ABSTRACT The absence of endogenous mouse mammary tumor viruses (MMTVs) in the congenic mouse strain, BALB/Mtv-null, restricts the early steps of exogenous C3H MMTV infection, preventing the superantigen (Sag) response and mammary tumorigenesis. Here we demonstrate that BALB/Mtv-null mice also resist tumor induction by FM MMTV, which encodes a stronger Sag compared to C3H MMTV. In contrast to infections with C3H MMTV, Mtv-null mice show FM-MMTV Sag-specific responses comparable to those observed in susceptible BALB/c mice. Neither virus shows significant replication in the spleen or mammary gland. Thus, Mtv-null mice restrict MMTV replication and mammary tumorigenesis even after a robust Sag response.

Abstract DDD/1 (DDD) mice contrast strikingly with DDD-mtv-2/mtv-2 (DDD-mtv-2) congenics in their marked lymph node (LN) T cell paucity. To clarify the possible difference in LN function between them, reciprocal LN grafting experiments were conducted. DDD-mtv-2 LN grafts in DDD recipients underwent hyperplasia as dramatic as 10-to 20-fold increase in weight between 3 and 4 wk after implantation. Lymphoid cells in hyperplastic LN grafts were of recipient origin. Similar hyperplasia of mtv-2-heterozygous LN grafts also occurred on various hybrid backgrounds involving DDD mice. Moreover, LN grafts from BALB/c mice infected with mtv-2-derived exogenous mouse mammary tumor virus (MMTV) swelled in MMTV-free BALB/c recipients. Genetic analysis of DDD x (DDD x DDD-mtv-2)F1 backcross progeny demonstrated that LN hyperplasia was closely linked to mtv-2. The frequencies of V beta 5+ and V beta 8+ T cells unresponsive to mtv-2-encoded superantigen (SAg) changed with practically the same kinetics in both LN grafts and recipients' LN. Thus, the cells responsible for LN hyperplasia were polyclonal. V beta 14+ CD4+ cells responsive to mtv-2 SAg were specifically stimulated in recipients' LN but selectively deleted in hyperplastic LN grafts. DDD mice carrying hyperplastic mtv-2+ LN grafts or pretreated with mtv-2+ spleen cells developed an unresponsive state in terms of influx of mtv-2- lymphoid cells into mtv-2+ LN grafts. These results indicate that mtv-2 gene products including SAg may stimulate mtv-2- lymphoid cells of recipients and cause them to migrate into mtv-2+ LN grafts in a nonspecific manner with resulting LN hyperplasia.

Mls-1 is an endogenous superantigen that leads to in vivo deletion and in vitro stimulation of T cell receptor (TCR) V beta 6-, 7-, 8.1-, and 9-expressing cells. The MA/MyJ mouse deletes the identical set of TCR from its mature T cell repertoire; however, it does not contain Mtv-7, the murine mammary tumor provirus (MMTV), whose sag gene encodes Mls-1. Interestingly, the superantigen activity of this mouse strain segregates with a new mammary tumor provirus, Mtv-43, not seen in other inbred strains. The predicted amino acid sequence of the sag gene of Mtv-43 was compared with that of Mtv-7. Strikingly, the COOH terminus of the two molecules is very similar, while all other MMTV-encoded superantigens differ 100% in this segment.

Murine T cell reactivity with products of the minor lymphocyte stimulatory (Mls) locus correlates with the expression of particular variable (V) domains of the T cell receptor (TCR) beta chain. It was recently demonstrated that Mls antigens are encoded by an open reading frame (ORF) in the 3' long terminal repeat of either endogenous or exogenous mouse mammary tumor virus (MMTV). Immature thymocytes expressing reactive TCR-V beta domains are clonally deleted upon exposure to endogenous Mtv's. Mature T cells proliferate vigorously in response to Mls-1a (Mtv-7) in vivo, but induction of specific anergy and deletion after exposure to Mtv-7-expressing cells in the periphery has also been described. We show here that B cells and CD8+ (but not CD4+) T cells from Mtv-7+ mice efficiently induce peripheral deletion of reactive T cells upon transfer to Mtv-7- recipients, whereas only B cells stimulate specific T cell proliferation in vivo. In contrast to endogenous Mtv-7, transfer of B, CD4+, or CD8+ lymphocyte subsets from mice maternally infected with MMTV(SW), an infectious homologue of Mtv-7, results in specific T cell deletion in the absence of a detectable proliferative response. Finally, we show by secondary transfers of infected cells that exogenous MMTV(SW) is transmitted multidirectionally between lymphocyte subsets and ultimately to the mammary gland. Collectively our data demonstrate heterogeneity in the expression and/or presentation of endogenous and exogenous MMTV ORF by lymphocyte subsets and emphasize the low threshold required for induction of peripheral T cell deletion by these gene products.

