Abstract:
El presente trabajo tiene como finalidad desarrollar la viabilidad en la creación de un producto natural hecho con insumos orgánicos oriundos del Perú, aprovechando la tendencia de alimentarse saludablemente, con la intención de llevar un estilo de vida saludable, ya sea por mantener una buena salud, hábito de consumo o simplemente por el hecho de querer verse bien. Natural Cake es un muffin hecho a base de ingredientes naturales nativos del Perú como la harina de tarwi que contiene un valor nutricional excepcional, pues presenta un alto porcentaje de proteínas, vitaminas y minerales; también está hecho con harina de quinua, rica en fibra y proteínas; cacao, rico en antioxidantes y vitaminas; todo esto endulzado con miel de caña, gran proporcionador de energía. Estos insumos, de calidad, son comercializados por comunidades campesinas del Perú quienes serán nuestros principales proveedores. El propósito es desarrollar y contribuir con el progreso de estas comunidades y servir como ejemplo de responsabilidad social. Nuestro producto es una alternativa ideal para aquellas personas que no solo quieran una cultura light, también buscan consumir productos naturales y beneficiosos para su salud. Nuestro público objetivo son personas de edades entre 18 y 45 años del NSE A y B que gracias a nuestra investigación de mercado pudimos observar sus características y necesidades. En conclusión, Natural Cake busca, en nuestros consumidores, no solo ofrecer un producto delicioso, sino contribuir a conservar la buena salud y mejorar la calidad de vida de las personas.<br>This study aims to develop the feasibility in the creation of a natural product made from organic inputs of Peru, taking advantage of the tendency to eat healthy, with the intention of putting a style of healthy lifestyle, either by maintaining a good health, habit of consumption or simply by the fact of wanting to look good. Natural Cake is a muffin made of native natural ingredients of Peru like flour tarwi containing exceptional nutritional value, because it has a high percentage of protein, vitamins and minerals; It is also made with quinoa, riched in fiber and protein meal; cocoa, rich in antioxidants and vitamins; all of this was sweetened with honey, great energy proportioner. Of quality, these inputs are marketed by the Peru communities who will be our main suppliers. The purpose is to develop and contribute to the progress of these communities, and serve as an example of social responsibility. Our product is an alternative for those who not only want a light culture, also seek to consume natural and beneficial for your health products. Our target audience are people aged between 18 and 45 of the NSE A and B which, thanks to our market research we could observe their characteristics and needs. Abstract:
En la actualidad, desde el mes de junio del presente año todos los alimentos industrializados que se ofrecen en el mercado están sujetos a llevar octógonos de advertencia por el contenido alto en azúcar, sodio, grasas transgénicas y grasas saturadas. Lo que conlleva a que el consumidor sea más cuidadoso y detallista al momento de realizar su compra. Por tal motivo, el proyecto de investigación busca validar la viabilidad de comercialización y tercerización de la bebida natural orgánica Kamuru, el cual es un producto beneficioso por contener nutrientes naturales obtenidos de la cascarilla de café, insumo principal en esta bebida la cual no es muy utilizada en la industria alimenticia, por el contrario, se desechan toneladas de cascarilla al año. Asimismo, el proyecto se sustenta a través de una investigación de mercado, donde se identifica que existe un target del 16% del mercado operativo el cual asciende a un total de 78,812 consumidores dispuestos a invertir en bebidas saludables con una frecuencia de una a dos veces por semana, observando este patrón se empleara la distribución semanal de bebidas a bodegas y supermercados. Finalmente, se observa la rentabilidad y viabilidad del presente proyecto, el cual contara con cinco años de validación financiera. Este sustento se apreciará en el valor actual neto, el cual tiene una rentabilidad de S/. 1,019,793.59 soles para los accionistas traídos al año actual; además, de una tasa interna de retorno de 285.45% mayor al 30 % del costo de oportunidad del accionista.<br>At present, since the month of June of this year all industrialized foods offered in the market are subject to carry warning octagons for the high content of sugar, sodium, transgenic fats and saturated fats. For this reason, the research project seeks to validate the feasibility of commercialization and outsourcing of Kamuru organic natural beverage, which is a beneficial product for containing natural nutrients obtained from the coffee husk, the main input in this beverage which is not very used in the food industry, on the contrary, tons of husks are discarded annually. Likewise, the project is supported through a market investigation, which identifies that there is a target of 16% of the operating market which amounts to a total of 78,812 consumers willing to invest in healthy drinks with a frequency of one to two times per week, observing this pattern, the weekly distribution of beverages to wineries and supermarkets will be used. Finally, the profitability and viability of this project is observed, which will have five years of financial validation. This support will be appreciated in the net present value, which has a profitability of S /. 1,019,793.59 soles for shareholders brought to the current year; In addition, an internal rate of return of 285.45% greater than 30% of the opportunity cost of the shareholder.<br>Trabajo de investigación

Kinesiology<br>Ph.D.<br>The prevalence of obesity among young children has markedly increased over the past two decades, with more than one-third of American preschoolers now overweight or obese and at risk for lifelong health problems. Physical activity is a recommended obesity prevention strategy, yet preschoolers typically fail to meet recommended daily physical activity guidelines, spending just 15 minutes engaged in moderate to vigorous physical activity, compared to six sedentary hours daily. Unstructured play in settings with varied features, such as childcare center playgrounds, potentially plays a significant role in increasing the amount of time preschoolers spend in moderate-to-vigorous physical activity (MVPA). The purposes of this study were first, to compare the intensity and type of preschoolers' physical activity across four distinctly different outdoor play settings; second, to identify particular features in each play setting associated with MVPA and sedentary behavior; and, third, to identify, test, and evaluate environmental modifications to increase preschoolers' MVPA in outdoor play settings. Seventeen 3-5 year-old children participated in repeated unstructured play sessions featuring 16 min of play in each of 4 novel settings: a traditional climber, a wooded natural area, a garden, and an adventure, or "loose parts" playground. Interventions to increase physical activity were introduced to the adventure playground during the first two phases, and to all four settings during the third phase. Physical activity intensity was measured using ActiGraph GT3x+ activity monitors and contextual information concerning motor skills was obtained by trained observers using the System for Observing Play and Recreation in Communities (SOPARC), adapted to the age and environment of the study. Results show that both play setting design theme and the composition of specific play features within the setting impact the type of motor skills children perform and the amount of MVPA young children accumulate during unstructured play. Findings additionally demonstrate that simple, low-cost modifications to play settings can increase MVPA for targeted subgroups and individual children; outcomes were setting-specific. Study results may be useful to public health and medical workers, parents, educators, playground designers, community planners, and policy makers who focus on increasing preschool children's daily MVPA and decreasing childhood obesity.<br>Temple University--Theses

Abstract:
Healthy ageing in the absence of neurodegenerative diseases is accompanied by a variety of perceptual and cognitive changes, which can occur on various distinct but interacting levels. Much research has been devoted to understanding how basic cognitive functions such as working memory and attention change with age. However, only more recently has age-related changes on perceptual functions been investigated, thus less is understood. My thesis aimed to address this gap in the literature, by exploring age-related changes in visual perception, specifically changes related to visual temporal processing of natural and biological stimuli. Also investigated was the extent to which age-related changes in perceptual and cognitive functions affect each other. Chapter 2 investigated the time course of visual processing of natural scene categorisation in older adults. Younger, young-old and old-old adults performed a go/no-go task, in which they had to respond to images of animals whilst ignoring images of landscapes. The results showed that the temporal processing of complex scenes is impaired in healthy older adults and this deficit becomes more pronounced with increasing age. Chapter 3 assessed whether age-related decline in biological motion perception is mediated by impaired attentional abilities. Younger and older adults performed a series of tasks assessing biological motion perception and visual attention. The results indicated that age-related changes in biological motion perception are not driven by general attentional decline. Lastly, Chapter 4 explored whether age-related changes in biological motion and attention tasks can be explained by differences in the allocation of attention. Younger and older adults performed a conjunctive visual search, and two biological motion tasks, while their eye movements were being tracked. The results illustrated that differences in the allocation of attention cannot explain age-related differences found on biological motion perception and attention tasks. Overall, my findings provide substantial evidence to suggest that both visual perceptual and cognitive abilities change with healthy ageing. However, my results also indicate that certain aspects of these two functions remain relatively preserved in older adulthood.

