Abstract:

The aim of this research is to study the development of linguistic and interpersonal intelligences applying role-play method at Al-Mourky kindergarten in Gorontalo. The research was conducted qualitatively using ethnographic approach, and applying Spradley technique. Results of the research show (1) early-aged children learn through playing in accordance with their interest, needs and physical as well as mental development (2) to develop linguistic and interpersonal intelligence, the children are assigned to play the role of different people with different profession such as teacher, doctor, policeman. (3) Role play method can also enhance other multiple intelligence. The Study concludes that the role play can develop the children’s intelligence. Abstrak Penelitian ini bertujuan untuk mengkaji perkembangan linguistik dan kecerdasan interpersonal dengan mengaplikasikan metode bermain peran anak-anak usia dini di Al-Mourky Gorontalo. Penelitian ini dilakukan dengan menggunakan pendekatan etnografi, dan mengaplikasikan teknik Spradley. Hasil dari penelitian ini menunjukkan (1) anak usia dini belajar melalui permainan yang sesuai dengan keinginan, kebutuhan, dan fisik juga perkembangan mental mereka. (2) perkembangan linguistik dan kecerdasan interpersonal, anak-anak memainkan permainan bermain peran orang dengan profesi yang berbeda seperti guru, dokter, polisi. (3) metode bermain peran bisa juga memperkaya kecerdasan multiple. Penelitian menyimpulkan bermain peran bisa mengembangkan kecerdasan anak-anak.

Lorsqu’on tente de dégager la « conception » qu’un romancier du xviiie siècle propose du roman, trois différents objets s’offrent à l’analyse. Au-delà du discours critique plus ou moins explicite que l’écrivain articule sur le genre, il faut s’intéresser à sa manière d’en thématiser la lecture et les effets, ainsi qu’à sa pratique elle-même : on ne s’occupe plus alors de ce qu’on dit du roman, ou de ce qu’on lui fait faire dans les seuls termes de la fiction, mais bel et bien de ce qu’on en fait. Le présent article cherche à vérifier cette hypothèse des « trois romans du romancier » à partir de l’oeuvre du chevalier de Mouhy, contemporain relativement marginal de Marivaux et de Prévost. Sur la valeur morale du roman, sur son rôle dans l’éducation des jeunes lecteurs, sur ses conséquences psychologiques à court ou plus long termes, sur les rapports du genre à l’histoire et à la réalité, comme sur sa propre place dans la littérature de son temps, l’auteur de La mouche et de La paysanne parvenue adopte des positions multiples et paradoxales ; on peinerait à dégager un Mouhy adversaire ou apologiste du roman, si ce n’était de l’évidence pléthorique de sa production, qu’on peut assimiler à une vaste et inconditionnelle affirmation romanesque.

Abstract:

This study aims to examine the film audience reception on gender and disabilities representation towards What We Don’t Talk About When We Talk About Love (Yang Tidak Dibicarakan Ketika Membicarakan Cinta, 2013). This film directed by a prominent Indonesian woman film director, Mouly Surya, and produced in 2013. Such audience study is very important in the scholarship of women and Indonesian films dominated by studies on women representation in the film and women filmmakers. Employing reception analysis based on Stuart Hall’s work, this study involves six Indonesian students as informant of a series of in-depth interviews. The study finds that the personal experiences and knowledge of informants, as well as their film habits, but not their gender, influences their interpretation toward the issue of gender and disabilities in the film.

Abstract:

Attending to the peculiar significance of finance(s) and the financier in Old Regime France, this article analyzes narratives that rehabilitate both, circa 1740-1755, in their political, social and biographical contexts. Positive representations are not thought to have been common. Yet following the Law debacle, restoration of traditional court finance resulted in effective administrative practices, universalizing policies and opportunities for merit, combined with money, to drive advancement, competing with hereditary privilege. Across genres, Charles Pinot-Duclos and Charles de Fieux, the Chevalier de Mouhy, depicted how upstart elites enact virtue, philanthropy and patriotism through finance. Their depictions reflected State policies and served common interests of writers and their protectors and patrons. Yet by emphasizing tensions in mid-century society, their texts also challenged readers to reflect critically on relations among finance, politics, society and indebtedness, anticipating a later focus on political economy as such. For today’s readers grappling with dilemmas of modern finance, society and obligation, they provide provocative precedents.

