Abstract:
Recent discoveries establish DNA and RNA as bona fide substrates for ADP-ribosylation. NADAR (“NAD- and ADP-ribose”-associated) enzymes reverse guanine ADP-ribosylation and serve as antitoxins in the DarT-NADAR operon. Although NADARs are widespread across prokaryotes, eukaryotes, and viruses, their specificity and broader physiological roles remain poorly understood. Using phylogenetic and biochemical analyses, we further explore de-ADP-ribosylation activity and antitoxin functions of NADAR domains. We demonstrate that different subfamilies of NADAR proteins from representative E. coli strains and an E. coli-infecting phage retain biochemical activity while displaying specificity in providing protection from toxic guanine ADP-ribosylation in cells. Furthermore, we identify a myxobacterial enzyme within the YbiA subfamily that functions as an antitoxin for its associated DarT-unrelated ART toxin, which we termed YarT, thus presenting a hitherto uncharacterised ART-YbiA toxin–antitoxin pair. Our studies contribute to the burgeoning field of DNA ADP-ribosylation, supporting its physiological relevance within and beyond bacterial toxin–antitoxin systems. Notably, the specificity and confinement of NADARs to non-mammals infer their potential as highly specific targets for antimicrobial drugs with minimal off-target effects.

Abstract:
The feelings, actions, and habits of the individual are the first steps in the development of culture. Another stage is the cultural development of societies formed by individuals living together. Culture is the third stage when the lifestyle, habits, artistic sensibility, and maturity of thought formed when many communities work together to form a community and intertwine as a community are all manifested as a kind of social thought. No mention of Nadars is found in Sangam Tamil literature. The state of the Nadars is known from the references of Christian missionaries in the early nineteenth century. At the beginning of the 19th century, social reform movements emerged in many places around the world, and there was awareness among the people of society. At that time, the status of the people of the Nadar community was backward in the social hierarchy. The word "wise men" is another name for Chanars. It is no exaggeration to say that in the social and religious hierarchies, the Chanars were slightly superior to the lower castes and a little inferior to the upper castes, and the status of the Nadars, who were denied entry into the temple to worship, due to the democratic renaissance social strata in today's era, the present state that this community has reached in the social strata and religious hierarchies is a pioneering society for the various ethnic groups that may exist in the world. In this article, we will see the importance of cultural elements such as economic resources, education, artistic development, morals, constitution and traditional elements of clan deity worship such as temple structure, worship methods, festivals, and social participation, etc. in making social reform ideas flourish.

The 1895 riot at Kalugumalai in the Tirunelveli District of Madras Presidency, South India, pitted the local Nadar community, then newly-converted to Roman Catholicism, against the main Hindu castes of Kalugumalai, particularly those associated with its Hindu temple and the Ettaiyapuram zamindari estate within which the town lay. It was the violent climax to a long-running dispute over the Nadars' right to take processions through the main streets, and one of the bloodiest episodes in a conflict which posed a severe threat to public order throughout South India in the late nineteenth century.

This article examines changes in the death rituals performed among Hindu Nadars in a South Indian village. It emphasises the importance of understanding ritual changes within their specific micro-level local contextual framework, including changing social structures at household and village level. This empirical evidence showcases how changing rituals connected to death reflect various adaptations through imitation, substitution and alteration of specific ritual elements and performants. It also identifies emerging class distinctions among Nadars and their connection with changes in rituals associated with death. This analysis of the changes depicts how Nadars use ritual actions in pragmatic ways, symbolically expressing and realising their aspirations for status enhancement through such ritual performances.

The Belgium-based Nadar Ensemble is a new music ensemble that specializes in performing curated and integrated concerts. In this case ‘integrated’ infers that all aspects of the concert are taken into consideration. Questioning, developing, and instrumentalizing video, light and sound design (including live electronics), costuming, decor, and even personnel such as a conductor and other roles typically inherent to an ensembles’ tradition, have become an integral part of Nadar’s and other similar ensembles’ concert programming. In this paper I will specifically lay out the manners in which the musicians of Nadar Ensemble approach what they have grown to see as their broadened instruments, including the use of live electronics, game controllers and newly made instruments. I will also discuss ideas surrounding the so-called ‘attacca concert’ and post-instrumental practice by delving into two cases, the event Dead Serious (2014) and the concert program Extensions (2015).

