O Ensino Médio, última etapa da Educação Básica, tendo em vista sua função educacional, deve ir além da formação profissional oferecendo aos jovens novas perspectivas culturais para que possam expandir suas potencialidades e oportunidades. É nesse contexto que o Estado de Minas Gerais implementa o Programa Reinventando o Ensino Médio (REM) a partir do ano de 2012 em 11 escolas piloto em Belo Horizonte, no ano de 2013 em mais 122 escolas e a partir do ano de 2014 nas demais escolas do estado que oferecem o Ensino Médio. A presente pesquisa analisa o processo de implementação do REM em duas escolas pertencentes à Superintendência Regional de Ensino de Carangola durante o ano de 2013. Para tanto, utilizamos a metodologia do estudo de caso, sendo as informações e dados utilizados coletados por meio do Sistema de Administração Escolar/SIMADE. Também foram feitas entrevistas aos diretores das Escolas “A” e “B”, ao Coordenador do REM da Escola “B”, aos Membros da Equipe NAPEM e aplicados questionários de pesquisa aos alunos matriculados no 1º ano do Ensino Médio Noturno do REM das Escolas “A” e “B” e ainda, a leitura do Livro de Atas de Reuniões Pedagógicas das Escolas “A” e “B”. A análise dos dados permitiu que fossem apresentadas considerações sobre a implementação do REM nas duas escolas da regional. Objetivou-se com essa pesquisa apresentar à Direção da SRE de Carangola e aos Diretores das unidades escolares um plano de ação que visa à interferência na realidade apresentada, organizar e intensificar as ações de implantação do Programa Reinventando o Ensino Médio almejando corroborar para que esse programa seja implementado de forma positiva e alcance seus objetivos de ressignificação de Ensino Médio.
Due to its educational aspects, the high school, last stage of basic education, needs to go beyond vocational training offering new cultural perspectives to the students in view of expand their potentialities and opportunities. In this scenario, Minas Gerais state implanted the REM program (Reinventing Minas Gerais Program) in 11 schools in 2012, 122 schools in 2013 and extending to all high schools in the state in 2014. This research analyzes the implementation REM process in two schools belonging to the SRE (Regional Office of Education) of Carangola during the year of 2013. Therefore, we use the methodology of the case study, analyzing information and data collected through the use of Sistema de Administração Escolar/SIMADE (Student Administration System), interviews with the principals of “A” and “B” schools, with the REM coordinator of “B” school, and with members of NAPEM team, questionnaires were applied to students of “A” and “B” schools enrolled in the 1st year of high school nocturne of REM and also the reading of the Pedagogical Meetings Book (Livro de Atas de Reuniões Pedagógicas) of the “A” and “B” schools. These data analysis allowed to be presented considerations about REM implementation in the two schools. The objective of this research was to provide to the schools principals and to the SRE direction an action plan that aims to intervene in presented reality, organize and intensify REM’s implementation actions and to corroborate in a positive and successful accomplishment of the program goals, reframing the High School system.

The diploma thesis is focused on the area of real estate agents. It deals with the analysis of awareness of the real estate offices with direct targeting on the real estate company Gaute a.s. It identifies the current situation on the real estate market and the respondents' satisfaction with the services offered. In the theoretical part, the reader is made familiar with key concepts related to the topic, followed by the analysis, which is composed of the performance of Gaute a.s. preparation questionnaire and evaluation. Based on the findings recommendations are proposed, which will serve to raise awareness of the real estate office.