ABSTRACT Mouse mammary tumor virus (MMTV) is a retrovirus encoding a superantigen that is recognized in association with major histocompatibility complex class II by the variable region of the beta chain (Vβ) of the T-cell receptor. The C-terminal 30 to 40 amino acids of the superantigen of different MMTVs display high sequence variability that correlates with the recognition of particular T-cell receptor Vβ chains. Interestingly, MMTV(SIM) andmtv-8 superantigens are highly homologous but have nonoverlapping T-cell receptor Vβ specificities. To determine the importance of these few differences for specific Vβ interaction, we studied superantigen responses in mice to chimeric and mutant MMTV(SIM) and mtv-8 superantigens expressed by recombinant vaccinia viruses. We show that only a few changes (two to six residues) within the C terminus are necessary to modify superantigen recognition by specific Vβs. Thus, the introduction of the MMTV(SIM) residues 314-315 into themtv-8 superantigen greatly decreased its Vβ12 reactivity without gain of MMTV(SIM)-specific function. The introduction of MMTV(SIM)-specific residues 289 to 295, however, induced a recognition pattern that was a mixture of MMTV(SIM)- and mtv-8-specific Vβ reactivities: both weak MMTV(SIM)-specific Vβ4 and fullmtv-8-specific Vβ11 recognition were observed while Vβ12 interaction was lost. The combination of the two MMTV(SIM)-specific regions in the mtv-8superantigen established normal MMTV(SIM)-specific Vβ4 reactivity and completely abolished mtv-8-specific Vβ5, -11, and -12 interactions. These new functional superantigens with mixed Vβ recognition patterns allowed us to precisely delineate sites relevant for molecular interactions between the SIM or mtv-8 superantigen and the T-cell receptor Vβ domain within the 30 C-terminal residues of the viral superantigen.

Abstract The genetic linkage of loci encoding stimulatory Mlsa and Mlsc determinants with proviruses of mouse mammary tumour viruses (MMTV) has been shown. We previously have reported that the ligand(s) for V beta 5, V beta 11, and V beta 12 behaves as a novel minor lymphocyte-stimulating (Mls) determinant(s), Mlsf, to induce the strong proliferation of unprimed T cells, and that this ligand(s) also functions as a self-Ag for the clonal deletion of self-reactive T cells. In the accompanying paper (Part I), a unique polymorphism characteristic of the Mlsf gene product is presented. In order to determine the genetic basis for this novel Mls system, we examined the progeny of multiple genetic crosses to identify the MMTV proviral loci involved in the clonal deletion of self-Mlsf-reactive T cells. Results from these investigations indicated that at least three known MMTV proviruses, Mtv-8, Mtv-9, and Mtv-11 are involved in the expression of Mlsf gene products. Presence of Mtv-9 results in the complete deletion of V beta 5, V beta 11, and V beta 12; Mtv-8 is associated with the complete deletion of V beta 12, but only a partial deletion of V beta 11 (primarily CD4-positive T cell subset) with little or no deletion of V beta 5; and Mtv-11 induces the complete deletion of V beta 11 and V beta 12, but no deletion of V beta 5. Given the significant sequence homology in the C-terminal portion of the open reading frame (ORF) region among these three MMTV and the almost equivalent effect of these three MMTV provirus upon the V beta 12 repertoire, their apparent hierarchic effect upon the V beta 5 and V beta 11 repertoires suggests that affinity differences in recognition of the same determinant by different TCR V beta may play a significant role in the clonal deletion of self-reactive T cells.

