Abstract:
Abstract MKC-1 is a novel, orally active cell cycle inhibitor with in vitro and in vivo activity against a wide range of human solid tumor cell lines, including multi-drug resistant cell lines. MKC-1 has been tested in over 270 patients to date and is currently in Phase II clinical trials. The strong pre-clinical activity of MKC-1 towards solid tumor lines and signs of efficacy in the initial clinical evaluation with lack of neuropathy and cardiotoxicity suggests that MKC-1 may also be of clinical benefit in the treatment of hematopoietic cancers. The antiproliferative activity of MKC-1 was examined against a panel of hematopoietic cell lines including HL-60, U937, MV4;11, THP-1, Jurkat, and OCI-AML 1–5. MKC-1 showed potent and dose-dependent activity towards these cell lines, with IC50 values in the range of 20 – 400 nM. MKC-1 also inhibited in vitro growth of primary cells derived from AML and CML patients. Additionally, MKC-1 showed enhanced activity with Ara-C in combination studies in vitro when added either simultaneously or sequentially using the cell line OCI-AML 4. Binding studies have shown that MKC-1 binds to the colchicine binding site of tubulin and to members of the importin beta family of proteins. Consistent with these results, cell cycle arrest in the G2/M phase of the cell cycle followed by apoptosis was observed in cell lines and patient samples treated with MKC-1. Immunofluorescence analysis of cells treated with MKC-1 revealed that the drug induced a disruption of the microtubule network and the formation of aberrant mitotic spindles. Furthermore, MKC-1 was also shown to induce a dose-dependent reduction in the levels of both phospho-Akt and phospho-p70S6K kinases through Western blot analysis of treated THP-1 cells. In conclusion, our results indicate MKC-1 arrests the cell cycle and disrupts multiple survival pathways to induce apoptosis in hematopoietic cell lines and patient samples. These results suggest that MKC-1 may have clinical potential in the treatment of leukemia either alone or in combination with other agents. Phase I trials in hematological cancers are currently being explored.

In this research we aim to elaborate a Didactic Sequence based on Rickenmann and always resorting to previous knowledge that favors the understanding of the notions of Minimum Common Multiple (CMM) and Maximum Common Divisor (MDC) inspired by the Sieve of Eratosthenes. In order to reach the objective, we will assume as theoretical support the Theory of Didactic Situations making use of Didactic Engineering aspects as research methodology. For the elaboration of the Sequence we carry out a historical study with epistemological bias covering the mathematical definitions in the light of Number Theory, as well as the extension of the same ones, the knowledge of the origins and how the objects have been researched and presented in didactic books.

Abstract:
Viruses have evolved a multitude of mechanisms, which allow immune evasion in both initial and persistent infection. Understanding the intricacies of these pathways is essential to our future ability to combat primary and reactive viral infections. The murine gamma-2 herpesvirus 68 (γHV68) encodes a protein mK3, which targets Major Histocompatibility Complex (MHC) class I heavy chains for ubiquitin-dependent proteasome degradation. MK3 is able to target and ubiquitinate MHC class I by binding to Endoplasmic Reticulum (ER) resident proteins tapasin, Transporter associated with antigen processing (TAP) 1 and TAP2 that are subunits in the complex known as the peptide-loading complex (PLC). The aforementioned characteristics of mK3 make this novel protein an excellent vehicle to study MHC class I assembly, immune evasion, and ER associated degradation (ERAD). Deepening our understanding of class I assembly and viral immune evasion will impact both the fields of immunology and virology. The homology between γHV68 and many of the human γ-herpesviruses makes this an indispensable model to clarify mechanisms that can then be applied to a broader spectrum of viruses. ERAD, an emerging field of study, is known to play a key role in numerous cellular housekeeping pathways as well as a number of disease states. Illuminating the mechanisms implicated in the mK3-mediated ubiquitination of MHC class I, specifically requirements for substrate recognition and degradation, will yield an increased understanding of cellular pathways involved in ERAD. The studies in this dissertation aim to expand our understanding of the relationship between mK3 and adapter proteins TAP/tapasin as well as mK3 and mK3-targeted substrates. The results show that TAP/tapasin act as adapter proteins by recruiting substrates for mK3. Further, mK3 ubiquitinates TAP/tapasin-associated substrates as long as the substrates have a tail greater than 6aa in length and the tail possesses an ubiquitin acceptor residue (lysine, serine or threonine). These studies also confirm that location of a protein within the PLC will determine the substrate’s susceptibility to mK3-mediated degradation. In the field of ubiquitin ligases and ERAD, these studies lend support to the concept of adapter mediated substrate recruitment.