Este trabajo de investigación fue realizado en Lima Metropolitana a los NSE “A” Y “B”; teniendo un principal enfoque en quienes tienen perfiles digitales y buscan dinamismo en sus compras. Este modelo de negocio es atractivo, ya que vemos un consumidor interesado en mejorar su alimentación y tener un estilo de vida saludable, pero que no encuentra un lugar que le brinde una oferta unificada de todo lo que busca. Aprovechando que en el mercado existen pocas opciones que vuelvan dinámica la interacción entre ofertantes y demandantes, creamos Natural Club. Es un emprendimiento que permite a los vendedores ofertar sus productos, selectos y de alta calidad, de una manera rápida y sencilla, así como para el consumidor adquirirlos de la misma manera. Encargándose, además, de todos los envíos a domicilio y asegurándose de que cumplan con los altos estándares de salubridad que requiere la coyuntura COVID-19. Para el desarrollo de este proyecto se efectuó un análisis exhaustivo que cubrió lo siguiente: Validación del Modelo de Negocio, Análisis Interno y Externo y Desarrollo de Plan Estratégico. Todo lo realizado se hizo con el objetivo de validar la viabilidad y sostenibilidad del presente proyecto.<br>The purpose of this entrepreneurial project is to implement a business focused on unifying the supply and demand of select healthy products through an online platform (marketplace), which seeks to promote and motivate our consumers to adopt a healthier lifestyle. This project was carried out in Metropolitan Lima at SES (Socioeconomic status) "A" and “B”. We adopted a main focus on consumers with digital profiles that seek for dynamism in their purchases. This business model is attractive, since we see there are consumers willing to improve their diet and adopt a healthier lifestyle, but they do not find a place that provides a unified offer of everything they are looking for. Taking advantage that there are few options in the market where the interaction between suppliers and demanders is dynamic, we created Natural Club. Abstract:
Hoy en día, existe una tendencia mundial a consumir alimentos saludables, que no tengan insumos artificiales -como preservantes o aditivos- y que sean bajos en azúcar. El Perú no es ajeno a esta tendencia, pues se aprecia que existen cada vez más consumidores que buscan lo más natural posible. En este contexto, los superfoods son una alternativa importante, pues se trata de alimentos naturales que cuentan con altos niveles de nutrientes. El presente proyecto toma en cuenta esta tendencia para proponer una bebida saludable elaborada con superfoods peruanos, bajo la marca Superfoods Drinks. La investigación realizada muestra que en el segmento de personas de 18 a 55 años del NSE A, B y C1 es de 54.9% de los cuales el 11% está interesado en consumir saludable y que su demanda por bebidas saludables y naturales no está siendo satisfecha por la oferta actual, lo que explica el alto grado de aceptación de nuestro producto. Para la puesta en marcha del proyecto se necesita una inversión inicial de S/ 191,693, financiada con S/ 111,693 mediante un préstamo bancario y S/ 80,000 con aporte de inversionistas, que se estima recuperar en el cuarto año de la operación.<br>Nowadays, there is a global trend towards consuming healthy foods, that don’t contain artificial ingredients -such as preservatives or additives- and don’t have added sugar. Peru is part of this trend, as there is a growing number of consumers that opt for more natural products. In this context, superfoods are an important alternative because they are a natural foodstuff that has a high concentration of nutrients. The present project takes this trend into account and proposes a healthy drink prepared with Peruvian superfoods, under the brand Superfoods Drinks. Research shows that in the segment of people aged 18 to 55 of social economic level A, B, C1 there is 54.9% and about 11% of this people is interested in eating in a healthy way, and whose demand for healthy and natural drinks is not being satisfied by the current offer, which explains the high level of acceptance of our product. To start up the project we need an initial investment of S/ 191,693 that will be funded with bank loans -S/ 111,693- and by investors -S/ 80,000-, and that is expected to be paid back by the fourth year of operations.<br>Trabajo de investigación

Abstract:
Este proyecto se centra en el impacto que genera la elaboración de una bebida que contribuya a la disminución de problemas estomacales como la indigestión o estreñimiento. Algunas de las causas de estos problemas de salud, se relacionan con los factores dietéticos, especialmente la falta de fibra en la alimentación. Otras veces este problema se relaciona enfermedades que padece la persona, especialmente en personas diabéticas y tiroideas. Se analizaron distintos procedimientos para un mejor aprovechamiento de los hongos del kéfir, el cual está compuesto principalmente por bacterias que fermentan el agua. Estos procesos de fermentación de la bebida de desarrollaron cumpliendo las normas de salubridad establecidas y de buenas prácticas de higiene y seguridad ambiental Se realizó un estudio del mercado limeño para conocer los posibles factores claves de la venta, evaluando los puntos socioeconómicos, conductual de los personas en el sector sur de Lima, como los distritos de VES, SJM, Barranco, Chorrillos, y Surco. Por último, se pusieron a la venta las bebidas embotelladas obtenidas del kéfir en dos tipos de sabores; manzana y lima, brindando nuevas alternativas dentro de los productos farmacéuticos contra el estreñimiento, al aprovechar los beneficios nutricionales, bromatológicos que aporta el kéfir. Este proyecto nos permitió concluir con la degustación y ventas de las bebidas fermentadas, ser aceptadas por las personas consumidoras, lo cual demuestra el valor nutricional que tiene el kéfir para su producción, distribución y comercialización para la salud de la flora intestinal.<br>This project focuses on the impact generated by the development of a beverage that contributes to the reduction of stomach problems such as indigestion, constipation. Some of the causes of these health problems are related to dietary factors, especially the lack of fiber in the diet. Other times this problem is related to diseases that the person suffers, especially in diabetic and thyroid issues. Different procedures were analyzed for a better use of kefir fungi, which is mainly composed of bacteria that ferment water. These beverage fermentation processes were developed in compliance with established health standards and good hygiene and environmental safety practices. A study of the Lima market was carried out to know the possible key factors of the sale, evaluating the socio-economic behavioral points of people in the southern sector of Lima, such as the districts of VES, SJM, Barranco, Chorrillos, and Surco. Finally, bottled beverages obtained from kefir were put up for sale in two types of flavors; apple and lime, providing new alternatives in pharmaceutical products against constipation, taking advantage of the nutritional, bromatological benefits that kefir provides. This project allowed us to conclude with the tasting and sales of fermented beverages, to be accepted by consumers, which demonstrates the nutritional value that kefir has for its production, distribution and marketing for the health of intestinal flora.<br>Trabajo de investigación