The demonstrated ability of MMTV superantigen proteins to react with TCRs has led to the postulate that other infectious retroviruses may use superantigen-like molecules to modify the host's immune system. In this report, successful retrovirus-mediated Mtv-7 sag gene transfer into pluripotent hematopoietic stem cells is described. In two different strains of Mls-1- host mice (CBA/Ca and BALB/c) reconstituted with Mtv-7 sag gene expressing bone marrow cells, low levels of ectopic Mtv-7 sag gene expression on syngeneic donor hematopoietic stem cell-derived population alone can induce partial clonal deletion of Mls-1 reactive V beta 6+ and V beta 8.1+ T cells, and complete clonal inactivation of V beta 8.1+ T cells.

Channel identity can create picture, image, character and identity of a television station. MTV has made the best use of this form of corporate promotion program. Through their channel identity, MTV created images and pictures in accordance with their basic concepts. This research analyzed the MTV (Music Television) channel identity and logo. The analysis procedure carried on the MTV logos including MTV Indonesia, MTV China, MTV Russia, MTV United Kingdom, MTV Japan, and MTV India. This study used a multidisciplinary approach namely the historical, aesthetics, and semiotics approach. The method used in this research was qualitative method, with analytic descriptive analysis. The data was collected through literature study and observation. The data then grouped according to the basic concepts of MTV local culture exploration, and studied by looking at the process of visual idiom design concepts, shapes, characters, colors, and work techniques. The result of this research is interpretation of the visual sign and meaning of the MTV logo as a visual communication media and mass communication which contained certain value and meaning to persuate their audiens.</p><p> </p><p><br /><strong>Abstrak</strong></p><p>Analisis Tanda Visual Channel Identity MTV (Music Television) Indonesia. Channel identity stasiun televisi dapat membentuk citra, image, karakter dan menjadi identitas sebuah stasiun televisi. MTV telah memanfaatkan sebaik mungkin bentuk program promosi korporat ini. Melalui channel identity, MTV membentuk image dan citra sesuai dengan konsep dasar yang dimiliki. Penelitian ini menganalisis channel identity dan logo stasiun televisi MTV (Music Television). Analisis makna dilakukan pada logo MTV beberapa negara yaitu MTV Indonesia, MTV China, MTV Rusia, MTV United Kingdom, MTV Jepang, dan MTV India. Kajian ini menggunakan pendekatan multidisplin, yakni pendekatan historis, estetika, dan semiotika. Metode penelitian yang digunakan adalah metode kualitatif, dengan analisis deskriptif analitik. Data dikumpulkan melalui studi pustaka dan observasi. Data dikelompokkan sesuai dengan konsep dasar eksplorasi budaya lokal MTV, yang akan dikaji dengan melihat pada proses konsep perancangan idiom visual, bentuk, karakter, warna, dan teknik pengerjaan. Superantigens (SAGs) represent a new class of antigens, characterized as T cell receptor (TCR) V beta-reactive elements. Bacterial toxins constitute the major group of exogenous SAGs, while the mouse mammary tumor virus (MMTV)-encoded Mls molecules represent the endogenous SAGs. Mls-1 is the prototype of the latter SAGs, because it elicits a very potent T cell stimulatory response in vitro in unprimed T cells expressing the TCR V beta 6 or 8.1 chains. In vivo, Mls-1 causes deletion of immature T cells bearing the V beta 6, 7, 8.1, or 9 chains. Although Mls-1 was functionally discovered &gt; 20 yr ago, it has not been possible to raise antibodies against this molecule. We have previously cloned and sequenced the Mtv-7 sag gene, which encodes Mls-1. Sequence comparisons with other MMTV sag genes suggested that the polymorphic 3' end encodes the TCR V beta specificity of these SAGs. We have, therefore, immunized hamsters with a 14-amino acid peptide from the deduced COOH-terminal sequence of the Mtv-7 sag gene. We describe here the production of a monoclonal antibody (mAb), 3B12, which is peptide specific and reacts with a recombinant baculovirus product of Mtv-7 sag. This mAb blocks Mls-1-specific T cell recognition and detects the Mls-1 protein on the surface of the B cell hybridoma LBB.A, but not on LBB.11, which is an Mtv-7 loss variant of LBB.A. Transfection of the Mtv-7 sag gene into LBB.11 renders this cell functionally Mls-1+ as well as positive for 3B12 binding, confirming the specificity of this mAb. It is well documented that B cells and CD8+ T cells express T cell stimulatory Mls-1 determinants, and we show here that this functional profile correlates with the expression of MMTV-specific mRNA. However, primary lymphocytes derived from Mls-1+ mice do not stain with 3B12, even after in vitro activation with mitogens or phorbol ester.

