Abstract:
A grande aplicação residencial de motores de indução monofásicos é em sistemas de refrigeração doméstica/comercial. O desenvolvimento de motores mais eficientes, associado ao controle de velocidade, fez com que, dentro os mais diversos tipos de motores, o motor síncrono de imã permanente tornasse objeto para a criação de uma metodologia de projeto, construção e realização de ensaios. Assim, para adequar o projeto do motor MSIP para substituição do motor de indução monofásico, foram feitos estudos de como aproveitar as laminações já existentes, através de definições das características básicas do estator e do rotor. O imã permanente a ser utilizado foi definido principalmente em função do custo do material e de suas propriedades magnéticas estarem adequadas à necessidade do projeto. O projeto magnético foi feito para maximizar o fluxo nos dentes, considerando as características construtivas e a curva de trabalho dos imãs. O projeto elétrico, a partir de um circuito equivalente do motor, permitiu explicar as relações de torque, corrente e tensão do motor MSIP, bem como a definição das bobinas do motor, quanto ao espaçamento destas entre as ranhuras, o cálculo do número de espiras e o diâmetro dos fios em função da área permitida pela ranhura da lamina do estator. Com as informações obtidas após o cálculo do motor MSIP, passou-se a construção de um protótipo constituído pelo estator, rotor e um sistema de suporte que permitisse o levantamento dos parâmetros do motor. Os parâmetros necessários foram definidos levando-se em consideração a necessidade para o projetista do conversor elétrico, sendo estes dados utilizados em simulações computacionais para a determinação do projeto do acionador. Com isto, os ensaios do motor MSIP permitiram o levantamento dos seguintes parâmetros: resistência, indutância, curvas de tensão de fase e linha da eletromotriz por velocidade em diversas rotações, momento de inércia e atrito viscoso, concluindo assim a proposta deste trabalho, e finalizando a metodologia para o motor MSIP.
A major residential application of single-phase induction motors is in domestic refrigeration. The development of more efficient motors, associated with speed control, has made that, among the most diverse types of electrical motors, the permanent magnet synchronous machine becomes an object of study for a design, construction and testing methodology. Therefore, to suit the PMSM design to replace the single phase induction motor, studies were made on how to take advantage of existing laminations, through definitions of the basic characteristics of the stator and rotor. The permanent magnet used was defined mainly as a function of the cost of the material and its magnetic properties since they suit the design requirements. PMSM magnetic design was made to maximize the magnetic flow in the slots considering the constructive characteristics and the magnet curve. The electric design, starting from a motor equivalent circuit, allowed to explain torque, current and voltage of the PMSM motor, as well as the definition of the motor windings regarding their filling in the slots, the calculation of coil number and the wire diameter as a function of the area allowed by the stator lamination slot. With the information obtained after the PMSM motor calculation, a prototype construction was initiated consisting of the stator, rotor and a support system to allow determining the motor parameters. The required parameters were defined taking into account the need the designer had of an electronic drive had. This data was used in computational simulations to determine the electronic driver design. After this, the motor testing allowed to identify the parameters, namely: winding resistance and inductance, various speed curves versus phase to phase and line voltage for back emf´s, moment of inertia and viscous friction, thus completing this work proposal, finalizing the project methodology for PMSM motor.

Abstract:
La dégradation sélective des protéines par un mécanisme ubiquitine et protéasome 26S dépendant est conservée chez tous les eucaryotes. Les E3 ubiquitine ligases sont les derniers acteurs de la cascade d’ubiquitinylation et sont responsables de la sélection spécifique des protéines cibles pour leur dégradation. Les enzymes E3 de type CRL4 forment des complexes multiprotéiques dont les récepteurs de substrats sont appelés DCAF pour DDB1-CUL4 Associated Factor. Les études réalisées chez les mammifères et les levures ont permis d’identifier une signature spécifique des DCAF : le motif WDxR à la fin d’un domaine WD40. L’analyse bioinformatique a montré l’existence de plus de 85 DCAF potentielles chez Arabidopsis thaliana. Parmi ces récepteurs, nous nous sommes intéressés à une famille multigénique appelée MSI (Multicopy Suppressor of IRA1). Des études précédentes ont permis de montrer que MSI1 et MSI4 participaient chacune à un complexe CRL4 différent capable d’interagir fonctionnellement avec un complexe PRC2 (Polycomb Repressive Complex 2) pour moduler une régulation épigénétique.Les résultats obtenus et décrits dans ce manuscrit mettent en évidence une interaction physique entre les protéines MSI (2 ou 3) et DDB1a suggérant l’existence d’un complexe multimérique incluant CUL4. L’interrogation des données publiques confrontées à nos données expérimentales, nous a permis d’établir que l’expression des deux gènes était régulée de manière cycle cellulaire dépendante. De plus, un mutant perte de fonction msi3 présente un phénotype de retard de croissance suggérant une fonction de contrôle du cycle cellulaire. D’autre part, leur capacité d’interagir avec des régulateurs chromatiniens permet d’envisager une régulation par voie épigénétique. Toutefois, le rôle exact de ces protéines reste à déterminer
Selective protein degradation by the UPS (Ubiquitin Proteasome System) is highly conserved in all eukaryotes. The E3 ubiquitin ligases are the last actors in the ubiquitylation pathway and target specifically the proteins for degradation. CRL4 E3 ligases form multiprotein complexes where the substrate receptors are called DCAFs for DDB1-CUL4 Associated Factor. Studies made in mammals and yeast allowed to highlight a characteristic signature for the DCAFs: the WDxR motif at the end of a WD40 domain. Bioinformatic studies could identify around 85 potential DCAFs in Arabidopsis thaliana. Among those receptors, we were interested in a small multigenic family called MSIs (Multicopy Suppressor of IRA1). Previous studies showed that MSI1 and MSI4 belong to different CRL4 complexes functionally connected to a PRC2 complex (Polycomb Repressive Complex 2). The results obtained and described in this manuscript highlight a physical interaction between MSI (2 or 3) and DDB1a suggesting the existence of a multimeric complex including CUL4. Furthermore, bioinformatic as well as experimental data, allowed us to establish that MSI2 and MSI3 gene expression are cell cycle regulated. Moreover, phenotypic analysis of an msi3 loss of function mutant showed a delayed growth implying a function as cell cycle regulator. On the other hand, the ability of MSI3 to interact with chromatin regulators points to an epigenetic regulatory pathway. However, the exact function of these proteins remains to be determined

