In the late medieval N-Town Cycle Passion Plays, the trial of Jesus is presented in the context of a medieval courtroom, where Jesus is brought by Annas and Cayphas to be judged by Pontius Pilate. However, while the priests through abuses of the legal procedures attempt to ensure Jesus’s demise, Pilate opposes their intent by remaining true to his judicial duties, which presents him as a lonely Other in the presented world of legal misconduct. This paper explores the concept of otherness in a legal context of the plays, as well as the legal and social significance of Pilate’s actions.

Abstract:
Abstract Towns are of particular interest to geography which examines them in various objective and theoretical aspects. The concept of function is associated with the role which a particular town plays in the area, and this function is the entirety of socio-economic activity carried out in the town. The concept of the economic base of towns, which is used in this paper, distinguishes two groups of town inhabitants, namely such whose work directly contributes to the development of the town, the so-called primary builders (exogenous), and secondary builders (endogenous) who support the first group. This article presents the results of the study of changes in the function of 30 towns in the Silesian voivodeship in which coal mining is still carried out or has ended (18 mining towns and 12 post-mining towns). These towns have different sizes: small ones (up to 20,000 inhabitants; n=6), medium (20 to 100 thousand inhabitants; n=15), and large (with population of over 100,000 inhabitants; n=9). The study was conducted with the use of the indirect measurement of economic base method based on the location ratio, but in a modified form - i.e. the employee surplus rate. The analysis of functional changes in the mining towns of the Silesian voivodeship was performed in five aspects, in relation to: 1) the opening of the economy indicator, with the use of data concerning employment in the exogenous and endogenous group; 2) the employee surplus rate, determining the functional type of towns according to the dominant PKD [Polish Classification of Business Activities] section on the basis of the exogenous group; 3) the structure of exogenous functions of towns; 4) change of the exogenous function of towns; 5) the employee surplus rate, determining the share of section C (mining) in the exogenous function of towns. The analysis showed that in the period of 1996-2009 there has been a change in the functional type, from industrial to service type, in 8 towns; however, mining is still the primary branch of business activity in 11 towns studied, especially in small ones. Today, many service-based towns specialise in trade (n=7), and a small group of towns specialises in non-market services (n=4), which shows that the process of changes in this respect is still ongoing and the towns studied cannot be regarded as towns having a substantial share of higher-order services.

Abstract:
The Town Energy Budget (TEB) model coupled with the Regional Atmospheric Modeling System (RAMS) is applied to simulate the Urban Heat Island (UHI) phenomenon in the metropolitan area of Beijing. This new model with complex and detailed surface conditions, called TEB-RAMS, is from Colorado State University (CSU) and the ASTER division of Mission Research Corporation. The spatial-temporal distributions of daily mean 2 m air temperature are simulated by TEB-RAMS during the period from 0000 UTC 01 to 0000 UTC 02 July 2003 over the area of 116°E~116.8°E, 39.6°N~40.2°N in Beijing. The TEB-RAMS was run with four levels of two-way nested grids, and the finest grid is at 1 km grid increment. An Anthropogenic Heat (AH) source is introduced into TEB-RAMS. A comparison between the Land Ecosystem-Atmosphere Feedback model (LEAF) and the detailed TEB parameterization scheme is presented. The daily variations and spatial distribution of the 2 m air temperature agree well with the observations of the Beijing area. The daily mean 2 m air temperature simulated by TEB-RAMS with the AH source is 0.6 K higher than that without specifying TEB and AH over the metropolitan area of Beijing. The presence of urban underlying surfaces plays an important role in the UHI formation. The geometric morphology of an urban area characterized by road, roof, and wall also seems to have notable effects on the UHI intensity. Furthermore, the land-use dataset from USGS is replaced in the model by a new land-use map for the year 2010 which is produced by the Institute of Remote Sensing and Digital Earth (RADI), Chinese Academy of Sciences (CAS). The simulated regional mean 2 m air temperature is 0.68 K higher from 01 to 02 July 2003 with the new land cover map.

Housing has always been a necessity in the lives of modern man. Anyone who was already working needs to own at least some form of housing. Development of a city plays a part in determining the prices of the housing sold whereas in the central and business district houses can be very expensive, the one’s at the outskirt of the town can be cheaper or it can be which will be shown in this research. The government concerned offered developers a project which is known as Subsidized Housing, a house which is cheap with lots of benefits to encourage low income buyers. The industrial sector is known for its High Workforce which implies that a lot of manpower is involved, this creates ample market opportunities. Why don’t we see a lot of subsidized housing? This concerns the fact that not all houses built for this manner yields enough profit to entice developers to actually build them. Thus the research for counting the actual benefit must be done in order to convince developers that there is an acceptable return to be gained when developing Subsidized Housing. The analysis conducted in this research include location analysis, site analysis, legality analysis, comparative object study analysis, preference analysis and financial analysis. The analytical tool used is the analysis of proximity, Discounted Cashflow, Sensitivity Analysis, descriptive and quantitative. The results of this study are the amount of returns obtained by the developer in n years, the length of the return and the percentage of profits. Keywords: building material; developer’s profit; industrial district; subsidized housing ABSTRAKRumah sudah menjadi sesuatu yang wajar dimiliki oleh masyarakat di berbagai kalangan, rumah tidak hanya berperan dalam memberikan perlindungan terhadap cuaca tetapi juga sebagai tempat untuk keluarga berkumpul dan menikmati waktu bersama dengan keluarga maupun orang lain. Tentu tidak semua orang bisa memiliki rumah dengan berbagai alasan, oleh sebab itu pemerintah melakukan intervensi dalam bentuk penyediaan subsidi. Namun mengapa tidak banyak perumahan subsidi dikarenakan pihak pengembang hanya melihat secara singkat seberapa besar pengembalian yang didapat pengembang tingkat pengembalian tersebut yang menentukan apakah pengembang mau atau tidak mengembangkan perumahan subsidi. Dengan pertanyaan tersebut dilakukanlah penelitian pengembangan perumahan perumahan subsidi untuk menunjukkan tingakt keuntungan yang akan didapatkan seorang pengembang beserta lamanya pengembalian modal. Analisis yang dilakukan yakni analisis lokasi, analisis tapak, analisis legalitan, analisis objek studi banding, analisis preferensi dan analisis keuangan. Alat analisis yang digunakan adalah analisis proximitas, Discounted Cashflow, Sensitivity Analysis, deskriptif dan kuatitatif. Hasil dari penelitian ini adalah besarnya pengembalian yang didapatkan pengembang pada tahun n, lama pengembaliannya dan persentase keuntungannya.

Abstract:
Following the results of a sociological study conducted in 2018 by means of questionnaire method with a probability quota sample (N=420), this article analyzes the various interpretations of patriotism according to public opinion in the city of Saratov. It has been established that, according to its inhabitants, the word “Homeland” most often implies Russia, “small homeland” – one’s own city or family (less often one’s home neighborhood). The meaningful interpretation of this term is associated with socially significant activity, activism, and social responsibility. Basic displays of patriotism, according to respondents, are considered to be army service, patriotic upbringing of one’s children, abidance to the law and diligent work for society’s benefit. Most of Saratov’s inhabitants consider themselves to be patriots to one degree or another. However, what’s concerning is the fact that one out of every three city residents – especially among youths – denies such values. Youth patriotism is more oriented towards love for Russia. The elder generation builds its patriotic sentiments around love for their families, close ones, their home town and the USSR. Meanwhile most citizens (albeit to various degrees) draw a distinction between the terms “patriotism” and “nationalism”. The latter bears more of a negative connotation. Generally citizens (especially among the elder generation) believe that, after the USSR’s downfall, the teaching of patriotic values has “degenerated”, despite the government and the president taking certain steps towards increasing the level of patriotism within society. It is during such periods of change, like we are witnessing today, when the values of preceding generations start to contradict reality, and when the younger generation assumes the role of bearer of new knowledge and aspirations, due to possessing a more acute sense of society’s major issues and the will to openly speak out. Thereby not only the elder generation, but also the younger generation plays an equal role as a subject of developing society’s patriotic values. However, the profound differences between generations are proportional to the dynamics and scale of change within society, and the degree of mutual influence between generations acts as one of the major factors for stabilizing society, which includes establishing patriotic values. Increasing the efficiency of practices in this area, together with more extensive coverage of such practices in mass media and the internet, would aid in the development of historical and socio-cultural memory.