Identification et caractérisation de la première N-acylphosphatidyléthanolamine synthase chez A. thaliana. Les N-acylphosphatidyléthanolamines (NAPE) sont des phospholipides complexes peu abondants au sein des membranes biologiques mais largement répandues dans différents organismes. Outre ses fonctions de stabilisation des membranes ce lipide est davantage connu pour être le précurseur des N-acyléthanolamines (NAE) qui sont impliquées dans de très nombreuses voies de signalisation chez les plantes (lors de la germination, du développement racinaire, de l’induction de gène de défense, etc.) comme chez les animaux (apoptose, ligand des récepteurs endocannabinoïdes, notion de satiété, etc.). Au début de ma thèse, les gènes codant pour les enzymes impliquées dans les différentes étapes de la voie métabolique des NAE (e.g NAPE-PLD, FAAH1 et 2) ont été caractérisées exceptées le ou les gène(s) codant pour l’enzyme catalysant la synthèse de NAPE précurseurs de ces lipides. Une étude bioinformatique a permis d’identifier de nouvelles séquences codantes pour des acyltransférases putatives chez A. thaliana dont celle du gène At1g78690. La caractérisation fonctionnelle de cette enzyme a été déterminée après son expression hétérologue chez E.coli sur fractions membranaires et protéines purifiées. Puis le profil d’expression génique, la localisation cellulaire de la protéine ainsi que son activité ont été étudiés chez les plantes à partir notamment de mutants d’A. thaliana (ADN-T et « 35S »). Les résultats obtenus au cours de cette étude ont permis d’identifier et de caractériser la première NAPE synthase chez les plantes
Discovery and characterization of an A. thaliana N-acylphosphatidylethanolamine synthase. N-acylphosphatidylethanolamine (NAPE) is a widespread, albeit minor, membrane phospholipid in various organisms. Besides its stabilizing properties to membranes bilayers, NAPE is known to be the precursor for N-acylethanolamine (NAE) synthesis. NAE have been shown to regulate a variety of physiological functions in both plants (germination, root development, gene induction, etc.) and animals (apoptosis, ligand for cannabinoid receptors, satiety properties, etc.) At the beginning of my PhD, the genes encoding the enzymes involved in the different steps of NAE metabolism were well characterized (e.g NAPE-PLD, FAAH 1 and 2), with the exception of the NAPE synthase gene(s). A bioinformatic study allowed the identification of coding sequences for putative new acyltransferases in A. thaliana, such as the At1g78690 gene. After expression in E. coli, the functional characterization of At1g78690p was carried out by analyses of the lipid content and by enzymatic assays using membrane fractions or purified proteins. The localisation of the protein and its activity were also studied in A. thaliana mutants (ADN-T and “35S”). This study shows the identification and characterization of the first NAPE-synthase in plants

Abstract:
The issue of competences of central bank in monetary or foreign exchange policy, the optimal adjustment of regulation and supervision of financial market and also the necessary independence of this institution is subjected to intensive discussion at the present time economic policy. In the theoretical part I will define basic concepts and definitions relating to this issue. From a theoretical point of view I will describe the concept and function of central banking, the independence of central bank, its types and optimal settings. I will also characterize the regulation and supervision of financial markets with regard to the definitions, models, systems, objectives and instruments of this issue. I will define the basic theoretical aspects of monetary and foreign exchange policy. In the analytical part I will process an analysis of selected indicators of activities and powers of the central bank in our country since 1989, mainly from the independence of the central bank, setting of monetary policy, financial market supervision and foreign exchange policy. I will make a comparison of changes in the development and activities of central banks in these areas with regard to trends in the legislation and their powers. The aim of this thesis is to analyze the development of selected indicators of activity of the central bank in our country with a focus on the transformation of the Czech economy with a summary of significant changes.

Abstract:
The Rufous-naped lark Mirafra africana complex consists of 22 subspecies spread across the African continent. Several of the subspecies have recently been suggested to potentially be treated as separate species. In this study a comparative analysis was done on the song from seven of the subspecies: M. a. africana, M. a. athi, M. a. grisescens, M. a. kabalii, M. a. nyikae, M. a. transvaalensis and M. a. tropicalis. The results showed that M. a. athi, M. a. kabalii and M. a. nyikae are all very divergent from each other as well as from the other four subspecies. In contrast, M. a. tropicalis, M. a. grisescens, M. a. africana and M. a. transvaalensis are not clearly separable from each other. Based on the results, I suggest that M. a. athi, M. a. kabalii and M. a. nyikae can be classified as separate species, with M. a. africana, M. a. tropicalis, M. a grisescens and M. a. transvaalensis forming a fourth species (M. africana sensu stricto). Finally, I conclude that this study shows that more studies need to be done on the subspecies of the Mirafra africana complex.

