Full textVisceral leishmaniasis, in its active clinical form, is characterized by long-lasting fever, hepatosplenomegaly and cachexia. In Brazil, the lethality is, on average, 7% and the main causes of death are hemorrhage, comorbidity with immunosuppressive diseases and bacterial infection. The mechanism of increased bacterial infection in LV is unclear and one of the hypotheses is that there may be bacterial translocation of the intestinal mucosa to the lumen of the blood vessels and cause a greater severity of the immune-inflammatory response and consequent clinical worsening. The objective of this work was evaluate the occurrence of microbial translocation in Leishmania (L.) infantum infected-hamsters and correlate with the histopathological changes found in the gut of infected animals. Hamsters (Mesocricetus auratus) were infected intraperitoneally with 2x107 amastigotes of L. (L.) infantum and euthanized after 48, 72 hours and 15, 45 and 90 days of infection. As a control group, hamsters were inoculated intraperitoneally with RPMI culture medium. Were collected: blood, feces, spleen, large and small intestines. To detection amastigotes in intestinal mucosa, real-time PCR (qPCR), immunohistochemistry and histopathological analysis were used, and histological alterations in intestinal tissue were also evaluated. Spleen was used to determine the parasitic load through the Stauber technique. To detection of microbial translocation or products related, was performed quantification of lipopolysaccharide (LPS) in plasma. In order to evaluate the possible change in intestinal bacterial flora, bacterial sequencing of sample faeces from control and infected hamsters was carried. We observed increased parasite load on spleen and intestine as the infection progressed, the difference being significant at 90 days of infection. At the same time, we observed increased circulating LPS in infected animals at different times, 48 hours, 72 hours, 45 days and 90 days, with decrease in the intermediate period of 15 days, howeversignificant difference was observed only at 90 days post-infection in relation to control group. Histopathological changes were observed in the large and small intestine, ranging from mild to severe inflammatory infiltrate, enteritis, histiocytosis, and amastigotes. The changes occurred from 48 hours of infection, differentiating the population of the inflammatory infiltrate between neutrophils, lymphocytes, and even eosinophils in the large intestine of animals with 90 days of infection. |Bacterial sequencing shows that there was a change in the microbiome of the animals, but there is no significant identity, ie, above 95% in most bacteria. We conclude that changes in mucosal histology, invasion of amastigotes in this tissue and increase in LPS, suggest that microbial translocation is an event occurring during L. (L.) infantum infection in this experimental model.

Full textTsetse flies are the vectors of African trypanosomes, the causative agents of human African trypanosomiasis (sleeping sickness)in sub-saharan Africa. New sleeping sickness control strategies plan to use tsetse gut symbionts to increase tsetse flies refractoriness to trypanosomes infection. Such approaches require good knowledge on the molecular and cellular basis of interactions between symbionts, tsetse fly and trypanosome. This thesis aimed to i) assess the evolution of Glossina palpalis gambiensis symbionts (Wigglesworthia glossinidia and Sodalis glossinidius) densities throughout the host fly development cycle and ii) to characterize genes of Sodalis, G. p. gambiensis and Trypanosoma brucei gambiense in interaction, which are differentially expressed during the infection. We showed that both symbionts are present in all tsetse fly development stages, allowing their use in the context of vector control. Subsequently, experimental infections were performed on colonies flies. G. p. gambiensis female flies were fed on T. b. gambiense hosting mice. Transcriptome of infected flies and flies that have cleared trypanosome they ingested were analysed. This allow us identifying genes of Sodalis, G. p. gambiensis and T. b. gambiense differentially expressed at the infection key stages. Our results highlight the complexity of interactions between Sodalis, G. p. gambiensis, T. b. gambiense and underline the involvement of bacteriophages hosted by S. glossinidius in tsetse fly refractoriness to trypanosome infection. Full textIn marine environment, artificial surfaces are promptly colonized by biofilms, which