Abstract:
Tracking transplanted stem cells is necessary to clarify cellular properties and improve transplantation success. In this study, we investigate the effects of fluorescent superparamagnetic iron oxide particles (SPIO) (Molday ION Rhodamine-B™, MIRB) on biological properties of human dental pulp stem cells (hDPSCs) and monitor hDPSCs in vitro and in vivo using magnetic resonance imaging (MRI). Morphological analysis showed that intracellular MIRB particles were distributed in the cytoplasm surrounding the nuclei of hDPSCs. 12.5–100 μg/mL MIRB all resulted in 100% labeling efficiency. MTT showed that 12.5–50 μg/mL MIRB could promote cell proliferation and MIRB over 100 μg/mL exhibited toxic effect on hDPSCs. In vitro MRI showed that 1 × 106cells labeled with various concentrations of MIRB (12.5–100 μg/mL) could be visualized. In vivo MRI showed that transplanted cells could be clearly visualized up to 60 days after transplantation. These results suggest that 12.5–50 μg/mL MIRB is a safe range for labeling hDPSCs. MIRB labeled hDPSCs cell can be visualized by MRI in vitro and in vivo. These data demonstrate that MIRB is a promising candidate for hDPSCs tracking in hDPSCs based dental pulp regeneration therapy.

Abstract:
Remote sensing products provide a vital understanding of wildfire effects across a landscape, but detection and delineation of low- and mixed-severity fire remain difficult. Although data provided by the Monitoring Trends in Burn Severity (MTBS) project are frequently used to assess severity in the United States, alternative indices can offer improvement in the measurement of low-severity fire effects and would be beneficial for future product development and adoption. This research note evaluated one such alternative, the Mid-Infrared Bi-Spectral Index (MIRBI), which was developed in savannah ecosystems to isolate spectral changes caused by burning and reduce noise from other factors. MIRBI, differenced MIRBI (dMIRBI) and burn severity indices used by MTBS were assessed for spectral optimality at distinguishing severity and the ability to differentiate between unburned and burned canopy in a conifer forest. The MIRBI indices were better at isolating changes caused by burning and demonstrated higher spectral separability, particularly at low severity. These findings suggest that MIRBI indices can provide an enhanced alternative or complement to current MTBS products in high-canopy-cover forests for applications such as discernment of fire perimeters and unburned islands, as well as identification of low-severity fire effects.

Abstract:
Introduction: Evaluating the attenuation of influenza viruses in animal studies is important in developing safe and effective vaccines. This study aimed to demonstrate that the microRNA (miRNA)-let-7b-mediated attenuated influenza viruses (miRT-H1N1) are sufficiently attenuated and safe in mice. Methodology: The pathogenicity of the miRT-H1N1virus was investigated in a mouse model, evaluated with median lethal dose (LD50). The replicative dynamics of the miRT-H1N1, wild type (wt)-H1N1, and scramble (scbl)-H1N1 viruses in the lungs of infected mice were compared. The degrees of lesions and the expression levels of IL-6, TNF-α, and IFN-β in the lungs of mice infected with different viruses were also analyzed. Results: In miRT-H1N1 virus-infected mice, 100% of mice survived, and a lower pathogenicity was characterized with non-significant weight loss when compared to mice infected with the control wt virus. The miRT-H1N1 virus was not fatal for mice, even at the highest dose administered. The viral load in the lungs of miRT-H1N1-infected mice was significantly lower than that of the wild-type virus-infected mice. Fewer pulmonary lesions and lower levels of selected pro-inflammatory cytokines in the lungs of the mice infected with the miRT-H1N1 virus were also observed. The virulence of the miRT-H1N1 virus reduced significantly, suggesting that the miRT-H1N1 virus was safe for mice. Conclusions: Our study demonstrated that the miRNA-mediated gene silencing is an alternative approach to attenuating the pathogenicity of wt influenza viruses that have potential in the development of influenza vaccines.

