Abstract:
Endonuclease VIII-like (NEIL) 1 and 3 proteins eliminate oxidative DNA base damage and psoralen DNA interstrand crosslinks through initiation of base excision repair. Current evidence points to a DNA replication associated repair function of NEIL1 and NEIL3, correlating with induced expression of the proteins in S/G2 phases of the cell cycle. However previous attempts to express and purify recombinant human NEIL3 in an active form have been challenging. In this study, both human NEIL1 and NEIL3 have been expressed and purified from E. coli, and the DNA glycosylase activity of these two proteins confirmed using single- and double-stranded DNA oligonucleotide substrates containing the oxidative bases, 5-hydroxyuracil, 8-oxoguanine and thymine glycol. To determine the biochemical role that NEIL1 and NEIL3 play during DNA replication, model replication fork substrates were designed containing the oxidized bases at one of three specific sites relative to the fork. Results indicate that whilst specificity for 5- hydroxyuracil and thymine glycol was observed, NEIL1 acts preferentially on double-stranded DNA, including the damage upstream to the replication fork, whereas NEIL3 preferentially excises oxidized bases from single stranded DNA and within open fork structures. Thus, NEIL1 and NEIL3 act in concert to remove oxidized bases from the replication fork.

Abstract:
DNA damage caused by reactive oxygen species may result in genetic mutations or cell death. Base excision repair (BER) is the major pathway that repairs DNA oxidative damage in order to maintain genomic integrity. In mammals, eleven DNA glycosylases have been reported to initiate BER, where each recognizes a few related DNA substrate lesions with some degree of overlapping specificity. 7,8-dihydro-8-oxoguanine (8-oxoG), one of the most abundant DNA oxidative lesions, is recognized and excised mainly by 8-oxoguanine DNA glycosylase 1 (OGG1). Further oxidation of 8-oxoG generates hydantoin lesions, which are recognized by NEIL glycosylases. Here, we demonstrate that NEIL1, and to a lesser extent NEIL2, can potentially function as backup BER enzymes for OGG1 upon pharmacological inhibition or depletion of OGG1. NEIL1 recruitment kinetics and chromatin binding after DNA damage induction increase in cells treated with OGG1 inhibitor TH5487 in a dose-dependent manner, whereas NEIL2 accumulation at DNA damage sites is prolonged following OGG1 inhibition. Furthermore, depletion of OGG1 results in increased retention of NEIL1 and NEIL2 at damaged chromatin. Importantly, oxidatively stressed NEIL1- or NEIL2-depleted cells show excessive genomic 8-oxoG lesions accumulation upon OGG1 inhibition, suggesting a prospective compensatory role for NEIL1 and NEIL2. Our study thus exemplifies possible backup mechanisms within the base excision repair pathway.

Abstract:
The effects of abnormalities in the DNA glycosylases NEIL1, NEIL2, and NEIL3 on human cancer have not been fully elucidated. In this paper, we found that the median somatic total mutation loads and the median somatic single nucleotide mutation loads exhibited significant inverse correlations with the median NEIL1 and NEIL2 expression levels and a significant positive correlation with the median NEIL3 expression level using data for 13 cancer types from the Cancer Genome Atlas (TCGA) database. A subset of the cancer types exhibited reduced NEIL1 and NEIL2 expressions and elevated NEIL3 expression, and such abnormal expressions of NEIL1, NEIL2, and NEIL3 were also significantly associated with the mutation loads in cancer. As a mechanism underlying the reduced expression of NEIL1 in cancer, the epigenetic silencing ofNEIL1through promoter hypermethylation was found. Finally, we investigated the reason why an elevated NEIL3 expression level was associated with an increased number of somatic mutations in cancer and found that NEIL3 expression was positively correlated with the expression of APOBEC3B, a potent inducer of mutations, in diverse cancers. These results suggested that the abnormal expressions of NEIL1, NEIL2, and NEIL3 are involved in cancer through their association with the somatic mutation load.