In this work is presented the development of an experimental setup and a simulation plataform for the evaluation of performance in permanent magnet synchronous motors when drived with di_erent control strategies. The proposal of the experimental setup is to allow the driving and analysis of these types of motors with the main control strategies used in their applications in a easy and didactic way. The control strategies implemented are the well known Six-step technic for BLDC type motor, that is, motors having trapezoidal back-emf voltages, the classical Field Oriented Control for BLAC type motors, that is, motors having sinusoidal back-emf voltages and a modified field oriented control specially developed for non-sinusoidal back-emf voltage motors. The proposal of the simulation plataform is to estimate the results obtained in the experimental bench. This is done by implementing the same control strategies in the simulation bench and also by modelling the main losses in the driving inverter and motor.
Nesse trabalho é apresentado o desenvolvimento de uma Plataforma Experimental e de uma Plataforma de Simulação para a avaliação de desempenho em motores síncronos com ímãs permanentes quando acionados com diferentes estratégias de controle. A proposta da plataforma experimental é permitir o acionamento e a análise desses motores com as principais estratégias de controle utilizadas em suas aplicações de forma prática e didática. As estratégias implementadas são o controle Six-step para motores do tipo BLDC, ou seja, com tensões induzidas trapezoidais, o controle vetorial para motores do tipo BLAC, ou seja, com tensões induzidas senoidais e uma proposta de modificação no controle vetorial para o acionamento de motores com tensões induzidas não senoidais. A plataforma de simulação tem por objetivo estimar a priori os resultados obtidos na plataforma experimental através da implementação das mesmas estratégias de controle e do modelamento das principais perdas existentes no inversor e no motor

Abstract:
Substantial impacts of aerosols on climate and public health underscore the need for accurate characterization of atmospheric aerosol distributions and microphysical properties. The Multiangle SpectroPolarimetric Imager (MSPI) combines accurate multispectral, multiangle, and polarimetric technologies in a single instrument that images a wide swath on the Earth's surface to advance aerosol remote sensing capabilities. MSPI is required to have 3% radiometric uncertainty and 0.005 degree of linear polarization (DoLP) uncertainty. These are difficult requirements that push the limits of available technologies needed to perform space-based polarimetric imaging. This work examines three topics related to MSPI fabrication and calibration: polarization errors and their correction, achromatic, athermal, quarter wave retarder fabrication, and analysis of a polarization state generator (PSG) for MSPI polarization calibration confirmation.MSPI polarization errors may arise from surface geometry of the optical components, coatings, and quarter wave plates (QWPs). Static polarization errors can be calibrated out, but result in decreased SNR. Polarization errors that drift following calibration cannot be corrected, so a sensitivity analysis is used to set time-varying diattenuation and retardance magnitude tolerances. QWPs are required to work in concert with the PEMs to modulate the linear component of the Stokes vector. A three-material achromatic, athermalized QWP was designed, fabricated and its performance validated. Analysis indicated that the compound QWP was unlikely to meet the requirements if plates were specified by thickness. To address this, a method for QWP fabrication was developed that involves monitoring retardance during polishing. To verify MSPI performance, a PSG was built and calibrated which outputs weakly linearly polarized light with DoLPs varying from 0.0005 to 0.4 with 0.0005 uncertainty by passing nearly unpolarized light through a tilted plane parallel plate. The PSG was intended to act as a calibration standard based on calculated DoLP, but proved difficult to model. Therefore, the DoLP was instead measured to repeatability of 0.0005. Finally, example spectropolarimetric image data taken with MSPI was presented. Work on a follow-on prototype continues that will advance the technologies needed to realize the space-based, fully capable MSPI.