Penelitian ini kualitatif deskriptif dan subjek penelitian ini adalah masyarakat Desa Lembupurwo yang melaksanakan kerja sama gaduh sapi. Teknik pengumpulan data yang digunakan adalah wawancara dan dokumentasi. Analisis data yang penulis lakukan adalah memilah data yang dihasilkan dari wawancara dan dokumentasi sebagai sumber utama sementara sumber pendukung menggunakan jurnal artikel, buku,dan laporan penelitian. Hasil penelitian menunjukan bahwa praktik gaduh sapi di Desa Lembupurwo, Kecamatan Mirit, Kabupaten Kebumen mengikuti kebiasaan masyarakat baik dari segi cara, modal dan pembagian keuntungannya. Model kerja sama gaduh sapi yang dilakukan menggunakan dua system yaitu penggemukan dan pembibitan. Dalam pandangan hukum Islam praktik kerja sama gaduh sapi di Desa Lembupurwo, Kecamatan Mirit, Kabupaten Kebumen telah sesuai dengan hukum Islam, yakni menggunakan akad muḍārabah muţlaqah.

ABSTRACTSiderophores have been identified as virulence factors in the opportunistic fungal pathogenAspergillus fumigatus. The 14-pass transmembrane protein MirB is postulated to function as a siderophore transporter, responsible for uptake of the hydroxamate siderophoreN,N′,N″-triacetylfusarinine C (TAFC). Our aim was to identify amino acids ofA. fumigatusMirB that are crucial for uptake of TAFC. Site-directed mutagenesis was used to create MirB mutants. Expression of wild-type and mutant proteins in theSaccharomyces cerevisiaestrain PHY14, which lacks endogenous siderophore transporters, was confirmed by Western blotting. TAFC transport assays using55Fe-labeled TAFC and growth assays with Fe-TAFC as the sole iron source identified alanine 125, tyrosine 577, loop 3, and the second half of loop 7 (Loop7Del2) as crucial for function, since their substitution or deletion abrogated uptake completely. Wild-type MirB transported ferricrocin and coprogen as well as TAFC but not ferrichrysin. MirB was localized by fluorescence microscopy using antisera raised against a MirB extracellular loop peptide. Immunofluorescence microscopy showed that in yeast, wild-type MirB had a punctate distribution under the plasma membrane, as did the A125D and Y577A strains, indicating that the defect in transport of these mutants was unlikely to be due to mislocalization or degradation. MirB immunolocalization inA. fumigatusshowed that the transporter was found in vesicles which cycled between the cytoplasm and the plasma membrane and was concentrated at the hyphal tips. The location of MirB was not influenced by the presence of the siderophore TAFC but was sensitive to internal iron stores.

As a method to derive a “purified” measure along a dimension of interest from response data that are potentially multidimensional in nature, the projective item response theory (PIRT) approach requires first fitting a multidimensional item response theory (MIRT) model to the data before projecting onto a dimension of interest. This study aims to explore how accurate the PIRT results are when the estimated MIRT model is misspecified. Specifically, we focus on using a (potentially misspecified) two-dimensional (2D)-MIRT for projection because of its advantages, including interpretability, identifiability, and computational stability, over higher dimensional models. Two large simulation studies (I and II) were conducted. Both studies examined whether the fitting of a 2D-MIRT is sufficient to recover the PIRT parameters when multiple nuisance dimensions exist in the test items, which were generated, respectively, under compensatory MIRT and bifactor models. Various factors were manipulated, including sample size, test length, latent factor correlation, and number of nuisance dimensions. The results from simulation studies I and II showed that the PIRT was overall robust to a misspecified 2D-MIRT. Smaller third and fourth simulation studies were done to evaluate recovery of the PIRT model parameters when the correctly specified higher dimensional MIRT or bifactor model was fitted with the response data. In addition, a real data set was used to illustrate the robustness of PIRT.