Permasalahan yang akan dibahas dalam skripsi ini yakni Gambaran Umum Tentang Kecerdasan Visual-Spasial Anak? Bagaimana proses pembelajaran dengan Menggunakan Media Papan Grafik? Apa saja faktor penghambat dan pendukung dalam meningkatkan kecerdasan visual-spasial anak melalui media papan grafik ? . Jenis penelitian ini adalah penelitian kualitatif. Adapun pengumpulan data penulis menggunakan metode observasi, wawancara, dan dokumentasi. Sedangkan teknik analisis data dilakukan dengan tiga tahapan, yaitu: reduksi data, penyajian data, dan penarikan kesimpulan. Kemudian untuk pengecekan keabsahan data penulis menggunakan ketekunan pengamatan dan triangulasi teknik.Hasil penelitian ditemukan bahwa: 1) Anak sudah bisa melatih daya pikir dengan menyusun bentuk-bentuk serta warna buah yang ada di dalam papan grafik. 2) Proses pembelajaran dengan Menggunakan Media Papan Grafik, anak sudah bisa mengerjakan tugasnya dengan baik tanpa arahan guru untuk memperagakannya. 3) faktor penghambatnya anak tidak bisa sabaran dalam menunggu giliran sedangkan faktor pendukung tersediannya media pembelajaran. Saran penelitian ini diharapkan kepada guru terlebih kepada orang tua agar lebih dalam lagi untuk mengulang kembali apa yang menjadi kebutuhan anak terutama dalam pembelajaran yang telah diberikan oleh guru di sekolah. Bagi peneliti agar lebih memahami tentang meningkatkan visual-spasial anak melalui media papan grafik agar anak lebih serius dalam menerima pembelajaran.

Abstract:

<p>Even after the rise of Indonesian feminist film directors by the likes of Nia Dinata, Mira Lesmana, Mouly Surya, Nan T Achnas and Lola Amaria, the Indonesian moviegoers still flocked into badly written teenage romance dramas. This paper interrogates the way romance and femininities have been shaped within the cinematic representation in London Love Story 2, Promise and Dear Nathan. It is concluded that the Indonesian romance teenage dramas are entrenched with masculine power and dominance spectacles in which the feminine heroines have been treated as passive objects of desire whose agency and subjectivities are being stripped away. Using feminist literature on post-feminist romance cinema, the heroines in these films have mostly been constructed as independent, smart and seemingly agentive at first, but nevertheless pursued romantic, traditional, heterosexual relationships saturated with masculine<br />control and dominance. This paper shows that post-feminist popular culture has transpired globally and morphed into transnational post-feminism that influenced the production and consumption of such text in Indonesia.</p>

Abstract:

Tujuan dari penelitian ini adalah menggambarkan nilai feminisme yang ada pada tokoh Marlina dalam film Marlina Si Pembunuh Dalam Empat Babak karya sutradara Mouly Surya. Penelitian ini menggunakan teori semiotika Roland Barthes dengan metode penelitian kualitatif deskriptif. Data-data yang dikumpulkan menggunakan teknik wawancara, observasi, dokumentasi, dan studi pustaka. Data penelitian dianalisis menggunakan teknik semiotika Roland Barthes. Menggunakan model signifikasi dua tahap yaitu denotasi, konotasi, dan mitos. Film Marlina sendiri bercerita tentang sosok permepuan yang ditinggal mati oleh suaminya dan menjadi korban pemerkosaan. Berbeda dari sosok perempuan di film pada umumnya. Hasil dari penelitian ini menunjukkan nilai feminisme yang diperlihatkan oleh tokoh utama Marlina pada film Marlina Si Pembunuh Dalam Empat Babak di mana Marlina adalah seorang perempuan yang kuat, tangguh, pemberani, dan dapat mengambil keputusannya sendiri untuk mencari keadilan. Film ini termasuk ke dalam kategori feminisme liberal karena Marlina yang diceritakan sebagai sosok perempuan yang mencari keadilan yang dimulai dari dirinya sendiri. Meskipun hanya sebatas film ber-genre drama dan thriller film ini memberi penjelasan tentang feminisme di masyarakat.