Introduction and statement of the problem. Before the revolution of 1917, artists, writers, musicians and teachers created a rich literary fund that could be the basis for professional musical children’s theater in Ukraine. That is why there is a need to study the children’s musical and dramatic heritage of the past, which is an inexhaustible treasury of cultural and educational ideas that can be creatively developed and successfully applied in modern conditions. The process of creative development of the genre of children’s musical performance is today one of the most pressing problems of professional theater for children, take in account its growing popularity, both in the world and in Ukrainian musical culture. The lack of scientific research that fully and comprehensively cover the scientific and practical significance of musical children’s plays by Marko Kropyvnytskyi for the development of musical children’s theater in Ukraine indicates the need for more in-depth researching of the chosen topic. In our research, we rely on the works and articles of authoritative experts – in particular, I. Franko (1910), M. Voronyi (1913), D. Antonovich (1925), P. Rulin (1929), I. Mar’ianenko (1953), P. Kyrychok (1985), N. Yosipenko (1958), P. Perepelitsa (1956), A. Novikov (2007; 2011), L. Moroz (1990). The vast majority of researchers noted the great merits of the artist to the national drama in particular and Ukrainian culture in general. Among the scientific works devoted to Kropyvnytskyi as a children’s playwright, one can distinguish the research by A. Novikov (2007), which focuses on the history of creation of the first children’s troupe in the country, which had no analogues in the history of the world theater, since the actors in it were peasant children. In mentioned critical and scientific works, the innovative features of the playwright’s creative heritage are outlined, attention is focused on the specifics of the genre and problem-thematic range, literary-aesthetic, socio-political, and pedagogical views. The literary and theatrical activity of M. Kropyvnytskyi has been thoroughly studied. However, there is still no work that comprehensively reveal his musical and dramatic creativity for children. The purpose of the article is to show the significant role of M. Kropyvnytskyi in the development of children’s musical theater in Ukraine based on the research of children’s musical and dramatic creativity by the artist. The research methodology is integrative. The work uses knowledge of various fields of art history and related sciences: history and theory of theater, music theory, music and theater psychology, vocal and theater pedagogy. Presentation of the main material. A great pride of the playwright is the foundation by him on the territory of his village Zatyshok of the children’s theater, “actors” in which were his own and peasant children. This event was and remains unprecedented, since nothing like this has been observed in the history of Ukrainian and European culture. The troupe consisted of peasant children aged 10–13. For performances, Kropyvnytskyi assigned the largest room (hall) in the old house, where, as in a real theater, the stage was equipped. The first performance, “Goat-Dereza” (“Koza-dereza”), took place on Christmas day, 1906. The playwright drew the scenery himself, and prepared the costumes together with the children. The play was a great success. A few days later, the children’s troupe was invited to a “tour” in the neighboring village, and the entire theater with the scenery on five carts went on a journey of six versts (Novikov, 2007: 33). In the children’s repertoire at that time, there was, in fact, only one work – the opera by M. Lysenko “Goat-Dereza” (“Koza-dereza”) (libretto by Dniprova Chaika). Ukrainian children’s repertoire did not exist at that time, and in 1907, Kropyvnytskyi created two plays for young performers based on folk tales – “Ivasyk-Telesyk” and “At the behest of the pike” (“Po shchuchomu velinniu”). The performances included vocal numbers composed by M. Kropyvnytskyi on the themes of Ukrainian folk melodies. In a letter to his good friend entrepreneur A. Suslov in January 1908, the writer, in particular, notes: “I have assembled a troupe of peasant children and I am staging in the villages: Goat-dereza, IvasykTelesyk, and At the behest of the pike (the latter both are my)” (Kropyvnytskyi, 1960: 530). Based on the plot of folk tales of the same name, he wrote original musical and dramatic works for children of great educational value. The plays are quite simple in meaning and clearly depict the images of all the negative and positive characters. The first represent such social vices as lies and insincerity, and the second are carriers of eternal positive qualities – sincerity, candor, hard work. The plays are written in an exquisite Ukrainian language, close to the oral poetic creativity. All this, as M. Yosypenko rightly notes, is evidence of “a serious approach of M. Kropyvnytskyi to the business of writing plays for children, a deep knowledge of the psychology of the young audience and its cultural and educational needs and demands” (Yosypenko, 1958: 265). The performances require participation of music, which organic include into the language range of the play itself. Music explains and complements the true meaning of the situation to the young audience. Ukrainian musical folklore material formed the basis of the musical solution of M. Kropyvnytskyi’s children’s performances. Most often, folk songs served as a means to create the image and were introduce before the dramatic action mainly by the method of self-presentation: performing a particular song, the characters showed certain traits of their nature. The songs help to reveal the inner world of the characters, to express their state of mind and moods; often they contributed to the creation of the necessary stage atmosphere: festivities, fun and jokes. A significant part of the characters could not be imagined without songs. Using some folk melodies, Kropyvnytskyi mainly wrote original music, close in melody to the folk-song sources. Solo numbers, ensembles, and choirs are organically woven into the dramaturgy of these plays. A clear reflection of the integrity and unity of the musical and dramatic process is the principle of end-to-end development of the main musical idea of performances. In preparation for productions of his children’s plays, Kropyvnytskyi wrote an orchestration for them also. Intending to put these plays on the professional stage, Kropyvnytskyi wrote down advice to future directors regarding the production of their children’s plays. He began to think of broader horizons for them. In the spring of 1910, small artists had to show their art to the audience of the neighboring county town Kupyansk. However, the premature death of the Ukrainian playwright did not allow this plan to be realized. The children’s troupe soon ceased to exist. Kropyvnytskyi children’s troupe and the repertoire he created for it became a prologue to the development of the Ukrainian theater’s creativity for young viewers. In nowadays from the repertoire do not go off the pearls of drama for children “Ivasik-Telesik’ and “At the behest of the pike”. Conclusions. Marko Kropyvnytskyi’s creative heritage and practical activities wrote the gold pages to the history of Ukrainian musical children’s drama and Ukrainian children’s theater. Children’s musical and dramatic works of the writer based on song folklore are the effective mean to educate positive attitude of young Ukrainians to folk tradition as well as to form positive nature traits: generosity, hospitality, goodwill, charity.

Abstract:
This article is an attempt to outline the difference between Breytenbach's poetic method and that of the Symbolists. Although it touches on aspects of the symbolist poetic method like the rich suggestiveness, the creation o f a meaningful alternative world (and the effort of doing this), it focuses mainly on Breytenbach’s use of metaphor to create an impossible alternative world in a poem, only to relativize and destroy it again in the end. This process is illustrated in an analysis of poem 8.1 from Lotus. This analysis also shows up five well-known cardinal traits of Breytenbach’s poetry, viz. its carnality, the universal analogy between body, cosmos and poetry and the great emphasis on journeys, discoveries and transformations by means of language. It is also claimed that the Zen-Buddhisi Void plays an analogous role in Breytenbach's poetry to the theory of correspondances in the Symbolists: it is a rich source of metaphor. Breytenbach's poetry shows a strong duality between the present world and a meaningful alternative sphere. Being in and of this alternative sphere only aggravates the poet’s isolation (a typically symbolist trait), making him literally and figuratively an exile, as exile poems like "tot siens, kaapstad" (see you again, cape town) and "Walvis in die berg" (Whale on the mountain) and, of course, his prison poetry, clearly show.

AbstractThe battle of Petropavlovsk in August/September 1854 was a significant though little known victory for the Russians during the Crimean War. Petropavlovsk, i n Kamchatka, was attacked by a Franco-British naval force intending to destroy Russian ships within the harbour and to render the port unusable as a naval base. Allied plans were disrupted by the death ofthe commander in chief, the British Admiral Price, just before the action was joined. A bombardment on 31 August badly damaged the harbour defences and, if followed up, would probably have resulted in success. After a gap of three days, the allies mounted a landing in the rear of the town which was, after severefighting, repelled by the Russians. After this defeat, the squadron dispersed t o ports on the other side of the Pacific.

Technology-based enterprises play a paramount role in blooming a country economically. Nevertheless, according to a society’s capacity to launch such enterprises in various eras, their volume is less than expected in many economies. Therefore, establishing such enterprises is necessary for developing any country, although its innovation system contributes to establishing them. This paper considers the impact of entrepreneurial education on technology-based enterprise development, including motivation as a mediator variable, in Esfahan Scientific and Industrial Town. Despite much research investigating the correlation between entrepreneurial education and technology-based enterprises’ progress, it seems that no study has already considered this correlation with remarking the motivation as a mediator variable. This applied research follows a quantitative research design. The statistical population includes 500 enterprises in the Esfahan Scientific and Industrial Town, and for sampling, Cochran’s formula was applied (n = 217). Additionally, the researcher-made questionnaire and PLS3 software were used for data gathering and analysis. The results demonstrated that entrepreneurial education elements (including entrepreneurial skill, entrepreneurial learning, and entrepreneurial intention) positively affect technology-based enterprises’ development, considering motivation as a mediator variable. However, the impact of entrepreneurial intention on technology-based enterprises was not supported. It reveals that the entrepreneurial intention of motivated individuals could have a meaningful effect on the development of technology-based enterprises. Therefore, motivation is a critical issue to be considered by managers and policymakers while considering entrepreneurial education-related policies and initiatives.

