Abstract:
Small extracellular vesicles (EVs) are able to pass from the central nervous system (CNS) into peripheral blood and contain molecule markers of their parental origin. The aim of our study was to isolate and characterize total and neural-derived small EVs (NDEVs) and their micro RNA (miRNA) cargo in Alzheimer’s disease (AD) patients. Small NDEVs were isolated from plasma in a population consisting of 40 AD patients and 40 healthy subjects (CTRLs) using high throughput Advanced TaqMan miRNA OpenArrays®, which enables the simultaneous determination of 754 miRNAs. MiR-23a-3p, miR-223-3p, miR-100-3p and miR-190-5p showed a significant dysregulation in small NDEVs from AD patients as compared with controls (1.16 ± 0.49 versus 7.54 ± 2.5, p = 0.026; 9.32 ± 2.27 versus 0.66 ± 0.18, p <0.0001; 0.069 ± 0.01 versus 0.5 ± 0.1, p < 0.0001 and 2.9 ± 1.2 versus 1.93 ± 0.9, p < 0.05, respectively). A further validation analysis confirmed that miR-23a-3p, miR-223-3p and miR-190a-5p levels in small NDEVs from AD patients were significantly upregulated as compared with controls (p = 0.008; p = 0.016; p = 0.003, respectively) whereas miR-100-3p levels were significantly downregulated (p = 0.008). This is the first study that carries out the comparison between total plasma small EV population and NDEVs, demonstrating the presence of a specific AD NDEV miRNA signature.

Abstract:
Individuals with Down syndrome (DS) exhibit Alzheimer’s disease (AD) pathology at a young age, including amyloid plaques and neurofibrillary tangles (NFTs). Tau pathology can spread via extracellular vesicles, such as exosomes. The cargo of neuron-derived small extracellular vesicles (NDEVs) from individuals with DS contains p-Tau at an early age. The goal of the study was to investigate whether NDEVs isolated from the blood of individuals with DS can spread Tau pathology in the brain of wildtype mice. We purified NDEVs from the plasma of patients with DS-AD and controls and injected small quantities using stereotaxic surgery into the dorsal hippocampus of adult wildtype mice. Seeding competent Tau conformers were amplified in vitro from DS-AD NDEVs but not NDEVs from controls. One month or 4 months post-injection, we examined Tau pathology in mouse brains. We found abundant p-Tau immunostaining in the hippocampus of the mice injected with DS-AD NDEVs compared to injections of age-matched control NDEVs. Double labeling with neuronal and glial markers showed that p-Tau staining was largely found in neurons and, to a lesser extent, in glial cells and that p-Tau immunostaining was spreading along the corpus callosum and the medio-lateral axis of the hippocampus. These studies demonstrate that NDEVs from DS-AD patients exhibit Tau seeding capacity and give rise to tangle-like intracellular inclusions.

Abstract:
Baseline entomological information should be collected before the implementation of industrial projects in malaria endemic areas. This allows for subsequent monitoring and evaluation of the project impact on malaria vectors. This study aimed at assessing the vectorial system and malaria transmission in two ecologically different villages of the South-Cameroon forest bloc targeted for the creation of an agro-industrial complex. For four consecutive seasons in 2013, adult mosquitoes were captured using Human Landing Catch in NDELLE village (located along a main road in a degraded forest with many fish ponds) and KOMBO village (located 5km far from the main road in a darker forest and crossed by the Mvobo River). Morpho-taxonomic techniques were used alongside molecular techniques for the identification of mosquito species. ELISA test was used for the detection of circumsporozoite protein antigen of <em>Plasmodium falciparum</em>. Mosquito biting rate was higher in NDELLE than in KOMBO (28.18 <em>versus</em> 17.34 bites per person per night). Mosquitoes had a strong tendency to endophagy both in NDELLE (73.57%) and KOMBO (70.21%). Three anophelines species were identified; <em>An. gambiae</em>, <em>An. funestus</em> s.s and <em>An. moucheti</em> s.s.. <em>An. gambiae</em> and <em>An. funestus</em> s.s. represented the bulk of aggressive mosquitoes in NDELLE (n=10,891; 96.62%). <em>An. gambiae</em> was responsible for 62.6% and 77.72% of malaria transmission in KOMBO and NDELLE respectively. Mean entomological inoculation rate recorded in KOMBO and NDELLE were 4.82 and 2.02 infective bites per person per night respectively. Vector control was mainly based on the use of long-lasting insecticidal nets and indoor residual spraying. The degraded forest environment added to the presence of fishponds resulted in the increase of aggressive mosquito density but not of malaria transmission. The managers should use these data for monitoring and evaluation of the impact of their project; malaria control strategies should be included in their project in order to mitigate the risk of increased malaria transmission as a result of the implementation of their projects.

