Abstract:
This paper investigates the design cycle life of commercially available 9/16 inch needle valves and 9/16 inch tees under tri-axial fatigue. The valves and tees investigated were manufactured from 316 stainless steel and are rated to 4100 bar (60,000 psi) maximum allowable working pressure. The testing was performed using a tri-axial fatigue machine originally designed by Dr. B. Crossland, Dr. J.L.M. Morrison and Dr. J.S.C. Perry in 1960 [1] and upgraded by Michael D. Mann of KMT Waterjet Systems, Inc. This investigation compares the fatigue life of the 9/16 inch needle valves and 9/16 inch tees from two different commercial sources via actual test results from the fatigue machine.

Abstract:
The purpose of this study is to develop a novel bone replacement using in situ polymerization of thiol-acrylate with adipose tissue derived adult stem cells (ASCs). Specifically, Poly(ethylene glycol) diacrylate-co-trimethylolpropane tris (3-mercaptopropionate) (PEGDA-co-TMPTMP) was synthesized with 10% Hydroxyapatite (HA) foam by an amine-catalyzed Michael addition reaction. Initial characterization studies were performed to determine the temperature profile during the exothermic reaction showing a peak temperature of 50°C. To prevent hyperthermic cell damage and death during the exothermic polymerization procedure, the hASCs were encapsulated in alginate. Characterization of the 3-D structure and interconnectivity of pores in the polymeric foam scaffolds were performed using FIB-SEM and Micro-CT showing uniform distribution of HA. Cell viability experiments within the polymeric scaffold were performed using Vybrant® MTT cell profileration method, as well as fluorescent dyes: Calcein-AM (live) and Ethidium homodimer-1 (dead) showing viability of cells inside the samples.

The Michaelis-Menten (Km ), catalytic (kcat ), and specificity (kcat/Km ) constants were determined for humSft α- and β-thrombins with the chromogenic substrate S-2238 (H-D-Phe-Pip-Arg-pNA), Chromozym-TH (Tos-Gly-Pro-Arg-pNA), and Spectrozyme-TH (H-D-HHT-Ala-Arg-pNA) in 0.15 M NaCl buffered with 10 mM HEPES ancTlO mM Tris-HCT at pH 7.4, 37°C. Spontaneous hydrolysis was insignificant for these substrates. Both S-2238 and Spectrozyme-TH exhibited limiting solubilities at ∼35 μM, while Chromozym-TH did not do so up to 50 μM. From initial estimates, Km's and k t's were refined by computer Gause-Newton iterations or nonlinearneast-square fits for the concentration of p-nitroanalide formed per second versus the initial substrate concentration. No major differences were found between a-thrombin (99% α, 91% esterolytically active enzyme, and > 3,500 kilo-U.S. clotting units/g with fibrinogen) and Yγ-thrombin (98% γ. 89% active enzyme, and < 10 kilo-units/g). For α- versus γ-thrombin and the three substrates, respectively, the Km 's were 6.75 ± 0.13 vs 7.62 ± 0.30, 18.4 ± 0.4 vs 23.0 ± 0.3, and 2.53 ± 0.02 vs 3.85 ± 0.51 μM; the kcat> s were 125 ± 1 vs 134 ± 2, 181 ± 2 vs 130 ± 1, and 35.5 ± 0.2 vs 52.4 ± 2.0 s−1 ; and the kcat/Km 's were 18.5 vs 17.6, 9.84 vs 5.65, and 10.1 vs 13.6 s−1 μm . These values closely approximate those of 7.2 ± 0.9 μM, 84 ± 4 s−1 , and 11.7 s−1 μM−1 determined by Higgins, Lewis, and Shafer (J. Biol. Chem. 258:9276-9282, 1983) for the Aα cleavage of human fibrinogen by human α-thrombin under physiologically relevant conditions. Thus, these chromogenic substrates have thrombin specificities similar to that of fibrinogen, although their amidolytic activities are independent of additional active-site regions required for fibrinogen clotting activity (α vs γ-thrombin). Fibrinogen interactions with such active-site regions might account for why the fibrinogen Aa site is an atypical thrombin susceptible bond and the high species variability of fibrinopeptides. Abstract:
Production of hybrid artificial organs for implantation is one of the main topics of tissue engineering. A large organ consisting of soft tissues requires a high cell density, c.a. 108 cells/mL, to satisfy the same physiological metabolic rate per organ-volume as an organ in vivo. Therefore, the supply of oxygen and nutrition to all the cells composing the soft tissue is always critical problem for the in vitro artificial organ production. Energy metabolic rates, such as oxygen and glucose metabolism rate, of single cell at various temperatures are the basic data for designing the oxygen and nutrition transport in an artificial organ. It is reported that several conditions including pH, temperature, oxygen or glucose concentration have effects on energy metabolism, although these interactions are not clearly quantitatively measured mainly because of the problems of measuring systems. In this study, convenient method to measure glucose consumption rate of hepatocyte (HepG2 cell line) at different temperature and glucose concentration is proposed. A device for the measurement was developed which consists of a small closed chamber with an inlet and an outlet of culture medium at the both ends of the chamber. On the one side of the walls in the chamber, confluent HepG2 on a coverslip was installed. Culture medium supplemented with various concentration of glucose was supplied to the open flow chamber in a constant flow rate. The whole chamber was in a thermostatic bath to keep the temperature in the chamber constant. Glucose consumption rate can be calculated by measuring the difference between glucose concentration of inlet culture medium and outlet culture medium, the flow rate and the number of cells in the chamber. Enzymatic analysis using D-Glucose-HK allows quantification of the sample glucose concentration. The advantages of the proposed method include; 1) small number of cells is required for the measurement, c. a. 105cells, 2) the flow pattern and the glucose supply are in steady state. Especially the latter advantage made it possible to evaluate the effects of different conditions on the glucose consumption rate. Since the most of the metabolic rate were measured under unsteady state, conditions, such as pH, oxygen concentration and glucose concentration, were changed sometime drastically during the measurement. The results provided the several parameters of Michaelis-Menten kinetics at various temperatures.

