Abstract:

Abstract:
In animal tissues, NAEs (N-acylethanolamines), including N-arachidonoylethanolamine (anandamide), are primarily formed from their corresponding NAPEs (N-acylphosphatidylethanolamines) by a phosphodiesterase of the PLD (phospholipase D) type (NAPE-PLD). Recently, we cloned cDNAs of NAPE-PLD from mouse, rat and human [Okamoto, Morishita, Tsuboi, Tonai and Ueda (2004) J. Biol. Chem. 279, 5298–5305]. However, it remained unclear whether NAPE-PLD acts on endogenous NAPEs contained in the membrane of living cells. To address this question, we stably transfected two mammalian cell lines (HEK-293 and CHO-K1) with mouse NAPE-PLD cDNA, and investigated the endogenous levels and compositions of NAPEs and NAEs in these cells, compared with mock-transfected cells, with the aid of GC-MS. The overexpression of NAPE-PLD caused a decrease in the total amount of NAPEs by 50–90% with a 1.5-fold increase in the total amount of NAEs, suggesting that the recombinant NAPE-PLD utilizes endogenous NAPE as a substrate in the cell. Since the compositions of NAEs and NAPEs of NAPE-PLD-overexpressing cells and mock-transfected cells were very similar, the enzyme did not appear to discriminate among the N-acyl groups of endogenous NAPEs. These results confirm that overexpressed NAPE-PLD is capable of forming NAEs, including anandamide, in living cells.

Abstract:

Abstract:
Poor maternal nutritional status and substandard antenatal care, which result in increased women's risk, low birth weight and stillbirth, afflict many countries with weak or emerging economies even today. Studies that address the effect of extending nutrition awareness among pregnant women to the net outcome of pregnancy remain scarce. We aimed to compare and contrast the effect of a pragmatic nutrition awareness program for expectant mothers (NAPEM) on birth weight of the newborn with a control group who received no such nutrition awareness activity. The effect of variables of mode of newborn delivery, associated complications at birth, and APGAR score of the newborn were also assessed. A pragmatic intervention trial of an antenatal care (ANC) program that consisted in nutrition awareness was conducted involving 53 pregnant women. Awareness was given through one-to-one interview and through informational literature provided to the participants in the local language. A hospital registry for deliveries undertaken during the study period was screened for identification of variables. A control group of matched pregnant women (n= 53) was obtained from the same hospital registry from preceding years, when the nutrition awareness program was not executed. A statistically significant improvement in birth weight of the newborn was observed in the intervention group, where expectant mothers were made aware about desired nutrition during pregnancy. A reduced incidence of complications associated with pregnancy was also observed in the intervention group. Providing awareness about nutritional requirements during pregnancy and suggesting the pragmatic ways to meet them was shown to be one possible effective measure to deal with pregnancy-related undernutrition. We show the efficacy of the intervention for underprivileged regions of India marked by inadequate health care delivery and lower socio-economical standards. We discuss our findings in the context of available evidence-based guidelines.