The Brazilian MTV is on air for more than 15 years in Brazil and, since the beginning of its transmissions, in 1990, it has considerably evolved the variety of the TV shows it presents day by day, leaving behind the simple broadcast of music videos. However, this variety of the MTV shows is characterized by a discursive unit which differentiates it from other open tv channels. While the other TV channels, especially non cable TV channels, have developed a set of TV shows directed to all sorts of spectators, the MTV channel was able to develop a huge set of TV shows entirely directed to the young spectator. The intent of this work is to perform a qualitative reading of the language of the simbolic material produced by MTV for a specific public, subsidized by the critical speech analysis, the semiotic and the theoretical studies of Social Communication. The analysis corpus is comprised by the shows and vignettes from the week of January 23rd 29th, 2006. Duplicated presentations of the same TV show were not considered. It was demonstrated that it is possible to identify a discursive structure that compounds a language, a speech, which characterizes the MTV channel and differentiates it from the other non cable and cable TV channels. A language that combines elements from the universe of its main spectator the youth. Key words: communication, MTV, speech analysis, semiotic, youth.
A MTV brasileira está no ar há 15 anos no Brasil e desde o início de suas transmissões, em 1990, evoluiu consideravelmente a variedade de programas que oferece diariamente em sua grade de programação, saindo da simples reprodução de videoclipes. Essa variedade de programas, no entanto, é caracterizada por uma unidade discursiva que diferencia a MTV das outras emissoras de televisão aberta. Enquanto outras emissoras trabalham com gêneros diferentes de programas, em especial as que compõem a grade de programação de televisão aberta, a MTV consegue criar uma grade de programação diversificada, sempre direcionada a um público específico: o universo jovem. Objetivou-se realizar uma leitura qualitativa da linguagem do material simbólico que esta emissora produz a uma faixa de público especializada, subsidiada pela análise crítica de discurso, a semiótica e os estudos teóricos da Comunicação Social. Compõem o corpus de análise, os programas e vinhetas inéditos da semana de 23 a 29 de janeiro de 2006. Não foram consideradas as reprises. Demonstrou-se que é possível identificar uma estrutura discursiva que compõe uma linguagem, um discurso, que caracterizam a MTV e a diferenciam de outros canais de televisão aberta e a cabo. Uma linguagem que combina elementos do universo de sua principal audiência, os jovens. Palavras-chave: comunicação, MTV, análise de discurso, semiótica, jovem.

Essa dissertação estuda as artbreaks, pequenas vinhetas produzidas e exibidas pela emissora MTV (Music Television), resgatando o seu contexto histórico e abordando os seus aspectos técnicos. Para tanto busca-se explicar as etapas de produção os idealizadores e as principais referências desse tipo de formato televisivo. Nesse sentido, o primeiro capítulo apresenta um histórico da emissora, desde o surgimento no cenário norte-americano até a sua chegada e consequente popularização no Brasil. A seguir, o segundo capítulo discorre sobre o gênero videoclipe e sua importância para a televisão, apresentando os formatos dele decorrentes, as vinhetas e as artbreaks. Desenvolve-se, então, a conceitualização da artbreak dentro desse universo, mostrando as principais diferenças entre cada um desses formatos, focalizando no referencial pós-moderno, no resgate de certas estruturas vanguardistas dos anos 20 e no processo de serialidade audiovisual. Politics has become apparently distant reality for Brazilian youth currently and the importance of mass communication in the democratic process that has become even more evident, has led us to question not only the means by which young people approach politics but also the role of the media within this process. Thus, the objective of this work is to analyze how the MTV Magazine ( Revista MTV), a means of mass communication aimed at a young public, approaches politics in its reports and how it seeks to promote knowledge of specific current issues which may be used as an indicator that the relationship between youth and politics definitely does not happen inside the parties and labor unions themselves but in more fragmented specific movements, for identity build-up, for individual actions. Using a methodology that permitted the selection and identification of the theme, eight reports and two columns which addressed the above topics were selected from the fifteen issues of the MTV Magazine that were analyzed. O aparente distanciamento da atual juventude brasileira em relação à política e a cada vez mais evidente importância dos meios de comunicação de massas no processo democrático, nos levou a questionar não apenas as novas formas que essa juventude estaria experimentando para fazer política como também qual o papel da mídia neste processo. Assim, este trabalho tem como objetivo analisar como a Revista MTV, um meio de comunicação de massas direcionado ao público jovem, aborda a política em suas matérias e como procura aproximar seu leitor de determinadas questões que estão colocadas hoje e que podem servir como indícios de que, definitivamente, a relação da juventude com a política já não se dá mais nas esferas institucionais dos partidos e sindicatos, e sim em movimentos mais fragmentados, de afirmação de identidades, de lutas individuais. Foram selecionadas nas 15 edições da Revista MTV que analisamos, a partir de uma metodologia que nos ajudou a recortar e identificar nosso tema de interesse, 8 matérias e 2 colunas que abordavam essas questões. Através delas foi possível perceber o posicionamento da publicação em relação à política e sua preocupação em fazer com que o jovem perceba sua responsabilidade em relação a determinados assuntos e, a partir disso, as possibilidades de ação e atitude que pode vir a ter

