AbstractGiven two k-graphs (k-uniform hypergraphs) F and H, a perfect F-tiling (or F-factor) in H is a set of vertex-disjoint copies of F that together cover the vertex set of H. For all complete k-partite k-graphs K, Mycroft proved a minimum codegree condition that guarantees a K-factor in an n-vertex k-graph, which is tight up to an error term o(n). In this paper we improve the error term in Mycroft’s result to a sublinear term that relates to the Turán number of K when the differences of the sizes of the vertex classes of K are co-prime. Furthermore, we find a construction which shows that our improved codegree condition is asymptotically tight in infinitely many cases, thus disproving a conjecture of Mycroft. Finally, we determine exact minimum codegree conditions for tiling K(k)(1, … , 1, 2) and tiling loose cycles, thus generalizing the results of Czygrinow, DeBiasio and Nagle, and of Czygrinow, respectively.

Abstract:
This work was originated from the increasing interest in several industries to implement voice based virtual assistant solutions powered by the Natural Language Processing field of study. This work is focused on the automotive industry Human Machine Interface related products, specifically the Instrument Panel. Nowadays people are constantly using virtual assistants like Google Assistant, Alexa, Cortana or Siri on their electronic devices. Furthermore, 31% of cars have a built-in virtual assistant, for example Ford uses Alexa, Merced­es-Benz and Hyundai use Google Assistant, BMW and Nissan use Cortana, GM uses IBM Watson, Honda uses Hana and Toyota uses YUI. Apart from the proprietary solutions described earlier, there are also contemporary open-source generic solutions available on the market, such as Mycroft AI which stands out from other technologies due to ready to deploy, well documented, simple installation on a Linux PC or RPI SoC, and simple execution. This paper presents a way to use Mycroft AI as an alternative to add artificial intelligence-based voice assistance to applications in the automotive domain. The voice communication module presented here drives notifications related to three different entities: seat belt, fuel level and battery level, all of them are telltales present in any automotive Instrument Panel. Since the Mycroft AI design approach is based on Human Centered Design (HCD), the voice communication module presented here provides real user experience (UX) based design. As a conclusion, Mycroft AI demonstrates great potential as an alternative to add voice assistance to automotive industry Human Machine Interface related products. About future work, due to the fact that Mycroft AI is based on Python, there are many possibilities for connecting and expanding the voice communication module by using countless Python libraries in order to import and process any type of information, in any format or source, for example the information from communication technologies like CAN, LIN, Ethernet, MOST, GPS or any other device or technology in order to create comprehensive automotive solutions.

Abstract:
Natural user interfaces are becoming popular. One of the most common today is interfaces with activated voice, including smart personal assistants such as Google Assistant, Alexa, Cortana, Siri, Alice, Bixby, Mycroft. This article presents the results of their evaluation in three dimensions: capabilities, language support, and how natural responses users experience. Evaluations were performed by analyzing existing reviews. The results show that Alexa and Google Assistant are much better than Siri and Cortana. However, there is no statistically significant difference between Alexa and Google Assistant, and neither of them integrates into modern messengers with a note-taking function, which is a significant disadvantage of such devices.

Given hypergraphs F and H, an F-factor in H is a set of vertex-disjoint copies of F which cover all the vertices in H. Let K−4 denote the 3-uniform hypergraph with four vertices and three edges. We show that for sufficiently large n ∈ 4ℕ, every 3-uniform hypergraph H on n vertices with minimum codegree at least n/2−1 contains a K−4-factor. Our bound on the minimum codegree here is best possible. It resolves a conjecture of Lo and Markström [15] for large hypergraphs, who earlier proved an asymptotically exact version of this result. Our proof makes use of the absorbing method as well as a result of Keevash and Mycroft [11] concerning almost perfect matchings in hypergraphs.

We say that a (di)graph G has a perfect H-packing if there exists a set of vertex-disjoint copies of H which cover all the vertices in G. The seminal Hajnal–Szemerédi theorem characterizes the minimum degree that ensures a graph G contains a perfect Kr-packing. In this paper we prove the following analogue for directed graphs: Suppose that T is a tournament on r vertices and G is a digraph of sufficiently large order n where r divides n. If G has minimum in- and outdegree at least (1−1/r)n then G contains a perfect T-packing.In the case when T is a cyclic triangle, this result verifies a recent conjecture of Czygrinow, Kierstead and Molla [4] (for large digraphs). Furthermore, in the case when T is transitive we conjecture that it suffices for every vertex in G to have sufficiently large indegree or outdegree. We prove this conjecture for transitive triangles and asymptotically for all r ⩾ 3. Our approach makes use of a result of Keevash and Mycroft [10] concerning almost perfect matchings in hypergraphs as well as the Directed Graph Removal Lemma [1, 6].