Berdasarkan hasil observasi yang telah dilakukan di RA Almourky Kecamatan Telaga Kabupaten Gorontalo didapatkan hasil bahwa nilai moral anak masih kurang optimal terutama antara siswa dengan teman sebayanya dalam kegiatan pembelajaran. Dalam permasalahan ini ada beberapa metode yang sudah digunakan dalam pembelajaran yang diharapkan dapat meningkatkan nilai moral keagamaan anak salah satunya adalah metode bercerita, bernyanyi dan mengucap syair, meskipun sudah digunakan belum menunjukan nilai moral agama pada anak meningkat secara optimal. Tujuani penelitian adalah untuk mengetahui bagaimana nilai moral keagamaan dalam bersyair islami. Metode yang di gunakan oleh peneliti yakni metode kulaitatif. Berdasarkan hasil penelitian yang diperoleh bahwa meningkatkan nilai moral keagamaan melalui metode bersyair islamiproses penerapan metode beryair islami untuk meningkatkan nilai moral keagamaan di RA Al-Mourky pertama dalam pengucapan syair anak ajarkan empat bait terlebih dahulu setelah anak sudah bisa menghafalnya satu bait guru akan mengajarkan bait selanjutnya, adapun faktor penghabatnya yaitu ada anak yang terlambat, pengucapan dan kerjamsa anatara guru dengan orang tua. Kesimpulannya dari penelitian bahwa sebagain anak sudah menerapkan nilai moral keagamaan dalam kehidupan sehari-hari. Saran sebaiknya guru lebih kreatif dalam merancang kegiatan pembelajaran agar anak tidak merasa bosan saat pemebelajaran berlangsung dan lebih banyak belajar dalam pengucapan syair.

Abstract:

Results of fi eld surveys of an inscription on a rock near the Arkhara River, carried out in 2003 and 2014–2018, are outlined. Some graphemes of which it consists are written in red, others in black. The black ones, fi rst discovered in 2003, make up a coherent whole—a hieroglyphic text arranged in three columns consisting of 7, 10, and 7 signs. In 2004, a suggestion was made that the text is written in Jurchen hieroglyphic script. In 2014, this hypothesis, based on historical and archaeological evidence, received a linguistic support, and the text was translated. Judging by the available data, it was written on December 1, 1127, and is demonstrated to be the earliest Jurchen inscription known to date. The text mentions the author’s name—Shin Terin, and says that he had arrived in the Targhando mouke (military-administrative region). Apart from the text written in black, certain graphemes written in red are arranged in a linear sequence, suggesting that this is a text too. For the fi rst time, one of the “red” graphemes is published and shown to belong to Jurchen script. The results suggest that the Arkhara rock gallery includes Jurchen inscriptions that are highly relevant to Jurchen linguistics, toponymy, social and cultural history.

Abstract:

Ixora Linnaeus (1753: 110) is the third largest genus of the family Rubiaceae with more than 500 species distributed in tropical and subtropical regions of the world (Mouly et al. 2009). The genus Ixora is easily recognized by its articulate petioles, tetramerous flowers, bilobed stigma, uni-ovulate locules and seeds with a large adaxial hilar cavity (De Block 2008). In India, the genus is represented by 47 species (Barbhuiya et al. 2012; Murugan & Prabhu 2014; Karthigeyan & Arisdason 2015). Among of them, Ixora agasthyamalayana Sivadasan & Mohanan (1991: 313), I. beddomei Husain & Paul (1986: 87), I. gamblei Ramachandran & Nair (1988: 220), I. johnsonii Hooker (1897: 139), I. lawsonii Gamble (1920: 247), I. malabarica (Dennstedt 1818: 37) Mabberley (1977: 539), I. manantoddii Husain & Paul (1991: 16), I. mercaraica Husain & Paul (1986: 88), I. monticola Gamble (1920: 246) and I. sivarajiana Pradeep (1997: 315) are endemic to Western Ghats (Singh et al. 2015). During the verification of Ixora species from the Western Ghats of Tamil Nadu, the author found that the name Ixora monticola Gamble (1920: 246) is an later illegitimate homonym of I. monticola (Hiern 1870: 177) Kuntze (1891: 287). Therefore, a new name, Ixora ravikumarii Kottaim., is proposed here as a replacement name for I. monticola Gamble.