Salsa Sumaq nace a partir del análisis de las diferentes necesidades dentro del mercado peruano, principalmente en las problemáticas presentadas en cuanto a temas relacionados a los hábitos alimenticios de las personas. El consumidor busca opciones saludables como acompañamiento para sus comidas para de esta manera darles un sabor agradable sin dejar de lado sus principales objetivos que se basan en mantener el equilibrio calórico en estas mismas. Al analizar el mercado, se pudo apreciar que el crecimiento en cuanto a la industria de productos saludables se encontraba en un crecimiento constante generando oportunidades para el desarrollo de nuevos productos, a su vez, la tendencia por lo orgánico y natural se hacía cada vez más popular. Luego de evaluar las preferencias de nuestros potenciales consumidores, se desarrolló la fórmula de las diferentes salsas , en donde se buscaba no solo brindar un agradable sabor de bajas calorías sino también que nuestros clientes puedan sentir el sabor casero tradicional de la comida peruana, por lo que se hicieron uso de insumos nativos de las diferentes regiones peruanas como también se buscaron insumos especializados que aporten una cantidad calórica menor en comparación a insumos tradicionales para la elaboración. En cuanto a la presentación, el diseño del envase resulta un factor bastante importante en cuanto a productos de calidad, por lo que se hizo uso de frascos de vidrio con tapa rosca de 250 gramos, los cuales podían ser reutilizados siguiendo a su vez con las tendencias eco amigables. Por otro lado, el etiquetado y rotulado son presentados en los colores característicos de las regiones y se desarrollaron los octógonos verdes donde se dan a conocer las diferentes bondades del producto en cuanto a su elaboración. Salsa Sumaq es un producto que pretende conectar con sus consumidores mediante la preocupación y la satisfacción de sus necesidades en cuanto a temas que implican la salud, ya que nuestro principal objetivo es brindar productos de calidad que puedan beneficiar a nuestros consumidores. A pesar del contexto, Salsa Sumaq está logrando un crecimiento progresivo y constante que de a pocos se encuentra generando beneficios para los inversionistas, ya que con el pasar de las semanas está tomando mayor posicionamiento en el mercado y pretender ganarse las preferencias de los consumidores.<br>Salsa Sumaq was set up from the analysis of the different needs within the Peruvian market, mainly in the problems presented with regard to issues related to people's eating habits. The consumer looks for healthy options as an accompaniment to their meals in order to give them a pleasant flavor without neglecting their main objectives that are based on maintaining the caloric balance in them. When analyzing the market, it could be seen that the growth in terms of the healthy products industry was in constant boost, generating opportunities for the development of new products, in turn, the trend for organic and natural was becoming more and more popular. After evaluating the preferences of our potential consumers, the formula of the different sauces was developed, where it was sought not only to provide a pleasant low-calorie flavor but also so that our clients could feel the traditional homemade flavor of Peruvian food, that is the reason why native ingredients from the different Peruvian regions were used as well as specialized ingredients that provide a lower caloric amount compared to traditional ones for the production. Regarding the presentation, the design of the package is a fairly important factor in the matter of quality products; consequently, 250-gram glass jars with screw caps were made, which could be reused following the ecofriendly trends. On the other hand, labeling is presented in the characteristic colors of the regions and green octagons were developed where the different benefits of the product in terms of its production are made known. Salsa Sumaq is a product that aims to connect with its consumers by caring for and satisfying their needs in respect of issues involving health as our main objective is to provide quality products that can benefit our consumers. Despite the context, Salsa Sumaq is achieving progressive and constant growth that, little by little, is generating benefits for investors, considering that as the weeks go by, it is taking a greater position in the market and trying to win the preference of consumers.<br>Trabajo de investigación

A method of natural transmission that ‘traverses’ all identified common cold viruses was evaluated in Leicester, UK during 2012 to 2015. The viruses were: adenovirus, coronavirus, enterovirus, influenza, metapneumovirus, parainfluenza, rhinovirus, and RSV. Mechanisms of transmission assessed included large droplet, hand contact, fomite, and aerosol. Aim: to augment current understanding of natural respiratory virus transmission using the TraVerse method. Methods: TraVerse method of 30-minute interactions between symptomatic paediatric inpatients and healthy young adults in a fully functioning hospital ward. Swab and aerosol samples were tested using qualitative and quantitative PCR. Illness severities were evaluated using daily symptom diary cards. Results: The overall RT-PCR positivity rates were 93% for children and 22% for adults. Rates of natural transmission, depending on the virus, were 4% - 24% by large droplet, 18% - 19% by hand contact, 50% - 100% by self-inoculation, and 8% - 33% by fomites. Aerosol transmission was not established. Adults did not shed flu, coronavirus, metapneumovirus, or parainfluenza; nevertheless, severe illness occurred in 26% whose child pairs had mono-infections of these viruses. When ill, only 7% of adults stayed home, and only 4% sought medical attention. Asymptomatic rhinovirus transmission occurred at a rate of 5%. Conclusions: TraVerse was fit-for-purpose in elucidating natural, human-to-human respiratory virus transmission rates. Aerosol transmission may be the primary mechanism in the natural transmission of flu, coronavirus, metapneumovirus, and parainfluenza but was not appropriately evaluated because of funding constraints. Paediatric mouthing has relevance in nosocomial transmission. Healthy adults develop severe illness but do not generally stay home, take medications, or seek medical advice. Recommendations: Sanitizing of paediatric patients’ hands, use of masks to discourage face-touching, flocked swabs to replace nasopharyngeal aspirates in respiratory sampling, use of virus molecular subtyping assays, increased efforts to include minorities in research, and better collaboration to encourage research funding.

Obesity is a contributing factor to many diseases. Unhealthy food choices are a behavior that can lead to obesity. Teaching children to make healthy food choices at an early age could lead to healthy food choices throughout a lifetime, which would mitigate potential for obesity. A number of different treatments have evaluated healthy food choices in children; however, many did not evaluate or show maintenance effects. The purpose of this study was to utilize correspondence training to acquire healthy food choices in preschoolers and to evaluate of natural communities of reinforcement as a maintenance procedure. Results showed that correspondence training is likely to increase healthy food choices in most preschoolers; however, maintenance of healthy snack choices is variable.