This study explores the portrayal of negative freedom as conceptualized by Isaiah Berlin in the character of Enola Holmes in the film Enola Holmes (2020). The research aims to identify aspects of Berlin’s liberalism reflected in the narrative, to describe how Enola exemplifies the principles of negative freedom, and to highlight the significance of achieving such freedom in a patriarchal society. Employing a qualitative method, this analysis focuses on narrative and visual elements that reflect Berlin’s concept of negative freedom. The data were collected by identifying scenes and applying the theory. Enola’s physical and mental transformation is central to her personal growth, demonstrating how the film explores the themes of self-discovery and autonomy. The findings reveal that Enola’s journey represents a compelling embodiment of Berlin’s negative freedom. At the film's outset, Enola is subjected to patriarchal restrictions, particularly through the control exerted by her brother Mycroft. Her decision to search for her mother marks a pivotal moment in reclaiming her autonomy, as she begins charting her own path. The analysis further demonstrates that Enola’s struggle against societal norms and her eventual self-acceptance signify a transformative journey toward individual freedom. Through her defiance of external controls, Enola exemplifies Berlin’s concept of negative freedom, underscoring the importance of autonomy in fostering personal growth and liberation. Abstract Alzheimer’s disease (AD) is the most common form of dementia and is associated with memory loss and cognitive impairments that affect daily life. Approximately 5.8 million older adults in the U.S. are living with AD. People with AD often require high levels of care and assistance to maintain daily activities. The majority of care provided to a person living with AD or other forms of dementia is from a family caregiver, representing 18.6 billion hours of unpaid care valued at $244 billion. The long duration, time-intensive nature of caregiving imposes high burdens on caregivers. We designed a socially assistive robot to engage in conversation with people with AD, by engaging in conversation, helping them conduct simple daily tasks, and relieving caregivers of some of their responsibilities. Using design-thinking methodology, a prototype social robot has been created using 3-d printing technology and a single board computer based on raspberry Pi. Interaction between human and robot is implemented using the Mycroft open source voice assistant. The authors demonstrated that the robot is able to have natural conversations with human users. The overall cost of the robot is estimated to be less than $300, rendering it possible for wide distribution among the public. Future research includes further implementation of various cognitive assessment and cognitive training programs using the social robot to improve the quality of life for people living with AD.

Abstract Our interdisciplinary team of designers, computer scientists, and engineers have begun the development of a low-cost social robot called FRED (the Friendly Robot to Ease Dementia). FRED has a conversational capability which will allow the device to be used as assistive companions for people with AD, engaging conversation, assisting with reminders, playing simple games and music, and relieving caregivers of some of their responsibilities. FRED was created using 3-d printing technology and a single board computer based on raspberry Pi. Interaction between human and the system was implemented using the Mycroft open-source voice assistant. The overall cost of the robot is estimated to be less than $300, rendering it possible for wide distribution among the public. Our presentation will share results from our participatory focus group which helped assist in the design and interactions of FRED. This human-centered design approach included neurologists, healthy older adults, as well as ADRD patients and their caregivers. A common language of important terms (e.g., feedback, interface, training, cognitive assessment) to aid in initial conversations were implemented. A thinking-aloud protocol was adopted once we had a prototype of FRED, where users were able to express freely any problems and/or concerns during interaction. Insights from the thinking-aloud results were used to improve the user interface design to enable the users and robot to interact and collaborate in an effective, natural way. Feedback from the focus group were carefully documented and the results will be shared during our presentation.