are communities of bacteria surrounded by matrix of extracellular polymeric substances (EPS). Formation of biofilm is a critical step of biofouling development, which corresponds to the accumulation of micro and macro-organisms on immersed surfaces and which can have important negative ramifications in particular in the marine sector. In this study, bacteria isolated from the Mediterranean Sea have been identified and characterized using different phenotypical tools. Their capacity to form a biofilm in vitro has been studied in different conditions, with a particular focus on their ability to produce abundant carbohydrate-rich EPS, the overall objective of the study being the isolation of original antifouling-active exopolysaccharides. Thirteen strains have been phylogenetically and phenotypically characterized. Seven genera and twelve species were identified among which two isolates were affiliated to a new species, named Persicivirga mediterranea. This genus has never been described in the Mediterranean Sea. Extraction of EPS of each strain, grown in biofilm conditions, allowed the determination of their general composition in carbohydrates, proteins, nucleic acids and lipids. One strain, Pseudoalteromonas ulvae TC14, was able to produce large quantities of exopolysaccharides, comprising in majority polymers of glucose whose chromatographic and spectroscopic analyzes revealed a diversity in size (Mw ~ 1-4000 kDa), charge (neutral or anionic) and associated function (acetate or lactate). These polysaccharides inhibited biofilm formed by other marine strains isolated from the Mediterranean Sea. They can also be synthesized by planktonic TC14, but in very different proportions

Full textThe filamentous fungus Aspergillus fumigatus and the bacteria Chlamydophila psittaci are two major pathogens in poultry. Although phylogenetically distant, these two microorganisms share the same aerial transmission. Mortality and morbidity associated to aspergillosis and chlamydiosis are responsible for economic losses, in particular in turkey farms. In France, investigations on human cases of chlamydiosis frequently reveal a link with duck farms. The bacteria C. Psittaci is carried in the gut of ducks without any clinical signs. In order to investigate the circulation of Aspergillus fumigatus and Chlamydophila psittaci in avian farms, we studied the genetic diversity of the two pathogens. Two new MLVA (Multiple Locus VNTR Analysis) typing methods were set up. These methods revealed to be very discriminant, rapid and easy to use. In the case of A. Fumigatus, the MLVA method made it possible to cluster the genotypes according to their geographic origin. It also permitted the analysis of contamination modes in a turkey hatchery. In order to analyze the pathogenicity of Aspergillus fumigatus and Chlamydophila psittaci in birds, two experimental infection models were developed. The first model was designed to reproduce aspergillosis in one-day old chickens inoculated with airborne A. Fumigatus conidia. During this experiment, the impact of immunosuppression and of chicken lineage was evaluated. For C. Full textThe Ararajuba or Golden conure (Guaruba guarouba) is a medium-sized parrot, endemic to the Amazon biome and threatened of extinction primarily due to habitat loss and poaching. Few scientific studies have examined the health conditions of these birds kept in captivity, which will be essential to future reintroduction efforts. This study aimed to evaluate the health status of Ararajubas maintained in zoos and private breeders in São Paulo state, Brazil, through haematological, microbiological and parasitological evaluations. Samples were obtained from 87 individuals held at six Parks Zoos, three Commercial Breeders and one Conservation Breeder. Average haematological results: packed cell volume 46.12%, total serum protein 3.55 g/dL, haemoglobin 12.68 g/dL, erythrocytes 3.52 x 106 cells/mm3, Leukocytes 13.41 x 103 cells/mm3, thrombocytes 26.03 x 103 cells/mm3, Mean Corpuscular Volume 133.3 fL, Mean Corpuscular Haemoglobin 36.58 pg, Mean Corpuscular Haemoglobin Concentration 27.65 g/dL, heterophils 56.62%, lymphocytes 42.22%, monocytes 0.9%, eosinophils 0.04%, basophils 0.20%. Microbiological cultures: Escherichia coli could be isolated from 50% of the cloacal swabs and Salmonella spp. was not isolated in none. Parasitology: all animals were negative for both intestinal parasites and haemoparasites. Plasma and erythrocytes obtained by centrifugation were stored for future research.