This is followed by business plan structure which serves as a base for the practical part. Last topic of the theoretical part summarizes current trends in the food industry and especially market with functional food, dietary supplements, powdered food replacement, and anticipated future developments. The practical part of the thesis is constituted by the business plan for the startup selling natural powdered meal-replacement on the Czech market. Para ello se han investigado las tendencias del entorno, tales como los hábitos de consumo de la población y la digitalización de la sociedad, la cual viene en aumento en nuestro país, con una proyección de crecimiento del e-commerce cada vez mayor. La Organización Mundial de la Salud recomienda el consumo de 400 gramos de frutas al día, el equivalente a 5 frutas; sin embargo, los limeños consumen solo 100 gramos de frutas y verduras como parte de su alimentación diaria. Teniendo en cuenta la tendencia de consumo de productos saludables y naturales, la cual está en crecimiento, es importante considerar la oportunidad de mercado en relación al bajo consumo de frutas. Así mismo, los estilos de vida cada vez más acelerados demandan ahorro de tiempo, por lo que el consumidor busca productos más prácticos. Como parte de la investigación se realizó un sondeo de mercado con la intención de validar el modelo de negocio. Para llevar este proyecto a cabo se ha desarrollado las cuatro variables del marketing mix, incorporando los resultados de la investigación realizada. Finalmente, se ha desarrollado la validación económica y financiera de manera que se demuestra la rentabilidad de la propuesta del plan de negocio.<br>In this research work we will focus on testing the viability of a business based on the marketing of chopped fruit by delivery, aimed at people working in companies within the district of San Isidro in the city of Lima. To this end, environmental trends have been investigated, such as the consumption habits of the population and the digitalization of society, which is increasing in our country, with a projection of growing e-commerce. The World Health Organization recommends the consumption of 400 grams of fruit per day, the equivalent of 5 fruits. However, Lima residents consume only 100 grams of fruits and vegetables as part of their daily diet. Taking into account the trend of consumption of healthy and natural products, which is growing, it is important to consider the market opportunity in relation to low fruit consumption. As part of the investigation, a market survey was carried out with the intention of validating the business model. To carry out this project, the four marketing mix variables have been developed, incorporating the results of the research carried out. Organik es un proyecto de negocio de comida saludable en base a insumos orgánicos y asesoría nutricional que va dirigido a personas que trabajan en oficinas y residen en el distrito de San Isidro. La preparación de las comidas se lleva a cabo en nuestro local ubicado en el mismo distrito cuyos platos cumplen con los controles de salubridad necesarios y con productos de alta calidad. La idea de negocio va orientada a personas entre 18 y 45 años de un nivel socioeconómico A y B que tienen un nivel de vida acelerado que no les permite preparar sus comidas por falta de tiempo. Objetivo: Evaluar la calidad de la dieta por región natural del Perú en adolescentes y adultos. Materiales y métodos: El presente es un análisis secundario de la base de datos del Estudio Latinoamericano de Nutrición y Salud (ELANS). La muestra estuvo compuesta por 1113 sujetos de diferentes regiones del país (Lima, resto de costa, sierra y selva). Para medir la calidad de la dieta se calculó el HEI (Índice de Alimentación Saludable), a través de la información del consumo de alimentos por recordatorio de consumo de 24 horas de dos días no consecutivos. El HEI se clasificó en no saludable (<60 puntos HEI), medianamente saludable (60-79 puntos) y saludable (> 80 puntos). Resultados: La media del puntaje HEI fue 71.0 (DE: 10,0), las mujeres tuvieron mayor puntaje que los hombres (72,2 (DE: 10,0) vs 69,5 (DE: 9,7); p<0.001, respectivamente). El consumo de lácteos obtuvo un mayor puntaje en la Sierra y menor en la Selva (6,76 (DE: 4,53) vs 2,86 (DE: 4,24); p<0.001, respectivamente). Asimismo, las carnes tuvieron mayor puntaje en la Costa y Selva y menor en la Sierra (4,76 (DE: 0,89), 4,76 (DE: 0,84) vs 4,24 (DE: 1,49); p<0.001, respectivamente). La regresión lineal ajustada mostró que la Sierra tiene mayor puntaje de HEI (coef: 1.89 EE: 0.75; p=0.012) y la selva el menor puntaje (coef: -5.31, EE: 1.01; p<0.001) en relación a Lima y Resto de costa. Conclusión: El lugar de residencia afecta el índice de calidad, siendo la Sierra la que presenta un mayor índice de alimentación saludable y la selva el menor en comparación a las otras regiones.<br>Objective: To evaluate the diet quality in Peru by region in adolescents and adults. The sample was composed of 1113 subjects from four different regions of Peru: Lima Metropolitana, Costa, Sierra and Selva. A multiple linear regression was performed to analyze the effect of region, adolescents and adults. HEI score. Results: The mean HEI score was 71,0 (SD: 10,0), women scored higher than men (72,2 (10,0) vs 69,5 (9,7); p<0.001). Dairy products scored higher in the Sierra region and lower in the Selva region (6,76 (4,53) vs 2,86 (4,24); p<0. 001). Similarly, meats scored higher on the Costa region and the Jungle region and lower in the Sierra region (4,76 (0,89) vs. 4,76 (0,84) vs. 4,24 (1,49); p<0.001). Purpose: Despite its many health benefits, moderate exercise can induce joint discomfort when done infrequently or too intensely even in individuals with healthy joints. This study was designed to evaluate whether NEM (R) (natural eggshell membrane) would reduce exercise-induced cartilage turnover or alleviate joint pain or stiffness, either directly following exercise or 12 hours post exercise, versus placebo. Patients and methods: Sixty healthy, postmenopausal women were randomly assigned to receive either oral NEM 500 mg (n=30) or placebo (n=30) once daily for two consecutive weeks while performing an exercise regimen (50-100 steps per leg) on alternating days. The primary endpoint was any statistically significant reduction in exercise-induced cartilage turnover via the biomarker C-terminal cross-linked telopeptide of type-II collagen (CTX-II) versus placebo, evaluated at 1 and 2 weeks of treatment. Secondary endpoints were any reductions in either exercise-induced joint pain or stiffness versus placebo, evaluated daily via participant questionnaire. The clinical assessment was performed on the per protocol population. Results: NEM produced a significant absolute treatment effect (TEabs) versus placebo for CTX-II after both 1 week (TEabs - 17.2%, P=0.002) and 2 weeks of exercise (TEabs - 9.9%, P=0.042). Immediate pain was not significantly different; however, rapid treatment responses were observed for immediate stiffness (Day 7) and recovery pain (Day 8) and recovery stiffness (Day 4). No serious adverse events occurred and the treatment was reported to be well tolerated by study participants. Conclusion: NEM rapidly improved recovery from exercise-induced joint pain (Day 8) and stiffness (Day 4) and reduced discomfort immediately following exercise (stiffness, Day 7). Moreover, a substantial chondroprotective effect was demonstrated via a decrease in the cartilage degradation biomarker CTX-II.

Abstract:
Situated in a time of advanced technoscience and new materialist feminist humanities/social sciences, this thesis explores how popular science renditions of biomechanics contribute to transforming imaginaries about “the natural” and “healthy”. It does so by zooming in on biomechanical scientist Katy Bowman’s pervasive and life-style commitment-requiring teaching. Her books and online material conceptualise and connect a bodily dependency on adequate physical load environments to an imagined natural health of our Palaeolithic ancestors. Drawing on several postconventional fields gathered under the banner of feminist posthumanisms and posthumanities (Braidotti 2013; Åsberg 2014), this thesis demonstrates how gendered and otherwise intersectionally interpreted fantasies intra-act with Bowman’s specific bodily practices, constructing a natural with both limiting and liberating consequences. Notions of the natural in popularised biomechanics are here explored foremost with a focus on the formative categories of gender and class. More explicitly, the thesis shows how Bowman’s teaching, on the one hand, links well with theorisings of corporeal, environmental and material feminist scholars, such as Elizabeth Grosz’s (1994) and Stacy Alaimo’s (2010) notions of environed corporeality and trans-corporeality. On the other hand, though, Bowman’s popularised biomechanics simultaneously reinforces a troublesome nature-culture divide and neo-liberal discourses on health as choice. However, while downplaying sociocultural and economical factors, and underpinning essentialist notions of motherhood, Bowman’s popular science also destabilises masculine understandings of the natural as tough; acknowledges material, individual and collective agency; and, offers effective techniques for managing various health conditions – all in ways that may well be interpreted and practiced within feminist registers. Based on this example from Bowman’s popular science, the author argues that contemporary Western understandings of the natural are influenced by a longing for self-commitment, control and connectedness.

The aging population grows every year, and certain nations have the largest percentage of elderly people. This population needs a growth in health care services since they are more likely to have chronic age-related disorders. "Healthy Aging" has been a research point to enhance the lives of the elderly. Several prior research on various nutraceuticals have been conducted to assess their potential as supplements to promote healthy aging and may even postpone the onset and progression of some age-related disorders. The antioxidant and anti-inflammatory effects of several nutraceuticals were investigated in various tissues (myoblasts, retinae, and brain). Muscles provide a variety of important functions, including glucose absorption, bone density regulation, and protein homeostasis. Exercise is critical for maintaining muscle function, avoiding hypercontraction injury and delaying the onset of age-related muscular diseases. The purpose was to evaluate the ability of a combination containing magnesium, potassium, vitamin D3, and curcumin (MKVC) to prevent or restore muscle damage on C2C12 murine myoblasts under normal conditions and in the presence of caffeine to mimic a hypercontractility condition by studying cell viability, mitochondrial activity during exercise, calcium and magnesium movements during the muscle contraction/relaxation cycle, the metabolic changes, alterations in the various intracellular pathways. All the data gained will aid in the development of a formulation that will assist athletes in maintaining peak performance with protection against muscle injury and decreasing muscle loss due to aging as well. As glaucoma is the leading cause of permanent blindness, defined by structural changes in the optical nerve, various nutraceuticals have demonstrated their ability to lower the oxidative stress associated with age- related eye illnesses. A hypothesis concerning a combination of gastrodin and vitamin D3 together with vitamin C, blackcurrant, and lycopene has been proposed to combat the early modifications of glaucoma that lead to degeneration. RGC cells and retinal tissues were employed to test our theory under normal or H2O2/NMDA-induced glaucoma circumstances by assessing cell survival, ROS generation, identifying changes in the retinal area, and examining the various intracellular pathways. All of these will contribute to the development of a formulation that will aid in the slowing of the many deteriorations associated with glaucoma. Concerning the many deteriorations affecting the brain as a result of aberrant protein depositions or a decrease in BDNF expression associated with multiple age-related neurodegenerative disorders and cognitive function loss. Because BDNF has a reduced ability to cross the blood-brain barrier, it was proposed that using a low-dose BDNF sequentially kinetic activated (SKA) formula could overcome this problem. This hypothesis was tested through BDNF quantification in both in vitro and animal models, as well as studying the safety of using this formula through cell viability, ROS production, mitochondrial activity, and determining the different intracellular pathways. Based on these findings, a combination of curcumin, vitamin D, and 6-shogaol (CVS) might delay and repair the damage caused by either oxidative stress or iron damage as two distinct characteristics of brain aging, and neurodegeneration were investigated. Another important point is to verify if CVS might promote BDNF expression in astrocytes and if it can aid with neuroprotection when administered to SH-SY5Y cells. These assumptions were tested by measuring cell survival and ROS generation, as well as identifying BDNF synthesis and distribution, and screening the several active intracellular pathways. All of this will pave the way for the development of a novel human anti-aging solution for neuroprotection.