A dissertação trata do processo que a produção cultural, no caso específico a música, passa para tornar-se pública, dentro de um contexto global no início do século XXI. Isso corresponde às configurações de um mercado oligopolista, organizado através de fusões e concentrações. É resgatada a relação que a música estabelece com a televisão brasileira, para finalmente colocá-la dentro da história e do desenvolvimento da MTV Brasil. Também apresenta as estratégias de comercialização do produto musical numa análise da programação dessa emissora. Ao mesmo tempo, contextualiza a cena musical brasileira, do industrial ao alternativo, dentro do desenvolvimento tecnológico e da expansão do capital. Realiza-se, então, um estudo de caso, no âmbito da Economia Política da Comunicação. Para isso, além da análise do conteúdo musical da MTV Brasil e revisão bibliográfica estruturante, procede-se a entrevistas, numa análise (dentre outros possíveis) da cena musical nacional. Esta pesquisa, enfim, é um canal que se abre àqueles que desejam conhecer o intrincado e complexo mundo que permeia os meio de comunicação e suas interfaces, o cenário em que ocorre e como chega ao topo do mercado alguns artistas, canções, CDs, DVDs e outros produtos.
This study is about the process that cultural production goes through in order to turn itself public within a global context in the beginning of the XXI century. It matches the configurations of an oligopolist market that keeps its organization three merger and concentrations. It rascues the link the music will bring up with the brazilian television to put it, at last, within history and development of MTV Brazil. Also it presents the strategics points of commercing musical products analyzing the MTV programming. At the same time, it contextualizes the brazilian musical scenary from the industrial to alternative way in the technological development. Achieving then a study of case to the Political Economy Communication. This work here goes further of an analysis of the musical MTV Brazil contents, and a structured bibliographyc revision proceeding interviews into an analysis (within possible others) of he national music scenary. The research, finishing, means an openned channel to those who wish to discover the complexed and implicated intricate world which permeates the mass media and its interfaces, the scenary where that happens and how some famous names, songs, CDs, DVDs and many other products reach the top of the market.