AbstractAlthough Margery Kempe (1373-1440) probably never had regular contact with a Jewish person, she depicts the Jews as symbols of evil in her autobiographical Book. Despite the rebellious nature Kempe displays to patriarchal authorities in the church by refusing to behave as a nun, to wear black, or to remain in a cloister, she is entirely orthodox in portraying the Jews as Christkillers in her vision of the Crucifixion. This anti-Judaic ideology appears in the Gospels and in official doctrine, beginning with Augustine, continuing with Pope Gregory I the Great, and worsening with Aquinas and the Franciscan and Dominican friars in the lourteenth century. Medieval drama, especially the N-Town Passion Play (which Kempe may have seen) provides a striking analogue tor Kempe's vision at Calvary. Kempe, extraordinary in some ways, is entirely conventional in her dratnatiration of the Jewish soldiers as dcicides.

Evacuation plans are critical in case of natural disaster to save people’s lives. The priority of population evacuation on coastal areas could be useful to reduce the death toll in case of tsunami hazard. In this study, the population density remote sensing mapping approach was developed using population records in 2013 and Defense Meteorological Satellite Program’s Operational Linescan System (DMSP/OLS) night-time light (NTL) image of the same year for defining the coastal densest resident areas in Pearl River Estuary (PRE), China. Two pixel-based saturation correction methods were evaluated for application of population density mapping to enhance DMSP/OLS NTL image. The Vegetation Adjusted NTL Urban Index (VANUI) correction method (R2 (original/corrected): 0.504, Std. error: 0.0069) was found to be the better-fit correction method of NTL image saturation for the study area compared to Human Settlement Index (HSI) correction method (R2 (original/corrected): 0.219, Std. error: 0.1676). The study also gained a better dynamic range of HSI correction (0~25 vs. 0.1~5.07) compared to the previous one [27]. The town-level’s population NTL simulation model is built (R2 = 0.43, N = 47) for the first time in PRE with mean relative error (MSE) of 32% (N = 24, town level), On the other side, the tsunami hazard map was produced based on numerical modeling of potential tsunami wave height and velocity, combining with the river net system, elevation, slope, and vegetation cover factors. Both results were combined to produce an evacuation map in PRE. The simulation of tsunami exposure on density of population showed that the highest evacuation priority was found to be in most of Zhuhai city area and the coastal area of Shenzhen City under wave height of nine meters, while lowest evacuation priority was defined in Panyu and Nansha Districts of Guangzhou City, eastern and western parts of Zhongshan City, and northeast and northwest parts of Dongguan City. The method of tsunami risk simulation and the result of mapped tsunami exposure are of significance for direction to tsunami disaster-risk reduction or evacuation traffic arrangement in PRE or other coastal areas in the world.

- The complex regulations for general plans which the Lombard Regional Law No. 12/2005 defines, seem suited above all to large urban and metropolitan realities. Planning in Lombardy therefore puts small municipalities, who often find difficulty in applying that law, to the test. Two cases of small municipalities illustrate the use they have made of Regional Law n. 12, the difficulties they encountered and the results achieved: Cassinetta di Lugagnano in the Province of Milan along the Naviglio Grande canal, with a town centre of great historical architectural value and Vaiano Cremasco in the Province of Cremona, where the consolidated agricultural land use and an attractive landscape sit alongside an active and heavily industrialised district. In both cases the general plan places the accent on respect for the environment, on safeguarding unbuilt land and on the need for urban planning to take back its role in directing and co-ordinating the environment. The main fundamental principle is that of the least consumption of land possible which is taken to the utmost in the case of Cassinetta di Lugagnano, where a principle of ‘zero growth' has been adopted for the municipality's urban planning policies.

Abstract Sickle cell disease (SCD) is characterized by hemolytic anemia in association with acute and chronic life-threatening clinical complications. Acute vaso-occlusive crisis (VOCs) are the main cause of hospitalization for SCD patients. In VOCs, amplified inflammatory response plays a key role in acute organ damage. Pro-Resolving lipid mediators such as resolvins (Rv) accelerate resolution of acute inflammation in different models, indicating that stimulation of endogenous resolution of inflammatory processes may be an additional strategy in limiting tissue damage. Recent data suggest beneficial effects of Rvs in hypoxia/reoxygenation (H/R) related tissue injury. Here, we study the effects of Rvs on a model of acute VOCs using humanized SCD mice (Hbatm1(HBA)Tow Hbbtm2(HBG1,HBB*)Tow). We treated SCD and control healthy mice (AA, Hbatm1(HBA)Tow Hbbtm3(HBG1,HBB)Tow) (n =6-7 animals in each group) with RvD1, 7S, 8R, 17S- trihydroxy-docosa-4Z, 9E, 11E, 13Z, 15E, 19Z-hexaenoic acid, 17R- RvD1, 7S, 8R, 17R- trihydroxy-docosa-4Z, 9E, 11E, 13Z, 15E, 19Z-hexaenoic acid; RvD2, 7S, 16R, 17S-trihydroxy-docosa-4Z, 8E, 10Z, 12E, 14E, 19Z-hexaenoic acid. Mice were treated with RvD1 and RvD2 at the dose of 2.5 ug/Kg by gavage 1 hour (hr) before H/R stress (10 hrs 8% oxygen followed by 3 hrs reoxygenation), which we have used in the past to mimic acute VOCs. We found that RvD1 and RvD2 significantly reduced the H/R-induced (i) increase in neutrophil count; (ii) lung inflammatory cell infiltrate, mucus and thrombi formation; (iii) glomerular inflammatory cell infiltration, glomerular sclerosis and thrombi formation. In the lung of H/R SCD mice, RvD1 prevented the H/R induced up-regulation of (i) cytokines such as MCP2, IL-6 and ET-1; (ii) vascular endothelial activation markers (VCAM-1 and ICAM-1; (iii) cytoprotective systems such as Prx6 and HO-1. In the kidney of H/R SCD mice, RvD1 significantly reduced H/R induced expression of IL-6 and ET-1 as well as HO-1. Our data indicate that RvD1 and RvD2 modulating inflammatory responses related to H/R in SCD, protect sickle cell target organs, and foster resolution. The Cai stream is a wastewater receiving stream from industrial zones in Tan Uyen town (Binh Duong province), which is facing to pollution risks, requiring appropriate management measures, especially in the context of socio-economic development. Accordingly, the study aimed to assess the current status and to forecast its water quality until 2020. By sampling, analysis, and water quality index (WQI), the results show that water quality in dry season of 2017 ranged from poor to medium level (WQI 23-79) and tended to increase towards the downstream. Some streams being concerns in terms of water quality were the Cho stream (WQI 18 - 31), the Cai stream flowing through VSIP II (WQI 26 - 35), the Tre stream flowing through waste water treatment plants of VSIP II (WQI = 40)... It could be explained by high turbidity, TSS and NH4-N concentrations. Water quality in 2020 was simulated by the Mike 11 model and demonstrated on maps using GIS. Simulation results show an increase in COD, BOD, TSS, total nitrogen, and total phosphorus concentrations in relation to local socio-economic development plans, especially COD and TSS (the highest concentrations would be 48 mg/L and 205 mg/L, respectively). In some areas, there is a clear increase in pollution levels, such as the Tre stream, the confluence of the Tre and the Cai streams, downstream of the Cai stream, the Vinh Lai stream, and the Cho stream. Research results could be an important basis for environmental management in general, water quality management in particular, contributing to sustainable development goals of the local.

Clinical, behavioural and structural interventions have each reduced these risks and improved health outcomes. However, combinations of these interventions have not been compared with each other or with no intervention at all. The ‘Girl Power’ study is designed to systematically make these comparisons.Methods and analysisFour comparable health facilities in Malawi and South Africa (n=8) were selected and assigned to one of the following models of care: (1)Standard of care: AGYW can receive family planning, HIV testing and counselling (HTC), and sexually transmitted infection (STI) syndromic management in three separate locations with three separate queues with the general population. No youth-friendly spaces, clinical modifications or trainings are offered, (2)Youth-Friendly Health Services (YFHS): AGYW are meant to receive integrated family planning, HTC and STI services in dedicated youth spaces with youth-friendly modifications and providers trained in YFHS, (3)YFHS+behavioural intervention (BI): In addition to YFHS, AGYW can attend 12 monthly theory-driven, facilitator-led, interactive sessions on health, finance and relationships, (4)YFHS+BI+conditional cash transfer (CCT): in addition to YFHS and BI, AGYW receive up to 12 CCTs conditional on monthly BI session attendance.At each clinic, 250 AGYW 15–24 years old (n=2000 total) will be consented, enrolled and followed for 1 year. Each participant will complete a behavioural survey at enrolment, 6 months and 12 months . All clinical, behavioural and CCT services will be captured. Outcomes of interest include uptake of each package element and reduction in HIV risk behaviours. One in five people in the world are said to have some type of disability. Disability is not merely individuals’ compromised capability in navigating the built environment, but rather the ‘misfit’ of capabilities with how a given living environment is organized. Planning, therefore, has a crucial role to play in responding to the needs of this significant population through changes to the built and social environment. However, discussion on planning theories and practices with a focus on persons with disability (PWD) has been limited to more specific realms of ‘design,’ and precariously absent in broader planning research. This systematic literature review aims to inform potential directions for planning scholarship by exploring the current and historic planning research investigating the needs of PWD. We compiled relevant papers from five prominent English language planning journals, some of which are long-standing (<em>Town Planning Review</em>, 1910–, <em>Journal of the American Planning Association</em>, 1935–). A very limited number of papers (n = 36) on topics related to PWD of any type have been published in the five journals throughout their existence, with even fewer focusing on the population. The subareas of planning these papers addressed include housing, transportation, land use, policy, and urban design. Many papers called for participation by PWD in the planning and decision-making processes, and some recent papers advocated for the production of evidence related to costs of creating accessible infrastructure. A critical look on some disciplinary divides and enhanced roles of planning research would be beneficial.

