Abstract:
Small extracellular vesicles (EVs) are able to pass from the central nervous system (CNS) into peripheral blood and contain molecule markers of their parental origin. The aim of our study was to isolate and characterize total and neural-derived small EVs (NDEVs) and their micro RNA (miRNA) cargo in Alzheimer’s disease (AD) patients. Small NDEVs were isolated from plasma in a population consisting of 40 AD patients and 40 healthy subjects (CTRLs) using high throughput Advanced TaqMan miRNA OpenArrays®, which enables the simultaneous determination of 754 miRNAs. MiR-23a-3p, miR-223-3p, miR-100-3p and miR-190-5p showed a significant dysregulation in small NDEVs from AD patients as compared with controls (1.16 ± 0.49 versus 7.54 ± 2.5, p = 0.026; 9.32 ± 2.27 versus 0.66 ± 0.18, p <0.0001; 0.069 ± 0.01 versus 0.5 ± 0.1, p < 0.0001 and 2.9 ± 1.2 versus 1.93 ± 0.9, p < 0.05, respectively). A further validation analysis confirmed that miR-23a-3p, miR-223-3p and miR-190a-5p levels in small NDEVs from AD patients were significantly upregulated as compared with controls (p = 0.008; p = 0.016; p = 0.003, respectively) whereas miR-100-3p levels were significantly downregulated (p = 0.008). This is the first study that carries out the comparison between total plasma small EV population and NDEVs, demonstrating the presence of a specific AD NDEV miRNA signature.

Abstract:
Individuals with Down syndrome (DS) exhibit Alzheimer’s disease (AD) pathology at a young age, including amyloid plaques and neurofibrillary tangles (NFTs). Tau pathology can spread via extracellular vesicles, such as exosomes. The cargo of neuron-derived small extracellular vesicles (NDEVs) from individuals with DS contains p-Tau at an early age. The goal of the study was to investigate whether NDEVs isolated from the blood of individuals with DS can spread Tau pathology in the brain of wildtype mice. We purified NDEVs from the plasma of patients with DS-AD and controls and injected small quantities using stereotaxic surgery into the dorsal hippocampus of adult wildtype mice. Seeding competent Tau conformers were amplified in vitro from DS-AD NDEVs but not NDEVs from controls. One month or 4 months post-injection, we examined Tau pathology in mouse brains. We found abundant p-Tau immunostaining in the hippocampus of the mice injected with DS-AD NDEVs compared to injections of age-matched control NDEVs. Double labeling with neuronal and glial markers showed that p-Tau staining was largely found in neurons and, to a lesser extent, in glial cells and that p-Tau immunostaining was spreading along the corpus callosum and the medio-lateral axis of the hippocampus. These studies demonstrate that NDEVs from DS-AD patients exhibit Tau seeding capacity and give rise to tangle-like intracellular inclusions.

Abstract:
Baseline entomological information should be collected before the implementation of industrial projects in malaria endemic areas. This allows for subsequent monitoring and evaluation of the project impact on malaria vectors. This study aimed at assessing the vectorial system and malaria transmission in two ecologically different villages of the South-Cameroon forest bloc targeted for the creation of an agro-industrial complex. For four consecutive seasons in 2013, adult mosquitoes were captured using Human Landing Catch in NDELLE village (located along a main road in a degraded forest with many fish ponds) and KOMBO village (located 5km far from the main road in a darker forest and crossed by the Mvobo River). Morpho-taxonomic techniques were used alongside molecular techniques for the identification of mosquito species. ELISA test was used for the detection of circumsporozoite protein antigen of <em>Plasmodium falciparum</em>. Mosquito biting rate was higher in NDELLE than in KOMBO (28.18 <em>versus</em> 17.34 bites per person per night). Mosquitoes had a strong tendency to endophagy both in NDELLE (73.57%) and KOMBO (70.21%). Three anophelines species were identified; <em>An. gambiae</em>, <em>An. funestus</em> s.s and <em>An. moucheti</em> s.s.. <em>An. gambiae</em> and <em>An. funestus</em> s.s. represented the bulk of aggressive mosquitoes in NDELLE (n=10,891; 96.62%). <em>An. gambiae</em> was responsible for 62.6% and 77.72% of malaria transmission in KOMBO and NDELLE respectively. Mean entomological inoculation rate recorded in KOMBO and NDELLE were 4.82 and 2.02 infective bites per person per night respectively. Vector control was mainly based on the use of long-lasting insecticidal nets and indoor residual spraying. The degraded forest environment added to the presence of fishponds resulted in the increase of aggressive mosquito density but not of malaria transmission. The managers should use these data for monitoring and evaluation of the impact of their project; malaria control strategies should be included in their project in order to mitigate the risk of increased malaria transmission as a result of the implementation of their projects.

