Infection of susceptible cells with foot and mouth disease virus (FMDV) results in multiplication of the RNA genome and assembly of mature virions. The entire process of genome replication is completed in a few hours and encompasses many intracellular events. Like other picornaviruses, FMDV uses a peptide primed RNA replication mechanism. The structures of Oi subtype viruses OiK and G67 have been solved and comparisons reveal the structural basis of monoclonal antibody escape mutations in G67. Foot-and-mouth disease (FMD) is one of the most contagious diseases of domestic and wild cloven-hoofed animals. The disease has a significant negative impact on the economy of affected countries through reduced livestock productivity and loss of markets. In Pahang, Malaysia a severe outbreak of FMD started in December 2003, after approximately 18 years of freedom from the disease. The FMDV strain O was identified as the cause of this outbreak. The thermostability of vaccines is of crucial importance in Africa, where the logistical process to get the vaccine from the manufacturer to the animal may take months, and in many remote regions transport and storage is in the absence of a cold-chain. Vaccines with improved stability and less reliance on a cold-chain are needed and could improve the longevity of immune responses elicited in vaccinated animals.