Research and practitioners alike have underscored the importance of customer value creation in marketing. For any marketing practice to be successful, it must first create value for customers. This is also true for the practice of relationship marketing, which is enjoying popularity among organizational marketers. However, there has been a lack of research done on the predictive effects of relationship marketing constructs in relation to buyer perceived value in organizational marketing. In other words, we still know little about the mechanism through which a good relationship enhances customer perceived value. The primary purpose of this study is to conceptually develop and empirically test a model that explains how the quality of a buyer-supplier relationship affects the buyer's value judgment in an organizational purchasing context. In the study, relationship quality is defined as comprising three different but mutually reinforcing dimensions: mutual trust, mutual commitment, and interdependence. Perceived value is conceptualized as an overall assessment of the utility of an offering based on the benefits and costs of accepting an offering. The conceptual model specifies the several routes through which relationship quality impacts buyer perceived value. First, a good relationship increases relationship benefits and reduces relationship costs, which in turn influences customer value perception - the higher the relationship benefits (and lower relationship costs), the higher the customer perceived value. Second, a good relationship reduces decision-making uncertainty. Lower decision-making uncertainty is hypothesized to increase the effects of perceived purchase episode benefits and perceived purchase episode costs on perceived value. The model was generally confirmed by an empirical test based on data collected from a national sample of purchasing managers in the United States.

Abstract:
Endocannabinoids (eCBs) are lipophilic signals that are produced by postsynaptic neurons in an activity-dependent manner, and signal in a retrograde fashion to modulate neurotransmitter release. As such, eCBs are highly involved in synaptic plasticity, a process that strengthens or weakens synapses. eCB-mediated synaptic plasticity is involved in many brain processes including learning, short-term memory, and adaptive reward, which are processed in the hippocampus and ventral tegmental area (VTA), respectively. However, the expression of eCB biosynthetic enzyme mRNA within hippocampal and VTA neurons, as well as the relationship between these mRNA species and the occurrence of synaptic plasticity, remains unclear. The goal of these studies was to demonstrate the expression pattern of eCB biosynthetic enzyme mRNA within hippocampal and VTA neurons, and to describe the relationship between synaptic plasticity and mRNA expression. Using whole-cell electrophysiology and real-time quantitative PCR, I tested hippocampal and VTA neurons for the presence of eCB biosynthetic enzyme mRNA and described the relationship between these enzymes and synaptic plasticity. The data presented herein demonstrate the importance of eCB signaling within the hippocampus and VTA and the expression patterns of eCB biosynthetic machinery within several neuron types. These data provide evidence that eCB signaling plays a critical role in learning, short-term memory, and adaptive reward.

Abstract:
Endocannabinoids are endogenous lipid mediators involved in various biological processes, and have immunomodulatory and anti-inflammatory activities. Anandamide (arachidonoyl ethanolamine, AEA) and 2-arachidonoyl glycerol (2-AG) are the main representatives of this group. The endocannabinoid receptors CB1 and CB2 with AEA have been found in human HaCaT keratinocytes and fibroblasts, but the metabolic pathway leading to endocannabinoid production in the skin has not been fully elucidated. This study aimed to investigate the profile of endocannabinoids and their main metabolizing enzymes in human skin cells and assess whether omega-3 polyunsaturated fatty acids (n-3 PUFA) altered these profiles. In addition, an investigation was carried out to check whether UV radiation could stimulate the production of endocannabinoids and N-acylethanolamines (NAE) in human skin cells. For this purpose HaCaT keratinocytes and 46RB.1N fibroblast cells were treated with 10 and 50µM of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA) or oleic acid (OA) in the presence or absence of UVR (15mJ/cm2). Data suggest that n-3 PUFA may both directly (by up-regulating NAPE-PLD levels) and indirectly (by decreasing FAAH levels) increased endocannabinoid and NAE levels in HaCaT keratinocytes and 46BR.IN fibroblasts. DHA treatment significantly decreased COX-2 expression in the absence of UVR and inhibited UVR-induced COX-2 overexpression in 46BR.IN fibroblasts. In contrast, DHA appeared