ABSTRACTThe NadA adhesin is a major component of 4CMenB, a novel vaccine to prevent meningococcus serogroup B (MenB) infection. Underin vitrogrowth conditions,nadAis repressed by the regulator NadR and poorly expressed, resulting in inefficient killing of MenB strains by anti-NadA antibodies. Interestingly, sera from children infected with strains that express low levels of NadA in laboratory growth nevertheless recognize the NadA antigen, suggesting that NadA expression during infection may be different from that observedin vitro. In a strain panel covering a range of NadA levels, repression was relieved through deletingnadR. AllnadRknockout strains expressed high levels of NadA and were efficiently killed by sera from subjects immunized with 4CMenB. A selected MenB strain, NGP165, mismatched for other vaccine antigens, is not killed by sera from immunized infants when the strain is grownin vitro. However, in anin vivopassive protection model, the same sera effectively protected infant rats from bacteremia with NGP165. Furthermore, we identify a novel hydroxyphenylacetic acid (HPA) derivative, reported by others to be produced during inflammation, which induces expression of NadAin vitro, leading to efficient antibody-mediated killing. Finally, using bioluminescent reporters,nadAexpression in the infant rat model was inducedin vivoat 3 h postinfection. Our results suggest that during infectious disease, NadR repression is alleviated due to niche-specific signals, resulting in high levels of NadA expression from anynadA-positive (nadA+) strain and therefore efficient killing by anti-NadA antibodies elicited by the 4CMenB vaccine.

Abstract:
O objetivo deste artigo é descrever os processos e significados envolvidos na aprendizagem do nado entre crianças ribeirinhas do médio rio Solimões. A pesquisa questiona os métodos de aprendizagem e o que aprender a nadar representa em termos de construção da pessoa. Assim, recorremos a um estudo exploratório, com abordagem qualitativa e estudo de caso. Realizamos registros fotográficos, entrevistas semiestruturadas e observação participante. As crianças aprendem a nadar por meio de brincadeiras, em espaços específicos nas beiras dos rios, no período de inundação. Aprender a nadar lhes permite circular de forma autônoma nos espaços comunitários e lhes torna aptos a participar também do mundo da pesca. Concluímos que a natação é fundamental para a formação da pessoa, construção e afirmação da territorialidade do grupo.

Antecedentes: La bibliografía y el sentido común señalan que saber nadar disminuye el riesgo de ahogamiento, pero no garantiza la supervivencia en el agua. Según informes de la OMS, se sabe que las muertes por ahogamientos son más frecuentes en países con rentas bajas y medias. Aunque España es un país con índice de desarrollo humano muy alto, en gran parte de sus currículos educativos no se contempla enseñar a nadar y a evitar el ahogamiento. Objetivo: El primer objetivo de este estudio es conocer los efectos de la aplicación del Currículo de Educación Primaria de Educación Física de la Comunidad Autónoma de Madrid sobre el nivel de competencia acuática de su alumnado. El segundo objetivo de estudio es comprobar si el nivel de competencia en el agua que posee el alumnado matriculado en colegios públicos es diferente al que posee el alumnado matriculado en centros privados o concertados. Método: Se ha comprobado la competencia en el agua de 1.274 estudiantes que cursan el tercer ciclo de Educación Primaria en la Comunidad Autónoma de Madrid. De ellos, 738 estudian en colegios privados o concertados y 536 estudian en centros públicos. Resultados: Los resultados de esta investigación muestran que, 102 estudiantes (8% del total) no saben nadar absolutamente nada y, 24 estudiantes más (1,9% del total) no son capaces de nadar 25 metros seguidos. Asimismo, el 100% del alumnado que no sabe nadar o que lo hace de forma insegura, está matriculado en colegios públicos. Conclusiones: Se podría destacar que el Currículo de Educación Primaria de Educación Física de la Comunidad Autónoma de Madrid no garantiza que el 100% de su alumnado sea mínimamente competente en el agua antes de los 12 años. También se concluye que, el Currículo de este territorio no disminuye el riesgo de ahogamiento de aquellas personas que, por una u otra razón, no disponen de las oportunidades necesarias para aprender a nadar fuera del ámbito escolar. Estos resultados confirman la existencia de un serio problema que la bibliografía especializada ha demostrado que afecta especialmente a la población más desfavorecida.