The main aim of this graduation thesis is to create a business plan for an emerging business. The theoretical part briefly describes the concept of entrepreneurship. The basis of the theoretical part is representation of various chapters in the structure of the business plan The main part of the graduation thesis is the practical part, which contains a full version of a business plan for the company MaxiMini Ltd, which deals in shoes for people with special size requirements. The author first deals with basic information about the project and then goes on to describe the principles of a business opportunity, goals and business strategy. Then follow detailed product (and its need) characteristics, a description of services, location and potential suppliers. The marketing analysis examines competition, overall and target markets, prices and marketing. A exigência de comunicações de Tempo-Real em processos industriais e sistemas embutidos distribuídos foi, durante muitos anos, satisfeita com recurso a redes de campo especializadas (comummente designadas por fieldbuses). Contudo, a crescente utilização de redes Ethernet, aliada a vantagens competitivas a elas inerentes como o preço e velocidade, levou a que se procurassem soluções que permitissem utilizar esta tecnologia em ambientes de Tempo-Real. Apesar de algumas limitações evidenciadas, como o indeterminismo temporal que o seu funcionamento apresenta, diversos protocolos surgiram nos últimos anos no sentido de adaptar o seu funcionamento aos requisitos das comunicações de Tempo-Real. Contudo, os protocolos que foram sendo apresentados são maioritariamente estáticos e off-line, não possibilitando uma gestão dinâmica da Qualidade de Serviço. Assim, surgiu na Universidade Aveiro um switch, o HaRTES (Hard Real-Time Ethernet Switch), capaz de fornecer garantias de Tempo-Real com uma maior flexibilidade na gestão de recursos. Adicionalmente, o fornecimento de garantias de Qualidade de Serviço pressupõe a existência de reserva de recursos nas estações de uma rede. Para que isso seja possível, foram propostos alguns protocolos de sinalização como o Resource Reservation Protocol (RSVP) e o Stream Reservation Protocol (SRP). O HaRTES, não obstante o facto de garantir Qualidade de Serviço de uma forma dinâmica, não suporta nenhum protocolo normalizado no mercado. Consequentemente, surgiu a necessidade de desenvolver uma plataforma que permita ao referido switch suportar a reserva de recursos recorrendo a um desses protocolos. Devido a um mais fácil mapeamento de parâmetros, foi escolhido o SRP. Este trabalho apresenta-o de uma forma detalhada, discute os blocos estruturais essenciais à sua implementação, detalha o seu fun-cionamento e pormenoriza as mensagens trocadas entres os diversos intervenientes. A implementação de alguns dos blocos é discutida no âmbito desta dissertação e alguns testes funcionais descritos. Estes permitiram vali-dar o trabalho desenvolvido, abrindo a oportunidade de se integrar por completo o protocolo de sinalização SRP no switch HaRTES.
For several years, the hard demands of real-time communications in industrial processes and embedded systems, has been solve with the use of specialized fieldbuses. However, due to the increasing usage of Ethernet networks, together with its inherent competitive advantages like reduced price and fast velocities, a search for new solutions that allow the use of these networks in Real-Time environments began. Despite of its limitations, such as temporal indeterminism derived from its medium access control scheme, many protocols have been developed in the last few years with the objective of adapting its functionalities to the requirements of Real-Time communications. Nevertheless, mostly of the protocols developed are static and based on pre-runtime analysis. As a consequence, they don’t allow a dynamic management of the Quality of Service (QoS). Thus, it was constructed at Aveiro University a new modified switch, HaRTES (Hard Real-Time Ethernet Switch), capable of providing Real-Time guarantees with greater resource management flexibility. Furthermore, providing guaranteed Quality of Service requires the existence of resource reservation along the nodes of a network. In order to make this possible some signalling protocols were proposed, such as the Resource Reservation Protocol (RSVP) and Stream Reservation Protocol (SRP). Even if the HaRTES switch guarantees Quality of Service in a dynamic way, it doesn’t support any protocol standard. Therefore, the need of developing a new platform that will enable the switch to perform resource reservations using standard protocols emerged. Due to the easier mapping of parameters, the SRP was chosen to be that platform. This work describes it in detail. It discusses the structural blocks crucial for its implementation, describes the protocol operation and details the messages exchanged between the different nodes in a network. The implementation of some blocks is discussed and some tests are performed allowing the validation of the work developed. The results of this work open a window opportunity for the total integration of the SRP protocol in the HaRTES switch.

Po vstupu České Republiky do EU se MSP (malé a střední podniky) musí potýkat s problémy, které přináší otevření trhů a příliv zahraniční konkurence. Pro působení podniku je podstatné také okolí v lokálním měřítku, ať již se jedná o okolí demografické, politické či sociální, které podstatným způsobem ovlivňuje veškeré procesy podnikání. Cílem diplomová práce je zhodnotit problematiku MSP s důrazem na analýzu příčin neúspěchu, která bude aplikována v podniku Mlékárna Varnsdorf s.r.o., jež se zabývá výrobou sýru eidam 30% a 40% ve dvou variantách ? klasik a bio, a spadá do kategorie MSP. Pro účely diagnostiky vnějšího i vnitřního prostředí a pro vytipování důležitých aspektů budoucího plánování bude využit zjednodušený postup tvorby SWOT analýzy. Domnívám se, že současný stav podniku není uspokojivý. V práci se pokusím zhodnotit současnou situaci mlékárny za účelem potvrzení nebo nepotvrzení stanovené hypotézy. Zamýšleným výstupem budou zjištěné příčiny dosavadní situace podniku se zaměřením na překážky okolního prostředí, které staví podnik do nepříznivé pozice na trhu, a nedostatky uvnitř podniku, které mohou být z pozice vedení korigovány. Výchozím bodem bude navržení přijatelného řešení pro podnik. Diplomová práce bude koncipována tak, aby našla praktické využití. Pevně věřím, že tato práce bude přínosem a obecným vodítkem pro vedení podniku Mlékárna Varnsdorf s.r.o.

The practical part was realized in the form of semistructured interview with a owner of Simplejohn, inc and the main goal of it was to verify the conclusions reached in the theoretical part.

