The similarity of John writings and the gnostics is still a lasting subject nowadays. All discoveries that attempt to clear things up get people’s attention. The discoveries of Nag Hammadi writings, whose some texts are gnostics, points at an Alexandrian II century AD Gnosticism that suffered from various influences, mostly Johannine. The exploration of such environment allow us not only to notice John richness, but also his diffusion in religious environments. Some of the writings of Nag Hammadi, as Trimorfaic Protenor, John Apocryphal and Gospel of Truth, have vocabularies and thematic affinities with the Fourth Gospel, especially with the Prologue. Through the similarity with the quoted texts, the question arises whether the texts of Alexandrian II century AD were used from John prologue to his composition. The hypothesis is that somehow the Gnostics had contact with the Johannine prologue and used their concepts to base their teachings. In order to answer this question, this work has bibliographic character, using the literature concerning the prologue of the Fourth Gospel and the analysis of the quoted texts and the prologue, from the tradition and the contemporary exegesis
A semelhança entre os escritos de João e os gnósticos é um assunto que continua em pauta atualmente. As descobertas que são feitas, em uma tentativa de lançar luz sobre o tema, chamam a atenção de todos. As descobertas dos escritos de Nag Hammadi, que possui alguns textos de autoria gnóstica, apontam para um gnosticismo alexandrino do século II d.C. que sofreu várias influências, mas principalmente a joanina. Explorar esse ambiente permite contemplar não apenas a riqueza de João como também sua difusão em ambientes religiosos. Alguns escritos de Nag Hammadi, como Protenoia Trimorfa, Apócrifo de João e Evangelho da Verdade, possuem afinidades vocabulares e temáticas com o Quarto Evangelho, sobretudo com o Prólogo. Através da semelhança com os textos citados, surge a pergunta se os textos do gnosticismo alexandrino do século II d.C. teriam se utilizado do Prólogo de João para sua composição. A hipótese é que de alguma forma os gnósticos tiveram contato com o Prólogo joanino e utilizaram seus conceitos para embasar seus ensinamentos. A fim de responder a tal questão este trabalho possui caráter bibliográfico, utilizando-se da literatura a respeito do Prólogo do Quarto Evangelho e da análise dos textos citados e do Prólogo, a partir da Tradição e da exegese contemporânea