The paper focuses on Dostoevsky’s influence in D. Tartt’s novel The Secret History. The author shows how the main plot collisions of Crime and Punishment are inverted in the book of the American writer. The character of the book is the collective antagonist of Raskolnikov who recognizes the right to kill. The Übermensch is portrayed in the novel as the impeccable rationalist Mycroft Holmes. The spiritual force that guides Tartt’s characters on the path of transformation into an Übermensch is the pagan god Dionysus, to whom the students go to kill. The novel shows the self-destruction of a murderer who refused to see a person in his victim. The author’s voice in the novel is designed on the model of Dostoevsky’s poetics, using the reception of strange phrases uttered by the hero for no reason, as it happens during the wake with the family of a murdered student. At this moment, a phrase pops up in Richard’s head by itself: “It was I who killed the old woman and her sister Lizaveta with an axe and robbed her”. This phrase of Raskolnikov is a brief synopsis of the plot of Crime and Punishment. In Donna Tartt’s novel, this phrase expresses the author’s position, the author’s explanation of what is happening in the book: other characters in another world repeat Raskolnikov’s fate, and the author shows what this leads to. Raskolnikov managed to escape by converting to Christ and accepting his human nature. Donna Tartt’s Übermensch could not be saved, because instead of Christ he communicated with Dionysus. Taking communion with Dionysus in the ancient ritual of homophagy, students go to murder. The novel shows the self-destruction of all five murderers who refused to see a person in their victim: suicide (Henry), suicide attempt (Francis), alcohol and drugs up to complete desocialization (Charles), selfpunishment in the form of rejection of the joys of life and communication with people (Camilla), loneliness (Richard). Thus, Donna Tartt proves from the opposite the same idea that Dostoevsky expressed in his Orthodox novel: every life is priceless, murder destroys people and the world.

Passive design strategy is the approach that maximizing utilization of natural resources to achieve occupancy comfort which covers the thermal comfort, visual comfort and acoustic comfort while minimizing secondary energy usage. Adapting passive design strategy during building design-stage is beneficial in reducing energy load of the building for long term, furthermore to reduce the energy consumption in day to day building operation. Applying Green Building Rating Tools, GBRT promotes passive design strategy for non-residential building and will assist the building designer to comply with the local and international standard. The minimum requirement of occupancy comfort is captured in GBRT rating system with different approach and intension of the system. This review summarize the coverage of passive design strategy sectors for non-residential building in Malaysia by comparing three GBRT system namely Green Building Index, GBI NCNR, MyCREST ICSAS and GreenRE NRB system. GreenRE NRB seems more aggressive in occupancy comfort evaluation compared to GBI NCNR and MyCREST ICSAS. GreenRE allocates 48.7% of total marks focus on occupancy comfort aspect while GBI and MyCREST allocates 14% and 13.2% respectively from the total score. MyCrest had enforce the mandatory compliance of OTTV, RTTV and Uroof to meet baseline requirement as per MS1525:2019 with zero marks allocation. Based on marks percentage allocation, GBI and MyCrest prioritizing visual comfort while GreenRE give more focus on thermal comfort with higher marks allocation. All rating system give less focus on acoustic comfort. On average, 16.3% of marks covers the thermal comfort, 9% covers visual comfort and 1% cover acoustic comfort. GBI and MyCrest focus more on green building materials which covers the building lifecycle includes water management and waste disposal. All system contributes to Malaysia-UNFCCC ratification of Paris Agreement on 45% GHG reduction in longer runs.

Abstract The synergies of the Building Information Modelling (BIM) process and green building assessment tools (GBATs) like the Malaysian Carbon Reduction and Environmental Sustainability Tool (MyCREST) known as green BIM, ensure that information is accurately exchanged during the green building design (GBD) practices. However, the issue of poor Information Exchange synthesis persists due to the lack of a standard model for green BIM implementation. Therefore, this study aims to develop the BIM-MyCREST Process Model (BMPM) for the concept design of green buildings. To actualize the research aim, an in-depth interview was conducted with 10 construction experts. Using the narrative analysis technique, the interview data were analyzed to identify the incidents that take place during the BIM process execution in GBD practices. With reference to MyCREST Guide, BIM standards; and the use of Integrated DEFinition (IDEF) language (IDEF0 and IDEF3), the BMPM was explicitly coordinated following the narratives of the interviewees. The developed BMPM will enable the multi-disciplinary GBD team to use the diverse domain BIM software to exchange accurate information and make critical decisions during the design of green buildings; thereby developing a building with high sustainability performance and accomplishing the business and commercial objectives of the MyCREST rating.