to induce COX-2 up-regulation in the absence of UVR and did not prevent UVR induced COX-2 up-regulation in HaCaT keratinocytes. EPA appeared to induce COX-2 down-regulation in the absence of UVR and did not prevent UVR induced COX-2 up-regulation in both HaCaT keratinocytes and 46BR.IN fibroblasts. UVR did not have any significant effect on endocannabinoid and NAE biosynthesis. However, UVR induced endocannabinoid production in some experiments of this study. A clinical study was carried on 16 volunteers from two different ethnic groups and two different skin types. The purpose was to assess the effect of UVR on the serum endocannabinoids and NAE, therefore, the volunteers were subjected to multiple doses (1.3, SED/ 6 min) of UVR for 6 weeks. Data showed that UVR did not have major effect on human serum NAE in both skin phototypes II and V but increased 2-AG in human serum in both skin types but the more pronounced effect was evident in skin phototypes V rather than in skin phototypes II. Human serum docosahxaenoylethanolamide levels were found to be higher in White Caucasians group (skin phototypes II). Based on these it can be concluded that n-3 PUFA and UVR alter the endocannabinoids and NAE profile in HaCaT keratinocytes and 46BR.IN fibroblasts. In addition, results of the clinical study indicated that UVR has no major effects on serum endocannabinoids or NAE therefore, further studies are required to address this question in vivo.

This diploma thesis deals with children remains in archeological cultures of the Bronze Age at the area of Czech Republic. 1318 findings have been gathered and put into a database together with all available details. Special attention has been set upon multiple child burials, deposition of child remains into settlement pits, presence of charities and individuals with exceptionally rich funerary goods. Individual chapter has been dedicated to phenomena connected with children in the Bronze Age such as burial of unburned bodies in jars, anthropophagy and human sacrifices. Terns usually will aggregate in the same reproduction area during breeding season. The color pattern of eggs may match the background, and it is important for breeding adults to distinguish their own eggs, especially in colonies with high density. This research was carried out at three islands, Huolung Shoal, Baisha and Jishan Islets of Penghu from May to August of 2007 and 2008. The experiment was divided into three parts, egg moving (EM) experiment, nest modification (NM) experiment and egg switching (ES) experiment. Terns spent more time for searching their own nests of each of the experiment situation. In the EM experiment 69.7% Black-naped Tern adults settled on original nests without eggs in 2007; 52.0% Roseate Tern in 2007 and 29.6 % in 2008 did the same. In the NM experiment, the behavior patterns of returning adults were significantly different between each operation and the control group. Adults were confused by the interference of the experimental conditions. In some occasions, adults settled on incorrect eggs quietly in ES experiment. In conclusion, terns have low ability of egg recognition but high ability of nest recognition. Terns presumably can recognize nests with the nearby environmental landmarks, but not be able to recognize eggs.

Black-naped Oriole (Oriolus chinensis), Oriolidae, Passeriformes, is a member of old world songbirds. It has a wide distribution all over the eastern Asia, and is divided into 18 to 20 subspecies by differences in breeding sites, residents or migrants, morphological characters and songs. The populations breeding in Taiwan are classified as subspecies Oriolus chinensis diffusus. Black-naped Orioles inhabit in wooded habitats with a preference for tall secondary forests and disturbed wooded habitats and the staple foods of Black-naped Orioles are fruits, larvae, pupae and imagines of insects. This study was intented to understand the habitat preference and to explore the fundamental ecological data of Black-naped Oriole in Taiwan. Breeding seasons of Black-naped Oriole started from March and ended in July, and reached its peak from April to June. Most of the birds had two broods each year. Females built nests and incubated for 17-21 days alone, and males shared feeding and protecting juveniles after hatching. The birds of Pingtung population often built their nests on tall Albizia falcate and those of Fenglin population on Pachira macrocarpa. The breeding success was 23.3% in Pingtung and 18.2% in Fenglin. The breeding failure were due to human disturbance, climate, and poaching. The materials used for nest were bark, grass, leaves, and some artificial materials. Black-naped Orioles prefer to build nests at the same tree but on different branches. The nests were often closed to nests of Dicrurus macrocercus. Cooperative defenses between these two birds were observed.