Abstract:
N-Acyl-phosphatidylethanolamine phospholipase D (NAPE-PLD) (EC 3.1.4.4) catalyzes the final step in the biosynthesis of N-acyl-ethanolamides. Reduced NAPE-PLD expression and activity may contribute to obesity and inflammation, but a lack of effective NAPE-PLD inhibitors has been a major obstacle to elucidating the role of NAPE-PLD and N-acyl-ethanolamide biosynthesis in these processes. The endogenous bile acid lithocholic acid (LCA) inhibits NAPE-PLD activity (with an IC50 of 68 μm), but LCA is also a highly potent ligand for TGR5 (EC50 0.52 μm). Recently, the first selective small-molecule inhibitor of NAPE-PLD, ARN19874, has been reported (having an IC50 of 34 μm). To identify more potent inhibitors of NAPE-PLD, here we used a quenched fluorescent NAPE analog, PED-A1, as a substrate for recombinant mouse Nape-pld to screen a panel of bile acids and a library of experimental compounds (the Spectrum Collection). Muricholic acids and several other bile acids inhibited Nape-pld with potency similar to that of LCA. We identified 14 potent Nape-pld inhibitors in the Spectrum Collection, with the two most potent (IC50 = ∼2 μm) being symmetrically substituted dichlorophenes, i.e. hexachlorophene and bithionol. Structure–activity relationship assays using additional substituted dichlorophenes identified key moieties needed for Nape-pld inhibition. Both hexachlorophene and bithionol exhibited significant selectivity for Nape-pld compared with nontarget lipase activities such as Streptomyces chromofuscus PLD or serum lipase. Both also effectively inhibited NAPE-PLD activity in cultured HEK293 cells. We conclude that symmetrically substituted dichlorophenes potently inhibit NAPE-PLD in cultured cells and have significant selectivity for NAPE-PLD versus other tissue-associated lipases.

Abstract:
The study investigated development gap in social policy and social problem interaction by examining impact of National Poverty Eradication Programme (NAPEP) on rural communities in Akwa Ibom State, Nigeria. The specific objectives were to examine the relationship between non-participation of beneficiary communities in NAPEP project design at community level, corruption and policy implementation strategy on rural poverty reduction. The Ex-post facto research design was adopted to study communities in Oron and Udung Uko local government areas in Akwa Ibom State. Using a multistage sampling procedure, data were obtained from 400 respondents using Taro Yamene sample size determination formula. Data were analysed using Chi-square (X2) statistical technique. Main findings indicated that significant relationship exists between non-participation of beneficiary communities in NAPEP project design, corruption, policy implementation and poverty reduction in rural communities. The findings imply that NAPEP has not significantly achieved its goal of poverty reduction in rural communities. It has failed to elicit economic wellbeing priorities directly from target communities. Poverty can be reduced through accommodative and humanistic arrangements by allowing communities to identify welfare projects and eligible recipients. Rural dwellers have not experienced poverty reduction because of NAPEP development strategy gap.

Abstract:
Diverse metabolic disorders have been associated with an alteration of N-acylethanolamine (NAE) levels. These bioactive lipids are synthesized mainly by N-acylphosphatidylethanolamine-selective phospholipase D (NAPE-PLD) and influence host metabolism. We have previously discovered that NAPE-PLD in the intestine and adipose tissue is connected to the pathophysiology of obesity. However, the physiological function of NAPE-PLD in the liver remains to be deciphered. To study the role of liver NAPE-PLD on metabolism, we generated a new mouse model of inducible Napepld hepatocyte-specific deletion (Napepld∆Hep mice). In this study, we report that Napepld∆Hep mice develop a high-fat diet-like phenotype, characterized by an increased fat mass gain, hepatic steatosis and we show that Napepld∆Hep mice are more sensitive to liver inflammation. We also demonstrate that the role of liver NAPE-PLD goes beyond the mere synthesis of NAEs, since the deletion of NAPE-PLD is associated with a marked modification of various bioactive lipids involved in host homeostasis such as oxysterols and bile acids. Collectively these data suggest that NAPE-PLD in hepatocytes is a key regulator of liver bioactive lipid synthesis and a dysregulation of this enzyme leads to metabolic complications. Therefore, deepening our understanding of the regulation of NAPE-PLD could be crucial to tackle obesity and related comorbidities.

Abstract:
In May 2016, the National Academies of Sciences, Engineering, and Medicine (NASEM) released the report “Genetically Engineered Crops: Experiences and Prospects,” summarizing scientific consensus on genetically engineered crops and their implications. NASEM reports aim to give the public and policymakers information on socially relevant science issues. Their impact, however, is not well understood. This analysis combines national pre- and post-report survey data with a large-scale content analysis of Twitter discussion to examine the report’s effect on public perceptions of genetically modified organisms (GMOs). We find that the report’s release corresponded with reduced negativity in Twitter discourse and increased ambivalence in public risk and benefit perceptions of GMOs, mirroring the NASEM report’s conclusions. Surprisingly, this change was most likely for individuals least trusting of scientific studies or university scientists. Our findings indicate that NASEM consensus reports can help shape public discourse, even in, or perhaps because of, the complex information landscape of traditional and social media.