Green Building rating tools are the essential need of this era, to cope up with the sustainable development goals, climate change, and natural resource degradation through buildings. Realization of green building incentives decently increased within past few decades with abrupt declination in real estate markets and economic depletion has decelerated the interest of investors towards the green building projects. This research calculates influence of costing elements in MyCREST (IS-design) using questionnaire survey distributed amongst qualified professionals (QP’S) of green buildings and expert practitioners. Firstly, factor score and then weightage factor was performed to produce the final result with weightage output for evaluating weighatge and ranking of the relevant criteria of MyCREST and life cycle cost elements respectively. It is found that the criteria of storm water management has weighatge of 0.236 as highest and criteria environmental management plan (EMP) as 0.061 as lowest. Research also identified another perspective by finding association of cost element at design stage of MyCREST and found that management cost is highly associated at design stage with the value of 87.7%. The outcome of this research will add value to green building development and map road towards sustainable development using green building tools to uplift quality of life. Furthermore, this paves a way to integrate various stages of MyCREST with life cycle costing tool to potentially contribute in evaluating cost association through green building rating tool.

Purpose Green building development practices reduce carbon footprint and promote sustainability in the built environment. To foster green building construction in Malaysia, several strategies and sustainability metrics like the Malaysian Carbon Reduction and Environmental Sustainability Tool (MyCREST) have been established. Yet, the implementation of green office building development in Sarawak still remains low. Therefore, the purpose of this paper is to develop MyCREST Embedded Framework (MEF) for enhancing the adoption of green office building construction in Sarawak. Design/methodology/approach To achieve the research aim, document analysis of the overarching MyCREST Reference Guide was conducted. Then, questionnaire was used to survey 120 green construction professionals in Kuching, the capital of Sarawak. Afterwards, data from the questionnaire was analyzed using Statistical Package for Social Science software version 22. As a result, the significant strategies for enhancing the adoption of green office building construction in Sarawak were identified, and ranked hierarchically using the Relative Importance Index. Findings The established significant strategies include: government policies and incentives, sustainable technological practices, defined process for executing green building projects and adequate information for the public. Moreover, this study recommends MyCREST as the suitable Green Building Assessment Tool for developing green office buildings in Sarawak due to its integration of carbon reduction strategies with sustainability indicators. Research limitations/implications This study is limited to Kuching; hence future study should be extended to other cities in Sarawak. Originality/value Based on the findings, the MEF is developed to enlighten the industry practitioners on the approaches that will elevate green office building development in Sarawak.

Carbon emission is released into the atmosphere as the result of various activities due to rapid urbanisation and thus contributed to global warming and climate change. The government has taken various initiatives to reduce the impact, including from the construction industry in order to support the carbon footprint reduction of 40% as pledged by the Prime Minister. Various strategies, such as the Malaysian Carbon Reduction and Environmental Sustainability Tool (MyCREST), have been established to promote green building development in Malaysia. Recent studies suggested that the selection of sustainable materials can reduce the overall carbon emission of a building, but the cost has been identified as the main barriers. This paper aims to analyse the potential of carbon footprint reduction by using sustainable material in mid-rise residential building and subsequently to evaluate the cost implication. The impact of the conventional and the selected sustainable materials was assessed using data from the MyCREST tool while the data for cost analysis were taken from various sources of cost data such as JKR Rates online (RATOL), JKR Sarawak Schedule of Rates (SOR), and previous research. The results show that the sustainable materials such as 30% of Blast Furnace Slag (BFS) concrete mixture, Aerated Autoclaved Concrete (AAC) block, and recycled steel roof truss has the potential to reduce the carbon emission. The findings also show that sustainable materials are slightly cheaper than the conventional materials except for the AAC block and clay roof tiles. Therefore, the potential of carbon emission reduction approach by using MyCREST as a guideline tool can assist in the reduction of the environmental impact of buildings.Keywords: Carbon emission, sustainable building materials, cost implication.