Objective Transcatheter aortic valve implantation (TAVI) relies on fluoroscopy and nephrotoxic contrast medium for valve deployment. We propose an alternative guidance system using augmented reality (AR) and transesophageal echocardiography (TEE) to guide TAVI deployment. The goals of this study were to determine how consistently the aortic valve annulus is defined from TEE using different aortic valve landmarks and to compare AR guidance with fluoroscopic guidance of TAVI deployment in an aortic root model. Methods Magnetic tracking sensors were integrated into the TAVI catheter and TEE probe, allowing these tools to be displayed in an AR environment. Variability in identifying aortic valve commissures and cuspal nadirs was assessed using TEE aortic root images. To compare AR guidance of TAVI deployment with fluoroscopic guidance, a TAVI stent was deployed 10 times in the aortic root model using each of the two guidance systems. Results Commissures and nadirs were both investigated as features for defining the valve annulus in the AR guidance system. The commissures were identified more consistently than the nadirs, with intraobserver variability of 2.2 and 3.8 mm, respectively, and interobserver variability of 3.3 and 4.7 mm, respectively. The precision of TAVI deployment using fluoroscopic guidance was 3.4 mm, whereas the precision of AR guidance was 2.9 mm, and its overall accuracy was 3.4 mm. This indicates that both have similar performance. Conclusions Aortic valve commissures can be identified more reliably than cuspal nadirs from TEE. The AR guidance system achieved similar deployment accuracy to that of fluoroscopy while eliminating the use and consequences of nephrotoxic contrast and radiation.

BackgroundLymphopenia is expected from the mechanism of action of cladribine tablets 3.5 mg/kg (CT3.5)ObjectiveInvestigate absolute lymphocyte counts (ALC; 312 weeks) and subsets (240 weeks) in RRMS patients receiving 2 annual courses of CT3.5.MethodsPooled data from patients randomised to CT3.5 over 2 years in CLARITY/CLARITY-Extension inclusive of the PREMIERE registry (n=685).ResultsBaseline: median ALC=1.86 × 109/L. Year-1: ALC reached nadir at 9 weeks post-treatment with CT3.5 (1.00 × 109/L). Year-2: ALC reached nadir at Week-55 (0.81 × 109/L), then recovered to the normal range (≥1.00 × 109/L; Week-96). ALC was in normal range in 75% of patients by Week-144. Baseline median CD4+ were 851 cells/µL. Nadirs occurred at Week-16 (385 cells/µL) in Year-1 and at Week-60 (292 cells/µL) in Year-2; values increased after nadirs and regained threshold (350 cells/µL, ~Week-120). Baseline median CD8+ were 378 cells/µL. Nadirs occurred at Week-16 (239 cells/µL; Year 1) and Week-72 (232 cells/µL; Year 2). CD8+ recovered quickly after treatment and remained above 200 cells/µL at all times. Baseline median CD19+ were 205 cells/µL. Macierzyństwo to zjawisko oczywiste i naturalne, które z reguły utożsamiane jest z wyjątkowymi doświadczeniami matek i pozytywnym postrzeganiem społecznym. Wyjątek może stanowić pełnienie roli matki dziecka z niepełnosprawnością. Wówczas macierzyństwo nabiera innych znaczeń, a sposoby doświadczania roli matki nie są już tak oczywiste, jak w przypadku dziecka pełnosprawnego. Celem artykułu jest zaprezentowanie wyników badań rekonstrukcji przeżyć i interpretacji doświadczeń na temat macierzyństwa z dzieckiem z niepełnosprawnością oraz zaproponowanie możliwości wsparcia matek wychowujących dziecko z niepełnosprawnością. Przedstawione w opracowaniu badania zostały osadzone w paradygmacie interpretatywnym, co w konsekwencji pozwoliło zastosować metodę biograficzną z wykorzystaniem autobiograficznego wywiadu narracyjnego. Kwestia macierzyństwa z dzieckiem z niepełnosprawnością pojawia się w dyskursie publicznym i naukowym dość często, jednak perspektywa biograficzna odsłania indywidualne sensy nadane macierzyństwu. Rekonstruowana przez matki rzeczywistość jest złożona i składa z negatywnych emocji będących skutkiem zagrożenia, niepewności, dezorientacji i braku kontroli nad sytuacją wywołaną niepełnosprawnością dziecka. Obok nich w życiu kobiet sytuują się pozytywne emocje wynikające z akceptacji niepełnosprawności, najmniejszych efektów dziecka, czy poczucia spełnienia w roli matki. Subiektywna interpretacja doświadczeń przeżywanych przez matki wychowujące dziecko z niepełnosprawnością ukazuje niepowtarzalne biografie i dowodzi, że matki na wiele sposobów doświadczają tego macierzyństwa: macierzyństwo osamotnione, obciążone ryzykiem czy zdominowane niepełnosprawnością, ale także znormalizowane, wspierane i spełnione. Opowiedzianym historiom maki nadają własne znaczenia, które z ich perspektywy są istotne.