Resumo: Esta tese tem como objetivo principal o estudo do narrador no evangelho de Mateus e de sua relação com o protagonista ¿ Jesus Cristo. Central em todo o trabalho é a constatação de que o narrador configura o texto de modo a produzir nos leitores a consciência de que a narrativa se refere não apenas a um personagem da história passada, mas a Jesus Cristo vivo. Este exerce a função de orientar a compreensão do texto. Para tanto, o narrador coloca-se em segundo plano e desenvolve técnicas para que o personagem principal ocupe espaço de proeminência. A minimização da presença do narrador, antes de se transformar em debilidade textual, produz abertura do texto ao leitor. Desse modo, as estratégias estabelecidas visam atrair o leitor para que participe da trama. Para chegar a tais conclusões, o trabalho discute variadas formas interpretativas pela quais o evangelho de Mateus é estudado na atualidade. Define o gênero literário ao qual pertence o evangelho como biografia greco-romana. Identifica o narrador, seu foco narrativo e a forma como organiza o evangelho em blocos narrativos e discursivos a partir da fonte principal, o evangelho de Marcos. Por fim, explicita estratégias literárias através da comparação exaustiva entre textos de Mateus e Marcos, demonstrando como elas apontam para propósitos retóricos específicos que o narrador deseja gerar nos leitores. O canal de discussão com biblistas esteve aberto, em alguns momentos utilizando interpretações e pontos de vistas, e em outros discordando de suas colocações. A principal delas diz respeito à declaração de que o evangelho apresenta um caráter catequético e desprovido de brilho. Em oposição, afirma-se que o evangelho de Mateus possui estratégias narrativas que o tornam extremamente persuasivo aos leitores. Torna-se claro que a desconsideração dos elementos de análise descritos no trabalho produz conseqüências nocivas à interpretação do texto bíblico
Abstract: The main goal of this thesis is the study of the narrator of the Gospel of Matthew and its relationship with its main character ¿ Jesus Christ. In the very heart of the dissertation is the observation that the narrator works his text in order to produce in the readers the consciousness that the narrative has to do with the living Jesus Christ, who plays the role of guiding the understanding the text, and not only with a mere character of a past history. To do so, the narrator put himself in a secondary place, and develops some mechanisms so that the main character may occupy a prominent role. The downsizing of the narrator is not a textual frailty. Rather, it produces an opening of the text to the reader. Thus, these strategies aim to attract the reader to participate in the plot. In order to reach these conclusions, the dissertation discusses several contemporary ways the Gospel of Matthew is interpreted at this moment. It also defines the literary genre of the Gospel as a Greek-Roman biography. Besides, it identifies the narrator, its narrative focus and how he organizes the gospel in discursive and narrative blocks from its main source, viz., the Gospel of Mark. As an ending, the dissertation explains the literary strategies through an exhaustive comparison between the texts of Matthew and Mark, giving evidence of how they point to specific rhetoric purposes the narrator wish to generate in the readers. There was an open channel of discussion with Bible scholars, sometimes agreeing, sometimes disagreeing with them. The main point of disagreement was related to the statement that the Gospel has a catechetical purpose and is deprived of brilliance. Arguing against this, it is stated that the Gospel of Matthew has narrative strategies that make it extremely persuasive to its readers. It is quite clear that the disrespect of the elements of analysis described in the dissertation produces harmful consequences to the interpretation of the biblical text

Abstract:
Glycoscience is important in many areas such as human health, energy and material science. Glycans have been shown to be involved in the pathophysiology of almost every major disease. Additional glycan structure knowledge is required to help advance personal medicine, and pharmaceutical developments, among others. For glycoscience to advance there is a need for large quantities of well-defined glycans and have quick access to glycosyltransferases for manipulating glycan synthesis. Herein, we will cover our efforts on studying the substrate specificities of human glycosyltransferases such as FUT8 and Gn-T V, and their application on N-glycan synthesis. Complex asymmetric N-glycan isomer structures have been related to many diseases such as breast cancer, among others. Synthesis of complex asymmetric N-glycan isomer structures including: alpha-1,6 core-fucosylated, and tri-antennary structures can be achieved by taking advantage of the high specificity of glycosyltransferases that can work as unique catalyst to generate well-defined glycan structures.

Nach grundsätzlichen Überlegungen zur Allgemeingültigkeit, Kultur- und Zeitbezo-genheit neutestamentlicher Aussagen untersucht die vorliegende Studie das Lehrver-bot der Frauen im gesamtbiblischen Kontext. Dadurch resultiert 1. Timotheus 2:12-14 als Schlüsseltext in der Frage, ob Frauen für den leitenden und lehrenden Dienst in der Gemeinde zugelassen sind. Hinweise für das richtige Verständnis von 1. Ti-motheus 2:12-14 sind Vergleiche mit anderen paulinischen Verweisen auf erzählte Ereignisse des Alten Testaments und ihre Funktion im jeweiligen Diskurs, die Wir-kungsgeschichte der Schöpfungsreihenfolge und von Evas Fall im Frühjudentum sowie die Funktion dieser Verweise in der Argumentation im 1. Timotheusbrief. Ab-schliessend wird 1. Timotheus 2:12-14 im Bezug auf den unmittelbaren Kontext un-tersucht. Die vorliegende Arbeit schließt, dass 1. Timotheus 2:12-14 trotz der Ver-weise auf die Schöpfungsreihenfolge und den Fall Evas nicht als allgemeingültig zu verstehen ist.
After general reflections on universal validity, and on the cultural and temporal set-ting of New Testament propositions, this study examines the prohibition on women teaching in Christian congregations in the context of the whole Biblical canon. From this perspective, 1 Timothy 2:12-14 offers a key role for the validity of women as leaders and teachers. Clues towards a correct understanding of 1 Timothy 2:12-14 are to be found in comparisons with references to Old Testament events and their par-ticular function in Pauline discourse, in reception of the order of creation and fall of Eve in early Judaism, and in the function of these references in the argument of 1 Timothy. Finally, 1 Timothy 2:12-14 is examined in view of its immediate context. The thesis concludes that, despite its reference to the order of creation and the fall of Eve, 1 Timothy 2:12-14 should not be understood as an absolute prohibition.