AbstractTo understand the mechanisms standing behind a successful stem cell-based therapy, the monitoring of transplanted cell’s migration, homing as well as the engraftment efficiency and functional capability in-vivo has become a critical issue. The present study was designed to track the labelled oADMSCs in-vitro and its visualization through MRI technique. oADMSCs from passage 4 (P-4) to passage 6 (P-6) were labelled with superparamagnetic iron oxide (SPIO) conjugated with rhodamine (Molday Ion Rhodamine-B - MIRB) at the concentration of 25μg Fe/ml in DMEM. Internalized MIRB was observed under fluorescent microscope after 72 hrs of incubation. Labelled oADMSCs showed Prussian Blue positive reaction demonstrating the iron uptake of the cells. The viability of the MIRB-labelled oADMSCs ranged between 98-99 per cent and Trypan blue exclusion test showed no significant difference in viability between labelled and unlabelled oADMSCs. MR signal in control group of cells was similar to that of water. MR signals or fluorescence in MIRB-labelled cells decreased with increasing concentrations of iron. The T2 weighted images of MIRB-labelled oADMSCs increased with increasing concentrations of SPIOs. The MIRB was found to be nontoxic, and did not affect proliferation capacity in-vitro.

Abstract:
Equating is a statistical process used to accomplish score comparability so that the scores from the different test forms can be used interchangeably. One of the most widely used equating procedures is unidimensional item response theory (UIRT) equating, which requires a set of assumptions about the data structure. In particular, the essence of UIRT rests on the unidimensionality assumption, which requires that a test measures only a single ability. However, this assumption is not likely to be fulfilled for many real data such as mixed-format tests or tests composed of several content subdomains: failure to satisfy the assumption threatens the accuracy of the estimated equating relationships. The main purpose of this dissertation was to contribute to the literature on multidimensional item response theory (MIRT) equating by developing a theoretical and conceptual framework for true-score equating using a simple-structure MIRT model (SS-MIRT). SS-MIRT has several advantages over other complex MIRT models such as improved efficiency in estimation and a straightforward interpretability. In this dissertation, the performance of the SS-MIRT true-score equating procedure (SMT) was examined and evaluated through four studies using different data types: (1) real data, (2) simulated data, (3) pseudo forms data, and (4) intact single form data with identity equating. Besides SMT, four competitors were included in the analyses in order to assess the relative benefits of SMT over the other procedures: (a) equipercentile equating with presmoothing, (b) UIRT true-score equating, (c) UIRT observed-score equating, and (d) SS-MIRT observed-score equating. In general, the proposed SMT procedure behaved similarly to the existing procedures. Also, SMT showed more accurate equating results compared to the traditional UIRT equating. Better performance of SMT over UIRT true-score equating was consistently observed across the three studies that employed different criterion relationships with different datasets, which strongly supports the benefit of a multidimensional approach to equating with multidimensional data.