Environmental, economic and raw material concerns encourage researchers to search for alternative pulping and bleaching methods. Organosolv pulping and bleaching are well-known alternative methods to traditional pulping and bleaching processes. The neutral alkali earth metal (NAEM) salts-catalyzed methanol pulping process produces high yield screened pulps from all lignocellulosic materials. However, when such refractory softwoods as spruce, are cooked by means of organosolv pulping at high alcohol concentrations (>70%), the penetration of liquor into the wood cell walls is partially blocked due to the frequent presence of aspirated pits in the air-dried fibers and air in the lumen. As a result of the decrease in liquor penetration, the diffusion of chemicals to the reaction sites is also decreased. Consequently, delignification and fiber liberation are delayed. To study whether increasing the cooking pressure changes the pulpability of air-dried wood chips (MC=7%), this study was set up in such a way that three major cooking variables (time, temperature, and particularly pressure) were varied in a split-plot experimental design with at least two replicates, and their individual and combined effects on delignification were assessed. The main objective of this research was to examine the effects of high pressure on spruce organosolv pulping behavior in terms of fiber liberation point (FLP), degree and selectivity of delignification, screened yield and residual lignin content of the resulting pulps at the FLP. The cooking conditions were optimized to achieve high-screened yield and low Kappa number pulp at the FLP. In this research, batch, isothermal delignification of spruce wood chips was carried out by means of high-pressure NAEM-catalyzed organosolv pulping. The cooking liquor contained methanol + water (80/20 by volume), and NAEM salts with liquor/wood ratio of 10 mL/g wood, all of which remained constant throughout the course of the cooks. The primary cooking variable was hydraulic reactor pressure (500-4000 psi) with cooking time (10-120 min) and temperature (190°-210°C) as secondary variables. Models were then developed to predict the responses of pulping, such as total yield, reject content, lignin-free yield, Kappa no., amounts of lignin and sugars dissolved and remaining. It was found that, in addition to cooking temperature and time, pressure had also a statistically significant effect on pulping behavior of spruce wood and on pulp chemical properties. Pulping was pressure sensitive particularly when cooking at low temperatures (190°-200°C) for short cooking times until the fiber liberation point was reached. Pressure was more effective in lowering reject content and consequently increasing screened-yield, than in reducing Kappa number. Thus, the issue of screen rejects could be resolved by applying high pressure (optimally 2000 psi) to the softwood organosolv pulping system while producing pulps of various qualities/grades that can be utilized for different applications. The high pressure did not affect residual mannose and xylose contents significantly; their dissolution depended on cooking temperature and time. Arabinose and galactose disappeared very early in the cook before the FLP. The topochemistry of delignification and fiber liberation (low reject) can be attributed to the removal of arabinose and galactose, in addition to the partial removal of pectins and lignins from the compound middle lamella. It was also found that there was a significant correlation between pH of spent liquor and chemistry of pulping (pulp composition). Dissolution of mannose and xylose were associated with the pH of the spent liquor during different stages of cooking. In all cases, the pH decreased from its initial value of 6.5 of the fresh cooking liquor and reached its optimum level of 4.2±0.2 at approximately the fiber liberation point, buffered by NAEM catalysts. Furthermore, dissolution of mannose and xylose were highly intercorrelated (r=0.99). Removal of xylose and mannose were also highly correlated with delignification, i.e. r=0.885 and 0.926, respectively. Moreover, the reject content of pulp was also correlated with xylose, mannose and lignin contents of the pulp, having a correlation coefficient (r) of 0.628, 0.677 and 0.838, respectively. Mathematical models were then developed, tested, validated and finalized. Using SAS software to run different subset selection procedures and taking several parameters into consideration, MAXR technique was found to be the most appropriate procedure to use when building the multiple regression models. All