According to previous research, participation in exercise training (ET) by South Korean firefighters varies with shift type, and the effect of shift type is greater in large cities than in small towns. However, shift types differ among regions, depending on the number of dispatches. Therefore, the present study examined the impact of the number of fire dispatches and other situational factors on ET. A series of multilevel logistic regression analysis was applied to analyze the data collected from South Korean firefighters (N = 5219) in 2017. According to the firefighters, participation in ET is higher among those who have someone to instruct their ET (Coefficient (Coef) = 0.057, SE = 0.017, p < 0.001) and who can do ET while on duty (Coef = 0.048, SE = 0.014, p < 0.001). The number of fire dispatches had a significant effect on participation in ET (Coef = −0.000, SE = 0.000, p < 0.01), meaning that the firefighters’ participation in ET varies with the number of fire dispatches in each region. Our main findings indicate that the number of fire dispatches is a key factor affecting ET participation among firefighters, and the other situational factors also play a role. Therefore, ET programs that firefighters can participate in between calls should be established.

The aim of this paper is to examine the organizational background of the protests ?Against dictatorship? that took place in several Serbian towns, in the spring of 2017. The absence of the official organizers and the role social networking sites played in terms of communication, organization and coordination of the protest events, raises the following question: Could the protest ?Against dictatorship? be considered as an example of a digitally enabled ?connective? action? According to the ?connective action? approach (Bennett & Segerberg, 2012, 2013), in the information society a new ?connective? form of collective action is emerging as a result of personalization of political action that occurs within new sociotechnological environment. Connective action is enabled by innovative technological opportunities for individual participation in collective ventures and supported by the role that ICTs play as ?stitching? and organizing agents of collective action. In such circumstances, conventional social movement organizations and solid collective identities seem to become far less necessary. The analysis of the data collected in the field (N = 175) and in the online survey of the experiences and attitudes towards the protest (N=225), leads to the conclusion that the organizational background of the protest ?Against dictatorship? is closest to the ideal-type of ?self-organized network?, as one of the two basic models of connective action. In the concluding part of this paper, the authors argue that the ?connective action? model is adequate for the interpretation of the emergence of the digitally mediated protests. However, with the transformation of protests into more solid and stable forms of collective action, this model loses its explanatory power.

Abstract. In order to determine whether the geographical distribution of autoimmune thyroid disease in Britain is influenced by the pattern of iodine intake, the prevalence of subclinical disease (detectable antithyroid antibodies in biochemically euthyroid individuals) has been measured in female blood donors from seven towns in England and Wales previously characterised in terms of past and present iodine intake. Thyroglobulin antibody and thyroid peroxidase antibody were measured by highly sensitive assays which are based on the direct interaction between antibody and radiolabelled antigen. Excluding cases of overt thyroid disease (biochemically hypo- or hyperthyroid with thyroid antibodies), the overall prevalences of the antibodies in sera from the 698 female blood donors were 17.8% for thyroglobulin antibody and 17.8% for thyroid peroxidase antibody. Both antibodies were found in 12.3% of the female blood donors. In contrast, the prevalences of thyroglobulin antibody and thyroid peroxidase antibody were 41 and 43%, respectively, in the 117 female relatives of 18 probands with autoimmune thyroid disease, but the highest prevalences were observed in groups of women patients with Graves' disease (N = 39) or Hashimoto's disease (N = 39) (51, and 97% for thyroglobulin antibody, respectively, and 72 and 97% for thyroid peroxidase antibody, respectively). Antibody prevalence increased with age in the female blood donors rising from 10.6% at age 18-24 to 30.3% at age 55-64 for thyroglobulin antibody and from 14.9% at age 18-24 to 24.2% at age 55-64 for thyroid peroxidase antibody. Geographical differences in the prevalences of both antibodies were not significant and did not correlate with either the previous goitre prevalence or with current differences in iodine intake. Consequently, it seems unlikely that environmental factors play a major role in the development of subclinical autoimmune thyroid disease in the geographical areas studied.

Greek cafés were a feature of Australian cities and country towns from the 1910s to the 1960s. Anglophile Australians, who knew the Greeks as dagos, were possessed of culinary imaginations that did not countenance the likes of olive oil, garlic, or lemon juice. As a result, Greek cafés catered to Australian tastes and became the social hubs of their communities. After establishing the diverse and evolving nature of food offered in Greek shops since their origins in the late nineteenth century – oyster saloons, cafés, fish shops, fruit shops, milk bars, snack bars, confectioneries – this article uses the concepts of “disgust” and “hunger” to offer new insights about food and identity in Australia’s Greek community and in the wider Australian culinary landscape. In particular, it applies Ghassan Hage’s work on nostalgia among Lebanese immigrants to the situation of Greek proprietors and reveals how memories of a lost homeland allowed café families to feel “at home” in Australia. In a land of “meat-n-three-veg,” a moussaka recipe the family had known for generations offered both a sense of identity and the comfort of familiarity, and Greek cafés, because they represented hope and opportunity, were familial spaces where feelings of nostalgia were affective building blocks with which Greeks engaged in homebuilding in a new land. And although their cafés did not serve Greek food, Greek proprietors and their families did eventually play a role in introducing the Australian palette to Mediterranean foods and foodways.

Abstract:
Rural primary care services have the potential to play a major role in reducing the gap in cardiovascular disease (CVD) outcomes between rural and metropolitan Australians, particularly in men at high risk of CVD. The aim of this study was to explore the self-reported behaviours and satisfaction with their general practice/practitioner of men at high risk of CVD, and attitudes of rural primary care clinicians regarding the role of primary care in CVD prevention. This observational research was addressed through survey questionnaires with rural men at high risk of CVD and semi-structured interviews with rural primary care clinicians. Fourteen rural primary care practices from towns with populations less than 25000 participated. One hundred and fifty-eight high-risk men completed the questionnaire. Their responses demonstrated poorly controlled risk factors despite a willingness to change. Alternatively, rural primary care clinicians (n=20) reported that patients were unlikely to change and that illness-based funding models inhibited cardiovascular preventive activities. Australians living in rural areas have worse CVD outcomes. In addition, there is a disparity in the assumptions of health providers and male patients at high risk of CVD in rural areas. This necessitates innovative rural primary care models that include a blended payment system that incentivises or funds preventive care alongside an emphasis on lifestyle advice, as well as an explicit strategy to influence clinician and patient behaviour to help address the disparity.

Background: One in 12 Australians is expected to develop bowel cancer by age 85. Over 90% of bowel cancers can be successfully treated if detected early yet participation rates in the National Bowel Cancer Screening Program (NBCSP) are currently only 39%. Research shows that general practitioner (GP) recommendation is a key predictor for bowel cancer screening uptake. However the current NBCSP invitation system does not involve GPs, making it difficult for them to know when a patient has received a test kit in the mail, and in turn play a role in screening behavior. Research suggests that GP point-of-care clinical prompts, encouraging GPs to discuss screening with their patients, can increase fecal occult blood testing. Such a ´reminder´ system is integral to other screening programs in Australia, however no such system exists for the NBCSP. The George Town Medical Centre located in rural Tasmania, has recently implemented a reminder prompt for bowel cancer screening as part of its standard clinical practice. This paper describes the protocol used to implement and evaluate this service. Aim: Our research aims to determine whether a clinical prompt delivered to GPs at point-of-care, can increase bowel cancer screening participation in patients attending a rural Tasmanian general practice. Secondary aims include assessing GP and patient attitudes toward the prompt and determining socio-demographic differences in the effect of the prompt on screening participation. Methods: The clinical prompt was implemented in January 2018. Active patients turning 50 (n=106), 60 (n=141) or 70 (n=103) in 2018 were eligible to receive the prompt. The prompt was created within the practice management software and associated with each eligible patient's file. GPs were provided with information on the NBCSP as well as resources to support their conversation with patients including a sample kit. The number of patients with whom the GP discussed screening and the number of patients who completed a kit will be collected after 12 months. Screening rates of patients eligible for the prompt will be compared with patients turning 54, 64 and 74 in 2018, controlling for potential covariates including age, gender and socioeconomic status. Interviews with all GPs (n=10) and a minimum of 10 patients will be conducted to gain an understanding of attitudes toward the prompt. Results: To date the prompt has been successfully integrated into the practice and GPs have begun encouraging patients to use their NBCSP kits. Attitudes of GPs and patients toward the prompt and preliminary data on kit usage will also be presented. Conclusion: A clinical prompt in general practice may be a simple and cost effective way to increase participation in the National Bowel Cancer Screening Program. The findings from this study will have implications for the program in terms of engaging GPs in the screening invitation process. The results will further be used to inform a pilot study in Tasmania.