Abstract:
Abstract The California net energy system (CNES) was the reference for the development of most energy requirement systems worldwide, such as Nutrient Requirements of Beef Cattle (NASEM, Nutrient requirements of beef cattle, 8th Revised ed, 2016) and Brazilian Nutrient Requirements of Zebu and Crossbred Cattle (Valadares Filho, S. C., L. F. C. Silva, M. P. Gionbelli, P. P. Rotta, M. I. Marcondes, M. L. Chizzotti, and L. F. Prados, BR-CORTE: nutrient requirements of zebu and crossbred cattle, 3rd ed, 2016). This review aimed to compare methods used by NASEM and BR-CORTE to estimate the energy requirements for beef cattle. The net energy requirements for maintenance (NEm) of BR-CORTE is based on empty body weight (EBW), whereas NASEM uses shrunk body weight (SBW), but the Bos taurus indicus presents 10% to 8% lower NEm than Bos taurus taurus. We have compared animals with different EBW and SBW but with same equivalent empty body weight/standard reference weight ratio (0.75), as both systems have suggested different mature weights. Both systems predicted similar net energy requirements for gain (NEg) for animals with 1.8 kg of daily gain. However, estimated empty body gain was lower for NASEM estimations when the same metabolizable energy for gain is available. For pregnancy and lactation of beef cows, the NEm and net energy requirements for pregnancy (NEp) of a Zebu cow estimated by BR-CORTE were lower than the values estimated by NASEM. Furthermore, the magnitude of differences between these systems regarding NEp increased as pregnancy days increase. The NASEM and BR-CORTE systems have presented similar values for energy requirement for lactation (0.72 and 0.75 Mcal/kg milk, respectively).

Abstract:
N-acyl-phosphatidylethanolamine phospholipase D (NAPE–PLD) is considered to be the principal enzyme that produces N-acylethanolamines (NAEs), a family of signaling lipids. NAEs are involved in numerous physiological processes such as appetite, satiety, pain, inflammation, fertility, stress, and anxiety. Furthermore, aberrant NAE levels are associated with metabolic syndrome and non-alcoholic steatohepatitis (NASH). Several inhibitors for NAPE–PLD have been reported. But most of the inhibitors showed poor to moderate potency for NAPE–PLD in vitro. Recently, Mario van der Stelt et al describe the SAR of NAPE–PLD inhibitors that afforded LEI–401 in vitro. However, no attempt was instigated to produce a consensus pharmacophore model of LEI–401 as inhibitors of NAPE–PLD. Pharmacophore modeling is an efficient and useful approach to identify important patterns in a series of molecules for optimizations. The consensus pharmacophore model revealed the importance of structural features and their correlation with the biological activity.

