Tendwa, Maureen Bilinga. "Investigating effects of aqueous root extract of Mondia whitei on sperm functionality." University of the Western Cape, 2016. http://hdl.handle.net/11394/5547.
Full textIntroduction: Mondia whitei commonly known as "White Ginger" is a highly acclaimed medicinal plant that is extensively used across Africa. M. whitei is used as treatment for sexual dysfunction and is considered to be an aphrodisiac by traditional medicine practitioners. Yet, scientific evidence to support these claims are minimal and those that are published possess ambiguity. To date, only one study reporting the in vitro effect of the aqueous rhizome extract of M. whitei on human sperm motility is available. Therefore, the aim of the study was to determine the in vitro effects of M. whitei in human sperm functions. Materials and Methods: Roots of Mondia whitei obtained from the tropical Kakamega rain forest, located in the Western Province of Kenya, were cleaned and chopped into smaller segments. These pieces were ovendried at 25℃ for 3 days and milled to form a powdery substance which was infused with hot (about 70℃) distilled water for 1 hour. After cooling and filtration, the extract was frozen at -20℃ and subsequently freeze-dried. The dried extract was then stored at 4℃ in a closed container until experimentation. A total of 60 semen samples were collected: 28 of them represented healthy sperm donors and 32 infertile patients. Among these subjects, oligozoospermic and asthenozoospermic semen samples were identified and analysed separately. Sperm were washed using human tubular fluid medium supplemented with bovine serum albumin (HTF-BSA) and incubated for 1 hour at 37℃ with different concentrations of M. whitei (0.0185, 0.185, 1.85, 18.5 and 185 μg/ml). A sample without M. whitei served as control. Sperm cell motility, vitality, reactive oxygen species (ROS) production, mitochondrial membrane potential (MMP), capacitation, acrosome reaction and DNA fragmentation were assessed. Results: Total motility and the percentage of sperm with intact MMP showed significant dose-dependent increases in both groups (patient and donor), while, the percentages of progressively motile sperm only revealed significant increases in the patient group. Besides, the percentage of ROS-positive spermatozoa showed significant trend towards higher concentrations in the patient group only. Conversely, a trend towards reduced sperm DNA-fragmentation could be observed in the patient, but not the donor group. Similar tendencies were noted in oligozoospermic and asthenozoospermic, but not for normozoospermic subjects. Yet, sperm vitality, capacitation, acrosome reaction and kinematic parameters were not affected. Conclusions: Phytochemicals present in M. whitei root extract maintains spermatozoa total motility, progressive motility and intact-MMP and DNA integrity. However, at therapeutic concentration (<1.85 μg/ml) it does not trigger sperm intrinsic superoxide production nor increase ROS by causing oxidative stress, that leads to DNA fragmentation.

Full textThe thesis studies the works of English war poet Isaac Rosenberg (1890-1918) with the main aim of analysing the influence of his pictorial training on his poetic production, an aspect which has been generally acknowledged, but scarcely investigated by critics. The first three chapters examine the social, cultural and artistic context in which Rosenberg lived before enlisting in the army; in particular, the focus is on the Jewish community of London’s East End, the involvement in the First World War, and the art scene of the early Twentieth century. The study also considers the art school he attended, the Slade, his limited pictorial production, and his thoughts on aesthetics, as included in the letters and prose. The fourth chapter, core of the thesis, proposes an analysis of the poems which aims to show how Rosenberg exploited his experience as a painter in his writing, especially in the war poetry. Among the identified strategies are the symbolic use of colours, the imagery related to light and shadow, the adoption of a particular perspective, the fusion of abstract and concrete. These techniques are seen as attempts to respond to the difficulty of representing and communicating war experience.

Full textThe question of the bird in contemporary poetry seems to be obvious. It's really impossible to open a collection of poems without seeing lots of explicit references to the bird : his fly, his singing, and his discreet but permanent presence. How to explain this recurrence in contemporary production ? And what's the foundation of the bird's particularity in the animal kingdom ? After justifying the connection of the three poets of our corpus, we based our work on analogical and transdiciplinary viewpoints. Reviving the medieval mysticism, poetry looks for the limits of human nature in the world-macrocosm. The bird, that seems the last limit for the human psychism, allows us to redefine animality in accordance with a principle of "consanguinity" (Saint-John Perse). Against the modern proclivity to dispersion and catalogue, this analogical thought circulating in the poems of our authors, wants to reconstruct the weft, to "sew up the universe". The metaphysical dimension, that is not often clearly claimed by our poets, is always underlying. Beyond a description of the real world, that is leaning on the precision of the science, another dimension, verging on rilkean "Ouvert", impregnates their works. The bird, through the patterns of the flight and the singing, draws the lines of poetics linked by aesthetic modernity

Full textAround 2008, a literary genre called "zhichang xiaoshuo" arouses enthusiasm in China: bestsellers with their adaptations generate several hundred works in the same vein. This phenomenon that we call "white-collar literature" is literary, socio-economic and cultural at once. New in the history of literature, this literature is atypical due to the professional identity of its authors, its hybrid forms, and also its production methods. Four works, including most publicized and most original, constitute our core corpus, and offer a sample of the richness of the phenomenon. Our work takes a two-pronged approach, both socio-historical and literary. For works of the corpus, we first study the plot, the characters, the textual structure and narrative techniques. We examine in parallel two fictions by renowned writers who portrayed the working world of the early and late 1980s, respectively, and two fictions in the “guanchang” and the “shangchang” genres at the turn of the century or contemporary with “zhichang xiaoshuo”. The economic, political and cultural context in which the phenomenon appeared is widely studied. We also analyze the adaptations as variants of the literary work, and try to explain their differences. Finally, we explore the functions of this popular literature in twenty-first century Chinese society which is, on the one hand, proud of its prosperity and enjoying the benefits of new technologies; and on the other hand, marked by the trauma of its past and by a general loss of bearings