Dans les oeuvres, le temps se brise: la linearite traditionnelle eclate en plusieurs modes d'apprehension du temps. Les œuvres de Neil Gaiman sont souvent qualifiées de postmodernes dans la mesure où elles lient expérimentation et autoréflexivité à une démarche de fiction populaire qui leur paraît antagoniste. A l’inverse d'une œuvre postmoderniste typique, qui met en avant sa mise à nu des mécanismes fabulateurs, les métafictions et les parodies gaimaniennes restent des fictions fantastiques et merveilleuses où les enjeux fabulateurs (psychologie et parcours des personnages, intrigue, émotion et suspens) gardent leur place primordiale. Elles ne sont donc pas des œuvres expérimentales mêlées d'éléments de fiction populaire, mais des œuvres de fiction populaire autoréflexive. Cette posture singulière fait de ces œuvres un terrain d'exploration fertile pour la réflexion sur les spécificités de la fiction populaire, et sur la place privilégiée qu'y occupe la fabulation ("storytelling"), notion qu’Henri Bergson ou Frank McConnell considéraient comme le fil rouge entre l’écriture de fiction d’aujourd'hui et la mythopoèse religieuse d’autrefois. Cette dialectique instaurée entre fabulation, mythe et fiction populaire constitue le cœur thématique des œuvres de Gaiman, qui abondent en réécritures de mythes anciens, modernes, religieux, populaires, et en représentations de personnages d’écrivains et de conteurs traditionnels, ou de personae gaimaniennes, qui, au plus fort de l’intrigue comme aux confins du paratexte, au détour d’une préface ou d’une vignette de bande dessinée, forgent une représentation de la fiction comme mythe, de l’écrivain comme figure mythique du conteur, et de la fabulation comme activité humaine quintessentielle<br>Neil Gaiman’s fantasy works are often deemed postmodernist since they experiment with self-reflexivity and mix it with a supposedly antagonistic impulse towards popular fiction. Typical postmodernist works mostly emphasize their attacks against the fictional illusions of referentiality, character or plot. On the contrary, Gaiman’s works unapologetically remain within the bounds of fantasy, a genre in which imagination and storytelling are paramount–along with their related issues: characterization, plot, emotion and suspense. Thus, Gaiman’s fiction is not experimental fiction enhanced with elements of popular fiction, but actual popular fiction made self-reflexive. Thanks to that peculiar mood, studying Gaiman’s work is a very fruitful means of investigating the distinctive features of popular fiction, and its inherent emphasis on storytelling–or, as Henri Bergson called it, “fabulation.” Both Bergson and Frank McConnell focused on this notion which they saw as the essential link between ancient mythmaking and today’s fiction writing. Gaiman’s fiction mostly relies on such a dialectics between myth, popular fiction and storytelling or fabulation. His fantasy stories constantly rewrite ancient, religious myths, modern myths or the “myths” of popular fiction, and feature many fictional and historical writers, traditional storytellers, or Gaiman’s own personae, so that in paratext as well as in the heart of a narrative, in the quiet and supposedly reliable words of an introduction as surely as on the most striking comics panels, fiction is portrayed as myth, writers as mythical avatars of some archetypal storyteller, and storytelling as the one, quintessential human act

A presente dissertação consiste em um estudo do romance Deuses americanos de Neil Gaiman levando em consideração suas conexões a outros textos bem como inserções de diversos textos provenientes de outros trabalhos na prosa do romance. A proposta de leitura do texto de Gaiman segundo este trabalho utiliza os conceitos de intertextualidade e arquétipos de forma a analisar a relação entre a trama de Deuses americanos às várias utilizações de textos cuja escrita “original” não é atribuída ao autor do livro inseridos (ou referenciados) na prosa do romance. Embora o objeto de estudo seja comumente visto como um livro difícil de ser categorizado dentre de um certo gênero, a proposta desta dissertação é demonstrar que o movimento e o fluxo contínuo de discursos (textos) e estilos na prosa do romance remonta a uma visão de um estrangeiro sobre os Estados Unidos e como o país foi criado: ou seja, que ele é não somente um ponto geográfico de confluência de muitos povos, mas também de muitas crenças e culturas que, de um modo ou