There is emerging a space for new possibilities such as the energy of future -- renewable sources of energy, which are taking the fourth place in the importance of the world and it's energy sources. It is mainly biomass, water, solar or wind energy. This diploma thesis is aiming at the production of biomass -- pellets, which are a biological source of energy (wooden or vegetative material) with number of positive features and parameters. The main objective of this diploma thesis is to offer excursion into another space of modern thinking, which is well-founded by economical facts and also explains the need of taking into the consideration other alternative energetic types, which should be a common thing in future.

The life-long production of blood cells is enabled by hematopoietic stem and progenitor cells (HSPC) residing in the bone marrow. An understanding of the genes that control how HSPCs work to sustain the continued production of blood cells will enable new techniques to expand them for life-saving transplantation therapies. We have used a retroviral integration screen to search for novel genes that regulate hematopoietic stem cell (HSC) function. One of the genes found was Musashi 2 (Msi2), an RNA binding protein that can act as a translational inhibitor. A gene trap mouse model that inactivates the gene shows that Msi2 is more highly expressed in long term (LT) and short term (ST) HSCs as well as in lymphoid myeloid primed progenitors (LMPPs), but much less in intermediate progenitors and mature cells. Mice lacking Msi2 are fully viable up to more than a year but exhibit severe defects in primitive precursors. Cell cycle and gene expression analyses suggest that the main hematopoietic defect in Msi2 defective mice consists in a decreased proliferation capacity of ST-HSCs and LMPPs. Moreover, HSCs lacking Msi2 are severely impaired in competitive repopulation experiments. We further found that Msi2 is expressed during embryo development in the CD41+ cells in the Aorta-Gonada Mesonephros (AGM), most probably corresponding to the earliest emerging HSCs. Also deficient Msi2 embryos have a decrease in the number of fetal liver HSPCs. Lastly, our experiments show that Msi2 deficient HSPCs have a defect in the Wnt and PTEN/PI3K/Akt pathways that could explain the phenotype observed. Altogether, my thesis provides novel insight in to the role and mechanism of action of Musashi-2 in mouse HSPCs.
La producción de células sanguíneas a lo largo de toda la vida se lleva a cabo gracias a las células madre hematopoyéticas y células progenitoras (HSPC) que residen en la médula ósea. El estudio de los genes que controlan cómo estas HSPCs trabajan para sostener la producción continua de las células de la sangre permitirán el desarrollo de nuevos protocolos basados en la expansión ‘in vitro’ de estas células para terapias de transplante. Hemos utilizado un cribado basado en el fenómeno de integración retroviral para buscar nuevos genes que regulan la función de células madre hematopoyéticas (HSC). Uno de los genes encontrados fue Musashi 2 (Msi2), una proteína de unión a ARN que puede actuar como un inhibidor de la traducción. El modelo de ratón desarrollado mediante “gene-trap” y que inactiva el gen, muestra que Msi2 está más expresado en HSCs de largo plazo (LT-HSC) y corto plazo (ST-HSC), así como en los progenitores linfoides-mieloides (LMPP), y su expresión disminuye en progenitores intermedios y células maduras. Los ratones deficientes para Msi2 son completamente viables, pero presentan defectos importantes en los precursores primitivos que se agravan con la edad. El análisis de ciclo celular y de expresión génica sugieren que el principal defecto hematopoyético en ratones con esta deficiencia consiste en una disminución de la capacidad de proliferación de ST-HSCs y LMPPs. Además, las HSCs con déficit de Msi2 no son capaces de repoblar la médula ósea cuando se transplantan junto a médula procedente de ratones “wild-type”. También hemos observado que Msi2 se expresa durante el desarrollo del embrión en las células CD41 + de la Aorta-Gonada Mesonefros (AGM), correspondiendo muy probablemente a las HSCs emergentes. Además los embriones deficientes para Msi2 tienen una disminución en el número de HSPC en hígado fetal. Por último, nuestros experimentos muestran que un déficit de Msi2 en las HSPCs provoca un defecto en las vías Wnt y PTEN / PI3K / Akt; esto podría explicar el fenotipo observado en estos ratones. En conjunto, mi tesis proporciona una nueva perspectiva sobre el papel y el mecanismo de acción de Musashi-2 en HSPCs de ratón.

The dissertation deals with the issue of potential for innovation in small and mid-sized business environments. Innovation is becoming ever more important as it helps businesses to stay competitive in the global economy. The same applies in this era of economic crisis. Innovation is very trendy, but huge sums spent on innovation, no matter how high, do not guarantee that it will be spent effectively. As a result, innovation must be deliberate and targeted. It assumes that a company knows its overall innovation potential and is capable of enhancing it if necessary. This dissertation proceeds within the aforementioned context. It begins with a comprehensive theory and definition of essential terms. This abstract of domestic and foreign bibliography reflects historic and contemporary notions related to innovation. The theoretical part is followed by the analysis of the current state of the issue from the perspective of small and mid-sized companies in the Czech Republic. The analysis consists in a modified research by the Czech Statistical Office and in my own primary questionnaire survey. The output of the aforementioned research consists in finding of weaknesses in the innovation potential of companies and acquiring of relevant data for corroboration or refutation of postulated hypotheses, based on partial goals of this paper. The next chapter proposes a model for the calculation of the summary corporate innovation potential index, whose aim is to find weaknesses in the company without having to carry out complex comparisons through long-term research. Another logical follow-up is the proposal section, providing small and mid-sized businesses with recommendations and solution proposals to strengthen individual partial factors of innovation potential. Application of the aforementioned recommendations in sequential steps, beginning with theoretic understanding, through analysis to implementation of proposed solutions should contribute to expansion of innovation potential in specific small and mid-sized businesses.