Lançada em 1990, a MTV Brasil representa o primeiro empreendimento de um grande grupo internacional de mídia no País - a americana Viacom Inc, a terceira maior empresa do mundo – a operar com um conceito similar às franquias de fast food. Inicialmente licenciada para o Grupo Abril, a marca introduz o conceito de canal segmentado de televisão no Brasil, apesar de ser anterior às transmissões a cabo. Em 1996, a holding adota uma estratégia de expansão mundial mais agressiva. É criada a MTV Brasil Ltda, uma parceria da Abril com a Viacom. Em 1999, a Rede MTV adota a filosofia Thinks Globally, Acts Locally, mais adequada ao seu projeto de expansão global. Em contrapartida, a afiliada brasileira, em sua luta para adequar-se ao mercado local, reformula sua política de programação e passa a privilegiar em sua grade os não-musicais. O formato da MTV Networks e sua forma de expansão fazem da rede um emblema do processo de aculturação imposto pela globalização, equilibrando modos de produção dos países em desenvolvimento com a alta tecnologia do "Mundo Rápido". A questão que emerge é a forma como o mercado brasileiro incorpora essa nova tecnologia, a partir da trajetória da MTV Brasil, e entender até que ponto a entrada de marcas estrangeiras no segmento pode contribuir para o desenvolvimento da mídia nacional.
Launched in 1990, MTV Brazil represents the first enterprise in our country of a big international media company – the American Viacom Inc, the third largest in the world – which operates similar to the fast food chains. The brand, initially just a license to Abril Group, introduces the segmentation media concept on TV channels, even before the introduction of cable TV in Brazil. In 1996, Viacom adopted aggressive global expansion plans. MTV Brazil Ltda is created in a partnership with Abril. After 1999, MTV Networks adopted the slogan Thinks Globally, Acts Locally appropriated to the new expansion strategy. On a counterpart, the Brazilian MTV franchisee reformulated the programmee policy, focusing the non-musical shows in the schedule, in an attempt to increase regional audience. MTV format and the way the network develops, became the brand MTV in one of the greatest symbols of the enculturation imposed by globalization, in the search for a balance between the primary productive manners in the renowned Third World, with the high technology of the "Speed World". The question that emerges is how Brazilian market embodies this new technology taking MTV's Brazil trajectory and analyzing the real contribution of international brands for Brazilian media segment.

Review question / Objective: The objective of this study was to investigate the natural pregnancy outcomes(IUP,REP) of ectopic pregnancy patients by comparing MTX versus surgery, MTX versus salpingostomy, MTX versus salpingectomy, salpingostomy versus salpingectomy, MTX versus expectant treatment. Condition being studied: Ectopic pregnancy(EP) remains one of the most common gynecological emergencies and a leading cause of maternal death in early pregnancy, Increase infertility and repeat ectopic pregnancy. Surgical treatment is considered the gold standard treatment, but with advances in early diagnosis, such as β -hCG levels and transvaginal ultrasound, EP can be diagnosed in the early stage, and the emergency operation rate and mortality rate of ectopic pregnancy are significantly reduced, and some patients can choose to expectant treatment or medical treatment, such as methotrexate (MTX). Naveed AK et al. suggests that expectant treatment is as safe and effective as MTX in EP patients with stable hemodynamic conditions and decreased or low β -hCG levels. The overall success rate of MTX is reported to be as high as 90% when choosing good indications. There is an increasing attention to reproductive outcomes for EP patients, especially for those who want to have a child, so it is crucial to clarify the impact of each treatment modality on natural pregnancy outcome.

This paper combines petrography with in situ laser-ablation inductively coupled plasma mass spectrometry to document trace-element variations in pyrite (Py) from Mississippi Valley-type (MVT) and fracture-controlled replacement (FCR) deposits in the Kootenay Arc, British Columbia. Three generations of pyrite are Py 1, Py 2, and Py 3. Pyrite 1, the earliest (occurring in MVT deposits only), has higher Ag, Ba, Cu, Ge, Pb, Sb, Sr, Tl, and V than adjacent Py 3. It has higher Ag, Au, Ba, Cu, Ge, Pb, and Tl than Py 2. Pyrite 2 occurs in MVT and FCR deposits. Relative to FCR Py 2, MVT Py 2 is enriched in Co, Ni, Mo, Ba, Tl, and Pb and depleted in other elements. The FCR Py 2 has growth-related compositional banding, which is absent in MVT Py 2. The FCR Py 2 has Ag, Cu, Ga, Ge, In, Sn, and Zn enriched cores, intermediate Au- and As-rich bands, and Co- and Ni-rich rims. Pyrite 3, the latest occurring pyrite, present in MVT and FCR deposits, is enriched in Co and Ni near overgrowths or infillings of sphalerite. Variations in composition of Py reflect mineralogy, characteristics of ore-forming fluids, and differences in physicochemical conditions between MVT and FCR deposits at the time of ore deposition.

