Full textThe alteration of mechanisms involved in intestinal homeostasis leads to the development of colorectal cancer (CRC). In France, CRC is the second leading cause of cancer mortality, and it is therefore necessary to improve our knowledge to treat it better. The objective of this project was to understand more precisely mechanisms involved in intestinal homeostasis through the atypical Cadherin MUCDHL whose expression seems decreased in CRC. However, the consequences of this loss, as well as the mode of action and the molecular relays of MUCDHL were largely unknown. Thus, the study of a large cohort of patients and of a murine model confirmed that the loss of MUCDHL enhances intestinal tumorigenesis. In addition, the functional characterization of the interaction between MUCDHL and β-catenin showed that the mode of action of MUCDHL is more complex than a simple membrane sequestration of β-catenin involving different signaling pathways. Finally, new interactants of MUCDHL were identified. The results obtained through this work show, for the first time, the tumor suppressor role of MUCDHL in the colon, and provide informations about its regulation and mode of action

Full textBecause of its frequency and severity, Colorectal Cancer (CRC) remains an important public health issue. Our objective is to understand mechanisms contributing to intestinal homeostasis through Mucdhl, a poorly characterized atypical cadherin that may play a unusual role in the intestinal , epithelium and be implicated in CRC. lnterestingly, its expression seems to be frequently reduced in CRC, while its retention in colon cancer cells decreases their tumorigenic potential.To better apprehend the mode of action of Mucdhl, a functional characterization of its interaction with oncogen,iç β-catenin was performed and new partners have been identified in intestinal cells.To understand the role of Mucdhl in intestinal physiology, mice genetically-invalidated at the Mucdhl locus were studied. Analysis of the consequences of Mucdhl loss of expression indicates that it is involved in the morphology and function of the mouse intestine, but also in metabolic processes. Moreover, Mucdhl loss of expression increases the sensibility of mice to the development of certain intestinal tumors. Thus, we generated new information on the physiopathological functions of Mucdhl, an intriguing atypical cadherin potentially involved in CRC

Full textMUCDHL is an atypical cadherin that has been poorly studied. The data obtained so far suggest that this gene has tumor suppressive activity in the intestine, namely by its interaction and inhibitory effect on β-catenin, and that its expression is frequently decreased in colorectal cancers (CCR). In parallel to this anti-oncogenic function, other studies have suggested that MUCDHL is involved in the assembly of the intestinal brush border (BB), by contributing to the formation of an inter-microvilli interaction complex. Our objective was to determine the function and mode of action of MUCDHL in the digestive system, Through a detailed characterization of the interaction with β-catenin, we showed that the anti-oncogenic mode of action of MUCDHL is more complex than a simple membrane sequestration of β-catenin. In addition, we confirmed the tumor suppressive function of MUCDHL on a very large cohort of human CCR and showed for the first time that its loss amplifies intestinal tumorigenesis in a murine model. Moreover, the study of the phenotype of Mucdhl-/- mice allowed us to demonstrate the importance of MUCDHL in the homeostasis of the digestive system. Indeed, the absence of MUCDHL causes morphological alterations of the intestinal BB, but also numerous metabolic disturbances. Thus, this work provides new information on the function and mode of action of MUCDHL in the digestive system

Full textThe pathogenesis of Inflammatory Bowel Diseases (IBD) remains poorly understood and there is currently no curative treatment. In addition, patients with colonic IBD are at increased risk of colorectal cancer (CRC). The atypical cadherin MUCDHL could be involved in IBD. We studied the role of MUCDHL in the pathogenesis of IBD, as well as the molecular mechanisms that restore its expression. We have shown that MUCDHL expression is decreased in the inflammatory intestinal mucosa. In addition, 5-aminosalicylates, used to treat IBD and prevent associated CRC, increase its expression by stimulating PPAR-γ and CDX2 or by inhibiting β-catenin. In Mucdhl -/- mice, the absence of MUCDHL accelerates and amplifies colonic inflammation induced by Dextran Sodium Sulfate and delays mucosal repair. Our data show MUCDHL's involvement in the pathophysiology of IBD and suggest that it could be a therapeutic target of interest

First of all, the state of the art was developed in a literature review which allowed to describe the main phenomena that occur during processing of a mixture of supercritical fluid and polymer in microcellular foam. Then, a study at the laboratory scale allowed to characterize and model the behavior of a thermoplastic polyurethane in contact with a supercritical fluid and deduct the characteristics and behaviors essential to the understanding of various phenomena encountered during the process(diffusion, solubility , nucleation...). Esta pesquisa tem o propósito de observar semelhanças e diferenças entre o jornalismo esportivo brasileiro e argentino, com ênfase na tensão entre objetividade e subjetividade, tanto em relação ao texto publicado quanto às condições políticas, sociais e econômicas que influenciaram e influenciam o produto jornalístico esportivo dos dois países. Utiliza-se o estudo comparativo como principal procedimento metodológico, sendo executado por meio de dez itens de comparação, divididos em três categorias: construção textual, linguagem e discurso. Adota-se como corpus as matérias veiculadas nos jornais Lance! e Olé ? maiores diários esportivos nos seus respectivos países ? durante a Copa do Mundo de 2014, além de entrevistas semiestruturadas com jornalistas esportivos vinculados aos referidos veículos.
Abstract : This research proposed to observe similarities and differences between Argentinean and Brazilian sports journalism, emphasizing the tension between objectiveness and subjectiveness, not just inside of texts that had been published, but also the political, social and economical conditions that have been influencing the sports journalistic products in both countries. We utilized comparative study as main methodology procedure, using ten comparative topics, divided in three categories: textual construction, language and discourse. Wnalyzed articles from Lance! and Olé ? biggest sports newspapers into its nations ? during the 2014 Fifa World Cup, in addition to semi-structured interviews with sports journalists from the named journals.