Full text
Abstract:
Microglia act as the immune cells of the central nervous system (CNS). They play an important role in maintaining brain homeostasis but also in mediating neuroimmune responses to insult. The interactions between neurons and microglia represent a key process for neuroimmune regulation and subsequent effects on CNS integrity. However, the molecular mechanisms of neuron-glia communication in regulating microglia function are not fully understood. One recently described means of this intercellular communication is via nano-sized extracellular vesicles (EVs) that transfer a large diversity of molecules between neurons and microglia, such as proteins, lipids, and nucleic acids. To determine the effects of neuron-derived EVs (NDEVs) on microglia, NDEVs were isolated from the culture supernatant of rat cortical neurons. When NDEVs were added to primary cultured rat microglia, we found significantly improved microglia viability via inhibition of apoptosis. Additionally, application of NDEVs to cultured microglia also inhibited the expression of activation surface markers on microglia. Furthermore, NDEVs reduced the LPS-induced proinflammatory response in microglia according to reduced gene expression of proinflammatory cytokines (TNF-α, IL-6, MCP-1) and iNOS, but increased expression of the anti-inflammatory cytokine, IL-10. These findings support that neurons critically regulate microglia activity and control inflammation via EV-mediated neuron–glia communication. (Supported by R21AA025563 and R01AA025591).

Abstract:
AbstractOverall healing, harmony, and balance within Ndee (Apache) communities can be understood through the Ndee term Gózhó—“beauty, balance, and harmony.” Beyond this Ndee cultural precept, however, how are such states of Gózhó achieved, whether in the past or today? Within archaeological research on Ndee lands, achieving healing and promoting health is crucial to overall individual and community well-being. Healing activities that contribute to overall states of Gózhó can take many forms. As Rybak and Decker-Fitts (2009:334) suggest, “Native American healing practices exemplify key cultural perspectives and influence the identity development of Native American individuals.” Such healing practices including the use of yellow cattail pollen (hádńdín; Typha spp.) for protection are based on traditions and perspectives typically outside the mainstream of Western psychological tenets, yet they can have significant impact on the sense of well-being for Ndee communities. Ndee cultural specialists with the White Mountain Apache Tribe demonstrate that such traditions and perspectives are not only necessary for continued community well-being but are powerful conduits amplifying the past as present, which contribute to the ongoing cyclical and reciprocal nature of the ways Ndee communities have related to Nígosdzán—Mother Earth—since time immemorial.

Abstract:

In studies concerning Islam and gender in West Africa, the expertise of women in Islamic esoteric practices is often overlooked. These practices, that include divination, dream interpretation and prayer sessions, are central in politics, economics and the daily life of most West Africans. Furthermore, their products (such as amulets) and their practitioners (marabouts) travel to Europe, the United States, and the Middle East. Despite the importance of Islamic esoteric practices in West Africa and the rest of the world, they are understudied. In this article, I focus upon the life and work of two marabout women living in Dakar: Ndeye Meissa Ndiaye and Coumba Keita. Their position is exceptional: Islamic esoteric knowledge is a particularly male-dominated field. This article describes how two women's Islamic esoteric expertise is negotiated, legitimated and publicly recognized in Dakar.

Abstract:
This article examines the role of radios in conflict by exploring the tenets of peace journalism in the United Nations sponsored Radio Okapi (Democratic Republic of Congo) and Radio Ndeke Luka (Central African Republic) run by the Swiss Fondation Hirondelle. It is a qualitative research that interviewed journalists on how they perceive their role in society and margin of autonomy. It aims at answering the question: To what extent do the conventions of professional practice of journalism affect the way newsmaking is shaped under the peace journalism approach in conflict-stressed environments? The findings pointed that peace journalism encompasses the idea of a symbolic rapprochement and reconcilement. Reporters stressed the notion of using journalism as a pedagogical tool. Many of the journalists have gone through life-threatening situations caused by opposition groups. Nonetheless, the testimonies accounted for a willingness to carry on with their commitment to a responsible journalism.