Abstract:
RESUMO A sociedade atual vive em um contexto tumultuoso, sendo comum as pessoas viverem de forma mais estressadas, se alimentando mal e convivendo cada vez menos com as pessoas, perdendo oportunidades de fazer e manter relacionamentos pessoais. Como este estilo de vida está saturado, gerando problemas de saúde aos indivíduos, existem diversos estudos que comprovam a tendência de que as pessoas buscarão um melhor aproveitamento do seu tempo, com um maior contato com a natureza, dando valor ao seu próprio bem-estar, prestando mais atenção no que consome e em como consome, assim como a valorização do prazer de estar em um meio social. Nesse contexto, o objetivo deste trabalho é contribuir por meio da visão sistêmica e do design, o incentivo e a prática da filosofia Slow Food, e dessa forma, promover a preocupação com uma saudável forma de alimentação e a ação colaborativa entre as pessoas, em que um indivíduo possa cooperar com o outro, resgatando o convívio e socialização. Assim, pretende-se atuar em pequenas unidades urbanas externas, como prédios, condomínios, praças e avenidas com intuito de criação de um artefato que motive a prática dessa filosofia, assim como impulsione também as práticas colaborativas nestes locais, buscando a integração social e desenvolvimento econômico e ambiental, atendendo os princípios da sustentabilidade e o desenvolvimento local. O método de pesquisa usado é descritivo, em que se observou e analisou as situações e relações dos indivíduos, tanto de forma individual quanto as relações em sociedade. Os instrumentos escolhidos para coleta de dados foram o questionário, entrevistas e observações abertas, feitas informalmente ao decorrer das entrevistas. Neste trabalho integrou-se dois métodos de projeto, a ferramenta HCD/IDEO (Human Centered Design - Design Centrado no Ser Humano), usado como base, com técnicas para o entendimento dos desejos e necessidades da comunidade estudada e o Guia de projeto NASDesign/UFSC (Núcleo de Abordagem Sistêmica do Design/Universidade Federal de Santa Catarina), dividido em três fases: Sentir, Agir e Realizar, como complementação. Ao final, conclui-se que o modelo físico do produto proposto atende uma lista de requisitos divididos em três grande grupo: requisitos de produto, referentes à forma, materiais e texturas do objeto; requisitos do usuário, referente a necessidade entendida a partir do tipo de ambiente em que o produto está, e de como ele está sendo usado e aos requisitos do contexto de uso, que consiste nas necessidades do usuário observado durante a pesquisa.Palavras Chaves: design de produto, slow food, sustentabilidade.