outro, trouxeram os seus deuses consigo. A análise do uso de intertextos, intratextos e arquétipos no romance está estruturada em três capítulos centrais: o primeiro contextualiza os mitos que aparecem no romance e discute a questão de gênero literário do livro, além do conceito de América no texto de Gaiman. O segundo capítulo examina o uso de mitos por Gaiman em relação a outros trabalhos, tanto os manuscritos antigos de crenças pagãs quanto instâncias mais modernas de mito e alegoria, além de estudar as conexões entre Deuses americanos e outros textos escritos por Gaiman de acordo com o conceito de intratextualidade proposto por Affonso de Sant’Anna. Por fim, o terceiro capítulo se concentra no uso pontual de intertextos no romance, organizando-os entre alusões literárias, referências à cultura pop, além de estudar o conflito entre a era digital e o antigo reinado da fé religiosa, sem deixar de investigar o uso de arquétipos e apropriação na prosa do romance. O trabalho, assim, tem como objetivo verificar a alegação de que a qualidade intertextual do romance é essencial tendo em vista sua trama e cenário, bem como a afirmação de que ele redefine o conceito da América do final dos anos 90 como um espaço multicultural, dinâmico e mítico.<br>This thesis consists of a study of Neil Gaiman’s American Gods in the light of its connections to other texts as well as the punctual insertions of various texts from other works in the novel’s prose. The proposed reading of Gaiman’s text employs the concepts of intertextuality and archetypes in order to further analyze the relation of the plot of American Gods to the various uses of texts - that were not originally written by the book’s author – which are inserted (or alluded to) in the novel’s prose. Although the object of study is generally seen as a book that is hard to brand within a certain genre, this thesis’ approach to the novel demonstrates that movement and the continuous flow of speeches (texts) and styles in the novel’s prose comprises an outsider’s view of America and how the country came into existence – that is, that it is the geographical conflux not only of many peoples, but also of many beliefs and cultures, which in some way or other brought their gods with them. This examination of the use of intertexts, intratexts and archetypes in the novel is structured in three main chapters: The first chapter contextualizes the myths that appear in the novel and discusses the issues of genre and the concept of America in Gaiman’s text. The second chapter analyzes Gaiman’s use of myths in relation to other works – the original manuscripts of ancient beliefs as well as modern instances of myth and allegory – along with the connections between American Gods and Gaiman’s other works according to Affonso de Sant’Anna’s concept of intratextuality. Finally, the third chapter focuses on the punctual uses of intertexts in the novel, breaking them down into literary allusions, references to pop culture and the conflict between the digital era and the age of religious faith, and the use of archetypes and appropriation in the novel’s prose. At the end of the work, I aim to assert my belief that the intertextual nature of the novel is essential to its plot and setting, and re-defines the concept of late-90’s/early 2000’s America as a multicultural, dynamic mythical space.

Abstract:
A systematic natural convection heat transfer experiment has been carried out of nanoparticle enhanced ionic liquids (NEILs) in rectangular enclosures (lengthxwidthxheight, 50×50×50mm and 50×50×75mm) heated from below condition. In the present experiment NEIL was made of N-butyl-N-methylpyrrolidinium bis{(trifluoromethyl)sulfonyl} imide, ([C4mpyrr][NTf2]) ionic liquid with 0.5% (weight%) Al2O3 nanoparticles. In addition to characterize the natural convection behavior of NEIL, thermophysical properties such as thermal conductivity, heat capacity, and viscosity were also measured. The result shows that the thermal conductivity of NEIL enhanced ∼3% from the base ionic liquid (IL), heat capacity enhanced ∼12% over the measured temperature range. The natural convection experimental result shows consistent for two different enclosures based on the degrading natural convection heat transfer rate over the measured Rayleigh number range. Possible reasons of the degradation of natural convection heat transfer may be the relative change of the thermophysical properties of NEIL compare to the base ionic liquid.