ABSTRACT During human cytomegalovirus (HCMV) infection, the periphery of promyelocytic leukemia protein (PML)-associated nuclear bodies (also known as PML oncogenic domains [PODs] or ND10) are sites for both input viral genome deposition and immediate-early (IE) gene transcription. At very early times after infection, the IE1 protein localizes to and subsequently disrupts PODs, whereas the IE2 protein localizes within or adjacent to PODs. This process appears to be required for efficient viral gene expression and DNA replication. We have investigated the initiation of viral DNA replication compartment formation by studying the localization of viral IE proteins, DNA replication proteins, and the PML protein during productive infection. Localization of IE2 adjacent to PODs between 2 and 6 h after infection was confirmed by confocal microscopy of human fibroblasts (HF cells) infected with both wild-type HCMV(Towne) and with an IE1-deletion mutant HCMV(CR208) that fails to disrupt PODs. In HCMV(Towne)-infected HF cells at 24 to 48 h, IE2 also accumulated in newly formed viral DNA replication compartments containing the polymerase processivity factor (UL44), the single-stranded DNA binding protein (SSB; UL57), the UL112-113 accessory protein, and newly incorporated bromodeoxyuridine (BrdU). Double labeling of the HCMV(CR208)-infected HF cells demonstrated that formation of viral DNA replication compartments initiates within granular structures that bud from the periphery of some of the PODs and subsequently coalesce into larger structures that are flanked by PODs. In transient DNA transfection assays, both the N terminus (codons 136 to 290) and the C terminus (codons 379 to 579) of IE2 exon 5, but not the central region between them, were found to be necessary for both the punctate distribution of IE2 and its association with PODs. Like IE2, the UL112-113 accessory replication protein was also distributed in a POD-associated pattern in both DNA-transfected and virus-infected cells beginning at 6 h. Furthermore, when all six replication core machinery proteins (polymerase complex, SSB, and helicase-primase complex) were expressed together in the presence of UL112-113, they also accumulated at POD-associated sites, suggesting that the UL112-113 protein (but not IE2) may play a role in recruitment of viral replication fork proteins into the periphery of PODs. These results show that (i) subsequent to accumulating at the periphery of PODs, IE2 is incorporated together with the core proteins into viral DNA replication compartments that initiate from the periphery of PODs and then grow to fill the space between groups of PODs, and (ii) the UL112-113 protein appears to have a key role in assembling and recruiting the core replication machinery proteins in the initial stages of viral replication compartment formation.