Both for Karol Wojtyła, the future Pope John Paul II, and Józef Tischner, freedom is a key matter. However, freedom must be e x p e r i e n c e d ; it is not revealed in objective metaphysics or in science.For Wojtyła, it is an experience of the moral a c t : freedom is a condition and in-gredient of the moral act and responsibility for it. It enters into the composition of the essential structure of the person, which means “self-restraint,” or shaping oneself through free choices. Freedom has the power to create man through itself: referring to others in morality, I at the same time refer to myself, deciding who I will be. This existential weight of freedom – responsibility for oneself before others, God, and oneself – is the price of freedom which, let us add, is inevitable. Even the rejection of freedom or resignation from it is still an expression of freedom. For Wojtyła, freedom shapes man through an instinc-tive, completely not induced reference to the truth about values. However, in this reference and in acting the subject is dependent only on itself. The weight of the action, whose truth we decipher in our conscience, is what most impresses Wojtyła. The lack of a need for the concept of grace in this vision is striking. The subject is independent, autonomous, and thus responsible. With regards to this point, Wojtyła is close to Thomism and its respect for the innateness of creation.For Tischner, freedom is key and is also an innate value that is experienced. Its essence is above all freeing, liberating; thus Tischner does not hesitate to discern it in extra-moral phenomena such as dancing, which liberates beauty, and even extra-human ones, such as the beauty of an elk jumping across a brook. Beauty and good require freedom. However, the true liberation of freedom is opening oneself to the freedom of the other in encoun-ter, as thanks to this I can enter into a relationship of love and fidelity; I can s a c r i f i c e myself and through this fulfill the highest act of freedom, an act that is not induced by pre-established responsibility for the other, as in Levinas’ philosophy. Although it is not induced, this act assumes in an essential way the relationality of the subject and at the same time its finiteness.

Alfonso Iadonisi at the University of Naples Federico II developed (Eur. J. Org. Chem. 2013, 3137) a procedure for the selective acetolysis of the perbenzylated sugar 1 to furnish 3 using isopropenyl acetate (2) instead of the more typical and high-boiling acetic anhydride. The (3,4-dimethoxylphenyl)benzyl (DMPBn) protecting group, which is removed (cf. 4 → 5) under acidic conditions in the presence of the cation scavenger 5, was developed (J. Org. Chem. 2013, 78, 5264) by David S. Larsen at the University of Otago as an alternative to the p-methoxybenzyl (PMB) group. Another new hydroxyl-protecting group, the AzDMB group, which can be installed by simple acylation of (7 + 8 → 9) and removed under reductive conditions, was developed by Gijsbert A. van der Marel and Jeroen D.C. Codée of Leiden University. Stefan Grimme at the University of Bonn and Armido Studer at Westfälische-Wilhelms-Universität Münster found (Chem. Sci. 2013, 4, 2177) that NHC precatalyst 11 in the presence of NaH, benzaldehyde, and the oxidant 12 allows for the selective O-acylation of aminoalcohol 10 to 13. The reductive deprotection of benzyl carbamate 14 using the strong organic reductant 15 under photolytic conditions was achieved (Angew. Chem. Int. Ed. 2013, 52, 2239) by John A. Murphy at the University of Strathclyde. Liang-Qiu Lu and Wen-Jing Xiao at Central China Normal University found (Chem. Asian J. 2013, 8, 1090) that mixed imide 17 could be detosylated under visible light photoredox catalysis in the presence of Hantzsch ester 18. Frank Glorius at Westfälische-Wilhelms-Universität Münster developed (Org. Lett. 2013, 15, 1776) a ruthenium-catalyzed procedure for the N-formylation of amine 20 using methanol as the source of the formyl group. Protection of the thymine derivative 22 with a 2-(methoxycarbonyl)ethenyl (MocVinyl) group to produce 23 was developed (J. Org. Chem. 2013, 78, 5832) by Jaume Vilarrasa at the University of Barcelona. Deprotection of the MocVinyl group is readily achieved by treatment with a nucleophilic reagent such as pyrrolidine. Robert H. Grubbs at Caltech demonstrated (Chem. Sci. 2013, 4, 1640) that ether 24 could be demethylated with triethylsilane and potassium t-butoxide at high temperatures.

