Abstract:
The trochophora larva, which is so well known from the marine plankton, is central to our understanding of the evolution of a large branch of the bilaterians. Two theories for this larval type have been prevalent, the trochaea theory and the theory proposed by Ivanova-Kazas. The embryology, or more precisely the cell-lineage, of these larvae seems to be central for our understanding of their origin, but important details have been missing. According to the trochaea theory, a circumblastoporal ring of blastomeres differentiates to follow the convoluted shape of the conspicuous ciliary bands of the larvae, with prototroch and metatroch around the mouth, forming a filtering system, and telotroch around the anus. According to the Ivanova-Kazas theory, the blastomeres with the ciliary bands develop through specialization of rings of cells of the general ciliation in a lecithotrophic larva. Now, a new cell-lineage study of the gastropod Crepidula has shown that the ring of cells at the edge of the blastopore develops into the band of cells carrying prototroch and metatroch, characteristic of the trochophora. This gives strong support to the trochaea theory.

Abstract:
The origin and radiation of the major metazoan groups can be elucidated by phylogenomic studies, but morphological evolution must be inferred from embryology and morphology of living organisms. According to the trochaea theory, protostomes are derived from a holoplanktonic gastraea with a circumblastoporal ring of downstream-collecting compound cilia (archaeotroch) and a nervous system comprising an apical ganglion and a circumblastoporal nerve ring. The pelago-benthic life cycle evolved through the addition of a benthic adult stage, with lateral blastopore closure creating a tube-shaped gut. The archaeotroch became differentiated as prototroch, metatroch and telotroch in the (trochophora) larva, but was lost in the adult. The apical ganglion was lost in the adult, as in all neuralians. Paired cerebral ganglia developed from the first micromere quartet. The circumblastoporal nerve became differentiated into a pair of ventral nerve cords with loops around mouth (the anterior part of the blastopore) and anus. Almost all new information about morphology and embryology fits the trochaea theory. The predicted presence of a perioral loop of the blastoporal nerve ring has now been demonstrated in two annelids. Alternative ‘intercalation theories’ propose that planktotrophic larvae evolved many times from direct-developing ancestors, but this finds no support from considerations of adaptation.

AbstractThis article traces the evolution of the archangel Metatron from his inception to his identification with Enoch. It argues that the name Metatron was derived from the term metator, which was used in Palestine to describe the role of the angel of the Lord in Exod 23:20-21. It was only in Babylonia that Metatron became a name of a specific angel, where, however, he was never identified with Enoch. The first attestations of Metatron in Palestinian sources are only from the beginning of the seventh century CE, and they know nothing of Enoch. The earliest evidence for Enoch-Metatron is only found in Palestinian sources from the eighth century. Enoch-Metatron is therefore neither an ancient esoteric Palestinian tradition nor a Babylonian creation, but rather a late Palestinian innovative synthesis of Babylonian Metatron and Byzantine Enoch trajectories, which resulted in a new hybrid figure.

The article examines the reception and transmission of traditions about the figure of Enoch/Metatron in Sasanian Babylonia, and particularly the emergence of Metatron speculation in the Babylonian Talmud and 3 Enoch, by reading these traditions in light of Zoroastrian and Manichaean reports of the Iranian hero, Yima. The figure of Enoch/Metatron was reimagined and reconfigured by the Babylonian authors so as to resemble local Yima traditions, though the process of translating and repackaging the figure of Enoch in the image of his Iranian counterpart was not merely a conscious act of comparison, in which an analogy is drawn in an attempt to highlight particular aspects common to both figures; it was an expression of a more comprehensive discourse of identification.Beyond close parallels in the depictions of these figures, the connections between Metatron speculation and the Zoroastrian and Manichaean Yima traditions are supported by an identification of Yima with the son of man implied in two Sogdian fragments of the Manichaean Book of Giants. The syncretic atmosphere that pervaded Sasanian culture in general and the Manichaean identification of Yima with the son of man in particular facilitated, and perhaps reinforced, the refiguring of Enoch/Metatron in the Babylonian Talmud and 3 Enoch in the image of local Yima traditions.