Abstract:
The construction industry is the biggest contributor to every country’s economy and development. It also uses many natural resources and produces much solid waste. It is important to apply environmental sustainability in the construction industry as construction activities contribute to environmental damage. This study investigates the impacts of construction activities in urban areas on environmental sustainability. The study adopts quantitative data collection via a questionnaire survey of the registered total population of 1,670 G7 Contractors in the Kuala Lumpur City Centre. A sample size of 313 was obtained and a total of 42 responded. The practice of sustainable construction sites in urban areas is still low. The construction sites in urban areas cause four main environmental impact. The highest is the ecosystem ecological impact followed by, public impact, and energy and natural resources impact. Additionally, the challenges of construction sites in urban areas to achieve environmental sustainability were higher initial costs, technical challenges, insufficient training, and lack of awareness and knowledge. The first potential solution was implementing green building assessment tools specifically using MyCREST followed by specific government roles, development of skills and education, and digitalization and Networking for construction site pollution management.

Abstract:
The purpose of this study is to report the results of a tension-free repair technique using expanded polytetrafluoroethylene Gore-Tex Mycro Mesh in patients with inguinal hernias. Over 3 years, 104 patients have undergone the aforementioned procedure for 118 hernias (85 indirect and 33 direct) in our clinic. Ninety-three of the patients were operated for the first time, whereas the remainder of them were operated for hernia recurrence. Fourteen patients had bilateral hernias, and six had incarcerated ones. The median patient's age was 53 years. Eighty-three patients were operated under general and 21 under local or epidural anesthesia. One hundred and two patients were offered a one-day surgery procedure, but three were readmitted with wound infection. Two of these patients were conservatively cured, and the third was submitted to mesh removal. Two patients developed hematoma in the scrotum and were discharged a week later after the absorption of the hematoma. All of the patients except the one who was submitted to mesh removal returned to normal life within a week after the operation. During the follow-up period, none of our patients experienced hernia recurrence. Our results are in accordance with those of the world literature. We perform the tension-free hernia repair by using the expanded polytetrafluoroethylene mesh in our patients because we believe it is a safe procedure without significant postoperative morbidity and with no hernia recurrences.

Automation has been developing since over the last four decades where advancement of computer hardware and in line with software developments has enabled various sort of design to be computerized. The design process is now more fast and accurate, and the entire process has been outstandingly improved by the available technology. However, the problems related to computerized design, started from the possibilities for unrecognized or unanticipated consequences as the design becomes more complicated. In line with it is the global warming issue that has been threatening the human being. Therefore, this paper reviews the design automation and its application in civil engineering design emphasizing in assessing the energy efficiency of buildings, which includes background information, its application, advantages, and challenges and also suggestions to further improve the automation in civil engineering design by artificial intelligence (AI). It was discovered that since over the past few years, BIM implementation in Civil Engineering design has started to be adopted massively in construction industry due to its tremendous advantages. In this paper, a new flow process in design automation in assessing building sustainability has been proposed by implementing AI BIM which is linked with MyCREST, a green rating tool in Malaysia in order to assist designers to conceptually design for a more energy efficient building.

The composition of chemical elements in rock samples can be used as a tool for classifying rock types, predicting the depositional environment, rock age, and the tectonic environment in which the rock is formed. X-Ray Fluorescence (XRF) is one of the instruments used to analyze rock samples. The advantage of using XRF analysis is that the analysis process can be done more quickly, easily, accurately, and does not damage the sample. However, before measurements are made using the XRF instrument it needs to be supported by an appropriate sample preparation process. In this study, optimal conditions in the standard (CRM) rock sample preparation of GBW 07105 and JR-1 have been studied. The parameters studied were variations in the composition ratio of Cellulose Mycro Crystalin (CMC) with standard (CRM) samples, namely 1: 4 and 1: 3. The technique of mixing the sample is done by using a mortar grinder and a shaker. Optimization of sample preparation was carried out using pressed powder pellet and fused glass bead techniques. Based on the data obtained from the analysis results the best ratio of binders to standard samples is 1: 3. The pressed powder pellet preparation technique is the best technique in analyzing samples using the X-Ray Fluorescence method. Further studies also need to be carried out to analyze minor elements and traces (REEs).