ABSTRACTObjective: to understand adolescents' knowledge about the nulliparous breastfeeding practices. Method: fieldwork, qualitative study in the town of Mossoró (RN), Brazil. The proposal was sent to the Ethics in Research State University of Rio Grande do Norte/UERN being approved as opinion n. 064/09. We applied a roadmap of semi-structured interviews with 10 female adolescents, aged between 12 and 17 years and had never gotten pregnant. The data were recorded, transcribed and analyzed by constructing categories. Results: about breastfeeding, young women interviewed demonstrated that they are "Knowing little or nothing, "Knowing what the program guide, "Knowing through other sources”. Perceives the need for greater coordination between family, school, health services and society as a whole, to enable information on the matter to adolescents, so that in future, when they came to be mothers, may play the act breastfeeding satisfactorily. Conclusion: Despite adolescents understand the importance of breastfeeding, their knowledge about the topic are very superficial and impregnated with acquired concepts and socio-culturally constructed. Abstract Introduction Innate immune complement activation may contribute to sickle cell disease (SCD) pathogenesis. The alternative complement pathway is abnormally activated in SCD and is additionally activated by phosphatidylserine (PS) on the outer leaflet of SS-red blood cells (SS-RBC). PS on the surface of SS-RBC and activated platelets accelerates the assembly of prothrombinase complexes leading to generation of thrombin, which can cleave circulating C5 protein into two biologically active fragments, C5a and C5b. C5a is an anaphylatoxin, a potent pro-inflammatory mediator, that can activate leukocytes, platelets, and endothelial cells, all of which play a role in vaso-occlusion (VO). Active C5b fragments stimulate the formation of membrane attack complexes (MAC) on SS-RBC that increases their susceptibility to lysis. We hypothesize that complement activation on the surface of SS-RBC may stimulate VO and RBC turnover in SCD. Methods and Results Whole blood from Townes-AA, -AS, and -SS mice (n=4) was collected in EDTA and RBC were immunostained with erythroid specific anti-Ter119 (Ly-76) IgG conjugated to PE-Cy7, anti-C5b-9 (MAC) IgG conjugated to Alexa Fluor 647, and anti-C3 (also specific for activation fragments C3b, iC3b, C3d and C3dg) IgG conjugated to PE. The percentages of Ter119-positive RBC that were positive for MAC were 6.3%, 6.5%, and 26.0% for AA-, AS-, and SS-RBCs, respectively (p<0.01 SS vs. AA and AS) suggesting enhanced C5 activation in Townes-SS mice. However, EDTA plasma C5a levels measured by ELISA were not significantly different between Townes-AA, -AS and -SS mice. C3 activation fragments were also found on a subset of MAC positive RBC. We used a mouse model of hypoxia/reoxygenation (H/R)-induced stasis to investigate the role of complement in VO. Townes-SS mice with implanted dorsal skin-fold chambers were infused with 30 µg of either anti-C5 IgG mAb BB5.1 (which blocks murine C5 cleavage ) (n=4), annexin V (n=2), or with isotype control IgG (n=4) 30 minutes prior to H/R (1 hour of hypoxia at 7% O2, followed by reoxygenation for 4 hours in room air). Percent microvascular stasis (% non-flowing venules) in the subcutaneous venules in the dorsal skin fold chamber window was measured using intravital microscopy at 1 and 4 hours post-hypoxia. Percent stasis was significantly lower in the Townes-SS mice receiving anti-C5 IgG or annexin V than the mice receiving the isotype control IgG at 1 and 4 hours post-hypoxia (Figure 1, means ± SD, *p<0.01 anti-C5 IgG or annexin V vs. control IgG). This finding implies that H/R may acutely activate C5 , thus leading to stasis. Initial studies suggest increased plasma C5a levels in Towne-SS mice treated with isotype control IgG compared to mice treated with anti-C5 IgG in response to H/R. Conclusions These results demonstrate an increased percentage of SS-RBC expressing MAC on their surface and inhibition of H/R-induced stasis with anti-C5 IgG. However, circulating C5a levels were not different between untreated Townes-AA, -AS, or -SS mice suggesting that C5a is not chronically elevated in SCD. Rather, the generation of C5a may occur locally and in the acute setting. Current studies underway are examining the effects of anti-C5 IgG on MAC deposition and RBC half-lives; the mechanism(s) of complement activation in SCD mice (classical, alternative, tic-over, etc); the ability of recombinant murine C5a to induce stasis; and complement activation markers on RBC and in plasma from SCD patients and controls. Abstract:
Environments containing significant concentration of NaCl such as salt lakes harbor extremophiles microorganisms which have a great biotechnology interest. To explore the diversity of Bacteria in Chott Tinsilt (Algeria), an isolation program was performed. Water samples were collected from the saltern during the pre-salt harvesting phase. This Chott is high in salt (22.47% (w/v). Seven halophiles Bacteria were selected for further characterization. The isolated strains were able to grow optimally in media with 10–25% (w/v) total salts. Molecular identification of the isolates was performed by sequencing the 16S rRNA gene. It showed that these cultured isolates included members belonging to the Halomonas, Staphylococcus, Salinivibrio, Planococcus and Halobacillus genera with less than 98% of similarity with their closest phylogenetic relative. The halophilic bacterial isolates were also characterized for the production of biosurfactant and industrially important enzymes. Most isolates produced hydrolases and biosurfactants at high salt concentration. In fact, this is the first report on bacterial strains (A4 and B4) which were a good biosurfactant and coagulase producer at 20% and 25% ((w/v)) NaCl. In addition, the biosurfactant produced by the strain B4 at high salinity (25%) was also stable at high temperature (30-100°C) and high alkalinity (pH 11).Key word: Salt Lake, Bacteria, biosurfactant, Chott, halophiles, hydrolases, 16S rRNAINTRODUCTIONSaline lakes cover approximately 10% of the Earth’s surface area. The microbial populations of many hypersaline environments have already been studied in different geographical regions such as Great Salt Lake (USA), Dead Sea (Israel), Wadi Natrun Lake (Egypt), Lake Magadi (Kenya), Soda Lake (Antarctica) and Big Soda Lake and Mono Lake (California). Hypersaline regions differ from each other in terms of geographical location, salt concentration and chemical composition, which determine the nature of inhabitant microorganisms (Gupta et al., 2015). Then low taxonomic diversity is common to all these saline environments (Oren et al., 1993). Halophiles are found in nearly all major microbial clades, including prokaryotic (Bacteria and Archaea) and eukaryotic forms (DasSarma and Arora, 2001). They are classified as slight halophiles when they grow optimally at 0.2–0.85 M (2–5%) NaCl, as moderate halophiles when they grow at 0.85–3.4 M (5–20%) NaCl, and as extreme halophiles when they grow at 3.4–5.1 M (20–30%) NaCl. Hyper saline environments are inhabited by extremely halophilic and halotolerant microorganisms such as Halobacillus sp, Halobacterium sp., Haloarcula sp., Salinibacter ruber , Haloferax sp and Bacillus spp. (Solomon and Viswalingam, 2013). There is a tremendous demand for halophilic bacteria due to their biotechnological importance as sources of halophilic enzymes. Enzymes derived from halophiles are endowed with unique structural features and catalytic power to sustain the metabolic and physiological processes under high salt conditions. Some of these enzymes have been reported to be active and stable under more than one extreme condition (Karan and Khare, 2010). Applications are being considered in a range of industries such as food processing, washing, biosynthetic processes and environmental bioremediation. Halophilic proteases are widely used in the detergent and food industries (DasSarma and Arora, 2001). However, esterases and lipases have also been useful in laundry detergents for the removal of oil stains and are widely used as biocatalysts because of their ability to produce pure compounds. Likewise, amylases are used industrially in the first step of the production of high fructose corn syrup (hydrolysis of corn starch). They are also used in the textile industry in the de-sizing process and added to laundry detergents. Furthermore, for the environmental applications, the use of halophiles for bioremediation and biodegradation of various materials from industrial effluents to soil contaminants and accidental spills are being widely explored. In addition to enzymes, halophilic / halotolerants microorganisms living in saline environments, offer another potential applications in various fields of biotechnology like the production of biosurfactant. Biosurfactants are amphiphilic compounds synthesized from plants and microorganisms. They reduce surface tension and interfacial tension between individual molecules at the surface and interface respectively (Akbari et al., 2018). Comparing to the chemical surfactant, biosurfactant are promising alternative molecules due to their low toxicity, high biodegradability, environmental capability, mild production conditions, lower critical micelle concentration, higher selectivity, availability of resources and ability to function in wide ranges of pH, temperature and salinity (Rocha et al., 1992). They are used in various industries which include pharmaceuticals, petroleum, food, detergents, cosmetics, paints, paper products and water treatment (Akbari et al., 2018). The search for biosurfactants in extremophiles is particularly promising since these biomolecules can adapt and be stable in the harsh environments in which they are to be applied in biotechnology.OBJECTIVESEastern Algeria features numerous ecosystems including hypersaline environments, which are an important source of salt for food. The microbial diversity in Chott Tinsilt, a shallow Salt Lake with more than 200g/L salt concentration and a superficies of 2.154 Ha, has never yet been studied. The purpose of this research was to chemically analyse water samples collected from the Chott, isolate novel extremely or moderate halophilic Bacteria, and examine their phenotypic and phylogenetic characteristics with a view to screening for biosurfactants and enzymes of industrial interest.MATERIALS AND METHODSStudy area: The area is at 5 km of the Commune of Souk-Naâmane and 17 km in the South of the town of Aïn-Melila. This area skirts the trunk road 3 serving Constantine and Batna and the railway Constantine-Biskra. It is part the administrative jurisdiction of the Wilaya of Oum El Bouaghi. The Chott belongs to the wetlands of the High Plains of Constantine with a depth varying rather regularly without never exceeding 0.5 meter. Its length extends on 4 km with a width of 2.5 km (figure 1).Water samples and physico-chemical analysis: In February 2013, water samples were collected from various places at the Chott Tinsilt using Global Positioning System (GPS) coordinates of 35°53’14” N lat. and 06°28’44”E long. Samples were collected randomly in sterile polythene bags and transported immediately to the laboratory for isolation of halophilic microorganisms. All samples were treated within 24 h after collection. Temperature, pH and salinity were measured in situ using a multi-parameter probe (Hanna Instruments, Smithfield, RI, USA). The analytical methods used in this study to measure ions concentration (Ca2+, Mg2+, Fe2+, Na+, K+, Cl−, HCO3−, SO42−) were based on 4500-S-2 F standard methods described elsewhere (Association et al., 