Abstract:
The diploma thesis deal with what ways drivers react to an unexpected stimulus. In the theoretical part, the analysis of the current state, reaction time, methods of reaction of the driver, stimuli from the environment and finally is being analyzed, the vehicle and its assistance systems. The analytical part of the diploma thesis was performed by driving tests, where it was found that the way the driver reacts from the distance to which they are obstructed and the speed of the vehicle. For that reason, a TTC dependency analysis was performed. It was found that drivers at low TTC values chose an obstacle in which they brake the need to move their foot from the accelerator pedal to the brake pedal and they have to brake sharply. At medium TTC values, drivers brake most often. In the case of a combination of conductors, they chose braking with avoiding at lower TTC values and avoiding with braking at medium TTC values. There was also a dependence of the method of reaction on driving experience. Inexperienced drivers rather choose braking, experienced drivers use combinations.

The final effects of the fast closing valve replacement will be examined on three different types of valves with regard to the turbine output in operation zone and without the influence of water intake runner and turbine draft tube. The Cardiovascular diseases may evocated the high percentual risk of sudden cardiac death in whole world. In several western countries is the number of death higher then number of cancer death. In this time is used a lot of methods for prediction of sudden cardiac death with focus on ECG T-wave alternance. The aim of the theses was to do stronger relation and cooperation with Internal Cardiac Clinic of Faculty Hospital Brno Bohunice on the risk analysis of sudden cardiac death. Secondly, we met the methods used for detection and quantification of simulated TWA. Last but not least was necessary to find TWA detection methods improvement and process the data on real signals obtained from Faculty Hospital Brno Bohunice. First part of the Thesis is focused on summary of pathologic artifacts in ECG signal, which are important for sudden cardiac risk stratification. There are described further known detection and quantification methods for TWA analysis. An interesting part for clinical practice is analysis of TWA trend in time and looking for the best method, which is able to catch and track the short TWA trend changes. Second part describes the new methods improvements, which were tested with interesting outputs. Further, there was developed method for TWA presence statement probability evaluation.

Abstract:
The thesis deals with the terminology used in forensic engineering practice at the analysis of road accidents. It means defining the concepts describing specific expert research in the area of car accident analysis such as the option of evasive action and the possibility of preventing collision. Further on, the thesis focuses on the terminology used in areas closely related to analysis of road accidents namely dealing with cases of criminal activities in the field of motor vehicle insurance from an expert point of view. In this area, terms such as extent of the damage, character of the damage, correspondence of the damage and the technical acceptability of accident, are solved. These terms constitute a formal process that can serve as a tool for experts when dealing with such cases. In addition, selected terms from the technical-legal area, which the experts often encounter, are processed here. At the same time, there is no uniform approach to their evaluation from a technical perspective. Here, there is a possibility of setting the limits of a sudden change of speed and a sudden change of direction.

This thesis studies the optimization of diagnosis defects of the photovoltaic cells in the darkroom by the help of new operating software for the used CCD camera. First the present situation and the possible solutions are analyzed. Then the attention is paid to the theory of light-sensitive CCD sensors, their construction, their principles of operation and the theory of scanning images. The thesis also focuses on the analysis of the function of the operating libraries which were provided by the equipment producer. The key part is the development of the application for the camera G2-3200 of the "Moravske pristroje" company which significantly accelerates the work with the camera in the darkroom.

Abstract:
This thesis deals with analysis of T-wave Alternans (TWA), periodical changes of T wave in ECG signal. Presence of these alternans may predict higher risk of sudden cardiac death. From the several possible ways of TWA classification, the training algorithms of self organizing maps are used in this thesis. Result of the thesis is a program, which in the first step detects QRS complexes in the signal. Then, in the next step, gained reference points are used for T-waves detection. Detected waves are represented by a vector of significant points, which is used as an input for artificial neural network. Final output of the program is a decision about presence of TWA in the signal and its rate.