Mycroft (JCTA, 2016) determined the asymptotic values of $t_{k-1}(n, F)$ for $k$-partite $k$-graphs $F$ and conjectured that the error terms $o(n)$ in $t_{k-1}(n, F)$ can be replaced by a constant that depends only on $F$. In this paper, we determine the exact value of $t_2(n, K_{m,m}^{3})$, where $K_{m,m}^{3}$ (defined by Mubayi and Verstraëte, JCTA, 2004) is the 3-graph obtained from the complete bipartite graph $K_{m,m}$ by replacing each vertex in one part by a 2-elements set. Note that $K_{2,2}^{3}$ is the well known generalized 4-cycle $C_4^3$ (the 3-graph on six vertices and four distinct edges $A, B, C, D$ with $A\cup B= C\cup D$ and $A\cap B=C\cap D=\emptyset$). The result confirms Mycroft's conjecture for $K_{m,m}^{3}$. Moreover, we improve the error term $o(n)$ to a sub-linear term when $F=K^3(m)$ and show that the sub-linear term is tight for $K^3(2)$, where $K^3(m)$ is the complete $3$-partite $3$-graph with each part of size $m$.

The size of the largest $P$-free family in $2^{[n]}$, denoted by $La(n,P)$, has been extensively studied since the 1980s. We consider several related problems. For posets $P$ whose Hasse diagrams are trees and have radius at most $2$, we prove that there are $2^{(1+o(1))La(n,P)}$ $P$-free families in $2^{[n]}$, thereby confirming a conjecture of Gerbner, Nagy, Patkós and Vizer [Electronic Journal of Combinatorics, 2021] in this case. For such $P$ we also resolve the random version of the $P$-free problem, thus generalising the random version of Sperner's theorem due to Balogh, Mycroft and Treglown [Journal of Combinatorial Theory Series A, 2014], andCollares Neto and Morris [Random Structures and Algorithms, 2016]. In the first five books, Shane Peacock developed the landscape of Holmes’ early life, preparing the reader for his transition into the adult genius-detective that we all know and love. Holmes’ creator, Sir Arthur Conan Doyle did not supply much information about Holmes’ childhood, so authors have great scope to imagine how such an extraordinary person as Sherlock Holmes grew up to become what he is. Shane Peacock is not the first to create a prequel series, nor is he likely to be the last. Prequels to much-loved stories are always a risk. However, Peacock has done a remarkably good job of imagining Holmes’ early years while staying true enough to Conan Doyle’s character that readers will not be jarred when they progress from this series to the original series. In this rendition of Holmes’ childhood, much is explained by the fact that he is a half-Jew and that his parents have died leaving the boy in the care of the apothecary, Sigerson Bell. Bell has taught Holmes much of his vast knowledge base as well as some disguise and martial arts techniques. In this series we also meet Holmes’ brother, Mycroft, already ensconced in government, as well as youthful versions of Inspector Lestrade and the arch-enemy, here called Malefactor. By the end of this book, Holmes has matured and thinks, “I am a man… It is just the beginning”, giving readers a segué into the original series. While other series about Holmes’ youth, for example Andrew Lane’s Sherlock Holmes, the Legend Begins Series address older teen readers, Peacock has written these volumes for 10 to 14 year olds. His use of language is age-appropriate and the final volume, like the earlier ones, is action-packed with lots of intrigue and just enough of the macabre to keep pre-teens and early teens engaged. Becoming Holmes is a good read that is highly recommended for public libraries and both elementary and junior-high school libraries everywhere. Pada industri minyak kelapa sawit, pengeringan Crude Palm Oil (CPO) merupakan tahap penting untuk mengurangi kandungan air (moisture) pada produk CPO. Kandungan air (moisture) yang tinggi pada CPO dapat mempercepat reaksi hidrolisa trigliserida yang disebabkan oleh mycroba sehingga mengakibatkan ketengikan atau bau yang tidak sedap dalam minyak. Salah satu faktor yang dapat mempengaruhi tingginya moisture pada CPO adalah kurang spray-nya minyak saat proses pemisahan minyak di dalam vacuum dryer. Kurang spray-nya CPO dapat diakibatkan oleh penggunaan nozzle. Penggunaan nozzle vacuum dryer dengan ukuran yang tidak tepat dapat mempengaruhi efisiensi pengeringan dan kualitas produk. Oleh karena itu peneliti melakukan penelitian untuk mengurangi moisture pada CPO dengan cara mengganti ukuran diameter nozzle vacuum dryer dengan menggunakan metode 8 steps. Penggunaan metode 8 steps memberikan panduan yang sistematis dalam proses penelitian ini. Dengan mengganti ukuran diameter nozzle vacuum dryer didapatkan hasil bahwa terjadi penurunan total defect moisture dan persentase moisture antara sebelum dan setelah dilakukannya perbaikan.