Abstract:
Nepeta deflersiana (Lamiaceae) is a well-known medicinal plant that grows in Saudi Arabia. This plant is used in Saudi and Yemeni folk medicine as an anti-inflammatory, carminative, and antirheumatic agent. In order to prove its use in folk medicine, four different extracts from the aerial parts of the plant: petroleum ether, chloroform, ethyl acetate, and n-butanol extracts were subjected to biological assays to screen PPARα and PPARϒ agnostic, antioxidant, anti-inflammatory, and cytotoxic activities. Ethyl acetate and n-butanol extracts of N. deflersiana NDEE and NDBE, respectively, showed a decrease in oxidative stress and inhibition of both NF-kB and iNOS activities with no cytotoxic effects on four human cancer cell lines. Both active extracts were standardized using two bioactive metabolites which were isolated from the aerial parts of the same plant [8-epi-7-deoxyloganic acid (compound 1) and Ursolic acid (compound 2)] by developing a validated HPTLC method. It was found to provide a sharp and compact band of compound 1 at Rf = 0.07 and Rf = 0.57 for compound 2, using chloroform, methanol, and formic acid (8.9:0.8:0.3, v/v/v) as mobile phase at 550 nm. Compounds 1 and 2 were found in NDEE by 9.59 %, w/w, and 84.63 %, w/w, respectively, and by 11.97 %, w/w, and 21.26 %, w/w, respectively, in NDBE.

Abstract:
Abstract The prehistoric settlement of the west coast of the Senegalese-Mauritanian basin is established from archaeological remains and coal samples collected, sometimes in a stratigraphic context. However, the chronology issued, in the Before Present (BP) age, does not take into account the taphonomic context of the sites and the local reservoir age. Therefore, this article revisits the chronologies obtained based on the 14C literature and dating(s) acquired. Changes in time and duration of human occupancy of the area are shorter or longer depending on adequate yields of local reservoir age (Ndeye, 2008), which is a relevant element for marine samples. Thus, the archaeological implications observed with the reservoir effect are the rejuvenation or ageing of the dates, the age of the sites, the duration of occupation prehistoric or historical sites studied. Using the calibration programmes, it is noted that for the site of Senegal (Khant), without taking into account the reservoir effect, the human occupation is a priori, from the fifth millennium (Ancient Neolithic) to the third millennium BC (Middle Neolithic). However, if this marine reservoir effect is applied, the chronological periodisation goes from the fourth millennium to the first millennium. For the Mauritanian sites, the reservoir age correction is necessary for the Chami site while for the Tintan site is not required. Therefore, the calibrated archaeological chronologies obtained after the application of the marine reservoir effect are more relevant.

Abstract:
Research on amphibian diversity in Rivers State, Nigeria, was conducted between 2015 and 2019. The survey covered seven Local Government Areas and three ecological zones (lowland forest, freshwater swamp and short mangrove). The surveyed areas were Ikwerre LGA (Isiokpo), Emohua (Rumuji and Ndele), Okrika (Fiberesima Polo, ATC sandfill, Greame Ama), Khana- Ogoni (Bori, Kaani 1), Ogba-Egbema-Ndoni (Omoku), Ahoada East (Ahoada main town, Ikata) and Obio-Akpor (Agbada, Rumuagholu, Rumuesara). The habitats included farmlands, fallow areas, swamps, streams and inundated areas, bush paths, human settlements and forested areas. Visual encounter and acoustic survey methods were used. Nineteen amphibian species were encountered including members of the Arthroleptidae, Bufonidae, Dicroglossidae, Ranidae, Hyperoliidae, Ptychadenidae and Pipidae. Freshwater swamps and short mangrove ecological zones (characterized by human settlements and farmlands) had the lowest diversity of two and five amphibian species, respectively. The highest diversity comprised of eighteen different amphibian species was recorded in forested habitats of the lowland forest/freshwater swamp ecological zone. 569 individuals were collected from lowland forest/freshwater swamp ecological zone; 215 from lowland forest; 60 from short mangrove; and 28 from freshwater swamp ecological zone. Shannon diversity was highest in the lowland/freshwater swamp ecological zone (H=2.12) and lowest in the freshwater swamp zone (H=0.52). It is concluded that a forested habitat comprised of the combination of lowland forest and freshwater swamp ecosystems hosts a greater diversity of amphibian species.