A literatura em Psicologia indica a carência de uma medida nacional que leve em consideração a personalidade do indivíduo no ambiente de trabalho. Considerando a qualidade psicométrica do teste dentro do contexto em que foi validado, o propósito deste estudo foi validar o Orpheus Business Personality Inventory (OBPI) para a população brasileira. Este teste foi desenvolvido especialmente para o ambiente organizacional e pode ser usado sob diversos propósitos, como seleção, promoção e desenvolvimento de colaboradores. O teste é composto por 190 itens, distribuídos em 16 escalas: cinco escalas maiores, sete escalas menores e quatro escalas de checagem. As escalas maiores, baseadas no modelo Big Five de personalidade, são Companheirismo, Autoridade, Conformidade, Emoção e Detalhe. As escalas menores, por sua vez, avaliam a integridade e foram baseadas nos traços do teste Giotto, são elas: Proficiência, Orientação para o trabalho, Paciência, Imparcialidade, Lealdade, Transparência e Iniciativa. Por fim, as escalas desenvolvidas para checagem de respostas contaminadas são: Dissimulação, Ambivalência, Desânimo e Falta de atenção. A coleta de dados foi realizada através da plataforma de testagem computadorizada Concerto, desenvolvida pela Universidade de Cambridge. Participaram deste estudo 938 indivíduos para a fase de testagem e 98 para a etapa de reteste. A validação de conteúdo contou com a participação de cinco juízes. Para a validade de construto, utilizou-se a Full Information Factor Analysis sob os pressupostos da Teoria de Resposta ao Item Multidimensional. O alfa de Cronbach e o coeficiente Omega foram calculados para medir a consistência interna de todas as escalas. Também foram avaliadas medidas de misfit de acordo com o modelo Rasch. Os resultados indicaram um conjunto de itens que apresentaram cargas fatoriais baixas, mas existem evidências de validade transcultural que podem ser 13 verificadas através dos resultados da validade de conteúdo e da fidedignidade do teste. A versão brasileira do OBPI não só suprirá uma lacuna na área de Avaliação Psicológica, como também auxiliará no desenvolvimento de um instrumento que seja multicultural em sua essência. Espera-se que em mão dos resultados aqui apresentados, estudos futuros possam encontrar evidências adicionais de validade de construto.
The literature in Psychology indicates the need for a national instrument that takes into consideration individual personality inside the workplace. Given the psychometric quality of this test within its validation context, the purpose of this study was to validate the Orpheus Business Personality Inventory to the Brazilian population. This test has been developed specially to the organizational environment and can be used under several purposes, such as selection, promotion and development of employees. The test is composed of 190 items divided through 16 scales: five major scales, seven minor scales and four audit scales. The major scales, based on the Big Five personality model, are Fellowship, Authority, Conformity, Emotion and Detail. The minor scale are a measure of integrity and were based on the traits stated on the Giotto test, they are: Proficiency, Work orientation, Patience, Fair-Mindedness, Loyalty, Disclosure and Initiative. Finally, the scales created to audit invalid responses are Dissimulation, Ambivalence, Despondency and Inattention. The data was collected through Concerto platform, developed by the University of Cambridge. In this study, participated 938 individuals for the testing phase and 98 for the retest phase. For the content validity study, five experts participated. For the construct valitidy study, a Full Information Factor Analysis was performed under the Multidimensional Item Response Theory underlying assumptions. Cronbach’s alpha and the Omega coefficient were calculated to evaluate the internal consistency for all scales. Misfit measurement were also investigated, according to the Rasch Model. The results indicated that a group of items presented low factor loadings, although cross-cultural validity can be inferred from the results for the content validity and reliability of the test. The Brazilian version of OBPI will not only fill a gap in the area of Psychological Evaluation, but will also assist the 15 development of an instrument that is multicultural in its essence. It is expected that further studies will support the results presented here with additional construct validity evidences.

The 1994 Beaver Mountain fire ignited the canopies of subalpine fir, Abies lasiocarpa, and spread ground fire into adjacent Douglas-fir forests, Pseudotsuga menziesii var. glauca. Despite shorter flight seasons due to lower annual temperatures and persistent snow, the Douglas-fir bark beetle, Dendroctonus pseudotsugae Hopkins, attacked a range of moderately fire-injured host conifers. Logistic regression models illustrated that in 1995 associated bark beetles selected large diameter Douglas-fir with 60-80% bole char, 60-80% crown volume scorch, and 50-70% probability of mortality due to fire. In 1996 beetle preference shifted to smaller diameter trees with lighter fire injury. Tree size was less significant for predicted attack in 1996 because most large fire-damaged conifers were colonized by beetles in 1995. Beetle populations did not reach outbreak proportions outside the fire boundary, but 53 green trees were also infested in 1997 along the burn perimeter. Log linear tests conducted to quantify beetle emergence supported conclusions that beetles were not only attracted to mature, moderately fire-weakened conifers, but also produced greater brood numbers with up to 60-80 emergence holes/ 1800 cm2. Fire-defoliated trees provided bark beetles with sufficient phloem and limited resistance, allowing beetles to aggregate on areas of viable stem tissue regardless of overall bole char extent.

Abstract:
For equating tests that measure several distinct proficiencies, procedures that reflect the multidimensional structure of the data are needed. Although there exist a few equating procedures developed under the multidimensional item response theory (MIRT) framework, there is a need for further research in this area. Therefore, the primary objectives of this dissertation are to consolidate and expand MIRT observed score equating research with a specific focus on the common-item nonequivalent groups (CINEG) design, which requires scale linking. Content areas and item types are two focal points of dimensionality. This dissertation uses two studies with different data types and comparison criteria to address the research objectives. In general, a comparison between unidimensional item response theory (UIRT) and MIRT methods suggested a better performance of the MIRT methods over UIRT. The simple structure (SS) and full MIRT methods showed more accurate equating results compared to UIRT. In terms of calibration methods, concurrent calibration outperformed separate calibration for all equating methods under most of the studied conditions.