Full textWith a new generation of lepton colliders being conceived, muons have been proposed as an alternative particle to electrons. Muons lose less energy to synchrotron radiation and a Muon Collider can provide luminosity within a smaller energy range than a comparable electron collider. This allows a circular collider to be built. As part of the accelerator, it would also be possible to allow the muons to decay to study neutrinos.
Because the muon is an unstable particle, a muon beam must be cooled and accelerated within a short amount of time. Muons are generated with a huge phase space, so radio frequency cavities placed in strong magnetic fields are required to bunch, focus, and accelerate the muons. Unfortunately, traditional vacuum RF cavities have been shown to break down in the magnetic fields necessary.
To successfully operate RF cavities in strong magnetic fields, the cavity can be filled with a high pressure gas in order to mitigate breakdown. The gas has the added benefit of providing cooling for the beam. The electron-ion plasma created in the cavity by the beam absorbs energy and degrades the accelerating electric field of the cavity. As electrons account for the majority of the energy loss in the cavity, their removal in a short time is highly desirable. The addition of an electronegative dopant gas can greatly decrease the lifetime of an electron in the cavity.
Measurements in pure hydrogen of the energy consumption of electrons in the cavity range in 10-18 and 10-16 joules per RF cycle per electron. When hydrogen doped with dry air is used, measurements of the power consumption indicate an energy loss range of 10-20 to 10-18 joules per RF cycle per ion, two orders of magnitude improvement over non-doped measurements. The lifetime of electrons in a mixture of hydrogen gas and dry air has been measured from < 1 ns, up to 200 ns. The results extrapolated to the parameters of a Neutrino Factory and Muon Collider indicate that a high pressure gas filled RF cavity will work in a cooling-channel for either machine.

Full textA Ilustração de Moda foi, desde o seu aparecimento, considerada tanto um meio publicitário para as criações de designers, como um registo histórico do traje, tendo portanto passado por variadas técnicas, da pintura à fotografia, cobrindo também o desenho digital. Actualmente a Ilustração de Moda incluí o cruzamento de diferentes técnicas, criando assim um maior valor na peça final. Este Projecto de Investigação teórico-prático pretende chegar a um Estudo Exploratório do cruzamento da forma ilustrativa de grandes nomes da história da pintura com a Ilustração de Moda Contemporânea. Como casos de estudo, foram abordadas as obras de três pintores – Alphonse Mucha, Gustav Klimt e Egon Schiele –, e posteriormente utilizadas para ilustrar o trabalho de três designers de Moda Contemporânea – Givenchy, Balmain e Ann Demeulemeester. Partindo de uma procura e revisão de conhecimentos teóricos sobre a vida e obra dos artistas em estudo, pretende-se posteriormente reflectir estes conhecimentos na prática de experiências ilustrativas. A exploração prática desta Dissertação é dividida em três momentos, cada um resultando numa série de ilustrações distintas. No primeiro momento cada ilustração será realizada apenas com a influência da obra de um pintor; no segundo cada uma das ilustrações será baseada nas características da obra de dois pintores distintos; o terceiro e último momento resultará em ilustrações obtidas através do cruzamento das características formais dos três casos de estudo.
ABSTRACT: Fashion Illustration, since created, has been considered both an advertising medium for creations of design and an historical record of the costume, using several different techniques from painting to photography, including digital design. Currently Fashion Illustration encompasses the intersection of different techniques, creating a greater value in the final piece. This Theoretical Practical Research Project, aims to develop an exploratory study of the intersection of great names in the History of Painting, convened to the Illustration of Contemporary Fashion. As case studies, the formal work of three artists was addressed - Alphonse Much, Gustav Klimt and Egon Schiele -, and formerly used to illustrate the work of three contemporary fashion designers: Givenchy, Balmain and Ann Demeulemeester. The Project begins with the search and review of theoretical knowledge of the life and work of the artists under study, so that later, using this base, it is possible to reflect this knowledge in practice of illustrative experiences. The practical exploration of this Dissertation is organized in three moments, each resulting in a series of distinct illustrations. At first each illustration will be drawn under the influence of a painter only; in the second moment, each the illustration will be based on the characteristics of the work of two distinguished painters; while the third and final part, will result in a set of figures obtained by crossing the formal characteristics of the three case studies.

Full textThis dissertation explores the so-called too-much-choice effect, according to which an overabundance of options eventually leads to negative consequences, such as a diminished motivation to choose any of them or a decreased satisfaction with the finally chosen alternative. While strong instances of this effect have been found in a small number of studies in the past, its theoretical underpinnings are still unspecific. Because the effect challenges basic axioms of rational choice theory and it also has important implications for applied fields such as marketing and public policy making, it is important to get a better understanding of the mechanisms that lead to the effect. As a starting point to test these mechanisms, an experimental paradigm is needed in which the effect reliably occurs. Therefore, I first strived to replicate previous experiments that reported large effect sizes. Yet in a series of three replications in the field and in the lab with a total of 850 participants, I did not find an effect of too much choice, suggesting that the effect is less robust than previously thought and that it depends on certain boundary conditions instead. To find out which specific conditions are necessary to reliably elicit it, I subsequently examined several boundary conditions in a series of six laboratory experiments. Based on the results of these experiments with a total of 595 participants, most of the tested boundary conditions could be ruled out as explanations of why and when the effect of too much choice occurs. The results of a meta-analysis of published and unpublished data including my own suggest that the effect of too much choice is smaller than previously thought and that the differences between the studies that found the effect and those that did not cannot be explained by mere chance. As a consequence, a further search for moderator variables in future research seems justified.