Abstract:
Objective: The aim of this research was to evaluate the acute and sub-acute oral toxicities of the nebulized dried extract of Myracrodruon urundeuva (NDEMU) leaf obtained by the spray drying technique on rabbits. Methods: In the acute toxicity study, the amount of nebulized dried extract (NDE) administered was adjusted to a dose of 2000 mg/kg of leaf powder of M. urundeuva to 6 rabbits once orally and were observed for 14 days. In the sub-acute study, the amount of NDEMU administered was adjusted to a dose of 2000 mg/kg/day of to 6 rabbits once daily for 30 day, orally. The appearance of toxic symptoms was observed every day, followed by each rabbits' food and drink intake. Haematological and biochemical analysis were observed and statistical analysis was performed on them. The rabbits were killed at the end of the study, and their organs were weighed and examined before organ histology were evaluated. Results: No toxic signs and no mortality were observed in the acute and sub-acute study. In the sub-acute study, the amount of dried extract administered was adjusted to a dose of 2000 mg/kg of leaf powder of M. urundeuva to 6 rabbits once daily for 30 days, orally. No toxic signs and no mortality were observed. There were no significant changes (p < 0.05) in the body weights, organ weights and haemato-biochemical parameters in any of the dose levels. No related histopathological lesions were observed. Conclusion: The results indicate that the treatment of repeated doses with the dried NDEME showed low toxicity in rabbits.

Abstract:
Just-in-time software defect prediction (JIT-SDP) is a fine-grained software defect prediction technology, which aims to identify the defective code changes in software systems. Effort-aware software defect prediction is a software defect prediction technology that takes into consideration the cost of code inspection, which can find more defective code changes in limited test resources. The traditional effort-aware defect prediction model mainly measures the effort based on the number of lines of code (LOC) and rarely considers additional factors. This paper proposes a novel effort measure method called Multi-Metric Joint Calculation (MMJC). When measuring the effort, MMJC takes into account not only LOC, but also the distribution of modified code across different files (Entropy), the number of developers that changed the files (NDEV) and the developer experience (EXP). In the simulation experiment, MMJC is combined with Linear Regression, Decision Tree, Random Forest, LightGBM, Support Vector Machine and Neural Network, respectively, to build the software defect prediction model. Several comparative experiments are conducted between the models based on MMJC and baseline models. The results show that indicators ACC and [Formula: see text] of the models based on MMJC are improved by 35.3% and 15.9% on average in the three verification scenarios, respectively, compared with the baseline models.

Abstract:
Unmanned Aerial Vehicles (UAVs) are emerging and promising platforms for carrying different types of cameras for remote sensing. The application of multispectral vegetation indices for ground cover classification has been widely adopted and has proved its reliability. However, the fusion of spectral bands and vegetation indices for machine learning-based land surface investigation has hardly been studied. In this paper, we studied the fusion of spectral bands information from UAV multispectral images and derived vegetation indices for almond plantation classification using several machine learning methods. We acquired multispectral images over an almond plantation using a UAV. First, a multispectral orthoimage was generated from the acquired multispectral images using SfM (Structure from Motion) photogrammetry methods. Eleven types of vegetation indexes were proposed based on the multispectral orthoimage. Then, 593 data points that contained multispectral bands and vegetation indexes were randomly collected and prepared for this study. After comparing six machine learning algorithms (Support Vector Machine, K-Nearest Neighbor, Linear Discrimination Analysis, Decision Tree, Random Forest, and Gradient Boosting), we selected three (SVM, KNN, and LDA) to study the fusion of multi-spectral bands information and derived vegetation index for classification. With the vegetation indexes increased, the model classification accuracy of all three selected machine learning methods gradually increased, then dropped. Our results revealed that that: (1) spectral information from multispectral images can be used for machine learning-based ground classification, and among all methods, SVM had the best performance; (2) combination of multispectral bands and vegetation indexes can improve the classification accuracy comparing to only spectral bands among all three selected methods; (3) among all VIs, NDEGE, NDVIG, and NDVGE had consistent performance in improving classification accuracies, and others may reduce the accuracy. Machine learning methods (SVM, KNN, and LDA) can be used for classifying almond plantation using multispectral orthoimages, and fusion of multispectral bands with vegetation indexes can improve machine learning-based classification accuracy if the vegetation indexes are properly selected.