model-building procedures were followed and explained for the future modeling uses. The final models can predict many properties of the softwood NAEM pulp from the cooking variables with high accuracy (R²>92%) and thus can be applied for process control purposes in the pulp mill. The response variables modeled include total yield, reject content, Kappa number, residual and dissolved lignin, lignin-free yield, and mannose and xylose contents of the NAEM pulp. The general form of the models for most responses is as follows: Y = b₀ - b₁T + b₂Tt – b₃t – b₄ t[sup ½] – b₅P R²=92-99%. Optimum cooking conditions to obtain early fiber liberation, low reject content (<2%), and high yield screened pulp were found to be: pressure (P) of 2000 psi for 40 min and 80 min (t) at 200° and 190°C (T), respectively. The resultant pulps had screened yields of 57.5 and 56.2% and Kappa numbers of 63 and 38, respectively, both with a lignin-free yield of >53%. Increasing the cooking temperature above 200°C is not recommended since it extensively dissolved hemicelluloses and degraded cellulose, resulting in a lower pulp yield and viscosity. The effects of the pulping conditions on the response variables, as predicted by the models, are consistent with the known physical and chemical reactions occurring during the organosolv pulping process. The distinctly different behavior of high-pressure NAEM-catalyzed organosolv pulping, compared to uncatalyzed organosolv pulping, seems to be due to the unique topochemistry of delignification (selective removal of lignins, arabinose, galactose, and pectins from the compound middle lamella), different mechanisms of reactions, and non-condensed structure of the dissolved and residual lignins.

The successful rates of Passeriformes reproductive are generally low. The factors include predation of predators, harsh climate, and reusable resources and so on. In order to understand how the relationship between nesting sites and environmental factors influence the reproductive success rates of Black-naped Blue Monarch (Hypothymis azurea), the investigation in nesting sites and habitat traits was started in Linnei Township and Douliu hilly area, Yunlin County in 2007 and 2008, setting up the basic data of bird nests. The composition of the nest was an open cupped and the layers from exterior to interior were named as attachment, out layer, structure, and the lining. The lining layer was not obvious, not all the nests had it. Each layers had various materials, and the bamboo leaf, moss and silk appear the most, and every nest had them. A total of 58 nests, with 22 successful and 36 failed. The main reason of failure comes from predators, secondly the harsh climate and other unknown reasons. The comparison of all the habitat traits shows a significant difference in the height of the nest and the distance from the canopy to the nest, the other traits shows no significant difference; A total of 36 nests collected and analyzed shows no significant difference between the nest size, weight and the success rates. Collected nests were then separated apart to see the composition. The birds used a variety of nesting plants, but the selection of the nesting branches had a common character, which was a Y-shaped twig or a H-shaped vine.

Le système endocannabinoïde (eCB) est présent dans le système nerveux central (SNC) de mammifères, incluant la rétine, et est responsable de la régulation de nombreux processus physiologiques. Bien que la présence du récepteur cannabinoïde de type 1 (CB1R) a bien été documenté dans la rétine de rongeurs et primates, il y a encore une controverse quant à la présence du récepteur cannabinoïde de type 2 (CB2R) au niveau du SNC. En utilisant la microscopie confocale, nous sommes les premiers à signaler les patrons d’expression du CB2R dans la rétine de singe. Nos résultats démontrent que le CB2R est exprimé exclusivement dans les cellules de Müller de la rétine du singe. En outre, nous avons comparé les différents patrons d’expression du système eCB dans la rétine de la souris, du toupaye, ainsi que du singe vervet et macaque. Nous rapportons que les distributions de CB1R, FAAH (fatty acid amid hydrolase), MAGL (monoacylglycerol lipase) et DAGLα (diacylglycerol lipase alpha) sont hautement conservées parmi ces espèces alors que CB2R et NAPE-PLD (N-acyl phosphatidylethanolamine phospholipase D) présentent différents profils d'expression. CB2R n'a pas été détecté dans les cellules neuronales de la rétine des primates. L’immunoréactivité de NAPE-PLD est présente dans les couches de la rétine de souris et toupayes, mais a été limitée à