The thesis deals with detection of T-wave alternans. The presence of T-wave in surface ECG is recognized as a marker of electrical instability of heart in stage his repolarization, arise increased risk of emergence ventricular fibrillation and sudden cardiac death. The goal of our project is familiarize with methods of detection T-wave alternans. In particular spectral method and spectral method which was realized in variant for running reading values in time (“sliding window”). To suggest a QRS complex detector, localize the T-wave and to make TWA detection using spectral method and modified spectral method. Monika Kuchařová, Ph.D. Title of Diploma Thesis: The determination of fatty acid levels in the tissues of healthy, suddenly deceased persons and polymorbid patients The aim of the thesis was to determine the levels of 14 fatty acids in tissues of suddenly deceased, otherwise healthy individuals and in the group of polymorbid, chronic patients. It was a comparison of fatty acid levels in seven tissues of the human body: subendocardial left ventricular tissue, liver parenchyma tissue, kidney cortex, adrenal tissue, skeletal muscle, abdominal subcutaneous adipose tissue, and brain tissue. Each group included 10 deceased patients. The theoretical part incudes fatty acids as the main component of lipids. It deals mainly with the group of polyunsaturated fatty acids and their relation to pathologies in the human body. This thesis monitors the levels of selected fatty acids in human tissues and the effect of long-term disease on their stores. In addition to the general characteristics, the theoretical part focuses on the intake of polyunsaturated fatty acids and their importance in the human body. It also deals with the effects of eicosanoids and other fatty acid metabolites. Methods used in fatty acid bioanalysis are also described. Sudden death in sport is a tragic event mediated primarily through the media. This thesis deals with sudden cardiac death from non-traumatic causes. The first section provides the overview of the issue, I mentioned specific cases of sudden death and the most common cause of sudden cardiac death among young and old athletes. The thesis focuses on the possibilities of preventive measures. In the second part, as part of my research, preparedness of Czech stadiums is evaluated situations of sudden cardiac death. In the conclusion section, I present the results of a questionnaire survey among representatives of clubs and stadiums and change proposals. According to recent research disturbances of self- awareness and conscious experience have a critical role in pathophysi- ology of schizophrenia, which in early stages of the disease mainly in- clude overactivation of the HPA axis. The dissertation thesis deals with short-term gas releases (puffs) in an urban canopy studied utilizing wind-tunnel modelling. The urban canopy was composed of buildings with pitched roofs organised into closed courtyards. Into it, a ground-level point gas source was placed. The first part of the thesis is focused on specific definitions of puff characteristics. New definitions of puff arrival and departure times are presented. Various definitions of puff arrival time were applied on the same datasets and the results were compared. Moreover, it was studied how slight changes in determination of puff departure time can affect its values and other derived puff characteristics. The second part of the thesis is focused on modelling of probability density functions of puff characteristics with knowledge of sampling positions towards the gas source and mean values of concentrations valid for long-term gas sources. The found equations will be utilized in an operational model. The outputs in the form of the probability density functions of puff characteristics distinguish my model from the usually utilized operational models, in which only the ensemble-averaged puff outline and concentration field can be predicted.