Abstract:

Abstract:
AbstractTo characterize neuronal mitochondrial abnormalities in major depressive disorder (MDD), functional mitochondrial proteins (MPs) extracted from enriched plasma neuron-derived extracellular vesicles (NDEVs) of MDD participants (n = 20) were quantified before and after eight weeks of treatment with a selective serotonin reuptake inhibitor (SSRI). Pretreatment baseline NDEV levels of the transcriptional type 2 nuclear respiratory factor (NRF2) which controls mitochondrial biogenesis and many anti-oxidant gene responses, regulators of diverse neuronal mitochondrial functions cyclophilin D (CYPD) and mitofusin-2 (MFN2), leucine zipper EF-hand containing transmembrane 1 protein (LETM1) component of a calcium channel/calcium channel enhancer, mitochondrial tethering proteins syntaphilin (SNPH) and myosin VI (MY06), inner membrane electron transport complexes I (subunit 6) and III (subunit 10), the penultimate enzyme of nicotinamide adenine dinucleotide (NAD) generation nicotinamide mononucleotide adenylytransferase 2 (NMNAT2), and neuronal mitochondrial metabolic regulatory and protective factors humanin and mitochondrial open-reading frame of the 12S rRNA-c (MOTS-c) all were significantly lower than those of NDEVs from matched controls (n = 10), whereas those of pro-neurodegenerative NADase Sterile Alpha and TIR motif-containing protein 1 (SARM1) were higher. The baseline NDEV levels of transcription factor A mitochondrial (TFAM) and the transcriptional master-regulator of mitochondrial biogenesis PPAR γ coactivator-1α (PGC-1α) showed no differences between MDD participants and controls. Several of these potential biomarker proteins showed substantially different changes in untreated MDD than those we reported in untreated first-episode psychosis. NDEV levels of MPs of all functional classes, except complex I-6, NRF2 and PGC-1α were normalized in MDD participants who responded to SSRI therapy (n = 10) but not in those who failed to respond (n = 10) by psychiatric evaluation. If larger studies validate NDEV MP abnormalities, they may become useful biomarkers and identify new drug targets.

Abstract:
IntroductionDiabetes increases the risk of Parkinson’s disease (PD). The phosphorylation of type 1 insulin receptor substrate (IRS-1) determines the function of insulin signaling pathway. Extracellular vesicles (EVs) are emerging as biomarkers of human diseases. The present study investigated whether PD patients exert altered phosphorylation IRS-1 (p-IRS-1) inside the blood neuron-derived extracellular vesicles (NDEVs).Research Design and MethodsIn total, there were 94 patients with PD and 63 healthy controls recruited and their clinical manifestations were evaluated. Blood NDEVs were isolated using the immunoprecipitation method, and Western blot analysis was conducted to assess total IRS-1, p-IRS-1, and downstream substrates level in blood NDEVs. Statistical analysis was performed using SPSS 19.0, and p &lt; 0.05 was considered significant.ResultsThe isolated blood EVs were validated according to the presence of CD63 and HSP70, nanoparticle tracking analysis and transmission electron microscopy. NDEVs were positive with neuronal markers. PD patients exerted significantly higher level of p-IRS-1S312 in blood NDEVs than controls. In addition, the p-IRS-1S312 levels in blood NDEVs was positively associated with the severity of tremor in PD patients after adjusting of age, sex, hemoglobin A1c, and body mass index (BMI).ConclusionPD patients exerted altered p-IRS-1S312 in the blood NDEVs, and also correlated with the severity of tremor. These findings suggested the association between dysfunctional insulin signaling pathway with PD. The role of altered p-IRS-1S312 in blood NDEVs as a segregating biomarker of PD required further cohort study to assess the association with the progression of PD.

The White Mountain Apache Tribe Cultural Heritage Resource Best Management Practices (WMATCHRBMPs) present and delineate in guideline form cultural heritage resource definitions, management, and necessary steps before, during, and after project implementation for any ground-disturbing projects potentially adversely affecting cultural heritage resources on Ndee (Apache) trust lands. However, since the tribe’s adoption of the practices, the application of Ndee tenets found within the guidelines to real-world cultural and archaeological methods and practices remains scant. Embedded in the Ndee cultural tenets is the tenet of “respect,” which I will argue can be used as a tool by non- Ndee researchers to critically reflect on their own research agendas and to guide research projects with Ndee communities. By foregrounding respect within various ongoing archaeological project-related occurrences, contemporary Ndee experiences, defining Ndee material trait lists, superiority statements, archaeological categorizations, and stereotypical underpinnings, better paths forward for collaborative research with Ndee and other Native American communities can be highlighted.