Background: Pediatricians often encounter children with heart murmurs during routine follow up visits or while assessment of intercurrent illness. Clinical differentiation between pathologic and innocent murmurs would be important. It is important to identify those children who need further evaluation. The aim of the study was to evaluate whether Nadas criteria can be applied as a screening test to decide on further workup.Methods: This was a prospective descriptive study done in Government T. D. Medical College, Alappuzha over a period of 12 months from July 2013 to June 2014. All children in the age group one month to 12 years, attending the outpatient department were screened for presence of murmur of grade 2 and more. They were further evaluated carefully. They were also studied for association of variables like maternal age, birthweight, dysmorphism and family history of heart disease. Nadas criteria was then applied to the study population. A score of 2 and 1 were given to major and minor criteria respectively.Results: 3070 children were screened. 150 children were detected to have murmur of grade2 and more.66 children were detected to have a structural heart disease by an echo study.74% of the study population in the age group <1 year had a heart disease.8 out of 78 children with a Nadas score of 1 and 10 out of 24 with a score of 2 had a heart disease. All with a score 3 and more had congenital heart disease (CHD).Conclusions: Nadas criteria can be used as screening test with a sensitivity of 87% and specificity of 83%.

In this paper, a method for extracting Fenglin and Fengcong landform units based on karst topographic feature points is proposed. First, the variable analysis window method is used to extract peaks, nadirs, and saddle points in the karst area based on digital elevation model (DEM) data. Thiessen polygons that cover the karst surface area are constructed according to the locations of the peaks and nadirs, and the attributes of the saddles are assigned to corresponding polygons. The polygons are automatically classified via grouping analysis according to the corresponding spatial combinations of peaks, saddles, and nadirs in the Fenglin and Fengcong landform units. Then, a detailed division of the surface morphology of the karst area is achieved by distinguishing various types of Fenglin or Fengcong landform units. Experiments in the Guilin research area show that the proposed method successfully distinguishes the Fenglin and Fengcong terrain areas and extracts Fengcong landform units, individual Fenglin units, and Fenglin chains. The Fengcong area covers approximately two-thirds of the whole area, the individual Fenglin area covers approximately one-fourth, and the Fenglin chain area covers approximately one-tenth. The development of Fenglin has different stages in the Guilin area. This study provides data support for the detailed morphological study of karst terrain, and proposes a new research idea for the division and extraction of karst landform units.