The basic-Helix-Loop-Helix PAS (bHLH-PAS) protein family is a growing family of transcription factors. Included in this family is the Drosophila single-minded (DSim) gene, which is critical for the development of the CNS midline cells.
We have isolated a murine homologue of the Drosophila Sim gene, mSim-2. The murine and Drosophila gene products share a high degree of homology in the bHLH and PAS regions but not within the carboxy terminus. Northern blot and RT-PCR analysis of adult murine tissues revealed that the mSim-2 is expressed at the highest levels in the kidneys and at lower levels is present in skeletal muscle, lung, testis, brain, and heart. In situ hybridisation experiments demonstrate that mSim-2 is also expressed in early fetal development in the central nervous system and in cartilage primordia.
We investigated the ability of mSIM-2 to associate with the Arnt gene product, a common dimerisation partner of a number of bHLH-PAS proteins. We found that the HLH and PAS motifs of both proteins are required for optimal association. We demonstrated the presence of two separable repression domains within the carboxy terminus of mSIM-2, in contrast to dSIM, which is a transactivator. We find that mSIM-2 is also capable of repressing activation by its binding partner, ARNT. We also demonstrate that mSIM-2 can functionally interfere with another bHLH-PAS transcription factor, HIF-1alpha, by competing for ARNT binding, providing a second mechanism by which mSIM-2 may inhibit transcription.
We also investigated the effects of mSIM-2 and its close paralogue mSIM-1, as heterodimers with ARNT, on reporter constructs containing native DNA binding sites. We find that mSIM-1 can effect transcriptional activation through it's association with ARNT. In contrast, mSIM-2/ARNT does not activate, as the mSIM-2 repression domains quench ARNT transactivation. We also find that the mSIM-2 can interfere with mSIM-1 mediated transactivation by competing for dimerisation with ARNT and for DNA binding site occupation. Our results suggest that mSIM-1 and mSIM-2 have similar dimerisation and DNA binding properties but different transcriptional effects and may therefore antagonise each other which may in turn be a mechanism of gene regulation by these two proteins.

Os mecanismos e técnicas do domínio de Tempo-Real são utilizados quando existe a necessidade de um sistema, seja este um sistema embutido ou de grandes dimensões, possuir determinadas características que assegurem a qualidade de serviço do sistema. Os Sistemas de Tempo-Real definem-se assim como sistemas que possuem restrições temporais rigorosas, que necessitam de apresentar altos níveis de fiabilidade de forma a garantir em todas as instâncias o funcionamento atempado do sistema. Devido à crescente complexidade dos sistemas embutidos, empregam-se frequentemente arquiteturas distribuídas, onde cada módulo é normalmente responsável por uma única função. Nestes casos existe a necessidade de haver um meio de comunicação entre estes, de forma a poderem comunicar entre si e cumprir a funcionalidade desejadas. Devido à sua elevada capacidade e baixo custo a tecnologia Ethernet tem vindo a ser alvo de estudo, com o objetivo de a tornar num meio de comunicação com a qualidade de serviço característica dos sistemas de tempo-real. Como resposta a esta necessidade surgiu na Universidade de Aveiro, o Switch HaRTES, o qual possui a capacidade de gerir os seus recursos dinamicamente, de modo a fornecer à rede onde é aplicado garantias de Tempo-Real. No entanto, para uma arquitetura de rede ser capaz de fornecer aos seus nós garantias de qualidade serviço, é necessário que exista uma especificação do fluxo, um correto encaminhamento de tráfego, reserva de recursos, controlo de admissão e um escalonamento de pacotes. Infelizmente, o Switch HaRTES apesar de possuir todas estas características, não suporta protocolos standards. Neste documento é apresentado então o trabalho que foi desenvolvido para a integração do protocolo SRP no Switch HaRTES.
The tools and mechanisms of the Real-Time domain are used when there is a need for a system, either an embedded or a large scale system, to possess certain characteristics that ensure the quality of service. The Real-Time Systems are defined as systems that possess strict time constraints, they require high levels of reliability as a way to ensure the timely operation in every instance of the system. With the growing complexity of the embedded systems, distributed architectures are normally employed, where each module is usually responsible for a single function. In these cases there is a need for a means of communication between them, so that they can communicate with each other and fulfill the desired functionality. Due to its high capacity and low-cost, Ethernet technology has been the target of study, aiming to make a means of communicating with the service quality feature of real-time systems. In response to this need, arose at the University of Aveiro the Switch HaRTES, which has the ability to manage their resources dynamically in order to provide the network where it is applied Real-time guarantees. However, for a network architecture to be able to provide such guarantees, it must be able to specify the flow of data, do a correct traffic routing, reserve resources, and do an admission control and packet scheduling. Unfortunately, the HaRTES Switch, despite having all these features, does not support a standard protocol. In this document is then presented the work that was developed for the integration of the SRP protocol in the HaRTES Switch.

In a near future several of the European countries will begin working after new guidelines concerning a declaration called the Bologna process. Everything will be done one step at a time and the declaration will change the way the students are being graded, all this for the sake of making the European labour-market more

opened for all of its millions of students.

