In this thesis we propose some new approaches to the study of complex networks, and apply them to multiple domains, focusing in particular on protein-protein interaction networks. We begin by examining the roles of individual proteins; specifically, the influential idea of 'date' and 'party' hubs. The adenovirus major late transcription unit (MLTU) encodes for most of the mRNAs that are translated into the structural proteins of the virus capsid. Transcription from the MLTU is directed by the major late promoter (MLP), which is highly activated during the late phase of infection. This thesis discusses how the adenovirus-encoded L4-22K protein regulates the MLP at both the level of transcription and pre-mRNA splicing. Amyloid-beta protein (Aß) is strongly linked to the aetiology of Alzheimer’s disease (AD). Even though Aß has a central role in AD, this protein is normally produced in healthy humans. It is the aberrant processing of Aß that determines its accumulation and aggregation into large oligomer species that evolve into the fibrillar structures deposited in amyloid plaques in the brain of AD patients. The neurotoxicity observed in AD has been attributed to the oligomeric intermediates, although their stoichiometry and structure still remain unknown. The genome constitution and phylogeny of hexaploid timothy (Phleum pratense L., 2n = 42) have been studied with C-banding and genome-specific DNA techniques. The cultivated species and two diploid wild relatives P. alpinum (2n = 14) and P. bertolonii (2n = 14) were karyotyped. In P. pratense, two sets of seven chromosomes could not be distinguished from each other either in gross morphology or in C-banding patterns and the third set was found to be differentiated from them.
