Abstract:
This thesis deals with automated tuning of drive controller. To achieve this goal, system identification is needed. Therefore, the issue of identification is described at the beginning of this thesis. Spectral analysis was selected from many described methods. It was implemented in Matlab and also in C# language where was used averaging and Blackman-Tukey method. The C# application is linked to Beckhoff TwinCAT 3 and TwinCAT 2 runtime sytems that enable connections with real drive. Next, the problem of drive regulation is discussed and the results of using spectral analysis on real drives are shown. At the end of the thesis is described the algorithm of setting the speed controller with different types of frequency converters.

Abstract:
The aim of this work is to investigate the energy consumption of the servo drive at different control settings. First, the theoretical part describes the servo drive as a mechatronic system with subsequent disassembly in terms of losses and efficiency, where all the losses that occur in the servo drives are defined. Subsequently, the theoretical part concludes with a chapter on controllers, thanks to which it is possible to control servo drives and describes their influence on the resulting control. In the practical part, a model of a DC motor with a linear axis and regulation was first created in the Simulink program. Subsequently, the Matlab program defined motion with linear acceleration and motion using S-curves, which controlled the model. Then the model was measured at different settings of the controllers and the resulting waveforms together with the energy values were compared with each other. In the second half of the practical part of this work, a real servo drive was then measured with a subsequent comparison with the created model. In this comparison, the resulting waveforms and energy values were also compared. The result of this work is then to determine the effect of changes on the resulting control, which are made in the controller settings. The functional models can then be used to determine how the various servo drives will behave under the given control conditions.

Abstract:
The thesis is focused on aircraft flight level control in case when the location of center of gravity changes during flight. The theoretical part of the thesis describes basics of aerodynamics, which are necessary to understand airplane's behavior and its control theory. Basics of state theory and stability of systems, as it is used for design of linear model. This part also mentions mathematical equations of nonlinear model of business jet and discusses conventional and adaptive methods used to design autopilot parameters. Several autopilots were designed in practical part of the thesis. The most important part is the logic used to determine location of center of gravity of the airplane. The models use nonlinear model of an airplane that better represents the real environment and conditions.

Abstract:
This thesis deals with the design of electrical control for a constant speed propeller. The objective of this paper is to demonstrate the propeller governor design process on a case example of a DMP-3 propeller paired with a Rotax 912 ULS, using knowledge of flight mechanics, electrical engineering and automation. The scope of this thesis addresses the problems of propeller pitch control using an electrically driven linear actuator. In conclusion, this paper compares the benefits of the DMP-3 constant speed propeller, considering its use in existing airplanes within the UL-2 and CS-LSA category. Furthermore, it evaluates the capabilities of the proposed electronic governor, and outlines possibility of further development in this field.

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmacology & Toxicology Student: Eva Troppová Supervisor: PharmDr. Marie Vopršalová, CSc. Specialized supervisor: Prof. Dr. Antje Gohla Title of diploma thesis: Establishment of screening methods to find new regulators of the activity of phosphoglycolate phosphatase This work deals with the siRNA-based genomic screening for the modification of phosphoglycolate phosphatase (PGP) activity. 235 proteins were affected by transient transfection of siRNAs in vitro. Each siRNA was used in duplicates and the control was carried out by two control siRNAs. After downregulation of protein by siRNA PGP activity was evaluated, whether any modifications of PGP activity have occurred. PGP was the main research target. The main goal of this study before the screening was to set up a method, to create a reliable protocol. The whole study was 96 plate well. It was necessary to find the right conditions to measure PGP activity in two cell types (HEK AD 293 and Hep G2). Subsequently, optimal conditions were set up to influence expression of the protein. The method was optimalized using PGP siRNAs and 2 types of transfection reagents were tested. During our study the following methods were used: PGP activity assay, Bicinchoninic acid...