Critically ill patients have been described to have blood coagulation abnormalities that predispose to bleeding and thrombosis.We have studied plasminogen activators (fibrin plate, enzyme-linked Immunosorbent assay using polyclonalantibodies for t-PA), X-oligomers fibrin fragments (using monoclonal antibodies in an enzyme-linked immunosorbent assay), octant i pi asmin, antithrombtai III and fibronectin (Laurel1 innaunoeleetrophoretic technique), fibrinogen (thrombin timeassay), plateLets count, kaolln-cephalin clotting time and prothrombin time on admission to the intensive care unit and sequentially after 24 and 48 hours in 39 adult patients: Adult respiratory distress syndrome (ARDS) (n:6), Trauma (n:12), Sepsis (n:8), and Miscellanea (n:13).A decrease in tissue plasminogen activator (ng/ml)(p<0.001, p<0.05, p<0.01, p<0.05, respectively in the four groups), associated to an increase in the earliest form of cross-linked fibrin degradation products, X-Oligomers concentration (ng/ml) (p<0.01), indicatethat fibrindeposition and fibrinolytic exhaustion is a widespread situation in the ICU patients. Fibronectin was significantly reduced (p<0.001) in ARDS and Sepsis patients, low fibronectin levels were related to prognosis (p<0.01).These findings suggest.that critically ill patients, must be evaluated in respect to fibrinolysis and supported when necessary with prophylactic treatment.

Abstract:
Introdução: Toxoplasmose é uma parasitose de ampla distribuição causada pelo Toxoplasma gondii. Esta zoonose pode causar abortos em várias espécies, além de desencadear alterações neurológicas e oculares, porém sabe-se pouco sobre seus efeitos nos órgãos da reprodução. Objetivo: Investigar o impacto da infecção crônica pelo T. gondii sobre a quantificação dos folículos ovarianos em camundongos adultos. Material e métodos: Camundongos c C57BL/6 fêmeas foram separadas nos grupos controle (GC, n=8) e infectado (GI, n=8). Enquanto o GC recebeu salina, o GI recebeu 1000 oocistos por gavagem para indução da infecção experimental (protocolo aprovado pela CEUA/UEM sob nº 4092040517). Após 60 dias houve análise do ciclo estral dos animais pelo smear vaginal e foram eutanasiados aqueles que se encontravam em estro, fase caracterizada pela presença de células epiteliais queratinizadas. Após o aprofundamento anestésico com vapor de isoflurano os eutanasiados tiveram os ovários dissecados, pesados, processados e incluídos em parafina. Cortes de 5µm foram corados em Hematoxilina-eosina para análise da morfologia geral da gônada e quantificação dos folículos ovarianos. Os resultados foram submetidos ao teste estatístico t student e considerados significativos quando p<0,05. Resultados: A infecção crônica pelo T. gondii não afetou o peso corpóreo ou do ovário dos animais. A gônada de ambos os grupos apresentou uma organização similar, contendo região cortical constituída por epitélio germinativo, túnica albugínea, tecido conjuntivo e folículos ovarianos em diferentes estágios de desenvolvimento. A região medular apresentou-se organizada em tecido conjuntivo contendo feixe vásculo-nervoso glandular. A análise quantitativa evidenciou um aumento em 32% no número de folículos totais na cortical ovariana do GI em relação ao GC (p<0,001). Sabe-se que o T. gondii é capaz de reduzir significativamente o FSH e o LH tanto em homens quanto mulheres. Assim, uma hipótese que justifica o resultado encontrado é de que o parasito foi capaz de promover desregulação hormonal e assim afetar a maturação folicular ovariana e a ovulação. Conclusão: A infecção crônica pelo T. gondii afetou a quantidade de folículos presentes no córtex ovariano, demonstrando que a infecção pelo parasito pode afetar os estágios mais precoces da reprodução em camundongos C57BL/6 fêmeas.