Amino acid changes within HIV protease or its substrate that decrease the susceptibility to protease inhibitors represent a highly complex issue still not yet fully understood. Various mechanisms by which this often complicated pattern of mutations influence drug binding needs to be analyzed on a molecular level by a series of methods including experiments with recombinant viruses, biochemical enzyme analysis, structural and thermodynamical studies or molecular dynamics. Each result may help to complete the overall picture of protease inhibitor resistance evolution and therefore contribute to the design of more powerful 3rd generation HIV/AIDS drugs. This thesis presents several analyses of HIV resistance development on molecular level. We have focused on the nelfinavir resistance pathway, lopinavir mutation score, emergence of amino acid insertions in HIV protease gene and their contribution to protease inhibitor resistance and finally we analyzed a highly mutated protease species isolated from patients failing darunavir therapy. Since we are able to accomplish a wide combination of techniques, we could explain and put together some pieces of viral evolution considering the final steps of HIV life cycle and also provide knowledge necessary for novel inhibitor design. In recent decades, it was shown that belowground competition for some plants may take form of the tragedy of the commons (TOC). In these plants, the competing neighbours invest more in root systems than would be appropriate for optimal nutrient uptake for the group and also more than they do when grown alone. However, there is also strong evidence that other species do not follow TOC, and tailor their root system to best nutrient exploitation irrespectively of competitor presence. The root investment strategy of these plants should correspond to the ideal free distribution (IFD). In my thesis I focus on two aspects:  I use game theoretical models to explore, whether those strategies can coexist within species and also whether different species can coexist if they have different strategy. From this model I draw predictions, which I test by meta-analysis.  Using Agrostis stolonifera as a model, I test assumptions on nutrient and neighbour perception, which underlie TOC and IFD models. I show that according to mathematical models, those two strategies can coexist in different species in a community, but cannot coexist within a species. The study is focused on advanced life support (ALS) performed by rescue team physicians in an out-of-hospital setting. The first part of the study analyzes diagnostic possibilities and correctness of assumed aetiology of the sudden cardiac arrest during cardiopulmonary resuscitation (CPR) in the field. It introduces an original method of "Crosscheck Tables" and applies this method to 211 cases of CPR provided by physicians of the Emergency Care Service. The study demonstrates that significantly misleading conclusions can result from a global analysis of a set of cases. It stresses importance of diagnostic analyses applied to individual cases. The second part of the study concentrates on changes in PETCO2 level as related to the return of spontaneous circulation (ROSC - Return of Spontaneous Circulation) as opposed to the circulation fully dependent on chest compressions. The study demonstrates that in constantly ventilated patients undergoing CPR in an out-of-hospital setting, PETCO2 is significantly higher (about 10 mmHg) after ROSC than before ROSC. It demonstrates that a sudden increase in PETCO2 exceeding 10 mmHg is likely to indicate the moment of ROSC. Vasopressin and its analogues in treatment of cardiac arrest Introduction: Recent guidelines for cardiopulmonary resuscitation (CPR) recommend use of drugs as an integral part of treatment. Vasoactive drugs have been used in resuscitation to increase myocardial and cerebral perfusion during chest compressions. Resuscitation research is trying to find alternative vasopressors to adrenaline, for which the long-term survival benefit has not been proven. Successful use of terlipressin in cardiac arrest (CA) was documented in small studies in children and some rare case reports. Terlipressin has never been tested in CA of cardiac aetiology in any clinical or experimental study. Objectives: We aimed to evaluate the role of catecholamines and non-adrenergic vasopressors, vasopressin and terlipressin, during CPR based on review of selected trials. Introduction of a standardized experimental porcine model of ventricular fibrillation (VF) was done for research of therapeutic interventions during CPR. We aimed to evaluate the effects of terlipressin with adrenaline on perfusion of vital organs during prolonged CPR compared to placebo with adrenaline. Title Sudden cardiac death in athletes in the Czech Republic and abroad Objectives The primary aim of the diploma thesis is to collect data about sudden cardiac death in athletes in the Czech Republic and abroad. The next aim is to create summary about health care concerning athletes in Czech sporting environment. The final step is to recommend some approaches and advices to prevent the sudden cardiac death in athletes. Methods The main method used diploma thesis is the analysis of documents, specifically the autopsy protocols providing professional information about sudden cardiac death in athletes in our country. Another method is questioning which allows to determine current state of athletic health in sports federations. The final method chosen