1920).Isolation of halophilic bacteria from water sample: The media (M1) used in the present study contain (g/L): 2.0 g of KCl, 100.0/200.0 g of NaCl, 1.0 g of MgSO4.7HO2, 3.0 g of Sodium Citrate, 0.36 g of MnCl2, 10.0 g of yeast extract and 15.0 g agar. The pH was adjusted to 8.0. Different dilutions of water samples were added to the above medium and incubated at 30°C during 2–7 days or more depending on growth. Appearance and growth of halophilic bacteria were monitored regularly. The growth was diluted 10 times and plated on complete medium agar (g/L): glucose 10.0; peptone 5.0; yeast extract 5.0; KH2PO4 5.0; agar 30.0; and NaCl 100.0/200.0. Resultant colonies were purified by repeated streaking on complete media agar. The pure cultures were preserved in 20% glycerol vials and stored at −80°C for long-term preservation.Biochemical characterisation of halophilic bacterial isolates: Bacterial isolates were studied for Gram’s reaction, cell morphology and pigmentation. Enzymatic assays (catalase, oxidase, nitrate reductase and urease), and assays for fermentation of lactose and mannitol were done as described by Smibert (1994).Optimization of growth conditions: Temperature, pH, and salt concentration were optimized for the growth of halophilic bacterial isolates. These growth parameters were studied quantitatively by growing the bacterial isolates in M1 medium with shaking at 200 rpm and measuring the cell density at 600 nm after 8 days of incubation. To study the effect of NaCl on the growth, bacterial isolates were inoculated on M1 medium supplemented with different concentration of NaCl: 1%-35% (w/v). The effect of pH on the growth of halophilic bacterial strains was studied by inoculating isolates on above described growth media containing NaCl and adjusted to acidic pH of 5 and 6 by using 1N HCl and alkaline pH of 8, 9, 10, 11 and 12 using 5N NaOH. The effect of temperature was studied by culturing the bacterial isolates in M1 medium at different temperatures of incubation (4°C–55°C).Screening of halophilic bacteria for hydrolytic enzymes: Hydrolase producing bacteria among the isolates were screened by plate assay on starch, tributyrin, gelatin and DNA agar plates respectively for amylase, lipase, protease and DNAse activities. Amylolytic activity of the cultures was screened on starch nutrient agar plates containing g/L: starch 10.0; peptone 5.0; yeast extract 3.0; agar 30.0; NaCl 100.0/250.0. The pH was 7.0. After incubation at 30 ºC for 7 days, the zone of clearance was determined by flooding the plates with iodine solution. The potential amylase producers were selected based on ratio of zone of clearance diameter to colony diameter. Lipase activity of the cultures was screened on tributyrin nutrient agar plates containing 1% (v/v) of tributyrin. Isolates that showed clear zones of tributyrin hydrolysis were identified as lipase producing bacteria. Proteolytic activity of the isolates was similarly screened on gelatin nutrient agar plates containing 10.0 g/L of gelatin. The isolates showing zones of gelatin clearance upon treatment with acidic mercuric chloride were selected and designated as protease producing bacteria. The presence of DNAse activity on plates was determined on DNAse test agar (BBL) containing 10%-25% (w/v) total salt. After incubation for 7days, the plates were flooded with 1N HCl solution. Clear halos around the colonies indicated DNAse activity (Jeffries et al., 1957).Milk clotting activity (coagulase activity) of the isolates was also determined following the procedure described (Berridge, 1952). Skim milk powder was reconstituted in 10 mM aqueous CaCl2 (pH 6.5) to a final concentration of 0.12 kg/L. Enzyme extracts were added at a rate of 0.1 mL per mL of milk. The coagulation point was determined by manual rotating of the test tube periodically, at short time intervals, and checking for visible clot formation.Screening of halophilic bacteria for biosurfactant production. Oil spread Assay: The Petridis base was filled with 50 mL of distilled water. On the water surface, 20μL of diesel and 10μl of culture were added respectively. The culture was introduced at different spots on the diesel, which is coated on the water surface. The occurrence of a clear zone was an indicator of positive result (Morikawa et al., 2000). The diameter of the oil expelling circles was measured by slide caliber (with a degree of accuracy of 0.02 mm).Surface tension and emulsification index (E24): Isolates were cultivated at 30 °C for 7 days on the enrichment medium containing 10-25% NaCl and diesel oil as the sole carbon source. The medium was centrifuged (7000 rpm for 20 min) and the surface tension of the cell-free culture broth was measured with a TS90000 surface tensiometer (Nima, Coventry, England) as a qualitative indicator of biosurfactant production. The culture broth was collected with a Pasteur pipette to remove the non-emulsified hydrocarbons. The emulsifying capacity was evaluated by an emulsification index (E24). The E24 of culture samples was determined by adding 2 mL of diesel oil to the same amount of culture, mixed for 2 min with a vortex, and allowed to stand for 24 h. E24 index is defined as the percentage of height of emulsified layer (mm) divided by the total height of the liquid column (mm).Biosurfactant stability studies : After growth on diesel oil as sole source of carbone, cultures supernatant obtained after centrifugation at 6,000 rpm for 15 min were considered as the source of crude biosurfactant. Its stability was determined by subjecting the culture supernatant to various temperature ranges (30, 40, 50, 60, 70, 80 and 100 °C) for 30 min then cooled to room temperature. Similarly, the effect of different pH (2–11) on the activity of the biosurfactant was tested. The activity of the biosurfactant was investigated by measuring the emulsification index (El-Sersy, 2012).Molecular identification of potential strains. DNA extraction and PCR amplification of 16S rDNA: Total cellular DNA was extracted from strains and purified as described by Sambrook et al. (1989). DNA was purified using Geneclean® Turbo (Q-BIO gene, Carlsbad, CA, USA) before use as a template in polymerase chain reaction (PCR) amplification. For the 16S rDNA gene sequence, the purified DNA was amplified using a universal primer set, forward primer (27f; 5′-AGA GTT TGA TCM TGG CTC AG) and a reverse primer (1492r; 5′-TAC GGY TAC CTT GTT ACG ACT T) (Lane, 1991). Agarose gel electrophoresis confirmed the amplification product as a 1400-bp DNA fragment.16S rDNA sequencing and Phylogenic analysis: Amplicons generated using primer pair 27f-1492r was sequenced using an automatic sequencer system at Macrogene Company (Seoul, Korea). The sequences were compared with those of the NCBI BLAST GenBank nucleotide sequence databases. Phylogenetic trees were constructed by the neighbor-joining method using MEGA version 5.05 software (Tamura et al., 2011). Bootstrap resembling analysis for 1,000 replicates was performed to estimate the confidence of tree topologies.Nucleotide sequence accession numbers: The nucleotide sequences reported in this work have been deposited in the EMBL Nucleotide Sequence Database. The accession numbers are represented in table 5.Statistics: All experiments were conducted in triplicates. Results were evaluated for statistical significance using ANOVA.RESULTSPhysico-chemical parameters of the collected water samples: The physicochemical properties of the collected water samples are reported in table 1. At the time of sampling, the temperature was 10.6°C and pH 7.89. The salinity of the sample, as determined in situ, was 224.70 g/L (22,47% (w/v)). Chemical analysis of water sample indicated that Na +and Cl- were the most abundant ions (table 1). SO4-2 and Mg+2 was present in much smaller amounts compared to Na +and Cl- concentration. Low levels of calcium, potassium and bicarbonate were also detected, often at less than 1 g/L.Characterization of isolates. Morphological and biochemical characteristic feature of halophilic bacterial isolates: Among 52 strains isolated from water of Chott Tinsilt, seven distinct bacteria (A1, A2, A3, A4, B1, B4 and B5) were chosen for further characterization (table 2). The colour of the isolates varied from beige, pale yellow, yellowish and orange. The bacterial isolates A1, A2, A4, B1 and B5 were rod shaped and gram negative (except B5), whereas A3 and B4 were cocci and gram positive. All strains were oxidase and catalase positive except for B1. Nitrate reductase and urease activities were observed in all the bacterial isolates, except B4. All the bacterial isolates were negative for H2S formation. B5 was the only strain positive for mannitol fermentation (table 2).We isolated halophilic bacteria on growth medium with NaCl supplementation at pH 7 and temperature of 30°C. We studied the effect of NaCl, temperature and pH on the growth of bacterial isolates. All the isolates exhibited growth only in the presence of NaCl indicating that these strains are halophilic. The optimum growth of isolates A3 and B1 was observed in the presence of 10% NaCl, whereas it was 15% NaCl for A1, A2 and B5. A4 and B4 showed optimum growth in the presence of 20% and 25% NaCl respectively. A4, B4 and B5 strains can tolerate up to 35% NaCl.The isolate B1 showed growth in medium supplemented with 10% NaCl and pH range of 7–10. The optimum pH for the growth B1 was 9 and they did not show any detectable growth at or below pH 6 (table 2), which indicates the alkaliphilic nature of B1 isolate. The bacterial isolates A1, A2 and A4 exhibited growth in the range of pH 6–10, while A3 and B4 did not show any growth at pH greater than 8. The optimum pH for growth of all strains (except B1) was pH 7.0 (table 2). These results indicate that A1, A2, A3, A4, B4 and B5 are neutrophilic in nature. All the bacterial isolates exhibited optimal growth at 30°C and no detectable growth at 55°C. Also, detectable growth of isolates A1, A2 and A4 was observed at 4°C. However, none of the bacterial strains could grow below 4°C and above 50°C (table 2).Screening of the halophilic enzymes: To characterize the diversity of halophiles able to produce hydrolytic enzymes among the population of microorganisms inhabiting the hypersaline habitats of East Algeria (Chott Tinsilt), a screening was performed. As described in Materials and Methods, samples were plated on solid media containing 10%-25% (w/v) of total salts and different substrates for the detection of amylase, protease, lipase and DNAse activities. However, coagulase activity was determined in liquid medium using milk as substrate (figure 3). Distributions of hydrolytic activity among the isolates are summarized in table 4.From the seven bacterial isolates, four strains A1, A2, A4 and B5 showed combined hydrolytic activities. They were positive for gelatinase, lipase and coagulase. A3 strain showed gelatinase and lipase activities. DNAse activities were detected with A1, A4, B1 and B5 isolates. B4 presented lipase and coagulase activity. Surprisingly, no amylase activity was detected among all the isolates.Screening for biosurfactant producing isolates: Oil spread assay: The results showed that all the strains could produce notable (>4 cm diameter) oil expelling circles (ranging from 4.11 cm to 4.67 cm). The average diameter for strain B5 was 4.67 cm, significantly (P < 0.05) higher than for the other strains.Surface tension and emulsification index (E24): The assimilation of hydrocarbons as the sole sources of carbon by the isolate strains led to the production of biosurfactants indicated by the emulsification index and the lowering of the surface tension of cell-free supernatant. Based on rapid growth on media containing diesel