Full textThe purpose of this research was to examine the acclimation processes occurring in the maize chloroplasts during growth of plants in different light intensity and quality. Photosynthesis of C4 plants involves mesophyll (M) and bundle sheath (BS) chloroplasts, which differ structurally and functionally, and cooperate with each other during photosynthesis. The main aim of the work was to explain the “state transitions” process, which is considered, a short-term response of a photosynthetic apparatus to light stress. In the literature studies focus on the C3 plants, thus the data are not available how this process works in C4 plants. In particular, there is lack of information concerning changes in the organization of photosystems and their antenna in agranal chloroplasts (BS) of maize. Research presents the acclimation of maize chloroplasts to the changes in light conditions. Experiments were carried out on the chloroplasts isolated from the leaves of plants grown in at low, moderate and high light intensity (70, 350 and 800 µmol photons m−2 s −1 , respectively). Also, was used the red and far red light which preferentially excite the photosystems (PSII and PSI, respectively) to induce the short-term response of photosynthetic apparatus. In the work, low-temperature fluorescence techniques were used allowing to examine the changes in the organization of thylakoid chlorophyll binding complexes. The activity of photosystems was done by measurements oxygen-evolving activities using a Clarktype oxygen-electrode under saturating white light. To identify the compositions of super-, and megacomplexes was used two-dimensional electrophoresis (BlueNative/SDS PAGE) and immunodetection with appropriate antibodies, and fractionation of complexes on sucrose gradients. It has been shown that in the mesophyll chloroplasts of maize occurs the classic “state transitions” process, where FR-light induced dephosphorylation of LHCII and the detachment of antenna from PSI occurred. In the BS thylakoids, the FR light initiated dephosphorylation of free and aggregated LHCII antenna, the dephosphorylated antenna can bound to PSII (State 1), thereby significantly increase fluorescence at 77K, while part of LHCII pool is bound with PSI. Thus, my data indicate for the first time that PSI in agranal bundle sheath chloroplasts of maize is locked in State 2. It has been detected in M chloroplasts three types of PSI complexes: PSI-LHCI monomer, PSI-LHCI-LHCII-Lhcb4 and PSI-LHCII-PSII. They are characterized by protein composition and spatial structure is proposed. It is shown that in PSI-LHCI-LHCII-Lhcb4 megacomplex Lhcb3 protein was not identified. So far it is considered, that the Lhcb3 is a key protein in the “state transitions”. Exposition of plants from high and low light growth condition on FR light induces different rearrangements in the composition of super- and megacomplexes, what indicate that it is related not only to balancing light absorption by photosystems, but these complexes also seems to play a role for PSI in excitation quenching and in PSII turnover. Is proposed for the first time, a method of the enzymatic kinetics of photosystems using light as substrate. It has been shown that for PSII it is Michealis-Menten kinetics (M-M) with the parameter Km related to the "efficiency" of the connected antenna. In the case of PSI, the Michaelis-Menten model was expanded, creating a new model in which the process of antenna association/dissociation occurs. In this model, the parameter [ ]1⁄2 was determined, which is equivalent to the kinetic parameter Km in the M-M model and is a functional association of LHCII antennas with PSI. Was determined the relationships between the amount of antenna associated with photosystem and kinetic parameters. The enzymatic kinetics of photosystems was also examined considering the concentration of artificial acceptors of electrons for M chloroplasts. For PSI, a two-substrate kinetics model was introduced in which the substrates were: oxygen and methylviologen competing for the PSI active site. It allowed to determined constant = 124 , which is related to the affinity of oxygen for PSI and is a quantitative value of the Mehler reaction. On this basis, it was found that the Mehler reaction can have a significant contribution (up to 33%) in electron transport via PSI. The M-M model was extended to describe the PSII enzymatic kinetics assuming that the substrate artificial electron acceptor 2,6-dichloro-1,4-benzoquinone (DCBQ), acts as both an acceptor and inhibitor of PSII. The conducted research and proposed experimental systems are innovative and provide information extended the state of knowledge about the light reactions of photosynthesis of C4 plants. The intensity and quality of light as a signal inducing functional and structural changes in chloroplasts have allowed to answer the question how maize C4 plant, having functionally and structurally differentiated chloroplasts (M and BS), acclimatize to changing light condition. These studies also provide information on the relationship between the location of chloroplasts and their function modified by light. Submolecular mechanisms of acclimation is proposed.

Full textThe presented dissertation comprises of a critical edition of an XVIII-century romance "Przykładne z wiernej i statecznej miłości małżeństwo" by Elżbieta Drużbacka and a monographic section dedicated to analysis of this romance in terms of persuasive strategies and didactics. The critical edition is based on 4 manuscripts of this romance available in Poland and it was prepared in accordance with generally accepted principles of editing old Polish texts. Mentioned principles are discussed in detail at the end of the Introduction. Below the text, there are explanations to specific parts of the romance and there is a glossary of more difficult words. The edition is based on a copy signed by the author's hand. The introduction to the work also includes a section dedicated to the collation of the manuscripts. The edition is intended to enable readers with varying degrees of experience in dealing with early Polish literature (i.e., both non-academic enthusiasts and, for example, students) to become familiar not only with the content of the romance itself (by reconstructing the text and explaining more difficult words and phrases), but also with its wealth of references to the culture and literature. The main goal of the study preceding the work is a thorough analysis of the romance from the perspective of both its ideological content and poetics. The structure of the work is discussed in relation to the theoretical principles of the genre and the poetic solutions used by the author in her other works. Particular attention was also given to reconstructing the author's didactic intentions. Drużbacka created a piece of literature that was not limited to "adventurous adventures", but she treated the romance form as an opportunity to implement educational agenda. The characters' discussions, plot construction, and narrator's statements in the romance's depicted world touch on themes of personal freedom, the right to love, the boundaries between persuading and imposing one's will on them, desirable and undesirable values (from a social and individual perspective), and the "correct" way to experience love. As a result of the research, it became clear that Drużbacka shaped large parts of the text according to the rules of rhetoric. Not only did she use rhetorical devices in the content, but she also shaped the characters' utterances according to speech patterns and incorporated minor rhetorical genres into the romance structure. These observations suggest that the poetess was not as poorly educated as it is generally assumed. The analysis of detailed poetic solutions of the heterogeneous form of the piece is included in the monographic part of the work. As far as the sources of "Przykładne małżeństwo" are concerned, the analysis revealed inspiration from M. Bielski's "Kronika wszytkiego świata", B. Chmielowski's "Nowe Ateny", the philosophy of J.J. Rousseau, poems of the descriptio gentium type, anti-Turkish poems, rhetorical dialogues, hagiographic adventures and S. Twardowski's "Nadobna Paskwalina". This allows us to assume that Drużbacka's work is not an adaptation of a foreign work, but an authorial work, and that she has processed the sources according to her own invention. The work also includes a chapter summarizing the state of research on Drużbacka's romance works, in which references to her poetry are dating back to the XIX century are analyzed. In the same chapter the author proposes modifications to the existing typology of the poet's works. Drużbacka's romances have not been thoroughly researched so far, which makes her insufficiently present in contemporary research on old Polish romances. This work meets the suggestions of scholars specializing in history of romance genre in Poland, who signal the necessity of a separate study of Drużbacka's works as an exceptional author, and aims at creating, on the basis of an extensive comparative analysis, a contribution to further research on the author's oeuvre.