for the diploma thesis is the interview which was arranged with football player Michal Meduna who survived a cardiac collapse during football match. Results The author of the thesis has found that the biggest problem of health care in athletes is the implementation and quality of sports health examination. In many cases the defect, bad hearth condition or congenital heart defect was found as late as autopsy was performed. Empirická část práce je věnována kvalitativnímu výzkumu se zaměřením na zjištění spokojenosti respondentek s poskytovanou ošetřovatelskou péčí po operaci mimoděložního těhotenství. Gene regulatory networks, underlying the molecular regulation of eye development are conserved across many animal phyla. Genes from the Pax family of transcription factors are one of the most conserved members through the evolution, regulating the development of crucial parts of eye, including the photoreceptor cells. Pax transcription factors are considered to be regulators of opsins, molecules providing the conversion of the light stimulus into the electrochemical signalisation in the photoreceptors cells. In this thesis, pax6 and pax2/5/8 transcription factors are investigated as potential regulators of eye development in Platynereis dumerilii. pax6 and pax2/5/8 transcription factors are tested as potential regulators of the r-opsin in Platynereis, based on the observed early expression onsets of these genes. Wild-type expression analysis of pax6 and pax2/5/8 using the whole mount RNA in-situ hybridization is provided, accompanied by the initial analysis of the Platynereis pax6 knockout line. pax6 heterozygote mutants are shown to be viable and able to reproduce, however, homozygote mutation of pax6 in Platynereis is lethal. Introduction: Induction of therapeutic hypothermia to target body temperature of 32 - 34 řC for 12 - 24 hours may reduce post-cardiac arrest brain injury in patients resuscitated from out- of-hospital cardiac arrest (OHCA). It has been recommended that the target therapeutic temperature should be reached as soon as possible. Thus, prehospital initiation of cooling appears to be a method of choice. Aim of the study: We performed three studies to assess a feasibility and clinical effectivness of prehospital therapeutic hypothermia in the setting of emergency medical service in the Czech republic and to optimize cooling procedure. The selected cooling method was rapid intravenous administration of 4 řC cold normal saline. Materials and methods: Forty consecutive cardiac arrest patients were treated by prehospital administration of 4řC cold normal saline with the target dose of 15 - 20 ml/kg in a prospective multicenter study PRE-COOL (Pre-Hospital Cooling in Cardiac Arrest Patients). The results were compared with 40 retrospective control group patients who did not undergo any cooling attempt in the field. Religious conversion is an interesting phenomenon that crosses religious beliefs. This phenomenon intersects with many various disciplines, from medicine through the scientific disciplines such as anthropology, sociology, religious studies, psychology and theology. Religious conversion may on the one hand be a demonstration of deep spiritual experience and on the other, the result of strong pressure from others. We can get closer to the truth only through the authentic testimonies of the converts. This is why I have chosen the theme of the religious conversion in the context of "pure experience". We can see differences among conversions to various religions. But it is necessary to say, that I am interested in conversion to Christianity. But the Christianity of two thousand years ago is not the Christianity of today. The Apostle Paul joined a marginal movement at a time when such acceptance was punishable by death. Today, however, converts have joined millions of faithful around the whole world. And this is why religious conversion to Christianity is a lively phenomenon. The Thesis deals with the issue of provision of advanced life support in a Hospital. The main focus is the process, algorithm, of successive individual steps, its correct performance in practice should lead to the provision of effective CPR or to minimize subsequent post-resuscitation complications. The Thesis is mainly focused on non-medical health worker, as an important member of the healthcare team, his tasks, responsibilities and competences. The theoretical part will be processed according to the current guidelines of the European Resuscitation Council. Within the subsequent empirical part is included the methodology of realized research, the process of data collection, and the final interpretation of the results, followed by analysis and discussion. The survey is conducted by quantitative research, using the questionnaire survey. The aim of the research was to determine respondents' level of theoretical training, ways of continuing education and current conditions at selected workplaces. Based on detected results, the steps for nursing practice are proposed. This Diploma thesis is focused on the issue of nursing care of acute abdominal pain in children under six years old. The abdominal pain of children can be the first symptom of an acute abdominal pain which can directly endanger a child´s life. Neglecting these troubles can lead to great health problems of a complicated character. Four research goals were set within the research. The first objective was to find specifics of acute abdominal pains of children up to six years old. The