Although consumers’ need for a healthy and sustainable lifestyle drives natural cosmetics consumption, various claims of natural cosmetics make consumers confused and distrustful. This study aimed to explore factors that affect consumers' purchasing behavior of natural cosmetics under the theoretical framework of planned behavior (TPB). The study adopted the qualitative method of purposive sampling. Semi-structured interview technique and thematic analysis were used to collect and analyze data from 21 consumers of natural or traditional cosmetics living in Uppsala. The results showed: consumers had uncertain attitudes towards buying natural cosmetics, which had no obvious impact on the purchasing behavior; the perceived social pressures from family, relatives, groups, etc. had a clear positive impact on the purchasing behavior; online purchasing, recommendation from others, and sustainable lifestyle facilitated the purchasing behavior; high price, inconvenient purchasing location, lack of knowledge, non-harmonized certifications, etc. led to the inconsistency between consumers' purchasing intention and actual purchasing behavior. Product knowledge was explored as an important resource that consumers need. The above findings can be used to improve marketing strategies like harmonized certifications. This study also paves the way for future quantitative verification of these factors.

Así mismo, en el mundo existe una alta preferencia en el consumo del café (95%), cuyos subproductos como la cascarilla del café no han sido bien aprovechados y en la actualidad se desperdicia más del 80% generando contaminación medioambiental y en otros casos se usa como abono, solo en algunos países del mundo y Latinoamérica están innovando con los subproductos y están usando la cascarilla para producir nuevos productos como infusiones, mermeladas, yogurt, etc. El Perú no está exento de ello, por eso nace Muru Café, una bebida natural a base de la cascarilla de café, cero azúcares, con altos contenidos de carbohidratos, proteínas y minerales, 500 veces más antioxidante que la misma vitamina C, con ligera dulzura y beneficiosa para la salud por su valor nutricional. En el mercado peruano existe este tipo de bebida natural, pero no ha sido industrializado para la comercialización en el mercado, por lo que representa una oportunidad para incursionar en el negocio. Además, por la investigación de mercado, se logró validar el público objetivo y los canales de distribución indirectos para vender las bebidas en minimarkets, tiendas por conveniencia, naturistas y supermercados. Así mismo, el equipo de trabajo está altamente capacitado y con experiencia en producción, ventas, administración y finanzas, lo que nos permite desarrollar un proyecto viable, rentable y sostenible en el tiempo.<br>The trend of healthy consumption is generating more people opt for more natural beverages, which is a trend in consumption worldwide. Likewise, in the world there is a high preference in the consumption of coffee (95%), whose by-products such as coffee husk have not been well used and currently more than 80% are wasted generating environmental pollution and in other cases use as fertilizer, only in some countries of the world and Latin America are innovating with by-products and are using the husk to produce new products such as infusions, jams, yogurt, etc. Peru is not exempt from this, that is why Muru Café is born, a natural drink based on coffee husks, zero sugars, with high carbohydrate, protein and mineral content, 500 times more antioxidant than vitamin C, with light sweetness and beneficial to health due to its nutritional value. In the Peruvian market there is this type of natural beverage, but it has not been industrialized for commercialization in the market, so it represents an opportunity to venture into the business. In addition, through market research, it was possible to validate the target audience and the indirect distribution channels to sell beverages in minimarkets, convenience stores, naturists and supermarkets. El presente trabajo de investigación se centra en el proyecto BioBakery, el cual consiste en la venta online de barras saludables de maní y almendras, hechos con ingredientes 100% naturales. Estas barras están dirigidas a hombres y mujeres del NSE A y B, entre las edades de 18 a 55 años, que residen en Lima Metropolitana y que tengan interés en llevar un estilo de vida saludable. Los experimentos llevados a cabo permitieron validar que el público objetivo presentaba un problema con respecto al consumo de postres tradicionales al momento de saciar un antojo, ya que las personas tomaban en cuenta que esos postres son dañinos para la salud a largo plazo, por lo que estuvieron dispuestos a probar un producto sano. Ante esto, se iniciaron las primeras ventas a través de las redes sociales de la marca, Facebook e Instagram, en donde se logró comprobar que se tiene la aceptación del público. Con el resultado obtenido, se buscó implementar el empaque biodegradable a las barras saludables. El contenido del trabajo se divide principalmente en la descripción del modelo de negocio, en la validación del mismo y en el desarrollo del plan de negocio, lo que incluye operaciones, recursos humanos, marketing, RSE y finanzas. Con todo lo mencionado anteriormente, se pudo justificar la viabilidad del proyecto. Finalmente, se pudo concluir que el negocio es rentable gracias al concierge realizado y al análisis de los estados financieros.<br>This research work focuses on the BioBakery project, which consists of the online sale of healthy peanut and almond bars, made with 100% natural ingredients. These bars are aimed at men and women of socioeconomic level A and B, between the ages of 18 to 55, who reside in Metropolitan Lima and who are interested in leading a healthy lifestyle. The experiments carried out allowed to validate that the target audience had a problem with respect to the consumption of traditional desserts when satisfying a craving, since people took into account that these desserts are harmful to long-term health, so they were willing to try a healthy product. Given this, the first sales began through the brand's social networks, Facebook and Instagram, where it was possible to verify that it has been accepted by the public. With the result obtained, it was sought to implement environmentally friendly packaging for healthy bars. The content of the work is mainly divided into the description of the business model, its validation and the development of the business plan, which includes operations, human resources, marketing, CSR and finance. With all the aforementioned, the viability of the project could be justified. Por esta razón, nuestro emprendimiento busca contribuir a que las personas tengan los nutrientes necesarios mediante el consumo de jugos naturales hechos a base de frutas que contienen diferentes propiedades que benefician la salud de las personas. Este proyecto está dirigido a hombres y mujeres de 18 a 50 años que pertenecen a niveles socioeconómicos A, B y C que estén interesados en mejorar su alimentación y llevar una vida mucho más saludable. Para el análisis de viabilidad de la propuesta se realizaron diversos experimentos para sustentar la propuesta de negocio y su rentabilidad. Asimismo, se realizaron validaciones de la venta de los jugos naturales y las interacciones con el cliente por medio de las redes sociales.Los canales digitales permitió conocer que esta idea de negocio era muy bien acogida por las personas que interactúan con las publicaciones de las páginas de Facebook e Instagram de Juicy Vit. Por otro lado, se ha podido determinar que el negocio será rentable,ya que de acuerdo a los diversos indicadores financieros se ha podido determinar la rentabilidad del proyecto. Por último, las ventas proyectadas para los próximos 5 años nos brindan resultados positivos por lo que el proyecto es rentable. El consumo per cápita de ají fresco en el Perú es de 4.75 Kg al año, según datos de la Asociación de Exportadores (ADEX) en el 2017, esto representa un incremento en comparación a años anteriores, el cual está relacionado con el boom de la gastronomía peruana que incentiva el uso de ajíes en pastas o salsas en los últimos años. Además, esta tendencia de crecimiento también se refuerza gracias a las actividades realizadas por MINAGRI, esta entidad pública promueve la producción y consumo interno del ají por medio de mecanismos que dan a conocer la diversidad, usos, beneficios del ají peruano y lo que representa en nuestra cultura gastronómica. Por otro lado, un estudio realizado por la consultora Arellano acerca de las tendencias de consumo saludable indica que los peruanos destacan la buena alimentación como parte de una vida saludable y que además, el 45% de los limeños se fija en el contenido nutricional de los productos. Otro dato asociado a esta tendencia es que el 54% de los hogares se considera saludable, de acuerdo a la consultora KWP, que además señala que los consumidores saludables son más conscientes y cuidadosos en elegir productos de consumo. Por ello, siguiendo las tendencias del mercado surge Salsa Jajay, una salsa picante compuesta por ají nativo peruano con pulpa de fruta, un producto natural. Actualmente, la alimentación no balanceada es uno de los mayores problemas de nuestro país. La mayoría de las personas no cuenta con educación alimenticia para el cuidado de su dieta diaria. Por ello, el consumo desordenado de alimentos con altos índices de azúcar, sodio, preservantes, entre otros influye constantemente en el desarrollo de enfermedades como diabetes, anemia y demás. Asimismo, se puede evidenciar que el mercado no cuenta con alternativas saludables, que satisfagan a clientes que demandan productos golosinaros que no perjudiquen su salud (snacks). Por ello, se ha realizado un plan de negocios que evalúa, desarrolla y genera gomitas saludables a base de insumo naturales, denominado GOMIFRUT. Cabe resaltar que cuenta con 2 presentaciones: Frutos Tropicales y Frutos silvestres, los cuales son elaborados salubremente. Asimismo, el objetivo del proyecto es que las personas puedan integrar el producto a su dieta diaria teniendo la seguridad de que no serán perjudicados por su ingesta. Doutoramento em Informática<br>Durante as ultimas décadas, os registos de saúde eletrónicos (EHR) têm evoluído para se adaptar a novos requisitos. O cidadão tem-se envolvido cada vez mais na prestação dos cuidados médicos, sendo mais pró ativo e desejando potenciar a utilização do seu registo. A mobilidade do cidadão trouxe mais desafios, a existência de dados dispersos, heterogeneidade de sistemas e formatos e grande dificuldade de partilha e comunicação entre os prestadores de serviços. Para responder a estes requisitos, diversas soluções apareceram, maioritariamente baseadas em acordos entre instituições, regiões e países. Estas abordagens são usualmente assentes em cenários federativos muito complexos e fora do controlo do paciente. Abordagens mais recentes, como os registos pessoais de saúde (PHR), permitem o controlo do paciente, mas levantam duvidas da integridade clinica da informação aos profissionais clínicos. Neste cenário os dados saem de redes e sistemas controlados, aumentando o risco de segurança da informação. Assim sendo, são necessárias novas soluções que permitam uma colaboração confiável entre os diversos atores e sistemas. Esta tese apresenta uma solução que permite a colaboração aberta e segura entre todos os atores envolvidos nos cuidados de saúde. Baseia-se numa arquitetura orientada ao serviço, que lida com a informação clínica usando o conceito de envelope fechado. Foi modelada recorrendo aos princípios de funcionalidade e privilégios mínimos, com o propósito de fornecer proteção dos dados durante a transmissão, processamento e armazenamento. O controlo de acesso _e estabelecido por políticas definidas pelo paciente. Cartões de identificação eletrónicos, ou certificados similares são utilizados para a autenticação, permitindo uma inscrição automática. Todos os componentes requerem autenticação mútua e fazem uso de algoritmos de cifragem para garantir a privacidade dos dados. Apresenta-se também um modelo de ameaça para a arquitetura, por forma a analisar se as ameaças possíveis foram mitigadas ou se são necessários mais refinamentos. A solução proposta resolve o problema da mobilidade do paciente e a dispersão de dados, capacitando o cidadão a gerir e a colaborar na criação e manutenção da sua informação de saúde. A arquitetura permite uma colaboração aberta e segura, possibilitando que o paciente tenha registos mais ricos, atualizados e permitindo o surgimento de novas formas de criar e usar informação clínica ou complementar.<br>Since their early adoption Electronic Health Records (EHR) have been evolving to cope with increasing requirements from institutions, professionals and, more recently, from patients. Citizens became more involved demanding successively more control over their records and an active role on their content. Mobility brought also new requirements, data become scattered over heterogeneous systems and formats, with increasing di culties on data sharing between distinct providers. To cope with these challenges several solutions appeared, mostly based on service level agreements between entities, regions and countries. They usually required de ning complex federated scenarios and left the patient outside the process. More recent approaches, such as personal health records (PHR), enable patient control although raises clinical integrity doubts to other actors, such as physicians. Also, information security risk increase as data travels outside controlled networks and systems. To overcome this, new solutions are needed to facilitate trustable collaboration between the diverse actors and systems. In this thesis we present a solution that enables a secure and open collaboration between all healthcare actors. It is based on a service-oriented architecture that deals with the clinical data using a closed envelope concept. The architecture was modeled with minimal functionality and privileges bearing in mind strong protection of data during transmission, processing and storing. The access control is made through patient policies and authentication uses electronic identi cation cards or similar certi cates, enabling auto-enrollment. All the components require mutual authentication and uses cyphering mechanisms to assure privacy. We also present a threat model to verify, through our solution, if possible threats were mitigated or if further re nement is needed. The proposed solution solves the problem of patient mobility and data dispersion, and empowers citizens to manage and collaborate in their personal healthcare information. It also permits open and secure collaboration, enabling the patient to have richer and up to date records that can foster new ways to generate and use clinical or complementary information.