Although green building assessment have been the study of many researchers over the past twenty years, it is still no systematic assessment of the detailed criteria for each assessment systems. Therefore, this research aimed to conduct critical review of green building assessment systems in Malaysia. This research is conducted through comparison analysis where the points allocated for each criteria is being measured and compared. It founds that energy efficiency criteria is the most widely used in green building assessment systems followed by water efficiency and innovations. This research showed that each green building assessment systems showed different culture and they used different criteria in evaluating the sustainability level of the green building projects. The outcomes of this research is expected to create awareness among construction industry players or decision makers to understand better and have a different perspective toward current green building assessment systems.

Abstract Background Owing to the lack of effective treatment options, early metastasis remains the major cause of pancreatic ductal adenocarcinoma (PDAC) recurrence and mortality. However, the molecular mechanism of early metastasis is largely unknown. We characterized the function of eukaryotic translation initiation factors (eIFs) in epithelial-mesenchymal-transition (EMT) and metastasis in pancreatic cancer cells to investigate whether eIFs and downstream c-MYC affect EMT and metastasis by joint interference. Methods We used The Cancer Genome Atlas (TCGA) and Genome Tissue Expression (GTEx) databases to analyze eIF4A1 expression in PDAC tissues and further validated the findings with a microarray containing 53 PDAC samples. Expression regulation and pharmacological inhibition of eIF4A1 and c-MYC were performed to determine their role in migration, invasion, and metastasis in pancreatic cancer cells in vitro and in vivo. Results Elevated eIF4A1 expression was positively correlated with lymph node infiltration, tumor size, and indicated a poor prognosis. eIF4A1 decreased E-cadherin expression through the c-MYC/miR-9 axis. Loss of eIF4A1 and c-MYC decreased the EMT and metastasis capabilities of pancreatic cancer cells, whereas upregulation of eIF4A1 attenuated the inhibition of EMT and metastasis induced by c-MYC downregulation. Treatment with the eIF4A1 inhibitor rocaglamide (RocA) or the c-MYC inhibitor Mycro3 either alone or in combination significantly decreased the expression level of EMT markers in pancreatic cancer cells in vitro. However, the efficiency and safety of RocA alone were not inferior to those of the combination treatment in vivo. Conclusion Overexpression of eIF4A1 downregulated E-cadherin expression through the c-MYC/miR-9 axis, which promoted EMT and metastasis of pancreatic cancer cells. Despite the potential feedback loop between eIF4A1 and c-MYC, RocA monotherapy is a promising treatment inhibiting eIF4A1-induced PDAC metastasis.

Commercial kits and instruments were used for cell apoptosis analysis, real-time PCR, western blotting, ELISA analysis and other experiments. Result Comparing the tumor tissues from prostatic cancer patients, we found elevated expression of tryptophan 2, 3-dioxygenase 2 (TDO2), and elevated Trp metabolism in chemo-resistant tumor tissues. In vitro, overexpression of TDO2 significantly promoted the Trp metabolism in prostatic cancer cell lines LNCaP and VCap, resulting in the multidrug resistance development. Mechanistically, we demonstrated that Trp metabolite kynurenine (Kyn) promoted the upregulation and nuclear translocation of transcription factor aryl hydrocarbon receptor (AhR). Subsequently, AhR collaborated with NF-κB to facilitate the activation of c-Myc. In turn, c-Myc promoted the up-regulation of ATP-binding cassette (ABC) transporters and Trp transporters, thereby contributing to chemoresistance and strengthened Trp metabolism in prostatic cancer. Interrupt of Trp/TDO2/Kyn/AhR/c-Myc loop with c-Myc inhibitor Mycro-3 efficiently suppressed the chemoresistance and improved the outcome of chemotherapy, which described a new strategy in clinical prostatic cancer treatment. Conclusion Our study demonstrates that elevated TOD2 expression promoted Trp metabolism and metabolite Kyn production, thus resulting in the activation of AhR/c-Myc/ABC-SLC transporters signaling pathway. Interrupt of Trp metabolism/c-Myc loop efficiently suppressed the drugs resistance induced by TDO2, which represented potential target to improve the outcome in drug-resistant prostatic cancer treatment.