In Växjö this process is in the making, but to adapt to the future demands the school must also change its ways of teaching.

To increase its quality the University has put together a procedure of actions to improve the pedagogical working method in all its institutions. In this essay we would like to elucidate how the work with the Bologna process is proceeding within the institution of Mathematics and System Engineering and what

they do improve their pedagogical teaching. By doing a qualitative case study on the student at the Computer Science Program we wish to find the answers to our questions.


Det pedagogiska arbetet inom MSI – Utifrån Bolognaprocessen och UPC.

Inom en snar framtid kommer flertalet länder i Europa jobba efter nya riktlinjer enligt en deklaration kallad Bolognaprocessen. Det hela kommer att ske i etapper och innebär att sättet på vilket studenter bedöms kommer ändras, allt för att göra Europas arbetsmarknad öppnare för alla dess miljoner studerande.

I Växjö pågår detta arbete för fullt, men för att anpassa sig till framtida krav måste skolan även förändra sin utbildningsform. För att höja sin kvalité har universitetet tagit fram en handlingsplan för att förbättra de pedagogiska arbetsmetoderna i alla institutioner. I den här uppsatsen vill vi belysa hur arbetet med Bolognaprocessen fortgår inom Matematiska och systemtekniska institutionen och vad de gör för att förbättra den

pedagogiska undervisningen. Genom att göra en kvalitativ undersökning med Systemvetenskapliga programmets studenter försöker vi finna svaren på våra frågor.

For this purpose, microspectrophotometry is a technique to be considered, because it can be used to obtain visible absorbance spectra allowing discrimination of macroscopically indistinguishable fibres. Although MSP (microspectrophotometer) records colour objectively (unlike humans), the spectra it produces (although objective) are being compared subjectively, therefore another system of comparison, based on objective criteria, must be determined. This research focuses on colour discrimination and considers several systems of comparison that can be used in order to determine some objective criteria of matching spectra. For this purpose, two approaches are envisaged: a colorimetric and a mathematical approach, and more than 230 fibres were studied by microspectrophotometry in order to determine some criteria of matching and non-matching. Firstly, the influence of some parameters was studied in order to establish a correct method of analysis. This study shows that the most important parameter to have good repeatability and good reproducibility is the light intensity because when the level of light intensity is raised, the ratio signal/noise follows the same pattern. experiments made, some criteria for matching and non-matching textile fibres were determined. A first discrimination can be made by the visible colour and the direct comparison of spectra, however this discrimination must be realised cautiously because they are not objective criteria. The calculation of the HQI values (Hit Quality Indices) then provides an objective criterion for the discrimination.

The aim of this study is to compare implementation, impact and sustainability of educational projects in small and medium enterprises which are funded by the European Social Fund. The results of the analysis should give us an answer to the question whether the financial support is used effectively and contributes to the fulfilment of its objectives or the potential of these funds remains untapped. The theoretical part deals with the current situation analysis of training and development of employees in SMEs, different methods of measuring the effectiveness of company training as well as the opportunities of training promotion in SMEs with support of the ESF. The practical part is focused on comparing the implementation of educational projects funded by the Operational Programme Prague -- Adaptability and Operational Programme Human Resources and Employment in chosen SMEs. These companies are compared with enterprises that fund their educational programmes with their own resources and with respect to differences resulting from their size measured by number of employees. This comparison enables us to assess the effectiveness of the use of public funds in this area.

This master thesis deals with the analysis of the possibilities of Cloud Computing for information system solutions in small and medium businesses. The main objective is to analyze these options, identify key areas for the transition to the cloud and propose an approach to develop Information system in this direction. The main contribution of this work is the practical experience which is involved in a view of the problems being solved and an emphasis on aspects that have been verified in practices. The main text of the work is divided into five chapters. The first chapter defines the basic concepts and limitations on which further work is based. The second chapter deals with the current form of IS in SMEs, its description and identifying problems. The third chapter analyzes the Cloud Computing from the perspective of the suitability of various models for SMEs, identifies the major trends that affect the Cloud Computing and describes some basic elements on which Cloud Computing is based. It also analyzes the issue of security. The fourth chapter deals with the state of Cloud Computing market and identification of key problem areas which need to be solved in various stages of market development. The last chapter uses several case studies to show a meaningfulness and readiness of Cloud Computing for running information system. In the end the recommended approach to the adoption of cloud services is defined.

A novel process for single-pot conversion of biomass to biofuels was developed called the molten salt catalytic pyrolysis (MSCP) method. The proposed single-pot MSCP process proved to be an inherently more efficient and cost-effective methodology for converting lignocellulosic biomass. In this study, several parameters that affect yield of bio-oil were investigated including carrier gas flow rate; pyrolysis temperature; feed particle size; varying types of molten salt and catalysts. Use of molten salt as the reaction medium offered higher liquid yield and experiments containing ZnCl2 showed higher yield than other chloride salts. The highest yield of bio-oil was up to 66% obtained in a ZnCl2-KCl-LiCl ternary molten salt system compared with 32.2% at the same condition without molten salts. In addition, the effect of molten salt on the composition of bio-oil was also studied. It was observed that molten salt narrowed the product distribution of bio-oil with furfural and acetic acid as the only two main components in the liquid with the exception of water. Finally, a thermogravimetric kinetic study on the pyrolysis of biomass in MSCP was conducted.