Abstract:
Objective To determine if models of human 'receptive' and 'non-receptive endometrium' differ in their responses to nitric oxide (NO) supplementation by measuring the levels of the enzymes of the endocannabinoid system (ECS) (fatty acid amide hydrolase (FAAH) and N-acylphosphatidylethanolamine-specific phospholipase D (NAPE-PLD)), which control the 'anandamide tone' essential for successful pregnancy. Design A study of FAAH and NAPE-PLD expression (using human endometrium) through the menstrual cycle and an in vitro using a model of 'receptive' (Ishikawa) and 'non-receptive' (HEC-1A) human endometrial cell lines treated with the NO-donating compound S-nitroso-N-acetylpenicillamine (SNAP). Results Immunoreactivity measured by optimised H-score for both FAAH and NAPE-PLD was reduced in secretory (receptive) endometrium compared to proliferative (non-receptive) endometrium (P = 0.0009 and <0.0001, respectively). FAAH and NAPE transcript levels were significantly higher in untreated Ishikawa cells than in HEC-1A cells (P = 0.0228 and 0.0001, respectively). Treatment of cultures with SNAP resulted in an increase in the amount of FAAH mRNA produced by Ishikawa cells and a decrease in NAPE-PLD mRNA. No effect of SNAP was observed in HEC-1A cells. Similarly, FAAH protein was significantly decreased in endometria representative of the receptive endometrium. Conclusion These data suggest that NO most likely affects the expression of ECS enzymes in the implantation site of a receptive endometrium; a phenomenon not seen in a non-receptive endometrium. These effects are most marked with FAAH expression, suggesting that FAAH may play the more critical role in ensuring the correct 'anandamide tone' for successful embryo implantation than NAPE-PLD. Lay summary Embryo implantation into the wall of the uterus is only successful when the inner wall of the uterus (the endometrium) is ‘receptive’, because if it is ‘non-receptive’, implantation will fail. Previous work showed that enzymes of the 'endocannabinoid system' are critical for implantation by maintaining the correct level of a fat called anandamide. This is by balancing its synthesis (by N-acylphosphatidylethanolamine specific phospholipase D, NAPE-PLD) and degradation (by fatty acid amide hydrolase, FAAH). Using immortalised cell lines as models of ‘receptive’ and ‘non-receptive’ human endometrium, we demonstrate a key stimulator of implantation, nitric oxide, has a positive effect on implantation by both increasing the mRNA levels of the degrading enzyme (FAAH) and decreasing the expression of the synthesising enzyme (NAPE-PLD). These effects are most marked with the degrading enzyme, suggesting that FAAH plays a more critical role than NAPE-PLD in ensuring the correct 'anandamide tone' for successful embryo implantation.

Abstract:

Abstract:
Nepal, a landlocked country between China and India, is developing disaster and emergency medicine. In 2007, the Nepal Disaster and Emergency Medicine (NADEM) Center was formed with the aim of developing this specialty in Nepal. The first hospital was built in July 1889. It wasn't until 1988 that a Disaster Response Team was organized following a stampede incident in the national stadium in Kathmandu. The country often experiences disaster and emergency situations due to geographic and natural hazards and political tensions. In 1984, the Institute of Medicine, Tribhuvan University Teaching Hospital created emergency services with general practitioners (GPs) directing and providing services. Since then, almost all emergency services of different hospitals are run by GPs with house officers, nurses, and paramedics. There still is a lack of training and proper management, and limited equipment and infrastructure to provide needed disaster and emergency services to the people. The NADEM Center is developing coordination objectives between different emergency service providers to organize ways of service providing. This will be done through NADEM's continuing medical education and publication of Journal of Nepal Disaster and Emergency Medicine (J-NADEM) and NewsHealth; coordination among emergency medical services (prehospital), in-hospital services, and disaster and critical care medicine; and planning and implementation of different research, training, workshops, seminars, and conferences in disaster and emergency medicine with cooperation from the world. The NADEM Center will develop International Institute of Disaster and Emergency Medicine.

Abstract:
As more than a billion people had done previously, on November 21, 2013, Ukrainian journalist and activist Mustafa Nayem wrote a Facebook post; this post, however, would have a much larger impact on subsequent political developments than most that had preceded it. Frustrated with President Viktor Yanukovych’s decision not to sign a long-promised association agreement with the European Union, Nayem asked others who shared his frustration to comment on his post. Even more importantly, Nayem wrote that if the post received at least 1000 comments from people willing to join him, they should all go to Independence Square to protest. And indeed they did: starting with just a few thousand people, the protests would swell to be the largest since Ukraine’s independence, particularly after police used force against protesters at the end of November 2013. Eventually, these protests led to the resignation of the government, the exile of the former president, and indirectly to the secession of Crimea and the ongoing conflict in the eastern part of the country.