Mental health and obesity were ranked among the health priorities of the 2014 and 2017 Community Health Needs Assessments in Kent County, Michigan. Exposure to nature is correlated to improved health outcomes across a variety of morbidities including poor mental health and obesity. This cross-sectional study set within the frameworks of attention restoration theory, environmental health, and pathways to health benefits from nature assessed county survey data including self-reported nature exposures/interactions separated into 3 domain areas: access to nature, attitudes about nature, and physical activity in nature or in nature-based activities. Binary logistic regression analyses of the 653 respondents found that those who self-reported higher frequency of physical activity in nature or in nature-based activities possessed lower odds of also reporting poor mental health (p < .001, OR .652, 95% CI .535, .795) and obesity (p < .001, OR .666, 95% CI .548, .808) with each ascending level of agreement with the physical activity statement question. Ascending levels of agreement with the ease of access to nature statement question was found to be associated with lower odds of poor mental health (p < .001, OR .585, 95% CI .470, .797); however, no correlation was found between this variable and obesity status. The attitudes about nature domain statement questions were not consistently found to be associated with either mental health or obesity status. The significantly associated independent nature variables demonstrated weak effects (Nagelkerke R� < .300) on their respectively linked health outcomes. These findings may equip public health officials with information to develop more effective interventions for addressing mental health and obesity in their respective communities.

