Recombinant mycobacteria expressing Mycobacterium tuberculosis extracellular proteins are leading candidates for new vaccines against tuberculosis and other mycobacterial diseases, and important tools both in antimycobacterial drug development and basic research in mycobacterial pathogenesis. Recombinant mycobacteria that stably overexpress and secrete major extracellular proteins of M. tuberculosis in native form on plasmids pSMT3 and pNBV1 were previously constructed by the authors. To better understand the biology and the virulence determinants of the two major mycobacterial human pathogens Mycobacterium tuberculosis and Mycobacterium leprae, their genome sequences have been determined recently. In silico comparisons revealed that among the 1439 genes common to both M. tuberculosis and M. leprae, 219 genes code for proteins that show no similarity with proteins from other organisms. Therefore, the latter ‘core’ genes could be specific for mycobacteria or even for the intracellular mycobacterial pathogens. To obtain more information as to whether these genes really were m

Mycobacterium tuberculosis is the leading cause of death from a single infectious agent. The varying efficacy of the BCG vaccine and the emergence of multi-drug resistant strains of M. tuberculosis have made it essential that novel drug and vaccine targets are identified. Antitermination is a mechanism by which RNA polymerase is able to transcribe through both Rho-dependent and-independent terminators.

Abstract:
A genetic test by PCR-reverse line blot hybridization was developed for rapid detection of mycobacterium tuberculosis and rifampin resistance simultaneously. Duplex PCR targeting IS 6110 and rpoB gene was employed to detect M. tuberculosis and the rpoB fragment with rifampin resistance hot spot region. The analytical sensitivity of duplex PCR for IS 6110 and rpoB gene was found to be 10 and 100fg of M.Tuberculosis H37Rv DNA, respectively. Since duplex PCR was specific to M. tuberculosis complex, it was thus able to be applied directly in clinical specimens. Oligonucleotide probes were designed

Abstract:
Leptospirosis is a worldwide zoonotic disease. Pathogenesis of leptospirosis is not well understood. Identification of virulence factors of pathogenic Leptospira and their roles in pathogenesis is crucial. Based on available whole-genome sequences of Leptospira, two presumptive virulence genes which are homologous to the invasionA (invA) gene of Rickettsia prowazekii and the mammalian cell entry (mce) gene of Mycobacterium tuberculosis, have been identified. The function of these genes may involve in the attachment and invasion of eukaryotic cells. We proposed that these gene homologues should