Abstract:
Arachidonylethanolamide (anandamide) acts as an endogenous ligand of cannabinoid receptors, while other N-acylethanolamines (NAEs), such as palmitylethanolamide and oleylethanolamide, show analgesic, anti-inflammatory, and appetite-suppressing effects through other receptors. In mammalian tissues, NAEs, including anandamide, are produced from glycerophospholipid via N-acyl-phosphatidylethanolamine (NAPE). The ɛ isoform of cytosolic phospholipase A2 (cPLA2) functions as an N-acyltransferase to form NAPE. Since the cPLA2 family consists of six isoforms (α, β, γ, δ, ɛ, and ζ), the present study investigated a possible involvement of isoforms other than ɛ in the NAE biosynthesis. Firstly, when the cells overexpressing one of the cPLA2 isoforms were labeled with [14C]ethanolamine, the increase in the production of [14C]NAPE was observed only with the ɛ-expressing cells. Secondly, when the cells co-expressing ɛ and one of the other isoforms were analyzed, the increase in [14C]N-acyl-lysophosphatidylethanolamine (lysoNAPE) and [14C]NAE was seen with the combination of ɛ and γ isoforms. Furthermore, the purified cPLA2γ hydrolyzed not only NAPE to lysoNAPE, but also lysoNAPE to glycerophospho-N-acylethanolamine (GP-NAE). Thus, the produced GP-NAE was further hydrolyzed to NAE by glycerophosphodiesterase 1. These results suggested that cPLA2γ is involved in the biosynthesis of NAE by its phospholipase A1/A2 and lysophospholipase activities.

Abstract:
In the era of the digital revolution and the global market economy, the economic interests of holders of industrial property rights face threats from new and advanced forms of infringement. In order to guarantee the safeguarding of the material status of the rightsholders, standards of protection have been set at the international level as well as within the European Union - with harmonization expected to take place on a global scale. These standards, related to compensation for material damages due to the infringement on industrial property rights, largely deviate from the general rules on compensation for damages. The reason for the deviations lie in the specifics of the authorizations that are inherent to these rights and the intangible nature of the intellectual property that is the subject of the protection: hence the need for specific methods devised to calculate the compensation of damages caused by the infringement on industrial property rights. This paper analyses special rules on compensation for material damages caused by infringement on industrial property rights, observed in domestic law as it has developed from 1995 and onwards. The subject of the analysis are the relics of previous positive law, i.e., triple compensation, and compensation for up to three times the amount of the license fee, as well as the current regulations on the damages set as a lump sum compensation, under which it may not be less than the compensation for the legal use of the subject of protection and compensation equal to the tortfeasor's gains. The aim of this paper is to propose de lege ferenda norms for enhanced regulation of the matter of compensation for material damages in the field of industrial property.

Abstract:
Purpose: To compare cannabis enthusiasts’ knowledge about cannabis risks and effectiveness in treating medical conditions with existing empirical evidence. Design: A brief survey assessed cannabis use, information sources, and knowledge about risks and effectiveness. Setting: A cannabis advocacy event in April 2019 in a state with legal medical and recreational cannabis. Participants: Demographically diverse adults (N = 472) who frequently used cannabis; 85% used cannabis for health or medical purposes. Measures: Participants reported the sources of their cannabis information, health conditions they thought cannabis was effective in treating (n = 10), and health risks increased by cannabis (n = 6). Conditions and risks were based on ratings of evidence (ie, from substantial to insufficient) for therapeutic effects and risks identified in a review by The National Academies of Sciences, Engineering, and Medicine (NASEM, 2017). Analyses: Chi-square tests examined the correspondence between participants’ knowledge and NASEM conclusions. Results: Most participants’ (95% confidence interval [CI]: 74%-81%) knowledge of cannabis was from their own experiences; 18% (95% CI: 14%-21%) received information from primary care providers. On average, participants’ beliefs matched NASEM conclusions for half of effectiveness (95% CI: 50%-53%) and risk items (95% CI: 55%-57%). Many (95% CI: 38%-42%) thought that cannabis use did not increase any risk. Contrary to NASEM conclusions, many thought cannabis was effective in treating cancer (76%), depressive symptoms (72%), and epilepsy (68%). Those who received cannabis information from their primary care providers had better knowledge of medical effectiveness. Medicinal cannabis use frequency inversely predicted knowledge of medical effectiveness and increased risks of adverse events. Conclusion: There were considerable discrepancies between cannabis users’ knowledge and available evidence, highlighting the need for more research and education (by physicians, caregivers, and dispensaries) on effectiveness and health risks, especially for users with specific health issues such as pregnant women and people with depression.