Natural killer (NK) cells possess the ability to destroy abnormal cells such as cancerous and virally-infected cells. A common feature of these cells is the downregulation of class I MHC molecules in an attempt to evade recognition by CD8+ T lymphocytes. "Missing-self" recognition allows NK cells to identify and destroy cells with decreased MHC-I expression. MHC-I receptors are expressed by NK cells, therefore allowing "missing-self" recognition. The major MHC-I receptors on human NK cells are KIR while mouse NK cells express Ly49 receptors. In addition to the recognition of MHC-I on target cells, Ly49 receptors are also involved in the education of NK cells, a process in which developing NK cells acquire effector functions.In human, KIR haplotypes vary greatly between individuals and the same phenomenon is observed in mice; Ly49 haplotypes differ in various mouse strains. Sequencing of the non-obese diabetic (NOD) Ly49 haplotype revealed that this strain encodes for the largest Ly49 haplotype known to date. The NOD Ly49 cluster is characterized by the largest number of genes coding for activating Ly49 receptors. The presence of a large number of Ly49 activating receptors may affect NK cell activation status and influence the development of diabetes in NOD mice.To gain insight in the in vivo role of Ly49 receptors in the control of NK cell activity, a NKC-knockdown mouse was generated. NK cells from these mice show greatly decreased expression of NKC-encoded receptors such as Ly49. NKC-knockdown NK cells have reduced lysis of MHC-I-negative target cells in vitro and in vivo. The ability of NK cells to destroy MHC-I-negative cells is restored by the re-introduction of a single Ly49 receptor. These results provide the first direct evidence that Ly49 receptors are involved in the in vivo surveillance of MHC-I molecules. Interestingly, the absence of Ly49 receptors results in defective "missing-self" recognition but activation of NK cells by MHC-I-independent mechanisms is not impaired.NKR-P1 receptors, which bind to Clr proteins, are members of another receptor family expressed by NK cells. Interestingly, the genes coding for NKR-P1 and Clr ligands are located in the same cluster and are therefore inherited together. Previous analyses demonstrated that the Nkrp1-Clr gene cluster is conserved in B6 and BALB/c mice. Mapping of the 129-strain cluster confirmed the high degree of conservation of the Nkrp1-Clr gene order in 129-strain mice. Gene sequencing revealed conservation of 129 and BALB/c Nkrp1 and Clr genes and divergence from the B6 genes, suggesting that the 129 cluster is more closely related to the BALB/c cluster than to the B6 cluster.Taken together, the Ly49 receptor family varies greatly in contrast with the highly conserved NKR-P1 receptors. Loss of Ly49 receptor expression reveals that Ly49 receptors are required for the acquisition of "missing-self" responses by NK cells.<br>Les cellules 'natural killer' (NK) possèdent l'habileté de détruire des cellules anormales telles que des cellules cancéreuses ou infectées par des virus. Chez l'humain, les haplotypes de gènes KIR varient grandement entre individus et le même phénomène est observé chez les souris; les haplotypes de gènes Ly49 diffèrent entre plusieurs souches de souris. Le séquençage de l'haplotype Ly49 des souris non-obèses diabétiques (NOD) a révélé que cette souche possède le plus grand haplotype de gènes Ly49 connu à ce jour. L'haplotype Ly49 des souris NOD est caractérisé par le plus grand nombre de gènes codant pour des récepteurs Ly49 activateurs. La présence d'un grand nombre de récepteurs Ly49 activateurs pourrait influencer le niveau d'activation des cellules NK et le développement du diabète chez les souris NOD.Afin de déduire le rôle in vivo des récepteurs Ly49 dans le contrôle de l'activité des cellules NK, une souris 'NKC-knockdown' a été créée. Les cellules NK de ces souris ont une expression grandement diminuée des récepteurs encodés dans le NKC dont les Ly49. Les cellules NK 'NKC-knockdown' possèdent une lyse réduite des cellules négative pour les CMH-I autant in vitro que in vivo. La capacité des cellules NK à détruire des cellules négatives pour les CMH-I est restaurée par la réintroduction d'un seul récepteur Ly49. Ces résultats fournissent la première preuve directe que les récepteurs Ly49 sont impliqués dans la surveillance des molécules CMH-I in vivo. Étonnamment, l'absence des récepteurs Ly49 entraine une reconnaissance de la 'perte de soi' défectueuse mais l'activation des cellules NK par des mécanismes n'impliquant pas des molécules CMH-I n'est pas diminuée. Les récepteurs NKR-P1, qui se lient aux protéines Clr, sont membres d'une autre famille de récepteurs exprimés par les cellules NK. Il est intéressant de noter que les gènes codant pour les NKR-P1 et leurs ligands Clr sont regroupés et sont donc hérités ensemble. Des analyses précédentes ont démontré que le groupe de gènes Nkrp1-Clr est conservé chez les souris B6 et BALB/c. La cartographie du groupe des souris de souche 129 a confirmé le haut degré de conservation de l'ordre des gènes dans le groupe Nkrp1-Clr des souris de souche 129. Le séquençage des gènes a démontré une conservation des gènes Nkrp et Clr chez les souris 129 et BALB/c tandis que ces gènes ont divergé chez les souris B6, suggérant que le groupe Nkrp1-Clr des souris de souche 129 est plus apparenté au groupe BALB/c qu'au groupe B6.En résumé, la famille de récepteurs Ly49 varie grandement contrairement au faible degré de diversité observé chez les récepteurs NKR-P1. La perte d'expression des récepteurs Ly49 révèle qu'ils sont requis pour l'acquisition de la réponse à la 'perte de soi' par les cellules NK.

Abstract:
This thesis addresses the problem of categorizing natural objects. To provide a criteria for categorization we propose that the purpose of a categorization is to support the inference of unobserved properties of objects from the observed properties. Because no such set of categories can be constructed in an arbitrary world, we present the Principle of Natural Modes as a claim about the structure of the world. We first define an evaluation function that measures how well a set of categories supports the inference goals of the observer. Entropy measures for property uncertainty and category uncertainty are combined through a free parameter that reflects the goals of the observer. Natural categorizations are shown to be those that are stable with respect to this free parameter. The evaluation function is tested in the domain of leaves and is found to be sensitive to the structure of the natural categories corresponding to the different species. We next develop a categorization paradigm that utilizes the categorization evaluation function in recovering natural categories. A statistical hypothesis generation algorithm is presented that is shown to be an effective categorization procedure. Examples drawn from several natural domains are presented, including data known to be a difficult test case for numerical categorization techniques. We next extend the categorization paradigm such that multiple levels of natural categories are recovered; by means of recursively invoking the categorization procedure both the genera and species are recovered in a population of anaerobic bacteria. Finally, a method is presented for evaluating the utility of features in recovering natural categories. This method also provides a mechanism for determining which features are constrained by the different processes present in a multiple modal world.

Abstract:
This thesis focuses on protein structure, dynamics and interaction and their relation to human disease. In particular, the biophysical and structural properties of both well-ordered and partially disordered proteins are studied using a range of biophysical techniques such as circular dichroism spectroscopy, fluorescence spectroscopy, mass spectrometry and nuclear magnetic resonance spectroscopy. Pseudomonas aeruginosa is a human pathogen due to its multidrug resistance (MDR) caused by overexpression of efflux pump systems. This thesis describes how MDR mutations within the MexR repressor of the MexAB-OprM system reduce the DNA affinity by altering its stability with maintained structure. The oncogenic protein c-Myc is involved in many essential biological functions such as cell proliferation, differentiation and apoptosis and is also highly associated with several forms of human cancers, and where the N-terminal domain is regulated by a plethora of protein interactions. In this thesis the intrinsically disordered N-terminal part of c-Myc and its interactions with the proteins Bin1 and TBP are described. Myc binds Bin1 with maintained disorder in a multivalent manner, which may explain why the onco-protein can interact with such a wide range of binding partners. A similarly dynamic interaction is observed for Myc with the TATA-binding protein (TBP). The essential human multidomain glutaredoxin Grx3 is associated with several biological functions such as redox signaling, proliferation and signal transduction. We have solved the structure and analyzed the dynamic properties in the ps-ns and ms time scale for the two N-terminal domains, providing a platform for further analysis of the Grx3 protein and its interactions. Taken together, this thesis emphasizes the importance of joint structural, biophysical and dynamic studies to better understand protein function in health and disease.