second objective was to find possible results of acute abdominal pains in children under six years old in the post-operative period. The third goal was to monitor the issue of acute abdominal pains in childhood from the point of view of a nurse and the fourth should have found the knowledge of parents about acute abdomeninal pains in childhood. The Diploma thesis strived to penetrate into the issue of this large chapter of acute medicine, highlight its specifics related to acute abdominal pains in childhood and find out how parents are informed about the existence of this type of illness. To complete the objectives of this Diploma thesis, we used a qualitative research model. This research was done by the technique of a semi-structured interview. The first research group consisted of eight nurses working in a standard paediatric ward or in a paediatric ward of the Intensive Care Unit (ICU). The second group consisted of parents, specifically mothers who responded based on their knowledge and general awareness gained. A technique of open coding a method of pencil and paper was used for processing the results of these interviews. Seven research questions were set: 1. What are the specifics of nursing care in acute abdominal pain in childhood? 2. What medical complications appear during acute abdominal pain in childhood? 3. What are the specifics of pre-operative and post-operative care in acute abdominal pain in childhood? 4. What is the post-operative period in children with acute abdominal pain? 5. What results can appear in acute abdominal pain in childhood? 6. How do nurses see the issue of sudden acute abdominal pain in childhood? 7. What is the parents´ knowledge about acute abdominal pains in childhood? Within the process of this Diploma thesis, a booklet was created. This should serve as information material for parents, which will help them to explain various terms and potential situations within this extensive diagnosis. This booklet will be at disposal in day nurseries and the outpatient department of the paediatric ward in České Budějovice.

In recent years, clinical studies have shown that the most effective method of treatment to reduce the incidence of sudden cardiac death in patients at increased risk of malignant arrhythmias is to provide the patient with an implantable cardioverter-defibrillator (ICD). Complications associated with the transvenous implanted system, especially lead disorders and systemic infections, and have led to the development of a so-called subcutaneous defibrillator, which is thought to minimize these complications. Despite the medical benefits of implantable defibrillator treatment, which is generally well accepted by patients, anxiety, depression, and deterioration in their quality of life have been reported in clinical trials examining patients' psychopathology and quality of life after defibrillator implantation. Methodology: The objective of the diploma thesis was to find out which group of patients better assesses the quality of life depending on the type of implanted defibrillation device (ICD and S-ICD). For the purposes of the survey, the method of quantitative research was chosen in the form of its own questionnaire survey. Testing Structural Changes Using Ratio Type Statistics Barbora Peštová Charles University in Prague, Faculty of Mathematics and Physics, Department of Probability and Mathematical Statistics, Czech Republic Abstract of the doctoral thesis We deal with sequences of observations that are naturally ordered in time and assume various underlying stochastic models. These models are parametric and some of the parameters are possibly subject to change at some unknown time point. The main goal of this thesis is to test whether such an unknown change has occurred or not. The core of the change point methods presented here is in ratio type statistics based on maxima of cumulative sums. Firstly, an overview of thesis' starting points is given. Then we focus on methods for detecting a gradual change in mean. Consequently, procedures for detection of an abrupt change in mean are generalized by considering a score function. We explore the possibility of applying the bootstrap methods for obtaining critical values, while disturbances of the change point model are considered as weakly dependent. Procedures for detection of changes in parameters of linear regression models are shown as well and a permutation version of the test is derived. Then, a related problem of testing a change in autoregression parameter is studied....

The thesis deals with philosophical aspect of Dzhgchen, which is the core of the Yungdrung Bon traditon. It is very special spiritual tradition because of it's very special nondualistic view of man and universe. First part deals with the history and division of it's teachings and it's Dzogchen lineages. Next part focuses on explanation of differences in views of sutra, tantra and Dzogchen, mainly from the standpoint of the view of base, path, fruit, emptiness, clarity, conscioussness and mind and it's nature. Next part provides translation of the text Twelve little tantras and it's deep explanation. Last part, and most important one, focuses on philosophical examination of some key aspects of Dzogchen, mainly on it's view of autenticity of mindnature. This part also compares Pramenides's fragments with some key points of view of Buddhism and Dzogchen, which may lead to some unexpected conclusions about man's thinking and the problem of it's intentionality.