Abstract:

Full text
Abstract:
The normal flora of the human skin is consisting of low pathogen bacteria, where the staphylococci are the most common bacteria. Studies show that antimicrobial substances can alter populations of skin bacteria and that these alterations can lead to critical consequences for the resistance of the skin. Propylene glycol is a substance that is classified as antimicrobial and bactericidal and the substance has a wide area of use and is frequently used as a supportive substance in various pharmaceuticals. Propylene glycol can be found in cosmetics and skincare products where it functions as moisture-binding and preservative. Normal concentrations of propylene glycol as moisture-binding substance in topicals is approximately 15 % and in cosmetics the concentration of propylene glycol is <0,1 % - >50 %. As of today, there are few scientific studies regarding the effects of propylene glycol to the bacterial flora of the human skin. The purpose of this study was to determine MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration) for normal staphylococci (S. epidermidis, S. aureus, S. hominis och S. capitis) included in the normal flora of the skin using various concentrations of propylene glycol. To be able to determine MIC and MBC the broth dilution method was used, where propylene glycol was diluted in various concentrations with broth in a microtiter plate. Hereafter, selected test bacteria were added and OD600 was measured during 24 hours. The results implicated that MIC and MBC for propylene glycol were 12,5 % and 25 % for common staphylococci located on the skin. At concentrations of propylene glycol of 12,5 %, visible bacterial growth of S. epidermidis, S. aureus, S. hominis and S. capitis was inhibited and at 25 % a bactericidal effect occurred on the bacteria. It shall be noted that further research is needed to find out how the skin's bacteria are affected by propylene glycol and its consequences.

Abstract:
It has often been assumed that hemopoietic progenitors undergo a rather uniform, deterministic number of proliferative cycles before they generate the elements which are recognizable by pano-ptiG and cytochemical methods. We now confirm and extend previous data on the distribution of the number of doublings undergone before polyploi'dization by day 7-MKC progenitors. Cultures of mouse bone marrow were stimulated by erythropoietin, WEHI conditioned medium (CM) and/or pokeweed CM. In all cases, cumulative doubling distributions of pure MKC colonies could be precisely fitted by exponential lines, whose slope and length depended on the stimulus or combination of stimuli used. The frequency of single MKC could be fitted by the same line as that plotting the number of doublings in MKC colonies, suggesting that single MKC and MKC colonies can be generated by the same class of progenitors. Exponential shape implies that the coefficient of variation (SD/mean) of doubling numbers is one, indicating significant variability in proliferative behavior among day 7-MKC progenitors. Such exponential distributions are best explained in the framework of Renewal Theory (Cox, Methuen, 1962). Whatever the distribution of the number of progenitor renewals in fixed time, the distribution of renewals will be geometric (i.e discontinuous exponential) if the total time spent by individual colonies in the proliferation phase (1) is distributed exponentially, and (2) is independent from the renewal process itself. These results suggest that the wave of MKC progenitor mitoses is randomly arrested by a differentia-tive event which strikes progenitor clones independently from their proliferative past.

Abstract:
Multiple kernel clustering (MKC) algorithms have been successfully applied into various applications. However, these successes are largely dependent on the quality of pre-defined base kernels, which cannot be guaranteed in practical applications. This may adversely affect the clustering performance. To address this issue, we propose a simple while effective framework to adaptively improve the quality of these base kernels. Under our framework, we instantiate three MKC algorithms based on the widely used multiple kernel $k$-means clustering (MKKM), MKKM with matrix-induced regularization (MKKM-MR) and co-regularized multi-view spectral clustering (CRSC). After that, we design the corresponding algorithms with proved convergence to solve the resultant optimization problems. To the best of our knowledge, our framework fills the gap between kernel adaption and clustering procedure for the first time in the literature and is readily extendable. Extensive experimental research has been conducted on 7 MKC benchmarks. As is shown, our algorithms consistently and significantly improve the performance of the base MKC algorithms, indicating the effectiveness of the proposed framework. Meanwhile, our framework shows better performance than compared ones with imperfect kernels.