oil as sole carbon source, the seven isolates were tested for biosurfactant production and emulsification activity. The obtained values of the surface tension measurements as well as the emulsification index (E24) are shown in table 3. The highest reduction of surface tension was achieved with B5 and A3 isolates with values of 25.3 mN m−1 and 28.1 mN m−1 respectively. The emulsifying capacity evaluated by the E24 emulsification index was highest in the culture of isolate B4 (78%), B5 (77%) and A3 (76%) as shown in table 3 and figure 2. These emulsions were stable even after 4 months. The bacteria with emulsification indices higher than 50 % and/or reduction in the surface tension (under 30 mN/m) have been defined as potential biosurfactant producers. Based on surface tension and the E24 index results, isolates B5, B4, A3 and A4 are the best candidates for biosurfactant production. It is important to note that, strains B4 and A4 produce biosurfactant in medium containing respectively 25% and 20% (w/v) NaCl.Stability of biosurfactant activities: The applicability of biosurfactants in several biotechnological fields depends on their stability at different environmental conditions (temperatures, pH and NaCl). For this study, the strain B4 appear very interesting (It can produce biosurfactant at 25 % NaCl) and was choosen for futher analysis for biosurfactant stability. The effects of temperature and pH on the biosurfactant production by the strain B4 are shown in figure 4.biosurfactant in medium containing respectively 25% and 20% (w/v) NaCl.Stability of biosurfactant activities: The applicability of biosurfactants in several biotechnological fields depends on their stability at different environmental conditions (temperatures, pH and NaCl). For this study, the strain B4 appear very interesting (It can produce biosurfactant at 25 % NaCl) and was chosen for further analysis for biosurfactant stability. The effects of temperature and pH on the biosurfactant production by the strain B4 are shown in figure 4. The biosurfactant produced by this strain was shown to be thermostable giving an E-24 Index value greater than 78% (figure 4A). Heating of the biosurfactant to 100 °C caused no significant effect on the biosurfactant performance. Therefore, the surface activity of the crude biosurfactant supernatant remained relatively stable to pH changes between pH 6 and 11. At pH 11, the value of E24 showed almost 76% activity, whereas below pH 6 the activity was decreased up to 40% (figure 4A). The decreases of the emulsification activity by decreasing the pH value from basic to an acidic region; may be due to partial precipitation of the biosurfactant. This result indicated that biosurfactant produced by strain B4 show higher stability at alkaline than in acidic conditions.Molecular identification and phylogenies of potential isolates: To identify halophilic bacterial isolates, the 16S rDNA gene was amplified using gene-specific primers. A PCR product of ≈ 1.3 kb was detected in all the seven isolates. The 16S rDNA amplicons of each bacterial isolate was sequenced on both strands using 27F and 1492R primers. The complete nucleotide sequence of 1336,1374, 1377,1313, 1305,1308 and 1273 bp sequences were obtained from A1, A2, A3, A4, B1, B4 and B5 isolates respectively, and subjected to BLAST analysis. The 16S rDNA sequence analysis showed that the isolated strains belong to the genera Halomonas, Staphylococcus, Salinivibrio, Planococcus and Halobacillus as shown in table 5. The halophilic isolates A2 and A4 showed 97% similarity with the Halomonas variabilis strain GSP3 (accession no. AY505527) and the Halomonas sp. M59 (accession no. AM229319), respectively. As for A1, it showed 96% similarity with the Halomonas venusta strain GSP24 (accession no. AY553074). B1 and B4 showed for their part 96% similarity with the Salinivibrio costicola subsp. alcaliphilus strain 18AG DSM4743 (accession no. NR_042255) and the Planococcus citreus (accession no. JX122551), respectively. The bacterial isolate B5 showed 98% sequence similarity with the Halobacillus trueperi (accession no. HG931926), As for A3, it showed only 95% similarity with the Staphylococcus arlettae (accession no. KR047785). The 16S rDNA nucleotide sequences of all the seven halophilic bacterial strains have been submitted to the NCBI GenBank database under the accession number presented in table 5. The phylogenetic association of the isolates is shown in figure 5.DICUSSIONThe physicochemical properties of the collected water samples indicated that this water was relatively neutral (pH 7.89) similar to the Dead Sea and the Great Salt Lake (USA) and in contrast to the more basic lakes such as Lake Wadi Natrun (Egypt) (pH 11) and El Golea Salt Lake (Algeria) (pH 9). The salinity of the sample was 224.70 g/L (22,47% (w/v). This range of salinity (20-30%) for Chott Tinsilt is comparable to a number of well characterized hypersaline ecosystems including both natural and man-made habitats, such as the Great Salt Lake (USA) and solar salterns of Puerto Rico. Thus, Chott Tinsilt is a hypersaline environment, i.e. environments with salt concentrations well above that of seawater. Chemical analysis of water sample indicated that Na +and Cl- were the most abundant ions, as in most hypersaline ecosystems (with some exceptions such as the Dead Sea). These chemical water characteristics were consistent with the previously reported data in other hypersaline ecosystems (DasSarma and Arora, 2001; Oren, 2002; Hacěne et al., 2004). Among 52 strains isolated from this Chott, seven distinct bacteria (A1, A2, A3, A4, B1, B4 and B5) were chosen for phenotypique, genotypique and phylogenetique characterization.The 16S rDNA sequence analysis showed that the isolated strains belong to the genera Halomonas, Staphylococcus, Salinivibrio, Planococcus and Halobacillus. Genera obtained in the present study are commonly occurring in various saline habitats across the globe. Staphylococci have the ability to grow in a wide range of salt concentrations (Graham and Wilkinson, 1992; Morikawa et al., 2009; Roohi et al., 2014). For example, in Pakistan, Staphylococcus strains were isolated from various salt samples during the study conducted by Roohi et al. (2014) and these results agreed with previous reports. Halomonas, halophilic and/or halotolerant Gram-negative bacteria are typically found in saline environments (Kim et al., 2013). The presence of Planococcus and Halobacillus has been reported in studies about hypersaline lakes; like La Sal del Rey (USA) (Phillips et al., 2012) and Great Salt Lake (Spring et al., 1996), respectively. The Salinivibrio costicola was a representative model for studies on osmoregulatory and other physiological mechanisms of moderately halophilic bacteria (Oren, 2006).However, it is interesting to note that all strains shared less than 98.7% identity (the usual species cut-off proposed by Yarza et al. (2014) with their closest phylogenetic relative, suggesting that they could be considered as new species. Phenotypic, genetic and phylogenetic analyses have been suggested for the complete identification of these strains. Theses bacterial strains were tested for the production of industrially important enzymes (Amylase, protease, lipase, DNAse and coagulase). These isolates are good candidates as sources of novel enzymes with biotechnological potential as they can be used in different industrial processes at high salt concentration (up to 25% NaCl for B4). Prominent amylase, lipase, protease and DNAase activities have been reported from different hypersaline environments across the globe; e.g., Spain (Sánchez‐Porro et al., 2003), Iran (Rohban et al., 2009), Tunisia (Baati et al., 2010) and India (Gupta et al., 2016). However, to the best of our knowledge, the coagulase activity has never been detected in extreme halophilic bacteria. Isolation and characterization of crude enzymes (especially coagulase) to investigate their properties and stability are in progress.The finding of novel enzymes with optimal activities at various ranges of salt concentrations is of great importance. Besides being intrinsically stable and active at high salt concentrations, halophilic and halotolerant enzymes offer great opportunities in biotechnological applications, such as environmental bioremediation (marine, oilfiel) and food processing. The bacterial isolates were also characterized for production of biosurfactants by oil-spread assay, measurement of surface tension and emulsification index (E24). There are few reports on biosurfactant producers in hypersaline environments and in recent years, there has been a greater increase in interest and importance in halophilic bacteria for biomolecules (Donio et al., 2013; Sarafin et al., 2014). Halophiles, which have a unique lipid composition, may have an important role to play as surface-active agents. The archae bacterial ether-linked phytanyl membrane lipid of the extremely halophilic bacteria has been shown to have surfactant properties (Post and Collins, 1982). Yakimov et al. (1995) reported the production of biosurfactant by a halotolerant Bacillus licheniformis strain BAS 50 which was able to produce a lipopeptide surfactant when cultured at salinities up to 13% NaCl. From solar salt, Halomonas sp. BS4 and Kocuria marina BS-15 were found to be able to produce biosurfactant when cultured at salinities of 8% and 10% NaCl respectively (Donio et al., 2013; Sarafin et al., 2014). In the present work, strains B4 and A4 produce biosurfactant in medium containing respectively 25% and 20% NaCl. To our knowledge, this is the first report on biosurfactant production by bacteria under such salt concentration. Biosurfactants have a wide variety of industrial and environmental applications (Akbari et al., 2018) but their applicability depends on their stability at different environmental conditions. The strain B4 which can produce biosurfactant at 25% NaCl showed good stability in alkaline pH and at a temperature range of 30°C-100°C. Due to the enormous utilization of biosurfactant in detergent manufacture the choice of alkaline biosurfactant is researched (Elazzazy et al., 2015). On the other hand, the interesting finding was the thermostability of the produced biosurfactant even after heat treatment (100°C for 30 min) which suggests the use of this biosurfactant in industries where heating is of a paramount importance (Khopade et al., 2012). To date, more attention has been focused on biosurfactant producing bacteria under extreme conditions for industrial and commercial usefulness. In fact, the biosurfactant produce by strain B4 have promising usefulness in pharmaceutical, cosmetics and food industries and for bioremediation in marine environment and Microbial enhanced oil recovery (MEOR) where the salinity, temperature and pH are high.CONCLUSIONThis is the first study on the culturable halophilic bacteria community inhabiting Chott Tinsilt in Eastern Algeria. Different genera of halotolerant bacteria with different phylogeneticaly characteristics have been isolated from this Chott. Culturing of bacteria and their molecular analysis provides an opportunity to have a wide range of cultured microorganisms from extreme habitats like hypersaline environments. Enzymes produced by halophilic bacteria show interesting properties like their ability to remain functional in extreme conditions, such as high temperatures, wide range of pH, and high salt concentrations. These enzymes have great economical potential in industrial, agricultural, chemical, pharmaceutical, and biotechnological applications. Thus, the halophiles isolated from Chott Tinsilt offer an important potential for application in microbial and enzyme biotechnology.
