The principle aim of this study was to isolate a chemical for the "safe anaesthesia" of abalone under commercial farming conditions. "Safe anaesthesia" implied that the anaesthetic had no immediate detrimental or long term sublethal effect on the abalone, that it was safe for the farmer, the consumer and the environment. Four chemicals, magnesium sulphate (MgS0₄), ethylenediamine tetra-acetic acid (EDTA), 2-phenoxyethanol and procaine hydrochloride were shown to effectively inhibit the in vitro contraction of isolated tarsal muscle of Haliotis midae. Thesis (MSc (Genetics))--Stellenbosch University, 2010.<br>ENGLISH ABSTRACT: Haliotis midae is the most important aquaculture species in South Africa, with abalone farming contributing 80% of the Rand value of the aquaculture industry. Although genetic research has benefited the abalone industry, several issues still hinder increases in abalone production. Progress towards an increase in H. midae growth rate by utilizing conventional genetic studies and selective breeding has been relatively slow. Gene transfer has therefore become a plausible option to address this problem. Genes that co

This study investigates aspects of the biology of C. impunctatus with a view to producing an optimised laboratory culture of this species. Artificial membrane feeding of C. impunctatus is shown to be enhanced by the addition of blood vestiges to the outer surface of the feeding apparatus. The effects of holding temperature, environment and conspecific density upon oogenesis in blood fed female midges are assessed through mortality, digestion of the blood meal and development of the eggbatch over time. Oviposition in C. impunctatus is investigated through choice and nochoice bioassays which sho

The abalone (Haliotis midae) culture industry in South Africa is becoming increasingly dependent on the use of formulated feeds, due to limitations in the supply of kelp. The bulk of the feeds that are currently available were developed based on the requirements of juvenile abalone cultured within the optimal temperature range for growth (18 - 20 °C). However, most abalone farming facilities are land-based pump ashore operations and are thus mostly exposed to temperatures outside of this range. In addition, these feeds have been found to be unsuitable for abalone cultured at elevated water tem

Two experimental diets, namely, a seaweed diet (SWD) Laminaria pallida (macrophytic algae) and a formulated diet (FD) (macro-algae), for use in abalone (Haliotis midae) feed development, were evaluated. The animals used in this study were juveniles (24.33 ± 3.14 mm shell length; 2.72 ± 0.83 g live weight, mean ± SE) and sub-adults (58.07 ± 10.33 mm shell length and 41.96 ± 20.61 g

In South Africa, Haliotis midae is the only commercially significant abalone species and it has become increasingly sold on the global market. The importance of abalone as a mariculture subject has triggered extensive research into maximizing production, with particular emphasis on optimizing growth rates. Animals generally do not have a requirement for protein, but instead have a requirement for specific essential amino acids (EAAs) and non-essential amino acids (NEAAs). The NEAAs are those that can be synthesised by the animal, however, EAAs cannot be synthesised and must therefore be supplied as part of the diet. When these amino acids (AAs) are supplied in the correct ratios and with the correct level of digestible energy, nutritionists can maximise somatic growth from proteins. This has resulted in increased research into the use of crystalline AAs as a tool in feed formulation research in

It is the only species of southern African Haliotidae to be commercially reared and has been successfully cultured for almost two decades. Its short history of domestication along with market demands and the need to develop efficiency in the production process has resulted in an increased interest in the possible genetic improvement of this species. Abstract:
The profitability of abalone farms is heavily influenced by their production per unit of grow-out space. With farms having physically expanded to the maximum, and with increasing production costs, one of the most realistic ways for farms to increase their production is through optimizing stocking densities. The effect of stocking density on Haliotis midae performance is undocumented and optimal stocking densities for this species have not been determined. Experiments were conducted under farm conditions to investigate the effects of four different stocking densities (16 %, 20 %, 22 % and 24 %