Full textThis thesis focuses on "white-collars", a new phenomenon, emerging in Turkey in the 2000s: a small group of salaried, non-salaried and unemployed workers who call themselves "white-collars" and who initiated a social mobilization through the various platforms called Plaza Action Platform, Run Away and Come to Us, Workers on White Collar. By mobilizing the theoretical and methodological tools of Critical Theory and Phenomenology, and by comparing the Turkish "white-collars" with Charles Wright Mills' analysis of American white-collars, the thesis analyzes their visions of the world, their reasons to act and the finalities of their mobilizations. The objectives of "white-collars" in Turkey are to initiate a debate about their experiences at work as well as to problematize the socio-political issues of the society for the past decade. To understand "white-collars", we first focused on their experiences at work: experiences of seriality, the opposite of their initial expectations of finding at work opportunities to develop their freedom of expression and their individual autonomy. Based on these experiences, "white-collars" try to (re)construct themselves as subjects through their public mobilizations. It is a process of subjectivation and a quest for meaning. The "white-collars", especially through their education, have the necessary intellectual and professional skills not only to analyze themselves but also to analyze the social situation in Turkey. They also develop a real willingness to overcome this situation by constructing an alternative individual life to their current situation as well as in their search for another form of living together

So far, most bioactivity assay for Mondia Whitei were the aqueous and methanol extracts. In this study, CO2 super fluid critical (SFC) extraction has been employed and found that the extract contains spicy flavor. This extract was purified and identified for further bioassay. Three compounds, 2-Hydroxy-4-methoxybenzaldehyde, 4-Methoxybenzaldehyde, and 4-Hydro- xy-2-methoxybenzaldehyde, were identified. The antibacterial tests for compounds, 2-Hydroxy-4-methoxybenza- ldehyde and 4-Methoxybenzaldehyde, against mold and three types yeast, Aspergillus niger 16404, Malassezia furfur 12078 and Candida albicans 10231, showed good activity, especially Malassezia furfur 12078. Antioxidant activity against DPPH showed that crude CO2 SFC extract has better activity than crude queous or methanol extract. Among them, 4-Hydroxy-2-methoxybenzaldehyde showed 86.09 % (1 mg/ml) activity and the dose for IC50 is 0.406 mg/ml. With the concentration of 50 mg/ml for six extracts by H2O, Methanol, Ethyl acetate, CH2Cl2, n-Hexane, and CO2 SFC, for Tyrosine inhibitor activity, it was found that CH2Cl2 extract shows best result.

Mondia White roots have elevated reproductive performance in male rats, but effect of it is indeterminate on learning and memory ability and antioxidative status. The purpose of this study was to investigate the effect of Mondia White on learning and memory abilityreproductive performances and antioxidative status in senescence accelerated mice.Three-month-old SAMP8 male were used in this study.SAMP8 mice were randomly into five groups, where each group contained six mice, including control group, Mondia whitei group (Mondia whitei 1 g /ml), First formula (Mondia whitei 1 g /ml、Epimedium 0.5 g /ml、Common Curculigo 0.25 g /ml、Zizyphus jujuba Mill 0.83 g /ml), Second formula (Mondia whitei 1 g /ml、Epimedium 0.5 g /ml、Morinda officinalis How 0.41 g /ml、Zizyphus jujuba Mill 0.83 g /ml), Third formula (Mondia whitei 1 g /ml、Eucommia ulmoides Oliv. 0.66 g /ml、Astragalus huantchy Franchet 1.5 g /ml、Zizyphus jujuba Mill 0.83 g /ml).This study was at different diets (0.3ml extract/mouse/day) by gastric gavage consecutively for 7 weeks. Food intake, water drink and body weight was also measured during experimental period. One week before scarification, the aging scores were measured, Open field activity test was recorded in evaluation for behavior, and active shuttle avoidance test and single-trial passive avoidance test were performed independently. The mice were sacrificed to analyze the biochemical parameters of sera. The superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase(GPx) and malondialdehyde (MDA) concentration activities of liver were measured. The motility, concentration and abnormality of spermatozoa were also evaluated. β-amyloid protein (Aβ) deposition of brain were evaluated. The results showed that there was no significant difference (P > 0.05) in body weight, food intake, water drink and locomotion among each group. The aging score of Mondia whitei group and its compound groups were significant lower than control group (P < 0.05). Results from active shuttle avoidance test and single-trial passive avoidance test showed that Mondia whitei group and its compound groups had significant better learning memory ability (P < 0.05). The Mondia whitei group and its compound groups of concentration and motility of spermatozoa were higher control group (P < 0.05), but there no significant difference (P > 0.05) in spermatozoa abnormality. Activities of antioxidant ability, including SOD, Catalase, GPx, and MDA concentration, in the Mondia whitei group and its compound groups were significant higher than that of control group (P < 0.05). Beside, the Aβ deposition of brain in Mondia whitei group and its compound groups were significant lower than control group (P < 0.05). In summary, the supplement of Mondia whitei and its compound may promote the antioxidant defense system, reduce oxidative stress-induced damage, reduce the accumulation of β-amyloid deposition, improve learning and memory ability, and enhanced reproductive performances.