Abstract:
Microglia act as the immune cells of the central nervous system (CNS). They play an important role in maintaining brain homeostasis but also in mediating neuroimmune responses to insult. The interactions between neurons and microglia represent a key process for neuroimmune regulation and subsequent effects on CNS integrity. However, the molecular mechanisms of neuron-glia communication in regulating microglia function are not fully understood. One recently described means of this intercellular communication is via nano-sized extracellular vesicles (EVs) that transfer a large diversity of molecules between neurons and microglia, such as proteins, lipids, and nucleic acids. To determine the effects of neuron-derived EVs (NDEVs) on microglia, NDEVs were isolated from the culture supernatant of rat cortical neurons. When NDEVs were added to primary cultured rat microglia, we found significantly improved microglia viability via inhibition of apoptosis. Additionally, application of NDEVs to cultured microglia also inhibited the expression of activation surface markers on microglia. Furthermore, NDEVs reduced the LPS-induced proinflammatory response in microglia according to reduced gene expression of proinflammatory cytokines (TNF-α, IL-6, MCP-1) and iNOS, but increased expression of the anti-inflammatory cytokine, IL-10. These findings support that neurons critically regulate microglia activity and control inflammation via EV-mediated neuron–glia communication. (Supported by R21AA025563 and R01AA025591).

AbstractOverall healing, harmony, and balance within Ndee (Apache) communities can be understood through the Ndee term Gózhó—“beauty, balance, and harmony.” Beyond this Ndee cultural precept, however, how are such states of Gózhó achieved, whether in the past or today? Within archaeological research on Ndee lands, achieving healing and promoting health is crucial to overall individual and community well-being. Healing activities that contribute to overall states of Gózhó can take many forms. As Rybak and Decker-Fitts (2009:334) suggest, “Native American healing practices exemplify key cultural perspectives and influence the identity development of Native American individuals.” Such healing practices including the use of yellow cattail pollen (hádńdín; Typha spp.) for protection are based on traditions and perspectives typically outside the mainstream of Western psychological tenets, yet they can have significant impact on the sense of well-being for Ndee communities. Ndee cultural specialists with the White Mountain Apache Tribe demonstrate that such traditions and perspectives are not only necessary for continued community well-being but are powerful conduits amplifying the past as present, which contribute to the ongoing cyclical and reciprocal nature of the ways Ndee communities have related to Nígosdzán—Mother Earth—since time immemorial.

Cette description de l'agni (langue kwa de cote d'ivoire) comprend quatre parties de longueurs inegales. La premiere (ch. 1) est consacree aux unites de deuxieme articulation : phonologie (systemes phonematique et tonal). Les trois autres (ch. 2 a 7) etudient les unites de premiere articulation. Tout d'abord la grammaire (ch. 2 a 5) etudie les lois tres generales et absolument contraignantes du fonctionnement de l'agni. Ceci est fait en deux niveaux : a) celui de la lexicologie(ch. 2 : lexicologie des bases ; ch. 3 : lexicologie du constituant nominal ; ch. 4 : lexicologie du constituant verbal) ; b) celui de la syntaxe (ch. 5) qui etudie la structure generale de l'enonce agni. Une autre partie (ch. 6) intitulee de la grammaire au lexique se situe a un niveau intermediaire entre celui tres general de la grammaire et celui des cas particuliers du lexique. A l'interieur du scheme d'enonce decrit en grammaire, elle dit quelles sont les structures d'enonces effectivement realisees par opposition aux autres structures egalement possibles mais qui ne sont pas attestees. Elle manifeste quels sont les choix ou habitudes enonciatives de la communaute linguistique agni. La derniere partie (ch. 7) intitulee lexique presente une conception du lexique articulee sur les resultats obtenus dans la partie precedente et s'attache plus particulierement a l'etude du lexique des verbes.

One of the key aspects of Shaaban Robert’s artistic techniques is his remarkable molding of winged creature images in the form of extraordinary migratory birds, songbirds, angels and winged termites to infuse the motif of ascendance, progress and journey. So far critics of Robert’s works have given this significant artistic tool a very low prominence in spite of its recurrence in his writings. This paper argues that Robert devulges two opposing forces through scary bird imagery. Robert successfully displays his creative artistry by supplanting the bird mythology drawn from the reservoir of folkloric knowledge of his people to appeal metaphorically to his readers’ senses.

The study dealt with enrichment of Tshivenḓa language through adoption of words from other languages such as English, Afrikaans, Sotho, Tsonga, Zulu and many more. The study discovered that no language can remain static forever and this applies to Tshivenḓa as well. Every successive generation makes its own small contribution to language change and when sufficient time has elapsed the impact of these changes becomes more obvious. It however cautions that borrowing of words should not be overdone as this may lead to the disappearance of Tshivenḓa as we know it.

The Western Apache Scouts of the 1870s who assisted the United States Army in tracking down the Chiricahua Apaches that had escaped from the federal reservations in the Arizona Territory laid the foundation for what became seventy-six years of military service in the U.S. Army. Consolidated and reassigned to Ft. Huachuca, Arizona in 1922, these scouts continued to serve with distinction long after the Army needed their skills as trackers. In 1947, the final four scouts retired from United States military service, each having served for over twenty-five years. This thesis explores how these men used their military service in order to survive, serving with honor while maintaining their cultural traditions within a changing world.

Abstract:
This chapter explores the use of aircraft to spray organochlorine pesticides (OCPs). It begins by tracing the origins of the practice in KwaZulu, South Africa, where ndege were adopted for pesticide spraying because of their ease of use, capacity for large-scale coverage, very few personnel required, and capability to reach mpukane habitats otherwise inaccessible by ground spraying. The second section examines the technical aspects of aerial spraying as an example of the extension of methods designed in the United States for agricultural or military purposes to deal with zvipukanana and with conditions for which they were not originally designed. In the final sections, the deployment and performance of first fixed-wing aircraft and then helicopters are closely examined.