The Malaysian Skills Training Programme (MSTP) is a sub-division of Malaysian Vocational and Training Education (VET) under the jurisdiction of the Ministry of Human Resource, Malaysia. The programme was developed to prepare skilled workers for employment in the ‘blue collar’ sector. The Malaysian Ministry of Higher Education and the UNESCO-UNEVOC believe that the VET should be upgraded to offer a more professional service in which ICT (Information and Communication Technology) applications such as electronic learning need to be explored and harnessed (Kafka & Dale, 2013). This thesis investigates the implementation of an electronic portfolio (E-portfolio) in one accredited Skills Training Institution in Malaysia. The research aim was to deploy an E-portfolio which was developed using MAHARA software with several skills training courses in one training semester. The idea of a ‘threshold concept’ was adopted to provide guidelines to direct the E-portfolio development and implementation. Kolb’s Experiential Learning (KEL) and Competency Based Training (CBT) were the main reference theories informing the design of the E-portfolio process of use. Based on quasi-experimental design, the system’s implementation included groups of students in 2013 and 2014 cohorts. Upon completion, the use of the E-portfolio was evaluated to determine the system’s impact and its effectiveness in the teaching and learning process. A mixed methods research approach was used, consisting of assessment rubrics, survey questionnaires, e-mail interviews, online communication and document reviews. A respondent’s validation based approach called a ‘report-and-respond enquiry’ was executed at the end of data analysis to validate the findings. Qualitative and quantitative data showed that the E-portfolio could contribute to the improvement of several aspects of teaching and training such as communication, reflection on practical work, motivation of learning, progress based learning, and the construction of new knowledge. In addition, senior staff believed that the use of an E-portfolio would benefit the institution by saving printing costs and enhancing the institution’s image to the public and to the awarding body. The findings suggested that, when blended with the current traditional training pedagogy, the E-portfolio system has the potential to be implemented as a showcase E-portfolio, used for online assessment or can be used as a teaching approach. The email interviews and Facebook communication revealed that user attitudes and the support required for the use of the E-portfolio are the biggest challenges. Based on these findings, a scaffolded approach to the implementation of E-portfolios in MSTP is recommended. The key outcomes of this research are the E-portfolio scaffolding strategies in the MSTP that could be deployed at other Skills Training institutions or organizations which intend to explore the use of E-portfolios in the future.

Within these studies the properties of a high repetition rate (≤ 25,000 Hz) Nd:YVO4 laser were investigated with a view to improving the throughput and quality of information obtained from MALDI MS data collected in continuous raster sampling mode. Initially, the nature of the laser beam profile was investigated and a novel fluorometric method developed for imaging these profiles. Analysis of thin film samples of α-cyano-4-hydroxycinnamic acid (CHCA) and lipid standard phophatidylcholine (PC) 34:1 were carried out. Under most conditions a lower repetition rate and slow stage raster speed were found to be optimal. However, in tissue based investigations repetition rates of 5-10 kHz and faster raster speeds were found to increase the detected ion intensity. Subsequent to this, the use of para-nitroaniline (PNA) as an effective matrix for high-repetition rate laser MALDI MSI applications was investigated and compared to CHCA. PNA was found to provide high quality MSI data, comparable to or better than that obtained with CHCA. The utility of CHCA and PNA were then investigated for the repeat analysis of single tissue sections with a view to increasing the amount of information obtained. Up to five analyses of the same tissue section were demonstrated. Repeat analysis was then applied to the acquisition of both lipid and protein data from a single tissue section using multiple matrices and tissue washing.

Microsatellites are short repetitive DNA sequences, which are liable to replication errors. Microsatellite instability (MSI) is controlled by the mismatch repair system, and accumulation of microsatellite mutations is used as a diagnostic criterion for tumours where this repair system is compromised, such as those which develop in Lynch Syndrome (HNPCC) patients. Currently, the Amsterdam II screening criteria and revised Bethesda Guidelines are used to identify tumours for MSI testing using both immunohistochemistry and fragment analysis tests. However, because Lynch Syndrome patients are being missed, testing for all colorectal and endometrial cancers is now being recommended. Faster and cheaper MSI testing methods are therefore desirable. Although PCR and sequencing error compromise sequence based typing of the repeats currently used for diagnosis, some short mononucleotide repeats have been identified which show low level instability, suggesting that sequence typing of short repeats may be possible. Here, I investigate the utility of high throughput sequencing (HTS) as the basis for MSI testing. As an initial assessment of the method, I used the MiSeq platform to type 22 previously published short mononucleotide repeats in 4 microsatellite unstable (MSI-H) tumours, and showed that MSI could be detected above background noise in 7-12bp repeats. To identify the most variable short repeat markers for MSI testing, I then analysed MSI in whole genome sequence data from The Cancer Genome Atlas network, and identified a panel of 120 7-12bp informative mononucleotide repeats which were subsequently evaluated on a panel of 5 MSI-H tumours and controls. The most informative 20 markers were further tested on a panel of 58 colorectal tumours to define thresholds for instability calling. Using a panel of eighteen 8-12bp mononucleotides it was possible to distinguish between MSI-H and microsatellite stable (MSS) tumours with a sensitivity and specificity of 100%. Flanking SNPs were also evaluated and identified an excess of allelic bias among MSI-H tumours compared to MSS tumours, a feature that could be integrated into the MSI test. Finally, short mononucleotide repeats with flanking SNPs were assessed for their potential to identify clonal variation in MSI-H tumours. Using a multiple biopsy approach evidence of different sub-clones was found in three MSI-H tumours, suggesting that these markers could be used for analysis of clonal variation and evolution.

