Full textVoronoi grids are generated under constraints to reduce the errors due to cells geometry during flow simulation in reservoirs. The Voronoi points are optimized by minimizing objective functions relevant to various geometrical constraints. An original feature of this approach is to combine simultaneously the constraints: - Cell quality, by placing the Voronoi points at the cell barycenters. - Local refinement according to a density field rho, relevant to permeability, velocity or vorticity. - Cell anisotropy according to a matrix field M built with the three principal vectors of the anisotropy, which one is defined by the velocity vector or by the stratigraphic gradient. - Faces orientation according to a matrix field M built with the three vectors orthogonal to the faces, which one is defined by the velocity vector. - Conformity to structural features, faults and horizons. - Voronoï points alignment along well path. The quality of the generated grids is assessed from geometrical criteria and from comparisons of flow simulation results with reference fine grids. Results show geometrical improvements, that are not necessarily followed by flow simulation results improvements

Full textGlycogen storage disease type 1a (GSD1a) is a rare metabolic disorder due to an absence of glucose‐6 phosphatase (G6Pase) activity. G6Pase is the key enzyme of endogenous glucose production (EGP) and catalyzes the last step before the glucose release into the bloodstream. This function to produce glucose is restricted to the liver, the kidneys and the intestine. GSD1a is characterized by chronic hypoglycemia, hepatomegaly associated with hepatic steatosis and nephromegaly. The longterm complications of G6Pase deficiency include hepatocellular adenomas. The available animal model of GSD1a rarely survive over three months of age and the study of mechanisms of hepatocellular adenomas development cannot be investigated. So, we generated an original mouse model of GSD1a with a liver‐specific invalidation of catalytic subunit of G6Pase gene by an inducible CRE‐LOX strategy (L‐G6pc‐/‐ mice). In this work, we demonstrated that L‐G6pc‐/‐ were viable and totally reproduced the liver pathology of GSD1a, including the late development of hepatocellular adenomas. Then, we have begun liver gene therapy treatment using lentiviral and AAV vectors to correct the hepatic pathology. Finally, concerning glucose homeostasis, we have demonstrated that L‐G6pc‐/‐ were able to regulate blood glucose, during prolonged fast, even in the absence of hepatic glucose production. Rapidly, L‐G6pc‐/‐ mice were able to induce renal and intestinal gluconeogenesis thanks to a key role of glucagon and the development of a metabolic acidosis. These results provide evidence that the major role of the liver for EGP during fasting requires re‐examination

Full textBased on proton conduction of polymeric electrolyte membrane (PEM) technology, the water electrolysis (PEMWE) offers an interesting solution for efficiency hydrogen production. During the electrolysis process of water in PEMWE, the anodic side is the place where the water is splitting into oxygen, protons and electrons. The aim of this study is to recognize the link between two-phase flows (anode side) and cell performance under low pressure conditions. We have developed three approaches: the analytical approach and the numerical approach validated by the experimental data. For the numerical model, we have developed a two-dimensional stationary PEMWE model that takes into account electro-chemical reaction, mass transfer (bubbly flow), heat transfer and charges balance through the Membrane Electrodes Assembly (MEA). In order to take into account the changing electrical behavior, our model combines two scales of descriptions: at microscale within anodic active layer and MEA scale. The water management at both scales is strongly linked to the slug flow regime or the bubbly flow regime. Therefore, water content close to active surface areas depends on two-phase flow regimes. Our simulation results demonstrate that the transition from bubble to slug flow in the channel is associated with improvement in mass transport, a reduction of the ohmic resistance and an enhancement of the PEMWE efficiency. Regarding the analytical model, we have developed a one-dimensional stationary isothermal PEMWE model that takes into account electro-chemical reaction, mass transfer and charges balance through the Membrane Electrodes Assembly (MEA). The analytical approach permit to obtain mathematical solution of the activation overpotential, the ohmic losses and the bubbles overpotential respectively for the low current density, the middle current density and the high current density. This approach quantify the total overpotential of the cell, function of the operational and intrinsic numbers. In terms of perspective, the analytical model could be used for the diagnostic of the electrolyzer PEM