Tumors in commercial poultry are caused mainly by infection with avian herpes and retroviruses, the herpesvirus Marek's disease virus (MDV) and the retroviruses, reticuloendotheliosis (REV), lymphoid leukosis, subgroups A-I and J (ALV and ALV-J) in chickens, or Iymphoprolipherative disease (LPDV) in turkeys. Infection with one virus aggravates the clinical outcome of birds that are already infected by another oncogenic virus. As these viruses do not interfere for infection, MDV and one or more retroviruses can infect the same flock, the same bird and the same cell. While infecting the same cell, herpes and retroviruses might interact in at least three ways: a) Integration of retrovirus genomes, or genomic fragments (mainly the LTR) into MDV;b) alteration of LTR-driven expression of retroviral genes by MDV immediate- early genes, and c) by herpesvirus induced cellular transcriptional factors. The first type of molecular interaction have been demonstrated to happen efficiently in vitro by Dr. Kung, in cases multiple infection of cell cultures with MDV and REV or MDV and ALV. Moreover, Dr. Witter showed that an in vitro-created recombinant, RM1, had altered in vitro replication and in vivo biological properties. A more comprehensive characterization of RM1 was carried out in the present project. We sought to highlight whether events of such integrations occur also in the bird, in vivo. For that, we had first to determine the prevalence of dually-infected individual birds in commercial flocks, as no systematic survey has been yet reported. Surprisingly, about 25% of the commercial flocks infected with avian oncogenic viruses had a multiple virus infection and 5% of the total samples ana lysed had multiple virus sequences. Then, we aimed to evaluate and characterize biologically and molecularly the resulting recombinants, if formed, and to analyse the factors that affect these events (virus strains, type and age of birds and time interval between the infection with both viruses). The perception of retrovirus insertions into herpesviruses in vivo is not banal, as the in vivo and in vitro systems differ in the viral-target cells, lymphocytes or fibroblasts, in the MDV-replicative type, transforming or productive, and the immune system presence. We realized that previous methods employed to study in vitro created recombinant viruses were not adequate for the study of samples taken directly from the bird. Therefore, the Hot Spot-combined PCR was developed based on the molecularly known RM1 virus. Also, the PFGE that was used for tissue cultured-MDV separation was inefficient for separating MDV from organs, but useful with feather tips as a source of bird original MDV. Much attention was dedicated now to feathers, because if a recombinant virus would be formed in vivo, its biological significance would be evident by horizontal dissemination through the feathers. Major findings were: a) not only in vitro, but also in vivo MDV and retrovirus co-infections lead to LTR integrations into MDV. That was shown by the detection of chimeric molecules. These appeared in low quantities and as quasispecies, thus interfering with sequence analysis of cloned gel-purified chimeric molecules. Mainly inserts were located in the repeat long MDV fragments. In field birds chimeric molecules were detected at a lower frequency (2.5%) than in experimentally infected birds (30-50%). These could be transmitted experimentally to another birds by inoculation with chimeric molecules containing blood. Several types of chimeric molecules were formed, and same types were detected in birds infected by a second round. To reproduce viral integrations, in vivo infection trials were done with field inoculate that contained both viruses, but the chimeric molecule yield was undetectable.

Farther west, platform carbonate rocks of the Kootenay Arc host MVT and fracture-controlled replacement (FCR) deposits. This is the first systematic LA-ICP-MS study of carbonates in MVT and FCR deposits. We investigated seven MVT deposits in the Rocky Mountains, and five MVT deposits in the Kootenay Arc. None of the post-Archean Australian shale (PAAS)-normalized REE profiles show light REE (LREE) depletion and strong negative Ce anomalies characteristic of modern seawater: some profiles are nearly flat; others show depletion in LREE similar to seawater but without negative Ce anomalies; others are middle REE enriched. Carbonates with a strong positive Eu anomaly precipitated from or interacted with different fluids than carbonates with flatter profiles without a strong positive Eu anomaly. REE signatures reflect crystallization conditions of primary carbonates, and crystallization and re-equilibration conditions of carbonates with ambient fluids during diagenesis, deep burial, and/or metamorphic recrystallization. Chemical evolution of fluids along their migration path, fluid-to-rock ratio, fluid acidity, redox, and temperature also influence REE profile shape, which helps establish genetic and timing constraints on studied deposits and improves knowledge of the metallogeny of the Kootenay Arc and Rocky Mountains.
