<p>Locusta migratoria apolipophorin III is an 18 kDa exchangeable apolipoprotein present in the hemolymph of locusts. In addition to its known role in diacylglycerol transport, the protein has antimicrobial properties targeting bacterial membranes. <p> <i>Locusta migratoria</i> apolipophorin III (apoLp-III) is an exchangeable apolipoprotein that aids in the transport of diacylglycerol through the hemolymph of insects. The x-ray structure shows five &alpha;-helices which form a five helix bundle. The NMR solution structure revealed the presence of a four-residue mini-helix that was not identified in the x-ray structure. It was hypothesized that this mini-helix initiates lipid binding. Specific inhibitors of protein kinase A (PKA), Rp-cAMP, protein kinase C (PKC), chelerythrine and staurosporine, and protein phosphatase 1 and 2A okadaic acid, inhibited fluid secretion whilst corpora cardiaca (CC) extract and cAMP were found to stimulate fluid secretion. Tubules inhibited by PKC inhibitors, were significantly stimulated when CC extract was included in the bathing medium, but cAMP did not stimulate under these conditions. Intracellular microelectrodes have been used in conjunction with ion substitution, and agonists and inhibitors of known transport processes to investigate the mechanisms whereby ions cross the basal and apical cell membranes of the Malpighian tubules of Locusta. Values for basal, apical and transepithelial potentials in 'Normal' saline were -71.6 ± 0.3 mV, -82.6 ± 0.8 mV and +5.7 ± 1.0 mV (lumen positive) respectively.