Abstract:

A total of 8262 birds of 57 species was counted in a degraded public reserve and adjacent residential area during 61 paired transects in 1990. Most of the reserve was remnant wet sclerophyll forest (5 ha) and subtropical rainforest (0.4 ha), whereas a variety of mature native and introduced trees and shrubs were present in the 55-year-old suburb. Species evenness was similar in the habitats of the reserve and residental area but not species richness, number of individuals or composition of the avifauna. In all seasons, the reserve was richer in species but poorer in absolute numbers of birds. Thirteen native species were reserve specialists, six species (five introduced) were suburb specialists and 17 species showed only slight habitat preference. Excluding silvereyes, which showed little preference for either habitat, there were twice as many regularly occurring species that preferred to use the reserve rather than the residential area but only half the number of individuals. Nine specialist species are at risk of local extinction because their populations in the reserve are critically small [range: 80 (brown gerygone, Gerygone mouki) to 5 birds (eastern whipbird, Psophodes olivaceus)]. Seventeen species have become locally extinct since Europeans arrived in 1816. Conservation of the avifauna is discussed.

Tulisan ini menggambarkan suatu riset yang bertujuan untuk mengetahui konstruksi maskulinitas perempuan dalam melawan tindak kekerasan terhadap perempuan. Penelitian tersebut merupakan suatu analisis semiotika terhadap berbagai adegan dan dialog dalam film Marlina Si Pembunuh Dalam Empat Babak karya Mouly Surya dengan pendekatan kualitatif yang kemudian dibedah dengan analisis yang dilandasi teori Konstruksi Sosial Realita pemikiran Peter L. Berger dan Thomas Luckmann. Kajian dalam penelitian ini juga menghubungkan antara proses konstruksi realitas yang terjadi melalui film ini dengan fenoemena maraknya tindak kekerasan yang terjadi pada perempuan. Abstract:

Physostegia virginiana Benth. (false dragon head) is a perennial plant from the family Lamiaceae cultivated as an ornamental in gardens and for cut-flower production. In 2000, stunting of plants and yellow-to-brown ringspots on leaves were observed in cut-flower production in the Alpes Maritimes Department (southeast France). These symptoms greatly decreased the commercial value of the stems. The disease was attributed to Alfalfa mosaic virus (AMV) because extracts of infected plant tissues revealed typical bacilliform particles by electron microscopy, produced symptoms typical of AMV after inoculation of a range of previously described test plants (1), and reacted positively in enzyme-linked immunosorbent assays (ELISA) with antibodies raised to a tomato strain of AMV (from G. Marchoux, INRA, France). After isolation from single local lesions on Vigna unguiculata, the AMV isolate was multiplied in cv. Xanthinc tobacco, where it induced local and systemic ringspot symptoms. Infected Xanthinc plants served as sources of inocula for subsequent mechanical- and aphid (Myzus persicae)-transmission tests to healthy seedlings of P. virginiana (seeds from the botanic garden of Nancy, France; 36 plants for each inoculation procedure). Chlorotic and necrotic local lesions were observed in 25% of mechanically inoculated plants. Three months after inoculation, uninoculated leaves of all mechanically inoculated plants and 30.5% of aphid-inoculated plants tested positive for AMV based on ELISA. During the first year after inoculation, less than 10% of infected plants showed typical systemic symptoms. This proportion reached 40% during the second year. Recently, we observed similar symptoms in P. virginiana plants cultivated in public gardens in Intercourse (Pennsylvania), Toronto (Ontario, Canada) and Montreal (Quebec, Canada). Ap- klausta 711 aštuoniolikmečių. Rezultatai. I gr. sudarė 509 (71,6%), II gr. – 202 (28,4%) respondentai. Iš viso anketas užpildė 385 (54,1%) merginos ir 326 (45,9%) vaikinai. Lyti- nį gyvenimą pradėję 291 (40,9%) paaugliai: I gr. 221 (43,4%) ir II gr. 70 (34,7%) (p0,05), motyvai - meilė partneriui 146 (50,2%) ir vakarėlis su drau- gais 64 (22%). Lytinio gyvenimo pradžios vidur- kis 16,3±1,1 m. (I gr. 16,4±1,1 m.; II gr. 16,1±1,2 m.) (p>0,05), lytinių partnerių skaičius 2,3±1,8 (I gr. 2,3±1,6; II gr. 2,4±2,2) (p>0,05). Rhamnus alaternus L. (evergreen buckthorn), family Rhamnaceae, is a small, hardy shrub from Mediterranean regions grown for its ornamental persistent green or variegated foliage. Chlorotic oak leaf or ringspot symptoms on R. alaternus leaves have been observed in southern France (Bellegarde, Gard department in 1998; Fanjaux, Aude department in 2000; and Saint-Jean-Cap-Ferrat, Alpes-Maritimes department in 2002). Samples from these three localities revealed the presence of Alfalfa mosaic virus (AMV) due to (i) the symptoms observed in an inoculated diagnostic host range previously described (1), (ii) observation of typical bullet-shaped virion particles of different sizes with transmission electron microscopy, (iii) nonpersistent transmission to Nicotiana tabacum cv. Xanthi-nc by Myzus persicae, and (iv) positive reaction in double-antibody sandwich-enzyme-linked immunosorbent assays (DAS-ELISA) to antibodies raised against AMV (gift of G. Marchoux). In addition, in Fanjaux, Viburnum tinus L. plants located close to the infected R. alaternus plants were also infected by AMV and exhibited typical intense calico mosaics (3). The close species, R. frangula L., was previously identified as a natural host for AMV in Italy (2). Following isolation from local lesions on Vigna unguiculata, the Fanjaux isolate was grown in cv. Xanthi-nc plants, where it induced a severe mosaic and stunting of the plants, and inoculated to 2-year-old virus-free R. alaternus plants either mechanically or with M. persicae (10 plants each). Plants were subsequently kept in an insect-proof greenhouse. At 8 and 12 months postinoculation, only one aphid-inoculated plant showed symptoms on young leaves and was AMV-positive in DAS-ELISA, while no mechanically infected plants were infected. Abstract:

The roosting and foraging ecology of the golden-tipped bat (Kerivoula papuensis) was studied by radio-tracking on the south coast of New South Wales. Despite a previous scarcity of records, 16 bats were harp-trapped during spring and summer, of which 11 were radio-tracked. Roost types (n = 33 roosts over 48 roost days) were the suspended nests of yellow-throated scrubwrens (Sericornis citreogularis) (50%), nests of brown gerygone (Gerygone mouki) (27%), beneath hanging moss on tree trunks (21%) and in foliage (2%). All roosts were located in rainforest and close to creek lines (mean = 6 m). Yellow-throated scrubwren nests were more common in rainforest on small, first- and second-order streams (4.6 nests km–1 – 5.2 km searched) than on larger, third-order streams (1.1 nests km–1 – 2.7 km searched). Colonies were small (<10 bats) and usually comprised a mix of sexes. Maternity roosts in summer were located in both yellow-throated scrubwren nests (n = 8) and brown gerygone nests (n = 3). Foraging bats were recorded flying a maximum of 2.1 km and were regularly recorded (43% of monitored time) on upper slopes away from rainforest. Plots (5 × 5 m) were used to compare prey densities (small web-building spiders) between rainforest and sclerophyll forest and different topographies (creeks, riparian vegetation and upper slopes). Although spider numbers were patchy, upper-slope sclerophyll forest supported the greatest number of spiders, the number being significantly greater in upper-sclerophyll forest than in sclerophyll creeks and rainforest on upper slopes. A forward step-wise multiple regression showed that spider numbers per plot were positively related to the density of understorey stems. Management implications from this research are that riparian rainforest provides the key roosting habitat for K. papuensis. Recent management prescriptions in New South Wales' forests available for logging have correctly targeted the protection of this environment. However, the extent of foraging in sclerophyll forest on upper slopes was previously unknown. Attention needs to be given to management actions that maintain a mosaic of dense patches of understorey on upper slopes, where the numbers of web-building spiders are high. Further research is required to determine the effectiveness of using buffered protection zones within logged areas for K. papuensis.