Abstract:
We are characterizing the specificity of the polyanion-binding domain of the heparin/dermatan sulfate-dependent plasma protease inhibitor, heparin cofactor II (HCII). Various phosphate-containing polyanions accelerate the HCII-catalyzed inhibition of thrombin (T). Phosvitin, a phosphoprotein, enhances the HCII/T reaction at 25°C and pH 8.0 with the apparent second-order rate constant value (K2) increasing from 5 × 104 M−1 min−1 (in the absence of phosvitin) to 8 x 10' M”1 min as phosvitin increased from 0.05 to 30 ug/ml and then decreases as phosvitin is increased above 30 ug/ml. Apparent dissociation constant values for phosvitin-HCII and phosvitin-T are 450 nM and 10 nM, respectively. Polynucleotides accelerate the HCII/T reaction and have the following specificity (concentrations examined from 1-200 ug/ml): poly(guanylic acid) >> poly(adeny-lic acid, guanylic acid) > poly(inosinic acid) > poly(guanylic acid, uridylic acid) > poly(uridylic acid) = poly(adenylic acid) > poly(cytidylic acid). Polyphosphate anions (phosphate chain length, n, ranging from 5-100) enhance the HCII/T reaction. When compared at an equimolar phosphate concentration (1 mM), the rate was saturated at n = 50 with a maximum V.2 of about 5 × 107 M−1 min−1. Ca2+ (or Mg2+)-phosvitin/polyphosphate anion complexes and salmon protamine-polynucleotide complexes have lost the ability to enhance the HCII/T reaction. Phospho-pyridoxylated-HCII (lysine modified), with greatly reduced heparin cofactor activity, has lost its accelerating effect with phosvitin, polynucleotides and the polyphosphate anions. None of the above mentioned polyphosphate-containing compounds are effective at accelerating either the HCII-catalyzed inhibition of chymotrypsin or the antithrombin Ill-catalyzed T reaction. Our results suggest that (i) HCII is activated by the multiple negative charges of phosphate polyanions but they alone are not sufficient; (ii) the effective phosphate polyanions must also possess a specific conformation for maximum activity; and (iii) the phosphoserine-containing protein, phosvitin, can serve as a "template" for HCII/T.