Abstract:
Multiple kernel clustering (MKC) algorithms have been extensively studied and applied to various applications. Although they demonstrate great success in both the theoretical aspects and applications, existing MKC algorithms cannot be applied to large-scale clustering tasks due to: i) the heavy computational cost to calculate the base kernels; and ii) insufficient memory to load the kernel matrices. In this paper, we propose an approximate algorithm to overcome these issues, and to make it be applicable to large-scale applications. Specifically, our algorithm trains a deep neural network to regress the indicating matrix generated by MKC algorithms on a small subset, and then obtains the approximate indicating matrix of the whole data set using the trained network, and finally performs the $k$-means on the output of our network. By mapping features into indicating matrix directly, our algorithm avoids computing the full kernel matrices, which dramatically decreases the memory requirement. Extensive experiments show that our algorithm consumes less time than most comparatively similar algorithms, while it achieves comparable performance with MKC algorithms.

Abstract:
When a cylindrical datum feature is specified at maximum material condition (MMC) or least material condition (LMC) a unique circumstance arises: a virtual condition (VC) cylindrical boundary must be defined [1]. The geometric relationship between a cylindrical point cloud obtained from inspection equipment and a VC cylinder has not been specifically addressed in previous research. In this research, novel approaches to this geometric analysis are presented, analyzed, and validated. Two of the proposed methods are new interpretations of established methods applied to this unique geometric circumstance: least squares and the maximum inscribing cylinder (MIC) or minimum circumscribing cylinder (MCC). The third method, the Hull Normal method, is a novel approach specifically developed to address the VC cylinder problem. Each of the proposed methods utilizes a different amount of sampled data, leading to various levels of sensitivity to sample size and error. The three methods were applied to different cylindrical forms, utilizing various sampling techniques and sample sizes. Trends across sample size were analyzed to assess the variation in axial orientation when compared to the true geometric form, and a relevant case study explores the applicability of these methods in real world applications.

Abstract:
The scientific and economic significance of underwater technology is considerable, particularly as the oceans are becoming more and more important not only as a source of energy, raw materials and food but also due to their influence on climatic changes. Underwater technology, especially at great depths, represents a very particular challenge which is being tackled by ATLAS ELEKTRONIK Autonomous Underwater Vehicles (AUVs) DeepC©, SeaOtter Mk1 and SeaOtter Mk2. Many actions that would normally require high cost ROV systems or which are not possible (e.g. under ice investigations) can now accomplished using AUVs, at less cost and manpower. DeepC© is a high efficient AUV system for ultra deep missions (4000m), with long endurance (40–60h) and for a lot of applications. The development objectives are: Low vehicle weight, modular design, high efficient energy generation, autonomous mission execution, obstacle avoidance, in mission re-planning capability, precise navigation, advanced diagnosis and fault recovery system, high manoeuvrability, hover capability and containerized payload. The system is final integrated and ready for water tests and demonstrations till end of 2004. The paper deals with the system performance and shows the development status. The AUV SeaOtter Mk1, based on the ATLAS Maridan M600 vehicle and adapted to modern requirements in navigation software and mission management, is the ideal payload sensor carrier for military and commercial purposes. It ensures precise positioning, stability and manoeuvrability for accurate data collection. The on-board system provides all necessary position, attitude, status and time information to ensure effective and efficient performance throughout the mission. High positional accuracy and accurate stabilization of the vehicle’s attitude throughout the collection of data allows the mapping of raw data to be implemented directly, without post-processing, to obtain the necessary correlation of the survey mosaic. The advanced version of the SeaOtter AUV, the Mk2 is designed for various purposes, strictly adhering to a modular approach with regard to superstructure, propulsion, energy, communication, navigation and payload. The basic vehicle can easily be extended by the means of one or two extension modules to carry heavier payloads. The paper shows the SeaOtter Mk1 performance and gives an overview over the Mk2 Philosophy and the development status.