Pesticides are widely used in agriculture to control weeds, pests and diseases. Successful control is dependent on the compound reaching the target site within the organism after spray or soil application. This is influenced by a range of physiological and environmental factors. One such factor is uptake into foliar tissue and roots and subsequent movement through the plant. Uptake and translocation (and metabolism) can affect the speed and persistence of action, crop selectivity and weed spectrum, thus understanding these issues is of great importance. Conventional methods for determining uptake and movement of herbicides and pesticides include autoradiography, liquid scintillation and chromatographic techniques such as HPLC. Autoradiography using radiolabelled compounds provides the best indication of a compound's movement within the plant system which is an established technique but relies on radiochemical synthesis. The cost and time taken means that only a relatively few agrochemicals can be studied in this way. MALDI is a highly adaptable soft ionisation technique that was established in the late 1980's through the developmental research of Tanaka and Hillenkamp. The versatility of MALDI-MS has been extended in recent years with the advent of protein profiling and imaging directly from the surface of thin biological tissue sections. Research by Caprioli et al has resulted in the successful profiling and imaging of proteins from thin sections of healthy mouse brain and mouse brain containing tumor and the profiling of proteins from a neurotoxin induced rat modelof Parkinson's disease. Recently the imaging technique has been further developed by research groups to include the detection and imaging of small organic molecules on the surface of porcine epidermal tissue and the surface of rodent brain tissue. In preliminary studies undertaken to support the grant application for this project the detection and imaging of compounds on the surface of a plant leaf or inside a plant stem using MALDI-MS was demonstrated. In this project the technique of imaging matrix assisted laser desorption ionisation mass spectrometry has been used to examine the uptake and distribution of a range of compounds of agrochemical interest into growing plants. The agrochemicals of interest are azoxystrobin, mesotrione, fluazifop-p butyl and nicosulfuron. Azoxystrobin is a broad spectrum fungicide which inhibits mitochondrial respiration in fungi; the compound is absorbed through the roots and translocated in the xylem. Mesotrione is a selective phloem mobile herbicide which inhibits HPPD, an enzyme involved in carotene synthesis and results in bleaching of the leaves. Fluazifop-p butyl is absorbed as its ester, which is quickly hydrolysised to its carboxylic acid (fluazifop acid) that is mobile in the xylem and the phloem. Fluazifop acid works as a fatty acid synthesis inhibitor, by inhibition of acetyl CoA carboxylase (ACCase). Nicosulfuron is both xylem- and phloem-mobile which inhibits branched chain amino acid synthesis by means of the acetolactate synthase (ALS) enzyme. MALDI-MS images of mesotrione in tissue underlying the waxy cuticle layer following removal using a tape stripping method have been generated. MALDI-MS images have been generated from azoxystrobin following uptake via the roots in soya and sunflower plants. Nicosulfuron translocation following uptake via the0roots and foliar application has been determined in sunflowers and a phase I metabolite has been identified 48 hours following uptake via the roots and 24 hours following foliar application using MALDI-MSI. Nicosulfuron and azoxystrobin translocation has been determined in the same plant following application of the compounds to the plant stem. The methods developed were used to determine the extent of translocation of four sulfonylurea herbicides 24 hours following foliar application. Additionally, a preliminary method was assessed to determine whether MALDI-MSI can be used to assess translocation of agrochemicals.

Leishmania chagasi is the causative agent of visceral leishmaniasis in South America. The most abundant glycoprotein on the surface of L. chagasi promastigotes is the glycosylphosphatidylinositol (GPI) anchored protease MSP (major surface protease), also called GP63. MSP is encoded by more than 18 tandem MSP genes on a single chromosome. MSP genes are classified according to unique sequences at their 3' ends and distinct expression patterns. The five MSPS genes (MSPS1, MSPS2, etc.) express 3.0 kb RNAs in stationary phase of promastigote growth in vitro in culture. The > twelve MSPL genes express 2.7 kb RNAs in logarithmic phase of promastigote growth, and the single MSPC RNA is constitutively expressed as two RNA species (2.6 and 3.1 kb) throughout promastigote growth. The progression from logarithmic to stationary phase is accompanied by an increase in parasite virulence for a mammalian host, and a 16-fold increase in the total MSP protein associated with the cell. As such, MSP has been called a virulence factor of leishmania. Little is known about the differences between isoforms of MSP proteins encoded by the three MSP gene classes, because they have a very similar amino acid sequences. The purpose of this thesis was to study the protein expression and localization of MSPS, MSPL, and MSPC in the promastigote and amastigote stages of the L. chagasi. We took three approaches to this problem. First, we produced constructs in which the fluorescent marker GFP was flanked by putative targeting sequences of the MSPs. Second, we generated Leishmania transfectants expressing Myc-tagged full-length MSPs and studied their localization in promastigote cells. Third, we generated antibodies to immunogenic peptides in the few regions with unique sequences that allowed us to distinguish between some of the MSP classes. One monoclonal anti-peptide antibody, named C51, recognized only MSPS1 and MSPL1. Data indicated that the product of the MSPC gene runs at a higher molecular size than products of the MSPL and MSPS genes, both of which localize to the promastigote surface. Overall the data set the stage for future studies of the properties and functions of specific MSP gene products.
