conditions: 440 mV/ENH, in our experiment. Assuming that biological activity of the soils could play a central role in Eh values, sunflower monitoring was also carried out on soils previously sterilized by drying (105°C for 5 days). The dissolved organic carbon (DOC) contents of these soils is increased compared to non-sterilised soils, due to degradation of the most labile organic matter. The consequence was a moderated and constant Eh value. These results open up perspectives on the impact of soil organic matter to maintain optimal soil redox potential values for plant growth

Full textOrganic functions of the human body are linked to biological constants. Variations of these constants induce pathological troubles. Among these constants, the pH is the central subject of this PhD work. In living beings, biological functions are related to either acid or alkaline constants. Indeed, the action of a protein depends on the surrounding pH. An inadequate value of the pH makes the proteins non active which is deleterious for the organism. There exist a need for pH sensors which can be used in the human body for clinical applications (macroscopic scale), on cells in culture for biology researches (mesoscopic scale) or at a cell membrane level for more fundamental studies (microscopic scale). Among the wide range of technologies potentially useful for these applications, fiber optic fluorescence pH sensing offers the possibility to be adapted to the three above mentioned dimensional scales. This PhD dissertation addresses these constraints by studying fluorescence fiber optics pH sensors using two kind of pH indicators: SNARF and fluorescein. Together with these experimental studies, mathematical descriptions of the fluorescence properties of these indicators are proposed. They allow progressing towards calibration free pH sensing

Full textThe considerable rise of eutrophication in water bodies has led to highly instrumented water resource recovery facilities (WRRFs) that can perform nutrient removal processes. To ensure the efficiency of these processes, several parameters that influence the performance of WRRFs need to be well thought out. The latter requires monitoring strategies composed of on-line and off-line methods. Lately, on-line measurements have contributed significantly to monitor and characterize the quality of water and wastewater. However, on-line measurements are not applicable or not implemented yet for some specific areas. For this, off-line laboratory methods are welcome alternatives. Titrimetry is one of the examples of a low-cost off-line method that allows characterizing aquatic streams. Concerning titrimetric monitoring of wastewater, a Titrino device was installed in the laboratory of the 12 m3 pilEAUte WRRF located at Université Laval. The wastewater feeding the plant is coming from a student residence building on campus. The water pumped to the station is feeding a storage tank with a volume of 5 m3 , from which it is then pumped to a primary clarifier. The influent stream is then split into two similar treatment lines, composed of several bioreactors. These two lanes are followed by two secondary settling tanks. In this work, sampling campaigns were performed to compare the sensor data and the off-line titrimetric measurements in a framework of research projects in which industrial partners are involved. The goal of this study was to investigate the efficiency of using titrimetry to analyze and characterize influent and effluent samples of a WRRF. Two data interpretation methods, buffer capacity and PHREEQC, were tested and their performances in estimating the concentration of the concerned buffers were evaluated. For better model estimation, first, some of the lab procedures were improved. It was found out that nitrogen blanketing and CO2 stripping are necessary to perform reliable titration, and thus, to measure other concerned buffers besides alkalinity, such as ammonia and volatile fatty acids (VFA). Moreover, to validate the results of the buffer capacity model and to have a complete description of the equilibrium reactions of the chemical system under study, a titration simulation model was successfully built in PHREEQC with the PHREEQXCEL interface. This titration lab simulation was extended with the OpenSolver, an open-source Excel add-in, which allows to reliably perform the parameter estimation needed to find the concentration of the different species in the sample. In addition, PHREEQC’s database was modified to include all the model components and their essential chemical information for the speciation calculations.After introducing the mentioned lab and modelling improvements, the reliability of the titrimetric measurements for monitoring the performance of nutrient removal was enhanced.