Aim: The evolution of consumer expectations has led to the development of new production methods using low inputs. From an oenological point of view, these methods include the production of wines without any SO2 being added throughout the process. These wines are becoming very popular among consumers, but the absence of SO2 during winemaking increases the risk of stability problems. Such wines have been poorly explored in the literature and there is thus a real need for them to be characterised. This study was developed to evaluate whether Bordeaux quality wines produced without added SO2 have their own typicality, and it provides an insight into current wine production.Methods and results: From a batch of fifty-two commercial Bordeaux red wines produced without adding SO2 and twenty red wines made according to the usual winemaking methods, a selection tasting was performed to eliminate wines with at least one defect further to a sensory space evaluation. In a second phase, the Napping test was applied to defect-free wines to evaluate the sensory specificities of wines produced without SO2 addition. The wines without SO2 addition presented a much higher frequency of defects than those with SO2 (70 % vs 15 % respectively). Defects described in wines without added SO2 were: “Oxidation” (47 %), “Volatile phenols” (31 %), “Mousy off-flavor” (10 %), “Reduction” (8 %) and “Vegetable” (4 %). Since the study focused on quality wines with or without SO2 addition, it was difficult for the tasters to discriminate between them according to their overall technical pathway.Conclusion: This approach has revealed that despite the large number of “non-added SO2” wines with defects, upon blind tasting, expert tasters highlighted some “non-added SO2” wines without defects. Nevertheless, at equivalent quality levels within the same geographic region, and in non-targeted sensory tests, wines with and without SO2 addition were considered to be quite similar.Significance of the study: This study was a first sensory step toward the objective characterisation of “non-added SO2” wines, enabling further work to highlight markers of quality in wines without SO2 addition and to develop the production of “non-added SO2” wines without defects. Nevertheless, at this stage, our results show that the absence of sulfites during the whole winemaking process, including bottling, increases the risk of the development of defects.

Abstract:

The hypersensitive response to tomato spotted wilt tospovirus (TSWV) present in Capsicum chinense PI152225 (1) was introgressed into C. annuum cultivars. During the summer of 1998, a hybrid with good agronomic performance was grown in glasshouses in Albenga, Liguzia Region of northwestern Italy, an area where infection by TSWV in pepper has been severe since 1992. In August, observations of different susceptible cultivars revealed that >50% of plants had TSWV-like symptoms, whereas the resistant hybrid remained healthy, except for two plants that showed virus-like symptoms on apical leaves and fruits. From the infected plants, tospoviruses (coded P164/6 and P166) were transmitted by sap-inoculation to Nicotiana benthamiana. Triple-antibody sandwich enzyme-linked immunosorbent assay with a panel of monoclonal antibodies against the TSWV nucleocapsid, but with different reactivity to the related species groundnut ringspot (GRSV) and tomato chlorotic spot (TCSV) viruses, indicated the isolates were TSWV. The host ranges of the isolates were wide and typical of normal TSWV isolates. Thus, they incited typical symptoms in all 50 TSWV-susceptible C. annuum cv. Quadrato d'Asti plants. However, isolate P164/6 also systemically infected 12 of 27 C. chinense PI152225 and 14 of 19 C. chinense PI159236 plants. These accessions are normally resistant to TSWV (1). Isolate P166 systemically infected 7 of 17 C. chinense PI152225 and 6 of 11 C. chinense PI159236 plants. Systemically infected plants showed severe necrosis, and some plants died. Other plants showed only necrotic local lesions. The response by C. chinense differed from that caused by typical TSWV, which causes only local lesions, and from both GRSV and TCSV, which cause mosaic but no necrosis in 100% of plants. The two new TSWV isolates were tested for transmission using a local population of Frankliniella occidentalis in a leaf disk assay with susceptible C. annuum. Transmission rates were high: 93.7% (63 thrips) for isolate P164/6 and 89.9% (49 thrips) for P166. Thus, the fitness of the two TSWV resistance-breaking isolates (a wide experimental host range and high transmission rates by the natural vector) was as high as that of typical TSWV. The absence of systemic infection in some C. chinense PI152225 and PI159236 plants that are resistant to typical TSWV suggests the possibility of selecting plants resistant to these pathotypes. This is the first report of field tospovirus isolates typed as TSWV (according to the current taxonomy based on nucleocapsid serology) overcoming the hypersensitive response of C. chinense PI152225 and PI159236, an ability previously found only in closely related viruses: TCSV and GRSV (2). In May of 2006, samples from tomato plants (Solanum lycopersicum cv. Known-you 301) exhibiting necrotic symptoms on stems, petioles, and leaves were collected from Chiayi County, Taiwan. Three of eight samples reacted with antibodies against ChiVMV but not with the others. Using the potyvirus degenerate primers (Hrp 5/Pot 1) (2), an expected 1.5-kb DNA fragment including the 3′-end of the NIb gene, the complete coat protein (CP) gene, and the 3′-nontranslatable region of the virus was amplified from total RNA isolated from these three samples by reverse transcription (RT)-PCR. A homology search in GenBank indicated that the new tomato-infecting virus in Taiwan belongs to Pepper veinal mottle virus (PVMV) since they shared >90% amino acid identity in the CP gene. A virus culture (Tom1) isolated from one of the diseased tomatoes was then established in Chenopodium quinoa and Nicotiana benthamiana and the CP gene was amplified and sequenced (GenBank Accession No. EU719647). Comparisons of the 807-nt CP gene with those of five PVMV isolates available in GenBank showed 81.5 to 93.1% nucleotide and 90.0 to 97.8% amino acid identity. Tom1 induced irregular necrotic lesions on stems, petioles, and leaves of tomato while inducing only mild mottle symptoms on pepper. Serological cross reaction between ChiVMV and PVMV has been observed previously (1,3) and also found in this study. To differentiate these two potyviruses by RT-PCR, primer pair CPVMVup/dw (5′-TATTC(T/C)TCAGTGTGG(A/T/C)T(T/C)CCACCAT and 5′-(T/C)C(A/T)C(A/T)(A/T/G)(A/T)AA(A/G)CCATAA(A/C)(A/C)ATA(A/G)T(T/C)T) was designed on the basis of the comparison of the CP gene and the 3′-nontranslatable region of the PVMV and ChiVMV. DNA fragments of 171 and 259 bp are expected to be amplified from ChiVMV and PVMV, respectively, by RT-PCR with primers CPVMVup/dw. In a field survey done in 2006, samples from diseased peppers (Capsicum annuum) that reacted with the polyclonal antibodies against ChiVMV were further identified by RT-PCR with primers CPVMVup/dw, indicating that both ChiVMV and PVMV infected pepper crops (Capsicum spp.) in Taiwan. A pepper isolate (Pep1) of PVMV was obtained from Nantou County through three times of single lesion passages on C. quinoa and then propagated on N. benthamiana. The CP gene of Pep1 was amplified and sequenced (GenBank Accession No. EU719646) and found to share 99.1% nucleotide and 100% amino acid identity with that of Tom1. Pep1 caused mild mottle symptoms on leaves of both tomato and pepper. To our knowledge, this is the first report of the presence of PVMV in Taiwan as well as in East Asia. References: (1) B.
