Abstract:
Abstract In mice, gene targeting has become a useful tool for resolving the functions of proteins and for creating new animal models. Cre/loxP technology has been used broadly for generation of genetically modified mice. The Cre recombinase recognizes a specific DNA sequence, called loxP, and removes any DNA fragment between two loxP-sites. The activity of the Cre recombinase can be controlled spatially and temporally with cell- or tissue-specific promoters and synthetic inducing agents, such as tamoxifen or tetracycline. In this thesis, we employed tamoxifen-induced Cre recombination in vitro

3D-tulostus on kehittyvä ala, jonka sovelluksilla on suuria tulevaisuuden mahdoll

Abstract The embryonic urogenital system generates the metanephric kidneys, the gonads and the adrenal glands, and its development is based on sequential and reciprocal cell and tissue interactions. The mechanisms which regulate urogenital ontogeny are still poorly understood. In this thesis, the roles of Wnt-4 and ErbB4 functions in gonad and kidney development were analysed by using in vivo functional genomic technologies. Wnt-4 is crucial in female development since its absence leads to a partial female to male sex reversal. We found that Wnt-4 mediated the interactions between the somatic

Abstract For decades, the mammalian kidney has served as a model system for studying developmental processes, such as induced epithelialization, branching morphogenesis, and cell differentiations. The possibility to recapitulate and follow the renal organogenesis ex vivo in organ culture set-ups has provided a large amount of molecular and cellular information about sequential events during development. However, certain limitations remain when combining traditional organ culture set-ups with modern molecular technology. This thesis seeks to address these disadvantages. In the experimental part

Abstract Wnt4, a member of the Wnt family of secreted factors, is essential for kidney organogenesis since the kidney fails to develop in its absence. Besides the kidney, Wnt4 signaling is involved in the control of development of several other organs such as the gonads, adrenal glands and pituitary gland. In the context of the embryonic kidney, Wnt4 signaling induces mesenchymal to epithelial transition of the progenitor cells in the metanephric mesenchyme, an early step in nephrogenesis. Wnt4 signaling may also be relevant in the development of a childhood kidney tumor, the Wilms’ tumor, tha

Abstract Kidney diseases affect an increasing number of people worldwide, and there is a growing demand to develop new treatments and increase the number of transplantable organs. New treatments can be designed when new knowledge is gained by studying the details of kidney development. The ex vivo culture techniques have been used for over a century to study the development of kidneys, but they are not optimal for long-term imaging and following the nephrogenesis process over time. Abstract The aim of this work was to evaluate the outcome of childhood Henoch-Schönlein purpura (HSP), the effectiveness of Cyclosporine A (CyA) for treating severe HSP nephritis (HSN), and more particularly the possibility for influencing the course of HSP disease by early prednisone treatment. A total of 47 adults who had had childhood HSP were evaluated after a mean of 24.1 years (16.4–35.6). The outcome was highly dependent on the renal symptoms at onset, since 7 out of 20 adults (20%) who had severe renal symptoms at onset had renal impairment as adults, compared with 2 out of 27 (7%) wit

Abstract Urinary tract infections (UTIs) are among the most common bacterial infections in childhood and have a tendency to recur. The ability of uropathogens to form biofilm may be important in the pathogenesis of UTI. Although childhood UTIs are thought to increase the risk of chronic kidney disease (CKD), evidence showing this association is scarce. Cranberry juice has been shown to prevent UTIs in women, but evidence of its efficacy in children is lacking. The aim of this work was to evaluate the significance of biofilm formation for the clinical presentation of UTI, the long-term conseque

Abstract This thesis focuses on mammalian kidney development and in particular on the question of how two Wnt signalling pathway genes, an antagonistic Dkk1 and an agonistic ligand Wnt5a, regulate the process. Wnts are secreted ligands that are involved in many developmental processes, including gonadal differentiation and kidney development, but also in various diseases and malformations. Wnts form a large signalling family containing 19 different glycoprotein ligands in mammals. Wnt signalling occurs via two different intracellular pathways. A canonical pathway proceeds via beta-catenin, and

Abstract Blood pressure is usually measured on a clinic visit as a momentary value. It can also be defined as a continuum based on several repeated measurements. Ambulatory blood pressure measurement (ABPM) is a method of repeated BP measurements targeted to evaluate the circadian blood pressure (BP). Nondipping, i.e., the lack of reduction of BP during the night, has been shown to associate with cardiovascular endpoints. The aim of this study was to investigate the association between 24-hour ABPM and cardio-metabolic confounders in a cross-sectional, population-based study design. Particular