In questo “position paper” si rappresentano obbiettivi modelli e metodi per contribuire ad&#x0D; avviare una campagna attiva di ricerca e sensibilizzazione sociale basata sull’interazione&#x0D; attraverso reti sociali e reti di automi basata nel Parco della Ricerca Enea Casaccia. Viene&#x0D; fornita un’anticipazione di quello che vuole essere il progetto “Sistemi Aperti Sapienza”; viene&#x0D; poi fornita una descrizione delle motivazioni di partenza e della riflessione che ha generato&#x0D; questa idea progettuale. Poi vengono considerate le ragioni per la convergenza di partner,&#x0D; identificati nei principali organismi della ricerca nel nostro territorio, in una organizzazione&#x0D; regionale guidata e ospitata da ENEA per raggiungere più rapidamente gli obiettivi dichiarati.&#x0D; La proposta prevede l’ implementazione presso ENEA, di un “FabLab”, un laboratorio&#x0D; finalizzato alla realizzazione di prototipi virtuali / reali, ovvero progettati attraverso software di&#x0D; modellazione 3d e stampabili attraverso stampanti 3d dall’utilizzatore finale. Ciò avverrebbe&#x0D; sulla scorta di quanto sta rapidamente diffondendosi in Italia e nel mondo sull’esempio del&#x0D; Center for Bits and Atoms (CBA) - MIT di Boston fondato da Neil Gershenfeld.&#x0D; Il FabLab Casaccia dovrebbe essere realizzato come una delle previste Officine della&#x0D; Sostenibilità specializzata nella produzione di soluzioni per la riqualificazione energetica degli&#x0D; edifici, per l’ applicazione delle energie sostenibili, per la diffusione virale di educazione e&#x0D; consapevolezza energetica e ambientale. Abstract:
Questo rapporto evidenzia come le discriminazioni strutturali e l'esclusione sociale influenzino le percezioni e gli atteggiamenti nei confronti del vaccino per il COVID-19 tra le comunità rom in Italia. Uno degli obiettivi è mettere in luce il ruolo che le autorità pubbliche e le comunità possono svolgere nel sostenere l'adozione del vaccino e nel contrasto ai più ampi processi di esclusione sociale.1 Le risposte contraddittorie che lo Stato italiano ha fornito durante la pandemia di Covid-19, insieme alle forme di esclusione già in atto, hanno comportato un aumento della sfiducia delle comunità rom nei confronti delle iniziative statali, impattando anche sull’adesione alla campagna vaccinale.2 Questo documento si propone di supportare e informare le amministrazioni locali e le istituzioni sanitarie pubbliche coinvolte nell’assistenza e nei processi di inclusione delle comunità rom in Italia. Il presente documento si basa su una ricerca condotta di persona e a distanza dal novembre 2021 al gennaio 2022 in Italia con le comunità rom e sinti di Milano, Roma e Catania. Sebbene queste comunità si caratterizzino per diversità storica e per differenti forme di identità linguistica, geografica, religiosa, sono state individuate delle somiglianze nel modo in cui hanno vissuto la pandemia di COVID-19 e nelle decisioni a proposito del vaccino. Questo documento è stato sviluppato per SSHAP da Iliana Sarafian (LSE) con i contributi e le revisioni di Elizabeth Storer (LSE), Tabitha Hrynick (IDS), Marco Solimene (University of Iceland), Dijana Pavlovic (Upre Roma) e Olivia Tulloch (Anthrologica). In the housing literature, Sweden is often portrayed as a so-called “cost-rental” society associated with tenure neutrality, where rental housing should be an attractive alternative – and not just a step in the way – to homeownership. A large and well-developed rental sector has traditionally made it possible for young adults to leave their family home at a relatively young age. However, this logic has been clearly disrupted as rental housing has become harder to access and homeownership has been favoured by incremental ideological political shifts and fiscal policy encouraging homeownership. As more households – also young ones – are steered into homeownership, Sweden has become one of the most mortgage-indebted nations in the OECD. This working paper on homeownership and mortgagization takes on the question of mortgaged indebtedness in discourse and practice. The working paper is the joint product of two different studies written as part of the research internship in the project “Financialisation of everyday life. Intersectional perspectives on housing and labor precarity” at Malmö university, led by Chiara Valli. In the first section, Neil Dunne presents a discourse analysis on how homeownership has been discussed in the largest newspapers in Sweden over the last decades, while Greta Cattabriga and Nathan O’Néill in the second section discuss perceptions and lived experiences of mortgaged homeownership on the basis of interviews with young adults. Put together, the two studies contribute with significant additions to the discussion about whether Sweden is moving towards a homeownership society and what the potential consequences are for young adults.