Abstract:
Abstract In 1996, the NASEM beef cattle committee developed and published an equation to estimate cow feed intake using results from studies conducted or published between 1979 and 1993 (Nutrient Requirements of Beef Cattle). The same equation was recommended for use in the most recent version of this publication (2016). The equation is sensitive to cow weight, diet digestibility and milk yield. Our objective was to validate the accuracy of this equation using more recent published and unpublished data. Criteria for inclusion in the validation data set included projects conducted or published within the last ten years, direct measurement of forage intake, adequate protein supply, and pen feeding (no tie stall or metabolism crate data). The validation data set included 29 treatment means for gestating cows and 26 treatment means for lactating cows. Means for the gestating cow data set was 11.4 ± 1.9 kg DMI, 599 ± 77 kg BW, 1.24 ± 0.14 Mcal/kg NEm per kg of feed and lactating cow data set was 14.5 ± 2.0 kg DMI, 532 ± 116.3 kg BW, and 1.26 ± 0.24 Mcal NEm per kg feed, respectively. Non intercept models were used to determine equation accuracy in predicting validation data set DMI. The slope for linear bias in the NASEM gestation equation did not differ from 1 (P = 0.07) with a 3.5% positive bias. However, when the NASEM equation was used to predict DMI in lactating cows, the slope for linear bias significantly differed from 1 (P < 0.001) with a downward bias of 13.7%. Therefore, a new multiple regression equation was developed from the validation data set: DMI= (-4.336 + (0.086427 (BW^.75) + 0.3 (Milk yield)+6.005785(NEm)), (R-squared=0.84). The NASEM equation for gestating beef cows was reasonably accurate while the lactation equation underestimated feed intake.

Abstract:
AbstractHabitat loss and degradation are critical threats for the globally threatened White-naped Crane Grus vipio. We estimated the size of the area used per day and the time budgets of parental and non-parental White-naped Crane pairs in north-eastern Mongolia during 2000 and 2001. Six parental crane pairs used an area of 11–155 ha per day. The maximum distance of a focal parental crane from the roosting-site was 3,030 m. Habitat conservation measures for breeding White-naped Cranes need to be targeted to within at least 3 km of the roosting-site or nest-site. Parental cranes spent 79.6 ± 4.8% of the daylight period foraging and had reduced preening and resting behaviour to 4.4 ± 1.9%. Pairs without juveniles showed a pronounced period of resting and preening behaviour during midday, which was absent in parental cranes. This indicates that parental cranes may be time-stressed. We conclude that increases in feeding-related activities (e.g. caused by a decrease in food availability) are likely to be at the expense of parental vigilance. Conversely, increases in vigilance (due to e.g. increased disturbance) may have a negative impact on feeding-related activities. Both increases can potentially negatively affect reproductive success in this Vulnerable species.