Abstract:
The polar lobe of Ilyanassa is asymmetrically partitioned into the D lineage of cells. Two of these cells, 3D and 4d, induce proper axial cell fate patterning in the embryo. Based on known embryological data in Ilyanassa, I hypothesized that Notch signaling would be required for this induction. I found that Notch signaling is required for cell fates induced by 4d and is temporally required well after 4d induction. Based on these results, I hypothesize that Notch signaling is involved in a reciprocal induction between the micromeres and the macromeres (endoderm) resulting in the maintenance of micromere fate induction and endoderm specification.Loss of the polar lobe results in the loss of cell fate induction by 3D/4d. Therefore, I hypothesized that proteins are asymmetrically bequeathed to the inducing D lineage cells by the polar lobe. To test this hypothesis, I compared global protein differences between two cell stage intact embryos, lobeless embryos, and isolated polar lobes by 2-Dimensional Electrophoresis analysis. I found several (12) quantitative differences between these samples including four spots enriched in the polar lobe isolates. I identified voltage-dependent anion-selective channel (VDAC) as one of the candidate proteins enriched in polar lobe isolates. I propose that VDAC is asymmetrically distributed by the polar lobe to the D cell and that it may function in D cell induction and mesendoderm fate specification.Lastly, I identify an acetylated tubulin antigen as a marker for cilia. I describe the pattern of cilia differentiation in the developing larvae that results in the formation of two ciliary bands, the prototroch and the metatroch, required for locomotion and feeding. These ciliary bands are conserved among annelid and mollusc larvae. Interestingly, the metatroch is derived from third quartet derivatives in the annelid Polygordius and from second quartet derivatives in the mollusc Crepidula. I provide evidence that the metatroch is derived from the first quartet derivatives in the mollusc Ilyanassa. Thus while the larval metatroch is conserved, its clonal origin is not. Based on these results, I provide support for the hypothesis that the metatroch is not homologous between annelids and molluscs or even among molluscs.

Abstract:
Nathan Neta ben Shlomo Shapira (1585-1633) is the most famous kabbalist stemming from the Jewish intellectual environment of Poland. His major treatise, Megaleh Amuqot, is among the most complex kabbalistic texts ever written. It combines variegated strata of older mystical traditions, to which the author applies diverse, often obscure modes of interpretation. For this reason, Nathan Shapira has remained one of the least studied figures in modern scholarship, despite the fact that he is generally acknowledged as the most important early-modern Ashkenazi kabbalist, whose influence on later Eastern-European mystical circles is well attested. Although there are some general accounts of Shapira’s religious activity in Kraków, and references have been made to his startling mathematical mind-set, scholarship still lacks a thorough examination of his literary legacy, and a detailed evaluation of his contribution to the development of Jewish mystical thought. My dissertation aims to integrate Nathan Shapira’s kabbalah within a broad panorama of Jewish mystical traditions of the early modern period. It challenges the notion of the dominance of Lurianic ideas in Shapira’s thought, arguing for a more pluralistic perspective of the historical development of the kabbalistic tradition. Recently, Yehuda Liebes and Moshe Idel have raised the possibility that Nathan Shapira’s kabbalah may have belonged to a tradition that sprang from a multifaceted cultural milieu of Ashkenazi mysticism, consisting of at least two distinct major strands. Following this notion, I propose to challenge the common view that the Ashkenazi mysticism was a homogenous entity, whose influences effectively ceased after 13th century. On the contrary, I claim that the medieval mystical Ashkenazi ideas underlie much of Nathan Shapira’s kabbalah. In considering medieval Ashkenazi mysticism as Shapira’s formative background, I focus on the ‘Enoch-Metatron’ cluster of traditions, which I claim was as central to Shapira’s thought as it was to his Ashkenazi predecessors.

Abstract:
This chapter focuses on Metatron, that enigmatic figure assuming the title “Lesser God.” It begins with an analysis of a midrash transmitted only in the Bavli, in which Rav Idith—a Babylonian amora of the fourth or fifth century—deflects the fierce attacks of certain heretics who insist on assigning Metatron divine status. No doubt, the notion of a second divine power alongside that of God has gained followers among the Babylonian Jews. In order to substantiate this claim, the chapter surveys all the relevant Metatron passages preserved in rabbinic literature. It turns out that almost all of them are found either in the Babylonian Talmud or in the Hekhalot literature, most notably in 3 Enoch. The Metatron of the Bavli and the Hekhalot literature is a deliberate response on the part of the Babylonian Jews to the challenges posed by Christianity.

Abstract:
Reinforcement learning (RL) has had many successes, but significant hyperparameter tuning is commonly required to achieve good performance. Furthermore, when nonlinear function approximation is used, non-stationarity in the state representation can lead to learning instability. A variety of techniques exist to combat this --- most notably experience replay or the use of parallel actors. These techniques stabilize learning by making the RL problem more similar to the supervised setting. However, they come at the cost of moving away from the RL problem as it is typically formulated, that is, a single agent learning online without maintaining a large database of training examples. To address these issues, we propose Metatrace, a meta-gradient descent based algorithm to tune the step-size online. Metatrace leverages the structure of eligibility traces, and works for both tuning a scalar step-size and a respective step-size for each parameter. We empirically evaluate Metatrace for actor-critic on the Arcade Learning Environment. Results show Metatrace can speed up learning, and improve performance in non-stationary settings.