Abstract:
Platelet count elevations are observed in various clinical situations. In essential thrombocythaemia (ET), this is believed to reflect a primary disorder of megakaryocytes (MK). In contrast, thrombocytosis in malignant lymphoma (ML) is thought to arise via a secondary effect on these cells. In this study, MK parameters of whole cell, cytoplasmic and nuclear volumes were evaluated in these disorders. A trephine biopsy was obtained from the iliac crest of 10 subjects with ET, and from 10 subjects with ML. All exhibited a thrombocy^osip (considered to be a platelet count in excess of 400×109 L−1 ). In addition, a biopsy was obtained from 5 volunteer, haematologically normal subjects. Platelet counts in all subjects were measured using Coulter S+. Biopsies were processed as described elsewhere( 1). MK and MK nuclear planimetric areas were measured and converted to associated volumes(l). Mean MK, MK cytoplasmic and nuclear volumes were significantly larger in ET(p<0.05)and ML(p<0.05) when compared to those in controls. In addition, MKs in ET showed morphological abnormalities which were not exhibited in ML. These results suggest that platelet count elevations in ML and ET arise from large MKs with increased amounts of cytoplasm. The increased nuclear size observed further suggests that this may be mediated by an increased DNA content.

Abstract:
Emulsification is an important process in various fields including foods, pharmaceuticals, cosmetics, and chemicals. Emulsification operation is commonly conducted using conventional emulsification devices, such as high-speed blenders, colloid mills, high-pressure homogenizers, and ultrasonic homogenizers. However, these emulsification devices result in the production of polydisperse emulsions with wide droplet size distributions and poor controllability in droplet size and its distribution. In contrast, monodisperse emulsions consisting of monosize droplets have received a great deal of attentions over the past decade due to their high-tech applications, e.g., monosize microparticles as spacers for electronic devices and monosize micro-carriers for drug delivery systems (DDS). Our group proposed microchannel (MC) emulsification as a promising technique to produce monodisperse emulsions in the mid 1990s. Micro/Nanochannel (MNC) emulsification enables generating monosize droplets with the smallest coefficient of variation (CV) of below 5% using MC and nanochannel (NC) arrays of unique geometry. The resultant droplet size, which ranged from 0.5 to 200 μm, can be precisely controlled by channel geometry. Droplet generation for MNC emulsification is very mild and does not require any external shear stress; a dispersed phase that passed through channels is transformed spontaneously into monosize droplets inside a continuous-phase domain. The aim of this paper is to present recent developments in MNC emulsification chips, particularly focusing on asymmetric straight-through MC arrays for large-scale production of monodisperse emulsions. Asymmetric straight-through MC array chips were fabricated using a silicon-on-insulator wafer. Numerous asymmetric straight-through MCs each consisting of a microslot and a narrow MC were positioned in the central region of the chip. Monosize droplets were stably generated via asymmetric straight-through MCs at high production rates. Below a critical droplet production rate, monosize droplets were generated via asymmetric straight-through MCs, with droplet size and size distribution independent of the droplet productivity. The use of a large asymmetric straight-through MC array chip achieved the mass production of monosize tetradecane oil droplets at ∼1 L/h. The simulation results using CFD (computational fluid dynamics) agreed well with the experimental results and provided useful information, such as the movement of the oil-water interface during droplet generation. Monosize submicron droplets were also obtained using NC emulsification chips made of single-crystal silicon.

Abstract:
With increasing demand of energy, attention to the alternative sources of sustainable energy is getting priority over the last decades. Offshore wind turbine is one of them. The most widely used foundation system for the wind turbine is the monopile, which is a large diameter single pile. In the present study, three-dimensional finite element (FE) analyses are performed to evaluate the capacity of large diameter monopiles in dense sand using the Arbitrary Lagrangian-Eulerian (ALE) approach available in Abaqus/Explicit FE software. The behavior of sand is modeled using the Mohr-Coulomb (MC) and a modified Mohr-Coulomb (MMC) model where the pre-peak hardening, post-peak softening and the effects of mean effective stress and relative density on stress-strain behavior of dense sand are considered. Comparison with physical model test results shows that the MMC model can simulate better the load-displacement response than that with the MC model. The mechanisms involved in soil deformation are also explained using FE results.