Doutoramento em Informática<br>Durante as ultimas décadas, os registos de saúde eletrónicos (EHR) têm evoluído para se adaptar a novos requisitos. O cidadão tem-se envolvido cada vez mais na prestação dos cuidados médicos, sendo mais pró ativo e desejando potenciar a utilização do seu registo. A mobilidade do cidadão trouxe mais desafios, a existência de dados dispersos, heterogeneidade de sistemas e formatos e grande dificuldade de partilha e comunicação entre os prestadores de serviços. Para responder a estes requisitos, diversas soluções apareceram, maioritariamente baseadas em acordos entre instituições, regiões e países. Estas abordagens são usualmente assentes em cenários federativos muito complexos e fora do controlo do paciente. Abordagens mais recentes, como os registos pessoais de saúde (PHR), permitem o controlo do paciente, mas levantam duvidas da integridade clinica da informação aos profissionais clínicos. Neste cenário os dados saem de redes e sistemas controlados, aumentando o risco de segurança da informação. Assim sendo, são necessárias novas soluções que permitam uma colaboração confiável entre os diversos atores e sistemas. Esta tese apresenta uma solução que permite a colaboração aberta e segura entre todos os atores envolvidos nos cuidados de saúde. Baseia-se numa arquitetura orientada ao serviço, que lida com a informação clínica usando o conceito de envelope fechado. Foi modelada recorrendo aos princípios de funcionalidade e privilégios mínimos, com o propósito de fornecer proteção dos dados durante a transmissão, processamento e armazenamento. O controlo de acesso _e estabelecido por políticas definidas pelo paciente. Cartões de identificação eletrónicos, ou certificados similares são utilizados para a autenticação, permitindo uma inscrição automática. Todos os componentes requerem autenticação mútua e fazem uso de algoritmos de cifragem para garantir a privacidade dos dados. Apresenta-se também um modelo de ameaça para a arquitetura, por forma a analisar se as ameaças possíveis foram mitigadas ou se são necessários mais refinamentos. A solução proposta resolve o problema da mobilidade do paciente e a dispersão de dados, capacitando o cidadão a gerir e a colaborar na criação e manutenção da sua informação de saúde. A arquitetura permite uma colaboração aberta e segura, possibilitando que o paciente tenha registos mais ricos, atualizados e permitindo o surgimento de novas formas de criar e usar informação clínica ou complementar.<br>Since their early adoption Electronic Health Records (EHR) have been evolving to cope with increasing requirements from institutions, professionals and, more recently, from patients. Citizens became more involved demanding successively more control over their records and an active role on their content. Mobility brought also new requirements, data become scattered over heterogeneous systems and formats, with increasing di culties on data sharing between distinct providers. To cope with these challenges several solutions appeared, mostly based on service level agreements between entities, regions and countries. They usually required de ning complex federated scenarios and left the patient outside the process. More recent approaches, such as personal health records (PHR), enable patient control although raises clinical integrity doubts to other actors, such as physicians. Also, information security risk increase as data travels outside controlled networks and systems. To overcome this, new solutions are needed to facilitate trustable collaboration between the diverse actors and systems. In this thesis we present a solution that enables a secure and open collaboration between all healthcare actors. It is based on a service-oriented architecture that deals with the clinical data using a closed envelope concept. The architecture was modeled with minimal functionality and privileges bearing in mind strong protection of data during transmission, processing and storing. The access control is made through patient policies and authentication uses electronic identi cation cards or similar certi cates, enabling auto-enrollment. All the components require mutual authentication and uses cyphering mechanisms to assure privacy. We also present a threat model to verify, through our solution, if possible threats were mitigated or if further re nement is needed. The proposed solution solves the problem of patient mobility and data dispersion, and empowers citizens to manage and collaborate in their personal healthcare information. It also permits open and secure collaboration, enabling the patient to have richer and up to date records that can foster new ways to generate and use clinical or complementary information.

Abstract:
A vaccine against the human immunodeficiency virus (HIV) is urgently needed. Recent estimates predict that 34 million people are currently infected with HIV. Attempts to induce potentially protective cytotoxic T-lymphocytes or broadly neutralizing antibodies by vaccination have either proven unsuccessful or failed to elicit the desired immune responses. However, the recent RV144 vaccine trial that provided partial protection against HIV infection appears to have induced antibodies that can utilize cells of the innate immune system, such as natural killer (NK) cells, to mediate antibody-dependent cellular cytotoxicity (ADCC) against HIV-infected cells. This potential mechanism of protection corroborates recent epidemiological and functional studies demonstrating that HIV-exposed seronegative individuals (HESN) and HIV-infected slow progressors (SP) have higher functioning NK cells and carry certain allelic combinations of killer immunoglobulin-like receptors (KIR) and their major histocompatibility complex class I (MHC-I or HLA-I) ligands that confer NK cells with enhanced functionality. Cumulatively, these observations suggest that understanding the conferral of functional potential during NK cell ontogeny could be important for designing anti-HIV vaccine constructs. In particular, data from HESN and SP have demonstrated that allelic combinations of KIR3DL1 and its HLA-Bw4 ligand are associated with protective outcomes in the context of HIV. Although previous work has demonstrated thatinteractions between these receptor ligand combinations during NK cell development confers NK cells with functional potential, the exact mechanism of the protective outcomes in the context of HIV remain unknown. For example, KIR3DL1+ NK cells have been demonstrated to be hypofunctional in the presence of autologous HIV-infected T cells. This suggests that if KIR3DL1+ NK cells are providing protection through mediating function, they require additional activating signals. As previously published data has demonstrated that activation through CD16a by antibody constant regions can overcome KIR-mediated NK cell inhibition and lead to ADCC of allogeneic cells, we hypothesized that ADCC could overcome inhibitory signalling and allow KIR3DL1+ NK cells to respond to autologous anti-HIV ADCC target cells. The data presented in this thesis reaffirms that HIV protective KIR3DL1/HLA-Bw4 allelic combinations confer enhanced functional potential upon NK cells. The results presented demonstrate that KIR3DL1/HLA-Bw4 combinations educate NK cells for enhanced anti-HIV ADCC against autologous target cells, and that this educational advantage is maintained after stimulation with function-conferring cytokines, such as IL-15. Furthermore, allelic combinations of KIR3DL1/HLA-Bw4 that are protective in the context of HIV are shown to confer the highest ADCC functional potential. These data suggest that the education of NK cells by allelic combinations of KIR3DL1/HLA-Bw4 could explain some of the protection observed in individuals with these combined genotypes. However, as we also observed anabrogation of this education-conferred functional advantage in HIV-infected individuals, we propose that the mechanisms of NK cell-mediated protection differ between uninfected HESN and infected SP.<br>Not enough space

Abstract:
This thesis investigates coreference and its resolution within the domain of health education. Coreference is the relationship between two linguistic expressions that refer to the same real-world entity, and resolution involves identifying this relationship among sets of referring expressions. The coreference resolution task is considered among the most difficult of problems in Artificial Intelligence; in some cases, resolution is impossible even for humans. For example, "she" in the sentence "Lynn called Jennifer while she was on vacation" is genuinely ambiguous: the vacationer could be either Lynn or Jennifer. <br/><br/> There are three primary motivations for this thesis. The first is that health education has never before been studied in this context. So far, the vast majority of coreference research has focused on news. Secondly, achieving domain-independent resolution is unlikely without understanding the extent to which coreference varies across different genres. Finally, coreference pervades language and is an essential part of coherent discourse. Its effective use is a key component of easy-to-understand health education materials, where readability is paramount. <br/><br/> No suitable corpus of health education materials existed, so our first step was to create one. The comprehensive analysis of this corpus, which required manual annotation of coreference, confirmed our hypothesis that the coreference used in health education differs substantially from that in previously studied domains. This analysis was then used to shape the design of a knowledge-lean algorithm for resolving coreference. This algorithm performed surprisingly well on this corpus, e.g., successfully resolving over 85% of all pronouns when evaluated on unseen data. <br/><br/> Despite the importance of coreferentially annotated corpora, only a handful are known to exist, likely because of the difficulty and cost of reliably annotating coreference. The paucity of genres represented in these existing annotated corpora creates an implicit bias in domain-independent coreference resolution. In an effort to address these issues, we plan to make our health education corpus available to the wider research community, hopefully encouraging a broader focus in the future.