la couche des photorécepteurs des singes vervet et macaque. Pour étudier les corrélats neuronaux et le rôle de la signalisation du système eCB dans la rétine, nous avons établi un protocole standard pour l'électrorétinographie (ERG), puis enregistré la réponse ERG de la rétine après le blocage des récepteurs avec des antagonistes spécifiques pour CB1R (AM251) et CB2R (AM630). Comparé au témoin, dans des conditions photopiques, et à certaines intensités faibles du stimulus, le blocage de CB1R diminue l'amplitude de l'onde-b, alors qu’à des intensités plus élevées, le blocage de CB2R augmente l'amplitude des deux-ondes a et b. De plus, le blocage des récepteurs cannabinoïdes provoque une augmentation de la latence des deux ondes a et b. Dans des conditions d’adaptation à l'obscurité, le blocage de CB1R et CB2R réduit l’amplitudes de l'onde a seulement à des intensités plus élevées et réduit l’onde b à intensités plus faibles. Des augmentations significatives de latence ont été observées dans les deux cas. Ces résultats indiquent que les récepteurs CB1 et CB2 chez les primates non humains sont impliqués dans la fonction rétinienne conditions photopiques. En outre, nous avons évalué le profil d'expression du CB1R, de FAAH et de NAPE-PLD au-delà de la rétine dans le corps géniculé latéral des singes et nous rapportons pour la première fois que CB1R et FAAH sont exprimés davantage dans les couches magnocellulaires. La NAPE-PLD a été localisée à travers les couches magno- et parvocellulaires. Aucune de ces composantes n’est exprimée dans les couches koniocellulaires. Ces résultats nous aident à mieux comprendre les effets des cannabinoïdes sur le système visuel qui pourraient nous mener à trouver éventuellement de nouvelles cibles thérapeutiques.
The endocannabinoid (eCB) system is present in the mammalian central nervous system, including the retina, and is responsible for the regulation of many physiological processes. Anatomical and functional data collected in the retina indicate that cannabinoid receptors are important mediators of retinal function. Although the presence of the cannabinoid receptor type 1 (CB1R) has been documented in the rodent and primate retina, there is still some controversy regarding the presence of the CB2 receptor (CB2R) within the central nervous system. By using confocal microscopy, we are the first to report the distribution patterns of CB2R in the monkey retina. Our results show that CB2R is expressed exclusively in the Müller cells of the primate retina. Furthermore, we compared the eCB system distribution patterns in the retinas of mice, tree shrews, and vervet and macaque monkeys. We report that CB1R, FAAH, MAGL, and DAGLα distributions are highly conserved among these 3 species whereas CB2R and NAPE-PLD exhibit different expression patterns. CB2R was not detected in the neuroretinal cells of primates. NAPE-PLD immunoreactivity was present in the retinal layers of mice and tree shrews but was restricted to the photoreceptor layer in both species of primates studied. To study the neural correlates and the role of eCB signaling in the retina, we first established a standard protocol for electroretinography (ERG) and then recorded the ERG response of the retina after blocking receptors with specific antagonists for CB1R (AM251) and CB2R (AM630). Compare to control, in photopic conditions, at certain low flash intensities, only the blockade of CB1R decreases the amplitude of the a-wave and b-wave, while at some high flash intensities, blockade of CB2R increase the amplitude of both a- and b-waves. Also the blockade of the cannabinoid receptors causes an increase in the latency of both a- and b-waves. In dark-adapted eyes, blockade of the CB1R and CB2R reduces the a-wave only amplitudes in the higher intensities and decrease the b-wave in lower intensities. Some significant increases in latency were observed in both cases. These results indicate that CB1 and CB2 receptors in primates are involved in retinal function under photopic and scotopic conditions. In addition, we assessed the expression pattern of eCB components CB1R, FAAH, and NAPE-PLD beyond the retina in the dorsal lateral geniculate nucleus (dLGN) of primates and report for the first time that while CB1R and FAAH are more abundantly expressed in the magnocellular layer, NAPE-PLD is distributed throughout both the magno- and parvocellular layers. None of these components are expressed in the koniocellular layer. These findings augment our understanding of the effects of cannabinoids on the visual system and may lead to novel therapeutics targeted to eCB signaling.