Abstract:
In the last 15 years, the west population of white-naped crane (Antigone vipio) decreased dramatically despite the enhanced conservation actions in both breeding and wintering areas. Recent studies highlighted the importance of protecting the integrity of movement connectivity for migratory birds. Widespread and rapid landcover changes may exceed the adaptive capacity of migrants, leading to the collapse of migratory networks. In this study, using satellite tracking data, we modeled and characterized the migration routes of the white-naped crane at three spatial levels (core area, migratory corridor, and migratory path) based on the utilization distribution for two eras (1990s and 2010s) spanning 20 years. Our analysis demonstrated that the white-naped crane shifted its migratory route, which is supported by other lines of evidences. The widespread loss of wetlands, especially within the stopover sites, might have caused this behavioral adaptation. Moreover, our analysis indicated that the long-term sustainability of the new route is untested and likely to be questionable. Therefore, directing conservation effects to the new route might be insufficient for the long-term wellbeing of this threatened crane and large-scale wetland restorations in Bohai Bay, a critical stopover site in the East Asian-Australasian flyway, are of the utmost importance to the conservation of this species.

Abstract:
Defective melanocortin signaling causes hyperphagic obesity in humans and the melanocortin-4 receptor knockout mouse (MC4R−/−). The human disease most commonly presents, however, as haploinsufficiency of the MC4R. This study validates the MC4R+/− mouse as a model of the human disease in that, like the MC4R−/−, the MC4R+/− mouse also exhibits a sustained hyperphagic response to dietary fat. Furthermore, both saturated and monounsaturated fats elicit this response. N-acylphosphatidylethanolamine (NAPE) is a signaling lipid induced after several hours of high-fat feeding, that, if dysregulated, might explain the feeding behavior in melanocortin obesity syndrome. Remarkably, however, MC4R−/− mice produce elevated levels of NAPE and are fully responsive to the anorexigenic activity of NAPE and oleoylethanolamide. Interestingly, additional differences in N-acylethanolamine (NAE) biochemistry were seen in MC4R−/− animals, including reduced plasma NAE levels and elevated hypothalamic levels of fatty acid amide hydrolase expression. Thus, while reduced expression of NAPE or NAE does not explain the high-fat hyperphagia in the melanocortin obesity syndrome, alterations in this family of signaling lipids are evident. Analysis of the microstructure of feeding behavior in response to dietary fat in the MC4R−/− and MC4R+/− mice indicates that the high-fat hyperphagia involves defective satiation and an increased rate of food intake, suggesting defective satiety signaling and enhanced reward value of dietary fat.

Abstract:

Abstract:
Abstract N-acylphosphatidylethanolamine phospholipase D (NAPE-PLD) catalyzes the cleavage of membrane NAPEs into bioactive fatty-acid ethanolamides (FAEs). Along with this precursor role, NAPEs might also serve autonomous signaling functions. Here, we report that injections of 6-hydroxydopamine (6-OHDA) into the mouse striatum cause a local increase in NAPE and FAE levels, which precedes neuronal cell death. NAPE, but not FAE, accumulation is enhanced in mice lacking NAPE-PLD, which display a substantial reduction in 6-OHDA-induced neurotoxicity, as shown by increased survival of substantia nigra dopamine neurons, integrity of striatal dopaminergic fibers, and striatal dopamine metabolite content. Reduced damage is accompanied by attenuation of the motor response evoked by apomorphine. Furthermore, NAPE-PLD silencing protects cathecolamine-producing SH-SY5Y cells from 6-OHDA-induced reactive oxygen species formation, caspase-3 activation and death. Mechanistic studies in mice suggest the existence of multiple molecular contributors to the neuroprotective effects of NAPE-PLD deletion, including suppression of Rac1 activity and attenuated transcription of several genes (Cadps, Casp9, Egln1, Kcnj6, Spen, and Uchl1) implicated in dopamine neuron survival and/or Parkinson’s disease. The findings point to a previously unrecognized role for NAPE-PLD in the regulation of dopamine neuron function, which may be linked to the control of NAPE homeostasis in membranes.