Introdução: A diabetes mellitus tipo 1 (DMT1) é uma doença metabólica, caracterizada pela destruição das células beta pancreáticas, levando a hiperglicemia crônica. Nas glândulas salivares pode provocar danos teciduais e prejuízo na produção de saliva. Objetivo: Este estudo tem como objetivo investigar se o DM1 altera a morfologia das glândulas salivares parótida e sublingual e submandibular. Materiais e métodos: Foram distribuídos 12 ratos Wistar adultos (250g) nos grupos: controle ou NG (normoglicêmico, n=6) e diabético ou DM1 (n=6). O DM1 foi induzido por injeção endovenosa de estreptozootocina (55 mg/kg de peso corporal) sendo considerados diabéticos os animais com glicemia de jejum e pós-prandial ≥ 300 mg/dL. Os animais foram eutanasiados 30 dias após a indução. As glândulas salivares foram dissecadas, pesadas, desidratadas em etanol, diafanizadas em xilol e emblocadas em parafina. Cortes de 5um foram corados em hematoxilina e eosina para análise histopatológica. O método de Weibel foi utilizado para analisar a proporção entre os compartimentos glandulares. Os dados foram submetidos a análise estatística pelo teste t e foi considerado significativo valores de p<0,05. Resultado: Dentre os principais resultados encontrados, pode-se listar a perda de polaridade das células epiteliais, vacuolização celular e diferença de distribuição entre os compartimentos glandulares. Conclusão: Conclui-se que o DM-1 alterou a morfológicas dos acinos e ductos das glândulas salivares.

Abstract:
The increase in von Willebrand factor (vWF) following desmopressin (DDAVP) (1-desamino-8-D-arginine vasopressin) infusion was markedly blunted in severe hemophiliacs who had high vWF levels after treatment with vWF rich plasma concentrates. Diabetics with microangiopathy appear to have disease-induced elevation of vWF. In the current study, the vWF response to DDAVP infusion was measured in 30 diabetics (12 type I, 18 type II) and 16 controls, matched for age, sex and weight. Extent of nephropathy, macroangiopathy, and neuropathy was evaluated. Diabetic retinopathy was assessed by indirect ophthalmoscopy and fluorescein angiography (n=8 proliferative retinopathy, n=6 background retinopathy, n=16 no retinopathy). Plasma samples were collected in the supine, overnight-fasted state. DDAVP (0.3 μg/kg) was infused over 30 min and samples obtained at 0-60 min. vWF antigen was assayed by Laurell rocket electrophoresis. Tissue plasminogen activator (t-PA) activity was measured by a coupled chromogenic substrate assay. Results; Basal vWF levels for type I diabetics (124±16%, MeantSEM) and type II (178±14%) were increased as compared to controls (94±6%), p<.05 and p<.005, respectively. vWF levels for diabetics with proliferative retinopathy (194±29%) were significantly higher than for diabetics with background retinopathy (106±13%) p<.01. Diabetics with elevated basal levels of vWF (>150%) showed less of an increase in vWF following DDAVP infusion than diabetics with normal basal levels (p<.01). The percent increase in vWF following DDAVP administration inversely correlated with basal vWF levels (type I, r=.51; p<.01 and type II, r=.46; p<.01). The basal vWF level was the significant determinant of DDAVP response, not the presence or absence of diabetic complications. Peak t-PA levels showed no difference in controls or diabetics. In contrast to the vWF response, a normal t-PA response to DDAVP infusion was observed in diabetics. Conclusion: Diabetics with microvascular complications and high circulating levels of vWF demonstrate a blunted vWF response to DDAVP. This supports the existence of a negative feedback mechanism as previously reported for the transfused hemophiliacs.

A series of well-known tubular joints tested in UKSORP II have been re-evaluated using the mesh-insensitive structural stress method as a part of the on-going Battelle Structural Stress JIP efforts. In this report, the structural stress based analysis procedure is first presented for applications in tubular joints varying from simple T joints, double T Joints, YT joints with overlap and K joints with various internal stiffening configurations. The structural stress based SCFs are then compared with those obtained using traditional surface extrapolation based hot spot stress methods. Their abilities in effectively correlating the fatigue data collected from these tubular joints are demonstrated. These tests are also compared with the T curve typically used for fatigue design of tubular joints as well as the structural stress based master S-N curve adopted by ASME Section VIII Div 2. Finally, some of the implications on fracture mechanics based remaining life assessment for tubular joints are discussed in light of the results obtained in this investigation.
